Biochemical effects of sub lethal doses LC10 and LC20 of cypermethrin were studied on some enzymes and macromolecule activities of adult beetles of Tribolium castaneum (Herbst.). Cypermethrin caused disturbances in levels of all biochemical components under study. The dose of 0.78 ppm caused abnormalities in α‐amylase and FAA by increasing their activities i.e., 45.45% and 21.97% significantly. The higher sub lethal dose of 2.62 ppm disturbed all the parameters (AcP, α‐amylase, soluble protein and FAA) except AkP, which was decreased by 93.06%. Moreover, sub lethal doses either increased or decreased the levels of all parameters non‐significantly except AkP and FAA which were effected significantly by 87.92% and 14.29% at lower and higher doses, respectively. In the present studies, cypermethrin significantly enhanced the activity of AkP in both susceptible and resistant strains of T. castaneum adult beetles while FAA contents were increased significantly in resistant strain only. The activity of α‐amylase was significantly lowered in susceptible strain only.  相似文献

In Tribolium castaneum adults, sublethal doses of 1 and 2 ppm permethrin and 300 ppm malathion led to significant changes in amylase, trehalase, acid phosphatase, alkaline phosphatase, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, and isocitrate dehydrogenase activities. Malathion at 150 ppm did not affect phosphatases and lactate dehydrogenase. Both malathion and permethrin significantly increased cholinesterase activity. Mixing of the two insecticides resulted in antagonistic action with reference to various enzymatic activities. Glucose and glycogen contents were at first mobilized for energy supply under insecticidal stress conditions followed by lipid and cholesterol. Soluble protein, total protein, free amino acids, and urea contents remained unaltered under all experimental conditions.  相似文献

The effect of exposure to sub-lethal concentrations of cypermethrin, a synthetic pyrethroid pesticide, on biochemical parameters of muscle, blood and enzyme activities in brain, liver and kidney of the Indian major carp, Labeo rohita was studied. The sub-lethal exposure studies were done for up to 45 days at 1/10 and 1/50 of 96 h LC(50) of cypermethrin. The 96 h LC(50) was found to be 0.139 ppm. RNA levels decreased while DNA levels were elevated. Acid phosphatase was unchanged while alkaline phosphatase was depleted. Brain acetylcholinesterase activity was decreased significantly (P<0.05) over a period of 45 days at both cypermethrin concentrations. Lactate dehydrogenase activity in brain and liver was elevated, but inhibited in kidney. Succinate dehydrogenase and ATPase activities were depleted in brain, kidney and liver. There was a decrease in serum protein level over control at both concentrations of the pyrethroid. Blood glucose level and total leucocytes were elevated compared with controls at either concentration from day 15 to day 45. Haemoglobin percentage and total erythrocytes decreased in both sub-lethal concentrations. Extracts of the herb Datura stramonium were effective in countering the toxicity of this pesticide. Our data suggest that sub-lethal exposure of cypermethrin alters the biochemical, haematological parameters and enzymes of organs tissue and exert stress on the fish. Plant extracts may be useful in counteracting some of these effects.  相似文献

BackgroundThe present work aimed to detect the toxicological effects of green synthesized silver nanoparticles (Ag-NPs) by using Moringa Oleifera leaves extract on hematological and biochemical parameters of Oreochromis niloticus.MethodsAdult fish were exposed to two sublethal concentrations (1.95 and 3.9 ppm) of Ag-NPs against sodium selenite (0.1 ppm) and biosynthesized selenium nanoparticles (Se-NPs); 0.1 ppm; protection role for 2 and 4 weeks. Hematological parameters; erythrocyte count (RBCs), hemoglobin content (Hb), haematocrit value (Hct), mean corpuscular volume (MCV), Mean Corpuscular Hemoglobin Concentration (MCHC), leucocytes (WBCs), with differential count Micronucleus (MN) and alerted cells and biochemical parameters; aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP) enzyme activities, serum protein (total protein, albumin and globulin) concentration, urea, creatinine, glucose, cholesterol (Cho) and triglyceride (Tg) were detected.ResultsThe present investigation showed that Ag-NPs in different doses led to a significant reduction (p < 0.05) in RBCs, Hb, Hct, MCV, WBCs, LYM and serum proteins concentration. However, MCHC, MN, alerted cells, NEUT, AST, ALT, ALP enzyme activities, urea, creatinine, glucose, Cho and Tg showed a significant increases (p < 0.05) when compared with control group. Sodium selenite (Se) and biosynthesized selenium nanoparticles (Se-NPs) play an optimistic role in detoxification of Ag-NPs toxicity.ConclusionThe results suggest the negative impact of Ag-NPs on hematology and biochemical parameters of fish. Moreover, Se-NPs showed a full improvement of hematological and biochemical parameters more than that of sodium selenite in elimination of Ag-NPs toxicity.  相似文献

Toxicological data on silkworm Bombyx mori are quite comparable to those of other lepidopteran pest insects, therefore, it is considered as a suitable model for exploring effects of any new synthetic formulations. In this study, female V instar larvae of silk moth B. mori were chosen to evaluate the lethal and sublethal toxicity effects of RH-2485 (methoxyfenozide), a non-steroidal ecdysteroid agonist and to substantiate the ecdysteroid mimicking action of RH-2485 on ovary development, vitellogenin incorporation and egg production in isolated pupal abdomen (IPA). Probit analysis was carried out to find the median lethal dose (LD50) from 96 h cumulative mortality percent. Protein profile of haemolymph, fat body, ovary and eggs were separated in SDS-PAGE. Western blot analysis was carried out to confirm vitellogenin in the ovary. Sublethal effects on feeding, cocoon spinning, pupation, adult emergence and egg production were studied at doses of 1/5th, 1/10th and 1/20th of LD50. Significant changes were observed in all these parameters at all three sublethal doses. The morphological effects were related to underlying biochemical changes by finding the changes in haemolymph, fat body, ovary and egg protein profile. Marked changes were observed in storage proteins (80 kDa) and 30 kDa proteins in the haemolymph at all three sublethal doses. The larvae that escaped the sublethal effects at a dose of 1/20 of LD50 and emerged as adults with malformed wings produced significantly lower number of eggs. The isolated pupal abdomen (IPA) treated with RH-2485 did not metamorphose into adult but the oocyte development and vitellogenesis were normal but the egg precursor processing was incomplete leading to failure in choriogenesis.  相似文献

Cypermethrin is a highly active type II pyrethroid that is widely used to control harmful insects. The present study was conducted to investigate the possible effects of the sublethal concentration of cypermethrin (0.05 μL/L) on the biometric, hematological, and biochemical parameters of Van fish (Alburnus tarichi Guldenstadt 1814) for 24, 48, 72, and 96 h. Behavioral and physiological changes were observed in the fish treated with cypermethrin. Hematological parameters revealed a significant decrease in the erythrocyte (RBC), hemoglobin (Hb), and hematocrit (Hct) values (p ≤ .05). Serum enzymes, aspartate aminotransferase (AST, EC 2.6.1.1), alanine aminotransferase (ALT, EC 2.6.1.2), and lactate dehydrogenase (LDH, EC 1.1.1.27) activities and serum cortisol level were significantly increased (p ≤ .05). The results showed that changes in the biometric, biochemical, and hematological values of the Van fish exposed to cypermethrin can be used as biomarkers for monitoring toxicity.  相似文献

Arsenic compounds are classified as toxicants and human carcinogens. Environmental exposure to arsenic imposes a big health issue worldwide. Sinapic acid is a phenylpropanoid compound and is found in various herbal materials and high-bran cereals. It has been reported that sinapic acid has antioxidant efficacy as metal chelators due to the orientation of functional groups. However, it has not yet been examined in experimental animals. In light of this fact, the purpose of this study was to characterize the protective role of sinapic acid against arsenic induced toxicity in rats. Rats were orally treated with arsenic alone (5 mg/kg body weight (bw)/day) plus sinapic acid at different doses (10, 20 and 40 mg/kg bw/day) for 30 days. Hepatotoxicity was measured by the increased activities of serum hepatospecific enzymes namely aspartate transaminase, alanine transaminase, alkaline phosphatase, gamma glutamyl transferase, lactate dehydrogenase and total bilirubin along with increased elevation of lipid peroxidative markers, thiobarbituric acid reactive substances, lipid hydroperoxides, protein carbonyl content and conjugated dienes. The toxic effect of arsenic was also indicated by significantly decreased activities of enzymatic antioxidants like superoxide dismutase, catalase, glutathione peroxidase, glutathione-S-transferase, glutathione reductase and glucose-6-phosphate dehydrogenase along with non-enzymatic antioxidant like reduced glutathione. Administration of sinapic acid exhibited significant reversal of arsenic induced toxicity in hepatic tissue. The effect at a dose of 40 mg/kg bw/day was more pronounced than the other two doses (10 and 20 mg/kg bw/day). All these changes were supported by reduction of arsenic concentration and histopathological observations of the liver. These results suggest that sinapic acid has a protective effect over arsenic induced toxicity in rat.  相似文献

The effects of sublethal treatments of malathion and malathion + permethrin combinations on activities of acetylcholine esterase (ChE), carbohydrate-metabolizing enzymes (amylase; lactate dehydrogenase, LDH), protein-metabolizing enzymes (alanine and aspartate aminotransferase, ALAT, ASAT), as well as acid and basic phosphatases (AcP and AkP, respectively), and Kreb's cycle enzymes (isocitrate dehydrogenase, ICDH) were studied on sixth instar larvae of the red flour beetle, Tribolium castaneum. In addition, the levels of lipid, cholesterol, glucose, glycogen, proteins, free amino acids (FAA), urea, and nucleic acids (DNA and RNA) were determined. Malathion (400 ppm) increased the activity of LDH (53%), as well as the concentrations of FAA (31%) and urea (39%). Malathion treatments of 400 ppm decreased the glucose (20%) and glycogen (24%) content but did not affect other enzymes and biochemical components. Permethrin (200 ppm) and malathion (20 ppm) mixtures increased the activities of ChE (708%) and LDH (55%), and raised the concentrations of FAA (26%), urea (24%), glucose (23%), lipid (14%), cholesterol (21%), DNA (24%), and RNA (8%). The decrease in AcP activity and in glycogen concentration observed with malathion was sustained by permethrin in the mixture. Permethrin + malathion mixtures also depressed the levels of AkP (30%), ICDH (24%), and glycogen (36%). The intensity of effects commensurated with the dose and duration of insecticide exposure. Refeeding showed tendencies towards normalization of various biochemical parameters.  相似文献

Effect of praseodymium nitrate on hepatocytes and Kupffer cells in the rat.   总被引:4,自引:0,他引:4  
Intravenous administration of the rare earth metal salt, praseodymium nitrate, induced hepatic damage in the rat, as assessed by morphologic examination (light and electron microscopy) and biochemical parameters (serum glutamic-pyruvic transaminase (EC 2.6.1.2) and glutamic-oxalacetic transaminase (EC 2.6.1.1) activity as well as hepatic triglyceride content). Praseodymium hepatotoxicity was only attained with lower doses (10, 20, or 40 mg/kg), whereas a larger dose (80 mg/kg) was inactive in this respect. As detected by electron microscopy, lower doses of the metal salt caused hepatocytic alterations consisting of degranulation and dilatation of rough endoplasmic reticulum, accumulation of smooth endoplasmic reticulum as well as numerous lipid droplets. No abnormalities were detected in the cell organelles following administration of a large dose of the metal salt; however, vacuoles containing markedly electron-dense material were seen in the cytoplasm of the hepatocytes and the sinusoidal Kupffer cells.  相似文献

Biochemical differences between crop tissue (CT) and crop milk (CM) of pigeons were evaluated in terms of activity of certain enzymes, content of protein and nucleic acids and mitogenisity in vitro. Whereas the activities of aspartate transaminase, alanine transaminase, maltase, trehalase and cellobiase were significantly higher in CT, those of alkaline phosphatase, leucine aminopeptidase, gamma glutamyl transpeptidase, acid phosphatase and lactase were greater in CM. The CM also showed significantly higher levels of protein, DNA and RNA than CT. In contrast, the in vitro mitogenic effect of CT was greater than that of CM, and this property was enhanced during the breeding period. The biochemical composition of crop milk did not differ significantly in the first 4 days of secretion. It appears that certain factors responsible for growth stimulation accumulate in the crop tissue of pigeons during incubation.  相似文献

The levels of lactate dehydrogenase, hydroxy butyric dehydrogenase, glutamic oxalacetic transaminase, glutamic pyruvic transaminase, glutamate dehydrogenase, alkaline phosphatase and leucine amino peptidase were determined in the plasma of rainbow trout. The protein concentration and the amount of alkaline phosphatase were reduced in starving trout. Fed trout showed reduced lactate dehydrogenase activity. There was a significant correlation between the condition factor, most of the enzyme activities and the protein concentration. At 10 parts per thousand salinity the activities of alkaline phosphatase and leucine amino peptidase were significantly elevated, while lactate dehydrogenase activity was significantly decreased.  相似文献

测定了氯氰菊酯、啶虫脒和椰甲清3种杀虫剂亚致死剂量对椰甲截脉姬小蜂Asecodes hispinarumBoucˇek寄生椰心叶甲Brontispa logissima(Gestro)功能反应的影响。结果表明:亚致死剂量杀虫剂没有改变椰甲截脉姬小蜂功能反应模型的基本结构,但影响到了模型的各项参数。药剂处理后椰甲截脉姬小蜂的最大日捕食量降低,处理猎物时间延长,搜寻效应减弱。氯氰菊酯和啶虫脒的处理后瞬时攻击率减小,但椰甲清处理后瞬时攻击率高于对照。可见,在亚致死剂量下,氯氰菊酯和啶虫脒对椰甲截脉姬小蜂的寄生作用存在不良影响,椰甲清(30%杀虫单+15%啶虫脒)可提高其瞬时攻击率但减弱其搜索效应。  相似文献

Vicolides A,B, C and D, the sesquiterpene lactones isolated from V. indica exhibited antiinflammatory activity against cotton pellet granuloma in rats at dose level of 10 mg/kg body weight, sc. Highly significant activity was observed with vicolides C and D. They reduced the protein content, acid and alkaline phosphatase, glutamate-pyruvate transaminase and glutamate oxaloacetate transaminase activities in liver and serum. Significant reduction in ascorbic acid content in adrenals was also observed in treated animals. The highly significant antiinflammatory activity of vicolides C and D can be attributed to their chemical structures. Vicolide D has an epoxy angeloyl group while vicolide C has 3,4 epoxy group and an ester moiety in the molecule. Vicolide D possesses antipyretic activity at 250 mg/kg body weight, po dose. It may be due to the presence of epoxy angeloyl group in the molecule.  相似文献

Studies on the effect of Catharanthus roseus alkaloids on the biology of Euproctis fraterna showed that at a dose of 10 ppm, only 19.25% fertile eggs were laid. The hatchability was totally suppressed at higher doses. There was a considerable reduction in total carbohydrate and protein content in treated larvae. Although acetylcholinesterase (AChE) is the primary target of organophosphorus insecticides (OPs), increasing evidence regarding their secondary effects suggests that OPs disturb homeostasis of insects by generating free radical intermediates that trigger lipid peroxidation. We therefore investigated alterations in lipid peroxidation product, malondialdehyde (MDA) content, and alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities, in conjunction with AChE activity as biochemical stress indicators in greater wax moth, Galleria mellonella (L.) larvae for OPs methyl parathion (MP) and ethyl parathion (EP). The effects of MP and EP were first investigated by rearing the young larvae on an artificial diet containing 0.01, 0.1, 1, 10, and 100 ppm of each insecticide. Second, the mature larvae were injected with 0.05, 0.5, 5, 50, and 500 ng of insecticides for determining the changes in biochemical stress responses. The diet with lowest level of MP significantly decreased the activities of all measured enzymes, whereas it increased MDA content. However ALT and AST were significantly higher in the larvae reared with the diet with high levels of MP than in control larvae. All tested levels of MP resulted in a decrease in AChE activity. The lowest level of EP in diet (0.01 ppm) significantly increased ALT activity, whereas it reduced that of AChE. This insecticide at 0.1 ppm resulted in reduced AST activity, but 1 ppm in diet elevated AST activity and MDA content. EP at 0.1 ppm and higher levels in the diet reduced ALT activity. All dietary EP levels significantly decreased AChE activity. ALT, AST, and AChE were lower in larvae fed with the diet containing 100 ppm ethyl parathion compared with larvae on control diet. MP at 50 ng per larva increased ALT and AST activities from 35.42 +/- 0.74 and 26.34 +/- 0.83 to 203.57 +/- 1.09, and 122.90 +/- 1.21 U/g, respectively, when the mature larvae were injected. All injected doses of EP dramatically reduced both ALT and AST activities, but only the lowest and highest levels of this insecticide decreased AChE activity. The lowest level of this insecticide also significantly increased MDA content in larvae. High levels of both insecticides increased MDA content. We observed a significant higher increase in MDA content in the larvae reared with 10 ppm EP (102.16 +/- 1.57 nmol/g protein) than the control group (30.28 +/- 1.42 nmol/g protein). These results suggest that OPs caused the metabolic and synaptic dysfunctions in greater wax moth and alter its biochemical physiology in response to oxidative stress.  相似文献

Forty newly hatched unsexed Japanese quail (Coturnix coturnix japonica) chicks were fed diets containing 3 ppm aflatoxin (AF) and 4 ppm T2 toxin either singly and in combination for 35 days. Sera samples were collected from six birds in each group at the end of the trial to study the effect of certain serum biochemical parameters. Highly significant (P < 0.01) differences were observed for serum total protein, albumin, globulin, glucose, cholesterol, aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP) and gamma glutamyl transferase (GGT) between control and toxin treated groups. Toxin treated groups did not reveal any significant difference for serum urea nitrogen, creatinine, uric acid, calcium, phosphorus, sodium and potassium. There was a reduction in the levels of serum total protein, albumin, globulin, glucose, cholesterol, ALT and elevation of AST and GGT and variable ALP levels observed in toxin treated groups. Effect of endotoxin on the immunity of Indian major carp, Labeo rohita   总被引:1,自引:0,他引:1  
Endotoxin, a lipopolysaccharide component of outer cell wall membrane of the Gram-negative bacteria is a factor responsible for a number of biological effects including immunostimulatory activities in different animal species including fish. In this study, L. rohita yearlings of weight ranging from 80 to 100g were injected intraperitoneally with 0.5, 1, 2, 5, 10 and 20 EU/fish dose of endotoxin to find out its effect on the immunity. The L. rohita yearlings were found to resist the endotoxin dose up to 20 EU/fish and at the lower doses, i.e., at 1 and 2 EU/fish; it acted as an immune potentiator. Different serum and immune parameters like protein, globulin, lysozyme, respiratory burst activity, myeloperoxidase activity, natural agglutination titre were found to be significantly high (p<0.01) at a dose of 1 EU/fish. While at 10 and 20 EU/fish, most of these parameters were lower thereby indicating the immuno-suppressive nature of the endotoxin at these higher doses.  相似文献

An increase in global food demand has resulted in a significant increase in the use of pesticides in agriculture. Synthetic pyrethroid pesticides account for over 30% of the global pesticide use; Pyrethroid pesticides were used preferably over organochlorines and organophosphates due to their high effectiveness, low toxicity to non-target organisms and easy biodegrability. It has widespread applications in agriculture through the world and as well in Algeria. Cypermethrin is one of the most insecticidal pyrethroids widely used in agriculture regions of Setif. to control wide range of insect pests in a variety of crops. The aim of this study is to investigate the effects of cypermethrin (Cyper-Ac 271 g/l from the active substance of the cypermethrin) on hematological, biochemical parameters, body weight loss, and histopathological study of some organs. Male mice weighing 30-40g were used, separated in 5 groups, n=6, two groups controls given vehicle (oil vegetable) and three experimental groups (Cypermetherin and vegetable oil). The animals were gavaged by 1/5 LD50 (LD50 = 485 mg/kg b/w) for 2 and 4 weeks respectively, and with 1/20 LD50 for 12 weeks, then the animals sacrificed at the end of the experiment.. Blood was collected. Enzyme activities were assayed in the plasma samples obtained. Glutamate oxaloacetate transaminase (GOT), Glutamate pyruvate transaminase (GPT), Alkaline phosphatase (ALPH) and Glucose. Red blood cells, (RBC), and white blood cells (WBC) were calculated too. The samples of liver and kidney were processed for histology. The results indicated a significant increase in transaminases GOT, GPT, and AlP. The decrease in Hb, RBC and WBC which are related to the immunity, this is probably due to cell lyses explain the effect of Cypermetherin on erythropoeisis. cypermethrin treatment exhibited severe histopathological changes, especially in the liver and kideney accompanied by weight loss of some organs. We conclude that cypermethrin induces oxidative stress and modifies biochemical parameters and histological aspects of liver and kidney.  相似文献

Three hundred and forty-eight Vencob broiler chickens were fed diets containingPenicillium griseofulvum rice culture material with 0, 12.5, 25 and 50 ppm of the mycotoxin cyclopiazonic acid (CPA) for 28 days. Serum samples were collected from 9 birds in each group at weekly intervals to study the effect of sublethal doses of CPA on certain serum biochemical parameters. Significant reductions in weight gains (p<0.01) and feed consumptions (p<0.05) were observed at 25 and 50 ppm. Exocrine pancreas showed degenerative and necrotic changes in CPA fed chickens. The CPA had significant (p<0.05) influence on serum total protein, albumin, cholesterol, amylase and lipase levels. CPA did not affect serum glucose levels. There was a decline in levels of total serum protein and albumin in CPA fed groups. A cypermethrin-mixed diet was fed uninterrupted to male albino rats for six months to evaluate toxicity in nontarget organisms. The rats consumed cypermethrin at a dose of 420 mg active ingredient (AI) per kilogram body weight per day. At the end of the stipulated period, the blood and liver were analyzed for insecticidal toxicity. The hemoglobin content and white blood cell (WBC) count remained unaltered, while the red blood cell (RBC) count and packed-cell volume (PCV) decreased significantly. The blood serum lactate dehydrogenase (LDH), isocitrate dehydrogenase (ICDH), and amylase activities were elevated 61%, 30%, and 46%, respectively, after six months of insecticide feeding, suggesting liver and possibly pancreas malfunction. The glutamate oxaloacetate transaminase (GOT) and creatine phosphokinase (CPK) activities, on the other hand, decreased 37% and 40%, respectively. The blood serum protein and free amino acids (FAA) content increased 12% and 31%, respectively, while cholesterol content decreased 49%. Consequent to cypermethrin administration the hepatic GOT, LDH, and ICDH activities increased 250%, 20%, and 30%, respectively. The soluble proteins, FAA, and glucose contents exhibited significant increases of 28%, 61%, and 71%, respectively. Histological changes were marked by hypertrophied hepatic cells and nuclei.  相似文献
