Abstract

Phytoremediation of heavy metal contaminated soils represents a promising technique and salt-tolerant hyperaccumulators for multiple metals are the need of time. Therefore, phytoremediation potential of four salt-tolerant grass species [Dhab (Desmostachya bipinnata), Kallar (Leptochloa fusca), Para (Brachiaria mutica) and Sporobolus (Sporobolus arabicus Boiss)] was evaluated for cadmium (Cd) and lead (Pb) in a hydroponic study. The plants were harvested after a growth period of 3 months in a nutrient solution containing different levels of Cd (0, 5, and 25?mg?L?1) and Pb (0, 25, and 125?mg L?1). Results indicated that Dhab grass showed the highest root and shoot dry matter yield followed by Para, Kallar and Sporobolus grass irrespective of metal or its level under which they were grown. All the grass species showed considerable Cd-accumulating potential with an accumulation of >150?mg kg?1of shoot dry matter at a higher level of Cd-contamination (25?mg?L?1). While in case of shoot Pb-accumulation only Para grass performed well and accumulated Pb >1000?mg kg?1 of shoot dry matter at the higher level of Pb-contamination (125?mg?L?1). Moreover, Para and Dhab grasses performed better for shoot Cd-uptake, while only Para grass showed promising shoot Pb uptake potential. In conclusion, these grass species could be penitentially used for phytoremediation of salt-affected Cd and Pb contaminated soils.  相似文献

A greenhouse experiment was conducted to evaluate the effectiveness of arbuscular mycorrhizal (AM) fungi in phytoremediation of lead (Pb)-contaminated soil by vetiver grass. Experiment was a factorial arranged in a completely randomized design. Factors included four Pb levels (50, 200, 400, and 800 mg kg?1) as Pb (NO3)2, AM fungi at three levels (non mycorrhizal (NM) control, Rhizophagus intraradices, Glomus versiforme). Shoot and root dry weights (SDW and RDW) decreased as Pb levels increased. Mycorrhizal inoculation increased SDW and RDW compared to NM control. With mycorrhizal inoculation and increasing Pb levels, Pb uptake of shoot and root increased compared to those of NM control. Root colonization increased with mycorrhizal inoculation but decreased as Pb levels increased. Phosphorus concentration and uptake in shoot of plants inoculated with AM fungi was significantly higher than NM control at 200 and 800 mg Pb kg?1. The Fe concentration, Fe and Mn uptake of shoot in plants inoculated with Rhizophagus intraradices in all levels of Pb were significantly higher than NM control. Mycorrhizal inoculation increased Pb extraction, uptake and translocation efficiencies. Lead translocation factor decreased as Pb levels increased; however inoculation with AM fungi increased Pb translocation.  相似文献

Coronopus didymus was examined in terms of its ability to remediate Pb-contaminated soils. Pot experiments were conducted for 4 and 6 weeks to compare the growth, biomass, photosynthetic efficiency, lead (Pb) uptake, and accumulation by C. didymus plants. The plants grew well having no visible toxic symptoms and 100% survivability, exposed to different Pb-spiked soils 100, 350, 1500, and 2500 mg kg?1, supplied as lead nitrate. After 4 weeks, root and shoot concentrations reached 1652 and 502 mg Pb kg?1 DW, while after 6 weeks they increased up to 3091 and 527 mg Pb kg?1 DW, respectively, at highest Pb concentration. As compared to the 4 week experiments, the plant growth and biomass yield were higher after 6 weeks of Pb exposure. However, the chlorophyll content of leaves decreased but only a slight decline in photosynthetic efficiency was observed on exposure to Pb at both 4 and 6 weeks. The Pb accumulation was higher in roots than in the shoots. The bioconcentration factor of Pb was > 1 in all the plant samples, but the translocation factor was < 1. This suggested C. didymus as a good candidate for phytoremediation of Pb-contaminated soils and can be used for future remediation purposes.  相似文献

Suitable plant species are able to accumulate heavy metals and to produce biomass useful for non-food purposes. In this study, three endemic Mediterranean plant species, Atriplex halimus, Portulaca oleracea and Medicago lupulina were grown hydroponically to assess their potential use in phytoremediation and biomass production. The experiment was carried out in a growth chamber using half strength Hoagland's solutions separately spiked with 5 concentrations of Pb and Zn (5, 10, 25, 50, and 100 mg L?1), and 3 concentrations of Ni (1, 2 and 5 mg L?1). Shoot and root biomass were determined and analyzed for their metals contents. A. halimus and M. lupulina gave high shoot biomass with relatively low metal translocation to the above ground parts. Metals uptake was a function of both metals and plant species. It is worth noting that M. lupulina was the only tested plant able to grow in treatment Pb50 and to accumulate significant amount of metal in roots. Plant metal uptake efficiency ranked as follows: A. halimus > M. lupulina > P. oleracea . Due to its high biomass production and the relatively high roots metal contents, A. halimus and M. lupulina could be successfully used in phytoremediation, and in phytostabilization, in particular.  相似文献

A pot experiment was conducted to investigate the effect of iron plaque on Pb uptake by and translocation in Carex cinerascens Kukenth. grown under open-air conditions. Using Scanning Electron Microscopy and Energy Dispersive X-Ray Spectrometry, iron plaque was present as an amorphous coating on root surfaces with uneven distribution. The amount of iron plaque increased significantly with increasing Fe additions regardless of Pb additions. The presence of iron plaque on the root surface of Carex cinerascens Kukenth. increased the concentrations of Pb adsorbed by iron plaque. The Pb percentage in whole roots increased by 14.52% at 500 mg kg?1 Fe treatment than at 0 mg kg?1 Fe, and the distribution coefficient (DC) of Pb and translocation factor (TF) root increased with Fe additions, but translocation factor (TF) shoot decreased with Fe additions. The results suggested that iron plaque could promote the translocation of Pb from soil to roots to some extent, and it played a role to reduce heavy metals pollution of Poyang Lake wetland.  相似文献

In a hydroponic setting, we investigated the possible role of phytochelatins (metal-binding peptides) in the lead (Pb) tolerance of vetiver grass (Vetiveria zizanioides L.). Pb was added to the nutrient medium at concentrations ranging from 0 to 1,200 mg L?1. Furthermore, we simulated the effect of soil phosphorus (P) on potentially plant available Pb by culturing vetiver grass in P-rich nutrient media. After 7 days of exposure to Pb, we evaluated the Pb uptake by vetiver grass. Results indicate that vetiver can accumulate Pb up to 3,000 mg kg?1 dry weight in roots with no toxicity. Formation of lead phosphate inhibited Pb uptake by vetiver, suggesting the need for an environmentally safe chelating agent in conjunction with phytoremediation to clean up soils contaminated with lead-based paint. Unambiguous characterization of phytochelatins (PCn) was possible using high pressure liquid chromatography coupled with electrospray ionization mass spectrometry (LC-ESMS). Vetiver shows qualitative and quantitative differences in PCn synthesis between root and shoot. In root tissue from vetiver exposed to 1,200 mg Pb L-1, phytochelatins ranged from PC1 to PC3. Collision-induced dissociation of the parent ion allowed confirmation of each PCn based on the amino acid sequence. Possible Pb-PC1 and Pb2-PC1 complexes were reported in vetiver root at the highest Pb concentration. The data from these experiments show that the most probable mechanism for Pb detoxification in vetiver is by synthesizing PCn and forming Pb–PCn complexes.  相似文献

Glutathione (GSH) plays a central role in the plant tolerance against the toxic effects of metals. It is a key antioxidant and acts as a cofactor for glutathione S-transferase (GST). The main objective of this study was to determine the Pb tolerance and bioaccumulation by Dodonaea viscosa (L.) Jacq. and their relation to GSH production and GST activity. The relationship between the Pb tolerance and bioaccumulation by D. viscosa and the effect of the exposure time on the GSH production or the GST activity was assessed in trials with perlite under different Pb treatments. D. viscosa showed a remarkable tolerance to Pb [half-inhibitory concentration (IC50) = 2,797 mg kg?1] and accumulated up to 11,428 mg Pb kg?1 in dry roots with a limited translocation to shoots without any signs of phytotoxicity after 105 days of exposure. The stress caused by the fast Pb uptake rate (489 mg kg?1 day?1) during the first 10 days of exposure was strongly correlated to increased GSH contents (~1.3-fold) and GST activities (~3.6-fold) in both shoots and roots. The results indicate that the Pb stress triggered a defense mechanism that involved increased contents of GSH and GST activities, suggesting that both variables are involved in the tolerance of D. viscosa against Pb toxicity.  相似文献

Interactive effects of two heavy metal pollutants Cd and Pb in the growth medium were examined on their uptake, production of reactive oxygen species (ROS), induction of oxidative stress and antioxidative defence responses in Indica rice (Oryza sativa L.) seedlings. When rice seedlings in sand culture were exposed to 150 μM Cd (NO3)2 or 600 μM Pb (CH3COO)2 individually or in combination for 8–16 days, a significant reduction in root/shoot length, fresh weight, relative water content, photosynthetic pigments and increased production of ROS (O2˙? and H2O2) was observed. Both Cd and Pb were readily taken up by rice roots and localisation of absorbed metals was greater in roots than in shoots. When present together in the growth medium, uptake of both the metals Cd and Pb declined by 25–40 %. Scanning electron microscope (SEM) imaging of leaf stomata revealed that Pb caused more distortion in the shape of guard cells than Cd. Dithizone staining of roots showed localisation of absorbed Cd on root hairs and epidermal cells. Both Cd and Pb caused increased lipid peroxidation, protein carbonylation, decline in protein thiol and increase in non-protein thiol. The level of reduced forms of non-enzymic antioxidants glutathione (GSH) and ascorbate (AsA) and their redox ratios (GSH/AsA) declined, whereas the activities of antioxidative enzymes superoxide dismutase (SOD) and guaiacol peroxidase (GPX) increased in metal treated seedlings compared to controls. In-gel activity staining also revealed increased intensities of SOD and GPX isoforms with metal treatments. Catalase (CAT) activity increased during early days (8 days) of metal exposure and declined by 16 days. Results suggest that oxidative stress is an important component in expression of Cd and Pb toxicities in rice, though uptake of both metals gets reduced considerably when present together in the medium.  相似文献

The current study evaluated the effect of groundwater and wastewater irrigation on lead (Pb) accumulation in soil and vegetables, and its associated health implications. A pot experiment was conducted in which spinach (Spinacia oleracea), radish (Raphanus sativus), and cauliflower (Brassica oleracea) were irrigated with groundwater and wastewaters containing varying concentrations of Pb. Lead contents were measured in wastewaters, soils and root and shoot of vegetables. We also measured health risk index (HRI) associated with the use of vegetables irrigated by wastewaters. Results revealed that Pb contents in groundwater and wastewater samples (range: 0.18–0.31 mg/L) were below the permissible limits (0.5 mg/L) set by the Food and Agriculture Organization (FAO). Application of Pb-containing groundwater and wastewater increased Pb concentration in soil and vegetables. Lead concentrations in all soils ranged from 10 to 31 mg/kg and were below the permissible limits of 300 mg/kg set by the European Union. Significant Pb enrichment was observed in the soils whereby all types of vegetables were grown and assessed for Pb risk. Our data showed that Pb contents, in all three vegetables (21–28 mg/kg DW), were higher than the permissible Pb limit of FAO (5 mg/kg Dry Weight (DW)). The HRI values were > 1.0 for radish and cauliflower. It is proposed that Vehari city wastewater/groundwater must be treated prior to its use for irrigation to avoid vegetable contamination by Pb, and as such for reducing Pb-induced human health risk.  相似文献

This study assessed the potential abilities of Scirpus triqueter for phytoremediation of soils contaminated with Pb-PAHs, amended with environment-friendly surfactant alkyl polyglucoside (APG). The effects of APG on the removal of PAHs from soil and the plant uptake and translocation of Pb were tested with plant growth and soil enzymatic activities. Experiments demonstrated that APG has an ability to facilitate PAH degradation and Pb uptake in the plant body at appropriate concentrations (20–40mg L?1). The highest PAH removal rate was observed in 30 mg L?1 APG treatment, and the highest accumulation of Pb was detected as 40 mg L?1 APG. Experiments documented the effects of APG on plant growth, soil enzymatic activity, bioaccumulation and translocation of Pb in Scirpus triqueter. Results indicated that the addition of appropriate APG enhanced PAH removal rate and increased plant uptake and translocation of Pb.  相似文献

This study was conducted to investigate the uptake of lead (Pb) and arsenic (As) from contaminated soil using Melastoma malabathricum L. species. The cultivated plants were exposed to As and Pb in separate soils for an observation period of 70 days. From the results of the analysis, M. malabathricum accumulated relatively high range of As concentration in its roots, up to a maximum of 2800 mg/kg. The highest accumulation of As in stems and leaves was 570 mg/kg of plant. For Pb treatment, the highest concentration (13,800 mg/kg) was accumulated in the roots of plants. The maximum accumulation in stems was 880 mg/kg while maximum accumulation in leaves was 2,200 mg/kg. Only small amounts of Pb were translocated from roots to above ground plant parts (TF < 1). However, a wider range of TF values (0.01–23) for As treated plants proved that the translocation of As from root to above ground parts was greater. However, the high capacity of roots to take up Pb and As (BF > 1) is indicative this plants is a good bioaccumulator for these metals. Therefore, phytostabilisation is the mechanism at work in M. malabathricum's uptake of Pb, while phytoextraction is the dominant mechanism with As.  相似文献

Iris pseudacorus L. (yellow flag) is a wide-use wetland plant for constructed wetlands for removing metals from wastewater. This study aims to understand effects of root iron plaque on sequestration and translocation of Cr and Ni in yellow flag seedlings using a hydroponic experiment. Yellow flag seedlings (4-week-old seedlings with 4–6 leaves) with or without iron plaque induction (at 50 mg Fe2+ L?1 for 72 hours) were spiked for 6 days in the Hoagland solution with Cr or Ni at 0.5, 5, and 50 mg L?1, equivalent to 1, 10, 100 times of thresholds of surface water quality, respectively. Results indicated that root iron plaque significantly reduced translocation of Cr and Ni to root but increased from root to shoot. Root iron plaque formation counteracted Cr toxicity to yellow flag seedlings while the control showed Cr toxicity to root at 5 mg L?1and to shoot at 50 mg L?1 with significant biomass loss. Neither Ni exposures caused significant biomass loss nor root iron plaque formation significantly changed Ni distribution among plant parts. Our study suggests that root iron plaque effects on metal sequestration and translocation in yellow flag seedlings were metal-dependent.  相似文献

Abstract

Kaempferia galanga is a monocotyledonous plant of the Zingiberaceae family, commonly utilized for medicinal purposes. This study evaluates the effect of different concentrations of sucrose, benzylaminopurine (BA) and photoperiod on in vitro propagation of K. galanga. Murashige and Skoog (MS) medium supplemented with 5 mg L?1 BA and 30 g L?1 sucrose, and a photoperiod with 4 h of light induced the highest shoot proliferation (7.4 ± 1.0 shoots/explant) and the highest number of roots/shoot (31.3 ± 3.2). On the contrary, the maximum shoot height (4.7 ± 0.7 cm) and the highest number of leaves/shoot (4.7 ± 0.2) were obtained from cultures using MS medium supplemented with 30 g L?1 sucrose but without BA, and exposed to 16 h of light. Hence MS medium supplemented with 5 mg L?1 BA and 30 g L?1 sucrose, and incubated under a 4 h light/20 h dark photoperiod was chosen as the optimal protocol for mass multiplication of K. galanga. This in-vitro technique can facilitate the production of a large number of uniform plants of K. galanga, irrespective of the seasonal factor, and could be used as a tool for conservation of the species.  相似文献

Pisum sativum plants were treated for 3 days with an aqueous solution of 100 μM Pb(NO3)2 or with a mixture of lead nitrate and ethylenediaminetetraacetic acid (EDTA) or [S,S]-ethylenediaminedisuccinic acid (EDDS) at equimolar concentrations. Lead decline from the incubation media and its accumulation and localization at the morphological and ultrastructural levels as well as plant growth parameters (root growth, root and shoot dry weight) were estimated after 1 and 3 days of treatment. The tested chelators, especially EDTA, significantly diminished Pb uptake by plants as compared to the lead nitrate-treated material. Simultaneously, EDTA significantly enhanced Pb translocation from roots to shoots. In the presence of both chelates, plant growth parameters remained considerably higher than in the case of uncomplexed Pb. Considerable differences between the tested chelators were visible in Pb localization both at the morphological and ultrastructural level. In Pb+EDTA-treated roots, lead was mainly located in the apical parts, while in Pb+EDDS-exposed material Pb was evenly distributed along the whole root length. Transmission electron microscopy and EDS analysis revealed that in meristematic cells of the roots incubated in Pb+EDTA, large electron-dense lead deposits were located in vacuoles and small granules were rarely noticed in cell walls or cytoplasm, while after Pb+EDDS treatment metal deposits were restricted to the border between plasmalemma and cell wall. Such results imply different ways of transport of those complexed Pb forms.  相似文献

Arbuscular mycorrhizal fungi (AMF) can promote plant growth and reduce plant uptake of heavy metals. Phosphorus (P) fertilization can affect this relationship. We investigated maize (Zea mays L.) uptake of heavy metals after soil AMF inoculation and P fertilization. Maize biomass, glomaline and chlorophyll contents and uptake of Fe, Mn, Zn, Cu, Cd and Pb have been determined in a soil inoculated with AMF (Glomus aggregatum, or Glomus intraradices) and treated with 30 or 60 µg P-K2HPO4 g?1 soil. Consistent variations were found between the two mycorrhizal species with respect to the colonization and glomalin content. Shoot dry weight and chlorophyll content were higher with G. intraradices than with G. aggregatum inoculation. The biomass was highest with 30 µg P g?1 soil. Shoot concentrations of Cd, Pb and Zn decreased with G. aggregatum inoculation, but that of Cd and Pb increased with G. intraradices inoculation. Addition of P fertilizers decreased Cd and Zn concentrations in the shoot. AMF with P fertilization greatly reduced maize content of heavy metals. The results provide that native AMF with a moderate application rate of P fertilizers can be exploited in polluted soils to minimize the heavy metals uptake and to increase maize growth.  相似文献

Activities at root-soil interface determine the solubility and uptake of metals by plants. Metal accumulation in plant species (Imperata cylindrical, Cynodon dactylon, Eleucine indica, Gomphrena celosoides, Sporobolus pyramidalis, Chromolaena odorata and Rhynchospora corymbosa) growing on Pb contaminated site as influenced by variations in physico-chemical characteristics, dissolved organic matter (DOM), Pb fractionation and different functional groups (using Fourier Transmittance Infra-red) of rhizospheric and non-rhizospheric soils was assessed. The electrical conductivity (2660–5520 µs) and Pb concentrations (51390.0–64080.0 mg/kg) were more in non-rhizospheric than rhizospheric soils having 276 µs to 3160 µs EC and 3289.0 to 44850.0 mg/kg Pb. More nutrients, DOM and carbohydrates functional groups (C-O; 1100 -1000 and O-H; 3700–3600) were found in rhizospheric compared to non-rhizospheric soils. The pH was slightly acidic (5.0–5.54) and E. indica with the lowest pH (5.0) accumulated highest Pb concentrations in shoot (8030 mg/kg) and root (16380 mg/kg) while C. odorata with highest values of pH, P, Ca and Mg in rhizospheric soil accumulated the least (root; 331.6 and shoot: 209.0 mg/kg). Pb was more in organic and residual fractions of rhizospheric and non-rhizospheric soils respectively. Reduction in pH, EC coupled with nutrients and DOM availability increased Pb uptake by plants.  相似文献

Background and Aims

The effects of Sb(V), alone or combined with Se, on the growth and root development of plants are unknown. The aim of this study is to investigate the interaction between selenite and different forms of Sb and the effects on their uptake in rice and on rice root morphology.

Methods

A hydroponic experiment was conducted that contained fourteen treatments. The treatment levels for Se were 0.5 and 1 mg L?1, and the treatment levels for Sb(III) and Sb(V) were 5 and 15 mg L?1.

Results

Sb(V) alone significantly reduced the surface area, mean diameter and volume of the roots, whereas Sb(III) alone reduced the values of most parameters of root morphology. The addition of 1 mg L?1 Se significantly enhanced the surface area, number of medium roots, and Sb concentration in the roots subjected to 15 mg L?1 Sb(V), but it decreased the number of root forks, the number and proportion of fine roots, and the shoot Sb concentration under exposure to 15 mg L?1 Sb(III). When the plants were subjected to 1 mg L?1 Se, the addition of 15 mg L?1 Sb(III) markedly reduced the shoot and root Se concentrations and the number of root tips, root forks, and fine roots and increased the mean root diameter. However, the addition of Sb(V) did not significantly affect the root and shoot Se concentrations but significantly decreased the number of root forks and fine roots and increased the proportion of medium roots.

Conclusions

Se and Sb(III) showed antagonistic effects on uptake in the shoots, but not in the roots, of paddy rice. A range of Se concentrations could stimulate the uptake of Sb in both the shoots and roots of paddy rice exposed to Sb(V).

The effects of growth-promoting hormone gibberellic acid 3 (GA3) on physiology, Pb phytoextraction, and metal detoxification mechanisms in Lolium perenne were studied. Results showed that addition of GA3 alone at lower doses (1 or 10 μM) facilitated antioxidant defense of L. perenne under Pb stress, decreased the toxicity of Pb in plant shoot by increasing the proportion of Pb in cell wall, hence significantly enhanced photosynthesis and plant growth, as well as Pb uptake and accumulation in L. perenne (P < 0.05). However, these indicators showed the opposite changes when treated with GA3 at a higher dose (100 μM). Of the total Pb in plant shoot, 36–51% was associated with cell wall, and 31–40% was soluble fraction, while 41.4–49.7% was NaCl extractable, 24.6–35.4% HAc extractable followed by other fractions. These findings suggest that Pb fixation by pectates and proteins in cell wall and sequestration in vacuole are responsible for Pb detoxification in plant, and the GA3 at 1 μM appears to be optimal for enhancing Pb phytoextraction by L. perenne from Pb polluted soils.  相似文献

Somatic embryogenesis and organogenesis in Lilium pumilum were successfully regulated by picloram, α-naphthaleneacetic acid (NAA), and 6-benzyladenine (BA). In organogenesis, the highest shoot regeneration frequency (92.5%) was obtained directly from bulb scales on Murashige and Skoog (MS) medium containing 2.0 mg L?1 BA and 0.2 mg L?1 NAA, while organogenic callus (OC) formed from leaves on MS medium supplemented with 1.0 mg L?1 BA and 0.5 mg L?1 NAA. Following subculture, 76.7% of OC regenerated shoots. In somatic embryogenesis, the combination of picloram and NAA increased the amount of embryogenic callus (EC) that formed with a maximum on 90.7% of all explants which formed 11 somatic embryos (SEs) per explant. Differences between EC and OC in cellular morphology and cell differentiation fate were easily observed. SEs initially formed via an exogenous or an endogenous origin. The appearance of a protoderm in heart-shaped SE and the bipolar shoot–root development in oval-shaped SE indicated true somatic embryogenesis. This protocol provides a new and detailed regulation and histological examination of regeneration pattern in L. pumilum.  相似文献

Polyvinyl alcohol–sodium alginate (PVA–SA) matrix was fabricated and red algae Jania rubens was embedded for removal of lead from aqueous solutions. The Pb(II) uptake rate was rapid primarily at 1 h and equilibrium was achieved within 2 h. The optimum pH was 5, the data were well fitted by Langmuir and Freundlich models, and RL values are in the range of 0.1–0.38. The sorption capacity (qe) of PVA–calcium alginate (CA)–J. rubens matrix increased from 10.77 to 37.195 mg g?1 with increasing Pb(II) concentration from 24.86 to 98.75 mg L?1 at the temperature of 30°C and pH 5. The sorption capacity (qe) and maximum biosorption (qm) were noted as 37.179 ± 0.32 and 71.43 mg/g, respectively. The adsorption process was well described by pseudo-second-order model. The reaction is endothermic, is spontaneous, and increases in randomness. The functional groups present on matrix, i.e., –OH, –C–N, –C–O,–CO–NH, –NH2, –SH, and –C–OH, were intensely involved in the process. Scanning electron microscopy results revealed the morphological changes due to adsorption of Pb(II) on and inside of PVA–CA–J. rubens matrix.
