A technique has been developed for artificially inseminating freshly molted female lobsters (Homarus). Using the criterion that eggs were fertilized if they showed development of normal cleavage patterns, at least 49.5% of the inseminated females extruded fertilized eggs. Sixty-two percent of the fertilized eggs developed to the eyespot stage, and 26.6% of these subsequently hatched. Eggs were not fertilized in 7.7% of the extrusions. In 42% of the samples, it was not possible to establish whether fertilization had occurred because either samples were collected prior to initiation of cleavage or eggs were too poorly preserved to assess cleavage with confidence. It is probable that many of the eggs that were evaluated as equivocal were also fertilized. This technique should be valuable in the experimental and commercial aquaculture of lobsters and might be directly applicable to other crustaceans.  相似文献

Despite being one of the most productive fisheries in the Northwest Atlantic, much remains unknown about the natural reproductive dynamics of American lobsters. Recent work in exploited crustacean populations (crabs and lobsters) suggests that there are circumstances where mature females are unable to achieve their full reproductive potential due to sperm limitation. To examine this possibility in different regions of the American lobster fishery, a reliable and noninvasive method was developed for sampling large numbers of female lobsters at sea. This method involves inserting a blunt-tipped needle into the female''s seminal receptacle to determine the presence or absence of a sperm plug and to withdraw a sample that can be examined for the presence of sperm. A series of control studies were conducted at the dock and in the laboratory to test the reliability of this technique. These efforts entailed sampling 294 female lobsters to confirm that the presence of a sperm plug was a reliable indicator of sperm within the receptacle and thus, mating. This paper details the methodology and the results obtained from a subset of the total females sampled. Of the 230 female lobsters sampled from George''s Bank and Cape Ann, MA (size range = 71-145 mm in carapace length), 90.3% were positive for sperm. Potential explanations for the absence of sperm in some females include: immaturity (lack of physiological maturity), breakdown of the sperm plug after being used to fertilize a clutch of eggs, and lack of mating activity. The surveys indicate that this technique for examining the mating success of female lobsters is a reliable proxy that can be used in the field to document reproductive activity in natural populations.  相似文献

The American lobster (Homarus americanus), a commercially important benthic marine crustacean, is widely distributed along the continental shelf of the western North Atlantic. The population substructure of this species remains poorly understood despite its economic value. Informative markers are required to clarify relationships between local populations. To this end, we developed eight polymorphic short sequence repeats (SSR) for the American lobster, which were derived from expressed sequence tags. Additionally, we tested four SSRs previously identified for the Norway lobster (Nephrops norvegicus L.) for cross‐species utility; only one of these showed polymorphism.  相似文献

Spermatozoa and stalked spermatophores in Baltic amber represent the first report of these reproductive structures in the fossil record. A comparison of this material with sperm and simple stalked spermatophores from extant invertebrates indicates that the fossil structures originated from a springtail (Collembola: Hexapoda). This conclusion is supported by the presence of a female springtail, Sminthurus longicornis Kock and Berendt (Sminthuridae: Symphypleona), in the same piece of amber. This specimen, which appears to have been molting at the time of death, is adjacent to objects interpreted as springtail eggs. The present find establishes the existence of the indirect method of sperm transfer by the late Eocene (40 Ma).  相似文献

Historical developments of the Republic of Ireland's Homarus gammarus fishery are reviewed and compared in the context of landings in Europe and the North American H. americanus fishery. H. gammarus fishing has provided essential income for over a century to many Irish coastal communities. There have been significant fluctuations (greater than 460%) in recorded landings over this time and the relative importance of H. gammarus fishing has changed with the establishment of ancillary crustacean fisheries. In 1994, H. gammarus landings reached an all-time high of 714 tonnes, with a landed value of IR £6.3 million; in 1999 landings were 597 tonnes valued at IR £6.15 million. Current Republic of Ireland lobster fishery regulations are: (a) minimum carapace length of 85 mm, (b) ban on landing lobsters that have been `V-notched' or have a mutilated tail fan, and (c) prohibition of capturing lobsters by SCUBA diving. Chronological details on recent legislation, regulations and landings relating to Ireland's lobster fishery sector are reviewed. In the last 40 years landings of H. gammarus in the Republic of Ireland exhibit signs of an overexploited stock characterised by a reduction in catch per unit effort and there has been a general decline of total landings for H. gammarus throughout Europe. This contrasts with North America, where many regulations govern the H. americanus fishery and landings appear to be in a healthier state.  相似文献

We have examined the formation of the fertilization envelope in the lobsters Homarus americanus and H gammarus. Oocytes were fixed for electron microscopy either in the ovary or following extrusion from the gonopore. Mature ovarian oocytes are surrounded by a coat (envelope 1), which is comprised of small electron-dense granules and structures resembling “bottlebrushes.” At least part of this coat is synthesized by the follicle cells of the ovary. The cortex of ovarian oocytes contains four types of vesicles that we refer to as high-density vesicles (HDV), low-density vesicles (LDV), moderately dense vesicles (MDV), and ring vesicles (RV). Oocytes that were electrically extruded from the gonopore and fixed immediately had an envelope identical to that of ovarian oocytes. The cortex of gonopore oocytes contained the four types of vesicles found in ovarian oocytes. When unfertilized gonopore oocytes were allowed to incubate in sea water, the oocyte cortex appeared unaltered, but envelope 1 swelled and the bottlebrushes dispersed. When recently fertilized oocytes were fixed during natural spawning or following in-vitro fertilization, each type of vesicle was released in sequence from the cortex of the oocyte. The contents of the HDV and LDV appeared first in the perivitelline space, but their fate could not be determined at later times. The ring-shaped elements of the RV and the moderately electron-dense material of the MDV were released exocytotically somewhat later; these materials coalesced in the perivitelline space to form a new coat (envelope 2). Envelope 1 subsequently condensed to its original thickness and appeared firmly attached to envelope 2. Our results show that the fertilized lobster egg is surrounded by two discrete coats. The outer coat, which is formed in the ovary, undergoes a swelling/condensation cycle at spawning. The inner coat originates from a complex cortical reaction. Together these coats comprise the fertilization envelope of the lobster egg.  相似文献

A north/south discontinuity along the northeastern coast of North America in the genetic structure of the American lobster ( Homarus americanus ) was detected using a suite of 13 microsatellite loci assessed using spatial analyses. Population genetic data laid over existing data on physiographic changes and sea-surface temperatures were used to reconstruct the Pleistocene distribution of this species. A postglacial northern-edge colonization model best explains the relative genetic homogeneity of the northern region compared to the southern region centred in the Gulf of Maine. Population genetic analyses identified significant structure (range of standardized theta 0–0.02) but no significant evidence for isolation by distance. The novel application of spatial genetic analyses to a marine species allowed us to interpret these results by providing a greater insight into the evolutionary factors responsible for shaping the genetic structure of this species throughout is natural range.  相似文献

Juvenile lobsters can be reared in hatcheries. Their use in ranching programmes has been demonstrated successfully and exploitation of the idea by fisheries is commencing. Studies are underway to investigate the use of these juveniles in restoring damaged or threatened populations. The European green crab (Carcinus maenas) is a newly invasive species in Newfoundland, where it has likely been present for ≤15 years. The green crab has been found in stomach contents of American lobster (Homarus americanus) in New England and Nova Scotia, Canada, but predation on this species has not yet been quantified in Newfoundland. We conducted feeding experiments to determine whether lobsters from Newfoundland were as likely as those from Nova Scotia (which have coexisted with green crabs for >60 years) to recognize and prey upon this new species. We also performed experiments to determine whether green crabs reach a size refuge from predation and whether factors including starvation, availability of alternate food sources, or habitat complexity would influence the probability of lobster attacking or feeding on green crabs. In our trials, lobster origin had no significant effect on crab predation; lobsters, irrespective of origin, were more likely to consume small (<40 mm carapace width [CW]) and medium (40–65 mm CW) crabs than larger (>65 mm CW) ones. Nevertheless, even small lobsters (73–76 mm carapace length, 300 g) were able to kill and consume the largest green crabs (78 mm CW, 100 g). Green crabs were less likely to be attacked or eaten when an alternative food source was present, suggesting that the lobsters were preying on the crabs, rather than simply killing them in a dispute over territory. The addition of a shelter provided a refuge for the green crabs; however, the crabs were only able to avoid being injured or eaten if this shelter was structurally complex. The green crab is slowly spreading westward around the island of Newfoundland, and so its long‐term effects, interactions with other organisms, and contribution to the diet of Newfoundland lobsters remain to be seen.  相似文献

The objectives of this study were to determine the effect of cryoprotectants on sperm viability and develop a freezing protocol for long-term storage of P. monodon spermatophores. Spermatophores suspended for 30 min in calcium-free saline (Ca-F saline) containing the cryoprotectants dimethyl sulfoxide (DMSO), ethylene glycol (EG), 1,2-propylene glycol (PG), formamide, and methanol at concentrations of 5, 10, 15, or 20% were studied using a modified eosin-nigrosin staining technique. The smallest reductions in apparent sperm viability occurred with DMSO; therefore, a freezing protocol was developed using Ca-F saline containing 5% DMSO. Spermatophores were cryopreserved using three protocols; cooling to a final temperature of -30, -80 or -80 degrees C and immediately stored in liquid nitrogen (cooling rates of -2, -4, -6, -8, -10, -12, -14 or -16 degrees C/min). Frozen spermatophores were thawed (2 min) at 30, 60, 70, or 90 degrees C. Successful cryopreservation of spermatophores in liquid nitrogen was achieved by a one-step cooling rate of -2 degrees C/min between 25 and -80 degrees C before storing in liquid nitrogen. Optimal thawing was in a 30 degrees C water bath for 2 min; this yielded live sperm after storage in liquid nitrogen for 210 days. Average sperm viability for fresh (97.8+/-2.9%) and cryopreserved spermatophores held for less than 60 days (87.3+/-4.1%) did not differ (P>0.05); however, that for spermatophores stored in liquid nitrogen between 90 and 210 days were lower (P<0.05) and varied from 27.3+/-3.4 to 53.3+/-4.3%. Thawed spermatophores previously held in liquid nitrogen for less than 62 days fertilized eggs (fertilization and hatching rates of 71.6-72.2% and 63.6-64.1%, respectively) at rates comparable to fresh spermatophores (70.8-78.2% and 66.3-67.8%, respectively). In conclusion, sperm within cryopreserved spermatophores stored in liquid nitrogen retained their viability for up to 210 days.  相似文献

The mean number of sperm ejaculated into the spermatophore by the male black fly, Simulium decorum Walker, was 4048, as determined from 43 counts. The completeness and rate of sperm transfer were estimated by counts from the spermatophore and female spermatheca at selected times after copulation ended. The beginning of transfer occurred at the earliest about 4 min after copulation, but could be delayed until 30 min. Transfer rate was interpreted as constant at about 73 sperm/min and was usually completed by about 55 min after copulation. All spermatozoa appeared to be moved to the single spermatheca.  相似文献

Lysosomes are known centers for sequestration of calcium and a variety of heavy metals in many invertebrate tissues, and as a result of this compartmentalization these organelles perform important detoxification roles in the animals involved. The present investigation uses a centrifugation method to isolate and purify hepatopancreatic lysosomes from the American lobster, Homarus americanus. Purified lysosomal preparations were used to characterize membrane transport mechanisms in these organelles for transferring and sequestering cytoplasmic copper following its absorption across the plasma membrane from dietary constituents. The copper-specific fluorescent dye, Phen Green, was employed to quantify transmembrane fluxes of this metal as has been recently used to investigate copper movements across hepatopancreatic mitochondrial and plasma membranes. Results indicated the presence of a vanadate-sensitive, calcium-stimulated, copper ATPase in the membranes of these organelles that displayed high affinity carrier-mediated transport kinetics and may significantly contribute to organismic copper homeostasis. Together with a putative bafilomycin-sensitive V-ATPase in the membrane of the same organelles, importing hydrogen ions into the organellar interior, this copper ATPase may function as part of a physiological mechanism for precipitate formation between metallic cations and anions. These ionic precipitate complexes may then act as a sink for excess metals and thereby reduce the circulating concentrations of these elements.  相似文献

Lobster olfactory sensory neurons have contributed to a number of advances in our understanding of olfactory physiology. To facilitate further study of their function, we have developed conditions allowing primary culture of the olfactory sensory neurons in a defined medium. The most common cells in the culture were round cell bodies with diameters of 10-15 micro m that often extended fine processes, features resembling olfactory sensory neurons. We discovered that acetylcholinesterase acted as a growth factor for these cells, improving their survival in culture. We also confirmed previous evidence from spiny lobsters that poly-D-lysine was a superior substrate for olfactory cells of this size and morphology. We then identified olfactory sensory neurons in the culture in two ways. Almost half the cells tested responded to application of a complex odorant with an inward current. An even more rigorous test was made possible by the development of an antiserum to OET-07, an ionotropic glutamate receptor homolog specifically expressed by Homarus americanus olfactory sensory neurons. It labeled a majority of the round cells in the culture, unequivocally identifying them as olfactory sensory neurons.  相似文献

Variation in the proportions of offspring fathered by a second male to mate (the P2 value) has been studied in two species of grasshoppers, Chorthippus parallelus (Zetterstedt) and Ch. biguttulus (Linnaeus), by means of the sterile‐male technique. In both species the P2‐values of the first egg pods laid were in the range of 50–100%, and the temporal variation of P2‐values appeared to follow two modes. In one, the P2‐value steadily declined with time, in the other it remained constant at a high level. It is concluded that sperm is passively lost between two matings. The remaining sperm then either mixes within the spermatheca or is stratified. Further variability in sperm precedence can possibly be explained by spermatophore ejection or differential use of stored sperm.  相似文献

Unraveling the structure of squids’ spermatophores: a combined approach based on Doryteuthis plei (Blainville, 1823) (Cephalopoda: Loliginidae). —Acta Zoologica (Stockholm) 93 : 281–307. Male coleoid cephalopods produce elaborate spermatophores, which function autonomously outside the male body during copulation, undergoing a complicated process of evagination. In order to contribute to the understanding of this unique structure, this study investigated the morphology of the spermatophore of Doryteuthis plei applying several microscopy techniques. A hitherto unreported, much more complex structural arrangement was revealed for the loliginid spermatophore, the most striking findings being: (1) the complex, layered structure of the middle membrane, which bears an additional, chemically distinct segment surrounding part of the cement body; (2) the presence of a space between the inner tunic and middle membrane filled with a fine reticulated material; (3) the presence of stellate particles not only embedded in the spiral filament, but also closely applied to the inner membrane at the level of the cement body; (4) the presence of a pre‐oral chamber in the cap region; and (5) the complex organization of the cement body, formed by two distinct layers encompassing contents of different chemical and textural properties. Careful literature reassessment suggests several of these features are common to loliginids, and to some extent to other squids. Their possible functional implications are discussed in light of our knowledge of the spermatophoric reaction mechanics.  相似文献

Understanding movements of palinurid (spiny) lobsters is important to fisheries management. However, movement data obtained with mark-capture techniques are often influenced by artefacts (e.g., displacement, tag-loss, tag-induced injury). To avoid these problems, a novel approach was employed to investigate the movements of an important coral reef palinurid, the painted lobster (Panulirus versicolor). Eighty adult individuals were captured by hand near Northwest Island, Australia. Each lobster was tagged internally (with elastomer) and externally (with plastic zip-ties) to enable both short- (1–10 days) and long-term (6–24 months) observations. Shortly thereafter, all lobsters were manually returned to their shelters (dens). After 6–24 months, 44 lobsters were recaptured, giving an overall recapture rate of 55%. All of these individuals were recaptured within 500 m of their original den, thus indicating a high level of site-fidelity for this species. Short-term observations revealed that individuals moved frequently to and from nearby dens, and that male lobsters typically moved further than female lobsters. It is hypothesized that male lobsters move about their home range more actively, perhaps to seek mating opportunities or to defend resources. The significance of these observations is discussed with respect to the management of this important fishery resource.  相似文献

Homarus americanus has a life history that is similar to other arthropods, including a pelagic larval phase and a benthic adult phase. The larval phase is divided into three morphologically distinct stages, followed by metamorphosis to the post-larval phase. H. americanus larval development has been studied previously, although the molecular mechanisms that regulate the consequent changes are not fully elucidated. This study is the first to use an oligonucleotide microarray to investigate global gene expression during H. americanus larval development. Stage-specific gene expression profiles of larvae and postlarvae from two-year classes were assessed. We found the expression levels of 1851 genes to be significantly different among larval stages. Functional annotations indicated that various differentially expressed genes were involved with immune function, energy regulation, and development. Ten target genes of interest were selected for expression verification using RT-qPCR.
