Wool carpet samples, untreated and treated with commercial levels of permethrin, were placed on a mite-inhabited carpet in a Melbourne house for 14 months. The levels of live house dust mites in the samples were found to be high at the end of this period and to exhibit no differences according to the presence of permethrin in the carpets.  相似文献

The determination of mite numbers in dust samples depends on the effectiveness of the sampling procedure. We report on mite isolation from dust samples by flotation distinguishing between living and dead mites. The results are varied and unreproducible; ageing processes are of relevance in the case of dead mites. Living mites can be determined more reliably when using mite mobility. Two methods based on this principle, the mobility test and the heat escape method, are described and discussed. In the case of a natural analogous mite development on carpets, mite numbers ranging from 80 000 to 200 000 mites per m2 are found after approx. 3 months. It is shown that the mobility of different mite species varies. The mobility test allows the detection of mites in textile objects, and shows how mites are distributed over these objects. It is demonstrated how the infestation of clothes by mites can be determined.  相似文献

House dust mites,Dermatophagoides species (Acari: Pyroglyphidae), produce allergens, known for the provocation of asthma and other allergic reactions. To determine the time needed for complete colonisation of a new house by house dust mites, dust samples were collected from carpets of houses varying from 2 weeks to 2 years in age. In contrast to the expectation, no relation was found between age of the houses on the one hand and average levels of mite-allergensDer pI andDer pII and mite numbers on the other. However, presence of dogs appeared to be positively related to allergen levels. Furthermore, carpets in bedrooms appeared to contain more allergens than carpets in living-rooms. Finally, the age of the mattress was not related to allergen levels of bedroom floors.  相似文献

House dust mites have been reported to be the most important allergen in human dwellings. Several articles had already shown the presence of different mite species at homes in Brazil, being Pyroglyphidae, Glycyphagidae and Cheyletidae the most important families found. Pupation sites of cat flea, Ctenocephalides felis (Bouché), larvae were determined in three styles of nylon and one style of wool carpet. Nylon saxony carpet had 59.3% of pupae at the top of the pile and 40% at the base of the pile. In nylon contract carpet, 55.2% of pupae were found at the top, 42.6% in the middle, and only 2.2% at the base of the pile. Nylon loop carpet contained 59.2% of pupae at the base, 25.5% in the middle, and 15.3% in the top of the pile. Wool loop carpet had 92.4% at the base and 3.8% both in the middle and top of the pile. Bioassays comparing the control of pupae manually placed at the base of carpets to that in carpets with natural pupation showed that control of pupae in the latter was 39-68% higher. Pupal control after natural pupation was greatest in nylon saxony and nylon contract carpets and lowest in nylon loop and wool loop carpets. Additional studies demonstrated that vacuuming provided the same level of pupal control on nylon saxony carpet as a spray application of permethrin to the carpet surface. Therefore, pupae that survived chemical and mechanical control treatments in nylon saxony carpet probably pupated away from the surface of the pile. Application of permethrin to the base of nylon saxony carpet did not significantly increase control. Future bioassays with cat flea pupae in carpet should be performed after natural pupation and consider carpet make and style.  相似文献

Summary The assessment of mite numbers in textile objects is important with regard to measures aimed at mite control. The standard method is to take a dust sample and to separate the mites from the dust by means of flotation. The results of mite assessment can be improved using methods based on the natural mobility of the mites; the so-called Mobility Test makes use of the intrinsic movement tendencies of the mites, whereas the Heat Escape Method uses the mobility of the mites in connection with the effects of heat. An example concernig two jackets shows the difference in results of mite assessment when using the conventional method (dust sample and flotation) on the one hand, and the new methods (Mobility Test and Heat Escape Method) on the other. It is shown that the conventional method can only supply relative mite numbers. The new methods also enable the user to monitor more efficiently the use of acaricide agents in mite control. This is demonstrated using the example of two beds and a carpet taken from a children's bedroom. The considerably higher numbers of mites suggest that the risk of the occurence of mite allergens and their penetration into the indoor air is even greater than previously assumed.  相似文献

The correlation between climatic conditions and mite numbers in houses from rural areas was studied in 13 agricultural communities (kibbutzim and moshavim) in nine geo-climatic subregions of Israel. Mites were present in 97% of the dust samples. The average number of mites per gram of dust in the different localities ranged between 84 and 2053. The maximum number of mites (7440/g dust) was found in a carpet from a house in Geva Carmel in the northern coastal region. The most prevalent species of mites were Dermatophagoides pteronyssinus and Dermatophagoides farinae, which were found in 85.6% and 71.3% of the samples, respectively. The house dust mites D. pteronyssinus, D. farinae and Euroglyphus maynei constituted 94.8% of the mites. Most of the mites were isolated from the carpets and sofas (37.0% and 33.7%, respectively), and a smaller number from beds (29.3%). The smallest number of mites (< or = 250/g dust) were found at a minimum relative humidity (RH) of 30% and lower, with a maximum temperature of 32 degrees C and higher, i.e. in the Jordan valley and Negev mountains. A greater number of mites (250-500/g dust) were found at a minimum ambient RH of 35-40% and a maximum temperature of 32 degrees C and higher, i.e. the Hula valley. A large number of mites (500-1000/g dust) were found at a minimum RH of 35-40% with a maximum temperature of 30 degrees C and lower, i.e. in the Judean and Samarian range, as well as in upper Galilee. The largest number of mites (1000-2000/g dust) was found at a minimum RH of 45% and higher, with a maximum temperature ranging between 30 and 32 degrees C. These conditions occur in the coastal strip, the coastal plain and in the Judean and Samarian foothills. A monthly examination of two houses in Zova, a kibbutz in the Judean hills next to Jerusalem, and two houses from Palmachim, a kibbutz in the coastal region, revealed that the highest prevalence of mites was found in the months April-November and May-November, respectively. In Zova, the highest number of mites were found during the months of June and July while the highest concentrations of D. pteronyssinus-antigen (Der p I) were measured during the month of September. A positive correlation between mite numbers and the quantity of Der p I in house dust was found.  相似文献

In order to evaluate the effect of continuous, thorough vacuuming on house dust organisms and mite allergen, a cotton carpet was vacuumed every other day, six times in total. Seven weeks later, the carpet was cleaned by means of spray extraction. Samples were taken before and after this wet cleaning.In total 1150 g of dust was collected, containing approximately 174 000 arthropods (dead and alive) and 9000×106 fungal spores. In the course of the experiment, the amount of dust collected at each vacuuming decreased. The number of extracted house dust mites did not decrease significantly, but that of the predator miteCheyletus did. The number of extracted fungal spores showed a significant decrease (from 142 to 16×106 spores/m2 per minute vacuuming), as did the extracted mite allergen per m2. After 7 weeks the number of mite eggs and complete house dust mites had increased enormously. After cleaning by spray extraction another increase in the number of complete mites and mite eggs was found, while the amount of mite allergen was diminished.The population growth of the house dust mite between the 6th and the 7th vacuuming is probably due to the decrease of their most important predator,Cheyletus. After the wet cleaning a number of extra eggs hatched, probably due to the high humidity in the carpet. The procedures used in this study to combat house dust mites may have an adverse effect in the long run.  相似文献

The work is devoted to analysis of various methods of assessment and control of house dust mites population and mite allergens in city dwellings using own results and the literature data. Data about in-house dust mites biology and ecology, mechanisms of their migration and circulation in modern city conditions are presented. The comparison of several methods of mites number assessment and mite allergens exposure (classical acarological analysis, colorimetric method of guanine detection in house dust, immunochemical methods) has been performed and their advantages and disadvantages analyzed. As choice of adequate avoidance measures is one of the key question, various such measures (mechanical, physical and chemical) have been compared.  相似文献

Studies of microbial contamination were carried out on a carpet strip (acrilan) installed in a laboratory corridor and on two carpet strips (acrilan and wool) installed in two rooms in a pediatric hospital. A sampling procedure of randomly removing 8-mm plugs from the carpets was used for subsequent enumeration and identification of contaminating microorganisms. Microbial counts increased with time, reached a "plateau" at about 4 weeks, and appeared to be related to the amount of activity present in the carpeted area. Vacuuming of carpets showed only a slight reduction in the number of recoverable microorganisms. Qualitative studies on a strip of acrilan carpet installed in a hospital room indicated that Staphylococcus epidermidis and gram-positive nonsporeforming rods were the predominating microorganisms. Coagulase-positive S. aureus organisms were isolated during every sampling period, and gram-negative rods were also regularly isolated. Most nitrate-reducing gram-negative rods belonged to the Enterobacter and Escherichia groups. Survival studies conducted on carpet strips after hospital installation showed relatively constant levels of contamination for about 6 weeks, followed by a gradual reduction in numbers; coagulase-positive S. aureus were found for 35 days, whereas the proportions of most organisms remained relatively constant with time.  相似文献

This study examined the question of what effect exposure to light might play in determining the vertical distribution of house-dust mites in carpet, and the degree to which light penetrates worn and unworn carpets of different pile conformation (loop- versus cut-pile), height and colour. The effect on population increase of a diurnal lighting cycle versus continual darkness was also investigated. It was found that the penetration of light into carpets was largely unaffected by pile colour or conformation. Pile height was an important factor, however, and for a given height within the pile, light intensity was higher in carpets subjected to a greater degree of wear. This corresponded to the reduction in effective pile height that occurs with carpet use. Whilst the differences observed were sometimes large (up to two-fold for a given height within the pile), Petri dish studies suggested no mite preference for habitation of areas of low light intensity compared to high intensity. Additionally, culturing mites under a diurnal light cycle was shown to be no more efficient than culturing in complete darkness. These results suggest that exposure to light is not an important determinant of house-dust mite behaviour, or their ability to colonise textile substrates.  相似文献

The objective of this study was to evaluate thefungal concentrations within carpets of the same typeand manufacture during their first 2 years of life.The carpets tested came from several different branchoffices of a bank.A total of 60 dust samples from 20 carpets (3samples per carpet) was analysed. The age of thecarpets varied from 50 to 666 days. Analysis of therelationship between fungal concentration and time wasmade by the Pearson test.Fungal concentrations were very different withina same carpet. Moreover, species distribution was alsoheterogeneous; thus the fungi in 1 m2 carpet didnot reflect mycobiota of the whole carpet. Very localfungal contaminations (>10 CFU/mg/dust) were builtup in a relatively short time (138 days). Thehighest concentrations (maximum: 200 CFU/mg dust) weremainly due to species of coelomycetes (mitosporicfungi forming conidia in closed bodies) and to variousyeasts. Comparisons of the 20 carpets sampled did notshow significant differences; the fungal level did notincrease with time (P > 0.05). A decrease waseven noticed. The age of carpets had no significanteffect either on the number of CFU/mg dust of the mainfungal taxa (P > 0.10).It was concluded that these carpets, which wereregularly cleaned, in this particular environment, donot favour important fungal growth. Moreover, sincethe species responsible for the highest contaminationobserved produce conidia or cells not easilytransported through the air, their role in respiratoryallergy would be relatively limited.  相似文献

House dust mites (HDM) have been shown to produce allergens causing atopic allergies in human beings, known in medicine as house-dust-mite allergy or atopy. Over the past 25 years (1981–2006) a survey of dust samples from different places was made in the southern part of Poland, to determine the diversity of indoor acarofauna. In total, 1,532 samples were taken from dwellings, hospitals, libraries, research laboratories, drug-stores, and offices and other workplaces. More than 30 mite species were found of which the most abundant and common were HDM, especially Dermatophagoides farinae. Highest mite densities (g?1 dust) were noted in dwellings. This survey revealed differences in the occurrence and prevalence of various species of domestic mites between and within (dwellings in) geographical areas. This knowledge may be useful in the field of forensic medicine.  相似文献

Glasgow's mild, high-rainfall climate, combined with a deteriorating quality of housing and low standards of living in many parts of the city, makes it a particularly suitable place for thriving populations of house dust mites. The acarofauna in 124 samples of house dust from beds and carpets in seventy-four homes in Glasgow, Scotland, comprised thirty-one species of which the most abundant were Dermatophagoides pteronyssinus (Trouessart) (64.3%), Glycyphagus domesticus (De Geer) (16.7%), Euroglyphus maynei (Cooreman) (11.6%), Tarsonemus sp. (1.6%), Cheyletus eruditus (Schrank) (1.5%), C. trouessarti Oudemans (0.9%), Tarsonemus fusarii Cooreman (0.8%) and Glycyphagus destructor (Schrank) (0.7%). Mites were present in all the homes surveyed and the mean population density was found to be 97/100 mg of dust (range 2-1210). Over 47% of homes visited showed signs of disrepair associated with damp, especially unmodernized flats in old tenement buildings and 1960s council housing stock, many of which contain deprived occupants. There was a high incidence of hygrophilic species such as Glycyphagus spp., Tarsonemus spp. and Euroglyphus maynei in such homes. Samples from homes of atopic asthmatics were found to contain significantly fewer mites than those from normal volunteers (chi 2 = 54.7). This was partly due to the use of house dust mite avoidance measures (e.g. regular vacuum cleaning of mattresses as well as carpets) by some of the asthmatics.  相似文献

For many years it has been suggested that allergens derived from the house dust mite played a major role in the pathogenesis of asthma, eczema and some cases of allergic rhinitis. Recently, house dust mite allergens have been purified and specific immunoassays developed with which exposure to house dust mites and their allergens can be more easily determined. Using these tools, epidemiological studies have provided confirmatory evidence that not only is house dust mite exposure associated with the majority of cases of asthma in children and young adults, but that it is causally related to the development of asthma.  相似文献

Specific IgE against Acarus siro, Glycphagus domesticus, Tyrophagus putrescentiae, and Lepidoglyphus destructor have been investigated by ELISA in sera of 92 children. Of them, 41 were found to be specific IgE positive (≥ 0.35 IU/ml) against at least one of house dust mite species, Dermatophagoides pteronyssinus and Dermatophagoides farinae, by an immunoblot. In 65.9% of the dust mite-sensitized children, specific IgE against at least one of these mite species was found. Sensitization levels, including co-sensitization cases were found to be 35.7% against A. siro, 24.4% against T. putrescentiae, 31.7% against L. destructor, and 26.8% against G. domesticus. In non-sensitized children, dust mite sensitization level was found to be 25.5%. Breakdown of sensitization by individual species in this group was; against A. siro and T. putrescentiae at 7.8%, against L. destructor at 13.7%, and against G. domesticus at 9.8%. When all children were reckoned, 43.5% was found to be sensitized against at least one storage mite species, with sensitizations against A. siro at 18.5%, T. putrescentiae at 26.1%, L. destructor at 21.7%, and G. domesticus at 17.4%. In dust samples collected from the dwellings of children, distribution of species was found to be A. siro (17%), G. domesticus (23%), T. putrescentiae (29%), L. destructor (25%), and unidentified (6%). In Fisher''s chi-square test on SPSS program, there was a relationship between dust mite sensitization and storage mite sensitization (P < 0.05), but no meaningful relationship was found on the basis of individual mite species.  相似文献

Over the past decade, there has been growing concern regarding the role of toxigenic fungi in damp indoor environments; however, there is still a lack of field investigations on exposure to mycotoxins. The goal of our pilot study was to quantify the proportion of toxigenic Aspergillus versicolor isolates in native carpet dust from damp dwellings with mold problems and to determine whether sterigmatocystin can be detected in this matrix. Carpet dust samples (n = 11) contained from <2.5 x 10(1) to 3.6 x 10(5) (median, 3.1 x 10(4)) A. versicolor CFU/g of dust, and the median proportion of A. versicolor from total culturable fungi was 18%. Based on thin-layer chromatography detection of sterigmatocystin, 49 of 50 A. versicolor isolates (98%) were found to be toxigenic in vitro. By using high-performance liquid chromatography-electrospray ionization tandem mass spectrometry, sterigmatocystin could be detected in low concentrations (2 to 4 ng/g of dust) in 2 of 11 native carpet dust samples. From this preliminary study, we conclude that most strains of A. versicolor isolated from carpet dust are able to produce sterigmatocystin in vitro and that sterigmatocystin may occasionally occur in carpet dust from damp indoor environments. Further research and systematic field investigation are needed to confirm our results and to provide an understanding of the health implications of mycotoxins in indoor environments.  相似文献

Since guanine is not only an essential constituent of vital nucleic acids, but also the main end product of nitrogenous waste excretion in arachnids, it is a potential candidate for a hygienic index for mite activity in house dust. The public health significance of these mites is based on their production of allergen in the home environment.In 101 mattress-dust samples, total number of mites and guanine content were positively correlated. The hygienic standard of 10 mites per 0.1 g dust (as derived from Danish investigations) was met in all samples with less than 0.06% guanine in the dry dust. Where the guanine content was more than or equal to 0.25% in the dry dust, mite numbers were higher than 10 mites per 0.1 g dust in 43 of the 44 samples. The three different groups (negligible mite content, possibly allergologically relevant mite concentration and allergologically unhygienic mite situation) could be easily discriminated on the color scale.The guanine-AZO-dye reaction in vacuum cleaner dust is a strong and promising candidate for an allergologically relevant hygienic index in the home.  相似文献

It is well known that dehydrating conditions for house dust mites can be created by simply raising the temperature, causing loss of body water and eventually death. Thus, it can be expected that conditions for dust mites are less favourable on floors supplied with sub-floor heating. This was examined in a study of 16 houses with sub-floor heating and 21 without. The pattern of changes in air humidity and temperature on the floors was investigated and compared to known data of the tolerance of dust mites. Also the resident mite populations were compared. Floors with sub-floor heating had, on average, fewer mites, but the difference with unheated floors was small. It was remarkable that mite numbers were also lower in upholstered furniture. Another important observation was that some houses with sub-floor heating had high mite numbers, indicating that this type of heating is compatible with a thriving mite population. Temperature and humidity conditions of heated floors may allow mites not only to survive, but also to remain active in winter. A moderate increase in temperature, a moderate decrease inf (absolute) air humidity, or a combination of both, will suffice to keep the humidity all winter below the Critical Equilibrium Humidity, the level of air humidity that is critical for mite growth and reproduction, hence for allergen production. However, it is argued that measures to suppress allergen production by house dust mites are likely to be far more effective if taken in summer rather than in winter. The trypsin-like protease Der p 3, a major allergen of the house dust mite Dermatophagoides pteronyssinus, is synthesized as a zymogen, termed proDer p 3. No recombinant source of Der p 3 has been described yet, and the zymogen maturation mechanism remains to be elucidated. The Der p 3 zymogen was produced in Pichia pastoris. We demonstrated that the recombinant zymogen is glycosylated at the level of its propeptide. We showed that the activation mechanism of proDer p 3 is intermolecular and is mediated by the house dust mite cysteine protease Der p 1. The primary structure of the proDer p 3 propeptide is associated with a unique zymogen activation mechanism, which is different from those described for the trypsin-like family and relies on the house dust mite papain-like protease Der p 1. This is the first report of a recombinant source of Der p 3, with the same enzymatic activity as the natural enzyme and trypsin. Glycosylation of the propeptide was found to decrease the rate of maturation. Finally, we showed that recombinant Der p 3 is inhibited by the free modified prosequence T(P1)R.  相似文献
