Treatment of two-week old soybean (Glycine max (L.) Merr. cv.A2) seedlings with 250 µg of either soil-applied paclobutrazolor XE-1019 (also known as S-3307) altered diurnal leaf movementswithin 48 h. Leaf blade and petiole angles of primary leaves,relative to the horizontal, were consistently lower in the triazole-treatedseedlings compared to controls during the daylight hours. Atnight, leaves were completely folded down in both treated andcontrol seedlings, but leaves on treated seedlings reached thefolded down position approximately two hours earlier than controls.A single foliar spray of 6.0 mg-liter–1 GA3 counteractedthe triazole effects on leaf blade and petiole angles. The properties and mode of action of a new plant growth retardant,(E)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol(S-3307), were investigated. When a dipping method was used,S-3307 at 2.2 ? 10-7 M (0.064 ppm) retarded the growth of riceplants by 50% of the value found for the control. The retardationof growth was removed by a gibberellin application (8.7?10-5M). S-3307 had nearly no effect on the shoot elongation inducedby gibberellin. The amounts of gibberellin-like substances inrice shoots were decreased by S-3307 treatment in proportionto the degree of growth retardation. This observation was confirmedwith GA1 and GA19, the main gibberellins in the rice plant.Our results indicate that S-3307 inhibits gibberellin biosynthesisin rice plants. Malondialdehyde, a product of lipid oxidation, increased graduallywhen Euglena gracilis cells were bubbled with 240 µ1.liter–1ozone (delivery rate of 1µmolO3.min–1) for 120 min.Simultaneously, the sulfhydryl group content decreased by 36%during the treatment, which was mainly due to oxidation of proteinsulfhydryl groups. The molar amount of SH groups oxidized was3 times higher than that of fatty acid oxidized, indicatingthat sulfhydryl groups were more accessible or more easily oxidizedby O3 than fatty acids. When Euglena cells were allowed to recoverunder autotrophic growth conditions following O3 treatment,viable cells were incapable of dividing during the first 5 hof the recovery period but regenerated SH groups nearly to thecontrol level. The increase of SH content during this periodpreceded the resumption of cell division and the restorationof normal growth. These results suggest that the regenerationof SH groups by Euglena cells is a part of a mechanism involvedin the repair of oxidative damage caused by ozone and is anessential step for the initiation of cell division. The relative growth rate of plants from 7-month-old potato seed-tuberswas significantly greater (37%) than that from 19-month-oldseed-tubers. To determine if loss in seed-tuber phospholipidcontent, and thus membrane integrity, is associated with age-reducedvigour, changes in fatty acyl composition of free fatty acid(FA) and phospholipid fractions were characterized in both seed-tuberages over a 19-d interval of plant establishment. The concentration(nmol g d. wt–1) of saturated and unsaturated FA withinthe phosphatidylcholine (PC) and phosphatidylethanolamine (PE)pools decreased an average of 27-fold over the initial 7 d ofestablishment. This decline was followed by a 19-fold (on average)increase to day 19. The change with time in saturated and unsaturatedFA concentration within the free FA pool was quadratic, butwas opposite to that displayed by the two phospholipid pools.The close correlation in fatty acyl content between the phospholipidand free FA fractions suggested high phospholipase activityduring early plant establishment. The double-bond index (DBI)of all three lipid fractions decreased from day 0 to 7 and thenincreased to day 19. More importantly, in older seed-tubers,the minima in phospholipid DBI and content occurred significantlyearlier (on a plant developmental scale) than in younger seed-tubers.A premature decrease in DBI is indicative of loss of membraneintegrity in potato, and undoubtedly has implications for efficiencyof substrate mobilization and energy metabolism during plantestablishment. Six-day-old rice plants (Oryza sativa L. cv. Bahia) were grownin the presence of 0.5 mol m–3 Ni or Cd for 5 or 10 d.Five days after treatment, some plants were transferred to amedium containing no heavy metal for 5 or 10 d. Plasmalemmavesicles from shoots of treated, transferred (recovery experiments)and control plants were isolated, ATPase activity and proton-pumpingwere assessed, and free sterols and phospholipids determined.The ATPase hydrolytic activity was increased by 37% and 34%in 5 and 10 d Cd-treated plants, respectively; and by 66% in5 and 10 d Ni-treated plants. However, neither the initial rateof H+ transport nor the proton-pumping rate at steady-statewere significantly affected by the treatments. The relativeproportion (%) of the plasmalemma sterols campesterol and 5-avenasteroldecreased while sitosterol increased during heavy metal treatment.The overall plasmalemma phospholipid fatty acyl chain lengthand degree of unsaturation were also reduced. In experimentswhere plants recovered from Cd and Ni treatment, differencesbetween treated and control plants were reverted, particularlyin 10 d Ni-recovered plants. The possible involvement of lipidsin the regulation of the plasmalemma ATPase as well as the relationshipbetween growth, ATPase and adaptations to stress conditionsare discussed. Nine 14C-labeled amino acids and 14C-acetic acid from root tipsof Fe-deficient Graminaceae plants (Hordeum vulgare, Oryzaesativa and Avena saliva) were surveyed to determine the precursoramino acid of phytosiderophores. The dominant precursor wasmethionine, which was incorporated into avenic acid deoxymugineicacid mugineic acid epihydroxymugineic acid and/or hydroxymugineicacid in this order. The compositions and positional distributions of fatty acidsin the major leaf phospholipids of phosphatidylglycerol, phosphatidylcholineand phosphatidylethanolamine were analyzed by gas-liquid chromatographyand enzymic hydrolysis, and chilling-sensitive and chilling-resistantplants were comparcd with respect to the relative contents ofpalmitic and trans-3-hexadecenoic acids in the separated phospholipids.A distinct difference between these plants was found in thefatty acid compositions of phosphatidylglycerol, in which thesum of palmitic and trans-3-hexadecenoic acids ranged from 60to 78% of the total fatty acids in 8 species of chilling-sensitiveplants, and from 50 to 57% in 11 species of chilling-resistantplants. The only exception among the chilling sensitive plantsin this respect was the tomato, in which the sum of palmiticand trans-3-hexadecenic acids in phosphatidylglycerol amountedto 54%. The fatty acid compositions and the positional distributionsof fatty acids in phosphatidylglycerol suggest that the occurrenceof high proportions of dipalmitoyl and 1-palmitoyl-2-(trans-3-hexadecenoyl)species in this lipid is correlated with the susceptibilityto chilling of the leaves of higher plants. (E)-1-(4-Chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazol-1-yl)-1-penten-3-ol(S-3307), a new plant growth retardant, was investigated todetermine the inhibitory sites in gibberellin biosynthesis andthe comparative effects of its stereoisomers in a cell-freesystem from Cucurbita maxima. After treatment with S-3307, theincorporation of [14C]mevalonic acid into GA12-aldehyde andGA12 clearly was inhibited, and kaurene accumulated. Feedingexperiments showed that S-3307 inhibited the oxidation of kaurene,kaurenol and kaurenal, but did not affect the oxidation of kaurenoicacid. Thus the reaction sites of S-3307 in gibberellin biosynthesiswere shown to be the three oxidation steps from kaurene to kaurenoicacid. Because S-3307 has an asymmetric center and a tri-substituteddouble bond, there are four stereoisomers; two optical isomersand two geometrical isomers. The activities of these isomersas a growth retardant and gibberellin biosynthesis inhibitorwere examined with a rice seedling assay and the cell-free system.The relative activity of these isomers was basically the samein the two assay methods. The (S)-(E) form was the most active,being 7-fold more active in the cell-free system and 70-foldmore active in the bioassay than the (R)-(E) form. In vitro treatment of microsomal membranes from the axes ofsoybean (Glycine max (L.) Merr.) seeds with free radicals simulatesthe type of membrane injury observed following a lethal desiccationstress—the accumulation of free fatty acids in the membranebilayer, the loss of lipid-P, and the formation of gel phasedomains. The major phospholipids in the microsomal fractionwere phosphatidylcholine, phosphatidylethanolamine and phosphatidylinositol.Although these treatments induced an extensive loss of totalphospholipid from the microsomal fraction following desiccation,the ratio of the major phospholipids remained unchanged. Neitherlysophosphatides nor phosphatidic acid accumulated in the fraction,but free fatty acid levels increased. Therefore, cleavage ofboth acyl chains from the phospholipid molecule occurred followingdesiccation of the axes and in vitro free radical treatmentof the membrane. Both treatments also promoted formation of gel phase domainsas shown by wide angle x-ray diffraction and increased microviscosityas determined by the fluorescent probe, DPH (1,6-diphenyl-1,3,5-hexatriene).This could be simulated in liposomes prepared from the totalmicrosomal lipid fraction by the addition of saturated freefatty acids (16:0 and 18:0) at the levels observed followingstress. In contrast, the addition of unsaturated fatty acidsperturbed the bilayer and reduced microviscosity. The inclusionof both saturated and unsaturated free fatty acids as observedin vivo promoted a response similar to that observed with onlythe addition of the saturated free fatty acids. Desiccation of the axes also promoted a loss of microsomal protein,which was recovered in the 165 000 x g supernatant, and an apparentloss of thiol groups from the membrane as determined by a thiolspecific fluorescence probe, dansylaziridine. This loss of thiolgroups could also be simulated by exposure of the membranesto gamma irradiation, which was used as a non-enzymatic sourceof free radicals. Collectively, these data support the hypothesisthat membrane disassembly following desiccation stress is mediatedby a free radical mechanism, and that the consequent de-esterificationof membrane phospholipid and accumulation of saturated freefatty acids alter the physical properties of the membrane. The effect of the substituted pyridazinone, 4-chloro-5-(dimethylamino)-2-phenyl-3(2H)-pyridazinone(Sandoz 9785), on fatty acid synthesis in two photosyntheticspecies (Chroomonas salina and Nannochloropsis oculata) andone non-photosynthetic species (Crypthecodiniun cohnii) of marinemicroalgae were examined. Effects were more obvious in C. salinathan in C. cohnii or N. oculata. In C. cohnii the relative distributionamongst polyunsaturated fatty acids (PUFA) of radioactivityincorporated from 14C-acetate was not influenced by the herbicideto any great extent and no major changes in the fatty acid compositionof lipid fractions were observed. In C. salina, Sandoz 9785reduced the proportions of radioactivity recovered in 20: 5(n–3) and 22: 6 (n–3) of the phospholipid fraction.The distribution pattern of radioactivity in the fatty acidsof monogalactosyldiacylglycerol (MGDG) in this species was notgreatly affected by the herbicide whereas its presence significantlyreduced the proportions recovered in 18: 4 and 20: 1 in digalactosyldiacylglycerol(DGDG). The level of 20: 5 (n–3) in DGDG of C. salinawas increased from 4.0 to 19.8% by growing the algae in thepresence of Sandoz 9785. The only notable effect of the herbicideon the synthesis of PUFA in N. oculata was a reduction from18.3% to 11.3% of the proportion of radioactivity recoveredin 20: 5 in phospholipids. The herbicide had no effect on thedistribution of radioactivity in PUFA of galactolipids or onthe fatty acid composition of lipid fractions. The results arediscussed in relation to the potential role of galactolipidsand phospholipids as substrates for desaturations involved inthe formation of long chain PUFA in marine microalgae. Antheridic acid was identified by retention time and full massspectra from GCMS analysis as an antheridiogen in Anemia rotundifoliaand A. flexuosa. In the dark spore germination assay, antheridicacid was active down to 10–10 and 5 ? 10–12g.ml–1in A. rotundifolia and A.flexuosa, respectively. Several orphan G protein-coupled receptors, including GPR40, have recently been shown to be responsive to fatty acids. Although previous reports have suggested GPR40 detects medium- and long-chain fatty acids, it has been reported to be unresponsive to short chain fatty acids. In this study, we have heterologously expressed mouse GPR40 in Xenopus laevis oocytes and measured fatty acid-induced increases in intracellular Ca2+, via two electrode voltage clamp recordings of the endogenous Ca2+-activated chloride conductance. Exposure to 500 µM linoleic acid (C18:2), a long-chain fatty acid, stimulated significant currents in mGPR40-injected oocytes (P < 0.01, ANOVA), but not in water-injected control oocytes (not significant, ANOVA). These currents were confirmed as Ca2+-activated chloride conductances because they were biphasic, sensitive to changes in external pH, and inhibited by DIDS. Similar currents were observed with medium-chain fatty acids, such as lauric acid (C12:0) (P < 0.01, ANOVA), and more importantly, with short-chain fatty acids, such as butyric acid (C4:0) (P < 0.01, ANOVA). In contrast, no responses were observed in mGPR40-injected oocytes exposed to either acetic acid (C2:0) or propionic acid (C3:0). Therefore, GPR40 has the capacity to respond to fatty acids with chain lengths of four or greater. Amino acid composition of the free amino acid pool and the TCA-insolubleprotein fraction were investigated in root tips of pea and Tamarixtetragyna plants grown at various levels of NaCl salinity. Salinitystress induced an increase of proline content, mainly in thefree amino acid pool in both plants, and of proline or hydroxyprolinecontent in the protein. Externally-supplied proline was absorbedand incorporated into protein, by pea roots, more effectivelythan by Tamarix roots. Salinity stress, apparently, stimulatedthe metabolism of externally-supplied labelled proline. Pearoots have a very large pool of free glutamic acid; however,70 per cent of the 14C from externally-supplied 14C-U-glutamicacid was released as CO2. Very small amounts of it were incorporatedinto protein. No measurable amount of radioactivity could bedetected in any one of the individual amino acids, either ofprotein hydrolysate or the free amino acid pool. Proline very effectively counteracted the inhibitory effectof NaCl on pea seed germination and root growth. A similar effectbut to a lesser degree was achieved with phenylalanine and asparticacid. Mung bean (Vigna radiata L. Wilcz. cv. Berken) seedlings wereraised hydroponically in 0, 5 and 15 mol m–3 choline.Fourteen-day-old plants were chilled at 5°C (under lightconditions) for 24 h and then returned to warm conditions fora further 24 h. Primary leaf lamina tissue was used for thedetermination of phospholipid, mole ratio of sterol to phospholipid(ST/PL), sterol composition, and ethylene-forming enzyme (EFE)activity for the various choline-temperature treatments employed.Chilling caused an irreversible loss of lipid in the absence,of choline. Differential loss of lipid resulted in an increasein ST/PL and a decline in the mole ratio of sitosterol to stigmasterol(S/S). There was no recovery of EFE activity following chillingin the absence of choline. Choline (5 and 15 mol m–3)enhanced phospholipid and sterol levels prior to chilling andmaintained lipid levels throughout the chill-warm cycle, withoutany significant change in ST/PL. At 5.0 mol m–3 choline,the chilling-induced decline in S/S was reduced, while at 15mol m–3 S/S increased following chilling. Choline treatment,though reducing EFE activity prior to chilling, allowed recoveryof EFE activity following transfer of plants from chilling towarm conditions. Broadbean plants (Vicia faba L.) were submitted to three differentlevels of steady state N limitation. Relative addition ratesof N were 0.06, 0.1 and 0.14d-1. Plants were harvested at fiveevenly distributed times over a 2d period. Shoot growth correspondedwell with the imposed treatment. Root growth, relative to shootgrowth, was highest at the 0.06d-1treatment. Diurnal patternsof soluble sugars, amino acids and starch were analysed. Onaverage, soluble sugar levels were highest in the plants ofthe 0.06d-1treatment whereas average free amino acid levelswere highest in the 0.14d-1treatment. Shoot growth increasedas the concentration of shoot amino acids increased. No suchcorrelation however could be found between root growth and freesugar levels in the root. The metabolism of [8-14C]adenine and [8-14C]hypoxanthine infour species of Camellia plants was investigated in relationto the synthesis of purine alkaloids, caffeine and theobromine.Young leaves of C. sinensis had the ability to synthesize caffeine,but in C. irrawadiensis, these labelled precursors were incorporatedinto theobromine, not caffeine. No synthesis of purine alkaloidscould be detected in C. japonica and C. sasanqua leaves. The antibiotic thiolactomycin inhibits the fatty acid synthesisfrom both [1-14C]- acetate and [2-14C]malonyl-CoA of spinachleaves, developing castor bean endosperms and avocado mesocarp.On the other hand, fatty acid synthetases of Brevibacteriumammoniagenes and Corynebacterium glutamicum are much less sensitiveto this antibiotic. As has been indicated that thiolactomycininhibits fatty acid synthetase of Escherichia coli but has littleeffect on the synthetases of yeast and rat liver [Hayashi etal. (1983) Biochem. Biophys. Res. Effects of uniconazole-P, a triazole-type growth retardant,on endogenous levels of IAA, cytokinins, ABA and ethylene inrice seedlings were investigated. Endogenous levels of IAA andABA were similar between control and uniconazole-P-treated riceshoots. Evolution of ethylene was promoted slightly, being 1.8times greater under 0.3 ppm uniconazole-P treatment than thatof control. The most obvious effect was the increase of trans-Zand trans-RZ in shoots. Shoots treated with uniconazole-P (10mg/m2 nursery box) contained 3.4 times and 3 times more trans-Zand trans-RZ than control, respectively. No significant differencesof cytokinin levels were recognized in roots except for cis-RZ.The increase of ethylene and active forms of cytokinins, andthe decrease of gibberellin in the shoots may be the basis forphysiological phenomena caused by uniconazole-P, namely thepromotion of flowering in woody plants and the enhancement offemaleness in cucumber. To gain more insight into the relation between plasma membranechanges and cold hardiness in mulberry trees (Morus bombysisKoidz. cv Goroji), biochemical and biophysical changes in theplasma membrane were studied during cold deacclimation in spring.The majority of the changes in the plasma membranes that occurredduring the cold acclimation process in the fall/winter werereversed following deacclimation in the spring. Significantdecreases in phospholipid content, degree of unsaturation inphospholipid fatty acids, and membrane fluidity were observedin the plasma membranes during cold deacclimation. The sterolto phospholipid ratio increased with decreasing cold hardiness.Reverse changes were also detected in the majority of proteinand glycoprotein components. The effect of low temperatures on the fatty acid compositionof phosphatidylglycerol (PG) in thylakoid membranes, in particularon the ratios of nmol% 16:1(3t) (mg fresh weight)–1 ofcotyledons and nmol 16:1(3t) (mg chlo rophyll)–1 weremeasured during squash seedling growth. Plants were germinatedand grown for one day at 30°C then were either kept at 30°C(control plants) or trans ferred to low temperatures (18, 14or 10°C). When plant were transferred from 30°C to lowtemperatures, the increase in fresh weight was gradually limited.The lowe the temperature, the smaller was the fresh weight.In contrast, the relative content of 16:1(3t) and 18:3, as wella the ratios of nmol 16:1(3t) (mg chlorophyll)–1 and mol%16:1(3t) (mg cotyledon fresh weight)–1 increased indicatingthat the increase of fresh weight and chlorophyll was mor sensitiveto low temperature than PG desaturation in thyla-koid membranes.Furthermore, low temperatures inducei an increase in 16:1(3t)and 18:3 (the final products of PC synthesis) at the expenseof 16:0 and 18:1 (the initial products of PG synthesis). However,within a range of temperature from 10 to 18°C, the extentof these changes (nmol% of 18:3 or 16:1(3t) per day) was graduallylimited by lower temperatures. We therefore propose that lowtemperature inhibit both fatty acid synthesis and desaturationactivities. However, at low temperatures the fatty acid synthesisis likely to be more strongly inhibited than the desaturationactivities, thus explaining the observed increase in the relativecontent of PG-18:3 and PG-16:l(3t).
