Recovery of photosynthesis in winter-stressed Scots pine   总被引:9,自引:5,他引:4  
Abstract. . Winter-induced inhibition of photosynthesis in Scots pine (Pinns sylvestris L.) is caused by the combined effects of light and freezing temperatures; light causes photoinhibition of photosystem II (Strand & Oquist, 1985b, Physiologic Plantarum, 65 , 117–123), whereas frost causes inhibition of enzymatic steps of photosynthesis (Strand & Öquist, 1988, Plant, Cell & Environment, 11 , 231–238). To reveal limiting steps during recovery from winter stress, the potential of photosynthesis to recover and the actual recovery outdoors during spring, were studied in Scots pine. Studies of light dependent O2-evolution under saturating CO2 and recordings of room temperature fluorescence induction kinetics were used. When branches of pine, in February and March, were brought into the laboratory and kept at 18°Cand 100μmol m?2 s?1, light saturated rates and apparent quantum yields of photo-synthetic O2-evolution recovered fully within approximately 48h. The photochemical efficiency of photosystem II, as measured by Fv/Fm ratios, recovered fully within 24h after an initial lag-phase of 2-3 h. Under natural winter conditions, the Fv/Fm ratio decreased more in exposed than in shaded pine, whereas the efficiency of photosynthesis was similarly inhibited in exposed and shadedpine. However, when recovery from winter stress occurred during spring, the Fv/Fm ratios of both shaded and exposed pine recovered well before photosynthesis. It is concluded that the light-induced photoinhibition component of winter stress in photosynthesis of pine recovers well before the frost induced component(s) of winter stress. In this context, reversible photoinhibition of photosynthesis in evergreen conifers is considered as a dynamic down-regulation of photosystem II to prevent more severe photodynamic damage of the thylakoid membrane when photosynthesis is inhibited by frost.  相似文献

Abstract. The kinetics of in vivo chlorophyll fluorescence of photosystem II (PS II) was measured at room temperature and 77 K during frost hardening of seedlings of Scots pine (Pinus sylvestris L.), and after exposure of frost-hardened shoots to sub-freezing temperatures. A more pronounced decrease in variable fluorescence yield for the upper exposed than for the lower shaded surface of the needles suggested that some photoinhibition occurred during prolonged frost hardening at 50 μmol photons m?2 s?1 and 4°C. Reversible inhibition of photosynthesis after exposure to sub-freezing temperatures was initially manifested as an increase of steady-state energy-dependent fluorescence quenching (qE) and a reduction in the rate of O2 evolution. Further inhibition after treatment at still lower temperatures caused a progressive decline of steady-state photochemical quenching (qQ) and the rate of O2 evolution, whereas qE remained high. This implies an inactivation of enzymes in the photosynthetic carbon reduction cycle decreasing the consumption of ATP and NADPH, which is likely to cause an increase of membrane energization and a reduction of the primary electron acceptor (QA) of PS II. Alternatively, the changes in qQ and qE might be attributed to an inhibition of photophosphorylation. Severe, irreversible damage to photosynthesis resulted in a suppression of qE and of variable fluorescence yield, probably because the photochemical efficiency of PS II was impaired. Changes in the fast fluorescence kinetics at room temperature after severe freezing damage were interpreted as an inhibition of the electron flow from QA to the plastoquinone pool. It is suggested that irreversible freezing injury to needles of frost-hardened P. sylvestris causes damage to the QB,-protein.  相似文献

Summary The apoplast of barley and oat leaves contained superoxide dismutase (SOD), catalase, ascorbate peroxidase, dehydroascorbate reductase, monodehydroascorbate reductase, and glutathione reductase activities. The activities of these enzymes in the apoplastic extracts were greatly modified 24 h after inoculation with the biotrophic fungal pathogenBlumeria graminis. The quantum efficiency of photosystem II, which is related to photosynthetic electron transport flux, was comparable in inoculated and healthy leaves during this period. Apoplastic soluble acid invertase activity was also modified in inoculated leaves. Inoculation-dependent increases in apoplastic SOD activity were observed in all lines. Major bands of SOD activity, observed in apoplastic protein extracts by activity staining of gels following isoelectric focusing, were similar to those observed in whole leaves but two additional minor bands were found in the apoplastic fraction. The apoplastic extracts contained substantial amounts of dehydroascorbate (DHA) but little or no glutathione (GSH). Biotic stress decreased apoplastic ascorbate and DHA but increased apoplastic GSH in resistant lines. The antioxidant cycle enzymes may function to remove apoplastic H2O2 with ascorbate and GSH derived from the cytoplasm. Winter-induced inhibition of photosynthesis in Scots pine (Pinus sylvestris L.) needles is accompanied by a 65% reduction of the maximum photochemical efficiency of photosystem II (PSII), measured as F v/F m, but relatively stable photosystem I (PSI) activity. In contrast, the photochemical efficiency of PSII in bark chlorenchyma of Scots pine twigs was shown to be well preserved, while PSI capacity was severely decreased. Low-temperature (77 K) chlorophyll fluorescence measurements also revealed lower relative fluorescence intensity emitted from PSI in bark chlorenchyma compared to needles regardless of the growing season. Nondenaturating SDS-PAGE analysis of the chlorophyll–protein complexes also revealed much lower abundance of LHCI and the CPI band related to light harvesting and the core complex of PSI, respectively, in bark chlorenchyma. These changes were associated with a 38% reduction in the total amount of chlorophyll in the bark chlorenchyma relative to winter needles, but the Chl a/b ratio and carotenoid composition were similar in the two tissues. As distinct from winter pine needles exhibiting ATP/ADP ratio of 11.3, the total adenylate content in winter bark chlorenchyma was 2.5-fold higher and the estimated ATP/ADP ratio was 20.7. The photochemical efficiency of PSII in needles attached to the twig recovered significantly faster (28–30 h) then in detached needles. Fluorescence quenching analysis revealed a high reduction state of Q A and the PQ-pool in the green bark tissue. The role of bark chlorenchyma and its photochemical performance during the recovery of photosynthesis from winter stress in Scots pine is discussed.  相似文献

Photoinhibition of photosynthesis in vivo is shown to be considerably promoted by O2 under circumstances where energy turnover by photorespiration and photosynthetic carbon metabolism are low. Intact protoplasts of Valerianella locusta L. were photoinhibited by 30 min irradiation with 3000 mol photons · m–2 · s–1 at 4° C in saturating [CO2] at different oxygen concentrations, corresponding to 2–40% O2 in air. The photoinhibition of light-limited CO2-dependent photosynthetic O2 evolution increased with increasing oxygen concentration. The uncoupled photochemical activity of photosystem II, measured in the presence of the electron acceptor 1,4-benzoquinone, and maximum variable fluorescence, Fv, were strongly affected and this inhibition was closely correlated to the O2 concentration. The effect of O2 did not saturate at the highest concentrations applied. An increase in photoinhibitory fluorescence quenching with [O2], although less pronounced than in protoplasts, was also observed with intact leaves irradiated at 4° C in air. Initial fluorescence, Fo, was slightly (about 10%) increased by the inhibitory treatments but not influenced by [O2]. A long-term cold acclimation of the plants did not substantially alter the O2-sensitivity of the protoplasts under the high-light treatment. Seedlings of Lodgepole pine (Pinus contorta L.) and winter wheat (Triticum aestivum L. cv. Monopol) were cold acclimated under controlled conditions to induce frost hardiness. Lodgepole pine responded to cold acclimation by partial inhibition of photosynthesis with an associated partial loss of photosystem II reaction centres, and a reduction in needle chlorophyll content. This was accompanied by a low daily carbon gain, and the development of a high and sustained capacity for non‐photochemical quenching of absorbed light. This sustained dissipation of absorbed light as heat correlated with an increased de‐epoxidation of the xanthophyll cycle pigments forming the quenching forms antheraxanthin and zeaxanthin. In addition, the PsbS protein known to bind chlorophyll and the xanthophyll cycle pigments increased strongly during cold acclimation of pine. In contrast, winter wheat maintained high photosynthetic rates, showed no loss of chlorophyll content per leaf area, and exhibited a high daily carbon gain and a minimal non‐photochemical quenching after cold acclimation. In accordance, cold acclimation of wheat neither increased the de‐epoxidation of the xanthophylls nor the content of the PsbS protein. These different responses of photosynthesis to cold acclimation are correlated with pine, reducing its need for assimilates when entering dormancy associated with termination of primary growth, whereas winter wheat maintains a high need for assimilates as it continues to grow and develop throughout the cold‐acclimation period. It appears that without evolving a sustained ability for controlled dissipation of absorbed light as heat throughout the winter, winter green conifers would not have managed to adapt and establish themselves so successfully in the cold climatic zones of the northern hemisphere.  相似文献

The effect of short-term drought stress on the water content, antioxidant system and photosynthetic characteristics was investigated using cucumber (Cucumis sativus L.) seedlings. The results indicated that polyethylene glycol induced water stress reduced water content in shoots of cucumber seedling after treatment of 36 hours, and caused obvious reductions in net photosynthetic rate, stomatal conductance, intercellular CO2 concentration and transpiration of leaves. In addition, water stress significantly reduced the photosynthetic pigment content and inhibited photochemical activity, including actual photochemical efficiency, maximal quantum yield of photosystem II photochemistry and coefficient for photochemical quenching. Meanwhile non-photochemical quenching increased. As responses to drought stress, significant increases in electrolyte leakage, malondialdehyde, superoxide anion and hydrogen peroxide levels were detected in leaves. The superoxide dismutases, catalase, glutathione reductase and dehydroascorbate reductase activities, protein, ascorbate and glutathione content, all decreased and peroxidases activity increased, while ascorbate peroxidase and monodehydroascorbate reductase activities exhibited different trend under different degree of water stress. Therefore, it can be concluded that water stress strongly disrupted the normal metabolism of leaves and restrained water absorption.  相似文献

Cr(VI) significantly reduced rates of net photosynthesis and transpiration and of stomatal conductance. Cr(VI) did not affect the Fv/Fm ratio of chlorophyll fluorescence implying that the primary photochemical processes in photosystem 2 were not affected. However, the efficiency of excitation capture by open PS2 centres, in vivo quantum yield of PS2 photochemistry, and electron transport rate were significantly reduced by Cr(VI). The coefficient of photochemical quenching was reduced with a concomitant increase in coefficient of non-photochemical quenching, suggesting reduced demand for ATP and NADPH due to inhibition of CO2 assimilation. Lipid peroxidation was increased by Cr(VI) and the activities of superoxide dismutase and catalase (CAT) were increased. However, the CAT activity was reduced by high Cr(VI) concentration. The activities of ascorbate peroxidase and glutathione reductase were significantly reduced by Cr(VI) treatment.  相似文献

The relationship between the redox state of the primary electron acceptor of photosystem II (QA) and the rate of O2 evolution in isolated mesophyll chloroplasts from Zea mays L. is examined using pulse-modulated chlorophyll a fluorescence techniques. A linear relationship between photochemical quenching of chlorophyll fluorescence (qQ) and the rate of O2 evolution is evident under most conditions with either glycerate 3-phosphate or oxaloacetate as substrates. There appears to be no effect of the transthylakoid pH gradient on the rate of electron transfer from photosystem II into QA in these chloroplasts. However, the proportion of electron transport occurring through cyclic-pseudocyclic pathways relative to the non-cyclic pathway appears to be regulated by metabolic demand for ATP. Analyses of chlorophyll fluorescence and photosynthetic oxygen evolution were conducted to understand why cold-hardened winter rye (Secale cereale L.) is more resistant to photoinhibition of photosynthesis than is non-hardened winter rye. Under similar light and temperature conditions, leaves of cold-hardened rye were able to keep a larger fraction of the PS II reaction centres in an open configuration, i.e. a higher ratio of oxidized to reduced QA (the primary, stable quinone acceptor of PSII), than leaves of non-hardened rye. Three fold-higher photon fluence rates were required for cold-hardened leaves than for non-hardened leaves in order to establish the same proportion of oxidized to reduced QA. This ability of cold-hardened rye fully accounted for its higher resistance to photoinhibition; under similar redox states of qa cold-hardened and non-hardened leaves of winter rye exhibited similar sensitivities to photoinhibition. Under given light and temperature conditions, it was the higher capacity for light-saturated photosynthesis in cold-hardened than in non-hardened leaves, which was responsible for maintaining a higher proportion of oxidized to reduced QA. 以冷敏感植物甜椒 (CapsicumannuumL .)抗冷性不同的两个品种为试材 ,利用CIRAS 1光合测定系统和FMS2调制式荧光仪 ,在控温控光条件下分析比较了冷锻炼苗与未经锻炼苗的叶片光合特性、叶绿素荧光参数对温度的响应。结果表明 ,随着温度的降低 ,无论是否经过锻炼 ,低温主要通过抑制碳同化能力来影响光合作用 ,并使光能过剩 ,导致低温光抑制。提高环境CO2 浓度以增强暗反应对光能的利用 ,低温光抑制减轻。 5d的亚适温锻炼过程中甜椒叶片已发生一定程度的光抑制 ,但锻炼苗叶片能在低温下维持较高的光系统II光化学效率(ФPSII)、光化学猝灭系数 (qP)和光适应下光系统II最大光化学效率 (Fv′/Fm)值 ;冷锻炼提高了两品种低温下对光抑制的抗性 ,而且对抗冷品种的作用效果更明显  相似文献

The relationship between susceptibility to photoinhibition, zeaxanthin formation and chlorophyll fluorescence quenching at suboptimal temperatures was studied in chilling-sensitive maize and in non-acclimated and cold-acclimated Oxyria digyna , a chilling-tolerant plant of arctic and alpine habitats. In maize, zeaxanthin formation was strongly suppressed by chilling. Zeaxanthin formed during preillumination at 20°C did not protect maize leaves from photoinhibition during a subsequent high-light, low-temperature treatment, as judged from the ratios of variable to maximal fluorescence, Fv/Fm. However, such preillumination significantly increased non-photochemical quenching (qN) at low temperatures, mainly due to an enhancement of the fast-relaxing qN component (i.e., of energy-dependent quenching. qE). In O. digyna , cold-acclimation resulted in an increased zeaxanthin formation in the temperature range of 2.5–20°C. Cold-acclimation substantially decreased the susceptibility towards photoinhibition at 4°C, but qN remained nearly unchanged between 2 and 38°C, as compared to control plants. Effects of cold acclimation on photosynthesis, photochemical quenching and quantum efficiency of photosystem II were small and indicated a slight amelioration only of the function of the photosynthetic apparatus at suboptimal temperatures (2–20°Ct. I) is concluded, that the xanthophyll cycle is strongly influenced by cold acclimation, while effects on the photosynthetic carbon assimilation only play a minor role in O. digyna.  相似文献

The photoinhibition of photosynthesis at chilling temperatures was investigated in cold-acclimated and unhardened (acclimated to +18° C) spinach (Spinacia oleracea L.) leaves. In unhardened leaves, reversible photoinhibition caused by exposure to moderate light at +4° C was based on reduced activity of photosystem (PS) II. This is shown by determination of quantum yield and capacity of electron transport in thylakoids isolated subsequent to photoinhibition and recovery treatments. The activity of PSII declined to approximately the same extent as the quantum yield of photosynthesis of photoinhibited leaves whereas PSI activity was only marginally affected. Leaves from plants acclimated to cold either in the field or in a growth chamber (+1° C), were considerably less susceptible to the light treatment. Only relatively high light levels led to photoinhibition, characterized by quenching of variable chlorophyll a fluorescence (FV) and slight inhibition of PSII-driven electron transport. Fluorescence data obtained at 77 K indicated that the photoinhibition of cold-acclimated leaves (like that of the unhardened ones) was related to increased thermal energy dissipation. But in contrast to the unhardened leaves, 77 K fluorescence of cold-acclimated leaves did not reveal a relative increase of PSI excitation. High-light-treated, cold-acclimated leaves showed increased rates of dark respiration and a higher light compensation point. The photoinhibitory fluorescence quenching was fully reversible in low light levels both at +18° C and +4° C; the recovery was much faster than in unhardened leaves. The change of chlorophyll fluorescence parameters in froze leaves of 3 leaf-age seedlings were examined using two winter barley cultivars (Chumai 1 and Mo 103) differing in cold tolerance to investigate physiological response to low temperature as affected by cold acclimation (under 3/1 degrees C, day/night for 5 days before freezing treatment) and irradiation size (high irradiance: 380+/-25 micromol m(-2)s(-1) and low irradiance: 60+/-25 micromol m(-2)s(-1)) during recovery. The results showed that non-lethal freezing shock (exposed to -8 degrees C for 18 h) did not obviously affect maximum quantum efficiency in photosystem II (PSII), but dramatically increased non-photochemical quenching and reduced effective quantum yield in PSII. Cold acclimation significantly improved stability of photosynthetic function of leaves after freezing stress through buffering excessive energy and alleviating photoinhibition during recovery, indicating it increased recovery ability of barley plants from freezing injury. High irradiance was quite harmful to the stability of PSII in barley plants during recovery from freezing injury. The electron transport rate of PSII varied with cold-acclimation, irradiance and genotype. Cold acclimation caused significant increase in electron transport rate of PSII for relatively tolerant cultivar Mo 103, but not for relatively sensitive cultivar Chumai 1. It can be concluded that some chlorophyll fluorescence parameters during recovery from freezing shock may be used as the indicators in identification and evaluation of cold tolerance in barley.  相似文献

Developmental changes of photochemical and non-photochemical processes and the antioxidant system in the shaded peel vs the sun-exposed peel of 'Gala' apple and their responses to sudden exposure of high light were determined to understand the susceptibility of the shaded peel to high light damage with fruit development. As fruit developed, actual PSII efficiency of the shaded peel decreased, whereas non-photochemical quenching (mainly the slow component) increased at any given PFD. Photochemical quenching coefficient of the shaded peel decreased at any given PFD with fruit development. As fruit developed, the activity of superoxide dismutase, ascorbate peroxidase and dehydroascorbate reductase and the level of reduced ascorbate and total ascorbate decreased; the activity of monodehydroascorbate reductase and glutathione reductase remained low, whereas catalase activity and the level of reduced glutathione and total glutathione increased in the shaded peel. Exposure to high light (1500 micromol m(-2) s(-1)) for 2 h significantly decreased the maximum quantum efficiency of PSII (F(V)/F(M)) in the shaded peel at each developmental stage, with the decrease being larger with fruit development. The F(V)/F(M) of the sun-exposed peel was also decreased by the high light treatment, but the decrease was much smaller than that in the shaded peel at each developmental stage. We conclude that the shaded peel of apple fruit becomes more sensitive to photoinhibition with fruit development, and this increased sensitivity is apparently related to the decease in the overall capacity for photosynthesis and photoprotection of the shaded peel with fruit development.  相似文献

以湖南省永州市冷水滩采穗圃中的美国山核桃为试材,研究了叶绿素荧光参数的日变化规律。结果表明:初始荧光(Fo)、最大荧光(Fm)、PSII原初光能转化效率(Fv/Fm)、光合量子产额(Yield)、光化学猝灭系数(qP)、非光化学猝灭系数(qN)和表观电子传递速率(ETR)均存在着明显的日变化。其中Fv/Fm、Fm、Yield、qP均呈先下降后上升的趋势,在中午强光下降低到最低值;qN则呈先上升后下降的趋势,在中午时分达到峰值;Fo呈下降趋势,部分品种傍晚稍有回升,但仍比早晨低;ETR日变化呈双峰曲线。不同品种间Fv/Fm、Yield、ETR、qP、qN对光强和温度的响应也存在着明显差异,可作为鉴定品种耐光抑制能力大小的指标。  相似文献

Comparative analysis revealed that a xantha rice mutant (cv. Huangyu B) had higher ratios of chlorophyll (Chl) a/b and carotenoids/Chl, and higher photosynthetic efficiency than its wild type parent (cv. II32 B). Unexpectedly, the mutant had higher net photosynthetic rate (P N) than II32 B. This might have resulted from its lower non-photochemical quenching (qN) but higher maximal photochemical efficiency (FV/FM), higher excitation energy capture efficiency of photosystem 2 (PS2) reaction centres (FV′/FM′), higher photochemical quenching (qP), higher effective PS2 quantum yield (ΦPS2), and higher non-cyclic electron transport rate (ETR). This is the first report of a chlorophyll mutant that has higher photosynthetic efficiency and main Chl fluorescence parameters than its wild type. This mutant could become a unique material both for the basic research on photosynthesis and for the development of high yielding rice cultivars.  相似文献

通过比较棉花(Gossypium hirsutum)幼叶和完全展开叶气体交换参数及叶绿素荧光特性的差异, 探讨高光强下幼叶的光抑制程度及明确光保护机制间的协调机理。在田间自然条件下, 以棉花刚展平的幼嫩叶片(幼叶)和面积已达到最大的完全展开叶片为研究对象, 通过测定不同发育阶段叶片气体交换参数及叶绿素a荧光参数的变化, 并运用Dual-PAM100对不同发育阶段的叶片进行快速光响应曲线的拟合。结果表明: 幼叶和完全展开叶片在光合、荧光特性方面表现出明显的差异。与完全展开叶相比, 较低的叶绿素(Chl)含量和气孔导度(Gs)是幼叶较低净光合速率(Pn)的限制因素, 从而直接导致其光系统II (PSII)实际光化学效率(ΦPSII)和光化学猝灭系数(qP)的降低。在1800 μmol·m-2·s-1光强以下, 完全展开叶具有较强的围绕PSI循环的电子流(CEF), 有利于合成ATP, 是其具有较高光合能力的原因之一。相同光强下, 幼叶较低的光饱和点(LSP)更易受光抑制, 但其PSII原初光化学效率(Fv/Fm)的日变化幅度显著小于完全展开叶, 说明强光下幼叶通过类胡萝卜素(Car)猝灭单线态氧、光呼吸(Pr)、热耗散(NPQ)以及PSI-CEF等光保护机制能有效地耗散过剩的光能, 从而避免其光合机构发生光抑制。  相似文献

Photoinhibition of photosynthesis was studied in intact barley leaves at 5 and 20°C, to reveal if Photosystem II becomes predisposed to photoinhibition at low temperature by 1) creation of excessive excitation of Photosystem II or, 2) inhibition of the repair process of Photosystem II. The light and temperature dependence of the reduction state of QA was measured by modulated fluorescence. Photon flux densities giving 60% of QA in a reduced state at steady-state photosynthesis (300 mol m–2s–1 at 5°C and 1200 mol m–2s–1 at 20°C) resulted in a depression of the photochemical efficiency of Photosystem II (Fv/Fm) at both 5 and 20°C. Inhibition of Fv/Fm occurred with initially similar kinetics at the two temperatures. After 6h, Fv/Fm was inhibited by 30% and had reached steady-state at 20°C. However, at 5°C, Fv/Fm continued to decrease and after 10h, Fv/Fm was depressed to 55% of control. The light response of the reduction state of QA did not change during photoinhibition at 20°C, whereas after photoinhibition at 5°C, the proportion of closed reaction centres at a given photon flux density was 10–20% lower than before photoinhibition.Changes in the D1-content were measured by immunoblotting and by the atrazine binding capacity during photoinhibition at high and low temperatures, with and without the addition of chloramphenicol to block chloroplast encoded protein synthesis. At 20°C, there was a close correlation between the amount of D1-protein and the photochemical efficiency of photosystem II, both in the presence or in the absence of an active repair cycle. At 5°C, an accumulation of inactive reaction centres occurred, since the photochemical efficiency of Photosystem II was much more depressed than the loss of D1-protein. Furthermore, at 5°C the repair cycle was largely inhibited as concluded from the finding that blockage of chloroplast encoded protein synthesis did not enhance the susceptibility to photoinhibition at 5°C.It is concluded that, the kinetics of the initial decrease of Fv/Fm was determined by the reduction state of the primary electron acceptor QA, at both temperatures. Spinach (Spinacia oleracea L.) plants were acclimated to 1° C or maintained at 18° C under the same light regime (260–300 mol photons·m–2·s–1). The cold acclimation led to several metabolic and biochemical changes that apparently include improved protection of the photosynthetic apparatus against active oxygen species. In particular, cold-acclimated leaves exhibited a considerably higher ascorbate content and significantly increased activities of superoxide dismutase, ascorbate peroxidase, and monodehydroascorbate reductase in the chloroplasts. The level of dehydroascorbate reductase did not alter. Catalase activity decreased. The photosynthetic pigment composition of cold-acclimated spinach was characterized by increased levels of the xanthophylls lutein + zeaxanthin and violaxanthin.
