The swimming excursions of individual perch, measured automatically, within either cages in Windermere or in a trough on the shore, showed increases at dawn and dusk and in the summer. Similar measurements in troughs in the laboratory showed increased activity following a change from light to dark and vice versa, a pattern that was obliterated by either continuous light or continuous darkness. These results are discussed in relation to water temperature and catches of perch in traps.  相似文献

This study evaluated physiological and functional sperm parameters and the seminal plasma proteome of Eurasian perch (Perca fluviatilis) over the course of their reproductive season. Spermatozoa velocity (169.56 ± 6.53 to 158.5 ± 7.4 µm sec?1), percent motility (95.89 ± 4.28% to 89.55 ± 4.5%), and osmolality of seminal plasma (290 ± 5 to 297 ± 12 mOsmol kg?1) remained stable throughout the reproductive season. Milt volume and protein concentration of seminal plasma gradually increased and reached the highest values late in the reproductive period. Spermatozoa concentration peaked in the mid‐reproductive season (66.90 ± 13 × 109 spermatozoa ml?1) and decreased towards the end (54 ± 10 × 109 spermatozoa ml?1). A proteomic analysis of seminal plasma using two‐dimensional polyacrylamide gel electrophoresis revealed 10 protein spots significantly altered over the course of the reproductive season. Subsequent protein characterization suggested that time in the reproductive season predominantly affected proteins involved in membrane trafficking, organization, cell motility, and oxido‐reductase activity. This study provides new data on physiological properties of sperm and protein patterns of seminal plasma over the course of the reproductive season that should be considered in the development of methods for artificial reproduction of perch. Despite the prevalence of zebrafish as a model scientific organism, understanding sperm function in this species is essentially limited to observations that osmotic shock initiates motility. During natural spawning, sperm encounter a range of environmental salinities as well as freshwater mixed with egg-associated ovarian fluid (OF), thus sperm are likely to be exposed to saline prior to egg contact. Effects of saline on sperm function in this model species are unknown, but likely to be important. Using computer assisted sperm analysis, this study addressed the effects of osmolality of spawning media and ionic composition and pH on the proportion of sperm becoming motile at activation (motility), as well as sperm velocity and path. When activated with tap water, motility was maximal (80%) at 10 s (earliest time measured), declining to 5% by 87 s postactivation. With activation at moderate osmolalities (∼160-200 mmol/kg) initial motility was decreased relative to low osmolality, increased from 10 to 30 s, and subsequently declined less rapidly (motility in 80 mM NaCl was 35%, 80%, and 60% at 10, 30 and 147 s, respectively). Thus, moderate osmolality increased duration, but introduced a temporal lag in motility onset. With moderate osmolalities, the rate of velocity decay was less than that with tap water activation. Sodium chloride and sucrose similarly impacted both motility and velocity. Replacement of NaCl with KCl, pH values ranging from 6.8 to 8.4, or the presence of gadolinium were without effect. Motility, but not velocity, was slightly supressed by Ca2+. Therefore, whereas pH and concentrations of Ca2+ or K+ of OF are unlikely to impact fertility via sperm motility, the OF contribution to spawning media osmolality may have pronounced effects on motility and velocity of sperm, factors previously correlated with fertility in other species.  相似文献

The objectives of the present study were to determine the relationships among length and weight of males, sperm volume, spermatozoa concentration, total number of spermatozoa, ionic contents and osmolality of seminal plasma in Barbus barbus. The effect of osmolality on sperm motility parameters after activation in NaCl, KCl, or sucrose solutions was also examined. There were significant correlations between spermatozoa concentration – length (R = + 0.7) and – weight (R = + 0.8) of males. No significant correlations were observed between the total number of spermatozoa, sperm volume, and length and weight of males. Seminal plasma osmolality was higher when the total number of spermatozoa (R = + 0.6) and sperm volume (R = + 0.6) were higher. Sperm motility and velocity was positively correlated with osmolality (R = + 0.5). The correlation between sperm motility and K+ was negative (R = 0.5), but positively correlated with Ca2+ (R = 0.8), Na+ (R = 0.8), and Cl (R = 0.8). There was a rapid decrease (P < 0.05) in sperm motility parameters after sperm activation. Just after sperm activation, beating waves propagated along the full length of flagella. At latter stages post sperm activation, the waves appeared only in proximal part of the flagellum. The highest spermatozoa velocity and percentage of motility were observed at 215–235 mOsmol kg− 1 in NaCl, KCl or sucrose. The tip of the flagellum became curled into a loop shape which shortened the flagellum after activation of sperm in distilled water. B. barbus sperm is very similar to that of other cyprinids in terms of ionic contents and osmolality of the seminal plasma, mechanism of sperm activation and behavior and motility of sperm during swimming period.  相似文献

The composition of seminal plasma and the effects on sperm longevity vary between fractions and individual stallions. The development of the retina of perch, Perca fluviatilis L., was studied for all developmental stages. The density of cones 100 μm-1 was found to decrease with age, although their actual numbers increased. All premetamorphosed perch had pure-cone retinae, the rods developing at, or after metamorphosis. Retinomotor responses were found to commence after metamorphosis when the rods developed. Visual acuity, measured as the minimum separable angle (α), was observed to improve exponentially with age, and its value was found to depend more on the focal length of the lens, than on the number of cones in the retina. The cones in the unspecialized part of the retina were predominantly of the twin type and were related to single cones in a ratio of 4 : 1.  相似文献

In Vimba vimba, GSI, sperm volume, and spermatozoa concentration range from 3.4-7.4 %, 0.1-1.1 ml, and 13.3-34.8 × 109 spz ml−1, respectively. Gonad mass (r = 0.90) and sperm volume (r = 0.35) significantly correlated with weight of males. Significant correlation was also found between gonad mass and length of males (r = 0.85). Sperm motility (r = 0.99) and velocity (r = 098) significantly decreased after activation in Tris-HCl 20 mM, pH 8.5. Osmolality of the seminal plasma was 273.2 mOsmol kg−1. Sperm motility and velocity were significantly affected by the osmolality of the activation medium (P < 0.01). Hyper-osmolality compared to seminal plasma osmolality totally suppressed the sperm activation. At 15 s post-activation, the sperm motility significantly decreased at 240 mOsmol kg−1 in KCl or NaCl media. The highest sperm motility and velocity (at 60 s post-activation) were observed at 200 mOsmol kg−1 in NaCl, KCl, or sucrose media. In all treatments, the tip of the flagellum of spermatozoa became curled into a loop shape after activation of sperm in distilled water containing 20 mM Tris-HCl, pH 8.5 that shortened the flagellum.  相似文献

Sperm plasma membrane lipids have an important role to play in determining membrane fluidity and sperm motility. The objective of the present study was to determine whether there are differences in the lipid and fatty acid (FA) composition of boar sperm and seminal plasma in the ejaculates of boars having different sperm motilities. Semen was collected from two groups of boars having normal (> 60%; n = 53) or low (< 60%; n = 53) motility sperm and the semen was evaluated for motility, morphology and vitality. The semen was then centrifuged to separate the sperm from the seminal plasma and both were kept at −20 °C until analyzed for lipid content and FA profile by gas chromatography. Total antioxidant status (TAS) of seminal plasma was determined using a commercial kit. There were differences (P ≤ 0.05) in sperm total lipids, cholesterol, saturated fatty acids (SFA), phospholipids, n-3 polyunsaturated fatty acids (PUFA), docosahexaenoic acid (DHA) and the ratio of n-6:n-3 PUFA between boars with normal and low motility sperm. Total lipids, cholesterol, phospholipids, PUFA, DHA and n-3 PUFA were positively correlated with sperm motility, viability, normal morphology and normal plasma membrane. In contrast, SFA and the ratio of n-6: n-3 PUFA were negatively correlated (P ≤ 0.05) with sperm motility, viability, normal morphology and normal plasma membranes. The TAS of seminal plasma from boars having normal motility sperm was higher (P ≤ 0.05) than that of boars having low motility sperm and TAS was positively correlated (P = 0.0001) with sperm motility, viability, normal morphology and normal plasma membranes. In summary, differences in sperm motility were related to n-3 PUFA content in the sperm plasma membrane and extracellular antioxidants in seminal plasma which protect sperm plasma membranes from lipid peroxidation during periods of oxidative stress.  相似文献

Perch, Percafluviatilis L. were studied in Slapton Ley which is a eutrophic freshwater lagoon in south Devon, England. The perch were caught in traps from February 1970 to June 1971. The traps selected males during the spawning season. The water temperature did not affect the numbers of trapped fish directly, but the sight of perch in the traps appeared to attract others. Sex ratios were found to be equal in the first 2 years of life but older year classes were predominantly female. The population consisted of young individuals. Males were found to mature in their first year and most females matured in their second. The fecundity varied from an average of 1000 eggs in perch n i the 9.75 cm length group to 30,500 eggs in fish in the 27.75 cm group. A few shoals of perch moved throughout the Ley although the majority of fish stayed in the same area. Population estimates showed a range from 10,000 to 95,000 for fish 9.75 cm and above. The large variations in population estimates and survival rates were attributed to the necessity of using traps which had variable catches and to the harmful effects of handling the perch. Chironomids were important in the diet of perch of all sizes. Plankton and Asellus were other important foods, the latter especially during the winter months. Perch 11.5 cm to 13.9 cm selected the widest range of food organisms. Only perch 14.0 cm and above ate fish.  相似文献

A significantly higher concentration of testicular spermatozoa was obtained from freshwater Oreochromis mossambicus (9·9×109 spermatozoa ml−1) than seawater O. mossambicus (4·6×109 spermatozoa ml−1). The mean osmolality of the urine of freshwater fish (78·5 mOsmol kg−1) was significantly different from that of seawater fish (304·8 mOsmol kg−1). The mean length of the mid-piece of the spermatozoa together with the tail was more variable in freshwater O. mossambicus (8·80±0·23μm) than in seawater specimens (8·27±0·18 μm). Stripped sperm of freshwater O. mossambicus was highly contaminated by urine which was a good activator of sperm motility in O. mossambicus held in both fresh and sea water. The osmolality for initiation of motility in freshwater O. mossambicus spermatozoa was from 0 to 333 mOsmol kg−1 while for seawater O. mossambicus spermatozoa it was from 0 to 1022 mOsmol kg−1. The optimum osmolality for motility was from 70 to 333 mOsmol kg−1 for freshwater O. mossambicus spermatozoa and from 333 to 645 mOsmol kg−1 for seawater fish. In freshwater O. mossambicus spermatozoa, the presence of 20 mM CaCl2 increased the permissive osmolality of NaCl from 184 to 645 mOsmol kg−1. For seawater O. mossambicus spermatozoa, solutions of NaCl devoid of CaCl2 were unable initiate motility, but the addition of 1·5 to 30 mM CaCl2 to the NaCl solution (0–934 mOsmol kg1) had a full motility initiating effect.  相似文献

Bovine seminal plasma proteins are in an aggregated form of high molecular weight in their native state. By immobilisation on a cation exchanger with exposure to disaggregating conditions (i.e., acetonitrile and low pH), the high-molecular-weight aggregates could be dissociated to slowly release the low-molecular-weight components. The anionic component released from the cation exchanger during disaggregation was collected by adsorption on a hydrophobic interaction column. The cationic component remaining on the cation exchanger was eluted with NaOH. Both components were found on gel permeation chromatography to be <5 kDa. SDS—PAGE of the various fractions showed that component of low molecular weight were still in an aggregated form. These components resulting from the disaggregation process have detrimental effects on sperm motility and the effects were more substantial compared with that of whole seminal plasma. All the cationic components were significantly detrimental to sperm motility, especially the fractions of low molecular weight. The anionic fractions reduced sperm motility when in an aggregated state. The isolated anionic peptide was not detrimental in its free form. In all fractions the peptides tended to re-aggregate to a higher molecular weight under neutral conditions, however, the isolated anionic peptide (molecular weight <1,500) failed to do so. © Wiley-Liss, Inc.  相似文献

The distribution of the enzyme leucine aminopeptidase (LAP, EC 3.4.1.1) in the digestive tract of perch, Perca fluviatilis L., was investigated histochemically. The enzyme was present in the mucosa of the entire intestine and was absent in the oesophagus, stomach and pyloric sphincter. In the intestine, the enzyme was localized in the supranuclear cytoplasm of the columnar cells and was strongest in the brush border area. Enzyme activity was also present in the lumen of the intestine. The activity of the enzyme in the intestine decreased slightly towards the rectum. In perch, the final digestion of peptides and aminopeptides is both extracellular and intracellular and takes place along the entire length of the intestine.  相似文献

The anatomy and enzyme histœhemistry of the œsophagus of a freshwater teleost perch ( Perca fluviatilis L.) has been studied. The mucosa is composed of an undifferentiated layer of basal epithelial cells, mucous cells and surface epithelial cells. The submucosa is compact and contains blood vessels, nerves and bundles of striated longitudinal muscles. The outer circular muscular layer contains both striated and smooth muscle fibres. Electron microscopy shows that the surface epithelial cells are degenerative and that they surround and support pores of the mucous cells. Undifferentiated epithelial cells undergo cytoplasmic changes and eventually become surface epithelial cells. Mucous cells arise from the undifferentiated epithelial cells and their droplets contain acid and neutral mucosubstances. Alkaline phosphatase, non-specific carboxylic esterases and aminopeptidase activity is absent in the mucosa. However, acid phosphatase activity is localized in the surface epithelial cells.  相似文献

Transferrin (Tf) is a major protein of carp (Cyprinus carpio) seminal plasma. Its relationship with milt quality is unknown. In this study, we sought to determine if Tf is polymorphic in carp seminal plasma and if this polymorphism is related to sperm motility characteristics. We screened males of purebred common carp line (Polish line R6) for Tf polymorphism in blood plasma. The majority of Tf genotypes represented only DD and DG variants. We then collected milt from preselected DD and DG genotypes and tested their sperm motility characteristics using computer-aided sperm analysis (CASA). Tf polymorphism in seminal plasma was found to be identical with that of blood. However, the relationships between Tf polymorphism and iron metabolic parameters were different for blood and semen. These data suggest different regulation of Tf in liver and testis. We found substantial differences in sperm motility characteristics between both genotypes. Spermatozoa of DG males were characterized by lower curvilinear velocity (VCL), amplitude of lateral head displacement (ALH), higher linearity (LIN) and straightness (STR) of movement as compared to DD males. No differences were found in other sperm characteristics such as sperm concentration and percentage of sperm motility. Our results suggest that sperm motility parameters are related to Tf polymorphism and therefore this polymorphism may be related to sperm competitive ability.  相似文献

The relationship between seminal plasma level (0, 10, or 20%) and extender type [Kenney type (EZ-Mixin-CST) or Kenney-modified Tyrodes-KMT] to the susceptibility of sperm DNA to denaturation and sperm motility measures were investigated in cooled (5 degrees C) stallion sperm. Three ejaculates from each of three fertile stallions were collected in an artificial vagina and processed as follows: diluted one part uncentrifuged semen with four parts of extender to a final concentration of 20% seminal plasma in either CST or KMT (20% CST; 20% KMT); diluted to a final concentration of 25 million sperm/mL in either CST or KMT (10% CST; 10% KMT); centrifuged to remove virtually all seminal plasma and resuspended in either CST or KMT (0% CST-Cent; 0% KMT-Cent); centrifuged semen to remove virtually all seminal plasma and resuspended with previously filtered seminal plasma from the same stallion in either CST or KMT to a final concentration of 20% seminal plasma (20% CST-Cent; 20% KMT-Cent). Sperm motion characteristics were determined by CASA and DNA integrity (%COMP, percent of cells outside the main population) evaluated by the Sperm Chromatin Structure Assay prior to cooling, and after 24 and 48 h cooled-storage at 5 degrees C. After 48 h of storage at 5 degrees C, extenders with 0% seminal plasma (0% CST-Cent, 0% KMT-Cent) maintained highest quality DNA (P < 0.05), but 0% KMT-Cent maintained higher velocity measures (P < 0.05) than 0% CST-Cent. Total sperm motility was highest (P < 0.05) in 0% CST-Cent, 0% KMT-Cent, 10% CST, 20% CST-Cent, and 20% CST compared to the other treatment groups. Progressive sperm motility was highest (P < 0.05) after 48 h of storage in the treatment with 10% seminal plasma in Kenney extender (10% CST), despite a reduction in DNA integrity. Regardless of extender type, addition of 20% seminal plasma following centrifugation resulted in almost a two-fold increase in %COMP(alpha t), even though one of the treatments (20% CST-Cent) maintained total and progressive motility similar to treatments with no seminal plasma, suggesting that sperm motility and DNA integrity may respond independently to environmental conditions. Overall, better quality sperm features (motility and DNA) were maintained in sperm from which seminal plasma was removed followed by resuspension in either Kenney extender or modified Kenney Tyrodes-type extender.  相似文献

The maximum growth rate of juvenile perch, PercaJuviatilis L., at different constant temperatures and in naturally changing day-lengths was studied in the laboratory. Standard metabolic rate was studied in starvation experiments at constant temperatures under short- and long-day conditions. Growth occurred in temperatures above 8 to 10°C. In winter, from mid-October until mid-April, maximal growth was considerably reduced and was relatively slow but constant. The standard metabolic rate was reduced c . 50% under short-day conditions. The seasonal change in metabolic rates, presumably controlled by an endogenous rhythm, was considered to be an adaptation to low food availability during the short winter days.  相似文献

Perca fluviatilis L. were caught by gill nets every 3 h over a 24 h period monthly from February 1975 until January 1976. The numbers caught increased with rise in temperature (correlation coefficient r = 0.86 P < 0.001). During the winter months perch migrated into deeper water although some fish were still caught in shallow water (1–6 m). Perch were active at very low light intensities. However changes in visual surface irradiance (400–700 nm) and water clarity caused inverse changes in activity. Only June did not conform to this pattern and it was presumed that spawning behaviour was of primary importance. It is suggested that increase in eutrophication with its effect on water clarity enables fish to become more active during the day in the summer months. They are thus able to utilise food sources to a maximum at an optimum period for feeding and growth.  相似文献

  • 1 The diet and daily food consumption (Ct) of 0+ perch were investigated in two shallow Scottish lakes.
  • 2 The first food comprised the rotifer Keratella sp., Volvox sp., copepod nauplii and stage I copepodites of Cyclops strenuus abyssorum Sars. Larger copepodites and adults of C. s. abyssorum and Diaptomus gracilis Sars were consumed as larvae grew and cladocerans were also eaten by larvae >11 mm. The diet of juveniles included a wider range of cladocerans and benthic invertebrates. Cannibalism was not observed among 0+ perch.
  • 3 The range of food items taken by larvae increased with length, and the size of food particles ingested was governed by jaw gape which was linearly related to fish length.
  • 4 Larvae fed during daylight hours with feeding maxima in midmorning and the evening.
  • 5 The arithmetic (constant) rate of evacuation R of the digestive tract of larvae was 0.59—1.16% of body weight h?1 and exponential R was 0.21–0.33% h?1. Daily Ct calculated using an arithmetic model was 14.2–23.4% body weight at water temperatures of 12.5–18.2°C, and 23.9–40.3% utilizing an exponential method. For larvae of 9 mm. this was 46 μg dry weight day?1 and 148 by an exponential model. Food conversion efficiency was 37–72% (arithmetic model) and 21–45% (exponential model).

An account is given of the histological changes associated with epidermal lesions on the Windermere perch, Perca fluviatilis L., between the months of January and March, 1976. During the later stages of the development of the lesions an apparent succession of fungal colonization was observed. A total of five genera of fungi were identified with Leptomitus lacteus appearing early in the infection and Achlya tending to dominate the more advanced lesions. The initial cause of the lesions has not been identified but the possibility of Argulus damage is discussed.  相似文献
