It was found that stomata on illuminated leaves, both of Pelargoniumand wheat, opened much wider where the leaf surface was enclosedin a small volume of air, as in a normal porometer cup, thanelsewhere. This was shown for both species by the infiltrationmethod, and for Pelargonium by Lloyd's method and direct microscopicalobservation also. The effect was shown not to be due to pressure of the porometercup or glass plate on the leaf, or to temperature differences,nor directly to the lack of movement or high humidity of theenclosed air. A considerable body of data was collected which appeared tosupport the hypothesis that the wide opening was due to accumulationof some volatile substance produced by the leaf, but all theresults were also consistent with the view that it was causedby reduction in the carbon dioxide content of the enclosed airbelow the normal 0·03 per cent. owing to photosynthesis.Further crucial experiments with both the porometer and infiltrationmethods left virtually no doubt that the latter hypothesis wascorrect. This extreme sensitivity of stomata to carbon dioxide concentrationwithin the range 0·03 per cent. to zero is discussedin relation to their operation in nature, and a possible biologicaladvantage is suggested. The bearing of the effect upon porometer investigations is alsodiscussed and it is concluded that for all quantitative or semi-quantitativeexperimentation it is essential to use a cup detached betweenreadings, or at least swept with air such as surrounds the restof the leaf, and to have the upper leaf surface above the cuparea freely exposed or similarly swept. For qualitative investigationof the light response of stomata the traditional form of cupmay be used. The importance is stressed of allowing porometer readings toreach equilibrium under one set of conditions before changingto another, when investigating the ‘closing’ or‘opening’ effects of external factors. Several subsidiary effects, observed in the course of the investigation,are discussed; in particular an effect of humidity upon therate of response to other factors.  相似文献

Putrescine, spermidine, and spermine content were analysed inzygotic embryos of barley (Hordeum vulgare L.). Changes in polyaminecontent were observed during zygotic embryo growth. In two cultivars,‘Bomi’ and ‘Golden Promise’, the totalpolyamine content in the embryos was 2.6–2.9 nmol mg–1fresh weight 10 d after anthesis, the highest content observed.It dropped to 1.3 nmol mg–1 fresh weight 14 d after anthesis.This drop was caused by decreases in all three polyamine concentrations.From 14 to 35 d after anthesis the putrescine content continuedto decrease while the spermidine and spermine content increased,thus the total polyamine content remained constant until 35d after anthesis. The mutant ‘Ris? 1508’ showeda constant polyamine content around 1.3 nmol mg–1 freshweight from 14 to 35 d after anthesis. The polyamine patternwas conserved in all three lines throughout the period of investigationshowing a spermidine content higher than putrescine contentwhich was, in turn, higher or equal to the spermine content.The polyamine content measured as nmol µg–1 proteindecreased from 14 to 21 d post anthesis in all three lines,because the protein content (µg mg–1 fresh weight)increased during the period. In dedifferentiating zygotic embryoscultured in vitro the putrescine content (nmol mg–1 freshweight) rose by a factor of nine and the spermidine contentdoubled within the first week of cultivation, whereas sperminecontent did not change. Recalling Taste Intensities in Sweetened and Salted Liquids   总被引:2,自引:1,他引:1  
The effect of training on recalling taste intensities over 6weeks was studied using an ad libitum mixing procedure. Subjectstasted sweet and salty standards labeled as ‘weak’and ‘strong’ (3 and 8% sucrose in redcurrant juice;0.4 and 1.2% NaCl in beef broth). They subsequently mixed unsweetenedand sweetened juice, and unsalted and salted broth, to producetaste intensities that corresponded to the standards. A minimumtraining (MT) group (n = 13) produced comparison stimuli bytasting and directly comparing with standards in one sessiononly; an extensive training (ET) group (n = 13) did this insix sessions before producing comparison stimuli based on memoryonly at 1 h, 1 day, 1 week and 6 weeks. An upward bias (chemicallydetermined concentrations of comparison stimuli exceeding thoseof standards) occurred at 1 day or 1 week in MT subjects for‘weak’ and ‘strong’ sweetness, and for‘strong’ saltiness, and sustained thereafter. Theupward tendency was also observed in ET subjects but was significantonly for ‘strong’ sweetness. The success of Triticum aestivumxZea mays crosses, used to producewheat doubled haploids, is influenced by light intensity. Toexamine the basis for this response, pollen tube growth, embryosurvival and indicators of photosynthetic rate were measuredin two wheat cultivars (‘Karamu’ and ‘Kotuku’)crossed with maize at two irradiance levels (250 or 750 µmolm-2s-1, PAR). Pollen tube growth was significantly affectedby light intensity in ‘Karamu’ plants but not in‘Kotuku’ plants, despite both cultivars being pollinatedby the same maize source. The percentage of pollen tubes reachingthe cavity between the ovarian wall and integuments, or in themicropyle of ‘Karamu’ plants at high light intensity(65%) was nearly three-times greater than that at low lightintensity (22%). Thus, either low light intensity can affectthe maternal wheat plant in a way that inhibits pollen tubegrowth and/or high light intensity may promote pollen tube growthin ‘Karamu’ plants. Significant differences in ratesof electron transport in plants grown at the two light intensitiesindicated that the rate of photosynthesis may also have an effecton pollen tube growth. These results have importance for improvingthe efficiency of wheat x maize crosses and other wide cerealcrosses. Growth analysis of plants raised under controlled environments(10–5, 12, 15, 18 and 20 °C, and 21 h photoperiod)was used to examine whether varietal differences in the minimumgermination temperature of four bean cultivars persist duringgrowth at suboptimal temperatures. A method to estimate theminimum vegetative growth temperature, based on axis relativegrowth rate, was developed. In order to compensate for ontogeneticdrift, the harvests were conducted at the same stage of developmentof the plants. Axis relative growth rates, reduction rates ofthe cotyledons and other growth parameters were calculated inorder to compare the cultivars. Cultivar ‘Marschall’showed better growth potential at 12 °C than the others,‘Pergousa‘ at 15 °C, and ‘Marschall’,‘Olsok’ and ‘Pergousa’ at 18 and 20°C. The effect of temperature on axis RGR was similar for‘Marschall’, ‘Olsok’ and ‘Pergousa’(Q10 = 2·1) and more pronounced than for ‘Processor’(Q10 = 1·3). Although there were significant differencesin the growth parameters among the cultivars within each temperatureused, the differences did not correspond with the differencesduring germination at low temperatures. The minimum vegetativegrowth temperature was close to 10 °C for all the cultivarstested. Spore discharge in Ascobolus crenulatus occurs both in the lightand in the dark. In a 12 h light: 12 h dark daily regime discharge-ratehas peaks in the dark periods, due apparently to light stimulationwith about half a day's interval between stimulus and response. Using a ‘spore clock’ the course of discharge hasbeen followed for a single apothecium on changing from darknessto light. Exposure to light (500 lux) of wave-lengths around400, 440 and 460 mµ immediately causes ’puffing‘,whilst light of longer wave-length (504 and 580 mµ) hasno effect. Change from darkness to white light has no immediateeffect, but there is a delayed stimulation with a marked increasein discharge-rate 10–14 h later. Simultaneous illuminationof an apothecium, which has been in darkness, by blue light(420 mµ, or 440 mµ, 500lux) and yellow light (580mµ, 500 lux) does not result in puffing. A series of experiments was conducted to assess net CO2assimilationand growth responses to waterlogging of grafted and seedlingtrees in the genus Annona. Seedlings of A. glabra, A. muricataandA. squamosa L., and scions of ‘Gefner’ atemoya(A. squamosaxA. cherimola Mill.), ‘49-11’ (‘Gefner’atemoyaxA. reticulata L.), ‘4-5’ (‘Priestley’atemoyaxA. reticulata), A. reticulata grafted onto either A.glabra, A. reticulata orA. squamosa rootstocks were floodedfor up to 60 d. Soil anaerobiosis occurred on the third dayof flooding. Seedlings ofA. glabra and A. muricata, and thescions ‘49-11’, ‘Gefner’ atemoya, andA. reticulata grafted onto A. glabra rootstock were consideredflood tolerant based on their ability to survive and grow inflooded conditions. Scions of the normally flood-sensitive A.reticulata, ‘Gefner’ atemoya, and ‘49-11’tolerated root waterlogging when grafted onto the flood-tolerantspecies, A. glabra. In contrast, flooding of A. squamosa seedlingsand rootstocks, and A. reticulata rootstocks greatly reducedgrowth and net CO2assimilation rates, and resulted in 20–80%tree mortality. Stem anatomical responses to long-term flooding(12 continuous months) were assessed in seedlings of A. glabraand A. muricata, and trees of ‘49-11’ grafted ontoA. glabra. Flooded trees developed hypertrophied stem lenticels,particularly in A. glabra, and enlarged xylem cells resultingin thicker stems with reduced xylem density. Flooding did notincrease air spaces in pre-existing xylem near the pith or inxylem tissue that was formed during flooding. Thus, flood tolerancedid not involve aerenchyma formation in the stem. Four experiments were conducted to determine the effect of tradewinds in Guam, USA, on growth and gas exchange of three papaya(Carica papaya L.) cultivars. ‘Known You 1’, ‘Sunrise’,and ‘Tainung 2’ papaya seedlings at two differentstages of development were exposed to 0, 36 or 100% ambientwind. Wind exposure reduced stem height and leaf or stem dryweight in most cases, but had little influence on root growth.Net CO2assimilation (ACO2) at midday was lower for seedlingsexposed to wind than for those protected from wind. Dark respirationof exposed seedlings increased as much as 120% above that ofthe protected seedlings during the night. Wind exposure decreasedwhole plant evapotranspiration by up to 36% during the photoperiod,but increased evapotranspiration by 58–87% during thenocturnal period. Responses to wind exposure were similar amongcultivars, except that growth of ‘Tainung 2’ seedlingswas less affected by wind than that of the other cultivars.Seedlings that were exposed to the various wind treatments fromgermination onwards were less influenced by wind exposure thanwere seedlings that were grown in a protected nursery beforebeing exposed to the various wind treatments. These data indicatethat: (1) ambient trade winds in Guam are strong enough to decreasethe growth of papaya seedlings; (2) plant age influences theresponse; (3) stem and leaf growth are more influenced thanroot growth; and (4) decreasedACO2 and increased dark respirationmay be partly responsible for growth reduction. Simulated swards of each of two selection lines of Lolium perennecv. S23 with ‘fast’ and ‘slow’ ratesof ‘mature tissue’ respiration were establishedin growth rooms at 20/15 °C day/night temperatures and studiedover four successive regrowth periods of 46, 30, 26 and 53 daysduration. The ‘slow’ line outyielded the ‘fast’,both in harvestable shoot (above a 5 cm cut) and in root andstubble. Its advantage increased over successive regrowth periodsto 23 per cent (total biomass). Gas analysis measurements onthe entire communities (including roots), during the final regrowthperiod, showed that the ‘slow’ line had a 22–34per cent lower rate of dark respiration per unit dry weight.This enabled it to maintain its greater mass of tissue for thesame cost in terms of CO2 efflux per unit ground area. Halfthe extra dry weight produced by the ‘slow’ line,relative to the ‘fast’, could be attributed to itsmore economic use of carbon. The rest could be traced to a 25per cent greater tiller number which enabled the ‘slow’line to expand leaf area faster (though not at a greater rateper tiller), intercept more light and fix more carbon, earlyin the regrowth period. Dry legumes of Sesbania punicea (Cav.) Benth. contain impermeableseeds which are either beige-greenish or reddish. When keptin water for over one year they do not swell. Quick imbibitionwas induced by a deep cut, abrasion with coarse abrasive paper,immersion in concentrated sulphuric acid, in boiling water andin boiling KOH solutions. Neither lipid solvents (acetone, chloroform,cyclohexane, diethyl ether, ethanol, xylene), nor, at room temperature,KOH solutions up to 50% per cent, permeabilized the seeds. Astrong barrier to water entry appeared to be located in thepalisade layer which showed ‘light points’ in places,and which seemed to be transversal thickenings. In the testa,‘ callose’, which may play a role in the impermeabilityof the Sesbania punicea seed, was detected. Plants of Plantago lanceolata L. and Zea mays L., cv. ‘Campo’were grown at two levels of light intensity. Especially in theroots, the rate of dry matter accumulation decreased at lowlight intensity. The carbohydrate content of both roots andshoots of P. lanceolata was not affected by light intensity.The relative contribution of SHAM1-sensitive respiration, thealternative chain, to total root respiration of both P. lanceolataand Z. mays, was not affected by light intensity during thedaytime. The alternative pathway was somewhat decreased at theend of the dark period, but not in the root tips (0–5mm) where it still contributed 56% in respiration. It was, therefore,concluded that photosynthesis is not a major factor in regulationof root growth in the species investigated. To see whether the effect of light intensity on root growthrate was via transpiration, plants of Z. mays were grown atdifferent air humidities. Both high humidity and low light intensityaffected the root morphology in such a way that the distancebetween the apex and the first laterals on the primary rootaxis increased. It is suggested that this effect on root morphologyis due to transpiration and the subsequent removal of root-producedinhibitors of lateral root growth; although light intensityalso affected the rate of dry matter accumulation of roots andthe rate was not affected by the humidity of the air. It is,therefore, concluded that the effect of light intensity on therate of dry matter accumulation of roots of Z. mays is not viaan effect on transpiration.  相似文献

Plants of two cultivars of Callistephus chinensis (Queen ofthe Market and Johannistag) were grown in 8 h of daylight perday with one of the following treatments given during the 16h dark period: (a) darkness—‘uninterrupted night’,(b) I h of light in the middle of the dark period—a ‘nightbreak’, (c) I min of light in every hour of the dark period—‘cycliclighting’, (d) light throughout—‘continuouslight’. The plants receiving uninterrupted dark periods remained compactand rosetted in habit with small leaves, while leaf expansion,stem extension, and flower initiation were promoted in all threeillumination treatments (b, c, d). Although these three treatmentsproduced similar increases in leaf area, continuous light wasthe most effective for the promotion of both stem growth andflower initiation while cyclic lighting was generally more effectivethan a I-h night break. Continuous light also caused more dry matter to be divertedto stems at any given vegetative dry weight and it was shownthat the stem weight ratio of both varieties was correlatedwith stem length.  相似文献

Extracts of small and mature-size lupin pods yielded four substancesaffecting the growth of wheat-coleoptile sections: one acidpromotor (A), two acid inhibitors(B and X), and one neutralinhibitor(Y). Inhibitor B was extremely active, however, coleoptile sectionsshowed no signs of toxic effects; they resumed growth at a rapidrate after rinsing them and adding ß-indolylaceticand (IAA) to the medium. 1 µg of IAA was required to counteractthe effect of ‘B’ extracted from 230 mg. Of tissue.On an equal fresh weight basis the inhibiting action of ‘B’in lupin pods was 500–1,500 times more potent than thatof ‘inhibitor ß’ in etiolated pea seedlings. Small pods of plants infected with pea-mosaic virus yielded3 times the amount of ‘A’ of healthy plants (equivalentto 1 µg. IAA 0.3 µg. IAA per 25 g. of tissue respectively),and approximately the amount of ‘B’. Mature podsof virus-infected plants again yielded more‘A’,but also 2? times more ‘B’ than pods of healthyplants. Healthy pods yielded more ‘A’ than virus-infectedpods, and there was no difference in ‘X’. A lupin abscission test was developed and the effects of proximaland distal application of -naphthyl acetic acid (NAA) are presented,and discussed with respect to results of other abscission tests. ‘A’ accelerated abscission when applied proximally,and delayed or prevented it when applied distally. ‘B’strongly accelerated abscission when applied in either way.A possible mechanism explaining the abscission-inducing effectof developing pods on later flowers is discussed in terms ofthe substances ‘A’ and ‘B’. Our goal was to construct a simple muskmelon phenology modelthat could be run with easily obtainable weather station dataand used by growers to quantify phenological development andaid in projecting harvest dates. A growth chamber experimentwas conducted with two cultivars of muskmelon (‘Gold Rush’and ‘Mission’) to determine how main vine leaf appearancerates responded to temperature. We identified three cardinaltemperatures for leaf appearance rate: the base temperature(10 °C) at which leaf appearance rate was zero; an optimumtemperature (34 °C) at which the rate of leaf appearancewas maximal; and an upper threshold temperature (45 °C)at which leaf appearance rate returned to zero. Using thesethree cardinal temperatures, we constructed a simplified thermalunit accumulator for hourly measurements of air temperature.Main vine plastochron interval (PI), thermal time to harvest,and final yield were determined for three cultivars of muskmelon(‘Explorer’, ‘Gold Rush’ and ‘Mission’)grown in the field at Overton, TX, USA, over six transplantingdates from March to June 1998. The flowering requirements of six European varieties of Loliumperenne L. were studied in controlled environments. In experimentson primary induction, flowering was recorded after transferto long days (LD) in a greenhouse at 12–24°C. In experimentson secondary induction, primary induction was first accomplishedat 6°C/10 h daylength for 12 weeks. When evaluated by the50% heading criterion, the requirement for duration of primaryinduction at 6°C/8 h daylength was <3 weeks in Mediterranean,5–6 weeks in Central European and 7–8 weeks in Scandinavianvarieties. While ‘Veyo’ (Italy) flowered profuselyregardless of temperature or daylength during primary induction,critical temperatures for primary induction in SD and LD were15 and 11°C in ‘Baca’ (Czech Republic) and 11and 7°C in ‘Falster’ (Denmark). The criticalphotoperiod for secondary induction at 15°C ranged from12 h in ‘Veyo’ and 14 h in ‘Baca’ to16.5 h in ‘Falster’ and 17.5 in ‘Kleppe’(Norway). The critical number of LD cycles varied correspondingly.While the Central and North European varieties required fewerLD cycles for 50% heading at 18 than at 12°C, ‘Veyo’showed the opposite response. It is concluded that the requirementsfor both primary and secondary induction of Lolium perenne increasewith increasing latitude of origin of the germplasm. In oneexperiment, 39–87% of the inflorescences came from tillersthat were not visible on transfer from primary to secondaryinduction, thus it is also concluded that there is no juvenilestage in tillers of Lolium perenne. Diurnal laminar reorientation was followed in solar-trackingleaves of Lavatera cretica L. under simulated conditions. Asimulated ‘sun’ was moved over the lamina in a 180?arc in the vertical plane of the mid-vein, at an angular velocityof 15? h–1 in a regime of 12-h photoperiods. In one groupof plants the petioles of the experimental leaves were arrangedto face ‘sunrise’, while in the other they werearranged to face ‘sunset’. At ‘sunrise’,the laminae in both groups, which were inclined towards theanticipated direction of ‘sunrise’, changed theirelevation towards the rising ‘sun’, resulting inprogressive reduction in the angle of incidence (AI) of lighton the laminar surface (AI= differential between laminar and‘solar’ elevation). As a result, laminar and ‘solar’elevation converged, and laminar reorientation gradually ceased,until the ‘solar’ elevation had passed the normalto the laminar surface (AI=0?). laminar reorientation was thenresumed, but its direction was reversed to follow the directionof ‘solar’ reorientation. During most of the remaining‘day’, laminar elevation (LE) trailed that of the‘sun’ by an average of 11?-14?. Laminar reorientationthen anticipated ‘sunset’ by starting to slow down60 to 90min in advance. During the 12-h dark period, the laminareoriented towards the anticipated direction of the subsequent‘sunrise’. The time-course of nocturnal reorientationwas qualitatively different in the two groups of experimentalplants. The time-course of diurnal phototropism under naturaland simulated conditions is analysed and compared and differencesand similarities between them are discussed. Exposure to light brings about a fall in the plasmolyticallydetermined osmotic pressure of the cells of the plumular hookof Vicia to about half the ‘dark’ value. Analysesand conductivity measurements show that light causes a smallrise in osmotically active solutes, which is confirmed by cryoscopicmeasurements. The contradiction lies in an abnormally high ‘dark’plasmolytic value, which is not attributable to entry of theplasmolysing solute; it is tentatively suggested that this isan imbibition phenomenon, and that the effect of light is todecrease the affinity for water by promoting a rise in molecularweight of the cell proteins.  相似文献
