Maitotoxin (MTX),a potent cytolytic agent, activatesCa2+ entry via nonselective cationchannels in virtually all types of cells. The identity of the channelsinvolved and the biochemical events leading to cell lysis remainunknown. In the present study, the effect of MTX on plasmalemmalpermeability of human skin fibroblasts was examined. MTX produced atime- and concentration-dependent increase in cytosolic freeCa2+ concentration that dependedon extracellular Ca2+ and wasrelatively insensitive to blockade by extracellular lanthanides. MTXalso produced a time- and concentration-dependent increase inplasmalemma permeability to larger molecules as indicated by 1) uptake of ethidium (314 Da),2) uptake of YO-PRO-1 (375 Da), 3) release of intracellular fura 2 (636 Da), 4) uptake of POPO-3 (715 Da), and, ultimately,5) release of lactate dehydrogenase (relative molecular weight of 140,000). At the single cell level, uptake of YO-PRO-1 correlated in time with the appearance of large MTX-induced membrane currents carried by the organic cation,N-methyl-D-glucamine (167 Da). Thus MTX initially activatesCa2+-permeable cation channels andlater induces the formation of large pores. These effects of MTX onplasmalemmal permeability are similar to those seen on activation ofP2Z/P2X7 receptors ina variety of cell types, raising the intriguing possibility that MTXand P2Z/P2X7 receptor stimulationactivate a common cytolytic pore.

Astrocytes are involved in normal andpathological brain functions, where they become activated and undergoreactive gliosis. Astrocytes have been shown to respond toextracellular nucleotides via the activation of P2 receptors, either Gprotein-coupled P2Y receptors or P2X receptors that are ligand-gatedion channels. In this study, we have examined the manner in whichactivation of the P2X7 nucleotide receptor, anextracellular ATP-gated ion channel expressed in astrocytes, can leadto the phosphorylation of ERK1/2. Results showed that theP2X7 receptor agonist2',3'-O-(4-benzoyl)benzoyl-ATP induced ERK1/2phosphorylation in human astrocytoma cells overexpressing therecombinant rat P2X7 receptor (rP2X7-R), aresponse that was inhibited by the P2X7 receptorantagonist, oxidized ATP. Other results suggest thatrP2X7-R-mediated ERK1/2 phosphorylation was linked to thephosphorylation of the proline-rich/Ca2+-activated tyrosinekinase Pyk2, c-Src, phosphatidylinositol 3'-kinase, and proteinkinase C activities and was dependent on the presence ofextracellular Ca2+. These results support the hypothesisthat the P2X7 receptor and its signaling pathways play arole in astrocyte-mediated inflammation and neurodegenerative disease.

Extracellular ATP is known to trigger apoptosis of thymocytesand lymphocytes through a P2Z receptor at which ATP is a partial agonist, giving only 70% of the maximum response of3'-O-(4-benzoyl)benzoyl-adenosine 5'-triphosphate (BzATP), a full agonist. This cytolytic receptor and its associated ion channel areCa2+ (andBa2+) selective but also passmolecules up to the size of ethidium cation (314 Da).RT-PCR showed identity between lymphocyte P2Z and thehP2X7 gene recently cloned fromhuman monocytes. When human leukemic B lymphocytes were incubated withATP and133Ba2+,an immediate influx of isotope occurred. It was augmented by 45% whenATP was added 10 min before isotope. Time-resolved flow cytometry wasused to examine kinetics of ethidium uptake in cells incubated withBzATP or the partial agonists ATP, 2-methylthioadenosine 5'-triphosphate, or adenosine5'-O-(3-thiotriphosphate).Maximally effective concentrations of BzATP (50 µM) induced immediateuptake of ethidium at a rate linear with time. In contrast, a delay was observed (30 s) before ethidium uptake commenced after addition ofmaximally effective ATP concentrations (500 µM) at 37°C, and thedelay was longer at 24°C. ATP addition 2-10 min beforeethidium abolished the delay. The delay was longer with other partialagonists and inversely related to maximal flux produced by agonist. Adelay was also observed for submaximal BzATP concentrations (10-20µM). P2Z/P2X7 inhibitors, KN-62and5-(N,N-hexamethylene)-amiloride, reduced the rate of agonist-induced ethidium uptake and lengthened thedelay. The results support a model in which agonists forP2Z/P2X7 receptor mediate animmediate channel opening allowing passage of small inorganic cations,followed by a slow further permeability increase allowing passage oflarger permeant cations like ethidium. The rate of the second stepdepends on time and temperature and the efficacy and concentration ofagonist and is slowed by antagonists, suggesting it depends on thefraction of P2Z/P2X7 channels held in the initial openstate.

Extracellular ATPelevates cytosolic Ca2+ by activating P2X and P2Ypurinoceptors and voltage-sensitive Ca2+ channels (VCCCs)in PC-12 cells, thereby facilitating catecholamine secretion. Weinvestigated the mechanism by which ATP activates VSCCs.2-Methylthioadenosine 5'-triphosphate (2-MeS-ATP) and UTP were used aspreferential activators of P2X and P2Y, respectively. Nifedipineinhibited the ATP- and 2-MeS-ATP-evoked cytosolic Ca2+concentration increase and [3H]norepinephrine secretion,but not the UTP-evoked responses. Studies with Ca2+ channelblockers indicated that L-type VSCCs were activated after the P2Xactivation. Mn2+ entry profiles and studies withthapsigargin revealed that Ca2+ entry, rather thanCa2+ release, was sensitive to nifedipine. AlthoughP2X2 and P2X4 receptor mRNAs were detected,studies with pyridoxal phosphate-6-azophenyl-2',4'-disulfonic acidrevealed that P2X2 was mainly coupled to the L-type VSCCs. The inhibitory effect of nifedipine did not occur in the absence ofextracellular Na+, suggesting that Na+ influx,which induces depolarization, was essential for theP2X2-mediated activation of VSCCs. We report thatdepolarization induced by Na+ entry through theP2X2 purinoceptors effectively activates L-type VSCCs inPC-12 cells.

Hyperpolarization in human leukemia THP-1 monocytes adherent tovascular cell adhesion molecule (VCAM)-1 is due to an induction ofinwardly rectifying K+ currents(Iir) (Colden-Stanfield M and Gallin EK,Am J Physiol Cell Physiol 275: C267-C277, 1998).We determined whether the VCAM-1-induced hyperpolarization issufficient to augment the increase in intracellular free calcium([Ca2+]i) produced by Ca2+ storedepletion with thapsigargin (TG) and readdition of external CaCl2 in fura 2-loaded THP-1 monocytes. Whereas there was a2.1-fold increase in [Ca2+]i in monocytesbound to glass for 5 h in response to TG and CaCl2 addition, adherence to VCAM-1 produced a 5-fold increase in[Ca2+]i. Depolarization of monocytes adherentto VCAM-1 by Iir blockade or exposure to high[K+] abolished the enhancement of the peak[Ca2+]i response. In monocytes bound toglass, hyperpolarization of the membrane potential with valinomycin, aK+ ionophore, to the level of hyperpolarization seen incells adherent to VCAM-1 produced similar changes in peak[Ca2+]i. Adherence of monocytes to E-selectinproduced a similar peak [Ca2+]i to cellsbound to glass. Thus monocyte adherence to the physiological substrateVCAM-1 produces a hyperpolarization that is sufficient to enhanceCa2+ entry and may impact Ca2+-dependentmonocyte function.

Lymphocytes from normal subjects and patients with B-chroniclymphocytic leukemia (B-CLL) show functional responses to extracellular ATP characteristic of the P2X7 receptor (previously termedP2Z). These responses include opening of a cation-selectivechannel/pore that allows entry of the fluorescent dye ethidium andactivation of a membrane metalloprotease that sheds the adhesionmolecule L-selectin. The surface expression of P2X7receptors was measured in normal leucocytes, platelets, and B-CLLlymphocytes and correlated with their functional responses. Monocytesshowed four- to fivefold greater expression of P2X7 than B,T, and NK lymphocytes, whereas P2X7 expression onneutrophils and platelets was weak. All cell types demonstratedabundant intracellular expression of this receptor. All 12 subjectswith B-CLL expressed lymphocyte P2X7 at about the samelevel as B lymphocytes from normal subjects. P2X7 function, measured by ATP-induced uptake of ethidium, correlated closely withsurface expression of this receptor in normal and B-CLL lymphocytes andmonocytes (n = 47, r = 0.70; P< 0.0001). However, in three patients the ATP-induced uptake ofethidium into the malignant B lymphocytes was low or absent. The lackof P2X7 function in these B lymphocytes was confirmed bythe failure of ATP to induce Ba2+ uptake into theirlymphocytes. This lack of function of the P2X7 receptorresulted in a failure of ATP-induced shedding of L-selectin, anadhesion molecule that directs the recirculation of lymphocytes fromblood into the lymph node.

We investigatedthe role of intracellular calcium concentration([Ca2+]i) in endothelin-1 (ET-1) production,the effects of potential vasospastic agents on[Ca2+]i, and the presence of L-typevoltage-dependent Ca2+ channels in cerebral microvascularendothelial cells. Primary cultures of endothelial cells isolated frompiglet cerebral microvessels were used. Confluent cells were exposed toeither the thromboxane receptor agonist U-46619 (1 µM),5-hydroxytryptamine (5-HT; 0.1 mM), or lysophosphatidic acid (LPA; 1 µM) alone or after pretreatment with the Ca2+-chelatingagent EDTA (100 mM), the L-type Ca2+ channel blockerverapamil (10 µM), or the antagonist of receptor-operated Ca2+ channel SKF-96365 HCl (10 µM) for 15 min. ET-1production increased from 1.2 (control) to 8.2 (U-46619), 4.9 (5-HT),or 3.9 (LPA) fmol/µg protein, respectively. Such elevated ET-1biosynthesis was attenuated by verapamil, EDTA, or SKF-96365 HCl. Toinvestigate the presence of L-type voltage-dependent Ca2+channels in endothelial cells, the [Ca2+]isignal was determined fluorometrically by using fura 2-AM. Superfusionof confluent endothelial cells with U-46619, 5-HT, or LPA significantlyincreased [Ca2+]i. Pretreatment ofendothelial cells with high K+ (60 mM) or nifedipine (4 µM) diminished increases in [Ca2+]i inducedby the vasoactive agents. These results indicate that 1)elevated [Ca2+]i signals are involved in ET-1biosynthesis induced by specific spasmogenic agents, 2) theincreases in [Ca2+]i induced by thevasoactive agents tested involve receptor as well as L-typevoltage-dependent Ca2+ channels, and 3) primarycultures of cerebral microvascular endothelial cells express L-typevoltage-dependent Ca2+ channels.

In a variety of disorders, overaccumulation of lipid in nonadipose tissues, including the heart, skeletal muscle, kidney, and liver, is associated with deterioration of normal organ function, and is accompanied by excessive plasma and cellular levels of free fatty acids (FA). Increased concentrations of FA may lead to defects in mitochondrial function found in diverse diseases. One of the most important regulators of mitochondrial function is mitochondrial Ca2+ ([Ca2+]m), which fluctuates in coordination with intracellular Ca2+ ([Ca2+]i). Polyunsaturated FA (PUFA) have been shown to cause [Ca2+]i mobilization albeit by unknown mechanisms. We have found that PUFA but not monounsaturated or saturated FA cause [Ca2+]i mobilization in NT2 human teratocarcinoma cells. Unlike the [Ca2+]i response to the muscarinic G protein-coupled receptor agonist carbachol, PUFA-mediated [Ca2+]i mobilization in NT2 cells is independent of phospholipase C and inositol-1,4,5-trisphospate (IP3) receptor activation, as well as IP3-sensitive internal Ca2+ stores. Furthermore, PUFA-mediated [Ca2+]i mobilization is inhibited by the mitochondria uncoupler carboxyl cyanide m-chlorophenylhydrozone. Direct measurements of [Ca2+]m with X-rhod-1 and 45Ca2+ indicate that PUFA induce Ca2+ efflux from mitochondria. Further studies show that ruthenium red, an inhibitor of the mitochondrial Ca2+ uniporter, blocks PUFA-induced Ca2+ efflux from mitochondria, whereas inhibitors of the mitochondrial permeability transition pore cyclosporin A and bongkrekic acid have no effect. A glutamateto alanine exchange at amino acid position 496 of the humanP2X7 receptor was recently shown to be associated with aloss of function in human B lymphocytes in terms of ATP-induced ethidium+ uptake, Ba2+ influx, and induction ofapoptosis (Gu BJ, Zhang WY, Worthington RA, Sluyter R, Dao-UngP, Petrou S, Barden JA, and Wiley JS. J Biol Chem 276:11135-11142, 2001). Here we analyzed the effect of theGlu496 to Ala exchange on the channel properties of thehuman P2X7 receptor expressed in Xenopus oocyteswith the two-microelectrode voltage-clamp technique. The amplitudes ofATP-induced whole cell currents characteristic of functionalexpression, kinetic properties including ATP concentration dependence,and permeation behavior were not altered by this amino acid exchange.Also in HEK293 cells, the Ala496 mutant mediated typicalP2X7 receptor-dependent currents like the parentGlu496 hP2X7 receptor. Because the function ofthe P2X7 receptor as an ATP-gated channel for small cationsincluding Ba2+ remained unaffected by this mutation, weconclude that Glu496 plays a critical role in poreformation but does not determine the ion channel properties of thehuman P2X7 receptor.

We previously reported that human growth hormone (hGH) increases cytoplasmic Ca2+ concentration ([Ca2+]i) and proliferation in pancreatic -cells (Sjöholm Å, Zhang Q, Welsh N, Hansson A, Larsson O, Tally M, and Berggren PO. J Biol Chem 275: 21033–21040, 2000) and that the hGH-induced rise in [Ca2+]i involves Ca2+-induced Ca2+ release facilitated by tyrosine phosphorylation of ryanodine receptors (Zhang Q, Kohler M, Yang SN, Zhang F, Larsson O, and Berggren PO. Mol Endocrinol 18: 1658–1669, 2004). Here we investigated the tyrosine kinases that convey the hGH-induced rise in [Ca2+]i and insulin release in BRIN-BD11 -cells. hGH caused tyrosine phosphorylation of Janus kinase (JAK)2 and c-Src, events inhibited by the JAK2 inhibitor AG490 or the Src kinase inhibitor PP2. Although hGH-stimulated rises in [Ca2+]i and insulin secretion were completely abolished by AG490 and JAK2 inhibitor II, the inhibitors had no effect on insulin secretion stimulated by a high K+ concentration. Similarly, Src kinase inhibitor-1 and PP2, but not its inactive analog PP3, suppressed [Ca2+]i elevation and completely abolished insulin secretion stimulated by hGH but did not affect responses to K+. Ovine prolactin increased [Ca2+]i and insulin secretion to a similar extent as hGH, effects prevented by the JAK2 and Src kinase inhibitors. In contrast, bovine GH evoked a rise in [Ca2+]i but did not stimulate insulin secretion. Neither JAK2 nor Src kinase inhibitors influenced the effect of bovine GH on [Ca2+]i. A postulated therapeutic avenue in cystic fibrosis (CF) is activation of Ca2+-dependent Cl channels via stimulation of Ca2+ entry from extracellular solutions independent of CFTR functional status. We have shown that extracellular zinc and ATP induce a sustained increase in cytosolic Ca2+ in human airway epithelial cells that translates into stimulation of sustained secretory Cl transport in non-CF and CF human and mouse airway epithelial cells, cell monolayers, and nasal mucosa. On the basis of these studies, the Ca2+ entry channels most likely involved were P2X purinergic receptor channels. In the present study, molecular and biochemical data show coexpression of P2X4, P2X5, and P2X6 subtypes in non-CF (16HBE14o) and CF (IB3-1) human bronchial epithelial cells. Other P2X receptor Ca2+ entry channel subtypes are expressed rarely or not at all in airway epithelia, epithelial cell models from other CF-relevant tissues, or vascular endothelia. Novel transient lipid transfection-mediated delivery of small interference RNA fragments specific to P2X4 and P2X6 (but not P2X5) into IB3-1 CF human airway epithelial cells inhibited extracellular zinc- and ATP-induced Ca2+ entry markedly in fura-2 Ca2+ measurements and "knocked down" protein by >65%. These data suggest that multiple P2X receptor Ca2+ entry channel subtypes are expressed in airway epithelia. The role of nitric oxide (NO) in the occurrence of intracellular Ca2+ concentration ([Ca2+]i) oscillations in pituitary GH3 cells was evaluated by studying the effect of increasing or decreasing endogenous NO synthesis with L-arginine and nitro-L-arginine methyl ester (L-NAME), respectively. When NO synthesis was blocked with L-NAME (1 mM) [Ca2+]i, oscillations disappeared in 68% of spontaneously active cells, whereas 41% of the quiescent cells showed [Ca2+]i oscillations in response to the NO synthase (NOS) substrate L-arginine (10 mM). This effect was reproduced by the NO donors NOC-18 and S-nitroso-N-acetylpenicillamine (SNAP). NOC-18 was ineffective in the presence of the L-type voltage-dependent Ca2+ channels (VDCC) blocker nimodipine (1 µM) or in Ca2+-free medium. Conversely, its effect was preserved when Ca2+ release from intracellular Ca2+ stores was inhibited either with the ryanodine-receptor blocker ryanodine (500 µM) or with the inositol 1,4,5-trisphosphate receptor blocker xestospongin C (3 µM). These results suggest that NO induces the appearance of [Ca2+]i oscillations by determining Ca2+ influx. Patch-clamp experiments excluded that NO acted directly on VDCC but suggested that NO determined membrane depolarization because of the inhibition of voltage-gated K+ channels. NOC-18 and SNAP caused a decrease in the amplitude of slow-inactivating (IDR) and ether-à-go-go-related gene (ERG) hyperpolarization-evoked, deactivating K+ currents. Similar results were obtained when GH3 cells were treated with L-arginine. To test thehypothesis that intracellular Ca2+activation of large-conductanceCa2+-activatedK+ (BK) channels involves thecytosolic form of phospholipase A2 (cPLA2), we first inhibited theexpression of cPLA2 by treating GH3 cells with antisenseoligonucleotides directed at the two possible translation start siteson cPLA2. Western blot analysis and a biochemical assay of cPLA2activity showed marked inhibition of the expression ofcPLA2 in antisense-treated cells.We then examined the effects of intracellularCa2+ concentration([Ca2+]i)on single BK channels from these cells. Open channel probability (Po) for thecells exposed to cPLA2 antisenseoligonucleotides in 0.1 µM intracellularCa2+ was significantly lower thanin untreated or sense oligonucleotide-treated cells, but the voltagesensitivity did not change (measured as the slope of thePo-voltagerelationship). In fact, a 1,000-fold increase in[Ca2+]ifrom 0.1 to 100 µM did not significantly increasePoin these cells, whereas BK channels from cells in the other treatmentgroups showed a normalPo-[Ca2+]iresponse. Finally, we examined the effect of exogenous arachidonic acidon thePoof BK channels from antisense-treated cells. Although arachidonic aciddid significantly increasePo,it did so without restoring the[Ca2+]isensitivity observed in untreated cells. We conclude that although [Ca2+]idoes impart some basal activity to BK channels inGH3 cells, the steepPo-[Ca2+]irelationship that is characteristic of these channels involves cPLA2.

Palytoxin-induced cell death cascade in bovine aortic endothelial cells   总被引:1,自引:0,他引:1  
The plasmalemmal Na+-K+-ATPase (NKA) pump is the receptor for the potent marine toxin palytoxin (PTX). PTX binds to the NKA and converts the pump into a monovalent cation channel that exhibits a slight permeability to Ca2+. However, the ability of PTX to directly increase cytosolic free Ca2+ concentration ([Ca2+]i) via Na+ pump channels and to initiate Ca2+ overload-induced oncotic cell death has not been examined. Thus the purpose of this study was to determine the effect of PTX on [Ca2+]i and the downstream events associated with cell death in bovine aortic endothelial cells. PTX (3–100 nM) produced a graded increase in [Ca2+]i that was dependent on extracellular Ca2+. The increase in [Ca2+]i initiated by 100 nM PTX was blocked by pretreatment with ouabain with an IC50 < 1 µM. The elevation in [Ca2+]i could be reversed by addition of ouabain at various times after PTX, but this required much higher concentrations of ouabain (0.5 mM). These results suggest that the PTX-induced rise in [Ca2+]i occurs via the Na+ pump. Subsequent to the rise in [Ca2+]i, PTX also caused a concentration-dependent increase in uptake of the vital dye ethidium bromide (EB) but not YO-PRO-1. EB uptake was also blocked by ouabain added either before or after PTX. Time-lapse video microscopy showed that PTX ultimately caused cell lysis as indicated by release of transiently expressed green fluorescent protein (molecular mass 27 kDa) and rapid uptake of propidium iodide. Cell lysis was 1) greatly delayed by removing extracellular Ca2+ or by adding ouabain after PTX, 2) blocked by the cytoprotective amino acid glycine, and 3) accompanied by dramatic membrane blebbing. The involvement of cAMP- andCa2+-mediated pathways in theactivation of tyrosine hydroxylase (TH) gene expression by nicotine wasexamined in PC-12 cells. ExtracellularCa2+ and elevations inintracellular Ca2+ concentration([Ca2+]i)were required for nicotine to increase TH mRNA. The nicotine-elicited rapid rise in[Ca2+]iwas inhibited by blockers of either L-type or N-type, and to a lesserextent P/Q-, but not T-type, voltage-gatedCa2+ channels. With continualnicotine treatment,[Ca2+]ireturned to basal levels within 3-4 min. After a lag of~5-10 min, there was a smaller elevation in[Ca2+]ithat persisted for 6 h and displayed different responsiveness toCa2+ channel blockers. This secondphase of elevated[Ca2+]iwas blocked by an inhibitor of store-operatedCa2+ channels, consistent with theobserved generation of inositol trisphosphate.1,2-Bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid-AM (BAPTA-AM), when added before or 2 h after nicotine,prevented elevation of TH mRNA. Nicotine treatment significantly raised cAMP levels. Addition of the adenylyl cyclase inhibitor2',5'-dideoxyadenosine (DDA) prevented thenicotine-elicited phosphorylation of cAMP response element bindingprotein. DDA also blocked the elevation of TH mRNA only when addedafter the initial transient rise in [Ca2+]iand not after 1 h. This study reveals that several temporal phases areinvolved in the induction of TH gene expression by nicotine, each ofthem with differing requirements forCa2+ and cAMP.

In cultured rat cerebellar granule cells, glutamate or N-methyl-D-aspartate (NMDA) activation of the NMDA receptor caused a sustained increase in cytosolic Ca2+ levels ([Ca2+]i), reactive oxygen species (ROS) generation, and cell death (respective EC50 values for glutamate were 12, 30, and 38 µM) but no increase in caspase-3 activity. Removal of extracellular Ca2+ blocked all three glutamate-induced effects, whereas pretreatment with an ROS scavenger inhibited glutamate-induced cell death but had no effect on the [Ca2+]i increase. This indicates that glutamate-induced cell death is attributable to [Ca2+]i increase and ROS generation, and the [Ca2+]i increase precedes ROS generation. Apoptotic cell death was not seen until 24 h after exposure of cells to glutamate. S-nitrosoglutathione abolished glutamate-induced ROS generation and cell death, and only a transient [Ca2+]i increase was seen; similar results were observed with another nitric oxide (NO) donor, S-nitroso-N-acetylpenicillamine, but not with glutathione, which suggests that the effects were caused by NO. The transient [Ca2+]i increase and the abolishment of ROS generation induced by glutamate and S-nitrosoglutathione were still seen in the presence of an ROS scavenger. Glial cells, which were present in the cultures used, showed no [Ca2+]i increase in the presence of glutamate, and glutamate-induced granule cell death was independent of the percentage of glial cells. Malignant hyperthermia (MH) is a potentially fatal pharmacogenetic syndrome caused by exposure to halogenated volatile anesthetics and/or depolarizing muscle relaxants. We have measured intracellular Ca2+ concentration ([Ca2+]i) using double-barreled, Ca2+-selective microelectrodes in myoballs prepared from skeletal muscle of MH-susceptible (MHS) and MH-nonsusceptible (MHN) swine. Resting [Ca2+]i was approximately twofold in MHS compared with MHN quiescent myoballs (232 ± 35 vs. 112 ± 11 nM). Treatment of myoballs with caffeine or 4-chloro-m-cresol (4-CmC) produced an elevation in [Ca2+]i in both groups; however, the concentration required to cause a rise in [Ca2+]i elevation was four times lower in MHS than in MHN skeletal muscle cells. Incubation of MHS cells with the fast-complexing Ca2+ buffer BAPTA reduced [Ca2+]i, raised the concentration of caffeine and 4-CmC required to cause an elevation of [Ca2+]i, and reduced the amount of Ca2+ release associated with exposure to any given concentration of caffeine or 4-CmC to MHN levels. Tonic contraction of corpus cavernosum smooth muscle cells (SMCs) maintains the flaccid state of the penis, and relaxation is initiated by nitric oxide (NO), leading to erection. Our aim was to investigate the effect of NO on the smooth muscle cellular response to adrenergic stimulation in corpus cavernosum. Fura-2 fluorescence was used to record intracellular Ca2+ concentration ([Ca2+]i) from freshly isolated SMCs from rat and human. Phenylephrine (PE) transiently elevated [Ca2+]i in the presence and absence of extracellular Ca2+, indicating release from intracellular stores. Whereas the NO donor S-nitroso-N-acetylpenicillamine (SNAP) with sildenafil citrate (SIL) caused no change in basal [Ca2+]i, the PE-induced rise of [Ca2+]i was reversibly inhibited by 27 ± 7% (n = 21, P < 0.005) in rat and by 55 ± 15% (n = 9, P < 0.01) in human SMCs. SNAP and SIL also reduced the contractile response to PE. To investigate the mechanism, we applied mediators alone or in combination. The soluble guanylyl cyclase inhibitor ODQ reduced the effect of SNAP and SIL. SIL, cGMP analogs, and NO donors without SIL did not reduce the PE-induced rise of [Ca2+]i. However, the combination of 8-bromo-cGMP with SNAP reduced the Ca2+ peak by 42 ± 9% (n = 22, P < 0.01). Our results demonstrate that NO and cGMP act synergistically to reduce Ca2+ release from intracellular stores. This study examines theCa2+ influx-dependent regulationof the Ca2+-activatedK+ channel(KCa) in human submandibulargland (HSG) cells. Carbachol (CCh) induced sustained increases in theKCa current and cytosolic Ca2+ concentration([Ca2+]i),which were prevented by loading cells with1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid (BAPTA). Removal of extracellularCa2+ and addition ofLa3+ orGd3+, but notZn2+, inhibited the increases inKCa current and[Ca2+]i.Ca2+ influx during refill (i.e.,addition of Ca2+ to cells treatedwith CCh and then atropine inCa2+-free medium) failed to evokeincreases in the KCa current but achieved internal Ca2+ storerefill. When refill was prevented by thapsigargin,Ca2+ readdition induced rapidactivation of KCa. These dataprovide further evidence that intracellularCa2+ accumulation provides tightbuffering of[Ca2+]iat the site of Ca2+ influx (H. Mogami, K. Nakano, A. V. Tepikin, and O. H. Petersen. Thyroid cells express a variety of P2Y and P2X purinergic receptor subtypes. G protein-coupled P2Y receptors influence a wide variety of thyrocyte-specific functions; however, functional P2X receptor-gated channels have not been observed. In this study, we used whole cell patch-clamp recording and fluorescence imaging of the plasma membrane marker FM1-43 to examine the effects of extracellular ATP on membrane permeability and trafficking in the Fisher rat thyroid cell line FRTL. We found a cation-selective current that was gated by ATP and 2',3'-O-(4-benzoylbenzoyl)-ATP but not by UTP. The ATP-evoked currents were inhibited by pyridoxal phosphate 6-azophenyl-2',4'-disulfonic acid, adenosine 5'-triphosphate-2',3'-dialdehyde, 100 µM Zn2+, and 50 µM Cu2+. Fluorescence imaging revealed pronounced, temperature-sensitive stimulation of exocytosis and membrane internalization by ATP with the same pharmacological profile as observed for activation of current. The EC50 for ATP stimulation of internalization was 440 µM in saline containing 2 mM Ca2+ and 2 mM Mg2+, and 33 µM in low-Mg2+, nominally Ca2+-free saline. Overall, the results are most consistent with activation of a P2X7 receptor by ATP4–. However, low permeability to N-methyl-D-glucamine+ and the propidium cation YO-PRO-1 indicates absence of the cytolytic pore that often accompanies P2X7 receptor activation. ATP stimulation of internalization occurs in Na+-free, Ca2+-free, or low-Mg2+ saline and therefore does not depend on cation influx through the ATP-gated channel.
