Background

Development of a non-hormonal long-acting reversible contraceptive for men could have a significant impact on reducing unintended pregnancies. Vasalgel? is a high molecular weight polymer consisting of styrene-alt-maleic acid (SMA) dissolved in dimethyl sulfoxide being developed as a reversible male contraceptive device. It forms a hydrogel when implanted into the vasa deferentia, which prevents the passage of sperm. Previous studies in the rabbit have proven its efficacy, durability and rapid onset. This study evaluates the capacity to restore sperm concentrations in ejaculates after a reversal procedure.

Methods

Sodium bicarbonate was injected into the vasa deferentia after fourteen months of azoospermia following the injection of two device variations (Vasalgel 100 and Vasalgel 80). Semen samples were then collected for six months and sperm characteristics were compared to baseline levels. Samples of vasa deferentia were obtained for histological examination.

Results

Spermatozoa were present in all subject ejaculates after the reversal procedure. Sperm concentration and sperm motility were similar to baseline levels after reversal, while sperm forward progression was significantly lower and normal acrosomes were not observed. Forward progression percentages increased linearly during six months of semen collection, however, normal acrosomes were not observed at the conclusion of the study. Histologically, several vasa deferentia were clear of the device and contained an intact epithelial lining. A smaller proportion of tissues contained residual test material. A secondary intraluminal inflammatory response was seen occasionally in the tissues containing residual material. There was no difference between the two device variations for studied parameters.

Conclusions

Vasalgel’s prevention of sperm transport for 14 months was reversed through an intravasal injection of sodium bicarbonate. Post-reversal sperm concentrations and motility returned to baseline levels during the six-month follow up. Residual material in the vas lumen or compromised epididymal and vas deferens function may be resulting in reduced forward progression and loss of acrosomes during transit through the vas. Reduced forward progression and the lack of normal acrosomes strongly suggest impaired sperm function.

We report the molecular characterization of a ring X chromosome that was transmitted from a mother to a male who has short stature and minor dysmorphic features. This represents only the second reported ring X chromosome in a male. The ring is derived from breakage within the Xp pseudoautosomal region (PAR) and just proximal to the Xq PAR. The total amount of deleted material is 700-900 kb DNA and includes six known transcribed genes. Interestingly, SHOX, a gene implicated in short stature, is not deleted from the ring chromosome. Possible pathogenetic explanations for the patient's clinical features include insufficient dosage of deleted genes, a position effect on SHOX expression, and cell death during development because of ring chromosome nondisjunction. The findings are also relevant to observations made of "complete" ring chromosomes.  相似文献

Male dimorphism has been reported across different taxa and is usually expressed as the coexistence of a larger morph with exaggerated male traits and a smaller one with reduced traits. The evolution and maintenance of male dimorphism are still poorly understood for several of the species in which it has been observed. Here, we analyse male dimorphism in several species of reptile parasitic nematodes of the genus Spauligodon, in which a major male morph (exaggerated morph), which presents the traditional male morphological traits reported for this taxon, coexists with a minor morph with reduced morphological traits (i.e. reduced genital papillae) resembling more closely the males of the sister genus Skrjabinodon than Spauligodon major males. Because of the level of uncertainty in the results of ancestral state reconstruction, it is unclear if the existence of male dimorphism in this group represents independent instances of convergent evolution or an ancestral trait lost multiple times. Also, although the number of major males per host was positively correlated with the number of females, the same did not hold true for minor males, whose presence was not associated with any other ecological factor. Nevertheless, the existence of male dimorphism in Spauligodon nematodes is tentatively interpreted as resulting from alternative reproductive tactics, with differences in presence and number of individuals as indicators of differences in fitness, with the lower numbers of minor males per host likely maintained by negative frequency‐dependent selection.  相似文献

Existing theory predicts that male signalling can be an unreliable indicator of paternal care, but assumes that males with high levels of mating success can have high current reproductive success, without providing any parental care. As a result, this theory does not hold for the many species where offspring survival depends on male parental care. We modelled male allocation of resources between advertisement and care for species with male care where males vary in quality, and the effect of care and advertisement on male fitness is multiplicative rather than additive. Our model predicts that males will allocate proportionally more of their resources to whichever trait (advertisement or paternal care) is more fitness limiting. In contrast to previous theory, we find that male advertisement is always a reliable indicator of paternal care and male phenotypic quality (e.g. males with higher levels of advertisement never allocate less to care than males with lower levels of advertisement). Our model shows that the predicted pattern of male allocation and the reliability of male signalling depend very strongly on whether paternal care is assumed to be necessary for offspring survival and how male care affects offspring survival and male fitness.  相似文献

Adult male rats with cannulated or ligated bile ducts were given S-(2-hydroxyoestradiol-1-yl)[(35)S]glutathione, S-(2-hydroxy[6,7-(3)H(2)]oestradiol-1-yl)glutathione or S-(2-hydroxyoestradiol-1-yl)[glycine-(3)H]glutathione by intraperitoneal injection. The recovery of radioactivity in the bile of bile duct-cannulated rats was 33-86% and in the urine of bile duct-ligated rats was 54-105%. Oestrogen thioether derivatives of glutathione, cysteinylglycine, cysteine and N-acetylcysteine were isolated from bile; only the N-acetylcysteine derivatives could be identified in the urine. The steroid moiety was characterized by microchemical tests before and after treatment with Raney nickel: 2-hydroxyoestradiol-17beta was released from the glutathione conjugate, and 2-hydroxyoestrone and 2-hydroxyoestrone 3-methyl ether from the other conjugates. From intact rats the recovery of administered radioactivity was about 15% in the urine and 5% in the faeces over a period of several days and the radioactivity appeared to be largely protein-bound. The results demonstrate that injected oestrogen-glutathione conjugate undergoes conversion into N-acetylcysteine derivatives in vivo. Oestrogen-glutathione conjugates formed in the intact rat may be excreted in an apparently non-steroidal, possibly protein-bound form, which would not be detected by current analytical techniques.  相似文献

Mammalian target of rapamycin (mTOR) is a central regulator of cellular metabolic phenotype and is involved in virtually all aspects of cellular function. It integrates not only nutrient and energy-sensing pathways but also actin cytoskeleton organization, in response to environmental cues including growth factors and cellular energy levels. These events are pivotal for spermatogenesis and determine the reproductive potential of males. Yet, the molecular mechanisms by which mTOR signaling acts in male reproductive system remain a matter of debate. Here, we review the current knowledge on physiological and molecular events mediated by mTOR in testis and testicular cells. In recent years, mTOR inhibition has been explored as a prime strategy to develop novel therapeutic approaches to treat cancer, cardiovascular disease, autoimmunity, and metabolic disorders. However, the physiological consequences of mTOR dysregulation and inhibition to male reproductive potential are still not fully understood. Compelling evidence suggests that mTOR is an arising regulator of male fertility and better understanding of this atypical protein kinase coordinated action in testis will provide insightful information concerning its biological significance in other tissues/organs. We also discuss why a new generation of mTOR inhibitors aiming to be used in clinical practice may also need to include an integrative view on the effects in male reproductive system.  相似文献

Males of the stream goby Rhinogobius sp. DA (dark type) court females in deep pools and care for the eggs under stones in shallow riffles. We studied male–male competition for access to females and nest sites to understand how male size influences the mating success of this species. In field observations, larger males won in fighting with other males. However, large males did not tend to monopolize courtship opportunities, and the frequency of successful courtships, after which males led the females to the nests, was not related to male body size. The fact that courted females always escaped from the fighting sites once males began fighting likely explains why male size was not positively related to courtship success. Large males occupied large nest stones, and the number of eggs received in the nest was correlated positively with nest size. In aquarium experiments with two tiles of different sizes provided as nesting materials, males always chose the larger nest and, when two males were introduced simultaneously, the larger one occupied the larger nest. These results suggested that male mating success of this goby is determined by male–male competition for large nests rather than for access to females. Conjugated and unconjugated urinary levels of phenylacetic acid (PAA), m-hydroxyphenylacetic acid (m-HPA) and p-hydroxyphenylacetic acid have been determined for 24-h urine samples obtained from a single healthy male over a 28-day period. Gas chromatographic—electron-capture and mass spectrometric—integrated ion current techniques incorporating appropriate internal standards were used. The average urinary excretion values obtained were (in mg/24 h): PAA unconjugated 0.67, conjugated 96.6; m-HPA unconjugated 7.3, conjugated < 0.1; p-HPA unconjugated 22.4, conjugated < 1.2. Following the ingestion of appropriate deuterated amino acid precursors the expected urinary deuterated trace acids were identified and quantitated; in the case of deuterated phenylethylamine, m-HPA and p-HPA as well as PAA were identified and quantitated. This is the first evidence of phenylethylamine hydroxylation in the human. The longitudinal excretion of the trace acids was compared with that of the trace amines.  相似文献

Male bushcrickets transfer a spermatophore at mating that consistsof a sperm-containing ampulla and a product of the accessoryglands, the spermatophylax, that is consumed by the female duringinsemination. Male Requena verticalis produce functionally differentspermatophores depending on the availability of sexually receptivefemales. They will maintain high mating frequency by providinga gift sufficient to ensure sperm transfer, or will invest parentallyin females when their mating frequency is low. We examined therelationship between male quality and nuptial feeding underconditions where males invest in ejaculate protection or inparental investment. When investing in ejaculate protection,males reduced the quality of the spermatophylax meal by reducingboth the concentration of protein and the absolute amount ofprotein it contained. There was no relationship between malephenotype and gift size or quality. Moreover, we could findno evidence for the recently advanced hypothesis that femalescan exercise mate choice by interfering with insemination. However,when males were investing parentally, we found a positive associationbetween spermatophylax size and male size, but no relationshipbetween protein content and male size. Males with high levels offluctuating asymmetry invested more heavily in the nutritionalcontent of their spermatophylaxes than did symmetrical males.Thus, male quality does influence nuptial feeding, but in amanner predicted by a model of indirect fitness benefits frommate choice.  相似文献

The mbuna cichlids of Lake Malawi are a diverse, monophyletic, and recently derived clade. Sexual selection is thought to have accelerated their rapid diversification, though the mechanism by which this has occurred remains unknown. In this study, we examine the effect that male–male interactions have on female preference. We first used a short interaction experiment as a proxy for male dominance. We then measured female preference for (1) a single, isolated male; (2) a group of three, highly ranked males, relative to a group of lower ranked males; and (3) a group of three, mixed-ranked males against a similar group of mixed-rank males. We found that male dominance was highly correlated with male standard length. Female preference for male standard length and for dominance rank was significant in both isolated and group interactions. However, females only showed preference for groups of interacting males when males were segregated by dominance rank, not when groups were composed of both dominant and subordinate males. The results suggest that male–male contests influence female mating decisions. By investigating the rules that dictate the outcomes of such interactions we can elucidate the role that behavior plays in the diversification of this species-rich lineage.  相似文献

Immature males of eastern mosquitofish Gambusia holbrooki start to be sexually active well before their copulatory organ (gonopodium) has completely developed and before they become able to transfer sperm. Sexual activity of males, consisting of copulatory attempts tending to bypass female acceptance, is intense (one attempt per minute) and is likely to be energetically very costly. The sexual behaviour of immature males relative to their maturation stage is described and tested against two possible adaptive explanations. Sexual activity was present in males from the beginning of the development of their gonopodium and increased during the following stages of maturation. Two to three weeks before gonopodium development was completed, sexual activity of immatures was as high as that of adults. Adult males showed aggressive behaviour against a male attempting a copulation, irrespective of the maturity of the latter. Since previous studies have shown that the reproductive success in this species is negatively correlated with male size when male–male competition is low (i.e. when the sex ratio is female biased), but decreases with male size when competition is high, the hypothesis was tested that sexual activity of immature males functions as a way to predict their future reproductive success if they mature at a given size. A second hypothesis tested was that precocious sexual experience improves the efficiency of copulatory attempts. Results were more in agreement with the first hypothesis, since size at maturity of males was influenced by the sex ratio experienced during maturation and precocious experience gave very little advantage.  相似文献

Duetting,or the stereotypical,repeated and often coordinated vocalizations between 2 individuals arose independently multiple times in the Order Primates.Across primate species,there exists substantial variation in terms of timing,degree of overlap,and sex-specificity of duet contributions.There is increasing evidence that primates can modify the timing of their duet contributions relative to their partner,and this vocal flexibility may have been an important precursor to the evolution of human language.Here,we present the results of a fine-scale analysis of Gursky’s spectral tarsier Tarsius spectrumgurskyae duet phrases recorded in North Sulawesi,Indonesia.Specifically,we aimed to investigate individual-level variation in the female and male contributions to the duet,quantify individual-and pair-level differences in duet timing,and measure temporal precision of duetting individuals relative to their partner.We were able to classify female duet phrases to the correct individual with an 80%accuracy using support vector machines,whereas our classification accuracy for males was lower at 64%.Females were more variable than males in terms of timing between notes.All tarsier phrases exhibited some degree of overlap between callers,and tarsiers exhibited high temporal precision in their note output relative to their partners.We provide evidence that duetting tarsier individuals can modify their note output relative to their duetting partner,and these results support the idea that flexibility in vocal exchanges—a precursor to human language—evolved early in the primate lineage and long before the emergence of modern humans.  相似文献

Mating system variation is profound in animals. In insects, female willingness to remate varies from mating with hundreds of males (extreme polyandry) to never remating (monandry). This variation in female behaviour is predicted to affect the pattern of selection on males, with intense pre-copulatory sexual selection under monandry compared to a mix of pre- and post-copulatory forces affecting fitness under polyandry. We tested the hypothesis that differences in female mating biology would be reflected in different costs of pre-copulatory competition between males. We observed that exposure to rival males early in life was highly costly for males of a monandrous species, but had lower costs in the polyandrous species. Males from the monandrous species housed with competitors showed reduced ability to obtain a mate and decreased longevity. These effects were specific to exposure to rivals compared with other types of social interactions (heterospecific male and mated female) and were either absent or weaker in males of the polyandrous species. We conclude that males in monandrous species suffer severe physiological costs from interactions with rivals and note the significance of male–male interactions as a source of stress in laboratory culture.  相似文献

To explore whether selection for testosterone-mediated traits in males might be constrained by costs of higher testosterone to females, we examined the effects of experimental elevation of plasma testosterone on physiological, reproductive, and behavioral parameters in a female songbird, the dark-eyed junco (Junco hyemalis). We used subcutaneous implants to elevate testosterone (T) in captive and free-living female juncos. In captive birds, we measured the effects of high T on body mass, feather molt, and brood patch formation. In the field, we monitored its effects on the timing of egg laying, clutch size, egg size, egg steroid levels, incubation, and nest-defense behavior. Females implanted with testosterone (T-females) had significantly higher circulating levels of testosterone than did control females (C-females). Captive T-females had lower body mass, were less likely to develop brood patches, and delayed feather molt relative to C-females. Among free-living females, the interval between nest completion and appearance of the first egg was longer for T-females than for C-females and egg yolk concentrations of testosterone were higher, but there were no significant differences in estradiol levels, clutch size, or egg size. Incubation and nest defense behavior were also similar between T- and C-females. Our results suggest that selection on males for higher testosterone might initially lead to a correlated response in females producing changes in body mass and feather molt, both of which could be detrimental. Other possible female responses would be delayed onset of reproduction, which might reduce reproductive success, and higher yolk testosterone, which might have either positive or negative effects on offspring development. We found no reason to expect reduced parental behavior by females as a negative fitness consequence of selection for higher testosterone in males.  相似文献

Abstract

This paper studies the effect of having at least one male offspring on a sex‐linked recessive disease and the fraction of affected males due to fresh mutations. The equilibrium frequency of heterozygous females depends not only on the intensity of the reproductive compensation, but also on the time of mutational change. It has been shown that the frequency ranges from 4u without reproductive compensation to √2u or √3u with strict compensation, where u is the mutation rate from the wild type allele to lethal gene. The frequency √2u is achieved when mutation occurs in mature germ cells, whereas, √3u achieved when mutation occurs in early development of germ cells. This increased frequency of heterozygous females due to reproductive compensation reduces considerably the proportion of affected males due to fresh mutation.  相似文献

Anther dehiscence is very important for pollen maturation and release.The mutants of anther dehiscence in rice (Oryza sativa L.) arefew,and related research remains poor.A male sterility mutant of anther dehiscence in advance,add(t),has been found in Minghui 63 and its sterility is not sensitive to thermo-photo.To learn the character of sterilization and the function of the add(t) gene,the morphological and cytological studies on the anther and pollen,the ability of the pistil being fertilized,inheritance of the mutant,and mapping of add(t)gene have been conducted.The anther size is normal but the color is white in the mutant against the natural yellow in the wild-type.The pollen is malformed,unstained,and small in the KI-I2 solution.The anther dehiscence is in advance at the bicellular pollen stage.A crossing test indicated that the grain setting ratio of the add(t) is significantly lower than that of the CMS line 2085A.The ability of the pistil being fertilized is most probably decreased by the add(t) gene.The male sterility is controlled by a single recessive gene of add(t).This gene is mapped between the markers of R02004 (InDel) and RM300 (SSR) on chromosome 2,and the genetic distance from the add(t) gene to these markers is 0.78 cM and 4.66 cM,respectively.  相似文献
