ABSTRACT. The oocytes of 3-day-old unfed Aedes aegypti mosquitoes are in a state of oogenic arrest, but microgram doses of ecdysterone stimulate their accumulation of a variable amount of yolk. We now find that these doses also induce the deposition of plaques of vitelline envelope by the follicle cells, and with transmission electron microscopy we have compared their formation with that in normal blood-fed females. Plaques in the experimental animals were abnormally large and irregular in shape and distribution. In part, these abnormalities were attributable to the fact that the follicle cells remain in close contact with the oocyte, whereas the space between follicle cells and oocyte increase significantly in the blood-fed female. Deposition of the plaques occurred earliest after the injection of 5 μg ecdysterone, but even at this high dose the amount of plaque material deposited was less than in the blood-fed controls. Induction of the deposition of abnormal vitelline envelope in unfed females was most clearly demonstrated after two injections, 1 μg ecdysterone each, 14h apart; 24h after the second injection, the plaques had prematurely fused into a thin disorganized envelope. When females were injected with ecdysterone immediately after a blood-meal, vitelline envelope plaques formed prematurely, and their structure became increasingly abnormal with time. This early onset of activity was characteristic of follicle cells adjacent both to the oocyte and to nurse cells. Thus, the factors that normally control the formation and organization of the vitelline envelope are absent in the unfed female stimulated with high doses of ecdysterone, while in the blood-fed females, excessive ecdysterone apparently interferes with the timing and orderly sequence of envelope formation.  相似文献

Egg development neurosecretory hormone (EDNH), a hormone that stimulates vitellogenesis in mosquitoes, was purified 10,000-fold from the mosquito Aedes aegypti. The purification procedure included chromatography on hydrophobic, ion exchange, and gel filtration columns, and preparative electrophoresis to give an almost homogeneous preparation. The hormone is a polypeptide monomer of molecular weight of 18,700 ± 500 as determined from SDS electrophoresis and gel filtration chromatography. Using lowpressure chromatography throughout the purification procedure, the hormone was recovered at a high yield (39%). The amount of EDNH in a mosquito is about 0.6-1.6 ng, corresponding to 32–85 fmol. Injection of purified EDNH into female mosquitoes resulted in the conversion of [14C]cholesterol into labeled ecdysone and 20-hydroxyecdysone which were separated and identified using thin-layer chromatography and high-performance liquid chromatography separation procedures. Egg development and vitellogenin synthesis were also induced when EDNH was injected into several mosquito species, indicating that the hormone is not species-specific. This report is the first to show that a purified preparation of EDNH has both steroidogenic and a gonadotropic effects on female mosquitoes.  相似文献

SYNOPSIS Herpetomonas sp. was found repeatedly in the Malpighian tubules of laboratory-reared male and female Aedes aegypti and Aedes albopictus in Malaysia. The flagellates occurred irregularly, in batches, and were absent during long periods. The data suggest an exogenous source of infection for the mosquitoes, presumably from another insect, probably of another genus. Thirty to 40% of the flagellates of Aedes contained intracytoplasmic rod-shaped structures strongly resembling bacteria. These were found often in groups suggesting intracellular multiplication. They were passed to the Herpetomonas daughter cells during division. Parasitism of Aedes by Herpetomonas is extremely unusual, only one previous record, an inconclusive one, having been found. Parasitism by Herpetomonas containing bacterium-like rods has apparently never been reported.  相似文献

Aedes albopictus (Diptera: Culicidae) was first reported in Central Africa in 2000, together with the indigenous mosquito species Aedes aegypti (Diptera: Culicidae). Because Ae. albopictus can also transmit arboviruses, its introduction is a public health concern. We undertook a comparative study in three Cameroonian towns (Sahelian domain: Garoua; equatorial domain: Douala and Yaoundé) in order to document infestation by the two species and their ecological preferences. High and variable levels of pre‐imaginal Ae. aegypti and Ae. albopictus infestation were detected. Only Ae. aegypti was encountered in Garoua, whereas both species were found in Douala and Yaoundé, albeit with significant differences in their relative prevalence. Peridomestic water containers were the most strongly colonized and productive larval habitats for both species. No major differences in types of larval habitat were found, but Ae. albopictus preferentially bred in containers containing plant debris or surrounded by vegetation, whereas Ae. aegypti tended to breed in containers located in environments with a high density of buildings. These findings may have important implications for vector control strategies.  相似文献

We asked whether climate change might affect the geographic distributions of Aedes aegypti (L.) and Aedes albopictus (Skuse) (Diptera: Culicidae). We tested the effects of temperature, diet and the presence of congeneric species on the performance of immature stages of these two aedine species in the laboratory. Mosquitoes in three different species-density combinations were reared at four constant temperatures (20 °C, 25 °C, 30 °C, 35 °C) on low- or high-level diets. Of the four temperatures tested, mortality increased only at 35 °C in both species. Mortality was higher on the high-level diet than on the low-level diet at 35 °C, but not at other temperatures. The presence of congeneric species had a significant positive effect on mortality in Ae. albopictus, but not in Ae. aegypti. Both species developed more quickly at higher temperatures within the range of 20-30 °C; development was not enhanced at 35 °C. Population growth of Ae. albopictus was more stable, regardless of diet and temperature; that of Ae. aegypti varied more according to these two factors. These species-specific attributes may help to explain the latitudinal distribution of the mosquitoes and degree of species dominance where they are sympatric.  相似文献

In Port‐au‐Prince, Haiti, the status of insecticide resistance has not recently been evaluated for Aedes aegypti (L) and Aedes albopictus (Skuse) populations. No prophylactics exist for dengue, so prevention is only through vector control methods. An earthquake occurred in Haiti on January 12, 2010, with a magnitude of 7.0 Mw that devastated the area. Dengue became a major concern for the humanitarian relief workers that entered the country. Bottle bioassays were conducted in the field on adult mosquitoes reared from larvae collected from the grounds of the U.S. Embassy and from an adjacent neighborhood in eastern Port‐au‐Prince, Haiti. At the CDC, Fort Collins, CO, bioassays, molecular, and biochemical assays were performed on mosquitoes reared from field‐collected eggs. A small percentage of the population was able to survive the diagnostic dose in bioassays run in Haiti. Mosquitoes tested at the CDC demonstrated no phenotypic resistance. A variety of factors could be responsible for the discrepancies between the field and lab data, but temperature and larval nutrition are probably most important. Knowledge of localized resistance and underlying mechanisms helps in making rational decisions in selection of appropriate and effective insecticides in the event of a dengue outbreak.  相似文献

Thirty‐two Aedes aegypti populations collected throughout Thailand and five populations of Aedes albopictus from southern Thailand were subjected to standard WHO contact bioassays to assess susceptibility to three commonly used synthetic pyrethroids: permethrin, deltamethrin, and lambda‐cyhalothrin. A wide degree of physiological response to permethrin was detected in Ae. aegypti, ranging from 56.5% survival (Lampang, northern Thailand) to only 4% (Kalasin in northeastern and Phuket in southern Thailand). All 32 populations of Ae. aegypti were found to have evidence of incipient resistance (62.5%) or levels of survival deemed resistant (37.5%) to permethrin. Four populations of Ae. albopictus were found with incipient resistance (97 – 80% mortality) and one with resistance (< 80%) to permethrin. The majority of Ae. aegypti populations (68.7%) was susceptible (> 98% mortality) to deltamethrin, with incipient resistance (observed 97–82% mortality) in other localities. In contrast, all populations of Ae. aegypti were completely susceptible (100% mortality) to the recommended operational dosage of lambda‐cyhalothrin. All five populations of Ae. albopictus were found completely susceptible to both deltamethrin and lambda‐cyhalothrin. Evidence of defined incipient or resistance to synthetic pyrethroids mandates appropriate response and countermeasures to mitigate further development and spread of resistance. In light of these findings, we conclude that routine and comprehensive susceptibility monitoring of dengue mosquito vectors to synthetic pyrethroids should be a required component of resistance management policies and disease control activities.  相似文献

ABSTRACT. . Azadirachtin in blood fed to adult female Aedes aegypti through an artificial membrane does not cause feeding inhibition over a wide dose range (0–200 ng/female), and high doses of ingested azadirachtin fail to inhibit or delay oviposition. However, significant, transient retardation of oocyte growth is observed for up to 72 h after feeding. Immature oocytes are observed in 86% of azadirachtin-fed females decapitated 10 h after a blood meal, whereas 96% of decapitated control females contain maturing oocytes. This suggests that azadirachtin delays the release of one or more factors from the head that regulate oogenesis. We propose that adult females overcome the effect of azadirachtin by rapid metabolism rather than by excretion of the compound, since by 2 h after a blood meal, only 0.1% of ingested azadirachtin was recovered from excreta and 5% recovered from the body.  相似文献

The synthesis of juvenile hormone III (JH III) by the isolated corpora allata (CA) of Aedes aegypti adult female was studied using an in vitro radiochemical assay. We dissected the corpora allata-corpora cardiaca (CA-CC) complex attached to a piece of aorta. The complex was left connected to the intact head capsule to facilitate the visualization and transfer of the glands. A linear increase in the cumulative amount of biosynthesized JH III was found for at least the first 6 h of incubation; approximately 45% of the synthesized JH III was present in the medium. There was a dependence of JH III synthesis on exogenous methionine supply. Using reversed phase high performance liquid chromatography two major labeled products biosynthesized by the CA were separated. They co-migrated with JH III and methyl farnesoate (MF). The identity of the biosynthesized JH III was confirmed by gas chromatography-mass spectrometry. JH III synthesis was only 2.0 fmol/pair gland/h immediately after adult emergence, but increased to 32.6 fmol/ pair gland/h 18 h later in sugar-fed females. Two days after emergence, the CA biosynthetic activity slowly started to decrease, and reached values of around 5.3 fmol/pair gland/h by one week after emergence. Synthesis of JH was similar from either sugar-fed females mated or unmated. A blood meal resulted in a decrease of JH III synthesis in CA from mated females by 12 h after feeding and from virgin females by 24 h after feeding. JH III biosynthesis remained low for at least 96 h in mated females, but was back to higher levels 72 h after feeding in virgin females. Rates of JH III biosynthesis closely reflected the hemolymph levels of JH III both after emergence and after a blood meal described by Shapiro et al. (1986). The activity of the CA in Aedes aegypti females seems to be regulated by developmental changes and nutritional signals, and to be independent of mating stimulus.  相似文献

Aedes aegypti and Aedes albopictus are invasive mosquito species with geographic ranges that have oscillated within Florida since their presence was first documented. Local transmission of dengue, chikungunya, and Zika viruses serves as evidence of the public health importance of these two species. It is important to have detailed knowledge of their distribution to aid in mosquito control efforts and understand the risk of arbovirus transmission to humans. Through a partnership involving the University of Florida Institute of Food and Agricultural Sciences Cooperative Extension Service and the Florida Medical Entomology Laboratory; the Florida Department of Health; and mosquito control agencies throughout Florida, a container mosquito surveillance program involving all life stages was launched in the summer of 2016 to detect the presence of Ae. aegypti and Ae. albopictus. Results from this survey were mapped to provide a picture of the current known distribution of Ae. aegypti and Ae. albopictus in Florida. Aedes aegypti and/or Ae. albopictus were detected in the 56 counties that were part of the survey. Only Aedes albopictus was detected in 26 counties, primarily in the panhandle region of Florida. The results of this work underscore the importance of maintaining container mosquito surveillance in a state where chikungunya, dengue, and Zika viruses are present and where there is continued risk for exotic arbovirus introductions.  相似文献

We investigated the oviposition behavior of Ae. aegypti and Ae. albopictus. In particular we examined whether small‐scale site characteristics and the presence of conspecifics or congeners altered oviposition by these mosquitoes. Various combinations of females of the two species were allowed to oviposit inside cages among either vegetation (potted plants) or structural components (wood and concrete blocks). Numbers of eggs deposited per female were compared between species, sides, and treatments. Most significant differences between treatments and species involved differences between single species and mixed species treatments. Ae. aegypti deposited more eggs/female in the vegetation side than in the structure side whereas the opposite pattern was evident for Ae. albopictus. Ae. aegypti females had higher frequency of skip oviposition than Ae. albopictus. An average of 63% of the containers in the two‐species treatments contained eggs of both species, with more frequent joint occurrences observed in the treatment with three females of each species than in the treatments with one of each. Our results point to the existence of various interactions between gravid Ae. aegypti and Ae. albopictus females at or near the oviposition sites but further experimental work is necessary to fully characterize the interactions and their specific mechanisms.  相似文献

Benzoylphenyl ureas inhibit chitin synthesis and interfere with the molting process in arthropods. In this study, the effect of diflubenzuron on third‐instar larvae of Aedes aegypti was evaluated. The susceptibility to the product was determined, and the alterations generated were shown through light and electron microscopy. LC50 and LC90 were 0.23 and 0.47 ppm, respectively. The main alterations observed were the incapacity to complete the molt, a reduction of mobility, the fragmentation of the old cuticle, a division of the body segments that was not evident, and the deformation of the caudal structures. Images of the ultrastructure are included, where breaking zones in the cuticle were observed, separation of the cuticle, the epidermis and the muscles, and these latter with a disorganized arrangement. In low concentrations, from 0.15 ppm, diflubenzuron causes alterations in the behavior and morphology of Ae. aegypti.  相似文献

Eleven microsatellite markers were used to determine the genetic population structure and spread of Aedes aegypti (Stegomyia aegypti) (Diptera: Culicidae) in Pakistan using mosquitoes collected from 13 different cities. There is a single genetic cluster of Ae. aegypti in Pakistan with a pattern of isolation by distance within the population. The low level of isolation by distance suggests the long‐range passive dispersal of this mosquito, which may be facilitated by the tyre trade in Pakistan. A decrease in genetic diversity from south to north suggests a recent spread of this mosquito from Karachi. A strong negative correlation between genetic distance and the quality of road connections shows that populations in cities connected by better road networks are less differentiated, which suggests the human‐aided passive dispersal of Ae. aegypti in Pakistan. Dispersal on a large spatial scale may facilitate the strategy of introducing transgenic Ae. aegypti or intracellular bacteria such as Wolbachia to control the spread of dengue disease in Pakistan, but it also emphasizes the need for simple measures to control container breeding sites.  相似文献

Aedes aegypti (L.) mosquitoes preferentially oviposit in natural and artificial receptacles where their eggs are able to withstand drying as water levels fluctuate. Desiccation‐resistant eggs also increase the potential for establishment in non‐native habitats while providing logistical impediments to control programs. Viability and mean survival times of eggs stored under three dryness conditions for up to 367 days were investigated among three field‐derived colonies of Australian Ae. aegypti to understand variation in desiccation survival. Further investigations compared egg survival between an established colony and its wild counterpart. Our results confirmed that Ae. aegypti eggs can withstand desiccation for extended periods of time with approximately 2–15% egg viability recorded after one year and viability remaining above 88% under all conditions through 56 days. Intraspecific variations in egg survival times were recorded, suggesting local adaptation while each of the colonies demonstrated a consistent preference for higher humidity. Egg volume varied between the populations, suggesting a relationship between egg volume and survival time, with the marginally larger eggs (Charters Towers and Innisfail) having greater desiccation resistance over the range of conditions. The strong survivorship of Charters Towers eggs in dry, warm conditions demonstrates the adaptive significance of a desiccation‐resistant egg.  相似文献

Abstract. The incidence of dengue haemorrhagic fever (DHF) in Thailand has increased cyclically since the first recognized outbreak in 1958. Without an effective vaccine against dengue, and considering the clinical difficulty of treating DHF cases, vector control is needed to prevent dengue transmission. Since the establishment of the WHO Aedes Research Unit in 1964, continued since 1973 as the WHO Collaborating Centre at the Department of Medical Research in Bangkok, much operational research has been carried out in Thailand on the bionomics and control of dengue vectors: Aedes aegypti and Ae.albopictus. This review shows that, as in most other countries, dengue vector control programmes in Thailand make little use of the procedures arising from research, nor have they reduced the upward trend of dengue or prevented DHF outbreaks. Implications of the reluctance to use results of operational research on vector control are considered and remedial suggestions made.  相似文献

All traditional surveillance techniques for Aedes aegypti have been developed for the cosmopolitan domestic subspecies Ae. aegypti aegypti, and not the sylvatic subspecies, Ae. aegypti formosus. The predominant form in Western Kenya is Ae. aegypti formosus that is rarely associated with human habitations but is linked to transmission of sylvatic dengue virus strains. We compared five surveillance methods for their effectiveness in sampling Ae. aegypti formosus with the goal of determining a sustainable surveillance strategy in Kenya. The methods included larval and pupal surveys, oviposition trapping, BG‐Sentinel trapping, resting boxes, and backpack aspirations. Larval and pupal surveys collected the highest number of Ae. aegypti formosus (51.3%), followed by oviposition traps (45.7%), BG‐Sentinel traps (3.0%), and zero collected with either backpack aspiration or resting box collections. No Ae. aegypti formosus larvae or pupae were found indoors. The results indicate that oviposition traps and outdoor larval and pupal surveys were better surveillance methods for Ae. aegypti formosus in Western Kenya.  相似文献
