OLAND生物脱氮系统中硝化菌群16S rDNA的DGGE分析   总被引:8,自引:0,他引:8  
为了考察生物脱氮系统中硝化菌群(氨氧化菌和亚硝酸氧化菌)的种群多样性及硝化菌群随溶解氧降低的种群变化规律,并建立一套行之有效的用于自养生物脱氮系统中功能微生物菌群的快速分子检测技术,采用DGGE(变性梯度凝胶电泳)分子检测技术对硝化菌群的16SrDNA的特异性PCR扩增产物进行了分析,结果表明:OLAND生物脱氮系统中氨氧化菌和亚硝酸氧化菌随溶解氧的降低表现出了不同的种群变化规律,氨氧化菌种群多样性受溶解氧的影响非常大,而非亚硝酸氧化菌的种群多样性比较单一,且不受溶解氧的影响。结合FISH(全细胞荧光原位杂交)分析结果表明,在OLAND限氧稳定运行后期,亚硝化单胞菌属(Nitrosomonas)是主要的氨氧化菌,占OLAND限氧亚硝化阶段反应器中总细菌数的72.5%左右。  相似文献

Ninety-six used personal care and topical OTC drug items collected from consumers in the USA were examined for the presence of microbial contaminants. Of the eye and face product type containing global preservative chemistries (i.e., acceptable for use in Japan without major restrictions), 55% yielded numbers of microorganisms in excess of 500 CFU/g (P < 0.1814). For the mascara products with global preservative chemistries, 79% yielded numbers of microorganisms in excess of 500 CFU/g (P < 0.024). Products containing global preservative chemistries accounted for 88% (n = 14) of the products that had microbial contents above 10(4) CFU/g (P < 0.001). Prominent contaminants were species of Staphylococcus, Pseudomonas, Klebsiella, Streptococcus, Lactobacillus, Bacillus, Corynebacterium, and yeast. In general, under the stress of consumer use, products preserved with global preservative chemistries did not maintain as adequate preservation as products with non-global preservatives.  相似文献

A procedure, preparing spheroplast of acetic acid bacteria, was established to elucidate the membrane structure of the organisms. Of the acetic acid bacteria, only Acetobacter aceti cells were converted into spheroplasts by the sucrose-EDTA-lysozyme system. To Gluconobacter suboxydans, a method exchanging sucrose in the system for NaCl was indispensable. This NaCl-EDTA-lysozyme system was adequate for almost all acetic acid bacteria, which were converted efficiently into spheroplasts. The existence of EDTA was not essential to the genus Gluconobacter.  相似文献

The impact of the Common Osier (Salix viminalis L.) root system on number (CFU) of heterotrophic bacteria and their production in a soil-willow filter was examined. The Osier rhizosphere was found to be suitable habitat for growth of the examined microbial group, and the root system stimulated development of heterotrophic bacteria. The rhizosphere bacteria to control soil bacteria (R:C) ratio oscillated between 2.48 and 2.75 depending on the location of sample collection. The highest abundance of bacteria as well as highest bacterial production was observed at location I, near sewage discharge onto the plot. There was a significant positive correlation between the number of heterotrophic bacteria and the bacterial production.  相似文献

通过在塑料水箱中水培空心菜(Ipomoea aquatica),研究浮床空心菜对氮循环细菌数量、分布和氮素净化效果的影响。研究结果表明,浮床空心菜氮循环细菌总数、氨化菌、亚硝化菌、硝化菌数量极显著高于空白对照(P<0.01),浮床空心菜反硝化菌数量与空白对照差异不显著(P>0.05);氨化菌在根内、根面、水体中的分布差异不大,亚硝化菌主要分布在根内和根面,硝化菌主要分布在根面,反硝化菌主要分布在水体中;试验结束时,浮床空心菜系统和空白对照对氨氮的去除率分别为91.8%和88.5%,对总氮的去除率分别为48.2%和62.1%。通过浮床空心菜各种氮循环细菌数量、分布与氮素浓度的相关性分析,发现浮床空心菜系统中氮素的去除是植物吸收,根系表面的氨化作用,以及水体中硝化、反硝化共同作用的结果,而空白对照系统中氮素的主要去除途径是微生物的硝化/反硝化作用以及氨挥发。  相似文献

比较分析投加不同微生态制剂的海水养殖系统硝化功能建立的过程,为实际应用提供依据。利用海水素构建4个海水养殖系统,通过投加硝化细菌、光合细菌、枯草芽胞杆菌3种微生态制剂以及纤维毛球作为生物膜载体,比较分析不同养殖系统硝化功能的建立过程及硝化强度差异。投加硝化细菌+光合细菌和硝化细菌+枯草芽胞杆菌系统硝化功能建立时间分别为108 h和96 h,氨氮初始质量浓度为6 mg/L时,氨氧化强度分别为1.69 mg/(L·d)和1.36 mg/(L·d);添加纤维毛球的生物膜系统与生物絮团系统硝化功能建立时间分别为96 h和120 h,氨氮初始质量浓度为6 mg/L时,氨氧化强度分别为1.36 mg/(L·d)和0.98 mg/(L·d);投加碳源系统和对照系统硝化功能建立时间分别为84 h和96 h,氨氮初始质量浓度为6 mg/L时,氨氧化强度分别为1.18 mg/(L·d)和1.36 mg/(L·d)。硝化细菌+枯草芽胞杆菌系统硝化功能建立时间更短,但系统硝化强度低于硝化细菌+光合细菌系统;生物膜系统硝化强度高于生物絮团系统且硝化功能建立更快;添加碳源能够加快系统硝化功能建立过程,但降低了硝化细菌+枯草芽胞杆菌系统的硝化强度。  相似文献

滩涂海水种植-养殖系统是一种新型的生态养殖模式.对种植红树林的滩涂种植-养殖系统中细菌的生物量分布和水质的分析结果表明,由于红树林的净化作用,该系统内的水质达到Ⅱ~Ⅲ类海水水质标准,而对照塘的处于Ⅳ、Ⅴ类水质标准.系统中处理塘的异养菌、弧菌、磷细菌和产酶类细菌的数量比未种红树林的对照塘低1~2个数量级.用CORREL软件分析了细菌与水质的相关关系,异养菌、弧菌、磷细菌和产酶类细菌的数量与水体中的氮磷含量呈正相关.其中异养菌与弧菌的相关系数为0.9205,与氮磷的相关系数为0.6535(N)、0.8342(P),表明异养菌和弧菌可作为滩涂海水养殖系统水质的生物监测指标.  相似文献

Investigations concerning the role of distribution system biofilms on water quality were conducted at a drinking water utility in New Jersey. The utility experienced long-term bacteriological problems in the distribution system, while treatment plant effluents were uniformly negative for coliform bacteria. Results of a monitoring program showed increased coliform levels as the water moved from the treatment plant through the distribution system. Increased coliform densities could not be accounted for by growth of the cells in the water column alone. Identification of coliform bacteria showed that species diversity increased as water flowed through the study area. All materials in the distribution system had high densities of heterotrophic plate count bacteria, while high levels of coliforms were detected only in iron tubercles. Coliform bacteria with the same biochemical profile were found both in distribution system biofilms and in the water column. Assimilable organic carbon determinations showed that carbon levels declined as water flowed through the study area. Maintenance of a 1.0-mg/liter free chlorine residual was insufficient to control coliform occurrences. Flushing and pigging the study area was not an effective control for coliform occurrences in that section. Because coliform bacteria growing in distribution system biofilms may mask the presence of indicator organisms resulting from a true breakdown of treatment barriers, the report recommends that efforts continue to find methods to control growth of coliform bacteria in pipeline biofilms.  相似文献

We have elaborated a system for testing the phagocytic activity of human blood platelets. Blood was sampled to heparin and ACD and centrifuged. This procedure yields platelet rich plasma (PRP). As a substrate for phagocytosis we used Staphylococcus aureus 209P. The ratio of platelets to bacteria is 1:1. Incubation of bacteria and platelets continued up to 10 minutes. Lysostaphine was introduced to the phagocytic system to destroy all bacteria outside the blood platelets. The maximal values for the percentage of phagocytizing blood platelets were 0.7% and the phagocytosis index of 1.0 was reached at 6 minutes with incubation of bacteria and platelets. The authors pay attention to the role of blood platelets in antibacterial mechanisms.  相似文献

Production and specific growth rates of attached and free-living bacteria were estimated in an oligotrophic marine system, La Salvaje Beach, Vizcaya, Spain, and in a freshwater system having a higher nutrient concentration, Butron River, Vizcaya, Spain. Production was calculated from [methyl-H]thymidine incorporation by estimating specific conversion factors (cells or micrograms of C produced per mole of thymidine incorporated) for attached and free-living bacteria, respectively, in each system. Conversion factors were not statistically different between attached and free-living bacteria: 6.812 x 10 and 8.678 x 10 mug of C mol for free-living and attached bacteria in the freshwater system, and 1.276 x 10 and 1.354 x 10 mug of C mol for free-living and attached bacteria in the marine system. Therefore, use of a unique conversion factor for the mixed bacterial population is well founded. However, conversion factors were higher in the freshwater system than in the marine system. This could be due to the different trophic conditions of the two systems. Free-living bacteria contributed the most to production in the two systems (85% in the marine system and 67% in the freshwater system) because of their greater contribution to total biomass. Specific growth rates calculated from production data and biomass data were similar for attached and free-living bacteria.  相似文献

AIMS: To develop a rapid and simple method for the assessment of metabolic activity of bacteria in natural environment. METHODS AND RESULTS: A rapid and simple multicolour digital image analysis system for enumerating viable bacteria based on active fluorescent staining has been developed. This system can accurately differentiate actively respiring bacteria and non-respiring bacteria by distinctive colour information in the digital image captured by an epifluorescence microscope equipped with low magnification objective lens. An algorithm to distinguish bacteria from considerable detritus, which produced bright fluorescence in different colours, by colour segmentation has also been developed. This system was applied to river water samples, and the total and respiratory active bacterial counts by digital image analysis were highly related to those by epifluorescence microscopy (r2 = 0.96 and 0.93, respectively). CONCLUSION: This system allowed the rapid and simple differentiation of bacteria from detritus and concurrent assessment of their metabolic activity, with results being available within 1 h. SIGNIFICANCE AND IMPACT OF THE STUDY: The low magnification image analysis allows more rapid and simple quantification of bacteria and could considerably decrease image data amount and acquisition time. This system could be easily applied to the rapid analysis of microbes in various environments.  相似文献

A two electrode system mounted as a single probe was developed to measure electrochemically the rate of reduction of a redox mediator (thionine) by bacteria. The system gave a rapid (2 min) bacterial-dependent current above 2.5 x 10(5) cfu/ml with pure cultures of bacteria, but when applied to the measurement of the bacterial contamination in samples of meat and milk it was unable to detect or quantify the contamination reliably. 为探讨间作大豆(Glycine max)对甘蔗(Saccharum officinarum)根际土壤细菌及固氮细菌多样性的影响, 收集和开发固氮菌资源, 筛选高效甘蔗联合固氮体系, 选用3个甘蔗栽培品种‘ROC22’、‘GT21’、‘B8’与大豆品种‘Guizao 2’进行间种栽培, 采用巢式PCR特异扩增细菌16S rRNA基因片段和固氮细菌nifH基因片段, 并结合变性梯度凝胶电泳(DGGE)技术, 对间作大豆的甘蔗根际土壤细菌及固氮细菌进行系统演化和多样性分析。聚类分析结果显示, 间作大豆改变了甘蔗根际土壤细菌及固氮细菌原来的群落组成结构, 尤其对固氮菌群落组成的改变更大, 但对群落物种的优势度影响较小。Shannon-Wiener多样性指数和Simpson多样性指数分析结果表明, 甘蔗-大豆间作显著影响甘蔗根际土壤中细菌和固氮菌的多样性, 其中对固氮细菌多样性的影响较大。不同甘蔗品种的根际土壤细菌和固氮菌在间作大豆条件下表现出不同的多样性, ‘ROC22’和‘GT21’间作处理甘蔗根际土壤固氮细菌的Shannon-Wiener多样性指数显著高于单作处理, 而‘ROC22’与大豆间作处理的甘蔗根际土壤固氮菌多样性最为丰富。在大豆生长盛期, 间作处理的甘蔗根际土壤细菌多样性最为丰富, 不同处理间的差异也最大, 随后下降。总体来看, 甘蔗-大豆间作显著地影响根际土壤细菌和固氮菌的群落结构和群落多样性, 有助于对甘蔗合理间作栽培模式的认识和筛选高效甘蔗联合固氮体系。  相似文献

Using a variety of techniques the distribution of the glutathione-regulated KefC K(+)-transport system among bacterial species was investigated. The presence of similar systems in a number of Gram-negative bacteria was demonstrated. In contrast, the system appeared to be absent from most Gram-positive bacteria tested with the exception of Staphylococcus aureus. Using the cloned Escherichia coli kefC gene as a probe for Southern hybridization it was shown that only limited DNA sequence homology exists with other bacteria, even when closely related members of the enteric group were examined.  相似文献

Relationship between bacteria and ciliates was investigated in continuous cultures with and without the recycling of flocculated biomass. Monocultures of ciliates of various ecological types were used together with mixed cultures of bacteria or bacteria and flagellates. The effect of ciliates on the distribution of flocculated and dispersed, biomass was tested with respect to characterizing an optimum mixed culture for activated-sludge treatment. Monoprotozoal populations were not successful in stabilizing the morphology of a bacterial community in a system with recycling of flocculated biomass. Stabilization was observed (with particular ecological types of ciliates) in the system with a high residence time without recycling and/or in the system with the recycling inoculated by both flagellates and ciliates.  相似文献

CRISPR (Clustered regularly interspaced short palindromic repeats)是近几年发现的一种广泛存在于细菌和古菌中,能够应对外源DNA干扰(噬菌体、病毒、质粒等),并提供免疫机制的重复序列结构。CRISPR系统通常由同向重复序列、前导序列、间隔序列和CRISPR相关蛋白组成。本研究以醋酸发酵中常见3个属醋杆菌属(Acetobacter)、葡糖醋杆菌属(Gluconacetobacter)和葡糖杆菌属(Gluconobacter)的48个菌株为研究对象,通过其基因组上CRISPR相关基因序列的生物信息学分析,探索CRISPR位点在醋酸菌中的多态性及其进化模式。结果表明48株醋酸菌中有32株存在CRISPR结构,大部分CRISPR-Cas结构属于type I-E和type I-C类型。除了葡糖杆菌属外,葡糖醋杆菌属和醋杆菌属中的部分菌株含有II类的CRISPR-Cas系统结构(CRISPR-Cas9)。来自不同属菌株的CRISPR结构中重复序列具有较强的保守性,而且部分菌株CRISPR结构中的前导序列具有保守的motif (与基因的转录调控有关)及启动子序列。进化树分析表明cas1适合用于醋酸菌株的分类,而不同菌株间cas1基因的进化与重复序列的保守性相关,预示它们可能受相似的功能选择压力。此外,间隔序列的数量与噬菌体数量及插入序列(Insertion sequence, IS)数量有正相关的趋势,说明醋酸菌在进化过程中可能正不断受新的外源DNA入侵。醋酸菌中CRISPR结构位点的分析,为进一步研究不同醋酸菌株对醋酸胁迫耐受性差异及其基因组稳定性的分子机制奠定了基础。  相似文献

An in vitro system to measure the adhesion of bacteria to human, eukaryotic cells was devised. Adhesion indices for test strains of bacteria could be calculated. Significant differences were then observed between various strains of Escherichia coli from a variety of sources, in their ability to adhere. The possible applications of the test, especially for the routine screening of bacteria for adhesion and for inhibitors of attachment, were considered.  相似文献
