A new compound named koninginin W ( 1 ) and four known polyketides ( 2 – 5 ) were isolated from endophytic fungus Trichoderma koningiopsis YIM PH30002 of Panax notoginseng. The structures of 1 - 5 , including absolute configuration of 1 , were elucidated on the detailed analysis of the HR-ESI-MS, 1D and 2D NMR, and X-ray crystallographic data. Koninginin W ( 1 ) presented weak antibacterial activity against Escherichia coli, Bacillus subtilis and Salmonella typhimurium.  相似文献

A new cytochalasan alkaloid, westalsan ( 1 ), along with two known cytochalasan compounds, phomacin B ( 2 ) and 19-hydroxy-19,20-dihydrophomacin C ( 3 ), were isolated from the solid rice culture of Westerdykella nigra, a marine-derived endophytic fungus, isolated from the roots of mangrove Avicennia marina (Forssk.) Vierh. The structures of compounds 1 – 3 were established on the basis of extensive 1D and 2D NMR spectroscopic techniques in combination with HR-ESI-MS. The ability of the isolated compounds to inhibit acetylcholine esterase activity was evaluated. Compound 3 showed the highest acetylcholine esterase inhibitory activity (IC50 0.056±0.003 μM), followed by compound 1 (IC50 0.088±0.005 μM) and compound 2 (IC50 0.140±0.007 μM) compared to donepezil (IC50 0.035±0.002 μM). This was further confirmed by molecular docking experiment.  相似文献

Bioassay‐guided isolation of a fungal strain Nigrospora sp. MA75, an endophytic fungus obtained from the marine semi‐mangrove plant Pongamia pinnata, which was fermented on three different culture media, resulted in the isolation and identification of seven known compounds, 2, 3 , and 5 – 9 , from a medium containing 3.5% NaCl, while a new compound, 2,3‐didehydro‐19α‐hydroxy‐14‐epicochlioquinone B ( 10 ) was obtained from the medium containing 3.5% NaI. In addition, two new griseofulvin derivatives, 6‐O‐desmethyldechlorogriseofulvin ( 1 ) and 6′‐hydroxygriseofulvin ( 4 ), were isolated and identified from the rice solid medium. Dechlorogriseofulvin ( 2 ) and griseofulvin ( 3 ) were the major components in fermentation extracts of all these culture media, while compounds 1 and 4, 5 and 6 , and 10 were only present in the extract of respective culture medium. The structures of these compounds were elucidated by detailed spectroscopic analysis, and the absolute configuration of 1 was determined by CD measurement. Compounds 9 and 10 exhibited antibacterial activities toward five tested bacterial strains, while compounds 5, 6 , and 8 selectively inhibited MRSA, E. coli, and S. epidermidis, and compound 3 showed moderate activity against V. mali and S. solani. Moreover, compound 10 potently inhibited the growth of MCF‐7, SW1990, and SMMC7721 tumor cell lines with IC50 values of 4, 5, and 7 μg/ml, respectively.  相似文献

Two new C13‐polyketides, aureonitols A and B ( 1 and 2 ), along with five known compounds ( 3 – 7 ), were isolated from the solid fermentation culture of the plant endophytic fungus Chaetomium globosum from the aerial parts of Salvia miltiorrhiza. The structures and absolute configurations of 1 and 2 were determined by comprehensive spectroscopic data analysis and computed methods. Compound 5 was found to display the remarkable antimicrobial activities against four multidrug‐resistant bacteria (Enterococcus faecalis, Enterococcus faecium, Staphylococcus aureus, and Staphylococcus epidermidis) with MIC values of 3.13–6.25 μg/mL (ciprofloxacin: 0.78–1.56 μg/mL), and also against all tested fungal strains with MIC values of 3.13–25 μg/mL (ketoconazole: 0.78–12.50 μg/mL).  相似文献

Two new sorbicillinoids, 1 and 2 , together with a novel benzofuranone derivative named phialofurone ( 3 ), were isolated from a deep‐sea sediment‐derived fungus, Phialocephala sp. Their structures were established on the basis of spectroscopic data. All compounds displayed cytotoxic effects against P388 (IC50 values of 11.5±1.4, 0.1±0.1, and 0.2±0.01 μM , resp.) and K562 (IC50 values of 22.9±0.8, 4.8±0.3 and 22.4±0.9 μM , resp.) cell lines.  相似文献

The first natural S‐containing benzophenone dimer, named guignasulfide ( 3 ), was isolated from the culture of Guignardia sp. IFB‐E028, an endophytic fungus residing in healthy leaves of Hopea hainanensis. Its structure was determined through correlative analyses of its MS, 1D‐ and 2D‐NMR spectroscopic data. Two other known benzophenone derivatives, monomethylsulochrin and rhizoctonic acid ( 1 and 2 , resp.) were also isolated. Guignasulfide ( 3 ) was more active against the human liver cancer cell line HepG2 (IC50 value: 5.2±0.4 μM ) than metabolites 1 and 2 (IC50 values: 63.5±0.6 and 60.2±0.5 μM ); compounds 1 – 3 showed also moderately inhibitory effects on the human bacterial pathogen Helicobacter pylori with MIC values of 28.9±0.1, 60.2±0.4, and 42.9±0.5 μM , respectively.  相似文献

Three new cytochalasins Z24, Z25, Z26 ( 1 – 3 , resp.) and one known compound, scoparasin B ( 4 ), were isolated from the fungus Eutypella sp. D‐1 isolated from the soil of high latitude of the Arctic. The structures of 1 – 3 were elucidated from spectroscopic data (NMR, MS). These compounds were evaluated for cytotoxic activities against several human tumor cell lines. Among them, compound 1 exhibited moderate cytotoxicity toward human breast cancer MCF‐7 cell line with IC50 of 9.33 μM .  相似文献

Three new diphenyl ether derivatives, talaromycins A–C ( 1 – 3 , resp.), together with six known analogs, 4 – 9 , were isolated from a gorgonian‐derived fungus, Talaromyces sp. The structures of the new compounds were determined by analysis of extensive NMR spectroscopic data. All of the isolated metabolites, 1 – 9 , were evaluated for their cytotoxic and antifouling activities. Compound 4 exhibited pronounced cytotoxicity against the tested human cell lines with the IC50 values ranging from 4.3 to 9.8 μM . Compounds 3, 5, 8 , and 9 showed potent antifouling activities against the larval settlement of the barnacle Balanus amphitrite with the EC50 values ranging from 2.2 to 4.8 μg/ml.  相似文献

Manipulation of the fermentation of the marine‐derived fungus Penicillium chrysogenum by addition of CaBr2 resulted in induced production of bromodiphenyl ether analogs. Two new free‐radical‐scavenging polybrominated diphenyl ethers, 1 and 2 , and three known diphenyl ethers, 3,3′‐dihydroxy‐5,5′‐dimethyldiphenyl ether ( 3 ), and an inseparable mixture of violacerol‐I ( 4 ) and violacerol‐II ( 5 ) were isolated. The structures of the two new polybromodiphenyl ethers 1 and 2 were assigned by combined spectroscopic‐data analysis, including deuterium‐induced isotope effect. Compounds 1 – 3 , and a mixture of 4 and 5 exhibited radical‐scavenging activities against 1,1‐diphenyl‐2‐picrylhydrazyl with IC50 values of 18, 15, 42, and 6 μM , respectively. With the exception of 3 , the compounds were, therefore, more active than the positive control, ascorbic acid (IC50 20 μM ).  相似文献

A new polyaromatic metabolite, ent-herqueidiketal ( 1 ), and a new phenalenone derivative, epi-peniciherqueinone ( 2 ), along with twelve known compounds 3 – 14 , were isolated from the fungus Penicillium herquei YNJ-35, a symbiotic fungus of Pulveroboletus brunneopunctatus collected from Nangunhe Nature Reserve, Yunnan Province, China. The structures of 1 – 14 and the absolute configurations of 1 and 2 were determined by their spectroscopic data or by their single-crystal X-ray diffraction analysis or optical rotation values. Compound 1 showed strong antibacterial activity against Staphylococcus aureus (ATCC 29213) with minimum inhibitory concentration (MIC) of 8 μg/mL. In the cytotoxicity assays, compound 1 showed weak inhibitory activity against breast cancer MCF-7 and mice microglial BV2 cells with half maximal inhibitory concentration (IC50) of 17.58 and 29.56 μM; compound 14 showed stronger cytotoxicity against BV2 and MCF-7 cells with IC50 values of 6.57 and 10.26 μM.  相似文献

Addition of the valproic acid (histone deacetylases inhibitor) to a culture of an endophytic fungus Diaporthe sp. harbored from Datura inoxia significantly altered its secondary metabolic profile and resulted in the isolation of three novel compounds, identified as xylarolide A (1), diportharine A (2) and xylarolide B (3) along with one known compound xylarolide (4). The structures of all the compounds (14) were determined by detailed analysis of 1D and 2D NMR spectroscopic data. The relative configurations of compounds 13 were determined with the help of NOESY data and comparison of optical rotations with similar compounds with established stereochemistry. All the isolated compounds were screened for antibacterial, antioxidant and cytotoxic activities. Xylarolide A (1) and xylarolide (4) displayed significant growth inhibition of MIAPaCa-2 with an IC50 of 20 and 32?µM respectively and against PC-3 with an IC50 of 14 and 18?µM respectively. Moreover, compound 1 displayed significant DPPH scavenging activity with EC50 of 10.3?µM using ascorbic acid as a positive control.  相似文献

Six new polyketides aplojaveediins A–F (16) were isolated from the endophytic fungus Aplosporella javeedii associated with the host plant Orychophragmus violaceus (Brassicaceae). The structures of the new metabolites were elucidated by analysis of their NMR and MS data. Compound 1 exhibited antifungal activity against the hyphae form of Candida albicans strain ATCC 24433 in the agar plate diffusion assay and the microbroth dilution assay. The kinetic of killing of C. albicans cells for compound 1 was considerably faster than that of the positive control hygromycin B. Compounds 1 and 6 also exhibited moderate antibacterial activities against sensitive (ATCC 29213) and drug-resistant (ATCC 700699) strains of Staphylococcus aureus.  相似文献

Activity‐guided purification of a MeOH extract of the Korean wild mushroom Boletus pseudocalopus afforded three new grifolin derivatives, 1 – 3 , along with four known phenolic compounds 4 – 7 . Their structures were established by a combination of 1H‐ and 13C‐NMR, NOESY, and extensive two‐dimensional NMR spectroscopic experiments such as gCOSY, gHSQC, gHMBC, and ROESY. The major metabolites 4 and 5 were subjected to reduction to provide the side chain‐reduced compounds 8 and 9 for biological testing. All of the compounds except compound 6 showed anticancer activities in the range of IC50 3.5–11.0 μg/ml against human lung carcinoma A549 and mouse melanoma B16F1 cell lines. In addition, all compounds showed moderate radical‐scavenging activities determined by DPPH assay.  相似文献

A new caryophyllene, named pulicaryenne A ( 1 ), along with four other known caryophyllene derivatives ( 2 , 3 , 4 and 5 ) were isolated from the ethyl acetate extract of aerial parts of Pulicaria vulgaris Gaertn . (Asteraceae). All compounds were isolated for the first time from this species. Compound 2 was identified as a new epimer of a known caryophyllene derivative isolated previously from P. dysenterica. Their structures were established by spectroscopic means including NMR analysis (1D‐ and 2D‐NMR) and ESI‐TOF‐MS. All compounds were evaluated for their anticholinesterase, antityrosinase and cytotoxic activities against two human cell lines (A549 and HeLa). Results showed that compound 5 exhibited the highest cytotoxic effect against A549 and anticholinesterase activity with IC50 values of 8.50±0.75 and 6.45±0.09 μg/mL, respectively. Furthermore, compound 5 showed also an interesting antityrosinase activity with percent inhibition value of 79.0±2.5 % at 50 μg/mL. The bioactivity and drug likeness scores of the isolated compounds 1–5 were calculated using Molinspiration software and discussed. These results may suggest that the five caryophyllene derivatives endowed with good biological properties, which could be used as bioactive alternatives in pharmaceutical preparations.  相似文献

A novel series of aminopyrimidinylisoindoline derivatives 1a-w having an aminopyrimidine scaffold as a hinge region binding motif were designed and synthesized. Among them, six compounds showed potent inhibitory activities against AXL kinase with IC50 values of submicromolar range. Especially, compound 1u possessing (4-acetylpiperazin-1-yl)phenyl moiety exhibited extremely excellent efficacy (IC50?=?<0.00050?μM). Their in vitro antiproliferative activities were tested over five cancer cell lines. Most compounds showed good antiproliferative activities against HeLa cell line. The kinase panel profiling of 50 different kinases and the selected inhibitory activities for the representative compound 1u were carried out. The compound 1u exhibited excellent inhibitory activities (IC50?=?<0.00050, 0.025, and 0.050?μM for AXL, MER, and TYRO3, respectively) against TAM family, together with potent antiproliferative activity against MV4-11 cell line (GI50?=?0.10?μM) related to acute myeloid leukemia (AML).  相似文献

Two new α‐pyrones (=2H‐pyran‐2‐ones), ficipyrones A and B ( 1 and 2 , resp.), and two new α‐furanones (=2H‐furan‐2‐ones), ficifuranones A and B ( 3 and 4 , resp.), together with three known metabolites, antibiotic F 0368 ( 5 ), hydroxyseiridin ( 6 ), and hydroxyisoseiridin ( 7 ), were isolated from solid cultures of the plant endophytic fungus Pestalotiopsis fici. Their structures were elucidated primarily by NMR spectroscopy, and the absolute configuration of 1 was deduced from the circular‐dichroism (CD) data. Two new steroids, named acrocalysterols A (1) and B (2), together with five known compounds were isolated and characterized from an endophytic fungus Acrocalymma sp., isolated from the tender stems of Sinomenium acutum. The structures of the isolated metabolites were identified based on UV, IR NMR (1D and 2D), HR-ESI-TOF-MS, single-crystal X-ray diffraction experiment using Cu Kα radiation and CD spectra associated with TD-DFT calculation. The bioassay showed that compound 2 displayed potent cytotoxicity against three cancer cell lines HeLa, HCC-1806, and RKO with IC50 values ranging from 18.37 to 19.64 μM.  相似文献

The chemical compositions of essential oils (EOs) extracted from Curcuma kwangsiensis rhizomes collected from six natural habitats in P. R. China were evaluated using gas chromatography/mass spectrometry (GC/MS). Fifty‐seven components were identified from the six EOs, and their main constituents were 8,9‐dehydro‐9‐formyl‐cycloisolongifolene (2.37 – 42.59%), germacrone (6.53 – 22.20%), and l ‐camphor (0.19 – 6.12%). The six EOs exhibited different DPPH radical‐scavenging activities (IC50, 2.24 – 31.03 μg/ml), with the activity of most of EOs being much higher than that of Trolox C (IC50, 10.49 μg/ml) and BHT (IC50, 54.13 μg/ml). Most EOs had potent antimicrobial effects against the tested bacteria and fungus. They also exhibited cytotoxicity against B16 (IC50, 4.44 – 147.4 μg/ml) and LNCaP cells (IC50, 73.94 – 429.25 μg/ml). The EOs showed excellent anti‐inflammatory action by significantly downregulating expression of pro‐inflammatory cytokines, cyclooxygenase‐2, and tumor necrosis factor‐α. This study provides insight into the interrelation among growth location, phytoconstituents, and bioactivities, and the results indicate the potential of C. kwangsiensis as natural nutrients, medicines, and others additives.  相似文献

A new fructofuranoside glycerol, dryoptkirbioside ( 1 ), along with thirteen known compounds ( 2 - 14 ), was isolated from the MeOH extract of Dryopteris kirbi rhizomes by silica gel column chromatography, Sephadex LH-20 column chromatography, and semipreparative HPLC. The structure of the new compound was determined by analyses of its spectroscopic data including nuclear magnetic resonance (NMR), and high-resolution electrospray ionisation mass spectrometry (HR-ESI-MS) and chemical conversions. The hexane-soluble portion and the EAFA fraction showed strong activities against lung (A549), breast (MCF-7), and cervical (HeLa) human cancer cell lines (IC50 values ranging from 4.0 to 8.8 μg/mL). Aspidinol P ( 5 ) and aspidinol B ( 6 ) exhibited moderate to low cytotoxicity on the three cell lines (IC50 values ranging from 20.4 to 58.7 μM). The MeOH extract and hexane-soluble portion had excellent activities against Staphylococcus aureus and Bacillus subtilis (MICs 11.7 and 23.4 μg/mL), whereas the AcOEt- and BuOH-soluble portions were significantly active on S. aureus (MICs 46.9 and 93.8 μg/mL). The main fractions EAFB, EAFC and nBFB displayed excellent activity against S. aureus (MICs 11.7 and 23.4 μg/mL). Aspidinol B ( 6 ) had significant activity, while aspidinol P ( 5 ) was moderately active against S. aureus and B. subtilis (MICs 42.0 and 89.5 μM).  相似文献
