Potato tubers artificially inoculated with Fusarium solani var. coeruleum or F. sulphureum 3 months after harvest were uniformly wounded and held at 5, 10 or 15°C for up to 21 days before immersion in fungicide suspensions. Holding tubers for 14 days at 15°C (curing conditions) or at 5°C did not alter the incidence of dry rot subsequently developing on tubers stored at 10°C, and holding tubers for up to 21 days at 15°C slightly increased disease caused by both pathogens. Thiabendazole, imazalil and prochloraz applied to tubers immediately after wounding almost completely prevented dry rot. Treatment after 3 days was less effective and the amount of disease increased with further delay; fungicides were more effective on tubers held at 5°C than at 10 or 15°C before treatment and storage, and efficacy of the fungicide was decreased by increasing the amount of inoculum on tubers. Wounds became less susceptible to infection by F. solani var. coeruleum and F. sulphureum when tubers were held at 15°C before inoculation, and the incidence of rots was decreased by 70–80% by delaying inoculation for 7 days. Treating tubers with dichlorophen immediately after wounding slightly increased the disease. The effects of fungicide treatment, curing conditions and wound healing are discussed.  相似文献

In 1982 – 88, potatoes were grown in 2-, 4- and 6-course rotations with spring barley on a field infested with Globodera rostochiensis. Severity of stem canker and black scurf increased with increasing frequency of previous potato crops, and seed tuber treatment with tolclofos-methyl became less effective in controlling diseases. This suggested that previous crops had increased the amounts of soil-borne inoculum of Rhizoctonia solani. Oxamyl soil treatment increased stem canker in one year and decreased black scurf in four years. Seed tuber treatment with imazalil or prochloraz decreased stem base infection by Polyscytalum pustulans and skin spot and silver scurf on tubers. Black dot was prevalent on tubers in all years and was not affected by seed tuber treatment or previous cropping. Oxamyl increased black dot and common scab in five years and decreased % tuber dry matter in six years. Cysts of G. rostochiensis were found attached to Désirée but not to Maris Piper tubers in August. At harvest tubers of both cultivars were affected by superficial pitting and its severity was related to soil populations of G. rostochiensis at planting. This damage was controlled by oxamyl. It is suggested that the pitting developed from holes made in the tuber skin at larval invasion. In 1989, Désirée seed tubers and healthy mini tubers were planted in all plots and severity of stem canker and black scurf increased with increasing proximity of previous potato crops and with the number of previous crops. Black dot on stems and tubers was not affected by previous cropping but was much less severe in a plot that had not grown potatoes during the seven years of the experiment. The severity of common scab generally decreased as the number of preceding potato crops increased.  相似文献

In a series of trials in the years of 1973-76 inclusive, chitted seed tubers of potato cvs Ulster Sceptre, Majestic and Pentland Crown were immersed immediately before planting, on three dates in each year, in water, suspensions of Erwinia carotovora var. atroseptica, Phoma exigua var. foveata or a mixture of both. The development of blackleg was greatest in cv. Ulster Sceptre and most rapid after late planting. Its final incidence was not closely related to date of planting except in cv. Pentland Crown which was least affected when planted late.
Combined inoculum of P. exigua var. foveata and E. carotovora var. atroseptica increased five-fold the number of plants that failed to emerge and often retarded early growth of the remainder. Failure to emerge was more frequent the earlier seed tubers were planted. Yield was affected most by blanking and blackleg in the Erwinia- inoculated plots and was also reduced by the pre-plant dip in a P. exigua var. foveata suspension, especially after early planting.
Seed dipped in the mixed suspensions yielded progeny that after wounding and cool storage developed a lower incidence of gangrene than progeny from seed dipped in P. exigua var. foveata alone. There was no evidence that planting time influenced the incidence of gangrene during storage.  相似文献

A method of measuring the water potential of stored potato tubers (Solanum tuberosum L.) was needed to investigate the relationship of bacterial soft rot in tubers to water potential. Pressure chamber measurements, while useful for tubers with functional stolons, cannot be made on stored tubers. Measurements could be made on excised tissue pieces in a hygrometer chamber and with hygrometers implanted into tubers. We report here our evaluation of these hygrometric methods using a comparison with the pressure chamber on tubers harvested with stolons intact.

In tubers of high water potential, measurements on excised tissue were as much as 0.5 megapascals lower than the pressure chamber, probably due to turgor-driven expansion of the sample when freed from constraints imposed by surrounding tissue. Good agreement (±0.05 megapascals) was found between the implanted hygrometer and the pressure chamber at potentials higher than −0.5 megapascals. At lower water potentials, both hygrometer measurements were higher than the pressure chamber. Respirational heating of the tissue contributed to the increase in the excised tissue samples, but not with the implanted hygrometers because of the hygrometer design. The osmotic pressure balanced the pressure chamber measurement of potential at −0.7 megapascals, but was too small to do so at lower potentials. At most, 25% of this discrepancy can be accounted for by dilution by apoplastic water. We believe that the pressure chamber measurement is too low at low water potentials and that the error is associated with air bubbles in the xylem. At low potentials air emerged from xylem vessels along with sap, and fewer xylem emitted sap as potentials decreased.

The susceptibility to gangrene infection of wounds of various shapes and depths on potato tubers was studied by inflicting wounds using differently-shaped brass teeth and rods of different diameters. Inoculating wounds with spore suspensions or damaging tubers which had been previously contaminated with Phoma exigua var. foveata or which had been recently lifted from plots of field experiments showed that wounds in which tissue was crushed were most susceptible to infection. Over a wide range of inoculum concentrations and in experiments using several different cultivars the incidence of infection of any wound type was compared to that of the standard severe cut and crush wound. Using a probit transformation a linear relationship was established, the slope of the line indicating the relative susceptibility of the wound. In 1977 and 1978, crops of cv. Pentland Dell were surveyed for damage incidence, inoculum and inoculum potential on arrival at a commercial bulk store. Nets of tubers buried among the tuber bulk were recovered after storage and gangrene incidence compared with damage and inoculum assessments. Inoculum potential and incidence of severe damage both influenced disease development but damage incidence was of greater importance, showing that priority should be given to decreasing damage and to curing to promote rapid wound healing in endeavours to control the disease.  相似文献

The nutrient contents of the two cultivare of Potato‐Irish Cobbler and Red Pontiac were quantitatively altered by Rhizoctonia bataticola during pathogenesis. The glucose content of both cultivare increased whereas sucrose, maltose and fructose contents decreased appreciably in quantity during the period of incubation. The total protein and lipid contents of both cultivare were also lowered quantitatively by the rot‐causing organism. The depletion of total carbohydrate, protein and lipid contents, was more in Irish Cobbler cultivar than in Red Pontiac cultivar.  相似文献

Under optimum growing conditions neither tuber- nor soil-borne Phoma exigua var. foveata inoculum appreciably affected stand or yield of the subsequent potato crop. Seed tubers with gangrene rots caused high levels of stem and tuber symptoms when planted in var. foveata contaminated or uncontaminated land; contaminated seed tubers with no rots also produced progeny with a high gangrene potential. Sufficient soil-borne inoculum was carried over in land that produced a gangrene affected crop in the previous year to override the effect of tuber disinfection. Effective gangrene control was achieved by a combination of tuber disinfection shortly after harvest over successive years with a 1 in 5 yr potato crop rotation. Gangrene rots usually developed through injuries to the tuber periderm, rots in other tubers being associated with pustules of powdery scab (Spon-gospora subterranea).  相似文献

The relationships of potato (Solanum tuberosum L.) tuber membrane permeability and membrane lipid composition to sugar accumulation were examined. Tubers from four potato cultivars were stored for 40 weeks at 3°C and 9°C. Rates of tuber membrane electrolyte leakage, total fatty acid composition, free fatty acid composition, and sugar content were measured throughout the storage period. Storage of tubers at 3°C caused dramatic increases in total fatty acid unsaturation, membrane permeability, and sugar content compared to tubers stored at 9°C. Cultivars with higher levels of fatty acid unsaturation had lower rates of membrane electrolyte leakage and lower sugar contents. We propose that high initial levels or high induced levels of membrane lipid unsaturation mitigate increases in tuber membrane permeability during storage, thus positively influencing the processing quality of stored potato tubers.  相似文献

Potato tubers of the cultivar Pentland Dell were investigated for ultrastructural changes following impact. Studies were carried out on tubers after 12 and 24 wk of storage using a falling bolt delivering 0.7 J of energy. Ultra-thin sections were examined by light and transmission electron microscopy. Tubers stored for 12 wk exhibited no visual bruise formation, whereas visible bruising resulted following impact of tubers stored for 24 wk. An ultrastructural sequence of events during bruising was established as (1) a collapse of intracellular compartmentation, (2) increased ribosomal and mitochondrial abundance within the cytoplasm, (3) increased density of cytoplasm adjacent to the cell wall and surrounding amyloplasts, and (4) the development of melanin in bruised cells. These observations are discussed in relation to tuber turgor and physiology during storage.  相似文献

In studies of the influence of haulm defoliants on gangrene incidence in storage the use of diquat dibromide was consistently found to exacerbate levels of Phoma exigua vzr.foveata infection particularly on tubers harvested 3–4 wk after burning down. Intermediate levels occurred where the haulm was chemically defoliated with dinoseb or was physically cut and removed and least where the defoliants were sodium chlorate or sulphuric acid. Pycnidia of P. exigua var. foveata developed within 10 days on stems desiccated with diquat dibromide, sulphuric acid or dinoseb and most prolifically on those treated with diquat dibromide but tuber infection was not always related to their abundance. Speed of kill was not considered to be important in determining effect on gangrene incidence.  相似文献

Heavy infestations of stored potato (Solanum tuberosum L.) tubers by the two potato tuber moth species Symmetrischema tangolias (Gyen) and Tecia solanivora (Povolny) frequently occur in Andean potato‐growing regions of Ecuador. The aim of the study was to develop a biological control strategy for both species using powder formulations made of inert substances, Phthorimaea operculella (Zeller) granulovirus (PhopGV) and Bacillus thuringiensis Berliner subsp. kurstaki (Btk). The LC50 of PhopGV on T. solanivora was 0.33 LE/L, and Btk caused 82.7% mortality at a concentration of 100 g/L in bioassays. The efficacy of talcum, kaolin, calcium carbonate and sand ranged between 76.2% and 98.7%. Calcium carbonate was highly effective to control both species; however, its efficacy was affected by the relative humidity and dropped to 55.4% at relative humidity of 100%. PhopGV at concentrations of five larvae equivalents (LE) per kg kaolin and Btk at a concentration of 60 g Btk/kg talcum caused 95.7% and 88.1% mortality of T. solanivora, respectively. In storage experiments, the efficacy of calcium carbonate alone and in combination with PhopGV (20 LE/kg) and Btk (15 g/kg) caused 95.0–99.8% mortality of T. solanivora in all treatments and reduced infestation on potato tubers by 83.6%–91.0%. In the case of S. tangolias, Btk significantly increased mortality to 96.5% compared with calcium carbonate alone and reduced tuber infestation by 83.4%. Storage of potato tubers in thin layers enhanced the efficacy of the calcium carbonate treatment compared with storage in bags. It was concluded that calcium carbonate alone seems to be appropriate for the control of T. solanivora, and an addition of 15 g Btk/kg would improve the control of S. tangolias. It is suggested to test these new formulations under on‐farm storage conditions.  相似文献

A filtrate, prepared from potato tuber by grinding in an isotonic medium, has been separated into a particulate and a ‘soluble’ fraction by ultracentrifugation. Following dialysis and lyophilization, both fractions catalysed the oxidation of l-DOPA, with approximately 30% of the l-DOPA: oxygen-oxidoreductase (EC 1.14.18.1; DOPA oxidase) activity being associated with the particulate fraction. When dithiothreitol (DTT, 10?2M was included in the grinding medium, much lower yields of DOPA oxidase were obtained and 80% appeared to be associated with the particulate fraction. DTT proved to be a powerful inhibitor of DOPA oxidase. With concentrations of DTT causing only partial inhibition, the kinetics of the inhibited rate of dopachrome formation from l-DOPA were complex. When oxygen consumption was measured inhibition was not transient. The degree of inhibition was inversely related to the DOPA oxidase activity, indicating interaction of a product of this activity with DTT. Direct determination of -SH groups in DTT using 5,5′-dithiobis(2-nitrobenzoic acid (DTNB) showed that they were all oxidised during the initial phase of inhibition of dopachrome formation. It is concluded that the first phase of inhibition involves oxidation of DTT by an intermediate between l-DOPA and dopachrome. The second phase of inhibition also appeared to require -SH groups initially, since trans-4,5-dihydroxy-1,2-dithiane (oxidized DTT) caused very little inhibition at all.  相似文献

A survey was made over 5 years of the incidence of contamination by Phoma foveata and Polyscytalum pustulans on 3-year-old virus-tested stem cutting (VTSC) stocks assessed on three occasions during the storage season, and on their daughter tubers at five farms. The contamination occurring on plants grown in small plots from stem-cutting/microplant tubers was also assessed at the farms.
Fungicide treatment soon after harvest reduced the incidence of both pathogens, especially P. pustulans . Tuber contamination was similar for samples collected after fungicide treatment and before planting. The incidence of contamination on daughter tubers, relative to that on the untreated mother tubers, was less with P. foveata but was similar or greater with P. pustulans . Contamination by both pathogens varied between farms and was reflected in the degree of contamination occurring on daughter tubers from stem-cutting/microplant tubers.
The incidence of P. foveata on stems, soil and tubers of seed stocks classified at Foundation Stock grade from various regions of Scotland was assessed over 2 years. The incidence of tuber contamination was correlated ( P < 0.05) with the amount of tuber-borne soil contamination for all comparisons, but was correlated with stem inoculum in only 7 out of 13 comparisons. The largest amounts of stem inoculum of P. foveata were found in stocks in Aberdeenshire.
These results are discussed in relation to the spread of both pathogens to healthy VTSC stocks.  相似文献

The changes in the components of the ascorbateglutathione systemduring the storage of potato (Solanum tuberosum L. cv. Spunta)tubers for 40 weeks at both 3C and 9C were studied in relationto lipid peroxidation. The level of malondialdehyde was foundto be higher at 3C than at 9C throughout storage. Thus, lipidperoxidation, which is the main cause of membrane deterioration,was accelerated at the lower temperature. Catalase activityincreased throughout storage independently of temperature. Theascorbate content of tubers decreased during storage both at3C and at 9C, as in other ageing processes. However, ascorbateperoxidase activity reached a maximum after about 8 weeks ofstorage, then declined at 9C, but held a higher level at 3C.The dehydroascorbic content also reached a maximum after about8 weeks and was significantly higher in tubers stored at 3C.These findings indicate a greater utilization of ascorbate byascorbate peroxidase at the lower temperature. Ascorbate freeradical reductase, dehydroascorbate reductase and glutathionereductase, the enzymes involved in the regeneration of ascorbate,were not affected by temperature and remained quite unchangedthroughout storage. It can be concluded that the ascorbate systemis involved in the scavenging of the free-radicals responsiblefor lipid peroxidation in stored potato tubers, at least atlow temperatures and in the first period of storage. Fusarium avenaceum is reported for the first time as a cause of rotting of potato tubers in Britain. The progress of rotting in tubers infected with F. avenaceum has been compared with dry rot due to F. caeruleum in the laboratory, clamp and potato store. Of the four varieties, Majestic, King Edward, Doon Star and Arran Pilot, tested for susceptibility, King Edward was the most susceptible to F. avenaceum and Doon Star to F. caeruleum.
Optimum temperatures for growth on potato-dextrose agar were 20-25 C. for F. avenaceum and 20 C. for F. caeruleum ; maximum temperatures were > 30 and 30 C. respectively. For infection of wounded potato tubers, cardinal temperatures for F. avenaceum were similar to those on agar, but for F. caeruleum the optimum was 15 C. and the maximum 25 C. The optimum temperature for rotting tended, with both species, to be higher in the more susceptible potato varieties. At low temperatures F. caeruleum caused quicker rotting than did F. avenaceum , even though its rate of growth on agar was scarcely more than half that of the latter.
High humidity favoured rotting especially by F. avenaceum; F. caeruleum was more tolerant of relatively low humidity. Both species caused quicker rotting in the clamp than in store, even though there was no appreciable difference in mean temperature between the clamp and the store. This was attributed to the higher atmospheric humidity in the clamp.  相似文献

In studies on the infection of potato cultivars with different field isolates of Phoma exigua var. foveata, cultivar × isolate interactions, although sufficiently large to influence the relative pathogenicities of isolates on different cultivars, were not substantial enough to affect cultivar ranking order. Cultivar rank was markedly influenced by pathogen isolate only when both field and culture collection isolates were compared. This suggests that the complications of cultivar × isolate interactions can be avoided in cultivar screening tests by the use of recent field isolates of high pathogenicity. It was found that cultivar was considerably more important than isolate in determining gangrene lesion size. The need to consider both lesion diameter and depth when estimating rot size following point inoculation of different cultivars was confirmed.  相似文献

To change the hexose-to-sucrose ratio within phloem cells, yeast-derived cytosolic invertase was expressed in transgenic potato (Solanum tuberosum cv. Desirée) plants under control of the rolC promoter. Vascular tissue specific expression of the transgene was verified by histochemical detection of invertase activity in tuber cross-sections. Vegetative growth and tuber yield of transgenic plants was unaltered as compared to wild-type plants. However, the sprout growth of stored tubers was much delayed, indicating impaired phloem-transport of sucrose towards the developing bud. Biochemical analysis of growing tubers revealed that, in contrast to sucrose levels, which rapidly declined in growing invertase-expressing tubers, hexose and starch levels remained unchanged as compared to wild-type controls. During storage, sucrose and starch content declined in wild-type tubers, whereas glucose and fructose levels remained unchanged. A similar response was found in transgenic tubers with the exception that starch degradation was accelerated and fructose levels increased slightly. Furthermore, changes in carbohydrate metabolism were accompanied by an elevated level of phosphorylated intermediates, and a stimulated rate of respiration. Considering that sucrose breakdown was restricted to phloem cells it is concluded that, in response to phloem-associated sucrose depletion or hexose elevation, starch degradation and respiration is triggered in parenchyma cells. To study further whether elevated hexose and/or hexose-phosphates or decreased sucrose levels are responsible for the metabolic changes observed, sucrose content was decreased by tuber-specific expression of a bacterial sucrose isomerase. Sucrose isomerase catalyses the reversible conversion of sucrose into palatinose, which is not further metabolizable by plant cells. Tubers harvested from these plants were found to accumulate high levels of palatinose at the expense of sucrose. In addition, starch content decreased slightly, while hexose levels remained unaltered, compared with the wild-type controls. Similar to low sucrose-containing invertase tubers, respiration and starch breakdown were found to be accelerated during storage in palatinose-accumulating potato tubers. In contrast to invertase transgenics, however, no accumulation of phosphorylated intermediates was observed. Therefore, it is concluded that sucrose depletion rather than increased hexose metabolism triggers reserve mobilization and respiration in stored potato tubers.  相似文献

When soil was maintained at a mean water potential (Slatyer & Taylor, 1960) of the order of -80 J kg-1 at 25 cm depth throughout the growing period, much infection of potato tubers by Streptomyces scabies occurred, but when soil was irrigated to maintain it at potentials greater than - 13 J kg-1 at 10 cm depth infection was negligible. Until about 5 weeks after initiation, tubers were very susceptible to infection; irrigation during this period reduced scab considerably, but subsequent irrigation reduced it only slightly. At the low water potential, actinomycete populations on lenticels were high and bacterial populations low; these effects were reversed at the high water potential. Since also actinomycetes were more frequently isolated in the absence than in the presence of bacteria it was deduced that there was an interaction between these two groups. It is suggested that irrigation may decrease the population of S. scabies in tuber lenticels by increasing populations of bacteria antagonistic to it.  相似文献

Potato tubers were subjected to cobalt-60 gamma-ray irradiationand stored at room temperature for from 10 days to 4 months.Effects of this irradiation on metabolic changes in potato tubersin response to cutting were investigated. The quantities ofpolyphenols, such as chlorogenic acid and isochlorogenic acid,in the tissue increased as compared with the non-irradiatedsample. Although the polyphenol content and activities of o-diphenoloxidase, peroxidase and phenylalanine ammonialyase increasedafter cutting, increases were lower in the irradiated sample.On the other hand, in the dose range between 5,000 and 12,400rad, the irradiated sample showed a larger defense action againstinfection by the black rot fungus, Ceratocystis fimbriata thandid the non-irradiated sample.
