Actinophage MSP2 is infectious for Streptomyces venezuelae S13. Based upon electron microscopy of coliphage T4 mixed with MSP2, MSP2 had a head about 48 +/- 2 nm wide and 87 +/- 5 nm long. DNA from polyoma virus and from coliphages T4 and T7 served as reference markers in estimating the molecular weight of MSP2 DNA from sedimentation in sucrose gradients. Denatured MSP2 DNA was estimated to be about 17 x 10(6) and double-stranded MSP2 DNA was about (36 +/- 1.6) x 10(6) in molecular weight. 相似文献

A prostaglandin F2 alpha receptor localized in plasma membranes of bovine corpus luteum cells was solubilized by treatment with Triton X-100. Sepharose chromatographies of ([3H]prostaglandin F2 alpha)-receptor complex gave a Stokes' radius of 630 nm. In the absence of detergent, aggregated forms of the receptor appeared. Sedimentation experiments of solubilized receptor in sucrose/H2O and sucrose/2H2O density gradients gave the following values: sedimentation coefficient (S20, w) 4.6 S; partial specific volume (VB) 0.78 cm3/g and frictional ratio (f/fo) 1.6. Based on the sedimentation coefficient and the Stokes' radius and assuming that the receptor is a non-glycosylated protein the molar mass of the receptor-(Triton X-100) complex was 144000 g/mol. The VB value indicated that ca. 26% of the weight represented bound detergent and that the molecular weight of the prostaglandin F2 alpha receptor is approximately 107000. 相似文献

Highly purified base plates of bacteriophage T4D were obtained from lysate of gene 19 am mutant of this phage by differential centrifugation and sucrose gradient. Base plates were studied by means of high speed sedimentation equilibrium. The molecular weight determined by this method is (6.7 plus or minus 0.2)-10-6. 相似文献

Ultrasonicated Toxoplasma gondii (RH strain) tachyzoites were fractionated into a water-soluble and a deoxycholate-soluble fraction. Polyclonal immune mouse serum was prepared by challenging chronically-infected mice with viable RH strain tachyzoites. The parasite fractions were labelled with 125I, and the radio-labelled antigens were precipitated by the immune mouse serum or a monoclonal anti-Toxoplasma antibody (FMC 20), that reacts only in the indirect hemagglutination antibody test. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and autoradiography of the immunoprecipitates showed that the water-soluble fraction contained 10 antigenic polypeptides, and the deoxycholate-soluble fraction contained seven antigenic peptides. The FMC 20 reacted against a 98,000-dalton antigen that was present in the water-soluble fraction only. 相似文献

1. Fibrin clots obtained from diluted human plasma with bovine thrombin often contain amounts of phospholipids that cannot be diminished by further plasma dilution. 2. The ;cold insoluble residue' obtained during fibrinogen preparation has a higher phosphorus content than the purified fibrinogen. 3. Evidence showed that adsorption of phospholipids or phosphorus-containing fibrinopeptides on purified fibrinogen or fibrin was unlikely. 4. O-Phosphorylserine was detected in acid hydrolysates of human fibrin. 5. On the basis of phosphorus determinations the average molecular weight of human fibrinogen cannot be less than 342000 (304000-383000) for a group of ten donors, and 265000 for two other persons, assuming 1 phosphorus atom/molecule and incomplete splitting of the phosphorus-containing fibrinopeptide. Complete splitting of the phosphopeptide would require molecular weights twice as high. 6. Fibrinolysis was a possible cause of lower phosphorus contents found in isolated fibrinogen and fibrin from a donor who showed apprehension during blood collection and in a fibrinogen preparation that had been submitted to prolonged dialysis. 相似文献

A low-angle light-scattering instrument has been used to measure molecular weights of native calf-thymus and T7 DNA. Molecular weights obtained by extrapolation of angular data to 0° from measurements above 30° are less than molecular weights from extrapolation of data taken at low angles (below 30°). The low-angle molecular weights determined for calf-thymus DNA and for T7 DNA agree well with estimates of weight-average molecular weight obtained with other techniques. The low-angle light-scattering molecular weight for calf-thymus DNA is higher than previously reported values by light scattering at angles above 30°. A concentration dependence in the scattering from DNA solutions is also observed. 相似文献

We have determined the molecular weight of some of the adenovirus serotype 2 structural proteins: penton, penton base and fibre. Physical techniques, namely neutron scattering and hydrodynamical measurements, indicate that the penton base is a trimer. This is confirmed by analysis of the virion composition based on quantitative gel scanning. This finding implies either that other proteins (e.g. protein IIIa) are essential in the architecture of the fivefold vertex of the virion, or that the usual assumption that icosahedral symmetry involves identical interactions related to the symmetry of the virion does not hold. 相似文献

Preliminary results of neutron quasi-elastic scattering experiments are reported for hydrated paracrystals of sodium deoxyribonucleic acid (NaDNA). The samples were investigated at two water contents: 3.5 +/- 1.0 and 9.5 +/- 1.5 mol H2O per mole nucleotide. The results of the scattering experiments were almost independent of whether the NaDNA fibers were oriented parallel or perpendicular to the momentum transfer. The data indicate that at the lower hydration the water molecules do not diffuse appreciably on the time scale of the neutron measurements (approximately 3 X 10(-10) s). At the higher hydration the water molecules diffuse isotropically in a sphere of 9 A in diameter with a diffusion coefficient of (5 +/- 2) X 10(-6) cm2 s-1. 相似文献

The triglyceride compositions of the milk fats of man, dog, guinea pig, cow, sheep, goat, and horse were compared by gas-liquid chromatography of the intact triglycerides and of the butyl esters of the component fatty acids. The milk fats of man, dog, and guinea pig, which were largely made up of long-chain fatty acids, showed a common pattern with major contributions made by the glycerides with 48-54 acyl carbon atoms. The milk fats of cow, sheep, and goat, which were rich in short-chain acids, showed significant proportions of triglycerides with 28-54 acyl carbon atoms. Horse milk, which contains large amounts of medium-chain fatty acids, gave a characteristic triglyceride pattern in the 26-54 carbon atoms range. The experimentally determined distributions of the molecular weights of the triglycerides of all milk fats deviated significantly from the distributions predicted by random association of the fatty acids from a single pool. The data suggest that in all species the milk fat may be formed by a partial resynthesis of preformed glycerides. 相似文献

The d. c. conductivity of five samples of NaDNA in dry state was investigated. All specimens show similar behavior, with specific resistivity at 40°C of the order of 1013–1014 ohm-cm and an energy gap of Ec = 2.04–2.25 eV. Based on the Eley's model for conductivity mechanism, the possibility of conservation of an ordered structure in the dry state (temperature range 20–80°C) is discussed. This is ascribed to the interaction of electron systems of individual bases. From the activation energy values it may be estimated that 17–19 π-electrons take part in the interaction. Photoconductivity measurements indicated activation energies Ef of 0.88–0.96 eV in the visible region. 相似文献

Arthrobacter ilicis T2 brings about a unique type of cometabolic structural rearrangement of longifolene, a sesquiterpene, resulting in the formation of an acid. The molecular weight of bovine heart mitochondrial cytochrome oxidase in 2% or 3% deoxycholate was determined by the sedimentation velocity method to be 228,000 daltons from an S20,w=8.44×10–13 sec, a D20,w=3.21×10–7 cm2 sec–1, and the
reported by others. The S20,w value was only slightly concentration-dependent. When the deoxycholate in our preparation was replaced with Tween 80 the S value increased to between 16 and 17. When one preparation in Tween 80 was allowed to stand at room temperature, the S value increased in successive determinations to reach 51.5 at the end of approximately 7 h. The minimum molecular weight of the enzyme as calculated from the heme content (determined from the absorbance at 603 nm) and total protein content (determined from total nitrogen) was 110,000. An amino acid analysis when related to the heme content yielded a minimum molecular weight of 85,000.Deceased. 相似文献
