In vitro-Verfahren zur Bestimmung der DNS-Synthese-Dauer einzelner Zellen Biochemische Voraussetzungen und Ergebnisse

Zusammenfassung Nach Hemmung der de novo-Synthese von TMP durch 5-F-UdR läuft die DNS-Synthese in vitro in Anwesenheit von 10^–5 M/l mit unveränderter Geschwindigkeit ab. Dadurch ist die DNS-Synthese-Dauer über die quantitative autoradiographische Messung des ¹⁴C-TdR-Einbaus bestimmbar. Diese in vitro-Methode wird an Rattenknochenmark und Zellen einer CML geprüft. Erste Ergebnisse zeigen eine gute Übereinstimmung mit in vivo-Messungen durch andere Methoden.

In vitro determination of the duration of DNA synthesis of individual cells

Summary Suspended haemopoietic cells are incubated in the presence of 5-fluoro-deoxyuridine (5-F-UdR) as an inhibitor of the de novo synthesis of thymidine monophosphate (TMP). The DNA synthesis rate remains undisturbed, if 10^–5 M/l thymidine (TdR) is added. Thus, the DNA synthesis rate of individual cells can be determined by means of a quantitative autoradiographic method in measuring the incorporation rate of ¹⁴C-TdR into the DNA of the cells. DNA synthesis rates are then converted into DNA synthesis times. This in vitro method has been checked in cells of rat bone marrow and of peripheral blood in a case of chronic myelocytic leukaemia (CML). Preliminary results correlate well with in vivo estimations obtained by other methods.

Zur Differentialdiagnose von Papillon-Léage-Psaume-Syndrom und Mohr-Syndrom

Zusammenfassung Das Papillon-Léage-Psaume-(Oro-facio-digitales-)Syndrom und das Mohr-Syndrom sind genetisch eigenständige und auch klinisch unterscheidbare Symptomenkomplexe. Einige Patienten, die früher als dem Papillon-Léage-Psaume-Syndrom zugehörig betrachtet wurden, müssen auf Grund heutiger Kenntnis dem Mohr-Syndrom zugeordnet werden. Die differentialdiagnostisch entscheidenden Gesichtspunkte werden diskutiert und zusammengestellt. Ein von Helbig (1958) und ein von Koberg u. Schettler (1966) beschriebener Patient gehören möglicherweise einem anderen, selbständigen Fehlbildungskomplex an.

Differential diagnosis between Papillon-Léage-Psaume-Syndrome and Mohr-Syndrome

Summary The oro-digito-facial syndrome (Papillon-Léage-Psaume) and the Mohr-Syndrome are genetically seperate entities and can be distinguished clinically. Several patients previously thought to belong to the Papillon-Léage-Psaume Syndrome can now be diagnosed to represent cases of the Mohr-Syndrome. The differentialdiagnosis is discussed. A case described by Helbig (1958) and a case published by Koberg and Schettler (1966) may represent another related type of malformation.

Localization exclusion of the HL-A genes from the short arm of human chromosome 5

Summary A cri du chat patient with deletion of more than 3/4 of the short arm of one chromosome 5 was found to be heterozygous at both of the closely linked HL-A gene loci, thus demonstrating that the HL-A genes cannot be located on the missing segment.

Zusammenfassung Bei einem Patienten mit Cri du chat-Syndrom und einer Chromosomendefizienz, die mehr als 3/4 des kurzen Armes eines Chromosoms 5 umfaßt, wurde Heterozygotie an beiden der enggekoppelten HL-A-Gene festgestellt. Damit können die HL-A-Gene nicht auf dem deletierten Abschmitt lokalisiert sein.

Demonstration of guanosinediphosphate-mannose-and uridinediphosphate-glucose pyrophosphorylase in Acetabularia

Summary The occurrence of guanosinediphosphate-mannose- and uridinediphosphate-glucose pyrophosphorylase is demonstrated in the green alga Acetabularia mediterranea. The possible role of the nucleotide sugars, which were synthesised by these enzymes, in the formation of the cell wall of the alge is discussed.

Fluoreszenzmarkierung von Tumor- und Leber-DNS der Ratte

Zusammenfassung Es wird eine Methode zur Fluoreszenzmarkierung von DNS, die aus Guérin-Rattentumoren und Rattenlebern isoliert wurde, mitgeteilt. Zur Kopplung wurde 1-Dimethylamino-naphthalin-sulfochlorid-5 (DIS) verwendet. Die Ratten intravenös applizierte markierte DNS stellt sich im Gewebsschnitt durch ihre gelbe bzw. intensiv gelbgrüne Fluoreszenz dar.

Summary This paper informs of a method of fluorescence marking of DNA isolated from Guérin rat tumors and rat livers. For coupling we used 1-dimethylaminonaphthalene-sulphochloride-5 (DIS). The marked DNA injected intravenously into rats presented itself by yellow or intense yellowish green fluorescence.

Giemsa banding of a human metacentric chromosome number 9

Summary Giemsa staining procedures have been used on human lymphocyte preparations to locate constitutive heterochromatin (C-bands) and to produce distinct banding patterns for each individual chromosome (G-bands). A metacentric C group chromosome has been shown by both techniques to be chromosome number 9.

Zusammenfassung An menschlichen Lymphocyten wurde eine Giemsafärbung durchgeführt, mit dem Ziel, konstitutives Heterochromatin (C-Banden) zu lokalisieren und für jedes einzelne Chromosom besondere Bandmuster herauszuarbeiten (G-Banden). Beide Techniken erlaubten, ein metazentrisches C-Chromosom als Nr. 9 zu identifizieren.

Zur transspezifischen Variabilität der Lactatdehydrogenase (E.C.: 1.1.1.27) der Säuger

Zusammenfassung Die Lactatdehydrogenasen in Erythrocyten von Säugern lassen eine transspezifische Variabilität erkennen, die bedingt wird durch: 1. Unterschiede in den Syntheseraten der A-und B-Polypeptidketten (unterschiedliche Aktivitäten der A-und B-Cistrons) und 2. Ladungsunterschiede der A-und B-Polypeptidketten (Äderungen in der Primärstruktur, Aminosäurensubstitutionen). Insectivora und Prosimiae besitzen ein gleichartiges Zymogramm, bei dem im wesentlichen nur das Isoenzym LDH 5 ausgeprägt ist, das Homotetramer der A-Polypeptidkette. In ihm erblicken wir ein phylogenetisch älteres Zymogramm einer gemeinsamen Vorfahrensgruppe, das in diesen Linien beibehalten wurde.

Transspecific variability of lactate dehydrogenases (E.C.: 1.1.1.27) in mammals

Summary The LDH isoenzymes in mammalian erythrocytes show a transspecific variability, caused by: 1. Differences in the rates of synthesis of the A-und B-chains (different activities of A-and B-cistrons), 2. Diferences in charge of the A-and B-chains (amino acid substitutions). Insectivora and Prosimiae possess a similar zymogram, in which LDH 5 is predominant, the homotetramer of the A-chain. This is supposed to be phylogenetically more ancient: the zymogram of an ancestral group, which is conserved in those branches.

Zusammenfassung Die Familienuntersuchungen einer galaktosämischen Patientin ergaben bei 5 Verwandten das gleichzeitige Vorkommen eines galaktosämischen Merkmals und eines varianten Enzyms (Duarte-Variante). Summary 5 relatives of a patient with galactosemia showed a simultaneous occurence of the characteristic sign of galactosemia and of a variant enzyme (Duarte variant). Zusammenfassung Die mittlere Dauer der Phasen des Zellzyklus von L-929-Zellen in Kultur wurde autoradiographisch und impulscytophotometrisch bestimmt. Die Mittelwerte von fünf Untersuchungen wichen biei beden Verfahren nur im Rahmen des statistischen Fehlers voneinander ab. Die Reproduzierbarkeit der ICP-Messungen war besser. Auf Konsequenzen für eine klinische Anwendung wird hingewiesen.

Duration of the phases of the cell cycle in L-929 cells

Summary The mean duration of the different phases of the mitotic cycle in L-929 cells cultivatedin vitro was determined autoradiographically and also by impulsecytophotometry. The mean values obtained with the two methods each involving 5 experiments were within the frame of statistical error. The reproducibility of the ICP measurements was better. The importance in clinical application is discussed.
