AIMS: The goal of this study was to monitor the quantitative and qualitative bacterial flora in the intestine of hybrid tilapia in fresh fish and fish kept in frozen storage conditions for 1 year. METHODS AND RESULTS: Quantitative and qualitative analyses of the bacterial flora associated with the intestine of hybrid tilapia (Oreochromis niloticus x Oreochromis aureus) in fresh fish and fish kept in frozen storage conditions for 1 year were carried out. In fresh and frozen fish, aerobic plate count (APC) ranged from 1.6 +/- 1.2 x 10(8) to 1.5 +/- 0.9 x 10(5) CFU g(-1) in the intestine of tilapia collected from pond 1, 8.7 +/- 2.3 x 10(7) to 6.5 +/- 3.8 x 10(4) CFU g(-1) in the intestine of tilapia from pond 2, and 1.9 +/- 2.9 x 10(8) to 6.2 +/- 2.8 x 10(4) CFU g(-1) in the intestine of tilapia from pond 3. APC for all the groups of fish decreased c. 2-log cycles after 1 months frozen storage; thereafter, counts slowly declined during frozen storage for 1 year. Altogether, 16 bacterial genera were identified: Gram-negative rods (67%) dominated. Both in fresh and frozen conditions, four bacterial species viz. Shewanella putrefaciens, Corynebacterium urealyticum, Aeromonas hydrophila and Flavobacterium sp. were always present, with a prevalence of 10% in most cases. Shewanella putrefaciens was the most dominant organism (15% of the total isolates) throughout the studied period. During frozen storage some of the bacteria were not recovered, but most of the bacteria survived after prolonged freezing. CONCLUSIONS: This study describes the aerobic heterotrophic microflora found in the intestine of fresh and frozen tilapia. The unique aspect of this study concerns the data revealing the micro-organisms, which are viable after prolonged freezing. Contamination of edible portions of fish could originate from gastrointestinal sources. SIGNIFICANCE AND IMPACT OF THE STUDY: The present results may enhance knowledge in controlling the storage life of fish, and fish product quality. Bacterial activity is by far the most important factor influencing fish quality, so bacterial numbers can be used as an index of quality. Storage of frozen tilapia without evisceration could be avoided. 相似文献

Nonadditive expression contributes to heterosis in hybrids. In this study, the expression profiles of twelve lipid metabolism pathway-related genes were investigated in the intestine of Nile tilapia (Oreochromis niloticus) ♀?×?blue tilapia (Oreochromis aureus) ♂ hybrid. The expression of genes from the hybrid were assigned to nonadditive and additive expression pattern groups and compared with expression patterns from Nile tilapia and blue tilapia. In the intestine of the hybrid, apoA4B was expressed at intermediate levels, but apoB and MTP were assigned to ELD-B and ELD-N categories, respectively. The LPL and LRP1 showed transgressive up-regulation in the hybrid, but LDLR was assigned to the ELD-B category. For fatty acid uptake related genes, only FABP11a was categorized as nonadditive expression with transgressive up-regulation, while CD36 and FABP3 were categorized as additive expression in the intestine of the hybrid. Two genes in triacylglycerol metabolism, namely, FAS and DGAT2, showed transgressive up-regulation in the hybrid. Most of the genes analyzed in the present study showed nonadditive expression (8 in 12), and five genes showed transgressive up-regulation. These results indicated that the stimulation of lipid metabolism in the hybrid compared to that of its parents. The hyperactive expression of these genes in the hybrid may be associated with the growth and lipid usage vigor. 相似文献

A feeding trial was conducted to evaluate the effects of dietary vitamin E contents on the growth, ascorbate induced iron-catalyzed lipid peroxidation in post-mortem muscle and liver tissue, and Raman spectral changes in lens of juvenile hybrid tilapia (Oreochromis niloticus x O. aureus). Experimental fish were fed practical diets supplemented with 0, 50, 100, 200, 450 and 700 mg alpha-tocopheryl acetate/kg diet for 14 weeks. There was no significant difference in weight gain, feed conversion ratio and protein efficiency ratio among fish fed test diets (P>0.05). Protein content of fish fed diet containing the lowest vitamin E level was the lowest (P<0.05) among all groups. No difference was found in other body constituents among test fish (P>0.05). The thiobarbituric acid-reactive substances produced by iron-catalyzed lipid peroxidation in muscle and liver tissue of fish fed the diet without alpha-tocopheryl acetate supplementation were significantly (P<0.05) greater than those from fish fed diets containing higher levels of alpha-tocopheryl acetate. Dietary vitamin E supplementation increased the antioxidant capability of tilapia tissues against lipid peroxidation. Further, dietary vitamin E supplementation also influenced the lens cortical membrane structure of tilapia. 相似文献

The effects of aluminum on plasma ion, lipid, protein and steroid hormone concentration were evaluated in Oreochromis niloticus broodstock females. Lipid and protein concentrations from the gonads and liver were also measured. Experiments were performed at neutral and acidic water pH. Four groups of fish were tested for 96 h: 1) control conditions at neutral water pH; 2) control conditions at acidic water pH (CTR-Ac); 3) aluminum at neutral water pH (Al-N); and 4) aluminum at acidic water pH (Al-Ac). Aluminum and acidic water pH exposure caused no ionoregulatory disturbances. Total lipid concentration increased in the mature gonads and decreased in the liver, suggesting an acceleration of lipid mobilization to the ovaries in animals exposed to aluminum. However, a decreased protein concentration in ovaries was also observed. Exposure of control fish to acidic water pH caused an increased concentration of plasma 17α-hydroxyprogesterone. However, females exposed to aluminum at acidic water pH showed a decreased of plasma 17α-hydroxyprogesterone and cortisol. No differences in plasma 17β-estradiol were observed. The physiological mechanisms underlying the disturbances observed are discussed focusing on reproduction. We suggest that aluminum can be considered an endocrine disrupting compound in mature O. niloticus females. 相似文献

Studies on age and growth in fishes are essential to establish models on population dynamics. The previous issues were determined in this study by scale growth analyses. Between September 2003 and May 2004, 382 organisms were captured using gill and atarraya nets. Eight samples were developed every month a long this study. Standard length in centimeters (L(e)), total weight in grams (P9t)), scale length in centimeters (L(esc)) and growth rings were determined. The size and weight ranged from 38 to 232 mm (L(e)) and 8.7 to 311.9 g (P(t)), respectively. The growth rings analyses reveal four age groups, being the second group the most representative with 34.3% of the total sampled. The scale ring analyses showed two growth rings, one develops between November-January and the other between April-May, in good correlation with low temperature and gonad development, respectively. The growth parameters were established as: L infinity = 281.1mm, W infinity = 877.1g, K = 0.33 and T(0) = 0.88 year(-1). The growth curves based on length and weight were described by using the von Bertalanffy model: L(e) = 28.11[1 -e (-0.33(t+0.88))] and P(t) = 877.17[1-e (-0.33(t+0.88))]3. Finally, it is concluded that commercial catches in this dam are between a year and year and half-old when exploited. 相似文献

The karyotype of Oreochromis species is considered to be highly conserved, with a diploid chromosome complement of 2 n = 44. Here we show, by analysis of mitotic and meiotic chromosomes, that the karyotype of O. karongae, one of the Lake Malawi chambo species, is 2 n = 38. This difference in chromosome number does not prevent the production of inter-specific hybrids between O. niloticus (2 n = 44) and O. karongae (2 n = 38). Analysis of the meiotic chromosomes of the O. niloticus × O. karongae hybrids indicates that three separate chromosome fusion events have occurred in O. karongae. Comparison of the O. karongae and O. niloticus karyotypes suggests that these consist of one Robertsonian fusion and two fusions of a more complex nature. 相似文献

应用腹腔注射染毒法,研究不同作用时间下微囊藻毒素-LR(MC-LR)在罗非鱼各组织器官的动态分布。结果表明:除罗非鱼肌肉中MC-LR含量低于检出限(用ND表示未检出)外,罗非鱼肝脏、血清、胆囊、鳃、肠道都有MC-LR检出;且各组织器官MC-LR残留量的分布有明显的区别,其中以肝脏中残留量的平均值为最高,其变化区间为0.902~4.938mg·kg-1,血清、胆囊、鳃、肠道的MC-LR残留量变化区间分别为ND~0.390mg·L-1、ND~0.236mg·kg-1、0.0134~0.0369mg·kg-1、0.007~0.016mg·kg-1;染毒后84h,罗非鱼肝脏、鳃、血清、胆囊中的MC-LR残留量都显著降低,且血清和胆囊中的MC-LR含量低于检出限,表明罗非鱼对MC-LR具有很强的解毒机能;染毒后36h,胆囊和肝脏中的MC-LR残留量骤然下降,由此推测胆囊和肝脏可能是罗非鱼对MC-LR的主要解毒器官;试验期间,肌肉中MC-LR含量始终低于检出限,表明低剂量的MC摄入不会在罗非鱼肌肉中造成明显的残留。 相似文献

East African cichlids have evolved feeding apparatus morphologies adapted to their diverse feeding behaviors. The evolution of the oral jaw morphologies is accomplished by the diversity of bone formation during development. To further understand this evolutionary process, we examined the skeletal elements of the jaw and their temporal and sequential emergence, categorized by developmental stages, using the Nile tilapia Oreochromis niloticus as a model cichlid. We found that chondrogenesis started in Stage 17. The deposition of osteoid for the dermal bones commenced in Stage 18. The uptake of calcium dramatically shifted from the surface of larvae to the gills in Stage 20. The bone mineralization of the skeleton began in Stage 25. These data provide important information regarding the sequential events of craniofacial development in East African cichlids and lay the groundwork for studying the molecular mechanisms underlying adaptation of jaw structure to feeding behavior. 相似文献

The effects of temperature on the salinity tolerance of Mozambique-Wami tilapia hybrids (Oreochromis mossambicus x O. urolepis hornorum) were investigated by transferring 35 g/l, 25 degrees C-acclimated fish to 35, 43, 51 or 60 g/l salinity at 15, 25 or 35 degrees C for 24 h, and by assaying gill tissue for branchial Na(+), K(+)-ATPase activity at the three temperatures after acclimating the fish to 15, 25 or 35 degrees C for 2 weeks. Tilapia survived all salinities at 25 and 35 degrees C; however, at 15 degrees C, mortality was 85.7% and 100% in the 51 g/l and 60 g/l groups, respectively. There was a significant interaction between temperature and salinity, as plasma osmolality, [Na(+)] and [Cl(-)] were significantly increased at 51 and 60 g/l salinity in 35 degrees C water (P<0.001). Additionally, muscle water content was significantly reduced at 43 g/l, 15 degrees C relative to pre-transfer values (P<0.001). Branchial Na(+), K(+)-ATPase activity was reduced at 15 degrees C regardless of acclimation temperature, and 25 degrees C-acclimated gill tissue did not show an increase in activity when assayed at 35 degrees C. Results indicate that the effects of a combined temperature-salinity transfer on plasma osmolality and ion concentrations, as well as muscle water content, are greater than when either challenge is given alone. Additionally, branchial Na(+), K(+)-ATPase activity is altered when assayed at varying temperatures; in the case of 15 degrees C, regardless of acclimation temperature. 【背景】罗非鱼作为联合国粮农组织(FAO)向全世界推广养殖的优良品种之一,有多个品系,其养殖范围已遍布85个国家和地区。1956年我国从越南引进莫桑比克罗非鱼,经过养殖及推广,2006年我国罗非鱼产量达到100万t。但是,该外来物种在给我国带来良好经济价值的同时,对土著种类及水环境造成了极大的影响。【方法】对近几年珠江水系渔获物的调查数据进行整理,并观察尼罗罗非鱼早期发育形态,统计尼罗罗非鱼苗对其他鱼苗的最大捕食量及捕食规格,以分析罗非鱼生物学特性及其在珠江水系的入侵现状。【结果】尼罗罗非鱼早期发育快,卵黄营养非常丰富,比珠江水系土著种类更有竞争性;罗非鱼苗呈现很强的攻击性与捕食性;尼罗罗非鱼已经扩散到珠江水系各主要河流,并在部分江段形成优势种群。【结论与意义】尼罗罗非鱼种群快速扩张,对土著种已构成严重威胁,有必要将其列为珠江水系高危入侵种。 相似文献

The purpose of this work was to investigate the circadian melatonin system in two tropical teleost species characterized by different behavioral habits, Nile tilapia (diurnal) and African catfish (nocturnal). To do so, fish were subjected to either a control photoperiod (12L:12D), continuous light (LL) or darkness (DD), or a 6L:6D photoperiod. Under 12L:12D, plasma melatonin levels were typically low during the photophase and high during the scotophase in both species. Interestingly, in both species, melatonin levels significantly decreased prior to the onset of light, which in catfish reached similar basal levels to those during the day, demonstrating that melatonin production can anticipate photic changes probably through circadian clocks. Further evidence for the existence of such pacemaker activity was obtained when fish were exposed to DD, as a strong circadian melatonin rhythm was maintained. Such an endogenous rhythm was sustained for at least 18 days in Nile tilapia. A similar rhythm was shown in catfish, although DD was only tested for four days. Under LL, the results confirmed the inhibitory effect of light on melatonin synthesis already reported in other species. Finally, when acclimatized to a short photo-cycle (6L:6D), no endogenous melatonin rhythm was observed in tilapia under DD, with melatonin levels remaining high. This could suggest that the circadian clocks cannot entrain to such a short photocycle. Additional research is clearly needed to further characterize the circadian axis in teleost species, identify and localize the circadian clocks, and better understand the environmental entrainment of fish physiology. 相似文献

为比较罗非鱼对晶体蛋氨酸、包膜蛋氨酸的利用效果, 设计了鱼粉含量为9%的鱼粉饲料、鱼粉含量为0%的无鱼粉饲料(以豆粕等蛋白替代鱼粉), 在无鱼粉饲料基础上分别添加晶体蛋氨酸、包膜蛋氨酸, 使其蛋氨酸含量与鱼粉饲料一致, 共4种饲料, 饲喂平均体重为(1.220.07) g/尾的奥尼罗非鱼(Oreochromis niloticus O. aureus) 8周。结果表明, 各组罗非鱼增重率分别为1642.7%、1250.3%、1362.0%、1451.7%, 饲料系数分别为1.08、1.45、1.30、1.21; 在无鱼粉饲料中添加晶体蛋氨酸、包膜蛋氨酸提高增重率8.9%、16.1% (P0.05), 降低饲料系数10.34%、16.55% (P0.05); 包膜蛋氨酸组较晶体蛋氨酸组具有更高增重率和更低饲料系数(P0.05)。对摄食后不同时间点肝脏和血浆谷草转氨酶(GOT)、谷丙转氨酶(GPT)活性变化的测定表明, 鱼粉饲料组、无鱼粉饲料组肝脏GOT、GPT活性分别在摄食后第2、第4h出现高峰; 晶体蛋氨酸组在摄食后1h出现高峰, 较无鱼粉组提前; 包膜蛋氨酸组在摄食后第5h出现高峰, 较无鱼粉组滞后; 血浆GOT、GPT活性变化趋势与肝脏的基本一致。消化率试验表明, 鱼粉饲料组干物质、粗蛋白质消化率最高, 无鱼粉饲料组最低; 在无鱼粉饲料中添加晶体蛋氨酸、包膜蛋氨酸后, 干物质、粗蛋白质消化率均得到显著提高(P0.05)。上述结果表明, 在蛋氨酸缺乏的实用饲料中补充晶体或包膜蛋氨酸, 均可显著提高罗非鱼生长性能和营养物质消化率, 包膜蛋氨酸较晶体蛋氨酸具有更高的利用效率和更好的促生长效果。

The humoral immune response of the tilapia Oreochromis niloticus was evaluated using a direct ELISA. Serum was tested from fish infected with Cichlidogyrus spp. (Monogenea) and from fish injected intraperitoneally with the Cichlidogyrus spp. antigenic extract, i.e., 150 microl of the Cichlidogyrus spp. saline extract diluted in Freund's complete adjuvant (FCA) (1:1) were inoculated intraperitoneally at day 0, followed by 2 dosages of 50 microl of the same Cichlidogyrus spp. saline extract diluted in Freund's incomplete adjuvant (FIA) (1:1) at weeks 2 and 4, respectively. The humoral response was also evaluated by the double immunodiffusion test (DID) and by serum protein and total immunoglobulin (Ig) determinations. The IgM OD values in the hyperimmune fish were significantly higher than in the infected and uninfected fish groups. In the DID test, a precipitation (antigen-antibody) band was observed between the Cichlidogyrus spp. saline extract and hyperimmune sera, but not with the other groups. Increases in serum protein concentration and total Igs were observed in the immunized fish at weeks 2 and 10 postinjection. Results from this study suggest that tilapia is capable of producing an induced humoral immune response against an antigenic extract of Cichlidogyrus spp. 相似文献

The red hybrid tilapia, Oreochromis niloticus (Linnaeus, 1758) x O. mossambicus (Peters, 1852) is a fertile hybrid used in the semi-intensive level of fish culture in the Northeast of Brazil. It is a territorial cichlid and is highly aggressive towards conspecifics during the breeding season. The purpose of this study was to investigate and describe the aggressive behaviour displayed by the males of this hybrid in non-reproductive and reproductive contexts. Behavioural observations revealed that aggression displayed by the reproductive males of red hybrid tilapia included threatening, undulation, parallel, lateral and frontal attacks, chasing, escape and submission. Possession of a territory influenced male aggressiveness, which was more intense in their own territory than that observed in a neutral situation. The males built nests, irrespective of female presence. All the behavioural patterns were in accordance with those previously described for one parental species, the Nile tilapia, O. niloticus. 相似文献

Transgenic tilapia were produced using a carp -actin lacZ gene construct and the transmission of the transgene to F 1 progeny was followed. Of 36 founder fish analysed, 9 (25%) were found to be transgenic in at least one tissue and two of these were found to transmit the transgene to progeny. One male produced only three out of 28 transgene DNA positive progeny (10.7%) but none of these were found to express the transgene. The other male produced 243 out of 1601 (15.2%) lacZ expressing embryos; 40 of these were analysed by Southern blotting and all were found to be positive for the presence of the transgene but revealed two different patterns of transgene arrangement. Three different expression patterns were also observed and presumed to be a result of multiple integration events and resulting position effects. 相似文献

The present experiment was conducted to study effects of Cu, Fe and Zn on activities of digestive enzymes of the hybrid tilapia Oreochromis niloticus× Oreochromis aureus. The acidic protease activities increased 65·5 and 55·1% by addition of homogenates of digesta‐containing stomach with copper (75 mg l −1) and zinc (50 mg l −1) respectively. Addition of Cu and Zn increased the activities of protease in the hepatopancreas homogenates by 132·7 and 38·1% respectively, and reduced the activity of protease in the digesta‐containing intestine homogenates by 11·0 and 13·8% respectively. Addition of Fe (50 mg l −1) increased the acidic protease activity by 96·7% but did not alter the activities of protease in the intestine and hepatopancreas. Addition of Cu markedly inhibited activities of amylase in intestine and hepatopancreas homogenates, while Zn addition showed no effects. Addition of Fe reduced activities of amylase in the intestine homogenates by 47·9% but had no effect on amylase activities in the hepatopancreas. When Cu (75 mg kg −1), Fe (50 mg kg −1) and Zn (50 mg kg −1) were supplemented to basal diet for 3 weeks, the activities of amylase in hepatopancreas homogenates increased 125·3, 215·6 and 70·0%, respectively, the activities of amylase in intestine increased 79·8, 74·6 and 48·5%, respectively, and the activities of lipase in intestine increased 90·5, 149·8 and 84·0%, respectively. Supplementation of Cu, Fe or Zn into diet had no effects on activity of protease in all digestive organs. Therefore, the results suggest that effects of Cu, Fe and Zn on activity of digestive enzymes in vitro were different from those seen in vivo, and that the positive effects of Cu, Fe and Zn supplemented to fish diet would be valuable information for formulating fish feed. 相似文献

California Mozambique tilapia (Oreochromis mossambicus x O. urolepis hornorum) are extremely saline tolerant and have been previously shown to reduce whole-animal oxygen consumption rate (MO(2)) upon exposures to salinities greater than that of seawater (SW). In this study tilapia were acclimated to 15, 30, 45, 60 and 75 g/L salinity for 1, 5, 14, or 28 days. There was little change in plasma osmolality or muscle water content in salinities below 60 g/L, and branchial Na(+), K(+)-ATPase (NKA) activity was low in 15 and 30 g/L relative to 60 and 75 g/L. When tilapia were exposed to 75 g/L, plasma osmolality and NKA activity were significantly increased within 5 days of exposure relative to those in 15 and 30 g/L, and remained elevated over the entire 28 days acclimation, indicating that short term salinity challenges (i.e., 5 days) are predictive of longer exposure durations in this species. MO(2) following transfer to 15 and 30 g/L was elevated, reflecting the high energy demand required for switching from a hyper- to a hypo-osmoregulatory strategy. The MO(2) of 60 g/L-exposed fish was significantly reduced at 1, 5, and 14 days, relative to 30 g/L-exposed fish; however by 28 days there were no significant differences. We investigated the potential for a metabolic basis for the salinity-induced MO(2) reduction, using forward stepwise linear regression to correlate enzyme activities of brain, liver, and kidney with MO(2). Brain NKA was correlated with MO(2) after 5 days (p<0.01, r(2)=0.944) and both brain NKA and hepatic total ATPase were correlated with the reduced MO(2) at 14 days (p=0.027, r(2)=0.980 and p=0.025, r(2)=0.780, respectively). These results may indicate a tissue-level metabolic suppression, which has not been previously described as a response to hypersaline exposure in fishes. 相似文献
