In order to improve vegetative propagation of Berberis thunbergii ‘Red Rocket’, stock plants were subjected to shading and IBA treatment, as well as to the application of two commercially available rooting powders. Spraying stock plants with IBA 24 hours prior to cutting harvest gave the effect comparable to the use of rooting powders. Reduction of light intensity by 50% and 96% of ambient prior to harvest of cuttings improved rooting. Positive effect of shading results in changes in shoot anatomy, i.e. a weaker sclerenchyma development. Enhanced rooting in cuttings from shoots grown under reduced light intensity was accompanied by a decrease in the concentration of soluble proteins and by an increase in total chlorophyll content while the concentration of total soluble sugars remained unaffected.  相似文献

The role of ethylene in adventitious root formation and its involvement in auxin-induced rooting were investigated in cuttings ofVigna radiata (L.). Treatment with 30 M indole-3-acetic acid (IAA) for 24 h slightly inhibited rooting, whereas the same concentration of indole-3-butyric acid (IBA) significantly stimulated it. Ethylene derived from 1-aminocyclopropane-1-carboxylic acid (ACC) increased the number of adventitious roots but inhibited their emergence and elongation. Endogenous levels of ethylene, ACC, and malonyl-ACC (MACC) were initially higher in cuttings treated with IAA. This trend was quickly reversed, and cuttings, particularly hypocotyls, treated with IBA produced higher levels of ethylene and had more ACC and MACC during most of the rooting process. Aminoethoxyvinylglycine significantly inhibited rooting, but its inhibitory effect could not be reversed by ACC. The data suggest that the stimulating effect of IBA on rooting is closely associated with its induction of ACC and ethylene biosynthesis.  相似文献

We have previously shown that both endogenous auxin and ethylenepromote adventitious root formation in the hypocotyls of derootedsunflower (Helianthus annuus) seedlings. Experiments here showedthat promotive effects on rooting of the ethylene precursor,1-aminocyclopropane-l-carboxylic acid (ACC) and the ethylene-releasingcompound, ethephon (2-chloro-ethylphosphonic acid), dependedon the existence of cotyledons and apical bud (major sourcesof auxin) or the presence of exogenously applied indole-3-aceticacid (IAA). Ethephon, ACC, aminoethoxyvinylglycine (an inhibitorof ethylene biosynthesis), and silver thiosulphate (STS, aninhibitor of ethylene action), applied for a length of timethat significantly influenced adventitious rooting, showed noinhibitory effect on the basipetal transport of [3H]IAA. Theseregulators also had no effect on the metabolism of [3H]IAA andendogenous IAA levels measured by gas chromatography-mass spectrometry.ACC enhanced the rooting response of hypocotyls to exogenousIAA and decreased the inhibition of rooting by IAA transportinhibitor, N-1-naphthylphthalamic acid (NPA). STS reduced therooting response of hypocotyls to exogenous IAA and increasedthe inhibition of rooting by NPA. Exogenous auxins promotedethylene production in the rooting zone of the hypocotyls. Decapitationof the cuttings or application of NPA to the hypocotyl belowthe cotyledons did not alter ethylene production in the rootingzone, but greatly reduced the number of root primordia. We concludethat auxin is a primary controller of adventitious root formationin sunflower hypocotyls, while the effect of ethylene is mediatedby auxin. We have analyzed the effect of N,N′-bis-(2,3-methylenedioxyphenyl)urea (2,3-MDPU) and N,N′-bis-(3,4-methylenedioxyphenyl)urea (3,4-MDPU), two symmetrically substituted diphenylurea derivatives with no auxin or cytokinin-like activity, on the rooting capacity of Pinus radiata stem cuttings. Results indicate that both diphenylurea derivatives enhance adventitious rooting in the presence of exogenous auxin (indole-3-butyric acid, IBA), even at low auxin concentration, in rooting-competent cuttings, but have no effect on the adventitious rooting of low or null competent-to-root cuttings. Histological analyses show that, in the simultaneous presence of MDPUs and low concentration of exogenous auxin, adventitious root formation is induced in the cell types that retain intrinsic competence to form adventitious roots in response to auxin. The time course of cellular events leading to root formation and the time of root emergence are closely similar to that observed in cuttings treated only with higher auxin concentration. In addition, the mRNA level of a P. radiata SCARECROW-LIKE gene, which is significantly induced in the presence of the optimal concentration (10 μM) of exogenous auxin needed for cuttings to root, is increased in the presence of MDPUs and low concentration of exogenous auxin (1 μM). The expression of a P. radiata SHORT-ROOT gene in rooting-competent cuttings during adventitious rooting is also affected by the presence of MDPUs when combined with auxin. As MDPUs do not affect the expression of either gene in the absence of exogenous auxin, but only in its presence, we suggest that MDPUs could interact, directly or indirectly, with the auxin-signalling pathways in rooting-competent cuttings during adventitious rooting.  相似文献

The effects of post harvest application of ethylene, abscisic acid (ABA), indole-3-butyric acid (IBA) treatments or dark storage on root induction and continued growth of regenerated roots in Pelargonium cuttings were investigated using hydroponics in the greenhouse. Ethylene markedly increased rooting percentage in ‘Greco’ and ‘Surfing’, reduced the number of roots per cutting in ‘Surfing’ and had no effect on the total root lengths in the two cultivars. Ethylene treatment reduced fresh root mass in ‘Surfing’, increased dry root mass and reduced root water content in both cultivars. ABA (50 μM) enhanced rooting percentage in ‘Greco’, reduced the number of roots per cutting, reduced total root lengths and fresh root mass in both cultivars. ABA increased dry root mass and reduced root water content in ‘Surfing’ but this effect was not apparent in ‘Greco’. Storing cuttings in the dark for 4 days had no effect on rooting percentage and number of roots per cutting in ‘Greco’ and ‘Surfing’. However, dark storage reduced total root lengths in ‘Surfing’ and reduced fresh root mass in ‘Greco’. Dark storage had no effect on dry root mass and water content in both cultivars. Applying 4 μl l−1 IBA in the rooting solution induced maximum (100%) root induction in ‘Surfing’. However, IBA reduced the number of roots per cutting in ‘Greco’, reduced total root lengths and fresh root mass in the two cultivars. IBA treatment profoundly increased and reduced dry root mass and root water content, respectively, in ‘Greco’ and ‘Surfing’. The enhanced root induction observed after IBA and ABA applications could be ascribed to their influence on ethylene biosynthesis, since ethylene treatment increased rooting percentage in both cultivars. However, high ABA (100 μM) and IBA (12 μl l−1) levels or dark storage reduced the ability of induced roots to continue growth. We attribute our results to plant stress-response mechanism and ethylene appears to play an important role in the process of root initiation and root growth in Pelargonium cuttings.  相似文献

We have re-examined the role of ethylene during rooting of mung bean cuttings. Cuttings were treated for 5 days with a low or a high concentration of NAA (naphthaleneacetic acid). During this 5 days period, we also applied STS (silverthiosulfate, an inhibitor of ethylene action) or ACC (1-aminocyclo-propane-l-carboxylic acid, a direct precursor of ethylene). At high NAA concentration, STS promoted and ACC inhibited rooting, respectively. At low NAA concentration, the effects were opposite, STS being inhibitory and ACC promotive. AVG (aminoethoxyvinylglycine, an inhibitor of ethylene synthesis) gave similar results as STS. Together, these data suggest supraoptimal and suboptimal ethylene levels in the tissue at high and low NAA concentration, respectively. We also examined whether the effect of ethylene varied during the successive phases of the rooting process. Thus, we gave 24 h pulses with either STS or ACC during the rooting treatment. During the first two days (0–48 h), ACC-pulses were promotive and STS-pulses inhibitory. Later on (48–168 h), the ACC-pulses were inhibitory and the STS-pulses promotive. Whether this effect was observed or not was dependent on the NAA concentration. These data indicate that ethylene promotes or inhibits rooting depending on the stage in the rooting process. When ACC was added only during the initial period, rooting was increased at all NAA concentrations in a NAA dose-response curve and the optimal NAA concentration remained the same. This suggests that ethylene renders more cells responsive to NAA.  相似文献

研究了日本落叶松母株年龄、插穗内源激素含量与生根之间的关系,以及外源IBA对插穗内源激素含量的影响及其对插穗生根的促进作用。结果表明:不同株龄插穗生根性状及插穗茎和叶中激素含量差异均达极显著水平,叶中激素含量对插穗生根力没有直接影响;插穗茎中生根抑制激素(ABA)含量随株龄增长而增加,生根促进激素与抑制激素的比值(IAA+GA+ZR)/ABA却随株龄的增长而递减,与生根力随株龄的变化趋势一致,且该比值与生根性状紧密相关,因此可作为评价母株(无性系)生根力的指标;插后13~32d是插穗愈伤组织形成和不定根诱导的关键期,此期生根促进激素消耗量大,茎中含量大幅度降低,进入根伸长生长阶段,含量上升;外源IBA促进插穗生根的机制在于通过外源激素的刺激,在不定根诱导期,插穗茎中ABA含量大幅度降低,从而有利于不定根的发生和发育。  相似文献

研究生长素、乙烯和一氧化氮(NO)对拟南芥下胚轴插条形成不定根的调节,以及生长素和乙烯信号转导成员在IAA促进不定根形成中的作用的结果表明:拟南芥切条以IAA和硝普钠(N0供体)单独处理7d后的不定根形成均受到促进,其中以50μmol·L^-1 IAAμmol·L^-1 SNP的促进作用为最强,乙烯的促进作用不明显;生长素运输和信号转导以及乙烯信号转导相关突变体对IAA促进生根作用的敏感性比野生型有所下降,特别是IAA14功能获得型的突变体。IAA和NO在促进不定根形成中有协同效应。  相似文献

Abstract Seedlings of Pinus sylvestris L. were grown under controlled conditions (temperature 20°C, photoperiod 17 h) at two irradiances, 8 or 40 W m-2. Hypocotyl cuttings were excised and rooted at different irradiances in tap water solutions of indolebutyric acid (IBA). The fastest rooting and highest rooting percentage were obtained with cuttings from stock plants grown at 8 W m-2 and treated with 10-5M IBA for 21 days. The concentration of 10-4M IBA inhibited root formation. In comparable treatments rooting was always better in cuttings from stock plants grown at 8 W m-2 than in cuttings from stock plants grown at 40 W m-2. The irradiance during the rooting period had only a minor influence on rooting. When cuttings from plants irradiated with 40 W m-2 were treated with 10-5M IBA for 21 days the rooting percentage almost reached the same level as in untreated cuttings from stock plants given 8 W m-2. In cuttings treated with IBA during the whole rooting period, rooting was depressed in comparison to untreated cuttings. Aeration of the 10-4M IBA solution increased the rooting percentage, but aeration had no effect on untreated cuttings and on cuttings treated with lower IBA concentrations.  相似文献

An attempt was made to induce rooting from single node cuttings of Camellia sinensis var. TV-20 under controlled conditions and study its biochemical changes during rooting. The nodal cuttings were pretreated with different concentrations of IAA, NAA and IBA and kept in a growth chamber (25 ±2 °C, 16 h photoperiod (55 μ mol m−2 s−1) with cool, white fluorescent lamps and 65% relative humidity) for 12 h. Among the three auxins used for pretreatment, IBA showed more positive response on rooting as compared to IAA and NAA within 2 weeks of transfer to potting medium. Among four concentrations of IBA tested, 75 ppm gave maximum percentage of rooting, number of roots and root length. Therefore, IBA was used further in experiments for biochemical investigation. The adventitious rooting was obtained in three distinct phases i.e. induction (0–12 days), initiation (12–14 days) and expression (14–18 days). IAA-oxidase activity of IBA-treated cuttings increased slightly as compared to control. The activity was found to decrease during induction and initiation phases and increase during expression phase. The peroxidase activity in IBA-treated cuttings increased up to initiation phase and declined at the expression phase. Polyphenoloxidase activity increased both in IBA-treated and control cuttings during induction and initiation phase but declined slowly during expression phase. Total phenolic content was higher in IBA-treated cuttings, particularly in initiation and expression phases and it also correlated with peroxidase activity. Phenolics might be playing key role for induction of adventitious rooting, and phenolic compounds can be used as rooting enhancer in tea plant.  相似文献

Cuttings of Arbutus unedo (strawberry tree) and Taxus baccata (yew) were treated with 8.0 and 10.0 g l–1, respectively, of KIBA, IBA, IAA, NAA and Paclobutrazol. No rooting occurred without growth regulator treatment. The effect of growth regulators on percentage of rooting followed the order KIBA > IBA > IAA = NAA = Paclobutrazol = 0% (for A. unedo) and KIBA > IBA > IAA > NAA > Paclobutrazol = 0% (for T. baccata). Genotypes of the above plant species had significant effects on the number and length of roots, percentage of rooting and peroxidase specific activity (PA) on KIBA-treated cuttings. High PA seems to be related with low percentage of rooting in the case of A. unedo cuttings while no similar results were noticed in the case of T. baccata. Electrophoretic analysis revealed the appearance of two to three anionic and one cationic peroxidase isoforms in A. unedo cuttings, while six to nine anionic and no cationic peroxidases isoforms appeared in the case of T. baccata genotypes. During adventitious rooting, the PA showed the three interdependent phases (induction, initiation, expression) in both K-IBA treated cuttings of A. unedo and T. baccata, but in a different time course.  相似文献

Untreated and indole-3-butyrie acid-treated (IBA) cuttings from 90-day-old Pinus banksiana Lamb, stock plants were propagated under normal greenhouse irradiance (max. 900 $$mol m-2 s-1) and shade (max. 120 $$mol m-2 s-1) to determine effects on adventitious rooting and on reducing sugar and starch concentrations in needles and basal stems. In one experiment, cuttings were assessed at days 15 and 25 of propagation for basal 1-cm stem fresh weight, proportion rooted, number of roots and longest root length. In a second experiment with cuttings, basal 1-cm stem fresh weight and concentrations of reducing sugar and starch in needles and basal stems were measured each day for the first 10 days of propagation. Carbohydrate measurements were also made for seedling stock plants as controls for the second experiment. Carbohydrate data for cuttings were primarily evaluated based on net (cutting minus seedling) concentrations, to correct for changes in cuttings not related to adventitious rooting. Increase of basal stem fresh weight and rooting of cuttings, based on all measured variables, occurred in the order: light + IBA > light > shade + IBA > shade. The best rooting required the greater irradiance. Compared to results from cuttings in the light, shading resulted in lesser accumulations of reducing sugars and starch in needles and basal stems. Reducing sugar: starch concentration ratios were significantly greater in shade- vs light-propagated cuttings, IBA treatment did not offset the effects of shade on rooting or on reducing sugar and starch concentrations or ratios. Overall, the results suggested that decreased reducing sugar and starch concentrations and/or their increased ratios are associated with shade-induced poor rooting of P. banksiana cuttings.  相似文献

For perennial woody plants, softwood cutting is an efficient technique for larger scale propagation and adventitious rooting of cuttings is one of the most crucial steps. To evaluate the significance of juvenility on adventitious rooting, rooting rates was compared between softwood cuttings collected from apomictic seedlings (juvenile), in vitro cultured plants (rejuvenated), suckers (juvenile like) and canopy shoots (adult) of reproductively mature trees in Malus xiaojinensis. After pre-treatment with indole-3-butytric acid (IBA) (3,000 mg L?1) + H2O2 (50 mM), rooting rates in cutting from juvenile, juvenile like and rejuvenated donor plants were significantly higher (>90 %) than that from adult trees. The effects of IBA on adventitious rooting were enhanced significantly by exogenous H2O2. After 15 passages of in vitro subculture, the micro-shoots from adult phase explants were rejuvenated successfully, marked by the elevated expression of miR156 in the leaflets of the micro-shoots. But the rooting ability of rejuvenated micro-shoots was recovered delayed at the 18th or 21st passage of subculture. During the process of rejuvenation, the leaf indole-3-acetic acid contents and the expressions of rooting related genes CKI1, ARRO-1, ARF7 and ARF19 increased significantly. In contrary, the leaf abscisic acid contents decreased. A lack of juvenility is the most important limiting factor governing adventitious rooting of softwood cuttings in apple rootstocks.  相似文献

Proliferating shoot cultures of black mulberry (Morus nigra), derived from axillary buds of two donor trees designated as Mn1 and Mn2, more than 80 years of age, were established in vitro. Subsequently, shoot-tips were used to induce both axillary and adventitious shoot regeneration following incubation on Murashige and Skoog medium containing 14 different treatments of various concentrations of plant growth regulators, including 6-benzyladenine (BA), thidiazuron (TDZ), and combinations of BA with indole-3-butyric acid (IBA) and TDZ with BA. The highest shoot proliferation of 5.3 shoots per explant for the Mn1 tree and 6.9 shoots per explant for the Mn2 tree were obtained when explants were incubated on a medium containing 0.5 mg l−1 BA and 0.1 mg l−1 IBA. The maximum frequency of adventitious rooting was similar for both genotypes. Changes in lignin and cellulose content, macromolecular properties of dioxane and Klason lignins, lignin monomer composition, and macromolecular properties of cellulose were determined in 1-year-old and 3 year-old micropropagated plants, as well as in the donor trees. Lignin and cellulose properties were significantly dependent on the genotype, the age and the mutual interaction of both these factors. The syringyl to guaiacyl weight ratio in lignin rose with the age of the micropropagated plants. Moreover, the tensile strength of wood in 1-year-old plants was supported by a high cellulose degree of polymerization. The highest polydispersity index of cellulose was detected in 3-year-old plants.  相似文献

Stem cuttings of Chrysanthemum morifolium Ram. were treatedwith aqueous solutions of (2-chloroethyl)phosphonic acid (Ethrel)and indolebutyric acid (IBA). Ethrel (1 mg per 1) as a dip oras two foliar sprays, promoted root length and branching inMrs. Roy and Clipper which are difficult-to-root cultivars,but had no effect on Improved Mefo, an easy-to-root cultivar.IBA increased root number in both Clipper and Improved Mefo.The results suggest that IBA and Ethrel act at different stagesof the rooting process, with IBA promoting initiation and Ethrelstimulating elongation and branching. Increases in root numberwith IBA treatment, and in root length with Ethrel treatment,were accompanied by decreases in the soluble carbohydrate concentration,particularly in stem bases.  相似文献

Paclobutrazol (PB), a triazole growth retardant and an inhibitor of gibberellin biosynthesis, reduced at 17 μM concentration the adventitious root formation of bean primary leaf cuttings. Treatments with 5 μM ABA or 4 μM Ethrel, an ethylenereleasing compound, restored the rooting of PB-treated cuttings. Ethylene production and the content of the precursor 1-aminocyclopropane-l-carboxylic acid (ACC) were enhanced in root-forming tissues of PB-treated petioles 48 h after ABA application. The effect of ABA could be abolished by 10 μM CoCl2, an inhibitor of ACC oxidase. Thus, ABA might stimulate rooting through its effect on ethylene release. 2 mM silver thiosulphate, an inhibitor of ethylene action, decreased the rooting of PB-treated cuttings similarly to Co2+, but failed to negate the ABA effect. Light-grown mung bean (Phaseolus aureus Roxb.) cuttings were treated with buffered and nonbuffered solutions of Ethephon, indole butyric acid (IBA), and the combination of both. Ethephon treatment resulted in increased tissue ethylene levels with increasing solution pH, but had no effect on rooting. IBA treatment had no effect on tissue ethylene levels, but strongly promoted rooting. Combinations of Ethephon and IBA had no effect on rooting of mung bean cuttings beyond that obtained by IBA alone.  相似文献

In order to improve vegetative propagation of a difficult to root Cotinus coggygria the stock plants were subjected to: etiolation, shading and spraying with IBA, combined with the application of two commercially available rooting powders. The IBA treatment was more suitable for rooting of C. coggygria cuttings than the NAA application and it enhanced rhizogenesis regardless of the form of auxin application (foliar application to a stock plant or a rooting powder used directly on cuttings) and the amount of light provided to stock plants. Etiolation did not improve rhizogenesis in stem cuttings, however, reduction of light intensity by 50% and 96% of the ambient prior to harvest of cuttings affected rooting positively. Positive effects of shading can be ascribed to changes in shoot anatomy, i.e. a weaker sclerenchyma development. Synergistic effect of shading and foliar auxin application can result from the increase in leaf blade area and/or thinner lower epiderm. Enhanced rooting in cuttings from shoots grown out under reduced light intensity was accompanied by decrease in the contents of total soluble sugars, soluble proteins and free ABA and by increase in total chlorophyll, free amino acids, polyphenolic acids and free IAA contents.  相似文献
