During 1986–87 57% of 300 chicken carcasses yielded salmonellas where tested by a swabbing method. Serotypes isolated were Salmonella enteritidis (66%), Salm. agona (12%), Salm. newport (6%), Salm. saintpaul (6%), Salm. derby (4%), Salm. typhimu-rium (3%), Salm. bardo (1%), Salm. ohio (1%) and untypable (2%). The results are compared with those of avian and human salmonellosis registered in Portugal during the same period.  相似文献

AIMS: To determine the prevalence of Salmonella enterica serovars in chicken carcasses in slaughterhouses in Spain and to examine genotypic relations among these serovars. METHODS AND RESULTS: A total of 336 chicken carcasses were collected from six slaughterhouses in Northwestern Spain. Salmonellae were isolated (ISO-6579-1993), serotyped, phage-typed, ribotyped and antibiotyped against 20 antibiotics. Salmonella strains were detected in 60 (17.9%) carcasses. Isolates belonged to nine different serotypes, with Salm. Enteritidis being the most common. Three strains (5%) were resistant to one antibiotic and 24 (40%) were multi-resistant (to more than one antibiotic). The most frequently encountered resistances were to sulphamides, fluoroquinolones and tetracycline. Ribotyping was able to differentiate isolates of the same serotype and phage type. CONCLUSIONS: The Salmonella serotypes and phage types detected are among those most frequently associated with human diseases in Spain. The large percentage of antimicrobial resistant strains is a matter for concern. A high genetic relationship between strains from different slaughterhouses was found. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides detailed information about Salmonella isolates from poultry in Spain. It emphasizes the importance of controlling this pathogen in poultry products, and suggests the need for more prudent use of antibiotics.  相似文献

Objective:  To determine the effect of refrigeration time and temperature on Salmonella cell numbers on inoculated chicken carcasses and their transfer to a plastic cutting board.
Methods and Results:  The survival of Salmonella on chicken skin and the transfer to a plastic cutting board when exposed to different refrigeration temperatures (2, 6 or 8°C) for 9 days were the two main issues on which this work focused. Two scenarios were carried out to ascertain these effects: carcasses treated with a decontaminating acetic acid solution and untreated carcasses. All of the contaminated carcasses remained contaminated after 9 days of refrigeration. However, on untreated samples, while Salmonella numbers increased almost 1·5 log at 8°C, the pathogen numbers decreased about 1 log at 2 and 6°C. On acid-treated samples, cell numbers slightly decreased at all of the temperatures studied. Temperature did not affect salmonellae transfer to the cutting board, but time did. Acid decontamination increased cell numbers transferred to the cutting board compared with untreated samples.
Conclusion:  Proper refrigeration at low temperatures did not allow Salmonella numbers to rise, regardless of which carcasses had been, or had not been, acid treated. Despite the fact that the rate of transfer was not affected by temperature, the acid treatment detached Salmonella cells from the chicken skin and, therefore, the probability of greater cross-contamination should be studied further.
Significance and Impact of the Study:  The results of this study may provide better information about the refrigeration conditions for fresh chicken storage and also determine if these, along with acetic acid decontamination of broiler chicken, would affect the pathogen transfer to a cutting board.  相似文献

While measures to control carcass contamination with Salmonella at the processing plant have been implemented with some success, on-farm interventions that reduce Salmonella prevalence in meat birds entering the processing plant have not translated well on a commercial scale. We determined the impact of Salmonella vaccination on commercial poultry operations by monitoring four vaccinated and four nonvaccinated breeder (parental) chicken flocks and comparing Salmonella prevalences in these flocks and their broiler, meat bird progeny. For one poultry company, their young breeders were vaccinated by using a live-attenuated Salmonella enterica serovar Typhimurium vaccine (Megan VAC-1) followed by a killed Salmonella bacterin consisting of S. enterica serovar Berta and S. enterica serovar Kentucky. The other participating poultry company did not vaccinate their breeders or broilers. The analysis revealed that vaccinated hens had a lower prevalence of Salmonella in the ceca (38.3% versus 64.2%; P < 0.001) and the reproductive tracts (14.22% versus 51.7%; P < 0.001). We also observed a lower Salmonella prevalence in broiler chicks (18.1% versus 33.5%; P < 0.001), acquired from vaccinated breeders, when placed at the broiler farms contracted with the poultry company. Broiler chicken farms populated with chicks from vaccinated breeders also tended to have fewer environmental samples containing Salmonella (14.4% versus 30.1%; P < 0.001). There was a lower Salmonella prevalence in broilers entering the processing plants (23.4% versus 33.5%; P < 0.001) for the poultry company that utilized this Salmonella vaccination program for its breeders. Investigation of other company-associated factors did not indicate that the difference between companies could be attributed to measures other than the vaccination program.  相似文献

This study was conducted to isolate psychrotrophic lactic acid bacteria (LAB) from chicken carcasses with inhibitory activity against strains of Salmonella spp. and Listeria monocytogenes. A total of 100 broiler samples were examined for the presence of LAB. Ninety-two LAB isolates that showed antimicrobial effects against Salmonella spp. and L. monocytogenes were further analysed to examine their LAB (Gram-positive, catalase negative, oxidase negative) and psychrotrophic characteristics (ability to grow at 7 °C). Fifty isolates were further selected and identified initially using standard biochemical tests in miniature (Micro-kits API CH 50) and then by sequencing of the 16s-23s rRNA gene boundary region (Intergenic Spacer Region). By molecular identification, these isolates were classified into 5 different LAB species: Lactobacillus salivarius, Lactobacillus reuteri, Lactobacillus johnsonii, Pediococcus acidilactici, and Lactobacillus paralimentarius. None of the isolates produced tyramine or histamine.  相似文献

Aim: To confirm the reliability and sensitivity of Salmonella testing of processed poultry in Australia. Methods and Results: The detection of Salmonella in a whole carcass wash of 90 randomly selected processed broilers was compared using the Australian Standard method, an Australian industry method used by a major processor and the United States Department of Agriculture method published in the Federal Register. The sensitivity of each method was determined using a carcass wash containing a known number of Salmonella Typhimurium to determine the minimum concentration to be able to be identified as positive. The two Australian methods were found to be comparable with both the Australian methods detecting more positive carcasses than the United States Department of Agriculture (USDA) method. The Australian methods were sensitive at the level of 1–3 CFU ml?1 and the USDA method was sensitive at 10–30 CFU ml?1. Conclusions: The Australian Standard method and the Australian industry method were both able to detect Salmonella reliably even at a low level of contamination. Significance and Impact of the Study: This study gives a high level of confidence both to the operators of poultry‐processing plants and to regulators dependent upon the outcome of Salmonella testing for process control in Australia.  相似文献

Plasmids conferring tellurite resistance were transferred at low temperature (27 degrees C) from Escherichia coli strains isolated from chicken carcasses at the time of slaughter and after storage. They belonged to group IncH, as evidenced by their large molecular weight and incompatibility with plasmid pIP233. From January 2007 until June 2008, 504 reptiles of four families and 57 species were examined for Salmonella by using cloacal or intestinal swabs. Salmonella was identified in 139 (28%) of the 504 animals tested. Of the 504 reptiles examined, 210 were captive and 294 were wild. Ninety-eight (47%) of the captive reptiles were shedding Salmonella at the time of sampling. In contrast, only 41 (14%) of the wild reptiles were shedding Salmonella. The higher prevalence of Salmonella in captive reptiles was statistically significant (P<0.0001). No Salmonella was found in 60 wild, freshwater chelonians or 48 wild southern water skinks (Eulamprus heatwolei). Our results suggest that some species of wild reptiles in Australia are not natural carriers of Salmonella and that diet and captivity may influence Salmonella excretion in other species.  相似文献

AIMS: A comparison of the prevalence of Salmonella in chicken carcasses with and without visible faecal contamination during commercial slaughter practice was made. The relationship between Enterobacteriaceae, coliform and Escherichia coli counts and Salmonella status was also evaluated to establish the likelihood of using these groups as 'index' organisms to predict the presence of pathogen. METHODS AND RESULTS: Samples were removed immediately after evisceration, after the inside-outside shower and after chilling from the processing line for microbiological analysis. Of the carcasses visibly uncontaminated with faeces after the evisceration step 20% harboured salmonellas and 20.8% of the visibly contaminated carcasses were positive for the pathogen. When E. coli, coliforms and Enterobacteriaceae were used as predictor variables the error rates ranged from 33.3 to 60% for both sample types. CONCLUSIONS: There was no indication that any of the groups of organisms analysed could predict the incidence of salmonellas on the samples studied. Positive results for the pathogen were obtained at every tested step of the slaughtering process regardless of whether or not faecal contamination was present. SIGNIFICANCE AND IMPACT OF THE STUDY: The present study demonstrated that carcasses not visibly contaminated with faeces carried Salmonella as well as the visibly contaminated carcasses.  相似文献

The distribution of species and serotypes of Salmonella among 2,498 cultures which were isolated in the United States and its territories is presented. These isolates were received for examination during the 12-month period between October 1, 1966 and September 30, 1967. These and other data obtained from the Salmonella Surveillance Summaries for the past five years indicate that a relatively small number of species and serotypes of Salmonella are regularly isolated from diagnostic specimens. Of approximately 1,300 presently known Salmonella species and serotypes, 33 account for almost 90% of the isolates reported from humans and approximately 80% of the isolates from nonhuman sources. The 50 most prevalent species and serotypes account for 97% of the isolates from humans. An abbreviated antigenic schema based on these 50 species and serotypes of Salmonella, in conjunction with adequate biochemical tests, permits complete bacteriological characterization of the common Salmonella  相似文献

Seventeen Salmonella enterica serovar Hadar isolates recovered from bison were found to possess a range of virulence genes and resistance to tetracycline, gentamicin, sulfamethoxazole, and streptomycin simultaneously. A 1-kb class 1 integron containing the aadA1 gene was identified in all isolates. Campylobacter jejuni was found in 82.9% of 94 chicken wing packages analyzed on the day of arrival at supermarkets and in 15.5% of 45 packages obtained from the supermarket shelves a few days later. Swarming behavior among 167 Salmonella sp. isolates, representing all eight groups, was assessed. Only eight strains failed to swarm under standard conditions. Four of the defective strains swarmed on alternate carbon sources, and four harbored general defects in motility or lipopolysaccharide. The presence of Salmonella and its relationship with indicator organisms of fecal pollution, such as total coliforms, fecal coliforms and fecal streptococci, was studied at two marine zones in Portugal. Seventeen different Salmonella serotypes were isolated and identified, S. virchow was the most frequently isolated (21.6%). In addition, a high percentage (35.1%) was recorded for some Salmonella serotypes of clinical significance, namely S. enteritidis, S. infantis, S. typhimurium and S. virchow. In any of the samples from the two zones Salmonella was not detected in the absence of any of the indicator organisms. However, the incidence of Salmonella as a function of indicator concentration intervals established by the EEC standards was 0, 10 and 19.3% at guide values of total coliforms, fecal coliforms and fecal streptococci, respectively in the Faro samples (south of Portugal). In contrast, Salmonella incidence rates of 37.5, 36.4 and 33.3% were recorded at the corresponding guide values the Caminha samples (north of Portugal). No significant correlations (p>0.005) were obtained between Salmonella and the indicators at the sampling stations; however, total coliforms and fecal streptococci were the indicators most closely related to Salmonella in Caminha and Faro samples, respectively. Survival experiments in Escherichia coli, Enterococcus faecalis and S. typhimurium, using diffusion chambers, were performed to verify whether the lack of correlation between indicators and Salmonella was due to different inactivation rates in seawater. The results indicate that survival percentages of the three microorganisms tested were similar after 48 h of exposure to seawater.  相似文献

Immunomagnetic separation techniques were used in the isolation of salmonella from raw chicken carcasses. Improved isolation rates were achieved with increased specificity and decreased processing time, although several technical difficulties remain to be addressed. Immunomagnetic separation offers significant potential for improvement on existing microbiological systems for the isolation of salmonella.  相似文献
