Pollen plantlets of Datura metel L. have been successfully rearedin vitro through anther culture on a nutrient medium supplementedwith coconut milk (15 per cent v/v) The early segmentation patternof the proembryos and their subsequent differentiation intoheart, torpedo, and cotyledonary stages were traced. More than100 plantlets were grown to maturity on soil. They grew normallyand produced flowers. Root-tip and leaf-squash preparationsshowed variable numbers of chromosomes in their cells, and thisindicated the occurrence of ploidy plants among the population.Frequency determinations showed the predominant types to bediploid (70 per cent), triploid (24 per cent), and haploid (6per cent). Differences in the morphology of mature plants inregard to floral size, meiotic irregularities in PMCs, formationof giant pollen grains, differential fruitset, somatic mosaicismin respect of chromosome number, and aneuploidy of cells, wererecorded. Supplementing pollen suspension cultures with CO2 (3–5per cent) caused a marked increase in germination and tube growthin vitro in Brassica campestris L. cv. toria. A weakening ofself-incompatibility by increased CO2 levels from 3–5per cent was observed. The percentage of pollen tubes whichpenetrated the cuticle layer of stigmatic papilla cells in self-pollinatedpistils was high when CO2 level was 5 per cent. Phosphoenolpyruvate (PEP) carboxylase activity was greater in the pollengerminated in 4 per cent CO2 as compared to air (0.03 per cent).A possible role of CO2 for self-recognition and control of pollentube growth is proposed, proposed. 1. Forty-three strains of moulds, selected from ten species,were examined to ascertain their capabilities as producers offat, when grown in four different solutions of nutrient inorganicsalts together with sucrose. The three highest yields of feltwere given by Penicillium javanicum van Beyma, P. Soppi Zaleski,and Aspergillus nidulans Eidam, in that order. The fat contenton felt weight was maximal at 34·8 per cent, with P.Soppi, at 28·4 per cent. with Fusarium lini(1), and at25·8 per cent. with A. nidulans; on sugar utilized itwas maximal at 11·4 per cent. with P. Soppi, at 7·9per cent, with A. nidulans, and at 5·6 per cent. withF. lini(1). 2. The mineral, carbohydrate, protein, amino acid, lipid and energycontents of the seven main pollens gathered by honey-bees insouth-east Scotland were determined. Neither dry matter (mean73 per cent) nor ash (mean 3 per cent) were very variable. Thecalcium content of all pollens was relatively similar as werethe magnesium, potassium and sodium contents but manganese whichwas relatively high in ericaceae and phosphorus which was lowin Trifolium were more variable. Acer pollen contained mostprotein and Fagus least, and of the amino acids only serine,cystine and histidine were significantly different in the differentpollens. Over 30 per cent of all pollens was a mixture of fructoseand glucose, ericaceae containing most and Acer least. Hemicellulose(mean 7 per cent) was high in Ranunculus and low in Acer, cruciferaeand Fagus. ‘Cellulose’ (mean 0•5 per cent)was high in cruciferae and low in ericacea. Ether extract (mean5 per cent) was high in cruciferae and Fagus. The mean grossenergy of pollen was 23 kJ g–1. Fresh pollen is comparable with lean meat as a source of energyand protein, and is more like dormant plant seeds than herbagein its water, ash and protein content but stores energy as sugarrather than starch or lipid.  相似文献

The uptake of 14C-glucose into cells of Nitella translucenshas been investigated under experimental conditions previouslyused in studies of the ionic relations of these cells. Glucoseentry was considerably stimulated by light, and under aerobicconditions the fluxes remained constant for many hours. Theinflux of glucose was inhibited by over 80 per cent at low temperature(4° C) and by over 90 per cent by the uncoupler carbonylcycanide-m-chlorophenylhydrazone. 2-Deoxy-D-glucose was a non-competitiveinhibitor of glucose uptake both in light and darkness. Cyclicphotophosphorylation promoted the influx (with decreasing efficiency)for several hours. It is suggested that an ATP-dependent transportprocess controls glucose entry to the cells, and that passivediffusion is of little significance.  相似文献

A study has been made, using C14 as a tracer, of the variationswhich occur according to the physiological condition of thecells in the distribution of carbon fixed, both in the lightand in the dark, by the diatom Navicula pelliculosa (Bréb.)Hilse during periods of 30 to 300 seconds. Fixation into thefollowing cell fractions was determined: (A) material solublein 80 per cent. ethanol but insoluble in benzene, (B) materialsoluble both in 80 per cent. ethanol and in benzene, and (C)material insoluble in 80 per cent. ethanol. Carbon fixed inphotosynthesis was incorporated, rapidly and in amounts representingup to 70 per cent. of the total fixation, into fractions B andC, as well as into fraction A. Considerable variation was foundin the proportions of carbon entering the three fractions inthe light; in actively growing cells the proportion enteringfraction C preponderated over that in B, corresponding to thesynthesis of protein, whereas in nitrogen-deficient cells fixationin B was the greater, corresponding to the synthesis of fat.These patterns changed only slowly, over periods of days, followingthe transfer of cells to altered conditions of nitrate supply.However, when ammonium nitrogen was supplied to nitrogen-deficientcells a marked change in distribution of carbon fixed occurredwithin 5 minutes, fixation in fraction B falling to a low valueand that in A rising correspondingly. In cells subjected toprolonged nitrogen-deficiency, fixation in fraction B fell toa relatively low value but the proportion which this fractionformed of the total dry matter in the cells rose as a resultof an increased rate of loss from the cells of constituentsother than lipides. The distribution of carbon fixed was alsodependent on light intensity. Fixation in fractions B and Crose relatively to that in A as light intensity was increasedup to 100 foot-candles but fell again at the highest intensityused, 2,000 foot-candles. These results are discussed with particularreference to the relationship between fat accumulation and photosynthesisin algae.  相似文献

Brassica oleracea pollen was applied to a basic medium of 1.5per cent agar and 15 per cent sucrose to which flavanoids wereadded at three concentrations. Two types of agar were used;with agar 1, quercetin at a concentration of 0.5 x 10–3per cent gave an increase in percentage germinating grains.With agar 2, an increase in germination occurred with kaempferoland naringin at concentrations of 0.5 x 10–3 and 0.5 x10–1 per cent respectively. Increase in pollen tube lengthoccurred with agar 2 and quercetin at a concentration of 0.5x 10–3 per cent. The stigma tissue of B. oleracea contains at least three andthe pollen at least one glycoside of quercetin. The sugars inthe glycosides were not identified. Pollen germination and pollentube extension were not stimulated exclusively by the flavanoidspresent in the stigma. The flavanoid composition of the stigmadid not vary amongst five different S-allele genotypes, indicatingthat flavanoids are probably not directly involved in the incompatibilityreaction of B. oleracea.  相似文献

A spontaneously occurring somatic mutant of Triticum turgidumdicoccoides showed close morphological resemblance to T. timopheevi(AAGO). The hybrid between the mutant and the T. turgidum dicoccoides‘ mother’ plant was completely sterile, with verylow pollen fertility (0·33 percent). It exhibited a reasonablyhigh frequency of trivalents and quadrivalents at first metaphaseof meiosis, indicating that the mutation involved substantiallevels of chromosome rearrangement. The hybrid between the mutantand T. timopheevi had reasonably high fertility (53·5per cent) and high pollen fertility (86·6 per cent) andalmost regular bivalent formation at first metaphase of meiosis. It is proposed that T. timopheevi could have arisen in consequenceof somatic macromutation from T. turgidum dicoccoides givingrise to spontaneous speciation. The G genome of T. timopheeviis possibly monophyletic in origin, arising from rearrangementof chromosomes of the B genome of tetraploid wheat. Nuclear volumes and cell areas were determined for seven regionsof the meristem of roots of Zea mays. Roots were fixed in 10per cent neutral buffered formalin, in 3 per cent glutaraldehydeor in acetic acid/alcohol; they were prepared as sections oralls were teased apart. Mean volumes of interphase nuclei weresimilar in all regions of the root except the vascular tissueof the stele. Mean nuclear volumes and the overall range ofvolumes were similar in sub-populations of cells with differentproportions of G1, S and G2 cells, e.g. in row I of root capinitials, whose cells lack a G1 phase, and in quiescent centrecells, which are mainly in G1. Nuclear volume does not appearto be closely correlated with DNA content. Nuclear volumes covereda 6 to 12-fold range within a meristem and even within specificregions, in which cells are part of the same cell lineages,there was a 4- to 9-fold range. Nuclear volumes were comparedin sister cells in rows I and II of the root cap initials. In10 per cent of the pairs, sister nuclei had identical volumes;the other pain had different volumes and mean difference was68 µm3. Mechanisms by which this variability could begenerated are discussed, particularly asymmetry, at mitoses,of factors that regulate nuclear growth. The distribution of radioactivity in Chlorella during dark 14CO2fixation was investigated either (a) in normal cells with andwithout added ammonium chloride, or (b) in nitrogen-starvedcells supplied with intermediates of the Krebs-Henseleit ureacycle. In the control experiments almost all the activity was presentin compounds of or associated with, the tricarboxylic acid cycle. The amino-acids citrulline and arginine became radioactive onlyin the presence of ammonia or ornithine where initially theycomprised 40–60 per cent. of the total activity, reactionsof the Krebs–Henseleit urea cycle being implicated intheir formation. No evidence could be found for a complete ureacycle. Unidentified compounds deriving their radioactivity fromthe C4 carbon of citrulline and/or arginine were detected andformed up to 40 per cent. of the total 14CO2 incorporated after25 min.  相似文献

The effect of genes ar (violet flowers, small hilum) and n (thick,fleshy pod wall) on whole plant in senescence peas was examinedby comparing Ar- with arar and N- with nn plants in segregatingprogenies. Homozygosity for ar or n significantly delayed the time whenthe plants were ready for harvest of their entire seed crop.These genes did not delay either the onset of reproduction orthe onset of apical arrest in the first instance. However, whereasAr- N- plants almost invariably senesced and died as the firstseed crop matured, the majority of arar and/or nn plants entereda period of secondary growth and a further fruiting cycle. Comparedwith Ar- plants, arar plants had over twice as many pods andseeds but individual seeds were 58 per cent lighter and totalseed yield (wt) was 19 per cent less. Pod length was unaffected.Compared with N-plants, nn plants had shorter pods (16 per cent),fewer seeds per pod (21 per cent), smaller seeds (20 per cent)and a lower total seed yield (wt 14 per cent less). It appearsthat ar and n impose a lower metabolic drain per reproductivenode as a consequence of their effects on hilum anatomy andpod morphology, respectively. These mutants disrupt the normalpattern of monocarpic senescence by breaking the coordinationbetween apical arrest and subsequent events. The developingseed crop delimited by the first arrest fails to cause plantdeath, possibly because sink size is less than in normal counterparts. The results of experiments on the effect of high pressures ofoxygen on apples are outlined. It is shown that partial pressuresof the order of 500 per cent, oxygen are toxic to the tissuesand that there is no evidence of an initial increase in CO2,output over that in 1 atmosphere 100 per cent, oxygen. It isfurther shown that the actual pressure (up to 20 atmospheres)is not responsible for the disorganization of the cells butthat there is specific oxygen toxicity.  相似文献

The metabolism of [2-14C]thymine, [2-14C]thymidine, [2-14C]uraciland [14C]uridine was investigated in protoplasts obtained fromsuspension cultures of Catharanthus roseus. Most of the exogenouslysupplied thymine, thymidine and uracil was degraded, and salvageof these pyrimidines accounted for 5–36 per cent of thetotal amount of 14C-labelled precursors which was metabolized.However, more than 80 per cent of the labelled uridine was utilizedfor the biosynthesis of nucleotides and nucleic acids, and therest was degraded. In contrast to the results from protoplastsof sugar cane cells in suspension culture, the data indicatethat protoplasts possess a pathway for the degradation of pyrimidines,and that the overall metabolism of these pyrimidines in protoplastsis very similar to the metabolism in the intact cells. Autoradiography and liquid scintillation techniques were usedto trace the pattern of photoassimilate translocation in twoperennial forest herbs, Aster acuminatus Michx. and Clintoniaborealis (Ait.) Raf. Vegetative shoots of C. borealis emergeearly each spring from the growing apices of a long-lived rhizomesystem. Vegetative shoots of A. acuminatus emerge in late springfrom rhizomes that decay within 2 years. In both species rametssurvive for only one growing season. Any connected plants aresisters. Mature leaves of these two species were exposed for1 h periods to 14CO2 during spring and summer. Radioactivitycould subsequently be detected in exposed leaves (16–84per cent, depending on the season), adjacent above ground plantparts (0.7–23 per cent), roots and rhizomes (4.9–84per cent) and, when present, flowers (4–8.5 per cent).Old rhizomes of C. borealis are prominent storage sites forphotosynthate. In A. acuminatus, no significant translocationbetween sister ramets (i.e. above ground shoots connected bya common rhizome) was observed. In C. borealis, there was small,but consistent translocation between sister ramets (0.2–4per cent). Disturbance of unexposed sister ramets by defoliation,shading or herbivory increased the flow of photoassimilatesto disturbed parts in C. borealis, but not in A. acuminatus.Based on the absence of translocation flow, ramets of A. acuminatusmay be regarded as physiologically independent. Connected rametsof C. borealis show physiological integration. These resultsare correlated with ecological differences between the two species. In oogonia of Saprolegnia terrestris some nuclei completed twoconsecutive divisions to form small gamete nuclei, others wererestituted at intermediate division stages. Possible multiplechromosome associations were seen in some late prophase figuresof the first division. Seven to eight chromatin bodies werepresent in pro-metaphase figures. All nuclei except a smallnumber of gamete nuclei were broken down prior to formationof oospheres. Antheridial nuclei either completed divisionsto form small, gamete nuclei or were restituted to form large,torus-shaped nuclei. In oogonia of S. diclina, nuclei enteredprophase but no subsequent division stages were seen. Torus-shapednuclei, but not gamete nuclei, were seen in antheridia. Oosporenuclei of both S. terrestris and S. diclina varied both in size(about 5, 10 or 50–100 µm3) and number (0, 1 or2); 7.5 per cent of oospores of S. terrestris and 54 per centof oospores of S. diclina contained no nucleus or one smallnucleus only. The oospore abortion rate was 7.3 per cent inS. terrestris and 50.4 per cent in S. diclina. A long-term, steady-state 13CO2 assimilation system at a constantCO2 concentration with a constant 13C abundance was designedand applied to quantitative investigations on the allocationof photoassimilated carbon in nodulated soya bean (Glycine maxL.) plants. The CO2 concentration in the assimilation chamberand its 13C abundance were maintained constant with relativevariances of less than ±0.5 per cent during an 8-h assimilationperiod. At the termination of 8-h 13CO2 assimilation by plantsat early flowering stage, the currently assimilated carbon relativeto total tissue carbon (measured by the degree of isotopic saturation)were for young leaves (including flower buds), 13.9 per cent;mature leaves, 15.7 per cent; stems+petioles, 5.9 per cent;roots, 5.4 per cent and nodules, 6.9 per cent, 48 h after theend of the 13CO2 assimilation period, they were 12.3, 7.5, 7.4,6.8 and 6.1 per cent, respectively. The treatment with a highconcentration of nitrate in the nutrient media significantlydecreased the allocation of 13C into nodules. Experiments on13CO2 assimilation by plants at the pod-filling stage were alsoconducted. Labelling by 13C was weaker than at the early floweringstage, but an intense accumulation of 13C into reproductiveorgans was observed. A morphometric analysis of the ultrastructure of columella statocytesin primary roots of Zea mays was performed to determine theprecise location of cellular organelles in graviperceptive cells.Vacuoles occupy the largest volume in the cell (11.4 per centof the protoplasm). The nucleus (9.51 per cent), amyloplasts(7.57 per cent), mitochondria (3.42 per cent), spherosomes (2.13per cent) and dictyosomes (0.55 per cent) occupy progressivelysmaller volumes of the statocytes. All organelles are distributedasymmetrically within the cell. Amyloplasts, spherosomes anddictyosomes are found in greatest numbers (and relative volumes)in the lower (i.e. ‘bottom’) third of the cell.The largest numbers and relative volumes of mitochondria arein the lower and middle thirds of the cell. Nuclei tend to befound in the middle third of the statocytes. Only the hyaloplasmis concentrated in the upper (i.e. ‘top’) thirdof Z. mays statocytes. When the sedimentation of amyloplasts(and the resulting exclusion of other organelles from the lowerthird of the cell) is corrected for, all cellular constituentsremain asymmetrically distributed within the cell. Therefore,the sedimentation of amyloplasts alone is not responsible forthe differential distribution of other cellular organelles inZ. mays statocytes. Inoculating Proctor barley leaves with Erysiphe graminis decreasedrates of photosynthesis, after an initial lag period, and increasedrespiration. Increasing the area inoculated progressively decreased ratesof photosynthesis, but the effects cannot be attributed to asimple loss of leaf area. When less than 30 per cent of a leafwas inoculated, decreases were equivalent to area losses greaterthan those inoculated; when more than 30 per cent was inoculatedthe photo-synthetic losses were equivalent to area losses lessthan those inoculated. Although the relative effects of E. graminis on photosynthesisand respiration were of the same order, the absolute effectson photosynthesis were greater than those on respiration. Inoculating30 per cent of a leaf decreased photosynthesis by 5–6mg CO2/dm2/hr from 12.9 in the uninoculated controls to 7.3.Respiration increased by 0.6 mg CO2/dm/hr, from 1.7 to 2.3-  相似文献

Development of 2-mm embryos of Lathyrus articulatus L. in vitrowas stimulated by placing them initially on an agar medium containing8 or 12 per cent sucrose. Germination took place after transferto a similar medium with 4 per cent sucrose. Successful cultureof 0.5-mm embryos was achieved under similar conditions butit was necessary to supplement the initial medium with 10 percent coconut milk. Using this technique, the hybrid L. clymenumL. x L. articulatus was raised from embryos which would haveotherwise aborted in vivo.  相似文献

Sycamore and soybean cell suspensions were subjected to osmoticstress by culturing for one passage in media supplemented with6 per cent mannitol or sorbitol. The effects on growth wereto reduce cell number and biomass (d. wt) production throughoutthe culture period by about 30 per cent. Ultrastructural studiesat the early exponential phase of culture growth indicated similarreductions in cell wall thickness in both species.
