采用SSR分子标记分析延胡索的遗传多样性,筛选出多态性高、稳定性好的12对引物,对19个居群360份延胡索样品进行了群体遗传分析。结果表明:(1)12对SSR引物共扩增出多态性位点227个,检测到4~9个等位基因,平均等位基因数目为5.25个,表现出丰富的多态性;延胡索居群具有较高的遗传多样性(Na=5.25,I=1.192 6,H=0.387 9),物种间遗传分化程度高(Fst=0.388 3),基因流较弱(Nm=0.393 8)。(2)UPGMA聚类分析和贝叶斯距离分析结果表明,19个居群明显聚为4大支;Mantle Test结果(r=0.326,P=0.01)表明,地理位置相近的种群亲缘关系更近。研究认为,延胡索的遗传结构是该物种自交为主的繁育系统、克隆生长、地理隔离以及居群间有限的基因流共同作用的结果,故对该物种的保护应以就地保护为主。  相似文献

为构建六角果鸢尾(Iris hexagona)的离体快繁体系,以其幼嫩根状茎为外植体,研究了培养基和植物生长调节剂对不定芽诱导、增殖和植株生根的影响。结果表明,根状茎用0.1%Hg Cl2消毒13 min的效果较佳;不定芽诱导最适培养基为MS+6-BA 1.5 mg L–1+NAA 0.5 mg L–1+蔗糖30 g L–1+琼脂7.5 g L–1,不定芽增殖的适宜培养基为MS+6-BA 0.5 mg L–1+NAA0.2 mg L–1+KT 0.3 mg L–1+蔗糖30 g L–1+琼脂7.5 g L–1;在MS+IBA 1.5 mg L–1+蔗糖30 g L–1+琼脂7.5 g L–1培养基上不定芽生根率可达100%;腐殖土和珍珠岩+泥炭土+蛭石(1∶2∶1)均可作为组培苗移栽的适宜基质,移栽成活率可达100%。  相似文献

Summary N6-Benzyladenine (BA; 0.04–4μM) application to germinated Quercus suber somatic embryos considerably increased caulinar apex elongation frequency and maintained active growth in the plantlets, although it did not have a significant effect on the percentage of shoots with normal morphology. The addition of 0.5 μM indoleacetic acid together with the cytokinin did not have any effect. The use of a low concentration (0.04 μM) of BA allowed the appropriate radicle elongation in all germinating somatic embryos, but higher concentrations arrested this elongation.  相似文献

Summary A protocol for in vitro propagation of Rollinia mucosa, an important medicinal plant, was developed. The presence of 500 mg l−1 polyvinylpyrrolidone (PVP) during explant excision was important to avoid browning. Axillary buds, adventitious buds, and shoot cluster proliferation were achieved from epicotyl and hypocotyl explants from nursery-grown seedlings. The highest direct organogenesis percentage from hypocotyl explants was obtained upon culture of explants on Murashige and Skoog medium supplemented with 2.2 μM benzyladenine (BA) plus 2.32 μM kinetin. Epicotyl explants display highest regeneration frequency on a medium containing 8.8 μM BA and 0.54 μM naphthaleneacetic acid. Gibberellic acid was necessary for shoot elongation. Root induction was observed when shoots were pretreated with activated charcoal for 7 d in the dark before culture on Woody Plant Medium supplemented with 49.21 μM indolebutyric acid for 10 d. Root development was observed when 20 g l−1 sucrose was used. Rooted plantlets were acclimatized and grown in the greenhouse.  相似文献

岩黄连是一种多年生草本植物,主要分布于我国西南部岩溶地区,具有重要的生态及医药价值,开发利用前景广阔。由于生存环境脆弱和人为采挖的影响使得野生岩黄连资源面临枯竭,因此被列入2021年国家重点保护野生植物名录。为探索该物种濒危的原因,该文研究了种子保存方法、化学试剂前处理、温度、光照、干旱、pH以及混沙保湿冷藏处理对种子萌发的影响。结果表明:(1)低温保存能够延长岩黄连种子的活性,保存2 a的种子萌发率仍能达到30%。(2)化学试剂HCl及NaClO对种子的前处理使其发芽率提升50%~60%;种子在20℃时萌发率能达到50%,而30℃时基本不能萌发;黑暗条件下的萌发率显著高于周期性光照条件;萌发率随着干旱程度加深不断下降;萌发率在pH值3.0~8.0的条件下无显著变化。(3)混沙保湿冷藏使种子发芽势和萌发率显著提高,萌发率达到对照的2倍。综上认为,岩黄连种子在室温条件下易失活且不能在30℃以上高温萌发的特性与其濒危有较大关系;高效的种子萌发方法可以为岩黄连保护与产业化应用提供有效途径。该研究结果为野生岩黄连的保育提供了理论和技术保障,为其大田栽培和产业化推广提供了技术支持。  相似文献

Summary The effect of abscisic acid (ABA) was evaluated during the maturation and germination of holm oak (Quercus ilex L.) somatic embryos. The addition of ABA to the culture medium significantly reduced unwanted recurrent embryogenesis in mature somatic embryos without affecting the germination of embryos subjected to stratification at 4°C. Stratification at 4°C for 2 mo. was the most efficient for stimulating somatic embryo germination of holm oak. The addition of 90 and 450 mM sucrose also improved germination, while higher sucrose concentrations were inhibitory.  相似文献

A procedure for rapid in vitro multiplication of Tylophora indica (Burm. f.) Merrill., an important indigenous medicinal plant, has been developed. Addition of ascorbic acid was essential to induce sprouting of axillary buds. Optimum multiplication was observed on MS medium containing 6-benzylamino purine (5.0 mg l–1), -naphathalene-acetic acid (0.5 mg l–1) and ascorbic acid (100 mg l–1). Rooting of in vitro produced shoots was readily achieved with indole-3-acetic acid alone (1.0 mg l–1) in MS. Summary The effects of abscisic acid (ABA) (0, 0.09 μM, 0.19 μM, 0.28 μM, and 0.38 μM) or ancymidol (0, 0.98 μM, 1.95 μM, 2.93 μM, 3.90 μM) in embryo germination medium on the conversion of primary embryos to plantlets and secondary embryogenesis were evaluated for asparagus. ABA and ancymidol each significantly enhanced both responses. ABA was more effective than ancymidol in promoting the conversion of primary embryos to plantlets, while the converse was true for the production of secondary embryos. The most effective treatments for embryo conversion were 0.19 and 0.28 μM ABA; 75–77% bipolar and 55–57% globular embryos converted to plantlets. For secondary embryogenesis, the most effective treatments were 1.95 and 2.93 μM ancymidol; 99–101 and 84–86 somatic embryos were produced from 10 globular and 10 bipolar embryos, respectively. Bipolar embryos generally converted to plantlets better than globular embryos, but more secondary embryos were produced from globular embryos than from bipolar embryos in all treatments. ABA and ancymidol also affected the morphology of the plantlets produced. The plantlets from the embryos incubated on the medium with ancymidol had strong and thick shoots and roots, while those on the medium with ABA had long, thin shoots and short thin roots.  相似文献

作为蚁播植物种子的重要传播者,蚂蚁不但取食种子上附着的油质体,也喜食其它富含蛋白质、脂类、糖和维生素等的食物,因此环境中其它可利用食物的存在可能会影响蚂蚁对种子的搬运进而影响种子散布,但目前对于这种影响是如何发生的仍不清楚。在野外研究了蚂蚁对小花宽瓣黄堇(Corydalis giraldii Fedde)种子、肉、蜂蜜、苹果、馒头等食物的趋性和偏好程度,以及添加食物后蚂蚁对种子的拜访频率和搬运效率,以揭示其它可利用食物如何影响蚂蚁觅食和取食偏好,进而影响小花宽瓣黄堇种子散布。结果显示,在所诱捕的8种蚂蚁中,玉米毛蚁(Lasius alienus(Foerster))和丝光蚁(Formica fusca Linnaeus)是小花宽瓣黄堇种子的主要搬运者,不同食物诱捕的玉米毛蚁数量无显著性差异(P0.05),但蜂蜜和苹果诱捕的丝光蚁数量均显著大于种子(P0.05)。玉米毛蚁和丝光蚁均为杂食性,在觅食中分别行使群体募集和简单协作性募集。在仅有种子的对照处理中,玉米毛蚁和丝光蚁对种子的拜访频率分别为(38.73±4.57)头和(30.8±2.87)头(40min,n=15),两种蚂蚁对种子的拜访频率差异不显著(P0.05);玉米毛蚁和丝光蚁搬运种子的效率分别为(33.87±4.22)粒和(16.27±3.35)粒(40min,n=15),玉米毛蚁的搬运效率显著高于丝光蚁(P0.05)。与对照相比,添加馒头、苹果和蜂蜜后丝光蚁对种子的拜访频率显著降低(P0.05),分别为(15.6±3.61)头、(9.07±1.4)头和(7.67±1.58)头(40min,n=15);添加苹果和蜂蜜后丝光蚁对种子的搬运效率显著降低(P0.05),分别为(3.47±1.17)粒和(2.87±0.9)粒(40min,n=15);添加不同食物后玉米毛蚁对种子的拜访频率和搬运效率均无显著变化(P0.05)。研究结果表明行使群体募集的玉米毛蚁比行使简单协作募集的丝光蚁有更高的种子搬运效率,添加食物后影响丝光蚁对种子的拜访频率和搬运效率,这说明其它可利用食物对小花宽瓣黄堇种子散布的影响与搬运蚂蚁的种类及其觅食的募集方式有关。研究结果可为进一步研究蚂蚁与植物(种子)间的互利共生关系及其影响因素提供资料。  相似文献

Somatic embryogenesis was induced from embryogenie cells derived from cotyledon expiants cultured on MS medium supplemented with 1 mg/L 2,4-D. In order to clarify the effect of abscisic acid (ABA) on the morphology of somatic embryos, embryogénie cell clumps or developing somatic embryos were treated continuously, or briefly, with ABA during culture. When embryogenie cells in MS medium without 2,4-D were treated with 0.04 mg/L ABA for the first week, normal embryos with two cotyledons increased slightly and embryos with anomalous cotyledons decreased. However when cell clumps in 2,4-D-free medium were treated with ABA in the second week normal embryos with two cotyledons decreased prominently and this decrease of normal embryos also occurred in the continuous ABA treatment during culture. Thus the morphological abnormalities in somatic embryogenesis occurred by exogenous ABA treatment beyond globular stage or by continuous treatment. The length of somatic embryos with anomalous cotyledons was larger than that of normal embryos with two cotyledons in control but both the normal and anomalous somatic embryos treated with ABA were almost similar in length. Somatic embryos formed in medium with ABA were larger in size than those in control due mainly to enlarged cotyledons. The enlarged cotyledons were composed of a greater number of cells than those of control. Therefore the enlargement of cotyledon by exogenous ABA seems to be not due to the enlargement of cells in cotyledons.  相似文献

Results of molecular studies regarding the phylogenetic placement of the order Ostropales and related taxa within Lecanoromycetes were thus far inconclusive. Some analyses placed the order as sister to the rest of Lecanoromycetes, while others inferred a position nested within Lecanoromycetes. We assembled a data set of 101 species including sequences from nuLSU rDNA, mtSSU rDNA, and the nuclear protein-coding RPB1 for each species to examine the cause of incongruencies in previously published phylogenies. MP, minimum evolution, and Bayesian analyses were performed using the combined three-region data set and the single-gene data sets. The position of Ostropales nested in Lecanoromycetes is confirmed in all single-gene and concatenated analyses, and a placement as sister to the rest of Lecanoromycetes is significantly rejected using two independent methods of alternative topology testing. Acarosporales and related taxa (Acarosporaceae group) are basal in Lecanoromycetes. However, if the these basal taxa are excluded from the analyses, Ostropales appear to be sister to the rest of Lecanoromycetes, suggesting different ingroup rooting as the cause for deviating topologies in previously published phylogenies.  相似文献

【目的】探究9种多糖对凝结芽孢杆菌(Bacillus coagulans)的增殖、产酶特性的影响。【方法】将凝结芽孢杆菌分别添加至菊粉多糖(inulin polysaccharide)、刺五加多糖(Eleutherococcus senticosus polysaccharide)、壳寡糖(chitosan oligosaccharide)、防风多糖(Saposhnikovia divaricata polysaccharide)、低聚木糖(xylo-oligosaccharide)、黄芪多糖(Astragalus polysaccharide)、甘露糖(D-mannose)、白术多糖(Atractylodes macrocephala Koidz polysaccharide)和玉屏风多糖(Yu Ping Feng polysaccharide)为唯一碳源的培养基中,通过菌株生长、酶活性及其体外厌氧发酵等作为指标,筛选出最优益生元。【结果】凝结芽孢杆菌能很好地利用防风多糖、黄芪多糖、白术多糖和玉屏风多糖;添加量为4%的防风多糖和白术多糖,pH差值差异最大,蛋白酶活性差异显著(P<0.05)。体外发酵乳酸活性和总蛋白酶活性均提高,4%白术多糖的乳酸和总蛋白酶活性差异显著(P<0.05);肠道内容物发酵液16S rRNA基因高通量测序结果表明,与对照组比较,添加黄芪多糖、防风多糖、甘露糖3种益生元发酵凝结芽孢杆菌显著降低了气单胞菌(Aeromonas)、α-变形菌(α-Proteobacteria)、链球菌(Streptococcus)、志贺氏杆菌属(Shigella)等致病菌的相对丰度,提高了乳杆菌(Lactobacillus)、厚壁菌门(Firmicutes)、乳酸乳球菌(Lactococcus lactis)、产酸杆菌(Acidobacteria)的相对丰度。【结论】凝结芽孢杆菌发酵4%白术多糖具有较好的产酶性能与益生特性,二者协同发酵添加至饲料中具有较好的发展潜力。  相似文献

外源ABA对大蒜试管苗玻璃化发生和抗氧化系统的影响   总被引:1,自引:0,他引:1  
以大蒜品种‘二水早’试管苗为材料,从活性氧代谢的角度研究了外源ABA、H_2O_2和H_2O_2+ABA处理下的试管苗玻璃化率、活性氧积累与组织定位和抗氧化系统的响应特征,探讨ABA缓解试管苗玻璃化过程的机理。结果表明:(1)外源H_2O_2处理可诱导大蒜试管苗玻璃化发生,外源ABA处理下玻璃化率最低,可以缓解H_2O_2诱导的玻璃化的发生。(2)试管苗O_2~产生速率和H_2O_2含量在H_2O_2处理下最高,在ABA处理下最低;在添加H_2O_2的培养基中同时添加ABA能显著减少因外源H_2O_2处理引起的O_2~产生和H_2O_2积累。(3)试管苗CAT、POD和APX活性在外源H_2O_2处理前期(0~8d)均上升并显著高于对照,但其CAT、APX活性在处理后期(8~16d)下降,其同期POD活性也增加缓慢;各抗氧化酶的活性在外源ABA与H_2O_2+ABA处理前期(0~8d)均呈直线上升趋势,而它们在H_2O_2+ABA处理后期(8~16d)均显著高于H_2O_2处理。(4)各处理试管苗抗坏血酸和谷胱甘肽含量随处理时间先升高后降低,并以外源ABA处理下最高,外源H_2O_2处理下最低。(5)试管苗O_2~和H_2O_2产生部位主要在基部和叶尖,且外源ABA处理下组织中ROS的积累最少。(6)在ABA+H_2O_2处理下,大蒜试管苗内丙二醛含量和膜相对透性显著低于对照和H_2O_2处理。研究发现,外源ABA处理可有效降低大蒜试管苗的内源O_2~产生速率和H_2O_2含量,提高抗氧化酶活性和抗氧化物质含量,抑制活性氧在试管苗内的产生和运输,显著降低试管苗玻璃化率;外源ABA可通过增强大蒜试管苗抗氧化能力来抑制玻璃化发生。  相似文献

Leveillula on monocotyledonous plants have been recorded as L. taurica by several authors, whereas the fungus on Allium has been described as an independent species, namely L. allii, by some authors. We sequenced ca 600 bp of the rDNA ITS region for two Leveillula specimens from Allium and Polianthes (both from monocotyledons) and compared them with several already published sequences from Leveillula isolates from dicotyledons. Pair-wise percentages of sequence divergences were calculated for all Leveillula isolates. The ITS sequence of the Polianthes isolate was identical to L. taurica on Helianthus and Vicia. The sequence of the Allium isolate was 99.5 % identical to L. taurica on Euphorbia, Haplophylum, Peganum, etc. These results suggest close relationships between monocot and dicot pathogenic Leveillula species. The identity between two monocot isolates was 98.4 %. Phylogenetic analysis revealed that the two monocot isolates do not group into a clade together. This result suggests that Leveillula acquired parasitism to monocots at least twice independently.  相似文献

Summary In vitro propagation of Quassia amara L. (Simaroubaceae) was attempted using mature and juvenile explants. Attempts to establish in vitro culture using leaf and internode explants from a plant more than 15yr old were unsuccessful due to severe phenolic exudation. Plant regeneration through direct and indirect somatic embryogenesis was established from cotyledon explants. Murashige and Skoog (MS) medium with 8.9 μM N6-benzyladenine (BA) and 11.7 μM silver nitrate induced the highest number (mean of 32.4 embryos per cotyledon) of somatic embryos. Direct somatic embryogenesis as well as callus formation was observed on medium with BA (8.9–13.3 μM). Semi-mature pale green cotyledons were superior for the induction of somatic embryos. Embryos developed from the adaxial side as well as from the point of excision of the embryonic axis. More embryos were developed on the proximal end compared to mid and distal regions of the cotyledons. Subculture of callus (developed along with the somatic embryos on medium with BA alone) onto medium containing 8.9 μM BA and 11.7 μM silver nitrate produced a mean of 17.1 somatic embryos. Primary somatic embryos cultured on MS medium with 8.9 μM BA and 11.7μM silver nitrate produced a mean of 9.4 secondary somatic embryos. Most of the embryos developed up to early cotyledonary stage. Reduced concentration of BA (2.2 or 4.4 μM) improved maturation and conversion of embryos to plantlets. Ninety percent of the embryos converted to plantlets. The optimized protocol facilitated recovery of 30 plantlets per cotyledon explant within 80d. Plantlets transferred to small cups were subsequently transferred to field conditions with a survival rate of 90%.  相似文献

Recent DNA sequence analyses indicated that Nigrosabulum globosum is a cleistothecial representative of the Bionectriaceae in the Hypocreales, but morphological characters supporting this relationship are unknown. Using light and electron microscopy we followed the development of the ascomata of this species, from the formation of gametangia through to the development of mature ascospores, and observed a series of characters that confirmed its hypocrealean affinities. These included the formation of a gel-filled centrum during early stages of ascoma development, the subsequent appearance of hyaline peridial tissue enclosed within a layer we interpret as representing a melanized uniloculate stroma, apically derived paraphyses, and an ascogenous system that gives rise to asci that were both cylindrical to clavate and globose. Ascospores, previously reported to be smooth, were ornamented with a honeycomb-like reticulum and were able to germinate within the ascoma. The carbonaceous outer (stromatic) walls of the mature, grit-like cleistothecia indicate possible resistance to UV radiation and desiccation. Furthermore, the complement of germinated ascospores would enable mature ascomata to function as propagules that could quickly initiate new growth when transferred to fresh substrate. Our reexamination of N. globosum also provides data that support the hypothesized close relationship with other bionectriaceous, cleistothecial coprophiles, i.e., species of Hapsidospora, and Bulbithecium in particular.  相似文献
