A helix-loop-helix within the group I intron has most of the canonical sequence elements of a kink turn (K-turn), yet it bends in the opposite direction. The reverse K-turn kinks toward the major rather than the minor grooves of the flanking helices. This suggests that there are two distinct subclasses of tertiary structures that a K-turn secondary structure can adopt. The final structure may be specified by external factors, such as protein binding or the tertiary structural context, rather than the intrinsic conformation of the RNA.  相似文献

Complex, process-based dynamic models are used to attempt to mimic the intrinsic variability of the natural environment, ecosystem functioning and, ultimately, to predict future change. Palaeoecological data provide the means for understanding past ecosystem change and are the main source of information for validating long-term model behaviour. As global ecosystems become increasingly stressed by, for example, climate change, human activities and invasive species, there is an even greater need to learn from the past and to strengthen links between models and palaeoecological data. Using examples from terrestrial and aquatic ecosystems, we suggest that better interactions between modellers and palaeoecologists can help understand the complexity of past changes. With increased synergy between the two approaches, there will be a better understanding of past and present environmental change and, hence, an improvement in our ability to predict future changes.  相似文献

In all living cells, DNA is constantly threatened by both endogenous and exogenous agents. In order to protect genetic information, all cells have developed a sophisticated network of proteins, which constantly monitor genomic integrity. This network, termed the DNA damage response, senses and signals the presence of DNA damage to effect numerous biological responses, including DNA repair, transient cell cycle arrests (“checkpoints”) and apoptosis. The MRN complex (MRX in yeast), composed of Mre11, Rad50 and Nbs1 (Xrs2), is a key component of the immediate early response to DNA damage, involved in a cross-talk between the repair and checkpoint machinery. Using its ability to bind DNA ends, it is ideally placed to sense and signal the presence of double strand breaks and plays an important role in DNA repair and cellular survival. Here, we summarise recent observation on MRN structure, function, regulation and emerging mechanisms by which the MRN nano-machinery protects genomic integrity. Finally, we discuss the biological significance of the unique MRN structure and summarise the emerging sequence of early events of the response to double strand breaks orchestrated by the MRN complex.  相似文献

Summary The ultrastructural distribution of fibronectin immunoreactivity was investigated in the chicken embryo during late gastrulation. Sites of binding of anti-fibronectin antibodies were ascribed to the basal lamina and associated structures, and to the cell surface. The fibronectin-rich basal lamina was resolved into (1) a lamina densa, which appears as a continuous, dense sheet, (2) a lamina lucida, consisting of anchoring cords between lamina densa and epithelial cells, and (3) a lamina intima, closely juxtaposed to the cell surface. Cell-surface labelling was also observed in mesoblast cells, and along the dorsal side of the deep-layer cells. The ventral side of the latter cells was poorly stained in the endophyllic crescent, except in coated pits, and more regularly stained at the level of definitive endoblast. Some structures associated with the basal lamina reacted intensely with anti-fibronectin antibodies. These are (1) the interstitial bodies, which are aggregates of extracellular material, and (2) a kind of fibril or tubule, embedded in a fibronectin matrix and mainly found in the endophyllic crescent. Some intracellular labelling was found in most deep-layer cells, in few epiblast cells, never in mesoblast cells. These results extend previous studies on the localization of fibronectin, and correlate its presence and surface topology with its postulated role in migration of mesoblast cells on the basal lamina which, chemically, constitutes an appropriate substrate.  相似文献

Calumenin belongs to a family of multiple EF-hand proteins that include reticulocalbin, ERC-55, and Cab45. Reticulocalbin and ERC-55 localize to the ER due to a C-terminal HDEL retrieval signal. Cab45 contains a HEEF C-terminal sequence and is localized to the Golgi apparatus. The murine homologue of calumenin is reported to be present in the ER due to a C-terminal HDEF retrieval signal. The human homologue differs from the murine at 7 amino acid positions but the HDEF signal is conserved. However, in the cultured human cell lines, HaCaT keratinocytes, normal and transformed MRC-5 fibroblasts, as well as in transfected COS-1 cells, human calumenin could be demonstrated in the ER as well as in the Golgi complex. Especially in MRC-5 cells, a certain heterogeneity was observed, with some of the cells having calumenin localized solely to the ER while in other cells calumenin could be demonstrated in the ER as well as in the Golgi complex. Immunoelectron microscopy of placental syncytiotrophoblast cells showed that a substantial fraction of calumenin is localized in close association with the ER membrane. In addition, the protein may be recovered from the medium of cultured cells in an endoglycosidase H-resistant form, suggesting that the glycosylated protein has been further modified in the Golgi apparatus and secreted to the medium.  相似文献

In eukaryotes, DNA replication requires the regulated assembly of pre-replicative complexes (pre-RCs) onto DNA during G1 phase. Pre-RCs render the chromatin competent to replicate, yet it is only at the G1-S phase transition that protein-kinase complexes trigger the transition to DNA replication. Central to the formation of pre-RCs and regulation of DNA replication is the Cdc6 protein. Two recent studies have shown that Cdc6 is the long-sought factor that confers the competence to replicate in unfertilized Xenopus eggs.  相似文献

目的:探讨双胶润通胶囊的润肠通便作用的效果及剂量.方法:实验采用动物试验,将小白鼠(雌雄各半,体重18~22 g)分成两个实验组,每组50只,实验一组进行小肠推进试验,实验二组进行排便试验,时间均为7 d.按推荐剂量每人每日5 g设计,分为低、中、高3个剂量组,另设空白对照和模型对照组.结果:在不影响小鼠体重的情况下,双胶润通胶囊高剂量组小肠推进率与模型对照组比较差异均有显著性变化(P《0.05);3个剂量组小鼠的首便时间、粪便粒数均高于便秘模型对照组,差异具有极显著性差异(P《0.01);结论:双胶润通胶囊对小鼠小肠运动有增强作用,并能缩短小鼠首便时间、增加小鼠的粪便粒数与重量,即:具有润肠通便作用.  相似文献
