三草芽孢杆菌(Bacillus subtilis)BM9602产生的中性内切β-甘露聚糖酶(endo-β-1,4-D-mannan mannanohydrolaes,EC,3.2.1.78)经硫酸铵分级沉淀、DEAE-纤维素(DE22)离子交换柱层析,得到电泳纯的样品,提纯了45.5倍,收率为5.9%。用SDS-PAGE测得该酶的分子量为35kD。用PAGEIEF测得其等电点pⅠ为4.5。酶反应的  相似文献

柑叶片蔗糖酶的分离纯化及其部分性质的研究   总被引:8,自引:0,他引:8  
柑(Citrusreticulata Blanco)幼叶中存在高活性的酸性蔗糖酶。经硫酸铵盐析、DEAE-琼脂糖离子交换层析、SephacrylS-200 凝胶层析纯化,活性回收率6.4% ,纯化倍数179.2 倍。纯化的酶经聚丙烯酰胺凝胶电泳显示单一蛋白带,SDS-PAGE显示1 条蛋白带,其亚基分子量40 kD。用SephacrylS-200 凝胶层析法测得分子量为80 kD。推测该酶由两个相同亚基构成。以蔗糖为底物测定该酶的表观Km 为1.6×10- 2 m ol·L- 1,Vm ax为100 m g 还原糖·m g- 1蛋白质·h- 1。最适pH 5.0,酸碱稳定区在pH 4.5—5.5 之间。最适温度55℃  相似文献

经SephadexG-75凝胶过滤,QAE-SephadexA-50和CM-SephadexC-25离子交换层析的步骤,从湖南产尖吻蝮(Dienagkistrodonacutus)蛇毒中纯化出两个出血毒素(DaHT-1和DaHT-2).SDS-PAGE测得分子量均为23.5kD,IEF-PAGE测得等电点分别为5.6和5.2,两者具有相似的氨基酸组成,其中酸性氨基酸(Asx,Glx)分别占23%和24%,DaHT-1和DaHT-2的最小出血剂量(MHD)分别为0.5μg和0.8μg。都具蛋白水解酶活性,无对TAME,BAEE的水解活性和PLA2酶活性.两者的蛋白水解酶活力与出血活性并非正相关.DaHT-1和DaHT-2的最适温度分别为35℃和40℃,最适pH为6-9,对热均不稳定,温度高于60℃活性完全丧失。金属离子的分析显示每摩尔毒素蛋白约含0.5mol的Zn,1mol的Ca,较多的Na、K、Mg,不含Co。  相似文献

本文从含ArgRS306KR基因args306KR的pUC18重组质粒的大肠杆菌TG1转化子中经DEAE-Sephacel和Blue-Sepharose两步柱层析,得到电泳一条带的ArgRS306KR。纯酶的比活为2790单位/毫克。该酶氨酰化和ATP~PPi交换活力的最适pH分别为pH8.3和pH7.5。氨酰化活力对ATP、Arg和tRNA的Km分别为2.6mmol/L、14.0μmol/L和5.0μmol/L:Vmax为7630单位/毫克;koat为9S-1。ATP~PPi交换活力对ATP和Arg的Km分别为8.3mmol/L和99μmol/L;Vmax为16320单位/毫克;kcat为18S-1。  相似文献

液体发酵得到的红栓菌(Pycnoporuscinnabarius)菌丝经沸水抽提,胰匀浆处理和Sevage法除蛋白,DEAESephadexA-25与SephadexG-150柱层析,乙醇沉淀得到二种多糖组分(简称PC_1、PC_2)。经聚丙烯酰胺电泳,冻融分析鉴定为均一体。PC_1和PC_2的,总糖含量分别为81.5%和77.3%,分子量分别为23000和13000.两者的红处光谱具多糖特征吸收峰,在紫外区无明显吸收现象,基本不含氮。PC_1单糖组成为D-葡萄糖、D-半乳糖及D-甘露糖,PC_2为D-半乳糖、D-甘露糖。  相似文献

蚯蚓体内一种纤溶酶原激活剂(e-PA)对BAEE的降解   总被引:2,自引:0,他引:2  
以苯甲酰-L-精氨酸乙酯(benzoyl-L-arginineethylester,BAEE)为底物,研究了蚯蚓体内纤溶酶原激活剂(plasminogenactivatorfromEiseniafetida,e-PA)的酶学性质.酶促反应的最适pH为8.4,e-PA降解BAEE的Km为1.24±0.16×10-5mol/L,Kcat为13.80±4.02s-1.测定了构成e-PA的大,小亚基分别降解BAEE的Km和Kcat.结果表明,大亚基的Km与全酶的Km相差不多,但比小亚基小约10倍,即对底物的亲和力比小亚基强约一个数量级.大小亚基的Kcat比较接近,分别是全酶的1/6和1/3.研究了8种抑制剂对e-PA降解BAEE活性的影响,其中pepstatin和E-64(一种巯基抑制剂)对酶促反应有激活作用,TPCK,TL-CK,PMSF,chymostatin和leupeptin对其有不同程度的抑制作用,EDTA对e-PA的活性没有影响.对e-PA的BAEE活性和e-PA的纤溶活性之间作了比较.  相似文献

The liver is thought to be involved in the systemic clearance and detoxification of lipopolysaccharide (LPS). Argininosuccinate synthase (AS), a liver cytosolic urea cycle enzyme, has been found to bind to and inactivate LPS and lipid A. To elucidate the participation of AS in the clearance of LPS by liver and hepatocytes, we investigated the correlation between AS content and the removal of lipid A and LPS in vivo and in vitro, tracing levels of biological activity. A hepatotoxic model in which mice were injected with CCl(4) revealed a significant reduction in lipid A clearance along with liver failure on day 1; total body clearance was changed to 0.534 ml/min from 1.42 ml/min. AS content in liver concomitantly decreased to about half and AS leaked to blood at about 6 microg/ml. Total body clearance of i.v. injected AS was estimated at 0.083 ml/min, which predicted about 24-h leakage of AS after CCl(4) injection. The treatment also reduced the clearance of R-type LPSs to a lesser degree the larger its polysaccharide portion. S-type LPS, which has a large O-antigen polysaccharide, exhibited enhancement of clearance on CCl(4) treatment. When pretreated in vitro with AS and injected into normal mice, lipid A and R-type LPS showed a similar pattern of clearance of residual activities to the untreated forms, but S-type LPS exhibited enhancement of clearance. Comparison between different strains of mice revealed a correlation of AS content in liver and lipid A clearance, where the higher AS strain C3H/He mice showed a more rapid clearance than the lower AS strains C57BL/6 and BALB/c. Primary spheroid cultures of hepatocytes treated with 0.1 microM dexamethasone and 1 microM glucagon showed about a 2-fold increase in AS amount and a more rapid clearance of LPS from culture medium than untreated cells. These results suggest that AS in hepatocytes may be involved in the process of lipid A and LPS clearance and the extracellular leakage of AS may also participate in the systemic detoxification.  相似文献

Adjustment of polyamine contents in Escherichia coli was studied with strains of Escherichia coli producing normal (DR112) and excessive amounts of ornithine decarboxylase [DR112(pODC)] or S-adenosylmethionine decarboxylase [DR112(pSAMDC)]. Although DR112(pODC) produced approximately 70 times more ornithine decarboxylase than DR112 did, the amounts of polyamines in the cells of both strains did not change significantly. The amounts of polyamines in DR112(pODC) were adjusted by excretion of excessive amounts of putrescine to the medium. When ornithine was deficient in cells, polyamine contents in DR112(pODC) were much higher than those in DR112, although polyamine contents were low in both strains. This indicates that large amounts of ornithine decarboxylase increased the utilization of ornithine for putrescine synthesis. During ornithine deficiency, strain DR112 produced 3.4 times more ornithine decarboxylase. Strain DR112(pSAMDC) produced seven times more S-adenosylmethionine decarboxylase than DR112 did. In DR112(pSAMDC) an increase (40%) in spermidine content, a decrease (35%) in putrescine content, and no significant excretion of putrescine and spermidine were observed. The amount of ornithine decarboxylase in DR112(pSAMDC) was approximately 30% less than that in DR112. In addition, S-adenosylmethionine decarboxylase activity was strongly inhibited by spermidine. A possible regulatory mechanism to maintain polyamine contents in Escherichia coli is discussed based on the results.  相似文献

用钳位均匀电场脉冲电泳(CHEF)系统分析了克鲁斯假丝酵母(Candida krusei),郎比可假丝酵母(C. lambica)和粗状假丝酵母(C. valiad)的模式菌株的电泳核型,发现这三种表型相似的假丝酵母却具有互不相同的染色体DNA分子带型,为其分类学研究提供了可靠的鉴别依据。在常规分类学研究的基础上,测定了AS 2.75(原定种名为(C. incospicua),AS2.1182(原定种名为 C. lambica)和AS 2.1772(未定种)等三株假丝酵母的G+C含量和脉冲电泳核型。通过对已报道的C. inconspicu的G+C含量及上述三种假丝酵母模式菌株的脉冲电泳核型的比较分析证明,AS 2.75和AS 2.1772为粗状假丝酵母(C. valida),AS 2.1182为克鲁斯假丝酵母(C. krusei)。  相似文献

Atherosclerosis (AS), the leading cause of morbidity and mortality in cardiovascular disease, needs an early detection for treatment and prevention of fatal events. Here, for the first time, we applied gold nanorods (GNRs)‐assisted diffusion reflection (DR), a noninvasive technique for in vivo detection of AS in a high‐fat‐diet‐induced c57bl mouse model, which resembles the manifestation of AS in humans. DR simply detects the change in light reflection profile of tissue due to the accumulation of GNRs in the AS plaques and enables clear detection of AS lesions in carotid and femoral arteries of these hyperlipidemic mice. After 24 hours post‐GNRs injection, DR showed the highest efficiency of AS detection. Moreover, the sensitivity of the DR method is much higher than computed tomography (CT) and is comparable to ex vivo high‐resolution CT. Our results strongly suggest that the DR method can detect early atherosclerotic lesions in a sensitive and specific manner.   相似文献

The effects of dietary restriction (DR) on the activities of liver superoxide dismutase (SOD), catalase (Cat), and glutathione peroxidase (GPX) and the level of lipid peroxidation (LP) in developing mice were investigated in this study. Male and female Kunmin mice were fed a standard rodent diet ad libitum (AL), 80% of AL food intake (20% DR), or 65% of AL food intake (35% DR) for 12 or 24 wk. Both 12 and 24 wk of DR resulted in retarded body weight gain in male and female mice. The activities of SOD, Cat, and GPX and the content of LP in DR male and female mice were not different (P > 0.05) from those in controls after 12 wk of DR. However, the SOD activity was increased at 24 wk in 20% DR (P < 0.05) and 35% DR (P < 0.01) male, but not in DR female, mice. The Cat activity was elevated at 24 wk in both DR male (P < 0.05 for 20% DR, P < 0.01 for 35% DR) and female (P < 0.01) mice with a greater increase in DR female (P < 0.05) than in DR male animals. GPX activity was also increased at 24 wk in DR male (P < 0.01) and female (P < 0.01) mice with a greater elevation in DR females (P < 0.05) than in DR males. Furthermore, LP was decreased at 24 wk in both DR male (P < 0.01) and female (P < 0.01) animals with a greater reduction in DR females (P < 0.01) compared with DR males. These findings indicated that 24 wk, but not 12 wk, of DR led to differential effects on liver SOD, Cat, and GPX activities and LP content in male and female mice during development, suggesting sex-associated modulations of DR on antioxidant systems in developing animals.  相似文献

Objectives

Diabetic retinopathy (DR) is a common diabetic eye disease which is well-known as the result of microvascular retinal changes. Although the potential biological functions of astragaloside IV (AS IV) have long been described in traditional system of medicine, its protective effect on DR remains unclear. This study aims to investigate the function and mechanism of AS IV on type 2 diabetic db/db mice.

Methods

Db/db mice were treated with AS IV (4.5 mg/kg or 9 mg/kg) or physiological saline by oral gavage for 20 weeks along with db/m mice. In each group, retinal ganglion cell (RGC) function was measured by pattern electroretinogram (ERG) and apoptosis was determined by Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining. Blood and retina aldose reductase (AR) activity were quantified by chemiluminescence analysis. The expressions of phosporylated-ERK1/2, NF-κB were determined by Western blot analysis. Furthermore, the expression of related downstream proteins were quantified by Label-based Mouse Antibody Array.

Results

Administration of AS IV significantly improved the amplitude in pattern ERG and reduced the apoptosis of RGCs.in db/db mice. Furthermore, downregulation of AR activity, ERK1/2 phosphorylation, NF-κB and related cytokine were observed in AS IV treatment group.

Conclusions

Our study indicated that AS IV, as an inhibitor of AR, could prevent the activation of ERK1/2 phosporylation and NF-kB and further relieve the RGCs disfunction in db/db mice with DR. It has provided a basis for investigating the clinical efficacy of AR inhibitors in preventing DR.  相似文献

Chronic dietary restriction (DR) is considered among the most robust life-extending interventions, but several reports indicate that DR does not always extend and may even shorten lifespan in some genotypes. An unbiased genetic screen of the lifespan response to DR has been lacking. Here, we measured the effect of one commonly used level of DR (40% reduction in food intake) on mean lifespan of virgin males and females in 41 recombinant inbred strains of mice. Mean strain-specific lifespan varied two to threefold under ad libitum (AL) feeding and 6- to 10-fold under DR, in males and females respectively. Notably, DR shortened lifespan in more strains than those in which it lengthened life. Food intake and female fertility varied markedly among strains under AL feeding, but neither predicted DR survival: therefore, strains in which DR shortened lifespan did not have low food intake or poor reproductive potential. Finally, strain-specific lifespans under DR and AL feeding were not correlated, indicating that the genetic determinants of lifespan under these two conditions differ. These results demonstrate that the lifespan response to a single level of DR exhibits wide variation amenable to genetic analysis. They also show that DR can shorten lifespan in inbred mice. Although strains with shortened lifespan under 40% DR may not respond negatively under less stringent DR, the results raise the possibility that life extension by DR may not be universal.  相似文献

根据酵母属( Saccharomyces Meyen ex Reess) 分类学研究最新进展,核实并更新了保藏于中国普通微生物菌种保藏中心的该属菌株的种类归属。在形态和生理生化性状,包括对6 种糖的发酵能力、对18 种碳源和3 种氮源化合物的同化能力、在无维生素培养基中和37 ℃下的生长情况、对放线菌素酮的抗性等常规分类学研究的基础上,对部分疑难菌株进行了脉冲电泳核型比较分析。酿酒酵母( Saccharomycescerevisiae) 、贝酵母( S.Bayanus) 和巴氏酵母( S.Pastorianus) 三者与少孢酵母( S.Exiguus) 在电泳核型上具有明显的差异,主要表现在前三者染色体DNA 分子的大小范围均为225 ~2200 kb ,而S.Exiguus 缺少小于365 kb 的染色体DNA 分子。S.Cerevisiae 的模式和权威菌株具有12 ~14 条染色体DNA 带;S.BayanusS.Pastorianus 的模式菌株均有17 条带,但在带型上存在一定差异。原归于S.Cerevisiae 的株菌AS2-100 具有16 条带,与S.Cerevisiae 区别明显而与S.…  相似文献

The early embryonic development of Pygoscelis adeliae, up to the stage with fused neutral folds, was investigated by means of serial sections and compared with the development of the chick (Gallus domesticus). The total incubation time in the penguin is about 35 days, compared with about 20 days in the chick (Herbert 1967). This superior rate of development in the chick is likewise recognized in the early period of development; for fusion of neural folds is achieved 45–49 hours after laying in the chick and 6–7 1/2 days incubation in the penguin. Furthermore the time of origin and shape of the neural tube, notochord and heart show recognizable differences in the two genera.  相似文献

对根据常规形态和生理生化性状难以确定分类学地位的8株假丝酵母菌,进行了以大亚基(26S) rDNA中D1/D2区域(约500~600 bp)的碱基序列分析为依据的分子分类学研究.根据系统树上所显示的供试菌株与假丝酵母属及相关子囊菌酵母已知种的亲缘关系,以及与最近缘种模式菌株D1/D2区域序列的相似性比较,确定了各个菌株的归属.本研究也显示了DNA序列分析在假丝酵母菌快速鉴定中的优越性.  相似文献
