Recent studies describe interactions of pollen surfaces with aerosol particles; pollen surfaces undergo morphological changes and the release of allergens and allergenic fragments from the pollen can be enhanced. Thus allergens from pollen can be found in particle size fractions much smaller than undamaged pollen (<5m). This may explain allergic reactions in parts of the lungs which cannot be reached by undamaged pollen. In Switzerland the birch tree (betula verrucosa) major allergen Bet v 1 and the grass (phleum pratense) pollen major allergen Phl p 5 are of particular relevance for inducing pollinosis. In this study aerosols of different aerodynamic diameters were sampled by Andersen-Impactors over 18 months. Sampling areas are subjected to different levels of air pollution (Zürich, Switzerland, urban; Payerne, Switzerland, rural: Davos, Switzerland, alpine). Samples were scanned by electron microscopy and submitted to specific allergen assays (ELISA) for birch pollen major allergen Bet v 1 and grass pollen major allergen Phl p 5 respectively. Particle and major allergen concentrations were highest in Zürich, followed by Payerne and, significantly lower, Davos. Scanning electron microscopy investigations showed interactions of aerosols with pollen surfaces in Zürich and Payerne. The presence of Bet v 1 in smaller aerosol fractions was demonstrated in Zürich and Payerne some weeks before and after birch pollen was counted.  相似文献

Cuttings of Populus nigra L. cv. Loenen and Populus maximowiczii Henri × Populus nigra L. cv. Rochester were exposed for 6 weeks in open-top chambers to investigate effects of realistic mixtures of ozone, sulphur dioxide, and nitrogen oxides on carbohydrate levels. Whereas the total main nonstructural carbohydrates in the leaves were reduced, those in the roots were nearly not affected. The reduction in leaf carbohydrates was associated with a significant decrease in starch. In contrast, sucrose and especially glucose were increased, causing a shift in the starch to sucrose and the starch to glucose ratios. The effects were strongest in the older leaves of both cultivars, with cv. Loenen, especially, responding to ozonic mixtures. In the petioles, sucrose and glucose were unchanged or, in some cases, decreased. The alterations in carbohydrate levels were coupled to a reduction in both total fresh weight of leaves and total dry matter of roots, and were attributed to an air pollution-dependent demand for energy and carbon in leaves.  相似文献

We investigated the physiological and biochemical differences in Pterocarpus indicus and Erythrina orientalis grown in four sites at different pollution levels in the Philippines: Makati, Pasig and Quezon (high pollution levels; HP) located in Metro Manila, and La Mesa Watershed (a non-polluted area; NP). Among these four areas, HP sites had higher net photosynthetic rates (P N) than NP sites, except for Makati. Among HP sites, Makati and Quezon had the lowest P N for P. indicus and E. orientalis, respectively. Chlorophyll (Chl) contents were significantly lower in HP than in NP sites. Trees in Makati had the lowest Chl contents among HP sites, and P. indicus had higher Chl contents than did E. orientalis. In addition, the chloroplasts in HP trees had small starch grains with numerous dark, large plastoglobuli. Furthermore, antioxidant enzymes, indicative of the defense mechanism, showed a significantly higher activity in HP than in NP trees.  相似文献

Cooking is a significant source of indoor air pollution (IAP) and is associated with significant morbidity and mortality. Liquefied petroleum gas (LPG; predominant cooking fuel of urban India), though safer than solid fuels, emits significant concentration of fine particles, NOx and several other pollutants, which may trigger respiratory ailments. Effect of exposure to pollutants, generated from fuel combustion and cooking activities, on subjects of different microenvironments has been assessed by carrying out a health survey using a questionnaire and by conducting spirometry. A significant increase in prevalence of cough and impairment of lung functions has been found among cooks compared to non-cooks. A significant difference in prevalence of cough between smoker and non-smoker non-cooks but no such difference in cooks suggests that kitchen pollutants and smoking have similar effects on respiratory system. Comparison of lung functions (average FEV1% predicted and average FVC% predicted) of cooks (90.6% and 82.6%) and non-cooks (93.7% and 86.2%) shows a significant difference in certain categories. The study brings out a close association between different kitchen microenvironments and specific respiratory parameters, signifying short- and long-term impairment. The impairment is prevalent in both rural and urban Indian cooks, signifying that fuel usage (biomass and LPG) and cooking practices play a prominent role in affecting respiratory health.  相似文献

BackgroundA growing number of researches indicated the association between plasma trace elements and blood lipids. However, the potential interaction and dose-response relationship were less frequently reported.MethodsIn this study, a total of 3548 participants were recruited from four counties in Hunan Province, South China. Demographic characteristics were collected by face-to-face interviews and inductively coupled plasma mass spectrometry (ICPMS) was used to determine the levels of 23 trace elements in plasma. We applied a fully adjusted generalized linear regression model (GLM) and a multivariate restricted cubic spline (RCS) to estimate the correlation, dose-response relationship and possible interaction between 23 trace elements and four blood lipid markers.ResultsThe results indicated positive dose-response relationships of plasma 66zinc with triglycerides (TG) and low density lipoprotein cholesterol (LDL-C), plasma 78selenium with LDL-C and total cholesterol (TCH), and plasma 59cobalt with high-density lipoprotein cholesterol (HDL-C). There was a negative dose-response relationship between 59cobalt and LDL-C. Further analysis found that 66zinc and 59cobalt had an antagonistic effect on the risk of increased LDL-C level.ConclusionsThis study added new evidence for the potential adverse effects of 66Zn and 78Se on blood lipids, and provided new insight into the threshold value setting for metals as well as the intervention strategy for dyslipidemia.  相似文献

The gene CYTOKININ INDEPENDENT-1 (CKI-1), previously isolated by enhancer trap screening, has been hypothesised to play a role in cytokinin perception. Alternative hypotheses suggest that it is required for the production of cytokinins or that it has no direct role in cytokinin signalling but simply interferes with the pathway when overexpressed. These hypotheses were investigated by producing transgenic Arabidopsis plants expressing CKI-1 cDNA in antisense orientation. In standard conditions, the phenotype of the plants was similar to wild type. Significantly higher amounts of the free base and riboside forms of cytokinin and lower amounts of membrane-impermeable cytokinins were found in the antisense lines. This supports the hypothesis that CKI-1 is involved in cytokinin perception and demonstrates the existence of a feedback loop altering cytokinin metabolism in response to the level of receptor abundance. An elevation in the content of free bases and ribosides of zeatin and isopentenyladenine, along with a reduction in the content of ribotide forms, suggests that a cytokinin ribotide 5'-ribonucleotidase may be a site at which CKI-1 exerts feedback control. When seed homozygous for the transgene was germinated on medium with reduced total mineral nutrient levels, the cotyledons of seedlings with reduced levels of CKI-1 failed to expand and green, and vegetative growth was inhibited. A similar phenotype was observed on low-phosphate media, suggesting that this failure resulted from an interaction between phosphate and cytokinins.  相似文献

The most consistent neurochemical finding in autism has been elevated group mean levels of blood platelet 5-hydroxytryptamine (5-HT, serotonin). The origin and significance of this platelet hyperserotonemia remain poorly understood. The 5-HT(1A) receptor plays important roles in the developing brain and is also expressed in the gut, the main source of platelet 5-HT. Post-natal tissue levels of 5-HT, 5-hydroxyindoleacetic acid (5-HIAA) and tryptophan were examined in the brain, duodenum and blood of 5-HT(1A) receptor-knockout and wild-type mice. At 3 days after birth, the knockout mice had lower mean brain 5-HT levels and normal mean platelet 5-HT levels. Also, at 3 days after birth, the mean tryptophan levels in the brain, duodenum and blood of the knockout mice were around 30% lower than those of the wild-type mice. By 2 weeks after birth, the mean brain 5-HT levels of the knockout mice normalized, but their mean platelet 5-HT levels became 24% higher than normal. The possible causes of these dynamic shifts were explored by examining correlations between central and peripheral levels of 5-HT, 5-HIAA and tryptophan. The results are discussed in relation to the possible role of 5-HT in the ontogeny of autism.  相似文献

The lysosomal neutral red retention time (NRRT) assay, a biomarker for lysosomal membrane stability, and the total immune activity (TIA) assay, a measure of non-specific immune system activity, were used in laboratory studies to assess the toxic effects of 2,4,6-trinitrotoluene (TNT) on earthworms (Eisenia andrei) in vivo. The results were compared with the concentration of TNT and its metabolites in earthworm tissue, as well as standard sublethal toxicity endpoints including growth (i.e. weight change) and reproduction effects from previously published studies. Filter paper experiments indicated a significant decrease in NRRT at ≥1.8 μg TNT cm-2, whereas sublethal (weight loss) and lethal effects to earthworms were detected at ≥3.5 and 7.1 μg TNT cm-2, respectively. Experiments in artificial soil showed that NRRT effects could be detected at lower TNT concentrations (≥55 mg TNT kg-1 soil dry weight) compared with other sublethal endpoints (effects on growth and reproduction). The TIA biomarker did not significantly respond to TNT. Copper (as CuSO4, filter paper contact tests) and 2-chloroacetamide (soil tests), which were used as reference toxicants, also decreased the NRRT. The use of the NRRT assay linked with tissue concentrations of TNT metabolites in earthworms was identified as a potentially appropriate biomarker approach for TNT exposure assessment under laboratory conditions and a novel tool for effects-based risk assessment.  相似文献

The high plasticity of the active-site cavity of cytochromes P450, permitting reactivity toward a vast array of compounds, makes these enzymes attractive targets for biotechnological application. Escalating attention in this area is driven by remarkable progress in the rational design by DNA shuffling of self-sufficient, multi-domain P450/electron donor constructs simplifying the composition of biocatalytic systems. Moreover, versatile approaches were undertaken to supersede the well-established, NAD(P)H-steered proteinaceous redox chains by cost-effective alternative electron transfer conduits constituted of organometallic mediators or photoactivatable redox triggers. Electrochemical techniques have proven particularly useful: employing different types of carbon- and metal-based electrodes for the fabrication of biosensors, the continuing challenge was to optimize the conductive properties of these devices by creating biocompatible interfaces for transferring electrons between sensor surfaces and redox proteins. The present review provides a critical update of the most significant breakthroughs in innovative manipulation of the redox machinery, giving an impulse to exploitation of P450s in fields such as the production of fine chemicals, drug processing, medicinal diagnostics and remediation of biotopes contaminated with harmful environmental pollutants.  相似文献

The incidence of pancreatic cancer is increasing in both developed and developing Nations. In recent years, various research evidence suggested that reprogrammed metabolism may play a key role in pancreatic cancer tumorigenesis and development. Therefore, it has great potential as a diagnostic, prognostic and therapeutic target. Amino acid metabolism is deregulated in pancreatic cancer, and changes in amino acid metabolism can affect cancer cell status, systemic metabolism in malignant tumor patients and mistakenly involved in different biological processes including stemness, proliferation and growth, invasion and migration, redox state maintenance, autophagy, apoptosis and even tumor microenvironment interaction. Generally, the above effects are achieved through two pathways, energy metabolism and signal transduction. This review aims to highlight the current research progress on the abnormal alterations of amino acids metabolism in pancreatic cancer, how they affect tumorigenesis and development of pancreatic cancer and the application prospects of them as diagnostic, prognostic and therapeutic targets.  相似文献
