Summary Geotrichum sp. and Trichoderma sp. isolated from petrochemical effluent treatment plant activated sludge completely degraded 2 g acrylic acid/l as sole carbon source in 4 d. However, after neutralization with Ca(OH)2, concentration as high as 10 g/l was degraded in 6 d. During the degradation acetic acid and -hydroxy propionic acid transiently accumulated.  相似文献

Summary The batch fermentation of whey permeate to lactic acid was improved by supplementing the broth with enzyme-hydrolyzed whey protein. A mathematical model based on laboratory results predicts to a 99% confidence limit the kinetics of this fermentation. Cell growth, acid production and protein and sugar use rates are defined in quantifiable terms related to the state of cell metabolism. The model shows that the constants of the Leudeking-Piret model are not true constants, but must vary with the medium composition, and especially the peptide average molecular weight. A strain of Byssochlamys sp. produced -hydroxypropionic acid when grown on media containing high concentrations of acrylic acid. The maximal production of -hydroxypropionic acid was 4.8% when the initial culture medium contained 7% acrylic acid and 2% glucose, and the initial culture pH was adjusted to 7.0. -Hydroxypropionic acid production from acrylic acid depended greatly on the pH of the culture medium. The interaction of free IAA and its amino acid conjugates on growth and development of cultured tomato hypocotyl tissue (Lycopersicon esculentum Mill. cv. Marglobe) was studied. In a nutrient medium containing 10 mol/L of benzyladenine, free IAA stimulated shoot and root development with little callus proliferation. In contrast, all IAA-amino acid conjugates tested supported mostly callus growth. Simultaneous application of free IAA and its conjugates resulted in the expression of mixed morphogenetic responses (i.e., both vigorous callus growth and organogenesis resulted). Summary The presence of abscisic and phaseic acid in a purified acidic extract from flowering plants of the long-short-day plant Bryophyllum daigremontianum [(R. Hamet and Perr.) Berg.] was conclusively established by combined gas chromatography-mass spectrometry (GC-MS) of their methyl esters. Gibberellin A20 (GA20) was identified by GC-MS of the methyl ester and the trimethylsilyl ether of the methyl ester. The following levels of the 3 compounds per kg fresh weight were estimated: Abscisic acid, 5.5 g; phaseic acid, 9.4g; gibberellin A20, 0.8 g. When GA20 and four other GAs were applied to Bryophyllum under shortday conditions, the order of effectiveness for induction of flower formation was: GA2>GA1>GA5=GA7>GA20. The low biological activity of the native GA20 is discussed.  相似文献

Combining different chromatography systems, unusual nonprotein amino acids were isolated and unequivocally identified from a small amount (less than 100 g fresh weight) ofAmanita gymnopus fruit body. Without obtaining crystals of these amino acids, on the basis of1H-NMR determination, high resolution mass spectrometry, chlorine analysis and oxidation with L-amino acid oxidase, one of them proved to be a new chloroamino acid, (2S)-2-amino-5-chloro-4-hydroxy-5-hexenoic acid (G2). The other three were (2S)-2-amino-5-hexenoic acid (G1), (2S)-2-amino-4,5-hexadienoic acid (G3) and (2S)-2-amino-5-hexynoic acid (G4). Amino acid (G1) was also encountered for the first time in natural products. Amino acid (G3) has been reported from several kinds of fungi belonging toAmanita, subgenusLepidella. Rhodotorula minuta, a basidiomycete fungus, prefers neutral pH for growth and its growth inhibition by food preservatives such as benzoic acid and cinnamic acid has not been reported. Cinnamic acid at 1 g l–1 arrested the growth and decreased the respiration of Rhodotorula but did not kill the yeast. The inhibitory effect was stronger in a mutant strain, 5-286, deficient in the -ketoadipate pathway than in the wild, suggesting that -ketoadipate pathway functions to detoxify this acid by restoring the decreased respiration.  相似文献

Summary The time course for the synthesis of glutamic acid and by-products from glucose was investigated using immobilized cell reactor of the bacterium C.glutamicum. Lactic acid, succinic acid, alanine acid and aspartic acid were formed early in the fermentation and during the active growth phase, whereas gluconic acid, -ketoglutaric acid and proline were produced late and during the active phase of glutamic acid synthesis. Oxygen transfer rate in fermentation broth had a pronounced effect on the nature and quantities of fermentation products. In continuous fermentation and at OTR of 102.5 mMO2/l.h., formation of by-products greatly decreased and up to 58.5 g/l of glutamic acid were produced with a conversion efficiency of 74.6% of the theoretical value and volumetric productivity of 6.2 g/l.h.  相似文献

Summary The present study describes the growth of Pseudomonas putida cells (ATCC 33015) in batch and continuous cultures on two toxic substrates; toluene and m-toluic acid as sole carbon and energy sources. In fed-batch cultures on m-toluic acid up to 3.55 g cell dry weight/1 were achieved with a maximal specific growth rate (max) of 0.1 h-1. The average cellular yield was 1.42 g cell dry weight/g m-toluic acid utilized. When liquid toluene was added to shake-flask cultures in the presence of 0.7 g/1 m-toluic acid, the average cellular yield obtained was 1.3 g cell dry weight/g toluene utilized and the max was 0.13 h-1. Growth on toluene vapour in the presence of 0.7 g/l m-toluic acid in batch cultures resulted in a cellular yield of 1.28 g cell dry weight/g toluene utilized, with growth kinetics almost identical to those with liquid toluene (max liquid=0.13 h-1, max vapour=0.12 h-1). The maximal biomass concentration was 3.8 g cell dry weight/l, obtained in both cases after 100 h of incubation. Pseudomonas putida was grown in a chemostat initially on 0.7 g/l m-toluic acid and vapour toluene and then in the steady state on toluene as the sole source of carbon and energy. Toluene was added continuously to the culture as vapour with the inflowing airstream. Chemostat cultures could be maintained at steady state for several months on toluene. The maximal biomass concentration obtained in the chemostat culture was 3.2 g cell dry weight/l. The maximum specific growth rate was 0.13 h-1, with a cellular yield of 1.05 g cell dry weight/g toluene utilized. Approximately 70% of the toluene consumed was converted into biomass, and the remainder was converted to CO2 and unidentified byproducts.  相似文献

An albino mutant was isolated after treating Monascus araneosus AHU9087 with N-methyl-N-nitro-N-nitrosoguanidine. All other physiological and biochemical characteristics were retained. The mutant did not produce any pigment but produced L-malic acid at 28 g/l, compared with 20 g/l by the parent strain, in media containing 10% (w/v) glucose after incubation under aerobic conditions for 5 days at 37°C.S. Summary Ethanol was produced by a strain ofPichia stipitis adapted to an inhibitory acid wood hydrolysate ofPinus radiata. The best ethanol productivity for batch cultures was 0.21 g/l h at 0.7% ethanol. Varying culture conditions increased ethanol concentration to 0.76%, however the productivity decreased to 0.18 g/l h. A decrease in ethanol concentration in the culture fluid was noted late in the batch which suggested ethanol catabolism. Values of kinetic parameters (K m,K s, max, andV max) were evaluated for this system. The use of calcium alginate immobilized cells in a continuous-flow stirred tank reactor lead to enhanced fermentative performance, namely a maximum productivity of 0.27 g/l h and 1.13% ethanol yield. The immobilized cells in continuous flow reactors represent an attractive option for fermenting sugars released by sulphuric acid hydrolysis ofP. radiata wood.  相似文献

Summary The contents of 2-heptanone and 10-hydroxy-trans-dec-2-enoic acid present in the mandibular glands of worker bees of various ages were determined by gas chromatography.The content of 2-heptanone was dependent on the physiological development rather than the chronological age of the bee. It remained at zero for hive bees and reached a maximum of ca. 40 40 g/bee after the bees began to forage.The content of 10-hydroxy-trans-dec-2-enoic acid found in worker bee heads increased with age and/or in connection with normal changes in the type of social activity of the bee. Only trace amounts of this acid were found in newly emerged bees, but up to 60 g/bee were found after about 3 weeks of age.
Zusammenfassung 2-Heptanon und 10-Hydroxy-trans-2-Decensäure, die in den Mandibeldrüsen verschiedenalter Arbeiterbienen vorkommen, wurden gas-chromatographisch mengenmäßig bestimmt.Das Vorkommen von 2-Heptanon hing von der physiologischen Entwicklung, nicht aber direkt vom Alter der Bienen ab. Stockbienen enthielten kein Heptanon, Flugbienen hatten bis zu ca. 40 g.10-Hydroxy-trans-2-Decensäure nimmt mit dem Alter der Bienen oder auch im Zusammenhang mit dem normalen Wechsel von einer sozialen Tätigkeit zur anderen zu. Bei neugeschlüpften Bienen fanden sich nur Spuren, aber nach 3 Wochen fanden sich bis zu 60 g pro Biene.

A procedure for rapid in vitro multiplication of Tylophora indica (Burm. f.) Merrill., an important indigenous medicinal plant, has been developed. Addition of ascorbic acid was essential to induce sprouting of axillary buds. Optimum multiplication was observed on MS medium containing 6-benzylamino purine (5.0 mg l–1), -naphathalene-acetic acid (0.5 mg l–1) and ascorbic acid (100 mg l–1). Rooting of in vitro produced shoots was readily achieved with indole-3-acetic acid alone (1.0 mg l–1) in MS. The inhibitory effect of propionic acid P and biomass concentration X is studied in batch and continuous fermentations with cell recycle.In batch fermentations, the specific growth rate decreases and cancels out at a critical propionic acid concentration Pc 1; the formerly decreasing specific production rate becomes constant after Pc 1 and cancels out when a second critical propionic acid concentration Pc 2 is reached.In continuous fermentation with cell recycle, a similar inhibition is observed with biomass. The specific rates decrease and become constant at a critical biomass concentration Xc. The growth of Clostridium populeti in 2% (w/v) glucose medium containing 0.2% (w/v) yeast extract was optimal with 10 mM NH4Cl as the nitrogen source. Although the maximum specific growth rate (=0.32 h-1) with 5 mM NH4Cl was similar, the biomass yield was about 30% lower than that at the optimum. Either sodium sulphide or cysteine-HCl at an optimum concentration of 0.33 mM and 5.0 mM respectively, could serve as the sole sulphur source for growth. The growth rate was unaffected by initial glucose concentrations of up to 10% (w/v), but in the presence of 15% glucose it declined by about 35%. The molar yield of butyric acid (mol/mol glucose) declined from 0.70 in 1% (w/v) initial glucose medium to 0.39 in 10% glucose medium. In 5.7% initial glucose medium, butyric acid levels of 6.3 g/l were obtained (0.56 mol butyrate/mol glucose) after 72 h of incubation in 2.5 l batch cultures. A decrease of about 50% in the maximum specific growth rate of C. populeti was observed in the presence of an initial concentration of either 1.2 g/l of butyric acid or 18.9 g/l of acetic acid.This paper is issued as NRCC No. 29032  相似文献

The on-line calculated specific rates of growth, substrate consumption and product formation were used to diagnose microbial activities during a lactic acid fermentation. The specific rates were calculated from on-line measured cell mass, and substrate and product concentrations. Summary Low concentrations of acetic acid decreased the growth rate of and astaxanthin production by Phaffia rhodozyma on glucose, with growth completely inhibited by 2 g acetic acid/l. Using H2SO4 for pH control after sugar depletion caused a decline in the biomass concentration, whereas using acetic acid as titrant resulted in an increase in the biomass with a high astaxanthin content of 1430 g/g cells. An extended culture with a continuous glucose feed failed to maintain a high astaxanthin content.  相似文献

A method for plant regeneration of Iris via somatic embryogenesis is described. Root and leaf pieces from in vitro-grown plants of several genotypes of rhizomatous Iris sp. were cultured in vitro. Callus induction occurred only on root cultures incubated under low light intensity (35 mol m-2 s-1) on two induction media containing 2,4-D (4.5 or 22.5 M), NAA (5.4 M) and kinetin (0.5 M). Somatic embryos developed after transfer of callus onto four regeneration media containing 9 or 22 M BA, or 5 M kinetin and 2 M TIBA or 9 M BA and 4 M TIBA. Plantlets could be obtained from these somatic embryos. Genotypic differences were found both in callus induction and somatic embryo formation, with I. pseudacorus responding better than I. versicolor or I. setosa. Biotransformation of cephalosporin C (CPS-C) to 7-aminocephalosporanic acid (7-ACA) was carried out with coimmobilized permeabilized cells of Trigonopsis variabilis and Pseudomonas species entrapped in Ca-pectate gel beads. Good aeration and stirring during the process was assured. The analysis of this complicated biochemical process in a heterogeneous system was based on the identification of individual effects (internal diffusion, reaction) running simultaneously. A spectrophotometric method was proposed for the determination of 7-(-ketoadipyl amido) cephalosporanic acid (CO-GL-7-ACA) and 7-ACA. The reaction-diffusion model containing dimensionless partial differential equations was solved by using the orthogonal collocation method. A good agreement between experimental values and values predicted by the mathematical model was obtained. Summary These experiments studied the metabolic formation of a,-dodecanedioic acid using the mutant S 76 developed from the wild strain Candida tropicalis 1230 (capable of producing large amounts of a,-dodecanedioic acid).Our results show for the first time that 12-hydroxydodecanoic acid was excreted into the medium as a free acid.n-Dodecanol and n-dodecanoic acid were also detected in the n-dodecane medium. The mutant S 76 was able to produce a,-dodecanedioic acid using either n-dodecanol or dodecanoic acid as the sole carbon source. Quantitative cahnges in the concentrations of 12-hydroxy-dodecanoic acid and other intermediates were recorded during the formation of a,-dodecanedioic acid. S 76 was rapidly able to convert large amounts of 12-hydroxy-dodecanoic acid to a,-dodecanedioic acid.The formation of a,-dodecanedioic acid from n-dodecane via the sequence n-dodecanoln-dodecanoic acid 12-hydroxy-dodecanoic acid was confirmed.  相似文献
