Abstract Respiration and photosynthesis in zoospores of Enteromorpha intestinalis (L.) Link and Ulothrix pseudoflacca Wille showed greater sensitivity to inhibition by triphenyltin chloride than in vegetative tissue. Photosynthesis in zoospores and vegetative tissue of U. pseudoflacca was more resistant to triphenyltin chloride inhibition than in zoospores and vegetative tissue of E. intestinalis. Respiration in zoospores and vegetative tissue of both species was equally sensitive to triphenyltin chloride.  相似文献

Abstract— The uptake of [14C]GABA, [14C]taurine, [3H] β -alanine and [14C]dopamine was compared in slices of rat cerebral cortex of three different sizes (0.1 × 0.1 × 2 mm, 0.2 × 0.2 × 2 mm and 0.4 × 0.4 × 2 mm prepared with a mechanical tissue chopper). [14C]Taurine and [3H] β -alanine uptake increased whereas [14C]GABA uptake decreased with increasing slice size. [14C]Dopamine uptake was optimal in 0.2 × 0.2 × 2 mm slices. Increasing slice size was shown to decrease inhibition of [3H] β -alanine and [14C]GABA uptake by l -2,4-diaminobutyric acid. Lactate dehydrogenase activity increased with increasing slice size indicating decreased tissue damage or increased cellular integrity. The possibility that varying slice size can be used to distinguish between neuronal and glial uptake is discussed. It is suggested that taurine uptake in the cerebral cortex is predominantly glial.  相似文献

Abstract— Fifty-two substances were tested as inhibitors of the uptake of [3H]GABA in slices of rat cerebral cortex. Among GABA analogues tested, only the 2-fluoro, 3-hydroxy and 2-amino compounds had affinities for the uptake mechanism comparable to that of GABA. [3H]GABA uptake was also potently inhibited by p -chloromercuriphenylsulphonate, N -ethylmaleimide, chlorpromazine and haloperidol. No inhibitors were found to act in a competitive manner with respect to GABA. [3H]GABA uptake was also examined in homogenates of cerebral cortex and other regions of CNS. There was a rapid uptake of [3H]GABA into particles when homogenate samples were incubated with the labelled amino acid; this uptake had similar kinetic properties and inhibitor sensitivity to that observed in slices of intact tissue. Density gradient centrifugation experiments indicated that the particles responsible for the uptake of [3H]GABA in homogenates were probably synaptosomes. Uptake of [3H]GABA also occurred in slices and homogenates of rat spinal cord, and evidence was obtained by the simultaneous labelling of homogenates with [14C]glycine and [3H]GABA that these two amino acids were taken up by different nerve terminals in this region.  相似文献

Abstract— At 25°C the accumulation of [3H] dl -2,4-diaminobutyric acid (DABA) into small rat cortical slices was linear with time and a tissue: medium ratio of 35:1 was attained after 60 min. At 37°C the uptake was no longer linear and the tissue: medium ratio at 60 min was 66:1. Uptake was unaffected by the addition of 10 μ m -AOAA and dependent on the presence of Na+ in the incubation media. The uptake was shown to have a high affinity component with a K m of 20.7 μ m and a V max of 28.6 nmol/g/min. IC50's for the inhibition of [3H]DABA uptake by dl -DABA, l -DABA and GABA were 80, 40 and 17 μ m respectively. Two m m β -alanine, however, caused less than 13% inhibition of [3H]DABA uptake. Electron microscopic autoradiographs showed the [3H]DABA to be accumulated by 22% of the identifiable nerve terminals and, after 14 days exposure, the density of silver grains over nerve terminals was 36–38 times higher than that over the rest of the electron micrograph. On the other hand, [3H]DABA was not taken up into rat sensory ganglia and light level autoradiography showed the small amount of [3H]DABA accumulated by the ganglia to be evenly distributed throughout the tissue. Both electrical stimulation for 30 s and exposure of the tissue to a medium containing 47 m m -K+ for 2 min caused a marked increase in the efflux of [3H]DABA from the tissue. Both these effects were abolished by a reduction in Ca2+ concentration and an increase in the Mg2+ concentration of the superfusing medium. These results suggest that l -DABA acts as a 'false transmitter' for the neuronal uptake, storage and release of GABA.  相似文献

Abstract: The accumulation of [3H]xylamine ([ring-G-3H]- N -2'-chloroethyl- N -ethyl-2-methylbenzylamine; [3H]XYL) by rabbit thoracic aorta and rat vas deferens was studied in vitro . [3H]XYL uptake in both tissues saturated at the same concentrations and displayed the same time course as that for maximal inhibition of [3H]noradrenaline ([3H]NA) accumulation by XYL. Hydrolysis of [3H]XYL or coincubation with Na2S2O3 reduced tissue accumulation of tritium by 80%. In aorta and vas deferens, about 45% and 65%, respectively, of the total [3H]XYL accumulation was blocked by 100 μmol/L desmethylimipramine (DMI), l -NA, or bretylium. In the absence of sodium ion, the uptake of [3H]XYL was reduced by approximately these same amounts. In both tissues nearly 70% of the [3H]XYL taken up was protein bound when measured as trichloroacetic acid-precipitated tritium. Of this radioactivity, the same proportion was sensitive to 100 μmol/L DMI or the absence of sodium ion as found for total tissue accumulation of [3H]XYL. Hydrolysis of [3H]XYL prior to incubation with vas deferens reduced the protein-bound tritium by more than 95%. The studies indicate that [3H]XYL interacts with NA uptake carrier in both rabbit aorta and rat vas deferens and that a substantial portion of the resulting protein-bound radioactivity is carrier-dependent.  相似文献

Abstract: The effect of melatonin on [3H]glutamate uptake and release in the golden hamster retina was studied. In retinas excised in the middle of the dark phase, i.e., at 2400 h, melatonin (0.1 and 10 n M ) significantly increased [3H]glutamate uptake, and this effect persisted in a Ca2+-free medium. On the other hand, melatonin significantly increased [3H]glutamate release in retinas excised at 2400 h, but this effect was Ca2+ sensitive. Melatonin significantly increased 45Ca2+ uptake by a crude synaptosomal fraction from retinas of hamsters killed at 2400 h. In retinas excised at 1200 h, melatonin had no effect on [3H]glutamate uptake, [3H]glutamate release, or 45Ca2+ uptake at any concentration tested. Cyclic GMP analogues, i.e., 8-bromoguanosine 3',5'-cyclic monophosphate and 2'- O -dibutyrylguanosine 3',5'-cyclic monophosphate, significantly increased [3H]glutamate uptake, [3H]glutamate release, and 45Ca2+ uptake by tissue removed at 1200 and 2400 h, suggesting that the effects of melatonin could correlate with a previously described effect of melatonin on cyclic GMP levels in the golden hamster retina. Taking into account the key role of glutamate in visual mechanisms, the results suggest the participation of melatonin in retinal physiology.  相似文献

The regulation by phytochrome of stem elongation in light-grown plants depends on gibberellins (GAs). To investigate whether this is mediated by a change in GA metabolism, the effect of the GA biosynthesis inhibitor LAB 198 999 (an acylcyclohexadione derivative) on the end-of-day far-red (FR) response in cowpea ( Vigna sinensis L.) epicotyl explants has been investigated. Growth of epicotyl explants of light-grown seedlings was enhanced when treated with far-red light before incubation in the dark (end-of-day FR effect). Low doses of LAB 198 999 (0.05 and 0.5 μg explant−1) reduced the effect of FR, whereas 5 to 50 μg explant−1 stimulated elongation of both red light (R)- and FR-treated epicotyl explants while nullifying the differences between R and FR treatments. In paclobutrazol-treated epicotyl explants, FR enhanced the response to applied GA1 and GA20, whereas LAB 198 999 increased the activity of GA1 and decreased that of GA20, [3H]Gibberellin A1, injected into the basal part of the epicotyl, was transported and metabolized mainly to [3H]GA8 in the apical 20 mm of the epicotyl. The conversion of [3H]GA1 to [3H]GA8 was dramatically reduced by both end-of-day FR treatments and LAB 198 999 applications. In addition, both treatments enhanced epicotyl elongation. It is proposed that the regulation of cowpea epicotyl growth by phytocrome is mediated, at least partially, by modifying GA1 degradation.  相似文献

Abstract: The posterior stomach nerve (PSN) is a crustacean sensory nerve containing about 60 cholinergic neurons, which are devoid of synaptic interactions. Kinetic analysis shows that the PSN takes up [3H]choline by both low-affinity ( K m= 163 μM) and high-affinity (Na+-dependent) ( K m= 1 μM) processes. The capacity of the high-affinity system is only about 1% that of the low-affinity system. The high-affinity system is not tightly coupled to acetylcholine (ACh) synthesis, and it appears that both ACh and phosphorylcholine are formed from an intracellular pool of choline, which is fed by both uptake systems. There are differences in the rates of [3H]choline uptake and 3H metabolite accumulation between regions of the PSN that contain neuronal cell bodies and those that do not. These differences may arise from differences in the relative proportion of neuronal to nonneuronal tissue in each nerve region.  相似文献

Abstract: Recent reports suggest that nitric oxide (NO) may contribute to several neurodegenerative diseases, e.g., focal cerebral ischemia, N -methyl- d -aspartate-mediated neurotoxicity, and experimental autoimmune encephalomyelitis. Accordingly, an understanding of the CNS transport processes of NO synthase (NOS) inhibitors has important therapeutic implications. The objective of the present study was to characterize the in vitro transport processes governing the uptake of l -[14C]arginine and the NOS inhibitor [14C]aminoguanidine in rat choroid plexus tissue. Consistent with previous reports, the uptake of l -[14C]arginine was mediated by both saturable and nonsaturable processes and was inhibited by the NOS inhibitors N G-methyl- l -arginine, N G-amino- l -arginine, and N 5-imidoethyl- l -ornithine. l -[14C]Arginine uptake was not inhibited by aminoguanidine or N G-nitro- l -arginine. Because aminoguanidine is an organic cation that bears some structural similarity to l -arginine, aminoguanidine might be transported by either an organic cation transporter or by the basic amino acid transporter governing arginine uptake. However, there was no evidence of a saturable uptake process for [14C]aminoguanidine in isolated rat choroid plexus, in contrast to that observed for l -[14C]arginine.  相似文献

Abstract— The characteristics of the uptake of l -[U-14C] glutamate into rat dorsal sensory ganglia were investigated. The uptake was mediated by two distinct kinetic systems, with apparent Km values of the order of 10−3 M (low affinity) and 10−5 m (high affinity). The high affinity uptake system was strongly dependent upon temperature and sodium ion concn, and was depressed by a number of metabolic inhibitors. Following uptake, [14C] glutamate was extensively metabolized, primarily to glutamine, although this was not so with cultured ganglia, where in addition to an increased uptake of [14C] glutamate, the specific radioactivity of glutamate was increased and that of glutamine decreased. The labelled substrates [U-14C]pyruvate and [U-14C] acetate were used to investigate this phenomenon and the results are discussed in relation to current knowledge of metabolic compartmentation in nervous tissue.  相似文献

In contrast to the light-promoted uptake by mesophyll cells, light slightly inhibited sucrose uptake by stripped leaf disks of Commelina benghalensis L. This phenomenon appeared to result from a light-promoted vein-associated release, as light stimulated photosynthate release from stripped disks and inhibited that from mesophyll cells. In the -presence of the resorption-blocker p -chloromercuriphenylsulfonic acid, (PCMBS) the release of preloaded [14C]-sugars (sucrose, glucose) and [14C]-amino acids (alanine, asparagine, proline, valine, α-aminoisobutyric acid) from stripped disks was doubled in the light. Illumination enhanced by 20 to 60% the release of endogenous leaf cell compounds (sucrose, H2PO-4, K+, Mg2+, Ca2+) from stripped disks in the presence of PCMBS. Light also increased the export of [14C]-assimilates from intact leaves by 20% after pulse-labelling with 14CO2. A model for loading is proposed, based on the differential light sensitivities of the plasma membranes in the mesophyll-to-sieve tube path.  相似文献

Abstract: We studied effects of Ca2+ in the incubation medium on [3H]dopamine ([3H]DA) uptake by rat striatal synaptosomes. Both the duration of the preincubation period with Ca2+ (0–30 min) and Ca2+ concentration (0–10 m M ) in Krebs-Ringer medium affected [3H]DA uptake by the synaptosomes. The increase was maximal at a concentration of 1 m M Ca2+ after a 10-min preincubation (2.4 times larger than the uptake measured without preincubation), which reflected an increase in V max of the [3H]DA uptake process. On the other hand, [3H]DA uptake decreased rapidly after addition of ionomycin in the presence of 1 m M Ca2+. The Ca2+-dependent enhancement of the uptake was still maintained after washing synaptosomes with Ca2+-free medium following preincubation with 1 m M Ca2+. Protein kinase C inhibitors did not affect apparently Ca2+-dependent enhancement of the uptake, whereas 1-[ N,O -bis(1,5-isoquinolinesulfonyl)- N -methyl- l -tyrosyl]-4-phenylpiperazine (KN-62; a Ca2+/calmodulin-dependent kinase II inhibitor) and wortmannin (a myosin light chain kinase inhibitor) significantly reduced it. Inhibitory effects of KN-62 and wortmannin appeared to be additive. N -(6-Aminohexyl)-5-chloro-1-naphthalenesulfonamide hydrochloride (W-7; a calmodulin antagonist) also remarkably inhibited the enhancement. These results suggest that Ca2+-dependent enhancement of [3H]DA uptake is mediated by activation of calmodulin-dependent protein kinases.  相似文献

Abstract: The characteristics of β-alanine transport at the blood-brain barrier were studied by using primary cultured bovine brain capillary endothelial cells. Kinetic analysis of the β-[3H]alanine transport indicated that the transporter for β-alanine functions with Kt of 25.3 ± 2.5 µ M and J max of 6.90 ± 0.48 nmol/30 min/mg of protein in the brain capillary endothelial cells. β-[3H]Alanine uptake is mediated by an active transporter, because metabolic inhibitors (2,4-dinitrophenol and NaN3) and low temperature reduced the uptake significantly. Furthermore, the uptake of β-[3H]alanine required Na+ and Cl in the external medium. Stoichiometric analysis of the transport demonstrated that two sodium ions and one chloride ion are associated with one β-alanine molecule. The Na+ and Cl-dependent uptake of β-[3H]alanine was stimulated by a valinomycin-induced inside-negative K+-diffusion potential. β-Amino acids (β-alanine, taurine, and hypotaurine) inhibited strongly the uptake of β-[3H]alanine, whereas α- and γ-amino acids had little or no inhibitory effect. In ATP-depleted cells, the uptake of β-[3H]alanine was stimulated by preloading of β-alanine or taurine but not l -leucine. These results show that β-alanine is taken up by brain capillary endothelial cells, via the secondary active transport mechanism that is common to β-amino acids.  相似文献

The effects of external K+, H+ and Ca2+ concentrations on the intracellular K+ concentration, [K+]i, and the K+-ATPase activity in 2-day-old mung bean roots [ Vigna mungo (L.) Hepper] were investigated. [K+]i, in mung bean roots was markedly decreased by external K+ or H+ stress and did not recover the initial value even after the stress was removed. This decrease in [K+]i, gradually disappeared with the addition of (Ca2+. Ca2+ may offset the harmful effects of ion stress. Ca2+ seems to have two effects on K+ transport; control of K+ permeability and activation of K+ uptake, although K+-ATPase activity was inhibited by Ca2+ concentrations higher than 10–4 M. We suggest that Ca2+ activates K+ uptake indirectly through the acidification of the cytoplasm.  相似文献

Abstract— The effect of amino-oxyacetic acid on the uptake of [3H]GABA by rat brain slices was studied. When added simultaneously with [3H]GABA, amino-oxyacetic acid had no significant effect on [3H]GABA uptake. However, preincubation of brain slices with amino-oxyacetic acid prior to addition of [3H]GABA produced inhibition of uptake, which increased with longer duration of preincubation. The inhibitory effect of amino-oxyacetic acid was maximal at 2 mM concentration and concentrations sufficient to inhibit significantly GABA:glutamate transaminase (10--6 M) had no effect on [3H]GABA uptake. D-Cycloserine and β-hydrazino-propionic acid also inhibited [3H]GABA uptake, but the amounts required were considerably in excess of those needed to inhibit GABA:glutamate transaminase. 4-Deoxypyridoxine inhibited [3H]GABA uptake, whether given in vivo or in vitro , and the inhibitory effect of amino-oxyacetic acid was reversed with pyridoxine. GABA transport appears to be dependent on pyridoxal phosphate and interference with this function of the vitamin is suggested as the basis for the inhibitory effect of amino-oxyacetic acid on [3H]GABA uptake.  相似文献

Abstract— [3H]Choline uptake has been measured in vivo in the rat hippocampus. Pharmacological agents and lesions which profoundly affect sodium-dependent, high-affinity [3H]choline uptake in vivo similarly affect [3H]choline uptake measured in vitro. Pentobarbital (65 mg/kg) and oxotremorine (0.75 mg kg) cause a decrease in [3H]choline uptake. Scopolamine (5 mg/kg) and iontophoretically applied extracellular potassium cause an increase in [3H]choline uptake. Septal lesions cause a decrease in [3H]choline uptake. Application of the general method may allow direct examination of presynaptic function and neural integration in the undisrupted living mammalian brain.  相似文献

The distinctive cortical uptake of the tracer 18F-FDDNP (2-(1-{6-[(2-fluoroethyl(methyl)amino]-2-naphthyl}ethylidene)malononitrile) in Alzheimer's disease (AD) is believed to be because of its binding to both neurofibrillary tangles (NFTs) and highly fibrillar senile plaques. We therefore investigated the binding of a tracer concentration of 3H-FDDNP to brain sections containing AD hallmark pathologies. Semi-adjacent sections were labelled with 3H-PIB (Pittsburgh compound-B, 2-[4'-(methylamino)phenyl]-6-hydroxybenzothiazole) and 14C-SB13 (4- N -methylamino-4'-hydroxystilbene) for comparison. Neocortical sections containing widespread senile plaques and cerebrovascular amyloid angiopathy, produced a sparse and weak labelling following incubation with 3H-FDDNP. Furthermore, in sections containing NFTs, there was no overt labelling of the pathology by 3H-FDDNP. In contrast, sections labelled with 3H-PIB displayed extensive labelling of diffuse plaques, classical plaques, cerebrovascular amyloid angiopathy and NFTs. 14C-SB13 produced a broadly similar binding pattern to PIB. Radioligand binding assays employing in vitro generated amyloid-β peptide fibrils demonstrated a ∼10-fold reduced affinity for 3H-FDDNP (85.0 ± 2.0 nM) compared with 3H-PIB (8.5 ± 1.3 nM). These data provide an alternative mechanistic explanation for the observed low cortical uptake of 18F-FDDNP in AD; in that the ligand is only weakly retained by the hallmark neuropathology because of its low affinity for amyloid structures.  相似文献

Embryonic shoots of Picea abies (L.) Karst, isolated from 10-year-old trees, were excised either with or without the crown. Various short-term uptake experiments (3, 6 and 24 h) and one long-term uptake experiment (4 weeks) were performed with these shoots to obtain information about the physiological role of the crown as translocation barrier for different substances. Transport through the embryonic shoots was followed in both acropetal and basipetal directions using radiolabelled substances supplied in an agarified Schenk and Hildebrandt medium. The medium was labelled with [14C]-IAA and/or [32P]-phosphate, or with [35S]-sulphate and 86Rb (as a tracer for K+). The experiments were conducted in light at 20°C, with the exception of one of the short-term experiments, which was carried out at 5°C to evaluate the connection between transport and metabolism. The main observation is that the crown in its collenchymatous stage of development acts as a selective barrier both acropetally and basipetally for transport of substances such as [14C]-IAA and [32P]-phosphate or their metabolized forms. This could explain why the embryonic shoot when cultured plus or minus its crown shows different growth and developmental patterns in vitro.  相似文献

Abstract: The synthesis of (2 S ,3 S ,4 S )-4-[1-(4-azidobenzamidomethyl)ethenyl]-2-carboxy-3-pyrrolidineacetic acid (ABCPA) is described. This novel kainic acid analogue, bearing a photolabile functionality on the isopropenyl side chain, was proven to be a good inhibitor of [3H]CNQX and [3H]kainic acid binding on chick cerebellar membranes. [3H]ABCPA was photoaffinity cross-linked on the membrane fraction of chick cerebellum. Electrophoretic analysis with sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed two major radioactive bands with apparent molecular masses of 45 and 33.5 kDa. [3H]ABCPA incorporation in both bands was completely blocked by 2 m M CNQX. When photoaffinity labeling was performed in the presence of 2 m M kainic acid, incorporation of [3H]ABCPA was blocked by ∼70% in the 45-kDa band and by 18% in the 33.5-kDa band. Incorporation of radioactivity in both bands was blocked by ∼30% with 10 m M glutamate.  相似文献

Abstract: In contrast to striatal membranes of adult rats, where high- ( K D1= 34 n M ) and low- ( K D2= 48,400 n M ) affinity binding sites for [3H]WIN 35,428 are present, in primary cultures of ventral mesencephalon neurons (CVMNs) only low-affinity binding sites were found ( K D= 336,000 n M ). The binding of [3H]WIN 35,428 in CVMNs prepared from rat embryos was reversible, saturable, and located in cytosol. Although dopamine (DA) uptake blockers inhibited [3H]DA uptake at nanomolar concentrations in CVMNs, the displacement of [3H]WIN 35,428 binding in CVMNs by DA uptake inhibitors required 100-8,000 times higher concentrations than were needed to displace [3H]WIN 35,428 binding in striatal membranes. Piperazine derivatives, e.g., GBR-12909, GBR-12935, and rimcazole, inhibited [3H]WIN 35,428 binding in CVMNs more effectively than did cocaine, WIN 35,428, mazindol, nomifensine, or benztropin. A positive correlation ( r = 0.779; p < 0.001) was found between drug affinities for the striatal membrane sites labeled by [3H]WIN 35,428 and their abilities to inhibit DA uptake in CVMNs, whereas no correlation existed between the IC50 values of drugs that inhibited [3H]WIN 35,428 binding and [3H]DA uptake in CVMNs. The cytosolic [3H]WIN 35,428 binding sites may be a piperazine acceptor and may not be involved in the regulation of the DA transporter.  相似文献
