The effect of chronic phostoxin administration on some tissue ATPases, hematology and tissue histopathology was investigated using a combination of gravimetric, enzymatic, colorimetric and histological procedures in New Zealand White rabbits after 2 weeks administration of 0.8mg phostoxin/kg body weight/day, po. The phostoxin treatment led to significant decreases in Na(+)-K+ ATPase activities in renal, hepatic and cardiac tissues. Similar decreases were obtained in the activities of Ca(2+)-ATPase and Mg(2+)-ATPase in liver. In addition, the phostoxin-toxified rabbits manifested significant decreases in hematocrit, red blood cell count, hemoglobin and platelets. Histological examination of the tissues revealed pronounced degenerative changes in liver, heart and kidney.  相似文献

Five erythrocyte proteins (Adenosine deaminase, Ada; 6-Phosphogluconate dehydrogenase, Pgd; Esterase 1, Es-1; Esterase 3, Es-3; NADH-Diaphorase 2, Dia-2) and a serum beta-globulin protein (Esterase 7, Est-7) were studied in rabbits using starch gel electrophoresis. Samples were obtained from 317 Spanish Common individuals (38 families, 317 individuals). For the analyses of linkage, Morton's sequential probability ratio test was applied. Clear evidence for linkage between Es-1 and Est-7 (theta 0.2) was obtained, and no evidence of linkage was obtained for the remaining pairwise combinations of loci studied.  相似文献

In prior work we demonstrated the implementation of logic gates, sequential computers (universal Turing machines), and parallel computers by means of the kinetics of chemical reaction mechanisms. In the present article we develop this subject further by first investigating the computational properties of several enzymatic (single and multiple) reaction mechanisms: we show their steady states are analogous to either Boolean or fuzzy logic gates. Nearly perfect digital function is obtained only in the regime in which the enzymes are saturated with their substrates. With these enzymatic gates, we construct combinational chemical networks that execute a given truth-table. The dynamic range of a network's output is strongly affected by "input/output matching" conditions among the internal gate elements. We find a simple mechanism, similar to the interconversion of fructose-6-phosphate between its two bisphosphate forms (fructose-1,6-bisphosphate and fructose-2,6-bisphosphate), that functions analogously to an AND gate. When the simple model is supplanted with one in which the enzyme rate laws are derived from experimental data, the steady state of the mechanism functions as an asymmetric fuzzy aggregation operator with properties akin to a fuzzy AND gate. The qualitative behavior of the mechanism does not change when situated within a large model of glycolysis/gluconeogenesis and the TCA cycle. The mechanism, in this case, switches the pathway's mode from glycolysis to gluconeogenesis in response to chemical signals of low blood glucose (cAMP) and abundant fuel for the TCA cycle (acetyl coenzyme A).  相似文献

We studied the vertical gradient in lung expansion in rabbits in the prone and supine body positions. Postmortem, we used videomicroscopy to measure the size of surface alveoli through transparent parietal pleural windows at dependent and nondependent sites separated in height by 2-3 cm at functional residual capacity (FRC). We compared the alveolar size measured in situ with that measured in the isolated lungs at different deflationary transpulmonary pressures to obtain transpulmonary pressure (pleural surface pressure) in situ. The vertical gradient in transpulmonary pressure averaged 0.48 +/- 0.16 (SD) cmH2O/cm height (n = 10) in the supine position and 0.022 +/- 0.014 (SD) cmH2O/cm (n = 5) in the prone position. In mechanically ventilated rabbits, we used the rib capsule technique to measure pleural liquid pressure at different heights of the chest in prone and supine positions. At FRC, the vertical gradient in pleural liquid pressure averaged 0.63 cmH2O/cm in the supine position and 0.091 cmH2O/cm in the prone position. The vertical gradients in pleural liquid pressure were all less than the hydrostatic value (1 cmH2O/cm), which indicates that pleural liquid is not generally in hydrostatic equilibrium. Both pleural surface pressure and pleural liquid pressure measurements show a greater vertical gradient in the supine than in the prone position. This suggests a close relationship between pleural surface pressure and pleural liquid pressure. Previous results in the dog and pony showed relatively high vertical gradients in the supine position and relatively small gradients in the prone position. This behavior is similar to the present results in rabbits. Thus the vertical gradient is independent of animal size and might be related to chest shape and weight of heart and abdominal contents.  相似文献

Serum triiodothyronine (T3) and thyroxine (T4) concentrations inMacaca fascicularis were determined at 4-hr interval for 24 hr using commercial radioimmunoassay kits. It was found that the highest concentrations of T3 and T4 in the animals were in the serum sample taken at 10:00. Variations in plasma thyroxine (T4) and triiodothyronine (T3) contents of pike, pikeperch, perch, bream and chub from a large reservoir (Pareloup, South of France) were investigated before and during the progressive emptying of the reservoir. Fish were caught by gill nets, except at the last time of emptying when controls were collected with landing nets. Both iodothyronine levels showed desynchronized seasonal cycles resulting in higher T3/T4 ratios in fish stressed by gill-netting in early summer. The aforementioned results principally emphasized the prevailing influence of the technique of catching on eliciting stress responses and subsequent thyroidal impairment in all five species. However, the much higher amounts of T4 and/or T3 observed in specimens caught in the later stages of draining gave evidence to suggest that the progressive alteration in habitat might also have indirectly affected the thyroidal status of these fish, this latter result leading to the consideration of fish plasma thyroid hormones as possible endocrine biomarkers of chronic perturbation.  相似文献

Two rabbits (RG-1, RG-2) were immunized with rabbit thyroglobulin (RTg) purified from thyroid glands of four other normal rabbits of the same strain, and bled serially. Antisera were obtained at different times after the first immunization and kept separately and studied. Production of anti-RTg as well as anti-thyroid hormone antibodies such as anti-thyroxine (T4) and anti-triiodothyronine (T3) antibodies was observed in both rabbits. Physicochemical parameters of anti-RTg antibodies with RTg, T4, and T3 were calculated in two selected antisera (70-day and 253-day) of each of the rabbits, using a Scatchard plot. Extraction of serial sera from both rabbits disclosed the presence of larger amounts of T3 and T4 in immune sera than in preimmune serum. Examination of pathology of thyroid glands and kidneys in both rabbits was negative for the lesions of autoimmune thyroiditis and immune nephritis. These results indicate that anti-Tg as well as anti-thyroid hormone autoantibodies can be raised without thyroid pathology in rabbit by immunization with autologous Tg.  相似文献

Purpose

The article aims to test indicators for assessing the environmental and social impacts of biorefineries. Testing environmental and social impact categories and indicators, and selecting the most suitable ones, will simultaneously contribute to the further development of social life cycle assessment (S-LCA) methodologies while assessing several dimensions of sustainability at biorefineries.

Methods

The work applies two methodologies, environmental LCA (E-LCA) and social LCA (S-LCA), to two hypothetical production processes of second-generation bioethanol and biochemical in two alternative locations (Norway and the USA). Five impact categories were chosen for the E-LCA. The S-LCA was performed in two stages: a generic assessment (top-down approach) using the social hotspot database (SHDB 2013) to screen for potential social issues in the stakeholder group Worker in Norway and the USA and a specific assessment (bottom-up approach) for collecting data and confirming or refuting the SHDB results in the Norwegian case only.

Results and discussion

Bioethanol produced in the Norwegian biorefinery would perform relatively well in relation to climate change targets, with emissions of approximately 11 g CO2-eq/MJ. The same production process located in the USA would produce emissions of approximately 29 g CO2-eq/MJ. Other biorefinery products are difficult to compare because of a lack of clear alternatives. Bioethanol and biochemicals produced in the hypothetical USA production process have higher burdens than those from the Norwegian production process in all environmental categories assessed. For both production processes, the main social risks were in the category Health and safety followed by Labor rights and decent work. More detailed investigations in an existing Norwegian biorefinery value chain confirmed some of the risk issues but discarded others, demonstrating the necessity of providing specific data and results for the social dimension.

Conclusions

E-LCA and S-LCA make it possible to highlight the main environmental and social challenges when producing biochemicals. The SHDB has potential as a social screening tool although social indicators are not yet well established. Hence, specific assessment is necessary for validating the results in the social dimension. S-LCA is still in its infancy and needs to be applied in order to develop the best practice. The two methodologies addressed bioethanol and biochemical production performance in two different dimensions (environmental and social), and their combination makes it possible to achieve results that integrate the product-oriented approach with the more location-specific approach.

Peripheral venous blood samples were drawn before and after induction of adjuvant arthritis in rabbits at intervals of 15 days. Using these samples, biochemical parameters were measured, including total protein, albumin, globulin, cholesterol, alkaline phosphatase, glutamic oxaloacetic transaminase and glutamic pyruvic transaminase. The various biochemical tests exhibited an abnormal pattern in adjuvant animals as compared to the normal rabbits. The significance of these findings has been discussed.  相似文献
