Amylose content and starch granule size in grains influence rice quality, which differs between the early (ES) and late season (LS). The objective of this study was to determine the variation of amylose content and starch granule size between seasons and find the main reasons (e.g., temperature and solar radiation) for the observed variation. Field experiments with six rice varieties (three high and three low amylose content rice) planted in the ES and LS were conducted in 2016 and 2017, respectively. The mean temperatures during the filling stage were higher in ES, however, the daily temperatures at 7-10 days after flowering (DAF) in 2016, and at 5-10, 13-14 DAF in 2017 were higher in LS. The results showed that amylose content in LS was lower than in ES with high amylose content rice varieties (HACV); the opposite trend occurred with low amylose content rice varieties (LACV). The mean starch granule diameter was higher in LS than ES in 2016, but the opposite result occurred in 2017 with all rice. Our results suggest that higher temperatures increased and decreased the amylose content in HACV and LACV, respectively. Temperatures at 5-15 DAF were important for the formation of starch granules: lower temperatures during 10-14 DAF increased the proportion of larger starch granules (d>6.21 μm, some with d>13.3 μm), and higher temperatures at 5-6 DAF increased the proportion of starch granules with diameter 4.24-6.21 μm.  相似文献

The synthesis of amylose in amyloplasts is catalyzed by granule-bound starch synthase (GBSS). GBSS gene expression was inhibited via antisense RNA in Agrobacterium rhizogenes-transformed potato plants. Analysis of starch production and starch granule composition in transgenic tubers revealed that reduction of GBSS activity always resulted in a reduction of the production of amylose. Field experiments, performed over a 2-year period, showed that stable inhibition of GBSS gene expression can be obtained. Microscopic evaluation of iodine-stained starch granules was shown to be a sensitive system for qualitative and quantitative examination of amylose formation in starch granules of transgenic potato tubers. In plants showing inhibition of GBSS gene expression, the reduced amylose content in tuber starch was not a consequence of a lower amylose content throughout the entire starch granule. Starch granules of transgenic tubers were found to contain amylose at a percentage similar to wild-type starch in a core of varying size at the hilum of each granule. This indicated that reduced GBSS gene expression results in amylose formation in a restricted zone of the granules. The size of this zone is suggested to be dependent on the GBSS protein level. During development of the granules, the available GBSS protein is thought to become limiting, resulting in the formation of starch that lacks amylose. RNA gel blot analysis of tuber tissue showed that inhibition of GBSS gene expression resulted in a reduced GBSS mRNA level but did not affect the expression level of other starch synthesizing enzymes. Antisense RNA could only be detected in leaf tissue of the transgenic plants.  相似文献

Two-year-old plants of Karwinskia humboldtiana Zucc. and Karwinskia parvifolia Rose grown from the seeds in greenhouse were transferred to the growth cabinet and cultivated for two months under different temperature regimes (35/20 °C - the summer temperature regime, SR, and 20/5 °C - the winter temperature regime, WR). These temperatures were similar to the temperature conditions in the natural areas of the species studied (Nuevo León, Mexico). The root respiration rate was higher in the plants cultivated under SR than in those under WR. Roots of K. parvifolia res faster in both temperature regimes than roots of K. humboldtiana. Starch content in roots was higher in the plan cultivated under SR, however, concentrations of the other investigated organic and inorganic compounds were higher in the plants cultivated under WR. In K. humboldtiana roots, higher concentration of reducing sugars, carbon and hydrogen were found than in K. parvifolia.  相似文献

Analyses were made of the fatty-acid composition of Candida utilis NCYC 321 grown in a chemostat at a dilution rate (equal to growth rate) of 0.1 hr−1 and at temperatures in the range of 30 to 15 C and dissolved oxygen tensions between 75 and <1 mm of Hg. Cells grown under glucose limitation or NH4+ limitation contained mainly C16:0, C16:1, C18:0, C18:1, C18:2, and C18:3 acids as detected by gas-liquid chromatography of methyl esters of the acids from lipids extracted with chloroform-methanol. The relative proportions of these acids varied with the growth temperature and the dissolved-oxygen tension in the culture. A decrease in growth temperature from 30 to 20 C led to an increased synthesis of unsaturated acids in cells grown under either limitation at a fixed-oxygen tension in the range of 75 to 5 mm of Hg. In cultures with a dissolved-oxygen tension of 1 and <1 mm of Hg, a further decrease in temperature to 15 C caused an increased synthesis of unsaturated fatty acids. A decrease in dissolved-oxygen tension led to a diminished synthesis of unsaturated fatty acids in cells grown at a fixed temperature under either limitation. Cells grown at a fixed temperature under glucose limitation synthesized a greater proportion of C16 acids at the expense of C18 acids as the dissolved oxygen tension was decreased from 75 to <1 mm of Hg. A preferential synthesis of C16 acids also occurred as the growth temperature was decreased from 30 to 15 C in cells grown under glucose limitation at a fixed-oxygen tension. The same effect was observed in cells grown under NH4+ limitation when the temperature was lowered from 30 to 20 C; but when the temperature was decreased further to 15 C, the cells synthesized a slightly greater proportion of C18 acids. Synthesis of a large proportion of C16 acids was accompanied by an excretion of pyruvate, and occasionally traces of 2-ketoglutarate, and an increased intracellular accumulation of certain amino acids.  相似文献

Successful inoculation of peanuts and cowpeas depends on the survival of rhizobia in soils which fluctuate between wide temperature and moisture extremes. Survival of two cowpea rhizobial strains (TAL309 and 3281) and two peanut rhizobial strains (T-1 and 201) was measured in two soils under three moisture conditions (air-dry, moist (−0.33 bar), and saturated soil) and at two temperatures (25 and 35°C) when soil was not sterilized and at 40°C when soil was sterilized. Populations of rhizobia were measured periodically for 45 days. The results in nonsterilized soil indicated that strain 201 survived relatively well under all environmental conditions. The 35°C temperature in conjunction with the air-dry or saturated soil was the most detrimental to survival. At this temperature, the numbers of strains T-1, TAL309, and 3281 decreased about 2 logs in dry soil and 2.5 logs in saturated soil during 45 days of incubation. In sterilized soil, the populations of all strains in moist soil increased during the first 2 weeks, but decreased rapidly when incubated under dry conditions. The populations did not decline under saturated soil conditions. From these results it appears that rhizobial strains to be used for inoculant production should be screened under simulated field conditions for enhanced survival before their selection for commercial inoculant production.  相似文献

The short-term relationships of monoterpene emission with temperature and relative humidity were studied in Pinus halepensis L. and Quercus ilex L. seedlings grown in air-conditioned chamber. In P. halepensis terpene emission rate increased with temperature (from 15 to 35 °C) and relative humidity (from 40 - 60 to 65 - 95 %). In Q. ilex, a terpene non-storing species, it increased with temperature only at high relative humidities but not at relative humidities lower than 60 %. 利用益生菌发酵制备具有生物学功能特性的发酵乳制品,特别是在构建功能性食品方面,对人类的健康及人体内共生菌群的开发具有重大意义。以瑞士乳杆菌(Lactobacillushelveticus)为研究菌种,对瑞士乳杆菌利用乳清粉的发酵条件进行了系统的研究,并对发酵条件进行了优化。实验结果表明,瑞士乳杆菌在33℃,发酵液的初始pH为5.5,接种量为1.5%时蛋白酶活力最强,发酵液中游离氨基酸含量为3.004μg/ml活菌数达到108cfu/ml。通过对瑞士乳杆菌的发酵动力学的研究,确定了该菌生长特性,以及乳清蛋白水解进程曲线和产酸情况。  相似文献

Cold-set whey protein (WP) gels with addition of xanthan or guar were evaluated by mechanical properties and scanning electron microscopy. Gels were formed after the addition of different amounts of glucono-δ-lactone to thermally denatured WP solutions, leading to different acidification rates and final pH values. At lower acidification rates and higher final pH, gels showed more discontinuous structure and weaker and less elastic network, which was attributed to a predominance of phase separation during gel formation due to slower gelation kinetics. In contrast, at higher acidification rates and lower final pHs, gelation prevailed over phase separation, favoring the formation of less porous structures, resulting in stronger and more elastic gels. The gels’ fractal dimension (D f; structure complexity) and lacunarity were also influenced by the simultaneous effects of gelation and phase separation. For systems where phase separation was the prevailing mechanism, greater lacunarity parameters were usually observed, describing the heterogeneity of pore distribution, while the opposite occurred at prevailing gelation conditions. Increase in guar concentration or lower final pH of xanthan gels entailed in D f reduction, while the increase in xanthan concentration resulted in higher D f. Such a result suggests that the network contour length was rugged, but this pattern was reduced by the increase of electrostatic interactions among WP and xanthan. Guar addition caused the formation of gel network with smoother surfaces, which could be attributed to the guar–protein excluded volume effects leading to an increase in protein–protein interactions.  相似文献

Inflammation is a major mediator of CKD progression and is partly driven by altered gut microbiome and intestinal barrier disruption, events which are caused by: urea influx in the intestine resulting in dominance of urease-possessing bacteria; disruption of epithelial barrier by urea-derived ammonia leading to endotoxemia and bacterial translocation; and restriction of potassium-rich fruits and vegetables which are common sources of fermentable fiber. Restriction of these foods leads to depletion of bacteria that convert indigestible carbohydrates to short chain fatty acids which are important nutrients for colonocytes and regulatory T lymphocytes. We hypothesized that a high resistant starch diet attenuates CKD progression. Male Sprague Dawley rats were fed a chow containing 0.7% adenine for 2 weeks to induce CKD. Rats were then fed diets supplemented with amylopectin (low-fiber control) or high fermentable fiber (amylose maize resistant starch, HAM-RS2) for 3 weeks. CKD rats consuming low fiber diet exhibited reduced creatinine clearance, interstitial fibrosis, inflammation, tubular damage, activation of NFkB, upregulation of pro-inflammatory, pro-oxidant, and pro-fibrotic molecules; impaired Nrf2 activity, down-regulation of antioxidant enzymes, and disruption of colonic epithelial tight junction. The high resistant starch diet significantly attenuated these abnormalities. Thus high resistant starch diet retards CKD progression and attenuates oxidative stress and inflammation in rats. Future studies are needed to explore the impact of HAM-RS2 in CKD patients.  相似文献

The growth and the content of endogenous cytokinins (CKs) of current-year-old shoots from juniper plants (Juniperus communis L.) growing over and off ore site were compared. The juniper shoots from ore site (M plants) had higher metal content and exhibited delayed growth. Less bases and nucleosides of Z- and iP- type CK and more iP-conjugates were present in the M shoots. These changes were probably due to inhibited CK export from the roots and/or altered CK metabolism forming less biologically active CKs.  相似文献

以蜡质玉米淀粉为原料在优化条件下制备具有乳化能力的低粘度辛烯基琥珀酸淀粉钠(SSOS)。用Brabender粘度仪测定淀粉糊的粘度及其稳定性,发现酶处理前SSOS糊的粘度较蜡质玉米淀粉提高,但稳定性变化不大。研究发现低粘度SSOS微胶囊包埋柠檬油的百分率在57.4%至87.6%(w/w)之间,并取决于SSOS的葡萄糖值。在壁材与芯材的质量比大于3比2,葡萄糖值为12.8时,微胶囊包埋的产率和效率达到最高值。微胶囊在40℃下放置贮存3周后,其微胶囊化效率仅降低3%。通过扫描电镜观察,发现微胶囊为球形的未粘连的颗粒,有些颗粒的表面并不平整。研究结果表明一定葡萄糖值的低粘度SSOS作为微胶囊壁材具有很好的包埋效果。  相似文献

The effects of day-length and temperature on flowering and dormancyinduction were studied in Anemone coronaria L., with plantsraised either from corms or achenes. An Israeli hybrid sourcewas used (de Caen cv. Hollandia x Israeli wild type). Dormancy onset is characterized by the cessation of foliageleaf production, the appearance of leaf scales protecting theperennating bud, and leaf senescence. Dormancy was induced byhigh temperature and long days but increasing temperatures (from17/12 °C to 32/12 °C) induced earlier dormancy thanprolonging the photoperiod (range 8–16 h). A significant(P = 0.01) interaction was found between these factors, withsmaller photoperiodic effects the higher the temperature. At22/17 °C the critical day-length for dormancy inductionwas between 11 and 12 h. The transition from the vegetative to the reproductive stageappears to be an autonomous process that occurs with developmentin plants raised from either corms or achenes and does not requireenvironmental induction. Photo- and thermoperiodic effects onflowering were indirect, being mediated through their influenceon dormancy induction. Relative electrical conductivity (RC) values and Tally acid levels were measured on apple leaves of different ages exposed to 0 and 20°C. RC values were measured at—3°C and high RC values indicate frost-sensitive tissue. A prolonged period at 0°C gave an increased RC value of the leaves, which indicates damage. At 20°C the RC values were lower in older leaves than in young leaves. The fatty acids level as well as the degree of saturation were different at different ages of the leaves. Young leaves showed a higher fatty acid level in plants held at 20°C than in plants at 0°C. The older leaves maintained the same level after 12 days at 20°C as after 3 days at 20°C. The fatty acid level decreased at 0°C. The linolenic acid level followed the same trend as total fatty acids, indicating that synthesis and degradation of linolenic acid can occur in the same plant depending on the age of the leaf and on the temperature. Cold resistance and linolenic acid levels were correlated in both old and young leaves at 20°C and in older leaves at 0°C. There was no correlation between cold resistance and levels of linotenic acid levels in young leaves at 0°C. Two hiosynthetic pathways for linolenic acid synthesis are discussed.  相似文献

The response of whole leaf photosynthesis of wheat (Triticum aestivum L.) in relation to soluble CO2 available to the mesophyll cells, under low (1.5%) O2 at 25, 30, and 35 C, followed Michaelis-Menten kinetics up to saturating CO2 but deviated at high CO2 levels where the experimental Vmax is considerably less than the calculated Vmax. The affinity of the leaves for CO2 during photosynthesis was similar from 25 to 35 C with Km (CO2) values of approximately 3.5 to 5 μM.  相似文献

探讨高乳清蛋白的肠内营养对大鼠创伤皮肤的促愈合作用.将36只SD大鼠随机分为3组,每组12只,即高乳清蛋白肠内营养组(A)、普通肠内营养组(B)、空白对照组(C),建立大鼠皮肤创伤模型后进行肠内营养支持,肠内营养用量为每天731.5kJ·kg-1,以灌胃的方式提供,分别于术后第7、14天测定血清蛋白、血红蛋白含量并计算创面愈合率.结果显示,术后第7、14天A组的血清蛋白、血红蛋白及伤口皮肤平均愈合率显著高于B、C组,差异具有统计学意义(p<0.05).表明高乳清蛋白肠内营养具有较好的促进伤口愈合的作用.  相似文献

Response surface methodology was applied to determine the effects of pullulanase debranching, microwave irradiation time (2–4 min) and power (20–100%) on resistant starch (RS) formation and in-vitro glycemic index (GI) values in high amylose corn starch, Hylon VII. Starch:water (1:10) suspensions were cooked and autoclaved, debranched with pullulanase (1000 PUN/g; 1500 U/kg starch) at 60 °C and then different microwave-storing cycles and drying (oven or freeze drying) processes were applied. In order to describe the relationship between the dependent and independent variables (microwave power and irradiation time), the response values were fitted by first order polynomial regression models. Significance analysis showed that microwave irradiation time had significant effect on RS content and GI value of the samples treated with one cycle of microwave-storing prior to freeze-drying. Microwave power had significant factor on the GI value of the samples that were oven-dried after one cycle of microwave-storing. Solubility and water binding capacity values of all heat treated samples were higher than those of native starch. On the other hand, RVA viscosity values were lower than native starch for oven-dried samples. Water binding capacity, solubility and final viscosity values of the freeze-dried samples were higher than those of oven-dried ones.  相似文献

Measurements were made of the transport of ions through excisedbarley roots. It is shown that addition of 10–5 M abscisicacid (ABA) may lead to inhibition or stimulation of transport.The effect of ABA depended on the conditions in which the plantshad been grown and on the temperature at which the experimentwas carried out. It is suggested that stimulation of transportis a direct effect of ABA, but inhibition may be an indirecteffect due to disturbance of endogenous levels of plant hormones.The results are considered in relation to regulation of iontransport in the plant as a whole.  相似文献
