A fine hexagonal network of microscopic ridges has been discovered on the internal shell surface of the cosmopolitan Lower Palaeozoic inarticulate genus Lingulella. The micro-ornamentation has been recognised on all well-preserved specimens examined, from Cambrian and Ordovician successions in Ireland, North America, Gt. Britain, Spitzbergen, and China. Examination of the mantle of the related Recent genus Lingula revealed that the outer epithelial cells are arranged in a hexagonal close-packed pattern comparable in size and form to the micro-ornamentation in the fossil shells. This fact, along with the discovery of a polygonal pattern on some organic layers in the Lingula shell, suggests that the micro-ornamentation in fossil Lingulella is the mould of the outer epithelium responsible for shell secretion. No trace of epithelial moulds has yet been found in specimens of Lingula from younger Palaeozoic successions. This difference may prove to be a useful diagnostic feature in distinguishing linguloid inarticulates from one another. The preservation of polygonal imprints of epithelial cells in acrotretoid brachiopods is reviewed and supplemented by new data from the Cambrian of southern Great Britain. The imprints are confirmed as representing moulds of epithelial cells rather than an artefact of microstructure or preserved soft tissues, as they are (1) recorded in most taxa reviewed, (2) best preserved in areas where the shell has been thickened and (3) similar in size to cells recorded in Lingula, the closest living relative to the now extinct acrotretoids. Analysis of the morphology and sizes of epithelial cell moulds demonstrates that there is no consistent relationship between cell width and valve size, and that epithelial cells are not a useful taxonomic character within this group.  相似文献

The enigmatic Cambrian animal chancelloriids were discovered in a wide range of taphonomic settings; however, preservation of biomineralized sclerite microstructure was solely known from secondarily phosphatized skeletal remains. Here, we investigate a uniquely pyritized chancelloriid from the lower Cambrian Guojiaba Formation in southern Shaanxi Province, China, using a combination of advanced analytic techniques. Results of the energy dispersive spectroscopy (EDS), X-Ray Fluorescence (XRF), and Raman spectrum show that the sclerites and scleritomes are preserved as pyritized internal moulds with a calcitic outer layer. The outer layer enveloping the internal moulds likely represents the recrystallized counterpart of the original biomineralized sclerite wall. Distinctive fibrous microstructures are discovered in the sclerites, which echo the features seen in the phosphatized fossils of chancelloriids. The typical microstructure, along with the recrystallized calcite, corroborate the interpretation that chancelloriid sclerites were originally constructed by fibrous aragonite. The stability of the microstructure and mineral composition in both carbonate and siliciclastic backgrounds indicate that chancelloriids were adapted to exploit aragonitic fibres to build their skeletons regardless of the change of their living environments.  相似文献

Internal moulds of the laterally compressed monoplacophoran Anabarella plana Vostokova, 1962, and likely earliest rostroconch Watsonella Grabau, 1900, from the Early Cambrian of the Siberian Platform, show similar microstructures. The moulds are covered with a thin phosphatic crust replicating the inner morphology and microstructure of the shells. The shells were completely removed during etching of the samples in 10% acetic acid, except for some moulds of Watsonella sp., which retained an incompletely preserved and recrystallized wall. Three types of microtexture were found in moulds of Anabarella and Watsonella : polygonal, spiny and step-wise. The polygonal texture is well exposed in the apical area and dorsal margin and is interpreted to represent a prismatic outer layer. The polygons can be partially overlapped by spiny and stepwise textures that may represent a crossed-lamellar inner layer. Prisms and lamellae were first-order structural units, probably consisting of fibers. The similar shell microstructures of Anabarella and Watsonella , especially at the dorsal margins, support the hypothesis that a laterally compressed monoplacophoran such as Anabarella plana was the first evolutionary step from monoplacophorans towards Early Cambrian bivalves via the earliest rostroconch-like Watsonella.  相似文献

Taphonomic analysis of Lower and Middle Oxfordian ammonites from the Cracow Upland, southern Poland (localities at Pod???e, Zalas, M?ynka) revealed differences in ammonite preservation. The studied ammonites, usually termed as external and internal moulds, show a more complex state of preservation. In the Middle Oxfordian glauconitic marls, ammonites are preserved as internal moulds with neomorphic calcite shells showing relics of the original internal structure. In the Middle Oxfordian platy peloidal limestones, ammonites are preserved mostly as external moulds, without septal suture, however under microscope might show relics of internal whorls and septa and/or subtle differences in sediment filling phragmocone chambers. In sponge–microbial bioherms and biostromes, ammonite internal moulds have shells, which in contrast to ammonites from glauconitic marls are not strictly neomorphic ones, but originated by shell dissolution and subsequent filling of moldic porosity by calcite cement. In sponge–microbial nodular limestones, the ammonites are strongly deformed and the outer wall is usually removed by dissolution under pressure. Other important taphonomic differences include the rate of compaction (highest in platy limestones), sedimentary infillings, microborings, encrustations and preservation of siphuncular tubes. The majority of the ammonites appear to be phragmocones; aptychi in all facies are rare. Siphuncular tubes are fossilized exclusively in oppeliids, only in specimens from glauconitic marls and platy limestones, although their other taphonomic attributes are different. Tubes seem to have fossilized due to microbially mediated phosphatization that could be favoured by a set of parameters which operated rather at the scale of ammonoid carcasses: closed, poorly oxygenated conditions, and reduced pH. Taphonomic processes were controlled by the sedimentary environment (fragmentation, sedimentary filling, phosphatization of siphuncular tubes), as well as by early and late diagenesis (neomorphic transformation, dissolution, cementation, compaction) influenced by lithology.  相似文献

Characteristics of Streptomyces coelicolor A3(2) aerial spore rodlet mosaic   总被引:5,自引:0,他引:5  
Cytochemical analysis of Streptomyces coelicolor (A3(2) indicated that the aerial growth rodlet mosaic is a polysaccharide. Statistical analysis of frequency distributions of individual rodlet lengths from control and ether-reoriented spore mosaics indicated that the rodlet fibrillar image is the result of individual particulates, rather than evaginations in a continuous sheet of material. A model of the mature sport envelope was developed from freeze-etch-replicated, thin-sectioned, and critical point dried S. coelicolor A3(2) mature spores. The rodlet mosaic was situated between the outer spore wall and an external granuloma matrix. Mixture spore envelope layers from the inner surface to the external surface are plasma membrane, inner spore wall, outer spore wall, rodlet mosaic, an undefined granular matrix, and the sheath. The granular matrix had an uneven thickness and much of the matrix was frequently absent from the interspore spaces of mature spore chains. Streptomyces coelicolor A3(2) mosaic rodlets were isolated by acetic acid refluxing, then ethanol precipitation. Complete acid hydrolysis of rodlets released on sugar which cochromatographed with D-glucosamine-HCl and released acetic acid at 139% of the expected level. Cell associated rodlet mosaics and isolated mosaic rodlets were hydrolyzed with chitinase. Infrared spectra of isolated rodlets were similar to crab chitin spectra.  相似文献

Retinal pigment epithelial cells selectively phagocytize rod outer segments by a process that may be mediated by specific cell surface receptors. Since many receptors are glycoproteins, we have studied the effect of tunicamycin, an inhibitor of N-linked oligosaccharide synthesis, and of castanospermine and swainsonine, which are inhibitors of oligosaccharide processing, on the ability of cultured retinal pigment epithelial cells to phagocytize rod outer segment. Tunicamycin inhibits the glycosylation of newly synthesized glycoproteins by 85-90%; concomitantly, the phagocytosis of rod outer segments is inhibited by 70-80%. The effect of tunicamycin is to initially reduce rod outer segments binding, and therefore the subsequent ingestion of rod outer segments. SDS-PAGE analysis and autoradiography of [35S]methionine labelled extracts of tunicamycin-treated cells, demonstrates the disappearance of a number of glycoprotein bands, and the appearance of a number of protein bands of lower Mr. Kinetic analysis of the disappearance and reappearance of specific glycoproteins suggests that the lower Mr bands are the non-glycosylated forms of the higher Mr bands. By contrast, castanospermine and swainsonine have no effect on the ability of retinal pigment epithelial cells to phagocytize rod outer segments, or on the SDS-PAGE pattern of treated cells, although they were shown to inhibit oligosaccharide processing as expected. These results support the hypothesis that rod outer segment phagocytosis by retinal pigment epithelial cells is mediated by specific glycoprotein receptors. N-Glycosylation of these receptors is required for their function, or for their insertion into the plasma membrane, whereas processing of the N-linked oligosaccharide chains of these receptors is not crucial for rod outer segment phagocytosis by retinal pigment epithelial cells.  相似文献

Background and Aims

Thousands of floor mosaics were produced in lands across the Roman and Byzantine empires. Some mosaics contain depictions of agricultural produce, potentially providing useful information concerning the contemporary presence and popularity of crop plants in a particular geographical region. Hundreds of floor mosaics produced in Israel during the Byzantine period have survived. The objective of the present work was to search these mosaics for Cucurbitaceae in order to obtain a more complete picture of cucurbit crop history in the eastern Mediterranean region.

Results and Conclusions

Twenty-three mosaics dating from 350–600 ce were found that had images positively identifiable as cucurbits. The morphological diversity of the cucurbit fruits in the mosaics of Israel is greater than that appearing in mosaics from any other Roman or Byzantine provincial area. The depicted fruits vary in shape from oblate to extremely long, and some are furrowed, others are striped and others lack definite markings. The cucurbit taxa depicted in the mosaics are Cucumis melo (melon), Citrullus lanatus (watermelon), Luffa aegyptiaca (sponge gourd) and Lagenaria siceraria (bottle gourd). Cucumis melo is the most frequently found taxon in the mosaics and is represented by round dessert melons and long snake melons. Fruits of at least two cultivars of snake melons and of watermelons are represented. To our knowledge, images of sponge gourds have not been found in Roman and Byzantine mosaics elsewhere. Indeed, the mosaics of Israel contain what are probably the oldest depictions of Luffa aegyptiaca in Mediterranean lands. Sponge gourds are depicted often, in 11 of the mosaics at eight localities, and the images include both mature fruits, which are useful for cleaning and washing, and immature fruits, which are edible. Only one mosaic has images positively identifiable as of bottle gourds, and these were round–pyriform and probably used as vessels.  相似文献

Although the unique organization of vertebrate cone mosaics was first described long ago,both their underlying molecular basis and physiological significance are largely unknown.Here,we demonstrate that Crumbs proteins,the key regulators of epithelial apical polarity,establish the planar cellular polarity of photoreceptors in zebrafish.Via heterophilic Crb2a-Crb2b interactions,the apicobasal polarity protein Crb2b restricts the asymmetric planar distribution of Crb2a in photoreceptors.The planar polarized Crumbs proteins thus balance intercellular adhesions and tension between photoreceptors,thereby stabilizing the geometric organization of cone mosaics.Notably,loss of Crb2b in zebrafish induces a nearsightedness-like phenotype in zebrafish accompanied by an elongated eye axis and impairs zebrafish visual perception for predation.These data reveal a detailed mechanism for cone mosaic homeostasis via previously undiscovered apical-planar polarity coordination and propose a pathogenic mechanism for nearsightedness.  相似文献

Mantle tissue plays an important role in shell biomineralization by secreting matrix proteins for shell formation. However, the mechanism by which it regulates matrix protein secretion is poorly understood, largely because of the lack of cellular tools for in vitro study and techniques to evaluate matrix protein secretion. We have isolated the outer epithelial cells of the mantle of the pearl oyster, Pinctada fucata, and evaluated cellular metabolism by measuring the secretion of the matrix protein, nacrein. A novel sensitive sandwich enzyme-linked immunosorbent assay (ELISA) was established to quantify nacrein. Mantle explant culture was demonstrated to provide dissociated tissue cells with high viability. Single dissociated cell types from explant culture were separated by density in a discontinuous Percoll gradient. The outer epithelial cells were isolated from other cell types by their higher density and identified by immunolabeling and ultrastructure analysis. ELISA assays revealed that the outer epithelial cells retained the ability to secrete nacrein in vitro. Moreover, increased nacrein secretion resulted from an increased Ca(2+) concentration in the culture media of the outer epithelial cells, in a concentration-dependent manner. These results confirm that outer epithelial cell culture and the ELISA method are useful tools for studying the regulatory mechanisms of shell biomineralization.  相似文献

Summary In the perifoveal retina of the monkey, Cercopithecus aethiops, the melanin granules are accumulated in apical cytoplasmatic protrusions of the pigment epithelial cells, facing the end of the cones. The rods are inserted deeper into the pigment epithelium than the cones; they reach the bottom of the infoldings of the apical surface membrane of the pigment epithelial cells. No melanin granules or other inclusions are situated at the end of the rods. The outer extremity of the rods is considerably inclined and in sections often appears as groups of rod discs which are incompletely or completely separated from the main part of the outer segments. This separation is regarded as an artifact caused by the inclination of the rods, and it is therefore not considered to represent phagocytosis of the outer segments by the pigment epithelium.The inclusions of the pigment epithelial cells are classified in five categories which seem to be related to each other owing to their shared structural characteristics. It is suggested that melanin granules are produced, modified and destroyed by the pigment epithelial cells of the adult.Because of the relations between the photoreceptors and the melanin granules it is suggested that light scattered by the melanin granules may pass backwards through the outer segments of the cones, but not of the rods.This investigation was supported in part by the Danish Foundation for the Advancement of Science and by the Danish Medical Research Council.  相似文献

The anatomy of the epithelial surface of the uterine cervix was investigated in 31 mice with scanning electron microscope (SEM). The cervox of 20 mice was painted twice a week during five months with 3,4-benzpyrene (BP) in acetone and in 11 mice with acetone alone. Only two of the 15 acetone or BP treated animals with histologic normal epithelium showed similar structures as those described earlier for the normal cervical epithelium of untreated mice. The remaining 13 animals had regular mosaics, intermediate or cobblestone structures covered with irregular microvilli with or without long, finger-like protrusions. Irregular mosaics, intermediate or cobblestone structures were seen in atypical epithelium. In atypias Grade III and in invasive carcinoma irregular cobblestone structures with cellular overlapping were present. Atypical and invasive carcinoma were usually furnished with irregular microvilli, irregular fragmented microrugae and finger-like protrusions. The proportion of mice having anisovillosis with finger-like protrusions increased with increasing degree of epithelial severity. Since pathologic SEM changes were observed in some treated mice having normal histology the possibility was entertained that alterations at the cellular membrane level may preceed the nucleocytoplasmic changes required for the recognition of atypical epithelium in conventional histologic preparations.  相似文献

Exfoliated vaginal epithelial cells with attached bacteria, termed 'clue cells', which were procured from a patient with non-specific vaginitis, were stained with ruthenium red and examined by transmission electron microscopy. The attached bacteria appeared to adhere by means of an outer fibrillar coat. An epithelial tissue culture cell line (McCoy) and human red blood cells to which strains of Gardnerella vaginalis attached were similarly examined. The adherence of G. vaginalis to the epithelial cell line appeared to be mediated by an outer fibrillar coat while adherence to red cells appeared to be mediated by fimbriae. Transmission electron microscopy was performed on the Gardnerella strains used. Thin sections of tissue-culture-adherent strains revealed a dense outer fibrillar coat whereas the surface of the haemagglutinating strains showed fine fimbriae. Negative staining of haemagglutinating strains demonstrated fimbriae on a minority of organisms.  相似文献

The juvenile shell of the brachiopod Discinisca consists of a mosaic of micrometer-sized siliceous tablets embedded in a chitinous substrate. The first-formed tablets are secreted on glycocalyx by a newly differentiated collective of outer epithelial cells. They are mainly rhombic but may also be ellipsoidal, discoidal, or deformed and sporadically overlap one another. On the surrounding juvenile shell, secreted by an incipient outer mantle lobe, the tablets are nearly all perfect rhombic plates in rhombic arrays. Their constant size, arrangement, and centripetal crystallization suggest intracellular assembly. The tablets, which are normally bilamellar, consist of discrete aggregates of crystalline spherules of silica in rhombic arrays within an organic matrix of fibrous protein and, presumably, a soluble polysaccharide(s). Mosaic secretion ceases at about the time when juveniles settle on the sea bed, which more or less coincides with the secretion of a ring of lamellae around the mosaic, induced by rapid advances and retractions of the outer mantle lobe prior to deposition of the organophosphatic mature shell. Energy dispersion X-ray analyses of pelagic and newly settled juveniles show that phosphatic secretion, even in the site of the first-formed outer epithelial collective, does not begin until all siliceous secretion has ceased.  相似文献

The spatial pattern of defoliation by mammalian herbivores across vegetation mosaics bas been frequently discussed, but rarely spatially quantified. Here we considered the role of plant-herbivore interactions in determining the spatial distribution of shrub defoliation by a large mammalian herbivore across a grass-shrub mosaic.
We investigated the spatial pattern of heather defoliation by sheep in heather-grass mosaics. Heather-grass mosaics are two-phased vegetation mosaics, in which a spatially localized plant community (grass) fulfils nutritional needs, whilst a spatially extensive plant community (heather) meets energy requirements but is nutritionally marginal.
We used a spatial analysis method, originating from human geography, to show that heather defoliation was not spread across the mosaic homogeneously, but that the spatial pattern was determined by geometric characteristics of the mosaic, grazing intensity, and the contrast between preferred and less preferred communities.
The spatial analysis method proved to be a powerful tool to describe the spatial pattern of shrub defoliation. Applications of the method are explored and the implications of the spatial distribution of shrub defoliation are discussed.  相似文献

Genetic analysis of muscle specification, formation and function in model systems has provided valuable insight into human muscle physiology and disease. Studies in Drosophila have been particularly useful for discovering key genes involved in muscle specification, myoblast fusion, and sarcomere organization. The muscles of the Drosophila female reproductive system have received little attention despite extensive work on oogenesis. We have used newly available GFP protein trap lines to characterize of ovarian muscle morphology and sarcomere organization. The muscle cells surrounding the oviducts are multinuclear with highly organized sarcomeres typical of somatic muscles. In contrast, the two muscle layers of the ovary, which are derived from gonadal mesoderm, have a mesh-like morphology similar to gut visceral muscle. Protein traps in the Fasciclin 3 gene produced Fas3::GFP that localized in dots around the periphery of epithelial sheath cells, the muscle surrounding ovarioles. Surprisingly, the epithelial sheath cells each contain a single nucleus, indicating these cells do not undergo myoblast fusion during development. Consistent with this observation, we were able to use the Flp/FRT system to efficiently generate genetic mosaics in the epithelial sheath, suggesting these cells provide a new opportunity for clonal analysis of adult striated muscle.  相似文献

Diploid-triploid mosaics are rarely found in vertebrates, and until now they were known to be common in only two vertebrate species complexes. Here we report that diploid-triploid mosaics are widespread among unisexual hybrids of the minnows Phoxinus eos and Phoxinus neogaeus, a complex already known to contain diploid and triploid forms. Using chromosome counts and flow cytometry, we show that the mosaics occur throughout the known range of the unisexuals and are abundant in many of these natural populations. The mosaics are highly heterogeneous, showing individual variation in the ratio of diploid to triploid cells, and as a group they appear to form a continuum between the pure diploid and triploid forms. Tissue-graft analysis shows that the third genome present in the triploid cells of a mosaic is expressed, because grafts made from the mosaics show an incidence of rejection intermediate between that of the diploid (clonal) and triploid (nonclonal) biotypes.  相似文献

Summary The fine structure of the pharynx is presented and demonstrates that the pharyngeal epithelial system is a continuous one. The epithelial lining of the pharyngeal cavity with its characteristic fibrous secretory bodies merges with the outer pharyngeal epithelium at the point of anchorage of the pharynx. A few of these cells are insunk, the nuclei occurring beneath the underlying muscular layers. The nature of the outer epithelium changes towards the free end of the pharynx; the cells become ciliated and in contents come to resemble the inner epithelium which it joins at the tip.The gut cells merge at a transitional zone with the inner pharyngeal epithelium and at this point both bear microvilli and contain rod-shaped apical bodies. Some of these cells are also insunk. Towards the mouth the epithelium shows a greater degree of insinking and exhibits microapocrine secretion. Both inner and outer epithelia bear sense receptors which are concentrated at the lip.At the point of pharyngeal insertion, the sub-epithelial tissue resembles planarian parenchyma, but is rich in gland cells.
