Analysis of growth during phototropic curvature of cress hypocotyls   总被引:3,自引:3,他引:0  
Abstract. Growth rates throughout an organ curving phototropically under continuous, unilateral while light were monitored by lime-lapse photography of cress hypocotyls marked into 1 mm sections by two rows of ion-exchange beads. Curvature resulted from an integrated sequence of changes in growth rate on each side of the organ, but the actual patterns of change and, therefore rales of curvature, differed within even this one species, depending upon the immediate pretreatment of the seedlings. Transference of seedlings from darkness to unilateral irradiation gave immediate growth inhibition on both sides of the organ. Curvature resulted from differential recovery of growth rate, being seen first on the shaded side, most prominently in the apical regions; only 2h after initial exposure to light did growth recover on the lit (lower) side. On the other hand, transfer of seedlings from omnilateral to unilateral irradiation of the same intensity resulted in simultaneous growth inhibition on the irradiated side and stimulated growth on the shaded side: this growth stimulation of the shaded side was greater than occurred in totally darkened control plants.  相似文献

Abstract. The phototropic and the geotropic sensitivities of cress hypocotyls differed in etiolated and in green seedlings. In etiolated seedlings, phototropism was poorly developed and geotropism seemed to be the dominant orientation mechanism. In green seedlings, although geotropic sensitivity was slightly greater, phototropic responses were markedly enhanced, such that photo-signals could over-ride, or even reverse, geocurvature. The 'de-etiolation' light pretreatments required to bring about these changes in the photo-and geo-systems were different. The slight enhancement of geocurvature could be achieved by short-term exposure to red light and was reversed by far-red irradiation. The marked stimulation of photocurvature required extensive irradiation pretreatments.  相似文献

Abstract. Growth in various regions of the hypocotyls of dark-grown cress seedlings before and after exposure lo continuous white light has been analysed by time-lapse photography. In the dark, growth in the hook was minimal, the upward growth of the seedling being sustained by extension of the shank, especially the uppermost zones. Following irradiation, the hook and the remainder of the hypocotyl showed dissimilar growth responses. Previously growing regions of the shank were inhibited while zones within the hook, especially the apical end of the inner (concave) side, showed marked growth stimulation. These changes in growth rates commenced within 1 h from exposure to the light stimulus and thus considerably preceded any observable changes in hook angle.  相似文献

Analysis of growth during geotropic curvature in seedling hypocotyls   总被引:4,自引:4,他引:0  
Abstract. The patterns of growth in organs curving under the influence of gravity were analysed by time-lapse photography of cress and cucumber hypocotyls which were delimited into 1 mm zones by ion-exchange beads. Geotropic curvature resulted from changes in growth rate on both sides of the organ. Growth inhibition of varying degrees of intensity occurred in all the previously growing zones of the upper (concave) side. An absolute reduction in length due to compression frequently occurred in some zones. Also, in both species growth stimulation was observed on the lower (convex) side. The disparity in growth rate between the upper and lower surfaces varied with time, being more apparent in the subapical region in the first hour of curvature. A later promotion of growth rate on the lower surface subsequently increased the curvature of the more basal zones. Autotropic straightening occurred as a consequence of growth changes, both inhibitory and stimulatory, in the apical zones. These events indicate a polarity of response in which apical zones have precedence over basal zones.  相似文献

To correlate endogenous growth inhibitors of dwarf pea to its dwarfism a thorough search of growth inhibitors was made in the neutral fraction of an acetone extract from the shoots of light-grown seedlings of Pisum sativum L. cvs Progress No 9 and Alaska. From both cultivars six inhibitors were separated and named A-1, A-2, A-3, B-1, B-2 and B-3 based on their order of elution from the silica gel column. Their contents as determined by cress root bioassay were: in cv. Progress 0.40, 16.5, 6.36, 1.02, 0.11 and 0.10, and in cv. Alaska 0.33, 2.35, 3.51, 0.95, 0.10 and 0.09 cress units (g fresh weight)−1. Their contributions to growth regulation of the relevant pea plants were estimated as the products of the above-stated contents times the ratios of the specific activities of each standard sample in the cress roots and the relevant pea cultivars, and they were; in cv. Progress A-2, 5.44 and A-3, 2.10, and in cv. Alaska A-2, 0.68 and A-3, 0.88, those of A-2 and A-3 constituting more than 90% of the total contribution of the six inhibitors in either cultivar. The great differences in the content and contribution to growth regulation of A-2 and A-3 between the two cultivars suggest that the higher contents of and greater responsiveness to the two inhibitors in cv. Progress may be causes of dwarfism of this cultivar. A-l and A-3 were spectroscopically identified with pisatin and a mixture of cis, trans– , and trans, trans xanthoxins.  相似文献

Hypocotyl growth in Sinapis alba L: the roles of light quality and quantity   总被引:3,自引:3,他引:0  
Abstract. A comparison is made of the relative effectiveness of light quality and light quantity on the elongation growth of Sinapis alba hypocotyls. The results show that hypocotyl extension rate in plants which have not previously been exposed to light is controlled primarily by the prevailing photon fluence rate when the phytochrome photostationary state lies between ∼0.033 and ∼0.81. Below ∼0.033, changes in photostationary state also have a marked effect on extension rate. Elongation growth in light-adapted plants is controlled by both photon fluence rate and the spectral quality of the incident radiation at all photoequilibria. Photosynthesis can modify these responses but is not essential as a prior condition for a green plant to respond to changes in light quality and quantity.  相似文献

Temporal course of graviperception in intermittently stimulated cress roots   总被引:2,自引:0,他引:2  
Gravitropic bending of Lepidium roots caused by intermittent stimulation lasting ≈ 1 h was the same for a particular sum of stimulation intervals and was independent of (i) the length of a single stimulation interval (from 1 to 12·2 s) during which the roots were exposed unilaterally and horizontally, and (ii) rest intervals (from 60 to 300 s) during which roots were horizontally rotated at two revolutions per minute on a clinostat. The same effectiveness of equal sums of short stimulations separated by relatively long rest intervals indicates that the signals into which the stimuli are transduced are: (i) additive; (ii) proportional to the duration of a single stimulation; and (iii) stable for at least 5 min. The perception time is shorter than 1 s, the presentation time is ≈ 10 s. The effects of intermittent stimulation fit the hypothesis that the gravity-induced movement of statoliths changes asymmetrically the stress in cytoskeletal actin filaments, thereby inducing gravitropic bending.  相似文献

A reduced rate of greening after continuous illumination was observed in dark-grown cress seedlings ( Lepidium sativum L.) incubated with 5-aminolevulinate (ALA) or the complexing agents 2,2'-bipyridyl, 8-hydroxyquinoline or 1,10-phenanthroline. This effect cannot be explained merely by photodynamic damage caused by chlorophyll precursors which are accumulated in the dark under these conditions. Flash light experiments revealed that photoconversion of protochlorophyll(ide) to chlorophyllide was not influenced by chelator treatment. The next step in the chlorophyll pathway, the esterification of chlorophyllide, however, was inhibited. Simultaneously applicated ALA and complexing agents did not result in a synergistic reponse; on the contrary, ALA seemed to render cress plants less susceptible to the treatment with complexing agents upon subsequent irradiation. Ultrastructural studies demonstrated that grana formation in light was inhibited after pretreatment with ALA or complexing agents.  相似文献

In order to clarify the role of endogenous growth inhibitors A-2α and A-2β in a dwarf pea plant, red light (emission peak 657 nm) treated, 9-d-old seedlings of dwarf pea (Pisum sativum L. cv. Progress No. 9) were transferred to darkness, and the resulting changes in growth rate and concentrations of A-2α and A-2β were monitored. The growth rate of the epicotyls increased, and the concentration of the inhibitors in the epicotyls decreased, according to sigmoidal time courses. The relationship between the logarithms of the concentration of the inhibitors and the corresponding growth rate was linear. These results suggest that A-2α and A-2β, may play an important role in the growth recovery process of the dwarf pea cultivar after termination of red light irradiation. Abstract. The growth response of etiolated cucumber ( Cucumis sativus L.) hypocotyls to gravitropic stimulation was examined by means of time-lapse photography and high-resolution analysis of surface expansion and curvature. In comparison with video analysis, the technique described here has five- to 20-fold better resolution; moreover, the mathematical fitting method (cubic splines) allows direct estimation of local and integrated curvature. After switching seedlings from a vertical to horizontal position, both upper and lower surfaces of the stem reacted after a lag of about 11 min with a two- to three-fold increase in surface expansion rate on the lower side and a cessation of expansion, or slight compression, on the upper surface. This growth asymmetry was initiated simultancously along the length of the hypocotyl, on both upper and lower surfaces, and did not migrate basipetally from the apex. Later stages in the gravitropic response involved a complex reversal of the growth asymmetry, with the net result being a basipetal migration of the curved region. This secondary growth reversal may reflect oscillatory and or self-regulatory behaviour of growing cells. With some qualifications, the kinetics and pattern of growth response are consistent with a mechanism involving hormone redistribution, although they do not prove such a mechanism. The growth kinetics require a growth mechanism which can be stimulated by two-to three-fold or completely inhibited within a few minutes.  相似文献

InHordeum vulgare cultivar “Kirin-choku No. 1”, the final length of intact coleoptiles of totally etiolated seedlings was approximately twice as long as that of those grown under continuous red light. The fluence response curve of the latter was biphasic; the low-energy effect was saturated by red light of ca. 50 J m−2 which gave rise to about 40% of the maximum inhibition by continuous irradiation with red light of 1.2 W m−2, whereas the high-energy effect was induced by irradiation for 1 hr or longer. Coleoptiles of 3-day-old seedlings were most sensitive to light causing the low-energy effect, which was repeatedly red/far-red reversible. The growth inhibition was correlated to the photometrically measured percentage of Pfr so that the maximum effect was induced by red light of 50 J m−2 which transformed 70% of phytochrome to Pfr in the coleoptile tip. Wavelength dependence of the high-energy effect showed that monochromatic light of 400, 600 and 650 nm greatly inhibited the coleoptile growth, whereas light of 700 and 750 nm promoted it instead. The effect was also induced by intermittent irradiation with red light, and the more frequently the intermittent treatment was given, the more the growth was inhibited.  相似文献

Effects of red light (R), far-red light (FR) and UV radiation on growth and greening of lettuce seedlings (Latuca sativa L., cv. Berlinskii) have been investigated. UV-B and UV-C inhibited hypocotyl elongation and stimulated cotyledonary growth. R in combination with UV-B and UV-C partly eliminated these effects, but FR increased those and reversed the R effect. Chlorophyll accumulation was inhibited by UV-B and UV-C. In comparison with cotyledonary growth, R strengthened the UV inhibitory effect, and FR reversed this effect of R. Thus, UV and phytochrome system modify the effects of each other on hypocotyl and leaf growth in lettuce seedlings depending on the level of active phytochrome formed.  相似文献

The stem elongation responses of etiolated peas (Pisum sativum L.) to fluorescent light (35–45 mol.mt-2.s-1) were recorded using high resolution position transducers. Continuous fluorescent light decreased growth by 70% within 9 min. The growth rate declined to 5% of the control over the next 2 h and remained at this level for 7 h. Pulses of fluorescent light ranging from 8 s to 34 min led to partial suppression of growth and resulted in a complex kinetic response. The distinctive kinetics of blue and red light inhibition were apparent as components of the responses to non-saturating levels of fluorescent light. The rapid suppression of growth by blue light was not affected by concomitant red light. The lag time for the onset of red light inhibition was not affected by concomitant blue, but the rate of inhibition appeared accelerated.  相似文献

The relationship between growth, change in cell osmotic pressure and accumulation of osmotic solutes was investigated in hypocotyls of sunflower (Helianthus annum L.) seedlings. During growth in darkness the osmotic pressure decreased by 50% between days 2 and 6 after sowing. After irradiation of dark-grown seedlings with continuous white light (WL) an inhibition of hypocotyl growth was measured, but the osmotic pressure of the growing cells was not lower than in the dark-grown control. Growth in darkness and after WL irradiation was accompanied by an increase in the amount of osmotic substances (soluble sugars) which was proportional to the increase in length of the organ. During growth in continuous WL the cell osmotic pressure decreased by 45 % between days 2 and 6 after sowing. The transfer of WL-grown seedlings to darkness (“re-etiolation”) resulted in a rapid acceleration of hypocotyl growth, but the cell osmotic pressure was the same as that of the WL grown control. Growth in continuous WL was accompanied by a corresponding accumulation of osmotic substances (soluble sugars). The transition from WL to darkness resulted in an enhanced accumulation of osmotica and an increase in cell-wall extensibility. The results indicate that the relative maintenance of cell osmotic pressure during rapid hypocotyl growth in darkness is caused by an enhanced accumulation of soluble sugars into the growing cells of the organ.  相似文献

A variety of electrolytes (10-30 mol m-3) increased the relative growth rate of etiolated cucumber (Cucumis sativus L. cv. Burpee's Pickler) hypocotyls by 20-50% relative to water-only controls. The nonelectrolyte mannitol inhibited growth by 10%. All salts tested were effective, regardless of chemical composition or valence. Measurements of cell-sap osmolality ruled out an osmotic mechanism for the growth stimulation by electrolytes. This, and the nonspecificity of the response, indicate that an electrical property of the solutions was responsible for their growth-stimulating activity. Measurements of surface electrical potential supported this reasoning. Treatment with electrolytes also enhanced nutation and altered the pattern of phototropic curvature development. A novel analytical method for quantitating these effects on growth was developed. The evidence indicates that electrolytes influence an electrophysiological parameter that is involved in the control of cell expansion and the coordination of growth underlying tropisms and nutations.  相似文献

Photoreceptors, phytochromes and cryptochromes regulate hypocotyl growth under specific conditions, by suppressing negative gravitropism, modulating phototropism and inhibiting elongation. Although these effects seem to be partially caused via the regulation of the phytohormone auxin, the molecular mechanisms underlying this process are still poorly understood. In our present study, we demonstrate that the flabby mutation enhances both phytochrome- and cryptochrome-inducible hypocotyl bending in Arabidopsis. The FLABBY gene encodes the ABC-type auxin transporter, PGP19, and its expression is suppressed by the activation of phytochromes and cryptochromes. Our current results therefore indicate that the phytochromes and cryptochromes have at least two effects upon the tropic responses of the hypocotyls in Arabidopsis: the enhancement of hypocotyl bending through the suppression of PGP19, and a PGP19-independent mechanism that induces hypocotyl bending. By the using an auxin polar transport assay and DR5:GUS expression analysis, we further find that the phytochromes inhibit basipetal auxin transport, and induce the asymmetric distribution of auxin in the hypocotyls. These data suggest that the control of auxin transport by phytochromes and cryptochromes is a critical regulatory component of hypocotyl growth in response to light.  相似文献

Brief irradiation of intact etiolated seedlings of maize (Zea mays L.) with red light (R; 30 W cm-2, 10 min) reduces the amounts of diffusible and free (solvent-extractable) indole-3-acetic acid (IAA) obtainable from excised coleoptile tips. The effect is transient, the lowest level (30% of the dark control) occurring at about 3 h after irradiation. The free-IAA content of the whole coleoptile and the diffusible-IAA yield from the base of the same organ are similarly reduced, whereas the conjugated-IAA content of the coleoptile is not affected. These results support the view that R inhibits the production of IAA at the coleoptile tip. It is further shown that R inhibits biosynthesis of [3H]IAA from [3H]tryptophan supplied to the coleoptile tip. The shapes of the fluence-response curves obtained for the reduction of the diffusible-IAA yield by R and far-red light (FR) indicate the participation of two photoreactive systems. One has thresholds at 10-3 W s cm2 of R, five orders of magnitude less than the minimum required for the appearance of spectrophotometrically measurable far-red-absorbing form of phytochrome (Pfr) in vivo, and 10-1 W s cm-2 of FR; its response is linear to the logarithm of fluence exceeding five orders of magnitude. The other system is seen above 102 W s cm-2 as an increase in the slope of the fluenceresponse curve; its response is FR reversible and related to the Pfr level of total photoreversible phytochrome. Both systems inhibit biosynthesis of IAA from tryptophan. Elongation of the coleoptile is stimulated by R; the stimulation is most apparent in the apical region, and is saturated with a fluence at which bo detectable pfr is formed. Farred light can also saturate this response. At a concentration of 17 µmol·L–1, paclobutrazol (PP), a triazole plant growth retardant, effectively reduced the elongation and increased the thickness of hypocotyls in 6-day-old Phaseolus vulgaris L. cv. Juliska seedlings, both in the light and in the dark. PP treatment did not increase the cell number in transverse sections of hypocotyls. The diameter of hypocotyls was uniform from the zone of intensive elongation along the whole hypocotyl in etiolated plants, but those grown in the light exhibited an additional lateral expansion at the base. Ethylene evolution was not reduced by PP in etiolated hypocotyls, and did not differ significantly in the elongating apical and fully grown basal zones. PP reduced the ethylene release by the growing zones in green hypocotyls, but not in the basal parts, which resulted in an increasing ethylene gradient towards the hypocotyl base. The level of 1-aminocyclopropane-1-carboxylic acid (ACC), the immediate precursor of ethylene, was much higher in retardant-treated hypocotyls than in the controls, which was due in part to the reduced malonylation. The swelling of the hypocotyl bases could be eliminated by inhibitors of ethylene biosynthesis or action, or could be induced by 10 µmol·L–1ACC in control plants in the light. None of these treatments had a significant effect on the lateral expansion of hypocotyls in etiolated seedlings. PP treatment induced a similar effect to that of white light in etiolated seedlings, and amplified the effect of light in green plants with respect to the ACC distribution, and consequently, the ethylene production in the hypocotyls of 6-day-old bean seedlings. It can be concluded that the lateral expansion of hypocotyl bases in PP-treated green plants is controlled by ethylene.  相似文献
