Summary This study describes a protocol for plant regeneration from cultured seedling explants of Arctium lappa. Hypocotyls and cotyledons of A. lappa were induced to form callus by culturing on Murashige and Skoog (MS) medium supplemented with 2.0mg l−1 2,4-dichlorophenoxyacetic acid and 0.5–2.0 mg l−1 benzyladenine (BA). Formation of adventitious buds could be induced from calluses or explants directly by culturing on MS medium containing 1.0–2.0 mg l−1 α-naphthaleneacetic acid (NAA) and 0.5–2.0 mg l−1 BA. These regenerated shoots were rooted on MS medium with 1.0 mg l−1 indole-3-butyric acid or indole-3-acetic acid in combination with 1.0 mgl−1 NAA. The regenerated plants acclimatized in soil were normal morphologically and in growth characters. They flowered and set seeds in the following year after acclimatization.  相似文献

An in vitro highly efficient plant regeneration system was established from hypocotyl segments in buckwheat (Fagopyrum esculentum Moench.). Calli were induced on Murashige–Skoog (MS) medium containing 1.0 mg l–1 to 2.0 mg l–1 2,4-dichlorophenoxyacetic acid and 1.5 mg l–1 6-benzylaminopurine. Shoot buds were formed on subcultured pieces of callus. A high frequency (over 80%) of shoot differentiation was obtained on MS medium supplemented with 2.0 mg l–1 6-benzylaminopurine and 1.0 mg l–1 6-furfurylaminopurine. The regenerated shoots rooted readily on MS medium plus 0.2 mg l–1naphthaleneacetic acid and 0.2 mg l–1 indole butyric acid. The regenerated plantlets were acclimatized and successfully transferred to pots. Chromosome examination showed that the regenerated plants had normal chromosome number (2n=16).  相似文献

Callus cultures were initiated from apical meristems of two commercially cultivated species of cocoyam, Xanthosoma sagittifolium and X. violaceum, on AZ medium supplemented with 2.0 mg 1–1 1-naphthaleneacetic acid (NAA). Organ differentiation was achieved from the friable, white callus by using MS medium containing 2.0 mg 1–1 kinetin (Kn) and 0.2–2.0 mg 1–1 NAA. This two-step system of plant regeneration could be reduced into one-step in X. violaceum by culturing the explants on MS medium supplemented with 5.0 mg 1–1 NAA, 100 ml 1–1 coconut-milk, and 2.0 mg 1–1 Kn, where compact, green callus formation and organogensis occurred within 8 weeks. The regenerated plants were free of dasheen mosaic virus disease. In X. sagittifolium, various phenotypic variation was noted, such as shape, size and total number of leaves and corms. These alterations may be used to improve cocoyam production.  相似文献

Summary Friable calli were induced on leaf segments of Saintpaulia ionantha Wendl. on B5 medium containing 1 mg l–1 2,4-D and 2 g l–1 casein hydrolysate. Cell suspension cultures were readily established from these friable calli and protoplasts could be isolated from the cells with yields of 1–3×107/g f. wt.. By culturing in 0.1 % gellan gum-solidified B5 medium supplemented with 1 mg l–1 2,4-D and 0.1 M each of sucrose and mannitol at a density of 1×105/ml, the protoplasts divided within 6 days and formed macro-colonies after 2 months of culture. Shoot regeneration from protoplast-derived calli was obtained by sequential treatment of the calli with plant growth regulators: initially with 1 mg l–1 each of NAA and BA for 2 months followed by 0.01 mg l–1 NAA and 5 mg l–1 BA for 4 months. Regenerated plants were established after rooting of the shoots on half-strength MS medium, and successfully transferred to the greenhouse. Summary The ability of embryos at different developmental stages to form plants in vitro has been studied in cultivated Cucumis sativus L. and in the wild species C. zeyheri 2 x Sond. and C. metuliferus Naud. On MS medium containing 3.5% sucrose, 0.1 mg 1–1 kinetin (Kn) and 0.01 mg 1–1 indoleacetic acid (IAA), proembryos (0.03–0.05 mm) and early globular embryos (0.05–0.08 mm) from the wild species developed into plants in low frequencies of 8% and 21%, respectively. These embryos should be surrounded by the embryo sac tissue. On the same medium late globular (0.08–0.1 mm) and early heart-stage embryos (0.1–0.3 mm) developed into plants in moderately high and high frequencies of 48% and 83%, respectively. The presence of the embryo sac at these stages was still beneficial, but no longer a prerequisite. Late heart-stage embryos (0.3–0.8 mm) also showed high frequencies of plant formation, 63%, if Kn was applied at a concentration of 1 mg 1–1. From the early cotyledon stage onwards, the frequency of plant formation gradually decreased, reaching a minimum at the late cotyledon stage. Subsequently it began to increase again up to the late maturation stage. The poor plant formation shown by the intermediate-aged embryos could be improved slightly by lowering the sucrose concentration to 0.5% and by increasing the Kn concentration to 10 mg 1–1. Relative to the wild species, embryos of C. sativus showed lower percentages of plant formation. The optimum sucrose concentration was 2% for the heart-stage C. sativus embryos. In all three species the ability to form plants strongly decreased with increasing embryo age, from early to late cotyledon. This is thought to be caused by the increasing tendency of the embryos at these stages to continue in vitro the normal embryo development.  相似文献

Callus cultures were raised from segments of inflorescence axis of Bowies volubilis Harv. (Liliaceae) on a modified basal medium of Murashige and Skoog (1962) (MS) supplemented with 1 mg 1–1 2,4-dichlorophenoxy acetic acid (2,4-D) + 15% v/v coconut milk. Shoot primordia developed after 2–3 subcultures when auxin concentration was lowered. Rooted bulbous plants were obtained in MS medium without any hormone.Shoots were induced directly on scales of regenerated bulbs used as secondary explants on modified MS medium supplemented with 2 mg 1–1 6-benzylaminopurine (BAP) + 0.05 mg 1–1 2,4-D. These shoots grew and multiplied rapidly in shake culture using liquid MS medium. From each scale, 400–600 bulblets could be produced in 16–20 weeks period. Eighty percent of plants have survived on transferring to potted soil.  相似文献

A protocol for multiple shoot induction from cotyledonary node explants of Terminalia chebula Retz. has been developed. Germination frequency of embryos (up to 100 %) was obtained on Murashige and Skoog (MS) medium supplemented with 0.5 mg dm–3 gibberellic acid (GA3). Maximum number of shoots (6.4 shoots per cotyledonary node) was obtained on half-strength MS + 0.3 mg dm–3 GA3+ 1.0 mg dm–3 indole-3-butyric acid (IBA) + 10.0 mg dm–3 benzylaminopurine (BAP) after 4 weeks of culture. When the cotyledonary nodes along with the axillary shoot buds were allowed to grow in the same medium upto 19.2 shoots were obtained after 8 – 9 weeks. Best rooting (100 %, 5.5 roots per shoot) was observed when shoots were excised and transferred to half-strength MS medium containing 1.0 mg dm–3 IBA + 1 % mannitol and 1.5 % sucrose. Survival of rooted plants in vivo was low (35 – 40 %) when they were directly transferred to soil in glasshouse. However, transfer to soil with MS nutrients and 1.0 mg dm–3 IBA in culture room for a minimum duration of 2 weeks increased the survival percentage of plants to 100 %.  相似文献

A rapid propagation method comprising initiation of in vitro shoot tip culture from field-grown flowering plants and reculture of the nodal segments of regenerated shoots in Schenk and Hildebrandt (1972) medium was developed for Woodfordia fruticosa (L.) Kurz., a rare medicinal shrub. A medium supplement of 6-benzylaminopurine (0.2 mg.l–1) induced high frequency (88%) development of axillary shoot buds (3.2) in 4–5 weeks. Subculture of the explants with multiple new shoots in fresh medium for 30 days yielded an even larger number (9.7) of shoots. Highest multiplication (26–35 shoots) was recorded when using culture initiation media with 0.5 mg.l–1 each of BAP and NAA followed by subculture in 0.2 mg.l–1 BAP. The shoot multiplication rate was further accelerated by reculturing 0.4–0.6 cm nodal segments of regenerated shoots in media with 1.0 mg.l–1 BAP. Shoot cuttings (3.5–7.0 cm) were rooted in 0.2 mg.l–1 IAA. In this work, in vitro clonal propagation of Drosophyllum lusitanicum (Dewy pine) was obtained from seedlings germinated in vitro. Seeds were collected in various populations identified in the Algarve region and germinated in vitro on MS medium supplemented with 0.5 mg l–1 BA (6-benzyladenine) and 0.1 mg l–1 GA3 (gibberellic acid). The obtained shoots were used in several multiplication assays. The best results were observed in MS medium supplemented with 0.2 or 0.5 mg l–1 zeatin. The highest rooting frequency (83%) was observed on 1/4MS medium supplemented with 0.2 mg l–1 IBA (indole-3-butyric acid). Fifty percent of the plantlets were successfully acclimatized to ex vitro conditions, exhibiting normal development. Plans are underway to reintroduce the in vitro produced plants from this study in selected locations in their natural habitat.  相似文献

Protoplasts were isolated from immature cotyledons of Glycine max L. Merr. cv. Clark 63 and cultured in liquid or in agarose-gelled modified KP8 medium. Plating efficiencies of 45–50% were obtained in liquid medium and 55–60% in 1.2% (w/v) agarose beads. Upon regular dilution with K8 medium rapidly growing green microcalli (1–2 mm in size) were obtained in 5–6 weeks, which upon transfer to MSB medium with 0.5 mg 1–1 each of 2,4-D, BA, Kn and 500 mg 1–1 CH produced compact green calli in 4–6 weeks. After 3–4 regular subcultures of 14 days each on MSB medium containing 0.5 mg 1–1 each of BA, Kn, ZT, 0.1 mg 1–1 NAA and 500 mg 1–1 CH, about 21% of the compact calli formed multiple shoots. Addition of glutamine, asparagine and GA3 enhanced shoot regeneration up to 30%. Shoots of 0.5–1.0 cm length were transferred to 1/2 MS medium with 0.01 mg 1–1 TH and 0.5 mg 1–1 GA3 for elongation. In 2 to 3 weeks, approximately 60% of the shoots were 2–3 cm in length. These shoots were rooted on 1/2 MS with 1% sucrose and 0.2 mg 1–1 IBA or 0.5 mg 1–1 NAA. Superoxide dismutase (SOD) plays an important role in cellular defense against oxidative stress in aerobic organisms. To generate cucumber (Cucumis sativus L.) fruits producing high yields of SOD for an anti-aging cosmetic material as a plant bioreactor, the CuZnSOD cDNA (mSOD1) from cassava was introduced into cucumber fruits by Agrobacterium-mediated transformation using the ascorbate oxidase promoter with high expression in fruits. The bialaphos-resistant shoots were selected on medium containing MS basal salts, 2 mg l–1 BA, 0.1 mg l–1 IAA, 300 mg l–1 claforan, and 2 mg l–1 bialaphos. After 6 weeks of culture on the selection medium, the shoots were transferred to MS medium containing 1 mg l–1 IAA, 300 mg l–1 claforan, 2 mg l–1 bialaphos to induce roots. Southern blot analysis confirmed that the mSOD1 gene was properly integrated into the nuclear genomes of three cucumber plants tested. The mSOD1 gene was highly expressed in the transgenic cucumber fruits, whereas it was expressed at a low level in the transgenic leaves. The SOD specific activity (units/mg protein) in transgenic fruits was approximately 3 times higher than in those of non-transgenic plants.  相似文献

A protoplast-to-plant regeneration system has been established for sweet potato (Ipomoea batatas (L.) Lam.) and its wild relative, I. lacunosa L. Viable protoplasts, isolated from preplasmolyzed stems and petioles of in vitro-grown plants, were cultured on liquid MS (Murashige & Skoog 1962) medium that supported cell division and colony formation. Embryogenic calli of sweet potato were induced on agar-solidified MS medium supplemented with 3% (w/v) sucrose, 50 mg l-1 casamino acids, 0.2–0.5 mg l-1 2,4-d, 1.0 mg l-1 kinetin and 1.0 mg l-1 ABA. On average, 3 plants were regenerated from a single sweet potato callus subcultured on semi-solid MS medium containing 3% (w/v) sucrose, 800 mg l-1 glutamine, 2.0 mg l-1 BA or 1.0 mg l-1 kinetin and 1.0 mg l-1 GA3. Embryogenic calli of I. lacunosa L. were initiated on semi-solid MS medium containing 0.2–0.5 mg l-1 IAA and 1.0–2.0 mg l-1 BA. Summary A method for obtaining a high frequency of haploid asparagus embryos through anther culture was developed. Flowers collected from plants in the field in July, August and September 1990, for the genotype G203, were stored at 5°C for 24 h. Anthers were placed on Murashige and Skoog medium (MS) containing 500 mg l –1 casein hydrolysate, 800 mg l–1 glutamine, 2 mg l –1 NAA, 1 mg l –1 BA and 5 % sucrose at 32 °C in the dark for three to four weeks to induce calli. Calli were then grown at 25 °C with a 16 h photoperiod for three to four weeks. Developing embryos and calli were transferred to embryo maturation medium, MS containing 6% sucrose, 0.1 mg l –1 NAA, 0.1 mg l –1 kinetin and 0.65 mg l –1 ancymidol, for four weeks. More than 50% of the recovered mature embryos germinated on MS containing l mg l –1 GA3. Anthers with microspores at the late-uninucleate stage had the highest frequency of total and embryogenic calli formation, 40% and 15%, respectively. Each embryogenic callus usually produced 10–15 embryos. Aproximately 75 plants per 100 anthers cultured were recovered: 76% haploid, 22% diploid and 2% triploid. Clonal propagation of Adhatoda beddomei C.B. Clarke (Acanthaceae), a rare medicinal shrub, was achieved through callus-free axillary meristem proliferation from stem node explants of field-grown plants cultured in SH medium. Shoot multiplication was a function of cytokinin activity but sustained growth of the shoots was dependent on the synergistic effect with the auxin, IAA. An optimum number of 5–10 shoots per explant were obtained in 6 weeks using 3.0 mg.l–1 BAP, 0.5 mg.l–1 2-ip and 1.0 mg.l–1 IAA, Upon subculture, vertical halves of the precultured node with the differentiated shoots yielded a larger aggregate number of shoots (23–27) than the uncut precultured node left intact (15–17). Shoot multiplication was rapid and consistent over prolonged periods when the hormonal concentrations were reduced to 1.0 mg.l–1 BAP and 0.2 mg.l–1 IAA during subculture, and reculture of the nodal explants derived from shoot cultures. Rooting of 3–5 cm shoots thus obtained was greatly accelerated in stationary liquid medium containing 0.2 mg.l–1 IBA or IAA. Hardening of the rooted plantlets in the humidity chamber was essential for high frequency (95%) survival. Summary We report on the regeneration of fertile Indica rice (Oryza sativa L.) plants from protoplasts isolated from scutellar tissue of immature embryos. The average yields of protoplasts after purification ranged from 2.8 × 105 to 3.5 × 105 protoplasts per fifty embryos. Protoplasts developed rapidly to colonies when cultured in maltose containing medium using the nurse culture method. Upto 146 or 39 visible colonies per 106 protoplasts were obtained for the varieties Basmati 370 and IR43 respectively. Of two basal culture media compared, R2 medium containing 3 mg l–1 kinetin, 1 mg l–1 naphthalene acetic acid (NAA), 30 g l–1 maltose and 3.0 g l–1 agarose was found to be more effective in producing green plants. Tissue culture propagation system was developed for zedoary (Curcuma zedoaria Roscoe), a valuable medicinal plant, using rhizome sprout cultures. Shoots were induced from rhizomes on basal MS medium containing 20 g l–1 sucrose and 5 g l–1 agar, supplemented with 20 (v/v) coconut water (CW) and benzylaminopurine (BA) concentrations from 0.5 to 5.0 m g l–1. The excised shoots were subcultured on Murashige-Skoog (MS) medium with 20 (v/v) CW and different concentrations of BA and kinetin (Kin), either alone or in combination with indolebutyric acid (IBA) or naphthaleneacetic acid (NAA). MS medium with 20 (v/v) CW, 3 mg l–1 BA, and 0.5 mg l–1 IBA resulted in a multiplication rate per shoot; 5.6 shoots per explant were obtained on average after 30 days of culture. Well-developed shoots (30–40 mm in length) were rooted on MS medium containing 20 g l–1 sucrose and 8 g l–1 agar, supplemented with 20 (v/v) CW and 2 mg l–1 NAA. Summary Mimosa tenuiflora (Willd.) Poiret (Leguminosae) was micropropagated throughin vitro culture of axillary buds on Murashige and Skoog (MS) medium. Shoot formation was achieved when the media were supplemented with 0.1 mg.L–1 IAA + 3 mg.L–1 KN.In vitro rooting of regenerated shoots was achieved when 0.1 mg.L–1 KN was combined with 1 mg.L–1 IBA in the absence of IAA. Ninety-four percent of the rooted plants were succesfully adapted to field conditions and grown in the soil. An efficient clonal multiplication system was developed for in vitro propagation of crocin — producing Gardenia jasminoides Ellis plants. Murashige and Skoog's (MS) medium containing 6-benzylaminopurine (BAP 1 mg l–1) and indole-3-butyric acid (IBA 1 mg l–1) resulted in multiple shoot initiation at the rate of 21 shoots per explant in 60 d of culture. Transfer of the microshoots into liquid MS medium supplemented with BAP (5 mg l–1) with two subcultures of 15 d duration in the same medium resulted in 400 ± 25 shoots per explant. Efficient rooting was achieved in MS medium supplemented with -naphthaleneacetic acid (5 mg l–1). The in vitro raised plants were hardened in a greenhouse and transplanted to the field successfully. Nodal explants, excised from young shoots from mature trees of Syzygium cuminii, were cultured on MS medium supplemented with different concentrations of BA or kinetin. Among these, BA (0.5 or 1.0 mg l–1) induced greening and opening of the incipient shoot buds, which however did not elongate. Elongation of the shoot buds was facilitated on MS medium with 1.0 mg l–1 BA supplemented with casein hydrolysate (1.5 g l–1) or glutamine (200 mg l–1). Nodal explants (microcuttings), taken from shoots developed in vitro, also developed multiple shoots when cultured on MS with1 mg l–1 BA. These explants did not require an additional supply of reduced nitrogen, for further normal development. Shoots developed from explants from mature trees and microcuttings were rooted by sub-culturing them on Knop's medium supplemented with 2% sucrose and 1 mg l–1 IAA. The plants that developed in vitro were planted in soil and were transferred to the field after an acclimatization period of 7–8 months. These plants have been thriving well for more than three years and have no apparent phenotypic aberrations.  相似文献

Pollen of Iochroma warscewiczii Regel (Solanaceae) produced embryogenic calli or embryos inside anthers cultured on Nitsch & Nitsch medium. Two distinct pathways could be recognized in this process, one involving mainly the vegetative cell, and the second starting with two equal cells in the pollen grains.In all media tested, androgenesis initiation was highest when anthers contained pollen at the first mitosis, or close to it, at inoculation. High sucrose (7%) and calcium (11.3 mM) concentrations were found to be highly desirable for the induction of androgenesis in this species. Addition of benzylaminopurine (0.5 mg l–1) to the culture medium seems to slightly improve callus or embryo production. When all three factors were present at optimal concentrations as much as 13.9% of inoculated anthers were found to be embryogenic.Plantlet development from pollen embryos required lower sucrose (3%) and a combination of 0.1 mg l–1 benzylaminopurine and 0.5 mg l–1 gibberellic acid in the culture medium. Cytological analysis of 55 regenerated plantlets showed that about 49% were haploids, but diploid (ca. 49%) and triploid (ca. 2%) plants were also obtained.  相似文献
