We succeeded in inducing conjugation of Spirogyra castanacea by incubating algal filaments on agar plate. Conjugation could be induced using clone culture. The scalariform conjugation was generally observed, while lateral conjugation was rarely. When two filaments formed scalariform conjugation, all cells of one filament behaved as male and those of other filament did as female. Very rarely, however, zygospores were formed in both of pair filaments. The surface of conjugation tube was stained with fluorescently labeled-lectins, such as Bandeiraea (Griffonia) simplicifolia lectin (BSL-I) and jacalin. BSL-I strongly stained the conjugation tubes, while weakly did the cell surface of female gamete first and then that of male gamete. Jacalin stained mainly the conjugation tubes. Addition of jacalin inhibited the formation of papilla, suggesting some important role of jacalin-binding material at the initial step of formation of the conjugation tubes.  相似文献

The tube building polychaete Lanice conchilega is a common and ecologically important species in intertidal and shallow subtidal sands. It builds a characteristic tube with ragged fringes and can retract rapidly into its tube to depths of more than 20 cm. Therefore, it is very difficult to sample L. conchilega individuals, especially with a Van Veen grab. Consequently, many studies have used tube counts as estimates of real densities. This study reports on some aspects to be considered when using tube counts as a density estimate of L. conchilega, based on intertidal and subtidal samples. Due to its accuracy and independence of sampling depth, the tube method is considered the prime method to estimate the density of L. conchilega. However, caution is needed when analyzing samples with fragile young individuals and samples from areas where temporary physical disturbance is likely to occur.  相似文献

Bioclast lined tubes of Schaubcylindrichnus coronus are not much reported around the world. As of now only two reports are available. Interestingly in both these reports and in the current find, tests of foraminifers are used as tube building material. Current report of Schaubcylindrichnus is from the subtidal deposits of the Naredi Formation (Ypresian) of Kachchh Basin, Western India. As the host rock here abounds in the tests of larger benthic foraminifera, concentration of same is noticed within the tubes. The natural selection of lighter and larger calcitic grains of tests over the heavier mineral grains is the main reason for presence of the tests in tube building; moreover, presence of inner smooth wall of tube also indicates organisms’ preference for comfort within their burrow.  相似文献

Summary Effect of cumin seed exudates on spore germination of Curvularia lunata, Alternaria solani and Colletotrichum capsici was studied in the laboratory. Three-hour exudate obtained for first 3-hour favoured initial protrusion but inhibited subsequent growth of the germ tubes. Two-hour exudate obtained between 5 and 7 hours after soaking was suggested to have no effect on initial protrusion but was found to have promotive effect on subsequent growth of germ tube. Short treatment with inhibitor for 45 minutes followed by a treatment with promotor ensured high germination and uniform germ tube growth. Abstract. 1. Investment in silken tubes may give tubicolous chironomid larvae more to lose than non‐tube building taxa when physical disturbances or the actions of other organisms induce dispersal. In this study, two chironomid species from flood‐prone Australian tropical streams were subjected to repeated experimental destruction of their larval and pupal tubes in laboratory experiments, with life‐history parameter responses recorded. 2. When subjected to the tube destruction treatments, Echinocladius martini (Orthocladiinae), whose larvae construct branched loose silken tubes in fast flow, showed no reduction in survivorship to adult, whereas Polypedilum australotropicus (Chironominae), which builds compact tightly woven tubes incorporating detrital material within pool leaf packs, showed reduced survivorship of about 25%. 3. For both species, tube destruction reduced pupal duration, regardless of whether destruction occurred in the larval stage only, or in both the larval and pupal stages. This may be a response to the risks of exposure outside tubes in nature, particularly for pupae, which are incapable of spinning silk. 4. Tube destruction caused little or no reduction in adult size, longevity, or female fecundity (oocyte number) for either species, but oocyte development and size was retarded in E. martini females, reflecting the energetic and nutritive costs of the tube destruction treatments. 5. Contrasts in the responses of the two species reflect differences in tube‐building behaviour and in situ habitat use. These results demonstrate that tube loss can affect the fitness of individual chironomids, though both species proved generally resilient, which may reflect their long‐term exposure to variable tropical lotic conditions.  相似文献

Rocellaria dubia bores into subtidal rocks of karsted limestone in the Adriatic Sea and elsewhere. It also bores into the shells of various bivalve species. The mechanism of boring has hitherto been debated, but examination of occupied shells suggest that this is achieved by mechanical (the shell) abrasion and chemical etching using secretions produced from glands in the anterior mantle. Fast‐growing bivalves such as Ostrea edulis and Pinna nobilis carry heavy R. dubia burdens, and encapsulate the borer in secreted calluses. Slow‐growing bivalves such as the burrowing Venus verrucosa and Glycymeris violacescens carry low R. dubia burdens, are less able to encapsulate the borers, and probably incur enhanced mortalities as a result. Individuals of R. dubia removed from their limestone boreholes re‐secreted adventitious tubes around their siphons, probably from glands in the posterior mantle. The lifestyle of R. dubia is now better understood, and its ability to bore bivalve shells in particular suggests how the more advanced tropical gastrochaenids Cucurbitula and Eufistulana have evolved from initial (as juveniles) bivalve shell borers into occupants of adventitious crypts and tubes, respectively. Analysis of 66 samples from DSDP Site 263 (Cores 263-4R-4 to 263-29R-4) reveals a unique faunal composition with a predominance of agglutinated taxa, many of them previously unrecorded from any other DSDP and ODP Indian Ocean sites. A total of 66 agglutinated and 31 calcareous taxa are documented and five new species are described: Hippocrepina gracilis n.sp., “Textulariopsiselegans n.sp., Aaptotoichus challengeri n.sp., “Gaudryinopsispseudobettenstaedti n.sp. and “Gaudryinacuvierensis n.sp. Three assemblages are recognized based on changes in the composition of dominant taxa and occurrences of stratigraphically important species: (1) a high-diversity Valanginian to Barremian Bulbobaculites-Recurvoides Assemblage (Cores 263-29R to -18R), comprised of numerous elongate agglutinated forms, rare nodosariids, and variable numbers of tubes and ammodiscids; (2) a moderately diverse Aptian to Albian Rhizammina-Ammodiscus-Glomospira Assemblage (Cores 26318R to -7R) with highly fluctuating numbers of the nominate taxa and Haplophragmoides, Trochammina, Verneuilinoides spp., and Vemeuilina howchini; (3) a very low diversity Albian or younger Assemblage (Cores 263-6R to -4R) containing sparse agglutinated foraminifera, rare nodosariids and rotaliids. We interpret the assemblages as shelf to lower slope and consider them to reflect a deepening palaeobathymetry as the Cuvier margin subsided after the initial breakup of East Gondwana during the Valanginian. Our interpretation is in sharp contrast with initial palaeodepth estimates of less than 100 m, as well as with original chronostratigraphic interpretations based on foraminifera and nannofossils which correlated the base of the recovered interval with the Aptian. The absence of many cosmoplitan forms, despite high diversity, suggests strong faunal differentiation in the Austral realm or endemisn within the Cuvier Basin during the Early Cretaceous.  相似文献

Summary The calcareous housing tubes of fossil species of the polychaete wormsDiplochaetetes andDodecaceria from the Tertiary of Lower California (Mexico) are composed, as well as the tubes of recent species ofDodecaceria, by micritic, peloidal lamellae. The wall ofDiplochaetetes spp. consists of only a few, that ofDodecaceria spp. of numerous lamellae. They form a stromatolitic fabric with intercalated lenses of fibrous calcite/aragonite. Histological investigation of the tubes ofDodecaceria showed that the tube formation is related to two processes. The initial process is weakly controlled by the worm itself (matrix mediated). The worm produces acidic organic mucus substances which are enriched between the soft tissue and the tube wall. The entire mucus inhibits the mineralization of the mucus for a certain time. The mineralization events of the mucus are responsible for the stromatolitic microfabric. Within the spaces between the primary lamellae non-specific EPS (extracellular polymeric substances)-rich mucus is enriched, which controls the formation of fibrous aragonitic crystals and peloidal fabrics. This is a very characteristic organomineralization process which is not controlled directly by the organism. The entire process is comparable with those seen in complex rigid modern microbialites from salt and alkaline lakes in Western Australia and Nevada.  相似文献

Two new polypores (Ceriporiopsis lavendula B. K. Cui sp. nov. and Skeletocutis inflata B. K. Cui sp. nov.) are described from the Guangdong Province in southern China. Ceriporiopsis lavendula is characterized by having resupinate basidiocarps with lavender to grayish blue pore surface and very thin to almost absent subiculum, a monomitic hyphal system with clamped generative hyphae, and oblong‐ellipsoid to ellipsoid basidiospores. Moreover, it grows on wood of Castanopsis exclusively. Skeletocutis inflata is characterized by having pileate basidiocarps, smaller pores (8–10 per mm), inflated and partly dissolved skeletal hyphae in KOH. In addition, its tramal skeletal hyphae are parallel along the tubes. Identification keys to species of Ceriporiopsis and Skeletocutis occurring in China are provided.  相似文献

The objective of this study was to evaluate the influences of detritus from the leaves of different species, and of exposure time on invertebrate colonization of leaves in a shaded Cerrado stream. We hypothesized that the exposure time is the main factor that influences the colonization of leaves by invertebrates. We used leaves of five tree species native to the Brazilian Cerrado: Protium heptaphyllum and Protium brasiliense (Burseraceae), Ocotea sp. (Lauraceae), Myrcia guyanensis (Myrtaceae), and Miconia chartacea (Melastomataceae), which are characterized by their toughness and low-nutritional quality. Litter bags, each containing leaves from one species, were placed in a headwater stream and removed after 7, 15, 30, 60, 90, and 120 days. The dominant taxon was Chironomidae, which comprised ca. 52% of all organisms and ca. 20% of the total biomass. The taxonomic richness of colonizing organisms did not vary among the leaf species. However, the density and biomass of the associated organisms varied differently among the kinds of detritus during the course of the incubation. The collector-gatherers and shredders reached higher densities in the detritus that decomposed more rapidly (Ocotea sp. and M. guyanensis), principally in the more advanced stages of colonization. The collector-filterers reached higher densities in the detritus that decomposed more slowly (P. heptaphyllum, P. brasiliense, and M. chartacea), principally in the initial stages of incubation. A cluster analysis divided the detritus samples of different leaf species according to the exposure time (initial phase: up to 7 days; intermediate phase: 7–30 days; advanced phase: 30–120 days), suggesting some succession in invertebrate colonization, with differences in taxon composition (indicator taxa analysis). These results suggest that regardless of the leaf-detritus species, exposure time was the main factor that influenced the colonization process of aquatic invertebrates.  相似文献

Summary In vitro penetration of the micropyle of freshly isolatedGasteria verrucosa ovules by pollen tube was monitored on agar medium. 40–60% of the micropyles were penetrated, comparable with in vivo penetration percentages. When germinated on agar,Gasteria pollen tube elongation lasts for up to 8 h while plasma streaming continues for about 20–24 h. The generative cell divides between 7 and 20 h after germination, and after 20 h the pollen tube arrives at one of the synergids. The sperm cells arrive after 22 h. The whole process takes more time in vitro than in vivo. In fast growing pollen tubes, a pulsed telescope-like growth pattern of tube elongation is observed. The formation of pollen tube wall material precedes tube elongation and probably prevents regular enlargement of the pollen tube tip-zone. Rapid stretching of the new pollen tube wall material follows, probably due to gradually increased osmotic pressure and the use of lateral wall material below the tip. The stretching ceases when the supplies of plasma membrane and excretable wall material are exhausted. Multiple pollen tube penetration of the micropyle occurs in vitro as it does in vivo. Most pollen tube growth ceases within the micropyle but, if it continues, the pollen tubes curl. Inside the micropyle the pollen tube shows haustorial growth. At the ultrastructural level, the wall thickening of in vitro pollen tubes is quite similar to that in vivo. Before transfer of pollen tube cytoplasm a small tube penetrates one of the synergids. Sperm nuclei with condensed chromatin are observed in the pollen tube and the synergid. In vivo prometaphase nuclei are found in the most chalazal part of a synergid, against the egg cell nucleus and nucleus of the central cell at a later stage. Using media forLilium ovule culture,Gasteria ovules were kept alive for at least 6 weeks. Swelling of the ovule depends on pollen tube penetration. The conditions for fertilization to occur after in vitro ovular pollination seem to be present.  相似文献

Despite the increasing importance of airlift fermentors, very little published information is available on how the geometric configurations of the draft tubes and the air-sparging system affect the mixing and oxygen transfer characteristics of the fermentor. A 14-L air-lift fermentor was designed and build with a fixed liquid height to diameter ratio of 1.5 utilizing four equally spaced air jets at the bottom. Two jet orifice sizes were used, 1.27 and 3.81 mm i.d., and for each jet size the following four geometric configurations were used: Single inner concentric draft tube, single outer concentric draft tube, two concentric draft tubes, and no draft tubes where the fermentor was operated as a shallow bubble column. It was found that the presence of draft tubes stabilized liquid circulation patterns and gave systemically higher mixing times than those obtained in the absence of draft tubes. In addition, the double draft tube geometry resulted in higher mixing times than the single draft tubes. For the power unit volume range 20 to about 250 W/m3 the larger 3.81-mm orifices gave systemically higher kL a values than the smaller 1.27-mm i.d. orifices. At 200 W/m3 the use of a single outer draft tube with the 3.81-mm orifices resulted in 94% increase in kL a values over that obtained with no draft tubes. However, the effect of draft tube geometry on kL a values when the 1.27-mm orifices were used was not significant. The air bubble formation characteristics at the jet orifices were found to be different, which reflected the differences observed in mass transfer and mixing characteristics. The power economy for oxygen transfer was found to be depend strongly on the orifice size and less on the geometric configuration of draft tubes.  相似文献

 Pollen tube and female gametophyte interactions in Douglas fir (Pseudotsuga menziesii) were examined in vitro. Formation of pollen tubes in Douglas fir occurred on a modified Murashige and Skoog medium in which concentrations of H3BO3 and Ca(NO3)2 were altered and supplemented with sucrose and polyethylene glycol. Addition of 100 μg/ml H3BO3 and 300 μg/ml Ca(NO3)2 resulted in optimum pollen viability. Lack of H3BO3 inhibited pollen tube formation. Addition of H3BO3 and Ca(NO3)2 significantly increased pollen tube formation within one week in culture. Using a medium supplemented with mannitol, viability of Douglas fir pollen can be sustained for 7 weeks in culture, about the same length of time as in vivo. However, pollen tubes are not formed. This suggests that the factors responsible for tube formation reside in the external environment of the pollen. Culture of female gametophytes to examine egg viability and longevity had not been done previously. We found that egg viability in culture is short-lived, and therefore the window to study and manipulate events of fertilization in Douglas fir is very limited. In spite of this, about 7% of the female gametophytes that were co-cultured became penetrated by pollen tubes. In vitro archegonial penetration has been repeatedly achieved, but pollen tubes also penetrated other parts of the female gametophytes. Pollen tubes also penetrated non-viable eggs. Most female gametophytes were not penetrated because of pollen tube branching and swelling, failure of tubes to orient towards the female gametophytes, or premature pollen tube death due to plasmolysis. Summary When the pollen tubes of Pisum sativum (initial line) and of its mutants are grown on a standard medium containing only sucrose, boric acid and agar-agar, no difference in maximum length was observed. But, while pollen tubes of the initial line took nine hours to reach this length, pollen tubes of the mutants needed only six hours. Growth seems to be faster in pollen tubes of the mutants than in those of the initial line.Further investigations examined the influence of twenty-one amino acids on pollen tube growth. With the initial line, these substances can be classified into three groups: those that promote pollen tube growth; those which have no influence upon its growth; and those which reduce its growth. The amino acids of each group are characterized by special structural properties. Those amino acids which accelerate pollen tube growth of the initial line show variable effects on the pollen tubes of the mutants. In some cases the same behaviour of pollen tubes can be observed whether amino acids are added or not, in others the addition of amino acids has a positive effect on pollen tube growth, though less than on pollen tubes of the initial line, and in a single case the addition of an amino acid is followed by a negative effect on growth.  相似文献

Abstract

The present paper examines some biological and ultrastructural aspects of fertilization and early development of the embryo in Cytinus hypocistis, a parasitic plant belonging to the Rafflesiaceae. The probable functions of a mucilaginous substance contained in the ovary and embedding the numerous pollen tubes coming from the style are discussed.

It was ascertained that pollen tubes pass through the micropyle and enter a synergid pushing, their way through the nucellar cells that show swollen walls owing to a probable enzymatic action whose function is to facilitate pollen tube penetration. It was hypothesized that the secretion of such enzymes is attributable to the numerous pollen present in the ovary or entering the mycropyle.

Since, in all the ovules observed, synergid degeneration was never found before the arrival of the pollen tube, this degeneration was interpreted as being caused by the material disharged by the pollen tube, rather than being an essential prerequisite for pollen tube penetration into the synergid.

Pollen tube content was observed to be made up of an intensely electron-dense substance surrounding many lipidic globules and numerous polysaccharide vesicles that fuse with the pollen tube wall, clearly contributing to its growth.

The sequence of the first divisions of the developing embryo was followed and the extreme reduction of the embryo is confirmed.

In Cytinus hypocistis starch is totally absent from all the sells belonging to the female gamethophyte as well as to those belonging to the embryo, but lipidic globules are very frequent; it is therefore supposed that these bodies constitute good material for the nutrition of the zygote and early embryo.  相似文献

Several studies have investigated the effect of nematodes on microbial degradation of macrophyte detritus, but little is known about the potential effect of bacterivorous nematodes on productivity of macrofaunal detritivores. We investigated the influence of the bacterivorous nematode Rhabditis (Pellioditis) mediterranea on the incorporation rate of isotopically-labelled macroalgal detritus by the amphipod Paramoera chevreuxi in a laboratory microcosm. The fatty acid composition of amphipods, nematodes, and macroalgal detritus was characterized to help determine the pathway of detritus incorporation by amphipods. The potential importance of R. mediterranea as a source of highly unsaturated fatty acids (HUFAs) to higher trophic levels was also investigated.We found no clear evidence for an effect of nematodes on the incorporation rate of fresh macroalgal detritus by amphipods, although there was some indication that the type of detritus (i.e. the green Chaetomorpha sp. vs the red Polysiphonia sp.) is important in determining the nature and extent of the relationship between nematodes and macrofaunal detritivores. Fatty acid data indicated that nematodes did not contribute significantly to the diet of amphipods when detritus was present, and there was no evidence that nematodes affected the pathway of detritus incorporation by amphipods. Amphipods incorporated Chaetomorpha sp. detritus about 10 times faster than Polysiphonia sp. detritus despite the higher C/N ratio and lower HUFA content of the former. This suggests that other factors, such as the presence of grazer deterrent compounds, are important in determining the palatability of macroalgal detritus. Amphipods fed exclusively on nematodes retained high HUFA levels but suffered high mortality. The burrowing behaviour of nematodes is suggested as the most likely factor limiting their availability to predators.Results suggest a limited interaction between amphipods and bacterivorous nematodes in detrital food webs. Further experiments are needed to test the wider applicability of these findings to different nematode and macrofaunal taxa, and for different types of detritus.  相似文献

Cellulose is the principal component of the load-bearing system in primary plant cell walls. The great resistance to tensile forces of this polysaccharide and its embedding in matrix components make the cell wall a material similar to a fiber composite. In the rapidly growing pollen tube, the amount of cellulose in the cell wall is untypically low. Therefore, we want to investigate whether the load-bearing function of cellulose is nevertheless important for the architecture of this cell. Enzymatic digestion with cellulase and inhibition of cellulose crystal formation with CGA (1-cyclohexyl-5-(2,3,4,5,6-pentafluorophenoxy)-1λ4,2,4,6-thiatriazin-3-amine) resulted in the formation of tubes with increased diameter in Solanum chacoense and Lilium orientalis when present during germination. In pre-germinated tubes, application of both agents resulted in the transient arrest of growth accompanied by the formation of an apical swelling indicating a role in the mechanical stabilization of this cellular region. Once growth resumed in the presence of cellulase, however, the cell wall in the newly formed tube showed increased amounts of pectins, possibly to compensate for the reduced amount of cellulose. Scanning electron microscopy of pollen tubes subjected to digestion of matrix polysaccharides revealed the mechanical anisotropy of the cell wall. In both Lilium and Solanum, the angle of highest stability revealed by crack formation was significantly below 45°, an indication that in the mature part of the cell cellulose may not the main stress-bearing component against turgor pressure induced tensile stress in circumferential direction.  相似文献

In angiosperms, pollen tube growth is critical for double fertilization and seed formation. Many of the factors involved in pollen tube tip growth are unknown. Here, we report the roles of pollen-specific GLYCEROPHOSPHODIESTER PHOSPHODIESTERASE-LIKE (GDPD-LIKE) genes in pollen tube tip growth. Arabidopsis thaliana GDPD-LIKE6 (AtGDPDL6) and AtGDPDL7 were specifically expressed in mature pollen grains and pollen tubes and green fluorescent protein (GFP)-AtGDPDL6 and GFP-AtGDPDL7 fusion proteins were enriched at the plasma membrane at the apex of forming pollen tubes. Atgdpdl6 Atgdpdl7 double mutants displayed severe sterility that was rescued by genetic complementation with AtGDPDL6 or AtGDPDL7. This sterility was associated with defective male gametophytic transmission. Atgdpdl6 Atgdpdl7 pollen tubes burst immediately after initiation of pollen germination in vitro and in vivo, consistent with the thin and fragile walls in their tips. Cellulose deposition was greatly reduced along the mutant pollen tube tip walls, and the localization of pollen-specific CELLULOSE SYNTHASE-LIKE D1 (CSLD1) and CSLD4 was impaired to the apex of mutant pollen tubes. A rice pollen-specific GDPD-LIKE protein also contributed to pollen tube tip growth, suggesting that members of this family have conserved functions in angiosperms. Thus, pollen-specific GDPD-LIKEs mediate pollen tube tip growth, possibly by modulating cellulose deposition in pollen tube walls.  相似文献

Summary The oxygen production rates for a cyanobacterial suspension flowing in straight and coiled tubes were measured to find a way of achieving higher efficiency of light utilization by means of convective mixing. The photosynthetic flow chambers were made of glass tubes and illumination was by fluorescent light. The cyanobacterium used was Spirulina platensis, which has a high growth rate. The oxygen production rate for fluid flow in straight and coiled tubes increase with the increase in Reynolds number. The maximum oxygen production rate was achieved at 30°C for both tube reactors, but the oxygen production rate was higher for the coiled tube unit than the straight tube unit at 30°C. Thus the convective mixing generated in the coiled tube reactor contributed to an increased in light utilization, which played an important part in improving the oxygen production rate. The mechanisms of penetration of Phytophthora cinnamomi Rands into seedling eucalypt roots were studied by light and electron microscopy. Culture grown seedlings of root-rot tolerant Eucalyptus st johnii and root-rot susceptible Eucalyptus obliqua were inoculated with both zoospores and mycelium. Zoospores encysted on roots of both species and the germ tubes penetrated without the formation of appressoria. Swellings, previously described as appressoria, were formed when the germ tube was slow to enter the host by intracellular penetration. Vegetative hyphae penetrated both inter- and intracellularly into the zones of root elongation and differentiation, often through root hairs. Evidence of hydrolysis of the host cell-wall at the point of penetration was observed in electron micrographs. Several hours after the germ tube penetrated the epidermis, a thick plug of amorphous material formed in the germ tube slightly below the level of the outer walls of the epidermal cells, sealing off the hypha within the root. Behaviour of zoospores and germ tubes and the mechanism of penetration were similar on both hosts. Micrographs do not suggest any kind of a hypersensitive reaction by the host cells during the early stages of infection.  相似文献
