Abstract:  Eggs of the potato tuber moth (PTM) Phthorimaea operculella (Zeller) obtained from 150 or 300 Gy-irradiated moths, either inbred or out-crossed to untreated counterparts, were offered as host material to three generalist Trichogramma species; Trichogramma cacoeciae , Trichogramma evanescens , and Trichogramma principium , to determine the ability of these egg parasitoids to develop in F1 sterile eggs. All eggs laid by the different parental crosses at 150 or 300 Gy, were acceptable and suitable for Trichogramma development. However, in no choice and paired comparison tests, eggs from 300 Gy-irradiated parents were significantly less preferred compared with eggs laid by non-irradiated moths. No intra-specific differences for PTM sterile eggs were recorded among the tested Trichogramma species. The mean number of wasps emerging from each egg types was not affected by an interaction between parental crosses and applied doses. Overall, this study demonstrates that release of irradiated and non-irradiated moths at a 5 : 1 over-flooding ratio and Trichogramma over potatoes in small plexiglass boxes, is complementary and more effective in reducing PTM F1-emerged progeny than either technique used alone. The results provide baseline data for developing an integrated control approach by using inherited sterility technique in conjunction with Trichogramma for PTM population suppression.  相似文献

Aims:  To assess the contribution of ozone to lethality of Salmonella enterica serovar Enteritidis in experimentally inoculated whole shell eggs that are sequentially treated with heat and gaseous ozone in pilot-scale equipment.
Methods and Results:  Whole shell eggs were inoculated with small populations of Salmonella Enteritidis (8·5 × 104–2·4 × 105 CFU per egg) near the egg vitelline membrane. Eggs were subjected to immersion heating (57°C for 21 min), ozone treatment (vacuum at 67·5 kPa, followed by ozonation at a maximum concentration of approx. 140 g ozone m−3 and 184–198 kPa for 40 min) or a combination of both treatments. Survivors were detected after an enrichment process or enumerated using modified most probable number technique. Ozone, heat and combination treatments inactivated 0·11, 3·1 and 4·2 log Salmonella Enteritidis per egg, respectively.
Conclusions:  Sequential application of heat and gaseous ozone was significantly more effective than either heat or ozone alone. The demonstrated synergy between these treatment steps should produce safer shell eggs than the heat treatment alone.
Significance and Impact of the Study:  Shell eggs are the most common vehicle for human infection by Salmonella Enteritidis. Many cases of egg-related salmonellosis are reported annually despite efforts to reduce contamination, including thermal pasteurization of shell eggs and egg products. Treatment with ozone-based combination should produce shell eggs safer than those treated with heat alone.  相似文献

Gastrointestinal (GI) parasitic infections of beef cattle were investigated in 15 farms from 3 different regions of Greece, characterized by temperate Mediterranean climate, during two years. A total of 262 faecal samples were collected. Helminth eggs were detected in 42 (16%) samples. Strongyle-type eggs were found in 28 (10.7%) samples, Strongyloides spp. and Toxocara spp. eggs in 8 (3.1%) samples each, and Capillaria spp. and Moniezia spp. eggs in 1 (0.4%) sample each. Coccidian oocysts were found in 123 (46.9%) samples. A logistic regression model showed that the age of animals, the number of employees, and the region of animals were associated with coccidian infections. There was almost a four-fold increase in the risk of coccidian infections in calves less than 12 months old compared with animals that were more than 36 months old (p < 0.05). The risk of coccidian infections was 0.07, 0.18 and 0.27-fold lower for farms with 1, 2 and 3 employees respectively compared to farms with 4 employees; farms with less than 4 employees had a protective effect (p < 0.05 in all cases). Almost a four-fold increased risk of coccidian infections was shown for animals from Epirus compared with those from Thessaly (p < 0.05). These findings are useful to formulate appropriate control strategies for coccidian parasites of beef cattle in Greece and other areas with similar climatic conditions.  相似文献

Many ecological studies use stress hormones to assess the condition, health or disturbance levels of wild organisms. Common blood sampling protocols for this research involve trapping individuals and taking blood within three minutes to obtain a “baseline” for analysis of stress hormones (“conventional method”). In some situations it may be difficult to get an accurate measure of baseline values; therefore, alternative sampling techniques may be preferable. We compared corticosterone levels in samples taken via a newly developed, minimally invasive blood sampling technique with corticosterone levels in blood taken via the conventional method. We collected samples from incubating adult common terns Sterna hirundo via blood sucking bugs (Heteroptera, Triatominae) contained in “dummy eggs” (“bug method”) and compared measured corticosterone concentrations to concentrations in blood taken from the same birds using the conventional method. We found no significant differences in mean or variance of baseline corticosterone levels between samples collected via the different methods. This suggests that the bug method offers a viable alternative for hormone sampling.  相似文献

In order to understand the molecular mechanism of development during early embryogenesis in diapause and non-diapause of the silkworm, mRNA from diapause and non-diapause eggs was compared using the differential display technique. We cloned the full length of a cDNA encoding a novel RNA helicase-like (RHL) protein by the RACE method using a cDNA fragment which was one of the specific cDNAs in the non-diapause eggs. A BLAST search using the predicted amino acid sequence of RHL revealed a low homology (21–25% identity of its partial length) with that of the DEAD-box RNA helicase. Gene expression of the RHL gene of the diapause and non-diapause eggs was investigated by RT-PCR until 60 h after oviposition. Amplified RHL cDNA was observed through all the stages in the non-diapause eggs, while in the diapause eggs, cDNA was found in eggs 0–12 h after oviposition but disappeared 24–60 h after oviposition. When the diapause eggs were activated by HCl treatment after chilling at 4 °C for 6 days from 48 h after oviposition (artificial diapause termination), cDNA was observed from 12 h after HCl treatment. We also investigated the immunohistochemical distribution and localization of RHL in non-diapause eggs using anti-recombinant His-tag RHL antiserum. RHL was distributed in blastoderm cells and yolk cells and was localized in the nucleus and the cytosol of yolk cells. These data suggest that RHL has an important role in the early embryo of the silkworm.  相似文献

ABSTRACT Collecting blood from neonatal or prehatched chicks is sometimes necessary, requiring specialized blood collection techniques and skills. However, such sampling can be difficult and could potentially have adverse effects. I developed a method for collecting blood from chicks still in an egg. The technique involves enlarging the pipping hole, removing the chick's head to take blood from a jugular vein, and returning the chick into its original position in the egg to complete hatching back in the nest. I used this technique on Crested Tern (Thalasseus bergii) chicks (N= 13), with no apparent adverse effects to growth or short‐term survival. The amount of blood extracted (0.1–0.15 ml) was sufficient for a blood smear, hematocrit, and total white blood cell count. This method should be useful for eggs as small as 40 mm in length; eggs smaller than this would likely not be good candidates for this technique because eggshells may be too thin to safely handle. This technique may be useful for investigators studying the immunological and endocrinological transition between embryos and hatchlings.  相似文献

Abstract.  Eggs laid by adult female Dianemobius nigrofasciatus , reared under long-day (LD 16 : 8 h, 25 °C) or short-day (LD 12 : 12 h, 25 °C) conditions from the nymphal stage, are kept at several constant temperatures. At 22.5–30.0 °C, eggs laid by long-day adults show lower incidences of diapause than those laid by short-day adults. In both eggs laid by adults under long-day conditions and those under short-day conditions, the higher the temperature at which the eggs are kept, the lower the incidence of diapause. When eggs of long-day adults are exposed to a low-temperature pulse (10 °C, 24 h) on the day of deposition (day 0), the incidence of diapause increases. The low-temperature pulse on day 1 does not increase the incidence of diapause. By contrast, when the eggs of short-day adults are exposed to a high-temperature pulse (35 °C, 24 h) on day 0 or day 1, the incidence of diapause decreases. The temperature pulses on day 0 are more effective at diapause prevention. Staining of diapause eggs by the Feulgen–Rossenbeck method shows that the eggs enter diapause at the blastoderm stage, which is on day 1 or day 2 at 25 °C. The exposure of adults to long days and higher temperatures prevents the eggs from entering diapause. In D. nigrofasciatus , embryonic diapause is controlled by maternal effects, adult photoperiod and temperature, and egg temperature before or at diapause.  相似文献

Abstract.  1.  This study investigated the interaction between eggs of the lizard Mabuya longicaudata Hallowell and two species of ant: a lizard egg commensalist ( Paratrechina longicornis Latreille), and an egg predator ( Pheidole taivanensis Forel). The ecological interaction between P. longicornis and lizard eggs was tested, and it was predicted that when the interaction was removed, lizard eggs would be attacked by P. taivanensis .
2. Field observations showed that both ant species actively searched for lizard eggs, and that P. taivanensis typically found new lizard nests earlier than P. longicornis did. Left undisturbed, P. taivanensis predation dramatically reduced lizard egg survival. While P. longicornis usually found nests later, they were able to displace P. taivanensis . As a result, proportional egg survival was higher in nests with P. longicornis (0.95 ± 0.04 eggs) compared to nests without either ant species (0.65 ± 0.09 eggs), or nests occupied by P. taivanensis (0.07 ± 0.02 eggs).
3. When P. longicornis ants were experimentally excluded from lizard nests, the proportion of eggs surviving significantly decreased because of increased P. taivanensis predation. Paratrechina longicornis benefits from water that condenses on the eggs. When this resource disappears P. longicornis abandons the nest, leading to predation of the eggs by P. taivanensis .  相似文献

Abstract.  1. Ladybird eggs are defended chemically against intra-guild predation, and are unsuitable to varying degrees as food for other ladybirds.
2. Ladybird eggs [ Coccinella septempunctata (Cs) and Harmonia axyridis (Ha); Coleoptera: Coccinellidae] from local Japanese versus North American populations were compared as food for two ladybirds (Cs and Ha) co-occurring in Japan.
3. Larvae of Cs exhibited high mortality, slow development, and reduced egg consumption and weight gain when they fed on Ha versus conspecific eggs, especially when Ha eggs were from the local (Japanese) population versus a distant (North American) population of Ha.
4. Larvae of Ha survived equally well on a diet of Cs or conspecific eggs, but developed more slowly and gained less weight on Cs eggs. Ha larvae were more reluctant to eat eggs from the local (Japanese) population of Cs than eggs from the distant (North American) population of Cs; however, other measures of performance did not differ significantly.
5. These results indicate greater egg chemical defence of Ha eggs against Cs larvae than vice versa, as expected from field observations of greater temporal overlap between Cs larvae and Ha eggs than between Ha larvae and Cs eggs. Furthermore, results also indicate that local populations of eggs are better defended than eggs from elsewhere against locally occurring intra-guild predators. Thus, it appears that the effectiveness of chemical defence of ladybird eggs reflects the degree to which specific pairings of ladybirds have the potential to interact in nature through egg predation.  相似文献

We present a comprehensive protocol for extracting DNA from egg membranes and other internal debris recovered from the interior of blown museum bird eggs. A variety of commercially available DNA extraction methods were found to be applicable. DNA sequencing of polymerase chain reaction (PCR) products for a 176‐bp fragment of mitochondrial DNA was successful for most egg samples (> 78%) even though the amount of DNA extracted (mean = 14.71 ± 4.55 ng/µL) was significantly less than that obtained for bird skin samples (mean = 67.88 ± 4.77 ng/µL). For PCR and sequencing of snipe (Gallinago) DNA, we provide eight new primers for the ‘DNA barcode’ region of COI mtDNA. In various combinations, the primers target a range of PCR products sized from 72 bp to the full ‘barcode’ of 751 bp. Not all possible combinations were tested with archive snipe DNA, but we found a significantly better success rate of PCR amplification for a shorter 176‐bp target compared with a larger 288‐bp fragment (67% vs. 39%). Finally, we explored the feasibility of whole genome amplification (WGA) for extending the use of archive DNA in PCR and sequencing applications. Of two WGA approaches, a PCR‐based method was found to be able to amplify whole genomic DNA from archive skins and eggs from museum bird collections. After WGA, significantly more archive egg samples produced visible PCR products on agarose (56.9% before WGA vs. 79.0% after WGA). However, overall sequencing success did not improve significantly (78.8% compared with 83.0%).  相似文献

ABSTRACT.   Egg volumes are most often estimated using a mathematical model that incorporates length and width measurements and a species-specific shape variable. Although adequate in many respects, this technique does not account for intraspecific variation in egg shape. We developed a computer-automated technique that uses calibrated digital photographs to render precise measurements of several egg-size parameters including length, width, volume, and surface area. The system extracts egg outlines from photographs, and divides each egg into latitudinal slices that are subsequently regarded as simple geometric shapes (cylinders or cone frustra) with volumes and surface areas that can be summed to generate size parameters for the entire egg. We tested this technique using 491 eggs from Florida Scrub-Jay ( Aphelocoma coerulescens ) nests and compared the resulting egg volumes with volumes calculated using the preeminent method of estimating volume from linear measurements. Our method was highly accurate, and differences between the volumes from our method and the alternative method were strongly associated with variation in egg shape. Advantages of our technique include decreased handling of eggs and increased accuracy. Summary 1. We identified temperature and photoperiod conditions under which the hatching of 45 cladoceran species could be elicited. Identification of appropriate hatching cues is of primary importance for the exploration of the ties between active and diapausing stages.
2. Incubation temperature affected the hatching success of resting eggs isolated from Danish, Belgian/Dutch and Spanish sediments. In general, most hatchlings and species were retrieved at 15 °C. Danish and Belgian/Dutch resting eggs hatched more successfully under a long day photoperiod than in continuous illumination.
3. Most species could be retrieved after incubation of resting eggs isolated from a limited amount of sediment (0.4 kg) under a single, well chosen combination of temperature and photoperiod. Processing additional sediment samples under seven more incubation regimes only allowed detection of 21% (Spain) to 34% (Denmark) more species.
4. The incubation period for resting eggs to hatch was strongly influenced by incubation temperature. Our results show that hatching experiments aimed at assessing cladoceran species richness and conducted at 15 °C should be continued for a period of at least 2 weeks, after which a random subset of hatchlings (e.g. n  = 100) can be selected from the total hatchling assemblage.  相似文献

1. The effect of temperature on embryonic development was compared in four populations, two bisexual and two unisexual, of Ephoron shigae , including one each near the northern and southern periphery of the species range in Japan.
2. Eggs from every population were chilled at 4, 8 or 12 °C for diapause development after 50 days at 20 °C for pre-diapause development (experiment I). Some eggs hatched during chilling at 8 °C or 12 °C, whereas no eggs hatched at 4 °C. The rate of hatching in a given condition of chilling was higher for the eggs from warmer winter environments.
3. Chilling at 4 or 8 °C effectively facilitated diapause development. Chilling at 12 °C was, in general, not so effective, but relatively effective for the eggs from warmer winter environments.
4. Eggs were incubated at 8, 12, 15 or 20 °C after chilling at 4 °C to examine the effect of temperature on post-diapause development (experiment II). The eggs incubated at higher temperature after chilling hatched quicker and more synchronously and had higher hatching success.
5. The relationship between temperature and the days required for hatching after chilling was well described by the power function. There was no significant difference in the slope of the regression lines (i.e. temperature dependency) among local populations. However, a longer time was required for hatching at a given temperature for the population from the colder winter environment.
6. There was no detectable difference in the observed intraspecific variations between unisexual and bisexual populations.  相似文献

The frequency of extreme events, such as cold spells, is expected to increase under global warming. Therefore, the ability of insects to survive rapid changes in temperature is an important aspect to investigate in current population ecology. The hemlock looper (HL), Lambdina fiscellaria (Guenée) (Lepidoptera: Geometridae), a defoliator of boreal balsam fir forests in eastern Canada, overwinters at the egg stage on tree trunks and branches where eggs can be exposed to very low subzero air temperatures. Using eggs from the island of Newfoundland (NL) and Quebec mainland (QC), we undertook field and laboratory experiments to determine: (1) their supercooling point (SCP) in mid‐January and mid‐February; (2) overwintering mortality; (3) cold tolerance to various combinations of subzero temperatures (?25, ?30, ?33, ?35, or ?37 °C) and exposure durations (2, 4, 8, 12, or 16 h); and (4) potential causes of death at subzero temperatures above the SCP. Regardless of population or sampling date, eggs supercooled on average at ?40.1 °C. In the field, 59% of eggs from either population that overwintered in Sainte‐Foy (QC) and Corner Brook (NL) hatched successfully, whereas none did in Armagh (QC) or Epaule (QC). In the laboratory, 50% of eggs survived after 4 h at ?34.4 °C or after 14 h at ?32.9 °C. In contrast, regardless of exposure duration, >50% of eggs hatched at temperatures ≥?33 °C, but <50% did so at ≤?35 °C, suggesting high pre‐freeze mortality. However, when eggs were attached to thermocouples and exposed to temperatures ranging from ?25 to ?37 °C for 16 h, 69% froze at temperatures of ?35 to ?37 °C, but only 2% did at ?25 or ?30 °C. Time to freeze decreased as subzero temperatures declined, and this was more evident in island eggs than in mainland eggs. Overall, eggs can freeze after a brief exposure to subzero temperatures higher than the standard SCP, and are thus highly vulnerable to cold spells.  相似文献

The effects of temperature on maintenance and termination of embryonic diapause were investigated in Jining (35.4°N, 116.6°E) and Sihong (33.5°N, 118.2°E) strains of the Chinese rice grasshopper, Oxya chinensis Thunberg (Orthoptera: Catantopidae). Eggs of both strains entered diapause when incubated at 30, 25, or 20 °C. Chilling at 8 °C had an evident effect on diapause termination and almost all eggs chilled for 60 days ended diapause development. Chilling of eggs at 8 °C for only 20 days failed to result in any hatching at 20 °C, suggesting that such level of chilling was not enough to induce diapause termination. However, the treatment combining incubation of eggs at 30 °C for varying lengths of time with subsequent incubation to 20 °C had a distinct effect on the completion of diapause of the eggs. The results indicate that there were two temperature optima, that is, low temperature (chilling) and high temperature, for diapause development in this grasshopper species. Incubation of chilled eggs at 20 °C for 5–15 days followed by further incubation at 25 °C reduced termination of diapause significantly compared with the eggs only chilled at 8 °C. Exposure of eggs chilled at 8 °C to a pulse of 25 °C from 1 to 7 days, separated by a 20-day interval at 8 °C, resulted in a decrease in the percentage of successfully hatched eggs as the length of the pulse of 25 °C increased. The results suggest that diapause intensity may be restored at moderately high temperatures. This reversible change in diapause intensity would play an important role in maintaining diapause before winter.  相似文献

The ability to monitor developing avian embryos and their associated vascular system via candling enables the application of important reproductive management techniques. Egg candling facilitates the confirmation of egg viability throughout the incubation process and identification of a precise position on a vein for the safe extraction of blood. Blood samples may then be analysed to retrieve vital health and genetic information to assist in conservation management. However, the thick or opaque egg shell characteristics of some avian species prevents the observation of egg contents using traditional candling methods, thus limiting management options. This paper tests a novel method of preparing thick-shelled or opaque eggs so that traditional egg candling and blood extraction methods may be applied. Eggs from captive emu (Dromaius novaehollandiae, Latham 1790) and southern cassowary (Casuarius casuarius johnsonii, Linnaeus 1758) were obtained, and partial fenestration was performed on two areas of shell either before incubation or at ⅓ of incubation. Hatchability and weight loss were examined as a measure of effect of the fenestration process on the developing embryo. Clear observation of vascular development was successful in 97% of viable fenestrated eggs, without affecting hatchability or weight loss. Blood samples were taken from developing embryos and DNA was successfully extracted for proof of concept of this new technique. The ability to observe vascular development and monitor the developing embryo in thick and opaque eggs will significantly improve both in situ and ex situ population management options such as in ovo sexing in species of concern.  相似文献

Background

As low pathogenic avian influenza viruses can mutate into high pathogenic viruses the Dutch poultry sector implemented a surveillance system for low pathogenic avian influenza (LPAI) based on blood samples. It has been suggested that egg yolk samples could be sampled instead of blood samples to survey egg layer farms. To support future decision making about AI surveillance economic criteria are important. Therefore a cost analysis is performed on systems that use either blood or eggs as sampled material.

Methodology/Principal Findings

The effectiveness of surveillance using egg or blood samples was evaluated using scenario tree models. Then an economic model was developed that calculates the total costs for eight surveillance systems that have equal effectiveness. The model considers costs for sampling, sample preparation, sample transport, testing, communication of test results and for the confirmation test on false positive results. The surveillance systems varied in sampled material (eggs or blood), sampling location (farm or packing station) and location of sample preparation (laboratory or packing station). It is shown that a hypothetical system in which eggs are sampled at the packing station and samples prepared in a laboratory had the lowest total costs (i.e. € 273,393) a year. Compared to this a hypothetical system in which eggs are sampled at the farm and samples prepared at a laboratory, and the currently implemented system in which blood is sampled at the farm and samples prepared at a laboratory have 6% and 39% higher costs respectively.

Conclusions/Significance

This study shows that surveillance for avian influenza on egg yolk samples can be done at lower costs than surveillance based on blood samples. The model can be used in future comparison of surveillance systems for different pathogens and hazards.  相似文献

Abstract  In this study we report the results from the first long-term (40 weeks) study of stick-insect fecundity and distribution under natural conditions of which we are aware. We used the number of eggs falling into 72 × 0.5 m2 traps to ask: 'Was egg production in Sipyloidea sp. uniform across the sample period'? and 'Was there evidence of host plant species preference or avoidance'? We collected a total of 213 Sipyloidea sp. eggs. The number of eggs caught per week was not uniform and an exponential decay model was the best-fit relationship between egg production and time, indicative of a steep decline from high to low (but continuous) egg production across the study period. Continuous egg production differs from other insect species in tropical areas that often show distinct seasonal differences between wet and dry seasons, timed to leaf production in host plants. The distribution of eggs within traps was aggregated, and more traps than expected from Poisson probabilities received no eggs, or six or more eggs. The concentration of eggs within particular traps was not related to the identity of canopy plant species, however. We suggest that continuous egg production in Sipyloidea sp. may be related to the wider range of plant species available as food resources for the polyphagous Sipyloidea , compared with other tropical insect species.  相似文献

Anthocoris nemorum L. and Anthocoris nemoralis Fabricius (Heteroptera: Anthocoridae) are important predators of insect pests in pome fruit. Females insert their eggs in leaf tissue. The females’ choice of oviposition site is important for the subsequent distribution of nymphs on host plants. Oviposition preference for apple and pear leaves was tested in the laboratory in four experiments (experiments 1–4). In three experiments it was tested whether simulated insect damage to leaves (experiments 5 and 6) or the presence of prey (experiment 7) influenced oviposition preference. The effect of the presence of prey was only tested for A. nemorum on apple leaves. There was a highly significant anthocorid species × plant interaction for the number of eggs laid on apple and pear leaves. Anthocoris nemorum laid more eggs on apple than on pear leaves, while A. nemoralis preferred pear. Anthocoris nemorum's preference for apple increased over the 6‐week period in which experiments 1–4 were performed, from 66% to 91% eggs laid on apple leaves. No change over time in preference was found for A. nemoralis. Across experiments 1–4, the majority of A. nemorum eggs were laid near leaf margins, whereas eggs of A. nemoralis were more commonly found in the leaf centre, 5 mm or more from the margin, with a highly significant leaf region × species interaction. There was no significant difference in preference for leaf side between A. nemorum and A. nemoralis, but there was a highly significant plant × leaf side × experiment interaction. Thus, more eggs were laid on the ventral than on the dorsal side of pear leaves in experiment 4, while significantly more eggs were laid on the dorsal side of apple leaves in experiments 3 and 4. Choice tests between damaged and healthy leaves showed that A. nemorum laid significantly more eggs on the damaged leaves, while A. nemoralis preferred healthy leaves. Anthocoris nemorum showed a near‐significant preference for ovipositing on leaves with eggs of Operophtera brumata (Lepidoptera: Geometridae). The oviposition preferences found correspond to the natural distribution of these predators in apple and pear orchards. The preference of A. nemorum for leaf margins, and of A. nemoralis for the leaf centre as an oviposition site, supports earlier observations. A preference for leaf side for oviposition site has not been reported earlier. Preference for damaged leaves could help A. nemorum to locate prey in a field situation.  相似文献

A modification of gDNA extraction was developed for the polymerase chain reaction (PCR) technique, intended for the detection and differentiation of Toxocara spp. eggs in soil or sediments. Sand samples from sandpits confirmed as being contaminated with Toxocara spp. eggs by the flotation technique were analysed by PCR. The use of proteinase K made it possible to obtain genomic DNA from the sample without needing to isolate eggs using flotation or to inactivate PCR inhibitors present in the sand. Specific primers in the PCR reaction allowed discrimination between T. canis and T. cati eggs. The modification simplified the procedure, thanks to eliminating the step of gDNA isolation from eggs, which is both laborious and difficult.  相似文献
