Effect of paclobutrazol (PBZ) treatment on salinity tolerance of wheat (Triticum aestivum) was investigated on a salt-tolerant (Karchia-65) and salt-sensitive (Ghods) cultivars. Salinity significantly reduced the investigated growth parameters such as plant height, length and area of sixth leaf, root length, fresh and dry weight of shoot, roots and sixth leaf, water content (WC) of plant and seeds weight in the both cultivars. The negative effect of salinity in Ghods cultivar was more than Karchia cultivar. However, PBZ treatment reduced the growth in both cultivars, the differences in plant growth among various levels of NaCl decreased in PBZ-treated plants. Salt stress resulted in high accumulation of Na+ in the sixth leaf and roots in both cultivars, particularly in Ghods cultivar. Against Karchia cultivar, salt stress decreased the storage of K+, P and N in sixth leaf and roots in Ghods cultivar. In the both cultivars, PBZ treatment enhanced the K+, P and N contents in sixth leaf and roots by increasing salinity. Although PBZ treatment decreased the growth of plants, it improved the weight of seeds against stress damage. PBZ treatment reduced the accumulation of harmful Na+ ion in plant tissues while increased the K+, P and N contents. These observations suggest that PBZ treatment may increase tolerance by diminishing ionic imbalance caused by salt stress.  相似文献

为研究盐胁迫下小麦幼苗生长及Na+、K+的吸收和积累规律,以中国春、洲元9369和长武134等3种耐盐性不同小麦品种为材料,采用非损伤微测技术检测盐胁迫2 d后的根系K+离子流变化,并对植株体内的Na+、K+含量进行测定。结果表明:短期(2d)盐胁迫对小麦生长有抑制作用,且对根系的抑制大于地上部,耐盐品种下降幅度小于盐敏感品种。盐胁迫下,小麦根际的 K+大量外流,盐敏感品种中国春K+流速显著高于耐盐品种长武134,最高可达15倍。小麦幼苗地上部分和根系均表现为Na+积累增加,K+积累减少,Na+/K+比随盐浓度增加而上升。中国春限Na+能力显著低于长武134,Na+/K+则显著高于长武134。综上所述,盐胁迫下造成小麦组织器官中Na+/K+比上升的主要原因是根系K+大量外流和Na+的过量积累,耐盐性不同的小麦品种间差异显著,并认为根系对K+的保有能力可能是作物耐盐性评价的一个重要指标。  相似文献

Salinity tolerance levels and physiological changes were evaluated for twelve rice cultivars, including four white rice and eight black glutinous rice cultivars, during their seedling stage in response to salinity stress at 100 mM NaCl. All the rice cultivars evaluated showed an apparent decrease in growth characteristics and chlorophyll accumulation under salinity stress. By contrast an increase in proline, hydrogen peroxide, peroxidase (POX) activity and anthocyanins were observed for all cultivars. The K+/Na+ ratios evaluated for all rice cultivars were noted to be highly correlated with the salinity scores thus indicating that the K+/Na+ ratio serves as a reliable indicator of salt stress tolerance in rice. Principal component analysis (PCA) based on physiological salt tolerance indexes could clearly distinguish rice cultivars into 4 salt tolerance clusters. Noteworthy, in comparison to the salt-sensitive ones, rice cultivars that possessed higher degrees of salt tolerance displayed more enhanced activity of catalase (CAT), a smaller increase in anthocyanin, hydrogen peroxide and proline content but a smaller drop in the K+/Na+ ratio and chlorophyll accumulation.  相似文献

Quinoa is regarded as a highly salt tolerant halophyte crop, of great potential for cultivation on saline areas around the world. Fourteen quinoa genotypes of different geographical origin, differing in salinity tolerance, were grown under greenhouse conditions. Salinity treatment started on 10 day old seedlings. Six weeks after the treatment commenced, leaf sap Na and K content and osmolality, stomatal density, chlorophyll fluorescence characteristics, and xylem sap Na and K composition were measured. Responses to salinity differed greatly among the varieties. All cultivars had substantially increased K+ concentrations in the leaf sap, but the most tolerant cultivars had lower xylem Na+ content at the time of sampling. Most tolerant cultivars had lowest leaf sap osmolality. All varieties reduced stomata density when grown under saline conditions. All varieties clustered into two groups (includers and excluders) depending on their strategy of handling Na+ under saline conditions. Under control (non-saline) conditions, a strong positive correlation was observed between salinity tolerance and plants ability to accumulate Na+ in the shoot. Increased leaf sap K+, controlled Na+ loading to the xylem, and reduced stomata density are important physiological traits contributing to genotypic differences in salinity tolerance in quinoa, a halophyte species from Chenopodium family.  相似文献

The study aimed to investigate the effects of an AM fungus (Glomus intraradices Schenck and Smith) on mineral acquisition in fenugreek (Trigonella foenum-graecum) plants under different levels of salinity. Mycorrhizal (M) and non-mycorrhizal (NM) fenugreek plants were subjected to four levels of NaCl salinity (0, 50, 100, and 200 mM NaCl). Plant tissues were analyzed for different mineral nutrients. Leaf senescence (chlorophyll concentration and membrane permeability) and lipid peroxidation were also assessed. Under salt stress, M plants showed better growth, lower leaf senescence, and decreased lipid peroxidation as compared to NM plants. Salt stress adversely affected root nodulation and uptake of NPK. This effect was attenuated in mycorrhizal plants. Presence of the AM fungus prevented excess uptake of Na+ with increase in NaCl in the soil. It also imparted a regulatory effect on the translocation of Na+ ions to shoots thereby maintaining lower Na+ shoot:root ratios as compared to NM plants. Mycorrhizal colonization helped the host plant to overcome Na+-induced Ca2+ and K+ deficiencies. M plants maintained favorable K+:Na+, Ca2+:Na+, and Ca2+:Mg2+ ratios in their tissues. Concentrations of Cu, Fe, and Zn2+ decreased with increase in intensity of salinity stress. However, at each NaCl level, M plants had higher concentration of Cu, Fe, Mn2+, and Zn2+ as compared to NM plants. M plants showed reduced electrolyte leakage in leaves as compared to NM plants. The study suggests that AM fungi contribute to alleviation of salt stress by mitigation of NaCl-induced ionic imbalance thus maintaining a favorable nutrient profile and integrity of the plasma membrane.  相似文献

Rice is a salt-sensitive crop whose productivity is strongly reduced by salinity around the world. Plants growing in saline soils are subjected to the toxicity of specific ions such as sodium, which damage cell organelles and disrupt metabolism. Plants have evolved biochemical and molecular mechanisms to cope with the negative effects of salinity. These include the regulation of genes with a role in the uptake, transport or compartmentation of Na+ and/or K+. Studies have shown that the arbuscular mycorrhizal (AM) symbiosis alleviates salt stress in several host plant species. However, despite the abundant literature showing mitigation of ionic imbalance by the AM symbiosis, the molecular mechanisms involved are barely explored. The objective of this study was to elucidate the effects of the AM symbiosis on the expression of several well-known rice transporters involved in Na+/K+ homeostasis and measure Na+ and K+ contents and their ratios in different plant tissues. Results showed that OsNHX3, OsSOS1, OsHKT2;1 and OsHKT1;5 genes were considerably upregulated in AM plants under saline conditions as compared to non-AM plants. Results suggest that the AM symbiosis favours Na+ extrusion from the cytoplasm, its sequestration into the vacuole, the unloading of Na+ from the xylem and its recirculation from photosynthetic organs to roots. As a result, there is a decrease of Na+ root-to-shoot distribution and an increase of Na+ accumulation in rice roots which seems to enhance the plant tolerance to salinity and allows AM rice plants to maintain their growing processes under salt conditions.  相似文献

The differential responses of the wheat cultivars Shi4185 and Yumai47 to salinity were studied. The higher sensitivity of Yumai47 to salinity was linked to a greater growth reduction under salt stress, compared to more salt-tolerant Shi4185. Salinity increased the Na+, proline and superoxide anion radical (O2 ?) contents in both cultivars. Leaf Na+ content increased less in the more salt-tolerant cultivar Shi4185 than salt-sensitive Yumai47. The proline content increased more significantly in Shi4185 than Yumai47; on the contrary, superoxide anion radical content increased less in Shi4185 than Yumai47. This data indicated that wheat salinity tolerance can be increased by controlling Na+ transport from the root to shoot, associated with higher osmotic adjustment capability and antioxidant activity. Although salinity increased aldehyde oxidase (AO) activity and abscisic acid (ABA) content in the leaves and roots of both cultivars following the addition of NaCl to the growth medium, AO and ABA increased more in the salt-sensitive cultivar Yumai47 than the more salt-tolerant cultivar Shi4185. Xanthine dehydrogenase (XDH) activity in the leaves of both cultivars increased with increasing concentrations of NaCl; however, leaf XDH activity increased more significantly in Yumai47 than Shi4185. Root XDH activity in Shi4185 decreased with increasing NaCl concentrations, whereas salinity induced an increased root XDH activity in Yumai47. The involvement of AO and XDH enzymatic activities and altered ABA content in the response mechanisms of wheat to salinity are discussed herein.  相似文献

Changes in respiratory pathway, dry weight, contents of proline, ATP, Na+and K+were investigated under five salinity treatments in the leaves of plants of spring wheat (Triticum aestivumL.). Two cultivars differing in salt resistance, namely, 89122 (salt-tolerant) and Longchun13 (salt-sensitive), were used. The decrease in dry weight and K+content was observed with the increasing NaCl concentrations, but more in cv. 89122 plants than in Longchun13 plants. Contents of proline and Na+in both cvs increased greatly, but the former increased more in 89122 while the latter more in Longchun13 plants. In all salinity treatments tested, a salt-induced increase in the activity of the alternative pathway was found, although cytochrome pathway (CP) still remained the main electron transport pathway. ATP production changed in parallel with CP operation. Cv. 89122 plants could produce more ATP than cv. Longchun13 plants exposed to each salinity treatment and their ATP generation could even be stimulated in contrast to its rapidly decline in Longchun13 plants with increased salinity stress. The possible relationship between respiration metabolism and above mentioned physiological changes is discussed.  相似文献

Sunflowers were treated with mixing proportions of NaCl, Na2SO4, NaHCO3, and Na2CO3. Effects of salt and saltalkaline mixed stress on growth, photosynthesis, chlorophyll fluorescence, and contents of inorganic ions and organic acids of sunflower were compared. The growth of sunflower decreased with increasing salinity. The contents of photosynthetic pigments did not decrease under salt stress, but their contents decreased sharply under salt-alkaline mixed stress. Net photosynthetic rates, stomatal conductance and intercellular CO2 concentration decreased obviously, with greater reductions under salt-alkaline mixed stress than under salt one. Fluorescence parameters showed no significant differences under salt stress. However, maximal efficiency of PSII photochemistry, photochemical quenching coefficient, electron transport rate, and actual PSII efficiency significantly decreased but non-photochemical quenching increased substantially under salt-alkaline mixed stress. Under salt-alkaline mixed stress, sunflower leaves maintained a low Na+- and high K+ status; this may be an important feature of sunflower tolerance to salinity. Analysis of the mechanism of ion balance showed that K+ but not Na+ was the main inorganic cation in sunflower leaves. Our results indicated that the change in organic acid content was opposite to the change of Cl, and the contribution of organic acid to total charge in sunflower leaves under both stresses decreased with increasing salinity. This may be a special adaptive response to stresses for sunflower. Sunflower under stress conditions mainly accumulated inorganic ions instead of synthesizing organic compounds to decrease cell water potential in order to save energy consumption.  相似文献

盐胁迫影响植物组织的离子分布,不同品种间存在差异。以玉米耐盐自交系81162和8723及盐敏感自交系P138为材料,研究了不同浓度(0、60、140、220 mmol/L)Na Cl胁迫下萌动期种子和幼苗的不同部位中Na+、K+、Ca2+含量以及K+/Na+和Ca2+/Na+比值的变化,旨在探讨不同自交系耐盐性差异的原因。结果表明,在萌动种子中,3个玉米自交系中的Na+积累量表现为种皮胚胚乳,K+累积表现为胚种皮胚乳;幼苗中,Na+积累表现为根茎叶。随着Na Cl浓度的增加,3个玉米自交系萌动种子和幼苗中的Na+含量逐渐升高,但是萌动种子中耐盐自交系81162和8723的Na+增加幅度小于盐敏感自交系P138,Na+含量小于盐敏感自交系P138;幼苗中耐盐自交系81162和8723的Na+增加幅度大于盐敏感自交系P138,幼苗根中Na+含量大于盐敏感自交系P138;茎叶中的Na+含量小于盐敏感自交系P138。随着Na Cl浓度的增加,萌动种子和幼苗中的K+和Ca2+含量逐渐降低。K+离子在耐盐自交系81162和8723萌动种子和幼苗中的降低幅度小于盐敏感自交系P138;Ca2+离子在耐盐自交系81162和8723幼苗中的降低幅度小于盐敏感自交系P138;而在萌动种子中3个自交系Ca2+的流失差异不大。耐盐自交系81162和8723萌动种子和幼苗中K+含量都大于盐敏感自交系P138。耐盐自交系81162和8723的萌动种子和幼苗根中Ca2+含量都大于盐敏感自交系P138;幼苗叶片中则小于盐敏感自交系P138。萌动种子和幼苗中K+/Na+和Ca2+/Na+均随着Na Cl浓度的升高而降低,K+/Na+比值表现为耐盐自交系81162和8723大于盐敏感自交系P138。耐盐自交系81162和8723通过调节离子平衡维持萌动种子和幼苗中较高的K+/Na+比值从而提高耐盐性。  相似文献

Salt stress is considered to be a major limiting factor for plant growth and crop productivity. Salt injuries in plants are mostly due to excess Na+ entry. A possible survival strategy of plants under saline environments is the effective compartmentation of excess Na+ by sequestering Na+ in roots and inhibiting transport of Na+ from roots to shoots. Our previous study showed that exogenous application of polyamines (PAs) could attenuate salt injuries in barley plants. In order to further understand such protective roles of PAs against salt stress, the effects of spermidine (Spd) on sodium and potassium distribution in barley (Hordeum vulgare L.) seedlings under saline conditions were investigated. The results showed that exogenous application of Spd induced reductions in Na+ levels in roots and shoots with comparison of NaCl-treated plants, while no significant changes in K+ levels were observed. Correspondingly, the plants treated with Spd exogenously maintained high values of [K+]/[Na+] as compared with salt-stressed plants. Moreover, it was shown by X-ray microanalysis that K+ and Na+ accumulated mainly in the exodermal intercellular space and cortical cells of roots under salinity stress, and low accumulation was observed in endodermal cells and stelar parenchyma, indicating Casparian bands possibly act as ion transport barriers. Most importantly, Spd treatment further strengthened this barrier effects, leading to inhibition of Na+ transport into shoots. These results suggest that, by reinforcing barrier effects of Casparian bands, exogenous Spd inhibits Na+ transport from roots to shoots under conditions of high salinity which are beneficial for attenuating salt injuries in barley seedlings.  相似文献

Dry matter changes and ion partitioning in two near isogenic barley cultivars Maythorpe (relatively salt sensitive) and Golden Promise (relatively salt tolerant) were studied in response to increasing salinity. Although the growth of both cultivars was significantly reduced by exposure to NaCl, the effect was greater in Maythorpe, whilst Golden Promise maintained an increased ratio of young to old leaf blade. Golden Promise maintained significantly lower Na+ concentrations in young expanding tissues compared with Maythorpe. Partitioning of Cl was evident in that both varieties maintained lower Cl concentrations in mesophyll than in epidermal cells. Golden Promise maintained higher K+/Na+ and Ca2+/Na+ ratios in young leaf blade and young sheath tissues than Maythorpe when exposed to salt. Differences in ion partitioning and the maintenance of higher K+ and Ca2+ to Na+ ratios, especially in young growing and recently expanded tissues, would appear to be important mechanisms contributing to the improved salt tolerance of Golden Promise.  相似文献

To assess whether foliar application of K+S as potassium sulfate (K2SO4) could alleviate the adverse effects of salt on sunflower (Helianthus annuus L. cv. SF-187) plants, a greenhouse experiment was conducted. There were two NaCl levels (0 and 150 mM) applied to the growth medium and six levels of K+S as K2SO4 (NS (no spray), WS (spray of water+0.1% Tween 20 solution), 0.5% K+0.21% S, 1.0% K+0.41% S, 1.5% K+0.62% S, and 2.0% K+0.82% S in 0.1% Tween-20 solution) applied two times foliarly to non-stressed and salt-stressed sunflower plants. Salt stress markedly repressed the growth, yield, photosynthetic pigments, water relations and photosynthetic attributes, quantum yield (Fv/Fm), leaf and root K+, Mg2+, P, Ca2+, N as well as K+/Na+ ratios, while it enhanced the cell membrane permeability, and leaf and root Na+ and Cl concentrations. Foliar application of potassium sulfate significantly improved growth, achene yield, photosynthetic and transpiration rates, stomatal conductance, water use efficiency, leaf turgor and enhanced shoot and leaf K+ of the salt-stressed sunflower plants, but it did not improve leaf and root Na+, Cl, Mg2+, P, Ca2+, N as well as K+/Na+ ratios. The most effective dose of K+S for improving growth and achene yield was found to be 1.5% K+0.62% S and 1% K+0.41% S, respectively. Improvement in growth of sunflower plants due to exogenously applied K2SO4 was found to be linked to enhanced photosynthetic capacity, water use efficiency, leaf turgor and relative water content.  相似文献

The Arabidopsis vacuolar H+‐pyrophosphatase (AVP1), when over‐expressed in transgenic (TG) plants, regulates root and shoot development via facilitation of auxin flux, and enhances plant resistance to salt and drought stresses. Here, we report that TG perennial creeping bentgrass plants over‐expressing AVP1 exhibited improved resistance to salinity than wild‐type (WT) controls. Compared to WT plants, TGs grew well in the presence of 100 mm NaCl, and exhibited higher tolerance and faster recovery from damages from exposure to 200 and 300 mm NaCl. The improved performance of the TG plants was associated with higher relative water content (RWC), higher Na+ uptake and lower solute leakage in leaf tissues, and with higher concentrations of Na+, K+, Cl and total phosphorus in root tissues. Under salt stress, proline content was increased in both WT and TG plants, but more significantly in TGs. Moreover, TG plants exhibited greater biomass production than WT controls under both normal and elevated salinity conditions. When subjected to salt stress, fresh (FW) and dry weights (DW) of both leaves and roots decreased more significantly in WT than in TG plants. Our results demonstrated the great potential of genetic manipulation of vacuolar H+‐pyrophosphatase expression in TG perennial species for improvement of plant abiotic stress resistance.  相似文献

Effects of seawater salinity (SWS) and pure NaCl on the intracellular contents of Na+, K+, Mg2+, Ca2+, chlorophylls (Chl) and carotenoids (Car) were studied in three submerged aquatic macrophytes, Hydrilla verticillata, Najas indica and Najas gramenia, which differed in their tolerance to salinity. NaCl resulted in significant increase in Chl/Car ratio in the salt-sensitive H. verticillata and moderately salt-tolerant N. indica, but not in the salt-tolerant N. gramenia. SWS treatment did not result in any significant change in the ratio. The intracellular content of Na+ increased significantly in all the test plants upon exposure to both NaCl and SWS. The content of K+ decreased significantly in these plants upon salinity treatment, except in N. gramenia. The contents of Ca2+ and Mg2+ decreased significantly upon NaCl treatment and remained unchanged or increased upon SWS treatment. No relationship between salt tolerance and K+/Na+ ratio was observed. The maintenance of a minimal level of K+ was observed to be the most probable requirement of salt tolerance in aquatic macrophytes.  相似文献

Na+ transport across the tonoplast and its accumulation in the vacuoles is of crucial importance for plant adaptation to salinity. Mild and severe salt stress increased both ATP- and PPi-dependent H+ transport in tonoplast vesicles from sunflower seedling roots, suggesting the possibility that a Na+/H+ antiport system could be operating in such vesicles under salt conditions (E. Ballesteros et al. 1996. Physiol. Plant. 97: 259–268). During a mild salt stress, Na+ was mainly accumulated in the roots. Under a more severe salt treatment, Na+ was equally distributed in shoots and roots. In contrast to what was observed with Na+, all the salt treatments reduced the shoot K+ content. Dissipation by Na+ of the H+ gradient generated by the tonoplast H+-ATPase, monitored as fluorescence quenching of acridine orange, was used to measure Na+/H+ exchange across tonoplast-enriched vesicles isolated by sucrose gradient centrifugation from sunflower (Helianthus annuus L.) roots treated for 3 days with different NaCl regimes. Salt treatments induced a Na+/H+ exchange activity, which displayed saturation kinetics for Na+ added to the assay medium. This activity was partially inhibited by 125 μM amiloride, a competitive inhibitor of Na+/H+ antiports. No Na+/H+ exchange was detected in vesicles from control roots. The activity was specific for Na+. since K+ added to the assay medium slightly dissipated H+ gradients and displayed non-saturating kinetics for all salt treatments. Apparent Km for Na+/H+ exchange in tonoplast vesicles from 150 mM NaCl-treated roots was lower than that of 75 mM NaCl-treated roots, Vmax remaining unchanged. The results suggest that the existence of a specific Na+/H+ exchange activity in tonoplast-enriched vesicle fractions, induced by salt stress, could represent an adaptative response in sunflower plants, moderately tolerant to salinity.  相似文献
