To further investigate the cyclopeptldes of the Caryophyllaceae family, two new cyclopeptldes, named Arenarlphilin E (compound 1) and Arenariphilin F (compound 2), were obtained from Arenaria oreophUe J. D. Hooker using some Isolation methods, e. g. normal and reverse silica gel. By detailed spectroscopic analysis, such as FAB^+-MS, 1 D NMR, 2D NMR, the structures of Arenariphilin E (compound 1) and Arenariphilln F (compound 2) were determined as cyclo(lle^1-Gly-Val^1-Ala-Leu-lle^3-lle^2-Val^2-Pro) and cyclo(Pro^2-Pro^1-Gly^2-lle-Val-Leu-Gly^1-AiaThr- Gly^3), respectively.  相似文献

以西伯利亚百合为试材,通过半巢式PCR和RT-PCR技术分别克隆了查尔酮合成酶基因(CHS)的DNA和cDNA.生物信息学分析显示,CHS的DNA序列全长1 397 bp(登录号HM622754),包含2个外显子和1个内含子;cDNA序列编码区全长1 182 bp(登录号HQ161731),编码393个氨基酸,具有3个典型的CHS蛋白结构域:N-末端结构域(Lys3-Pro229)、C-末端结构域(Gln239-Pro389)和聚合酶Ⅲ结构域(Met1-Thr391);不同百合品种的CHS基因编码的氨基酸序列相似性高达98%,表明百合CHS基因在进化上呈现出十分保守的趋势;不同植物CHS基因序列的系统进化邻接树结果表明:百合与单子叶植物鸢尾及禾本科的水稻、大麦、玉米等亲缘关系更为接近.  相似文献

从四棱草(Schnabalia oligophylla Hand.-Mazz.)全草的甲醇提取物中分离得到一个新的环八肽,命名为四棱草环肽(1)。用波谱方法鉴定了1的结构,并以通过HPLC手性柱分析指定了除脯氨酸和甘氮酸外的氨基酸的绝对构型。活性实验显示,四棱草环肽(1)对T/B淋巴细胞有免疫抑制作用,另外,从该植物中还分离鉴定了7个已知化合物。  相似文献

【目的】南方根结线虫(Meloidogyne incognita)是一种危害严重的土传性植物病原线虫,给农业生产造成了巨大的经济损失,前期研究发现多粘类芽胞杆菌(Panebacillus polymyxa) KM2501-1具有很好的温室防治南方根结线虫效果,且可产生多种挥发性杀线虫活性物质,但对其非挥发性产物是否有杀线虫活性没有研究。本研究拟进一步分离鉴定其产生的杀线虫活性代谢产物,发掘新的杀线虫药物。【方法】对菌株KM2501-1进行液体发酵并离心收集发酵上清液,通过硅胶柱层析、高效液相色谱分离等方法得到高纯度的杀线虫活性物质,并通过液相色谱质谱联用分析、核磁共振等技术鉴定杀线虫活性物质的结构。【结果】生物活性检测显示,多粘类芽胞杆菌KM2501-1发酵上清液具有较强的南方根结线虫触杀活性,并能有效抑制南方根结线虫卵孵化,体外杀线虫效率高达87.66%,抑制卵孵化效率达92.26%。结构鉴定结果显示多粘类芽胞杆菌产生的杀线虫活性物质为环二肽类物质cyclo (Pro-Phe),800 mg/L的cyclo(Pro-Phe)杀线虫效率达84.75%。进一步的显微观测结果表明,与对照组相比,活性物质cyclo(Pro-Phe)处理后的根结线虫肠道组织紊乱、结构发生破坏。【结论】多粘类芽胞杆菌KM2501-1产生的cyclo (Pro-Phe)是一个具有杀线虫新功能的活性物质,其可能通过破坏线虫肠道杀死线虫。  相似文献

HEV 239是福建省医学分子病毒学研究中心实验室研制的一种戊型肝炎病毒(HEV)重组颗粒性蛋白疫苗,该文旨在研究HEV239蛋白疫苗在小鼠体内诱导产生特异性免疫应答的情况.将5μg HEV 239蛋白疫苗(239-Pro)、加铝佐剂疫苗(239-Vac)或加弗氏佐剂疫苗(239-CFA)肌肉注射免疫BALB/c鼠3次,第8周检测鼠血清抗HEV抗体及其亚类,同时用ELISPOT方法检测细胞毒性T细胞(CTL)应答.结果显示:239-Vac诱导的抗体滴度与239-CFA相当,高于无佐剂的239-Pro.239-Vac诱导的抗体中,IgG1/IgG2a比值显著高于239-CFA和239-Pro,主要为Th2型应答.除239-CFA之外,239-Vac和239-Pro也可诱导出一定的HEV抗原特异性I型Tc应答.提示:重组抗原HEV 239能诱导良好的抗体应答及一定的Tc1应答.  相似文献

从海洋来源的放线菌Streptomyces sp.3275中分到7个化合物,经IR,ESI-MS,NMR等光谱数据鉴定及与文献对照,确定其结构为环(脯-缬)(1),环(脯-酪)(2),环(脯-甘)(3),环(脯-异亮)(4),环(脯-亮)(5),胆甾醇(6)及1-甲基-1,2,3,4.四氢咔啉-3-羧酸(7);并采用SRB法对其抗肿瘤活性进行了测试,其中化合物2,3,5对温敏型小鼠乳腺癌细胞tsFT210显示弱的增殖抑制活性。  相似文献

The 18-membered Aib-containing cyclohexapeptides, cyclo(-Gly-Aib-Aib-Gly-Aib-Phe-) (22), cyclo(-Gly-Aib-Phe(2Me)-Gly-Aib-Aib-) (24a), cyclo(-Gly-Phe(2Me)-Aib-Gly-Aib-Aib-) (24b), and cyclo(-Gly-Phe(2Me)-Aib-Gly-Aib-Phe-) (25), have efficiently been synthesized by solution-phase techniques. The linear precursors 1a-1d were prepared by combining the 'azirine/oxazolone method' for incorporation of alpha,alpha-disubstituted alpha-amino acids (Aib, Phe(2Me)) into the peptide chains by classical peptide coupling methods for segment condensations. Deprotection of the amino and carboxy termini of 1a-1d, followed by cyclization with DEPC as the coupling reagent, gave the above-mentioned cyclic hexapeptides 22, 24a, 24b, and 25 in good yields (26-57%). The solid-state conformations of the linear hexapeptides 1d, 16 and 27, and of the cyclohexapeptides 22 and 25 have been established by X-ray crystallography.  相似文献

During 1982, structural and functional aspects of the epiphytic component in a tropical mixed seagrass meadow, have been investigated for each seagrass species separately. This meadow consisted of the seagrasses Thalassia hemprichii (Ehrenb.) Aschers., Cymodocea serrulata (R.Br.) Aschers. et Magnus, C. rotundata Ehrenb. et Hempr. ex Aschers., Halodule uninervis (Forssk.) Aschers. and Syringodium isoetifolium (Aschers.) Dandy.No significant differences were observed in floristic composition, number of algal species, abundance and diversity of the epiphytic component. On an area basis, annual mean above-ground biomass (seagrass leaves and epiphytes), amounted to 82 g ADW, of which 18% could be ascribed to the epiphytic component. The contribution of the epiphytic component to the annual mean above-ground production ranged from 16% on leaves of Thalassia hemprichii to 33% on leaves of Cymodocea serrulata. Total annual mean epiphyte production was 4.6 g ADW m−2 sediment surface day−1 (19%).When including the macroalgal component of this mixed seagrass meadow, total annual mean above-ground plant biomass amounted to 93 g ADW (212 g DW) on an area basis, of which the epiphytes contributed 15.5% (28.5% DW), the macroalgal component 12% (32.5% DW) and the seagrass leaves 72.5% (39.5% DW). Aspects of the epiphytic component (e.g., floristic composition, abundance, biomass and production) in monospecific and mixed seagrass communities are discussed.  相似文献

Three cyclic hexapeptides bearing carboxyl groups, cyclo(L-Asp-L-Phe-L-Pro)2, cyclo(L-Aad-L-Phe-L-Pro)2 (Aad represents alpha-amino adipic acid residue), and cyclo(D-Asp-D-Phe-L-Pro)2 were synthesized and investigated on conformation, complexation with metal ions, and interaction with lipid membrane. These cyclic hexapeptides were found to take several different conformations, though their reference compounds, cyclo(L-Leu-L-Phe-L-Pro)2 and cyclo[D-Asp(OMe)-D-Phe-L-Pro]2, took a single C2 symmetric conformation. Cyclo(D-Asp-D-Phe-L-Pro)2 formed complexes with Ba2+ and Ca2+, whereas cyclo(L-Asp-L-Phe-L-Pro)2 and cyclo(L-Aad-L-Phe-L-Pro)2 did not. The latter two take amphiphilic structures and were distributed to lipid membrane more easily than cyclo(D-Asp-D-Phe-L-Pro)2. Cyclo(D-Asp-L-Phe-L-Pro)2 binds Ca2+ on the lipid membrane.  相似文献

We report the detection by gas chromatography/mass spectrometry and liquid chromatography/mass spectrometry analyses of the secreted 2,5-diketopiperazines (DKPs) cyclo(-Ala-Pro), cyclo(-Gly-Pro), cyclo(-Val-Pro), cyclo(-Ile-Pro), cyclo(-Leu-Pro), cyclo(-Pro-Pro), cyclo(-HyP-Pro), cyclo(-Met-Pro), and cyclo(-Phe-Pro) produced by Bacillus pumilus. The study focuses on a marine isolate and a laboratory test strain of B. pumilus with capabilities to lyse pregrown living cell lawns of different bacterial species, among them Arthrobacter citreus. Chromatographic methods were used to analyze induced bioactive compounds. At least 13 different DKPs are produced by B. pumilus. Both strains respond with an increased production of the DKPs cyclo(-Gly-Pro), cyclo(-Ala-Pro), and cyclo(-Val-Pro) to the presence of pasteurized A. citreus cells after 4 h in a nutrient-poor liquid medium. In agar diffusion assays, these DKPs did not cause lysis zones in living cell lawns, but they did inhibit further growth of several pregrown test bacteria in microplates even at concentrations as low as 1 μg ml?1. Antibiotic substances produced by B. pumilus after 20 h of cultivation in a special lysis medium showed lytic activity in cell-free extracts of B. pumilus culture supernatants.  相似文献

Paraquat (1,1'-dimethyl-4,4'-bipyridinium), a widely used non-selective herbicide, is a redox cycling agent with adverse effects on dopamine systems. Epidemiological data have shown that exposure to paraquat is one of the several risk factors for Parkinson's disease. We have already shown that cyclo(His-Pro), an endogenous cyclic dipeptide produced by the cleavage of the thyrotropin releasing hormone, has a cytoprotective effect through a mechanism involving Nrf2 activation that decreases production of reactive oxygen species and increases glutathione synthesis. Using primary neuronal cultures and PC12 cells as targets of paraquat neurotoxicity, we addressed whether and how cyclo(His-Pro) causes cellular protective response against paraquat-mediated cell death. We found that cyclo(His-Pro) attenuated reactive oxygen species production, and prevented glutathione depletion by up-regulating Nrf2 gene expression, triggering its nuclear accumulation and activating the expression of heme oxygenase1. These protective effects were abolished by RNA interference-mediated Nrf2 knock down whereas were unaffected by RNA interference-mediated Keap1 knock down. Inhibition of heme oxygenase activity decreased cyclo(His-Pro)-induced neuroprotection. These results suggest that cyclo(His-Pro), acting as a selective activator of the brain modulable Nrf2 pathway, may be a promising candidate as neuroprotective agent that act through induction of phase II genes.  相似文献

Two new cyclic peptides, named sajaponicin C (1) and sajaponicin D (2), were isolated from the whole plants of Sagina japonica (Caryophyllaceae). Their structures were determined as cyclo(Pro(2)-Leu(2)-Tyr-Leu(1)-Phe(1)-Pro(3)-Phe(2)-Pro(1)) (1) and cyclo(Pro(1)-Pro(2)-Pro(3)-Pro(4)-Phe(1)-Gly-Thr-Ser-Phe(2)-Ile-Tyr) (2) on the basis of spectroscopic data, especially by two-dimensional (2D) NMR techniques.  相似文献
