Actin is an abundant protein in most nonmuscle cells. It has often been observed in isolated nuclei, yet cytoplasmic contamination was of course initially regarded as the most plausible origin. Numerous studies on nuclear actin appeared in the 1970s and 1980s, but the picture remained rather muddy. The viewpoint at that time was that actin-shown to move freely between cytoplasm and nucleus-was a mere "thermodynamic wanderer," transiently occupying the nucleus. More recently, evidence has been mounting that actin's presence in the nucleus is not simply governed by the laws of diffusion. The same holds true for the finding of various actin-related proteins in the nucleus, and the case for nuclear myosin, specifically myosin I, is now quite convincing. Moreover, the first intimations of functional roles of nuclear actin are now emerging. Here we examine the overall subject from cell biological and chemical perspectives. The major issue is no longer the presence of actin in the nucleus but rather its supramolecular organization, intranuclear locations, and, of course, functions. These issues interface with recent findings that reveal a surprisingly diverse repertoire of actin conformations and oligomer and polymer forms beyond monomeric G-actin and polymeric F-actin. We present ideas for advancing the nuclear actin field and call for a renewed attack on this major problem in cell biology.  相似文献

Understanding the consequences of elevated CO2 (eCO2; 800 ppm) on terrestrial ecosystems is a central theme in global change biology, but relatively little is known about how altered plant C and N metabolism influences higher levels of biological organization. Here, we investigate the consequences of C and N interactions by genetically modifying the N-assimilation pathway in Arabidopsis and initiating growth chamber and mesocosm competition studies at current CO2 (cCO2; 400 ppm) and eCO2 over multiple generations. Using a suite of ecological, physiological, and molecular genomic tools, we show that a single-gene mutant of a key enzyme (nia2) elicited a highly orchestrated buffering response starting with a fivefold increase in the expression of a gene paralog (nia1) and a 63% increase in the expression of gene network module enriched for N-assimilation genes. The genetic perturbation reduced amino acids, protein, and TCA-cycle intermediate concentrations in the nia2 mutant compared to the wild-type, while eCO2 mainly increased carbohydrate concentrations. The mutant had reduced net photosynthetic rates due to a 27% decrease in carboxylation capacity and an 18% decrease in electron transport rates. The expression of these buffering mechanisms resulted in a penalty that negatively correlated with fitness and population dynamics yet showed only minor alterations in our estimates of population function, including total per unit area biomass, ground cover, and leaf area index. This study provides insight into the consequences of buffering mechanisms that occur post-genetic perturbations in the N pathway and the associated outcomes these buffering systems have on plant populations relative to eCO2.  相似文献

Visceral leishmaniasis (VL) is a systemic protozoan disease that is transmitted by phlebotomine sandflies. Poor and neglected populations in East Africa and the Indian sub-continent are particularly affected. Early and accurate diagnosis and treatment remain key components of VL control. In addition to improved diagnostic tests, accurate and simple tests are needed to identify treatment failures. Miltefosine, paromomycin and liposomal amphotericin B are gradually replacing pentavalent antimonials and conventional amphotericin B as the preferred treatments in some regions, but in other areas these drugs are still being evaluated in both mono- and combination therapies. New diagnostic tools and new treatment strategies will only have an impact if they are made widely available to patients.  相似文献

Spatialassociations among overstory and understory species tend to increase ongradients from wet to dry climates. This shift in the strength of spatialassociations has usually been attributed to shared abiotic requirements betweencanopy species and understory assemblages within communities and/or to anincrease in habitat heterogeneity in dry climates and therefore higher betadiversity. On another hand, more important positive effects of tree canopies onunderstory species in drier climates may also explain stronger associations andhigher beta diversity. We examined these three hypotheses along a strongrainshadow gradient that occurs from the wet external Alps to the dry innerAlpsby analyzing with correspondence analysis and canonical correspondence analysisthe species composition of 290 relevés of forests dominated to differentdegrees by Abies alba and Piceaabies.We found important differences in climatic requirements forAbies and Picea, withAbies occurring in warmer and drier habitats thanPicea. The understory species associated with these twospecies showed similar correlations with temperature but not with moisture,withunderstory species of Picea-communities having strongerxeric affinities than understory species ofAbies-communities. We found no significant associationsbetween canopy species and understory composition in the external Alps despitethe fact that Abies and Piceaoccurredin substantially different environments. In contrast,Abiesand Picea occurred in more similar environments in theinner Alps, but the understory assemblages associated with eitherAbies or Picea were significantlydifferent. This increase in canopy-understory associations was in partdetermined by strong differences in moisture between southern and northernaspects in the inner Alps, which affected both canopy and understory speciesdistributions. However, differences between the canopy effects ofPicea and Abies also appeared tocontribute to stronger associations between canopy and understory species, andconsequently to increase beta diversity. This pattern only occurred on southernaspects of the inner Alps but was highly significant. Our results suggest thatspecies distributions may be continuous on the wet ends of moisture gradientsbut discrete on dry ends. Relatively discrete communities at stressful ends ofgradients appear to develop as a result of both habitat differentiation and thepositive effects of overstory species.  相似文献

In landscape ecology, correlational approaches are typically used to analyse links between local population abundance, and the surrounding habitat amount to estimate biologically-relevant landscape size (extent) for managing endangered or pest populations. The direction, strength, and spatial extent of the correlations are then sometimes interpreted in terms of species population parameters. Here we simulated the population dynamics of generalized species across spatially explicit landscapes that included two distinct habitat types. We investigated how characteristics of a landscape (structure), including the variation in habitat quality and spatial aggregation of the habitat, and the precise population-dynamic properties of the simulated species (dispersal and growth rates) affect the correlation between population abundance and amount of surrounding favourable habitat in the landscape. To evaluate these spatial extents of correlation, proportions of favourable habitat were calculated within several circles of increasing diameter centred on sampling patches of favourable habitat where population abundance was recorded.We found that the value of the correlation coefficients between population abundance and amount of surrounding favourable habitat depended on both population dynamic parameters and landscape characteristics. Coefficients of correlation increased with the variation in habitat quality and the aggregation of favourable habitat in the landscape, but decreased with the dispersal distance. The distance at which the correlation was maximized was sensitive to an interaction between the level of aggregation of the habitat and the dispersal distance; whereas the greatest distance at which a significant correlation occurred was more sensitive to the variation in habitat quality. Our results corroborate the view that correlational analyses do provide information on the local population dynamics of a species in a given habitat type and on its dispersal rate parameters. However, even in simplified, model frameworks, direct relationships are often difficult to disentangle and global landscape characteristics should be reported in any studies intended to derive population-dynamic parameters from correlations. Where possible, replicated landscapes should be examined in order to control for the interaction between population dynamics and landscape structure. Finally, we recommend using species-specific, population-dynamic modelling in order to interpret correctly the observed patterns of correlation in the landscape.  相似文献

The worldwide incidence of breast cancer affects 1.2 million women each year. In contrast to the high occurrence of this malady, a decline in mortality is reported among industrialized countries. In this respect, both awareness campaigns and substantial progress achieved in therapy and diagnosis allowed for the enhancement of the survival rate in patients with breast cancer. Undoubtedly, oncology research programs played a relevant role in the improvement of therapeutics and diagnostics for breast cancer. Major strides were reported, especially over the last decade and a half, in better understanding molecular and cellular biology events involved in breast cancer pathogenesis and progression of the disease. However, therapeutic approaches for the treatment of patients with breast cancer need further improvement. Therapeutic interventions can chronically compromise both the state of health and quality of life of breast cancer survivors. In addition, current therapeutic approaches have not significantly improved the survival rate in patients with metastatic disease. On these grounds, it is necessary to develop more efficient therapeutics and diagnostic tools, which can improve the health and quality of life of breast cancer survivors and increase the survival rate in patients with metastatic disease. In this respect, the field of cancer research has placed a particular emphasis on the elucidation of genetic and epigenetic alterations that may lead to the pathogenesis of breast cancer and contribute to its progression.  相似文献

The kinship theory of genomic imprinting predicts that conflicts of interest between parents can promote the evolution of opposed expression patterns of maternally and paternally derived alleles in the offspring. The social Hymenoptera (ants, some bees, and some wasps) are particularly suitable to test this theory, because a variety of social conflicts are predicted due to relatedness asymmetries between female and male nestmates that are a corollary of haplo-diploid sex determination. Here I argue that the kin-selection predictions for genomic imprinting in social Hymenoptera might in many cases be more complex than previously suggested, because the optimal strategy will have to take fitness effects in different castes and sexes into account.  相似文献

Plants generally react to the attack of non-host and incompatible host microorganisms by inducing pathogenesis-related (PR) genes and localised cell death (LCD) at the site of infection, a process collectively known as the hypersensitive response (HR). Reactive oxygen species (ROS) are generated in various sub-cellular compartments shortly after pathogen recognition, and proposed to cue subsequent orchestration of the HR. Although apoplast-associated ROS production by plasma membrane NADPH oxidases have been most thoroughly studied, recent observations suggest that ROS are generated in chloroplasts earlier in the response and play a key role in execution of LCD. A model is presented in which the initial outcome of successful pathogen detection is ROS accumulation in plastids, likely mediated by mitogen-activated protein kinases and caused by dysfunction of the photosynthetic electron transport chain. ROS signaling is proposed to spread from plastids to the apoplast, through the activation of NADPH oxidases, and from there to adjacent cells, leading to suicidal death in the region of attempted infection.Key words: biotic stress, chloroplasts, flavodoxin, hypersensitive response (HR), reactive oxygen species (ROS), ROS signaling  相似文献

In 1992, in a special paper in the American Journal of Botany, Ernst Mayr attempted to ‘prove’ the biological species concept (BSC) worked as well in plants as it did in animals by analyzing the flora of the Concord region of northern Massachusetts. He concluded that there were minimal difficulties when applying the BSC for the plants of this particular area, and concluded that botanists were misguided in not accepting the BSC. He suggested that what he called ‘typological’ thinking was prevalent in the taxonomic community, and that this was a factor in botanical resistance to the BSC. Typology, as defined by Mayr in his 1992 foray into botany, is to a certain extent a straw‐man and, by the late 20th Century, no longer a way of thinking in widespread use in the taxonomic community in any organismal group. Here, I examine his analysis in the light of current interest in plant diversity. Species can be characterized as hypotheses about the distribution of variation in nature, subject to test with new data of many kinds. Climate change driven advances in the date of sea ice breakup will increasingly lead to a loss of spring polar bear foraging opportunities on ringed seal pups creating a phenological trophic ‘mismatch’. However, the same shift will lead to a new ‘match’ between polar bears and ground nesting birds. This new match will be especially prevalent along the Cape Churchill Peninsula of western Hudson Bay where both polar bears and nesting snow geese are abundant. Easily foraged goose eggs will provide at least some of the earlier arriving polar bears with compensation for the energy deficit accrued through lost seal hunting opportunities. We examine the potential impact of changes in the extent and pattern of polar bear egg predation on snow goose abundance using projection models that account not only for increases in the temporal overlap of the two species but also for autocorrelation and stochasticity in the processes underlying polar bear onshore arrival and snow goose incubation. Egg predation will reduce reproductive output of the nesting lesser snow geese and, under all but trivial rates, will lead to a reduction in the size of their nesting population on the Cape Churchill Peninsula. Stochasticity associated with the asymmetrical advances in polar bear onshore arrival and the snow goose incubation period will lead to periodic mismatches in their overlap. These, in turn, will allow snow goose abundance to increase periodically. Climate driven changes in trophic matches and mismatches may reduce snow goose numbers but will not eliminate this over‐abundant species that poses a threat to Arctic landscapes.  相似文献

The composition of termite assemblages was analyzed in three caatinga sites of the Esta??o Ecológica do Seridó, located in the municipality of Serra Negra do Norte, in the state of Rio Grande do Norte, Brazil. These sites have been subjected to selective logging, and cleared for pasture and farming. A standardized sampling protocol for termite assemblages (30h/person/site) was conducted between September 2007 and February 2009. At each site we measured environmental variables, such as soil pH and organic matter, necromass stock, vegetation height, stem diameter at ankle height (DAH) and the largest and the smallest crown width. Ten species of termites, belonging to eight genera and three families, were found at the three experimental sites. Four feeding groups were sampled: wood-feeders, soil-feeders, wood-soil interface feeders and leaf-feeders. The wood-feeders were dominant in number of species and number of encounters at all sites. In general, the sites were not significantly different in relation to the environmental variables measured. The same pattern was observed for termite assemblages, where no significant differences in species richness, relative abundance and taxonomic and functional composition were observed between the three sites. The agreement between composition of assemblages and environmental variables reinforces the potential of termites as biological indicators of habitat quality.  相似文献

At the start of the 21st century, respiratory syncytial virus (RSV) remains a serious global health concern. Although RSV has traditionally been acknowledged as a leading cause of morbidity and mortality in the paediatric population, the elderly and people with suppressed immune systems are now also recognised as being at risk from serious RSV infection. This problem is currently exacerbated by the lack of an effective vaccine to prevent RSV infection. Although the virus proteins play a variety of roles during the virus replication cycle, in many cases these tasks are performed via specific interactions with host-cell factors, including proteins, carbohydrates and lipids. The way in which RSV interacts with the host cell is currently being examined using a battery of different techniques, which encompass several scientific disciplines. This is providing new and interesting insights into how RSV interacts with the host cell at the molecular level, which in turn is offering the hope of new strategies to prevent RSV infection.  相似文献

Concentrations of uranium, thorium and rare earth elements (REE) in 36 species of ectomycorrhizal (26 samples) and saprobic (25 samples) macrofungi from unpolluted sites with differing bedrock geochemistry were analyzed by inductively coupled plasma mass spectrometry (ICP-MS). Analytical results are supported by use of certified reference materials (BCR-670, BCR-667, NIST-1575a) and the reliability of the determination of uranium was verified by epithermal neutron activation analysis (ENAA). It appears that data recently published on these elements are erroneous, in part because of use of an inappropriate analytical method; and in part because of apparent contamination by soil particles resulting in elevated levels of thorium and REE. Macrofungi from unpolluted areas, in general, did not accumulate high levels of the investigated metals. Concentrations of uranium and thorium were generally below 30 and 125 μg kg−1 (dry weight), respectively. Concentrations of REE in macrofungi did not exceed 360 μg kg−1 (dry weight) and their distribution more or less followed the trend observed in post-Archean shales and loess.  相似文献
