Marques, I. A., Oberholzer, M. J. and Erismann, K. H. 1985.Metabolism of glycollate by Lemna minor L. grown on nitrateor ammonium as nitrogen source.—J. exp. Bot. 36: 1685–1697. Duckweed, Lemna minor L., grown on inorganic nutrient solutionscontaining either NH4+ or NO3 as nitrogen source wasallowed to assimilate [1-14C]- or [2-14C]glycollate during a20 min period in darkness or in light. The incorporation ofradioactivity into water-soluble metabolites, the insolublefraction, and into the CO2 released was measured. In additionthe extractable activity of phosphoenolpyruvate carboxylasewas determined. During the metabolism of [2-14C]glycollate in darkness, as wellas in the light, NH4+ grown plants evolved more 14CO2 than NO3grown plants. Formate was labelled only from [2-14C]glycollateand in NH4+ grown plants it was significantly less labelledin light than in darkness. In NO3 grown plants formateshowed similar radioactivity after dark and light labelling.The radioactivity in glycine was little influenced by the nitrogensource. Amounts of radioactivity in serine implied that thefurther metabolism of serine was reduced in darkness comparedwith its metabolism in the light under both nitrogen regimes.In illuminated NH4+ plants, serine was labelled through a pathwaystarting from phosphoglycerate. After [1-14C]glycollate feedingNH4+ grown plants contained markedly more radioactive aspartateand malate than NO3 plants indicating a stimulated phosphoenolpyruvatecarboxylation in plants grown on NH4+. The duckweeds Lemna gibba L. and Lemna minor L. only grew wellin undisturbed culture under axenic conditions in low lightintensity when provided with a suitable energy source such asglucose. In media containing N03-N gibbosity (a convex ventralsurface) was induced in the presence of the chelating agentethylene-diamine-di-o-hydroxyphenylacetic acid (EDDHA). In nutrientsolutions containing NO3-N as the only N source, but withoutEDDHA, L. gibba occasionally exhibited gibbosity in culturesolutions of 40 cm3 volumes. More fronds were induced to exhibitgibbosity when the volume of the culture medium was increasedfrom 40 cm3 to 200 cm3. Gibbosity was never induced in L. minor,neither was it induced in L. gibba in media containing NH4-N,even in the presence of NO3-N. There was no direct correlationbetween the occurrence of gibbosity and frond growth rate, butgibbosity occurred only when there was good frond growth. In the absence of a sugar, frond growth was enhanced by bubblingair through the culture solution in the light. Increasing theCO2 concentration in the air up to 1% enhanced growth and inducedgibbosity. Carbon dioxide did not induce gibbosity in mediacontaining NH4-N. Low levels of ubiquitin conjugating activity are typically detectedin the green tissues of plants, an observation that may at leastpartially explain why no method to purify multi-ubiquitinatedproteins from photosynthetic cells has been reported in theliterature. The present paper provides a contribution to improvethe available methodology for the isolation of an efficientubiquitin conjugating system from photosynthetic cells. We haveselected Lemna minor L. as a plant system and have developeda simple and rapid methodology to synthesize and purify highmolecular mass ubiquitin-protein conjugates, formed with endogenoussubstrates and exogenous 125I-ubiquitin, using small amounts(<2g) of green tissue. It is demonstrated that L. minor possessesan ATP-dependent activity capable of forming ubiquitin conjugateswith endogen ous proteins in vitro. Anion exchange chromatographyon diethylaminoethyl-cellulose provides a simple and rapid techniqueto remove endogenous ubiquitin and to concentrate and partiallypurify the enzyme system responsible for ubiquitin conjugatingactivity. This enriched fraction has therefore been utilizedto syn thesize high molecular mass 125I-ubiquitin conjugatesformed with L. minor proteins. These conjugates were subsequentlypurified by directly loading the reaction mixture on a SephacrylS-300 gel filtration column, with no requirement for additionalconcentration or purification steps. The experiments described in this paper concern the nature ofthe K+-influx mechanism in Lemna minor L. It is establishedthat K+ influx is ‘site restricted’ and that theaffinity of the system involved is in close agreement with thatof the well-documented System I mechanisms that are found inmany plants (Epstein, 1972). Experiments with ATP and CCCP suggestthat K+ influx is an active process, that the influx machineryresides at the plasmalemma and that this machinery containsan ATPase activity. Membrane vesicles isolated from the plantsabsorb K+(Rb+) with an affinity comparable to that of the systeminvolved in physiological K+ influx.  相似文献

The flower-inducing activities of benzoic and salicylic acidsadded to the medium differ with the species (Lemna paucicostataand L. minor), and even with the strains used. The type andpH of the medium used, full or 1/10 strength M medium at pH3.8, 4.4 or 5.1, or 1/2 or 1/20 strength NH4+-free Hutner'smedium at pH 5.0, 6.0 or 7.0, also modify their activity. L.paucicostata, strain 151 is the most sensitive of the strainsused to both benzoic and salicylic acids followed by strain381. Such dramatic flowering responses were not obtained withthe other strains, but even strain 321, reportedly insensitiveto benzoic acid, could be induced to flower by adding benzoicacid to a modification of the medium. Benzoic acid is more effectivethan salicylic acid for all strains of L. paucicostata, butthe contrary is true for two L. minor strains tested. A higherpercentage of flowering is obtained in L. paucicostata in 1/2strength NH4+-free Huter'sn medium than in M medium, exceptfor strain 151. When diluted, both media enhance flowering inall L. paucicostata strains. Generally, a lower concentrationof benzoic acid or salicylic acid is enough to induce floweringwhen the pH of the medium is lower. (Received March 30, 1981; Accepted May 16, 1981)  相似文献

Electron transport across the plasma membrane of Valerianellalocusta mesophyll cells and intact fronds of Lemna gibba, inducedby 10–3 M ferricyanide, was inhibited by tetcyclacis,an inhibitor regarded to be specific for cytochrome P-450 mono-oxygenases.The effect was dependent on the concentration of tetcyclacisand the duration of preincubation. The apparent rate of trans-membraneelectron transport increased in the presence of catalase, indicatingtetcyclacis-induced H2O2-production or additional tetcyclacis-independentH2O2 release. The findings suggest an interaction of cytochromeP-450 with the plasma membrane-located electron transport chain.This redox-chain could be involved in the degradation of abscisicacid, being located at the plasma membrane. Leaf segments prepared from the first leaves of barley (Hordeumvulgare) exhibit a rapid loss of protein when given a matricstress with polyethylene glycol. Protein synthesis was reducedby the stress but a greater effect of stress was seen on proteindegradation. Growing leaves were exposed to 3H2O for 4 d ormore to label total protein, and the half-life of protein 2-3H,in the isolated segments prepared from such leaves, was shownto be c. 140 h in the absence of stress. Stress reduced thisto c. 62 h. A short pulse with 3H2O preferentially labels rapidlyturning-over protein and a 24 h pulse given to isolated leafsegments labelled proteins with a half-life of c. 64 h in thepresence or absence of stress. Degradation of the 24 h pulse-labelledproteins was inhibited by cycloheximide. Proline accumulationoccurred in the stressed segments and was inhibited by cycloheximide.The results are discussed in the light of current views concerningprotein degradation and possible relationships between proteolysisand proline accumulation.  相似文献

The relative requirement of N and P (the optimum N:P ratio)by Dunaliella tertiolecta, Phaeodactylum tricornutum, Prymnesiumparvum and Thalassiosira pseudonana was studied under variouslight intensities and spectra. The ratio was determined as theratio of the minimum cell N and P concentrations (q0N and q0pwhen either nutrient was limiting. The ratio varied widely amongspecies; under light-saturation for growth (116 µEin m–2s–1 it ranged from 11.8 in D. tertiolecta to 36.6 in P.tricornutum. The ratio appeared to be higher at a sub-saturatingintensity (24 µEin m–2 s–1 in all except P.tricornutum, mainly because of higher qoN with little changein qoP. In T. pseudonana QoP also increased, resulting in aninsignificant change in the ratio. The ratio varied little withinthe range of saturation intensity. Light quality affected qoNand qoP as well as the ratio, and the pattern of change variedfrom species to species. The optimum ratio of individual specieswas linearly correlated to their qoN except in P. tricornutum.qoN for all species showed a linear correlation with cell proteinconcentrations irrespective of light conditions. The changeof optimum N:P ratios in the three species thus appears to berelated to changes in cell protein contents. The ratio of carbohydratesto protein remained constant regardless of light intensity orquality and was higher in P-limited cultures. We conclude thatchanges in light regime can strongly influence algal nutrientrequirements and species interrelationships by altering theoptimum cellular N:P ratio.  相似文献

Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. Species-specific differences in the assimilation of atmosphericCO2 depends upon differences in the capacities for the biochemicalreactions that regulate the gas-exchange process. Quantifyingthese differences for more than a few species, however, hasproven difficult. Therefore, to understand better how speciesdiffer in their capacity for CO2 assimilation, a widely usedmodel, capable of partitioning limitations to the activity ofribulose-1,5-bisphosphate carboxylase-oxygenase, to the rateof ribulose 1,5-bisphosphate regeneration via electron transport,and to the rate of triose phosphate utilization was used toanalyse 164 previously published A/Ci, curves for 109 C3 plantspecies. Based on this analysis, the maximum rate of carboxylation,Vcmax, ranged from 6µmol m–2 s–1 for the coniferousspecies Picea abies to 194µmol m–2 s–1 forthe agricultural species Beta vulgaris, and averaged 64µmolm–2 s–1 across all species. The maximum rate ofelectron transport, Jmax, ranged from 17µmol m–2s–1 again for Picea abies to 372µmol m–2 s–1for the desert annual Malvastrum rotundifolium, and averaged134µmol m–2 s–1 across all species. A strongpositive correlation between Vcmax and Jmax indicated that theassimilation of CO2 was regulated in a co-ordinated manner bythese two component processes. Of the A/Ci curves analysed,23 showed either an insensitivity or reversed-sensitivity toincreasing CO2 concentration, indicating that CO2 assimilationwas limited by the utilization of triose phosphates. The rateof triose phosphate utilization ranged from 4·9 µmolm–2 s–1 for the tropical perennial Tabebuia roseato 20·1 µmol m–2 s–1 for the weedyannual Xanthium strumarium, and averaged 10·1 µmolm–2 s–1 across all species. Despite what at first glance would appear to be a wide rangeof estimates for the biochemical capacities that regulate CO2assimilation, separating these species-specific results intothose of broad plant categories revealed that Vcmax and Jmaxwere in general higher for herbaceous annuals than they werefor woody perennials. For annuals, Vcmax and Jmax averaged 75and 154 µmol m–2 s–1, while for perennialsthese same two parameters averaged only 44 and 97 µmolm2 s–1, respectively. Although these differencesbetween groups may be coincidental, such an observation pointsto differences between annuals and perennials in either theavailability or allocation of resources to the gas-exchangeprocess. Flat forms of the fronds of Lemna gibba L., used widely in laboratoryexperiments, can be easily confused with Lemna minor Chemicalanalyses of the two species, grown under defined laboratoryconditions, showed clear distinctions between the two speciesin relation to total protein content and the content of individualmonosaccharides. Infra-red spectral analysis confirmed the chemicaldata for protein content and also revealed differences in thetypes of proteins present in the two species. Chemical and infra-redspectral analyses, particularly the amide I: carbohydrate absorbanceratio, provide a rapid and reliable means of verifying betweenthe two species, which are often difficult to identify withcertainty from morphological differences. The occurrence and endogenous level of various plant hormoneswere measured for the short-day plants Lemna paucicostata 151and 381 and the long-day plant Lemna gibba G3 to determine whetherany of them are involved in the photoperiodic control of flowering.ABA, IAA, GA1, GA29, GA34, GA53, trans- and cis-zeatin, trans-and cis-ribosyl zeatin, N6-(2-isopentenyl) adenine and N6-(2-isopentenyl)adenosine were definitely detected in each species, while GA4was only detected in L. gibba G3 and GA20 was only detectedin L. paucicostata 151. The endogenous levels of ABA and IAAwere in the range of 1–7 ng/g fr wt and were not significantlydifferent in vegetative and flowering plants. The endogenousgibberellin levels were generally higher in Lemna grown underlong-day rather than short-day conditions. The endogenous cytokininlevels were almost the same in both flowering and vegetativeplants of L. paucicostata 151 and 381. In L. gibba G3, however,the level of cis-ribosyl zeatin, N6-(2-isopentenyl) adenineand N6-(2-sopentenyl) adenosine were higher in vegetative thanin flowering plants. These results indicate that there is not necessarily a directrelation between endogenous plant hormone levels and flowering,and that the chemical basis for the photoperiodic control offlowering cannot be explained solely by changes in hormone levels.The possibility remains, however, that one or more of the planthormones has some influence of secondary importance on the floweringprocess in Lemna. The relationship between chlorophyll a (Chl a) and primary productivity(PP) in the uppermost water layer and the water column-based(0–15 m) integral values of those variables were examinedusing measurements taken in Lake Kinneret (Israel) from 1990to 2003. In 81% of all Chl a profiles examined, the distributionwas fairly uniform within the entire 0–15 m water column,and 12.3% of instances showed a prominent subsurface maximum,when the lake phytoplankton was dominated by the dinoflagellatePeridinium gatunense. Chl a can be reliably estimated by remotesensing techniques in the productive and turbid water of LakeKinneret, since Chl a concentration at surface layers can beextrapolated to the entire water column. Light vertical attenuationcoefficient average for wavelengths from 400 to 700 nm, Kd,ranged from 0.203 to 1.954 m–1 and showed high degreeof temporal variation. The maximal rate of photosynthetic efficiency,PBopt [average 3.16 (±1.50)], ranged from 0.25 to 8.85mg C m–3 h–1 mg Chl a–1. Using measured dataof Chl a, PBopt, and light as an input, a simple depth-integratedPP model allowed plausible simulation of PP. However, a lackof correlation between photosynthetic activity and temperature(or other variable with remotely sensed potential) renders theuse of models that require input of photosynthetic efficiencyto calculate integrated PP of little value in the case of productiveand turbid Lake Kinneret.  相似文献

Conditions and maintenance of growth were chosen so that plantsof Clusia minor L. were obtained which showed the C3- and CAM-modes of CO2-exchange, respectively. C. minor is known to accumulateconsiderable amounts of citric acid in addition to malic acidduring the dark-phase of CAM. 14CO2-pulse-chase experiments were performed with these plants.Patterns of labelling during the pulse and redistribution oflabel during the chase in the C3-mode were as expected for C3-photosynthesis.Pulse-labelling in the CAM-mode during the last hour of thelight period, during the first part of the dark period and duringthe last hour of the dark period always led to an almost exclusiveincorporation of label into malate. Redistribution of labelfrom malate after the pulse at the end of the dark period duringthe chase in the subsequent light period followed the patternexpected for light-dependent reassimilation of CO2 remobilizedfrom malate in CAM during the light period. During the chasesin the dark period, label was transferred from l4C-malate tocitrate. This suggests that during accumulation of citric acidin the dark period of CAM in C. minor, citrate is synthesizedin the mitochondria from malate or oxaloacetate after formationof malate via phosphoenolpyruvate carboxylase. The experiment also showed that no labelled compounds are exportedfrom leaves in the CAM-mode during the dark period. In plantsof the C3-mode the roots proved to be strong sinks. Effects of removal of external Ca2+ on the cytoplasmic pH (pHc)of Chara corallina have been measured with the weak acid 5,5-dimethyl-oxazolidine-2,4-dione(DMO) as a function of external pH (pH0) and of the externalconcentration of K+. Removal of Ca2+ always decreased pHc whenpH0 was below about 6.0; the decrease was about 0.2–0.4units at pH0 5.0, increasing to about 0.5 units at pH0 4.3.When pH0 was 6.0 or higher the removal of Ca2+ had little orno effect on pHc. This situation was not altered by changingthe concentration of K+, though in some experiments at pH0 5.0–5.2there was a slight decrease in pH0 (about 0.2 units) when K+was increased from 0.2 to 2.0 mol m–3, an effect apparentlyreversed when K+ was higher (5.0 or 10.0 mol m–3). Theresults suggest that H+ transport continues in the absence ofexternal Ca2+, despite previous suggestions to the contrary,and that the H+ pump does not necessarily run near thermodynamicequilibrium with its chemical driving reaction. They indicate,rather, that the H+ pump is under kinetic control and providefurther evidence for the inadequacy of present models for theoperation of the H+ pump in charophyte cells, especially inrelation to its proposed role in regulating pHc. Regulatory effects of light on senescence of rice leaves wereinvestigated by measuring degradation of chlorophyll and proteinsin leaf segments which had been kept in the dark or under illuminationwith light of different intensities and colors. When leaveshad been left in total darkness for three days at 30°C,there was an initial long lag that lasted for one whole dayand then chlorophyll was rapidly degraded in the second andthird days. Breakdown of chlorophyll was strongly retarded bycontinuous illumination with white light of intensity as lowas 0.5 µmol photons m–2 s–1 but the effectof light decreased at intensities above 10 µmol photonsm–2 s–2. The initial lag and subsequent degradationof chlorophyll in the dark were little affected by illuminationwith red or far red light at the beginning of dark treatment.However, a brief illumination with red light at the end of thefirst and/or second day significantly suppressed degradationof chlorophyll during subsequent dark periods and the effectof red light was nullified by a short irradiation with far redlight. Thus, degradation of chlorophyll is regulated by phytochrome.Thylakoid membrane proteins and soluble proteins were also largelydegraded during three days in the dark. Degradation of membraneproteins such as the apoproteins of light-harvesting chlorophylla/b proteins of photosystem II and chlorophyll a-binding proteinsof reaction center complexes showed a long lag and was stronglysuppressed by illumination with weak white light. Thus, theloss of chlorophyll can be correlated with degradation of chlorophyll-carryingmembrane proteins. By contrast, light had only a weak protectingeffect on soluble proteins and ribulose-1,5-bisphosphate carboxylase/oxygenaserapidly disappeared under illumination with weak white light.Thus, breakdown of thylakoid membrane and soluble proteins aredifferently regulated by light. Gas exchange and organic acid accumulation of the C3-CAM intermediateClusia minor L. were investigated in response to various day/nighttemperatures and two light regimes (low and high PAR). For bothlight levels equal day/night temperatures between 20°C and30°C caused a typical C3 gas exchange pattern with all CO2uptake occurring during daylight hours. A day/ night temperatureof 15°C caused a negative CO2 balance over a 24 h periodfor low-PAR-grown plants while high-PAR-grown plants showeda CAM gas exchange pattern with most CO2 uptake taking placeduring the dark period. However, there was always a considerablenight-time accumulation of malic acid which increased when thenight-time temperature was lowered and had its maximum (54 mmolm–2) at day/night temperature of 30/15°C. A significantamount of malic acid accumulation (23 mmol m–2) in low-PAR-grownplants was observed only at 30/15°C. Recycling of respiratoryCO2 in terms of malic acid accumulation reached between 2·0and 21·5 mmol m–2 for high-PAR-grown plants whilethere was no significant recycling for low-PAR-grown plants.Both low and high-PAR-grown plants showed considerable night-timeaccumulation of citric acid. Indeed under several temperatureregimes low-PAR-grown plants showed day/night changes in citricacid levels whereas malic acid levels remained approximatelyconstant or slightly decreased. It is hypothesized that lowand high-PAR-grown plants have different requirements for citrate.In high-PAR-grown plants, the breakdown of citrate preventsphotoinhibition by increasing internal CO2 levels, whereas inlow-PAR-grown plants the night-time accumulation of citric acidmay function as an energy and carbon saving mechanism.
