Five tropical epiphytes were examined for evidence of CrassulaceanAcid Metabolism (CAM), namely the orchids Eria velutina Lindl.,Dendrobium tortile Lindl. and Dendrobium crumenatum Sw., andthe ferns Pyrrosia adnascens (Forst.) Ching and Pyrrosia angustata(Sw.) Ching, family Polypodiaceae. Diurnal variations in leaftitratable acidity, diffusive conductance and water potentialwere measured at various levels of water stress. The three orchidsshowed typical CAM behaviour, namely large diurnal fluctuationsin leaf acidity, day-time closure and night opening of stomataand a very slow decline in water potential under stress. Theferns showed some evidence of CAM, but this was not as well-developedas had been reported for two other tropical epiphytic membersof the same family. Relative water content R, water potential and leaf diffusiveconductance C5 were monitored while five tropical epiphyteswere subjected to an extended period of drought. The speciesstudied were: ferns Pyrrosia adnascens (Forst.) Ching and Pyrrosiaangustata (Sw.) Ching, family Polypodiaceae; orchids: Eria velutinaLindl., Dendrobium tortile Lindl. and Dendrobium crumenatumSw. The ferns reached zero turgor rapidly and after only small declinesin relative water content (R) and . Beyond this point stomatalmovement seemed strongly suppressed, but the leaves continuedto lose water vapour until very low values of R were reached.Nevertheless, on re-watering water potential (), R and diffusiveconductance (C5) returned to pre-stress levels within 3 d. The orchids showed a more gradual decline in R and , stomatalactivity was not so strongly suppressed, and night opening ofstomata was observed under stress. The relationship between and R was found for each species,curves being fitted to the data points by non-linear regression.From these analyses it was concluded that these species hadvery dilute cell sap, and consequently the change in for agiven decline in R was smaller than for most other species recordedin the literature. Osmotic Adjustment and Stomatal Response to Water Deficits in Maize   总被引:1,自引:1,他引:0  
A pot experiment was carried out using five maize {Zea maysL.) cultivars under three soil moisture levels (MPa 0 to –0.05,–0.3 to –0.9 and –1.2 to –1.5) to investigatethe effects of water deficits on osmotic adjustment and stomatalconductance. The degree of leaf rolling and the sugar and nutrientconcentrations in leaf cell sap were measured. Leaf water potential and osmotic potential decreased and stomatalconductance decreased with increasing water deficits. Stomatalconductance correlated positively with leaf water potentialand osmotic potential. Degree of leaf rolling was lower in cultivarswhich maintained higher turgor. Osmotic adjustment of 0.08 to0.43 MPa was found under the lowest soil moisture level in fivecultivars used. Sugar and K were the major osmotic substancesin the maize plant. Sugar, K and Mg concentrations increasedunder water deficit, and correlated negatively with a decreasein osmotic potential. Shoot Resistance to Water Flow in Cotton   总被引:2,自引:0,他引:2  
Studies using excised cotton (Gossypium hirsutum L.) plants,attached to a free water source and undergoing transpirationcycles, were conducted at intervals over a 2 year period inorder to quantify shoot resistance components of cotton canopies.Leaf water potential was found to be a linear function of transpirationrate at rates above 0.1 mm h–1, so shoot resistance wasevaluated as the slope of this function. The value of 4.8 104h (0.48 MPa h mm–1) total shoot resistance was consistentfor 1.10 m tall, well irrigated, fruit-bearing cotton plants.Further tests, with pre-wrapped and exposed leaves, revealedthat total shoot resistance was comprised of an axial component(40%) and a leaf component (60%). The total shoot resistanceof 0.48 MPa h mm–1 is likely to be relevant for modellingcotton water relations when LWP is evaluated on exposed, topof the canopy leaves, such as in the ‘big leaf’type models. Controlled environment experiments were performed to determinethe effects of temperature and water potential on germination,radicle elongation and emergence of mungbean (Vigna radiata(L.) Wilczek cv. IPB-M79-17-79). The effects of a range of constant temperatures (15–45°C) and water potentials (0 to –2.2 MPa) on germinationand radicle elongation rates were studied using an osmoticumtechnique, in which seeds were held against a semi-permeablemembrane sac containing a polyethylene glycol solution. Linearrelationships were established between median germination time(Gt50) and water potential at different temperatures, and betweenreciprocal Gt50 (germination rate) and temperature at differentwater potentials. Germination occurred at potentials as lowas –2.2 MPa at favourable temperatures (30–40 °C),but was fastest at 40 °C when water was not limiting, withan estimated base temperature (Tb) of about 10 °C. Subsequentradicle elongation, however, was restricted to a slightly narrowertemperature range and was fastest at 35 °C. The conceptof thermal time was used to develop an equation to model thecombined effects of water potential and temperature on germination.Predictions made using this model were compared with the actualgermination obtained in a related series of experiments in columnsof soil. Some differences observed suggested the additionalimportance of the seed/soil/water contact zone in influencingseed germination in soil. Seedling emergence appeared to reflectfurther the radicle elongation results by occurring within anarrower range of temperatures and water potentials than germination.Emergence had an estimated Tb of 12.6 °C and was fastestat 35 °C. In this study, a matric-potential control system was used todetermine the effect of matric-priming duration and primingwater potential on the germination response of Bouteloua curtipendula(Michx.) Torr., Cenchrus ciliaris L., Eragrostis lehmannianaNees, and Panicum coloratum L. Seeds were primed at water potentialsof –1·5 to –7·7 MPa for up to 14 d.Optimum germination generally occurred in treatments primedat high water potential for the shortest period. Germinationof seeds primed at lower water potential and for longer periodsexhibited a negative germination response relative to the control.Seeds were not redried after the priming treatment. The epidermal structure of the five species of ferns, Arthromeriswallichiana (Spr.) Ching., Drymoglossum piloselloides (Prest.),Drynaria quercifolia (L.) J. Smith, Lepisorus nudus (Hook.)Ching. and Pyrrosia nuda (Gies.) Ching., has been investigated.Fifteen types of stomatal structures have been identified ofwhich copolo-desmocytic and coperi-desmocytic are new types.Four more possible stomatal structures: ccpolo-peri-, codesmo-polo-,codesmo-peri- and duplodesmocytic, are suggested. Localizationof starch, insoluble polysaccharides, protein and lipids hasbeen examined histochemically in the guard cells, subsidiarycells and epidermal cells. In Drynaria starch plastids and plastidscontaining both starch and protein are present in guard cells.Starch plastids are present in the subsidiary cells of all speciesexcept in Arthromeris, whereas, they are present in epidermalcells of only Drymoglossum and Lepisorus. Granular or amorphousinsoluble polysaccharides (other than starch) are present inguard cells of all the species, in the subsidiary cells of Arthromeris,Drynaria and Pyrrosia, and in the epidermal cells of Pyrrosia.Except in Pyrrosia lipids are present in the guard cells. Subsidiarycells of Drynaria and the epidermal cells of Arthromeris andDrynaria show lipid bodies. The presence of plasmodesmata andectodesmata is demonstrated in the epidermal cells of Drymoglossum.  相似文献

A comparison of plant hydraulic conductances in wheat and lupins   总被引:3,自引:1,他引:2  
Previous studies have shown similar water use for lupins (Lupinusangustifolius L.) and wheat (Triticum aestivum L.), despitea considerably smaller root system in lupins. A field studyand an experiment under controlled conditions using pressure-fluxrelationships were conducted to examine whether higher hydraulicconductances were responsible for the greater water uptake perunit root length in lupins. In the field experiment, the fluxof water and differences in water potential through the soil-plantsystem were measured for both species and used to calculatethe hydraulic conductance through the plant and through theroot and shoot. The hydraulic conductance for the whole plantwas 3–5 times greater in lupins than in wheat. This relativedifference between the species was similar when plant hydraulicconductance was expressed per unit of root length. This occurreddespite the difference in midday water potential between soiland leaves, being consistently greater in wheat (–1.0MPa) than in lupins (–0.7 MPa). When the total plant conductancewas separated into its components, the combined soil and rootconductance and the shoot conductance were 2 and 6 times greater,respectively, in lupins than in wheat. In the experiment undercontrolled conditions, hydraulic conductance for the entireroot system was determined using a pressure chamber. The specificroot hydraulic conductances were 4 times greater in lupins thanin wheat. The results from both field and controlled conditionsexperiments suggest that the greater water uptake per unit rootlength in lupins compared to wheat results from appreciablylarger root and shoot hydraulic conductances. Responses of Vegetable Seeds to Controlled Hydration   总被引:1,自引:1,他引:0  
Leek, onion and carrot seeds were imbibed in water and in solutionsof polyethylene glycol (PEG) 6000 over the range –0.5to –4.0 MPa osmotic potential, for periods up to 28 dat 15 C. Seeds started to germinate after 7 and 14 d at –0.5MPa and –1.0 MPa PEG, respectively, but in the lattercase, germination did not exceed 5%. No germination occurredin solutions of lower (more negative) osmotic potential. Seedmoisture content increased with osmotic potential in all threespecies and the relationships were unaffected by the durationof imbibition in solutions of –1.0 to –4.0 MPa,though leek seeds had higher moisture contents than the otherspecies for any given osmotic potential. Linear relationships between response to priming (differencebetween mean germination times of primed and untreated seeds)and seed moisture content were obtained for each species, positiveresponses being obtained above 30–35% seed moisture contentwith optima at 46, 44.5 and 44% seed moisture contents in leek,onion and carrot, respectively. Priming had no effect on embryovolume or cell number per embryo in leek and onion. Carrot embryovolume increased by 43% and cell number per embryo doubled inprimed compared with untreated seeds, whereas seeds imbibedin water showed only a slight increase in cell number per embryoat the stage when radicles were beginning to penetrate the seedcoat. Salicylic Acid Levels in Thermogenic and Non-Thermogenic Plants   总被引:7,自引:0,他引:7  
The natural trigger for heat production in the thermogenic inflorescencesof Sauromatum guttatum Schott (voodoo lily) was recently identifiedas salicylic acid (SA), which induced heat production at levelsas low as 13 ng g f. wt–1. Since then the levels of SAwere determined in other thermogenic and non-thermogenic plantspecies. In thermogenic inflorescences of five aroid species,and in male cones of at least four thermogenic cycads SA levelsduring heat production exceeded 1 µg g f. wt–1.SA was not detected in the thermogenic flowers of a water lily,Victoria regia Lindl. (Nymphaeaceae), and Bactris major Jacq.(Palmae). Levels of salicylic acid varied substantially in thefloral parts of seven non-thermogenic species and in the leavesof 27 non-thermogenic species. The vulnerability of xylem vessels to water stress-induced cavitationwas studied by measuring hydraulic conductivity and ultrasoundacoustic emissions (AEs) in Fagus sylvatica L. f. purpurea (Ait.)Schneid. and Populus balsamifera L. The occurrence of xylemembolism in summer was investigated in relation to leaf waterpotential and stomatal conductance. Populus was extremely vulnerableto cavitation, losing functional vessels due to embolism atwater potentials lower than –0.7 MPa. Fagus experiencedembolism when water potential fell below –1.9 MPa. Middaywater potentials often approached these threshold values. Whenevaporative demand increased rapidly on sunny days, water lossbecame limited by low stomatal conductance. Thus water potentialsfell only slightly below the threshold values inducing cavitation.Despite the differences in vulnerability, both species tolerateda similar embolism rate of about 10% in the summer. There wasno embolism reversal during prolonged periods of rain. AEs werepredictive of loss in hydraulic conductivity, indicating thatAEs were mainly confined to vessels. Finally, vessel lengthdistribution, vessel diameter (tangential axis), vessel density,and vessel wall thickness had been determined for both speciesinvestigated. Irradiation of spores of the fern Mohria caffrorum Sw. witheither red light (67.4 µW cm–2) or far-red light(63.2 µW cm–2) for a period of 24 h induced maximumlevels of germination. Brief irradiations with blue light (127.6µW cm–2) administered before or after photoinductioncompletely nullified the effects of red or far-red light; however,with prolonged exposure to blue light, germination levels roseto near maximum. The similar effects of red and far-red lightin promoting spore germination makes the involvement of phytochromein this process questionable. Based on energy requirements,the promotive and inhibitory phases of blue light appear toinvolve independent modes of action. The effects of root age, temperature, and soil water statuson root hydraulic conductivity (LP) were investigated for twocactus species, Ferocactus acanthodes and Opuntia ficus-indica.The volumetric flux density of water was measured for excisedroot segments, either using negative hydrostatic pressures appliedto the proximal end or using reverse flow of water from theroot to the soil. For both species, LP at 20 ?C increased withroot age, average values reaching a maximum of 3.9 ? 10–7m s–1 MPa–1 for F. acanthodes and 5.2 ? 10–7m s–1 MPa–1 for O.ficus-indica at 11 to 17 weeksof age; LP subsequently declined with increasing root age forboth species. LP was maximal at a temperature of about 10 ?Cfor the youngest roots (1–3 weeks), this optimum shiftingto 40 ?C for 8-week-old roots of both species. For older roots(up to 1.5-years-old), LP increased with temperature from 0?C to 50 ?C, with a Q10 of 1.3 between 20 ?C and 30 ?C. At asoil water potential (soil) of –0.016 MPa, root LP wasindependent of the direction of water flow for both species.Depending on root age, LP declined 45- to 500-fold for F. acanthodesand 90- to 800-fold for O.ficus-indica as soil was reduced from–0.016 to –1.06 MPa, consistent with a rectifier-likebehaviour with respect to water movement between soil and roots.Incorporation of such responses into water uptake models shouldlead to a better understanding of root function. Relative water content (RWC) and water potential as measuredwith the pressure chamber were evaluated as indicators of waterstatus of tissue-cultured apple shoots and plantlets (shootswith roots). During the hydration required for RWC measurement,both water content and water potential exhibited the same hydrationkinetics, indicating that 10 h were required for full hydration.Once full hydration was reached, shoot mass remained relativelyconstant. Moisture release characteristics were also constructedand the associated shoot and plantlet water relations parameterswere estimated. Underin vitroconditions, both shoot and plantletwater potential were similar to the water potential of the culturemedium in which they were grown. The moisture release characteristicof shoots and plantlets was consistent with that expected fortypical plant tissues, and gave estimates of maximum modulusof elasticity (6.201.14 MPa), osmotic potential at saturation(–0.85 0.10 MPa), osmotic potential at zero turgor (–1.16 0.14 MPa) and RWC at zero turgor (78 2%) which were similarto values in the literature. Higher values of leaf conductanceand RWC were found in shoots and plantlets placed at 95% RH(21 C) compared to those at 90% RH. Plantlets had higher valuesof both conductance and RWC compared to shoots, suggesting thatinvitroroots are functional in water uptake. Relative water contentwas related to measures of physiological activity such as leafconductance, and it was also easier to measure than water potential.Relative water content is suggested as a sound index of waterstatus in tissue culture plants. The charophyte Lamprothamnium papulosum (Wallr.) J. Gr. is foundat salinities varying from nearly fresh water to twice thatof sea water. It can maintain its turgor constant at 302 mosmolkg–1 (0.73 MPa) when exposed to external osmotic pressuresof 550 to 1350 mosmol kg–1 (1.3–3.3 MPa). Turgorshows a tendency to rise slightly at lower osmotic pressure(388 mosmol kg–1 of turgor at 150 mosmol kg–1 externalosmolality). K+ and Cl are the main solutes in the vacuole,and are most important in controlling internal osmotic pressure.Mg2+, Ca2+, and SO2–4 are present in significant amountsbut their concentrations do not change with changes in externalsalinity. Na+ is present in lower concentration than K+, andplays a minor role in regulating turgor. Sucrose is presentin significant concentrations, but changes little with changesin salinity. Two enzymes involved in sucrose metabolism, sucrosephosphate synthetase (EC 2.4.1.14 [EC] ), and sucrose synthetase (EC2.4.1.13 [EC] ) are active in whole cell extracts of Lamprothamnium.As in the fresh water charophytes, Lamprothamnium membrane potentialmay be depolarized (close to EK) or hyperpolarized, and presumablyof electrogenic origin. The leaf to air water vapour deficit was increased suddenlyfrom about 1·0 to 2·5 IcPa for single leaves ofsoybean (Glycine max L. Merr.) plants held at 30 °C, 2·0mmol m –2 s–1 photosynthetic photon flux density(PPFD) and carbon dioxide pressures saturating to photosynthesis.After a lag of about 10 min, photosynthetic rate and stomatalconductance to water vapour began to decrease, and then cycledin phase with each other. The period of the cydes was about20 min. During these cycles the substomatal carbon dioxide pressurewas constant in the majority of leaves examined, and was alwaysabove saturation for photosynthesis. Epidermal impressions showedthat most stomata changed in aperture during the cycles, andthat very few were ever fully closed. Water potential measuredon excised discs changed by at most 0·1 MPa from theminima to the maxima in transpiration rate. In contrast, forleaves of sunflower (Helianthus animus L.) grown at low PPFD,the increase in VPD led to leaf wilting and decreased photosynthesis,followed by recovery of turgor and photosynthesis as stomatalconductance began to decrease. In these leaves photosynthesisand conductance then cycled approximately 180° out of phase.It is suggested that in soybeans decreased leaf conductanceinduced by high VPD provided a signal which decreased the rateof photosynthesis at carbon dioxide saturation by a mechanismthat was not related to a water deficit in the mesophyll. The effects of storage conditions on the germination of developingmuskmelon (Cucumis melo L.) seeds were tested to determine whetherafter-ripening is required to obtain maximum seed vigour. Seedswere harvested at 5 d intervals from 35 (immature) to 60 (fullymature) days after anthesis (DAA), washed, dried, and storedat water contents of 3·3 to 19% (dry weight basis) at6, 20, or 30°C for up to one year. Germination was testedin water and in polyethylene glycol 8000 solutions ( –0·2to –1·2 MPa osmotic potential) at 15, 20, 25 or30°C. Germination percentages and rates (inverse of meantimes to radicle emergence) were compared to those of newlyharvested, washed and dried seeds. For 40 and 60 DAA seeds,one year of storage at 20°C and water contents <6·5%significantly increased germination percentages and rates at20°C, but had little effect on germination at 25 and 30°C.Storage reduced the estimated base temperature (Tb) and meanbase water potential (b) for germination of both 40 and 60 DAAseeds by approximately 5°C and 0·3 MPa, respectively.Immature 35 DAA seeds showed the greatest benefit from storageat 3 to 5% water content and 30°C, as germination percentagesand rates increased at all water potentials (). Storage underthese same conditions had little effect on the germination ofmature seeds in water, but increased germination percentagesand rates at reduced 's. Accelerated ageing for one month at30°C and water contents from 15 to 19° increased germinationrates and percentages of mature seeds at reduced 's, but longerdurations resulted in sharp declines in both parameters. Immatureseeds lost viability within one month under accelerated ageingconditions. An after-ripening period is required at all stagesof muskmelon seed development to expand the temperature andwater potential ranges allowing germination and to achieve maximumgerminability and vigour. Post-harvest dormancy is deepest atthe point of maximum seed dry weight accumulation and declinesthereafter, both in situ within the ripening fruit and duringdry storage. Two succulent perennials from the Sonoran Desert, Agave desertiEngelm. and Ferocactus acanthodes (Lem.) Britton and Rose, loselittle water through their roots during drought, yet respondrapidly to light rainfall. Their roots tend to be shallow, althoughabsent from the upper 20 mm or so of the soil. During 12–15d after a rainfall, new root production increased total rootlength by 47 per cent to 740 m for A. deserti and by 27 percent to 230 m for F. acanthodes; root dry weight then averagedonly 15 per cent of shoot dry weight. The annual carbon allocatedto dry weight of new roots required 11 per cent of shoot carbondioxide uptake for A. deserti and 19 per cent for F. acanthodes.Elongation of new roots was greatest near a soil temperatureof 30°C, and lethal temperature extremes (causing a 50 percent decrease in root parenchyma cells taking up stain) were56°C and -7°C. Soil temperatures annually exceeded themeasured tolerance to high temperature at depths less than 20mm, probably explaining the lack of roots in this zone. Attached roots immersed in solutions with osmotic potentialsabove -2·6 MPa could produce new lateral roots, with50 per cent of maximum elongation occurring near -1·4MPa for both species. Non-droughted roots lost water when immersedin solutions with osmotic potentials below -0·8 MPa,and root hydraulic conductance decreased markedly below about-1·2 MPa. Pressure-volume curves indicated that, fora given change in water potential, non-droughted roots lostthree to five times more water than droughted roots, non-droughtedleaves, or non-droughted stems. Hence, such roots, which couldbe produced in response to a rainfall, will lose the most tissuewater with the onset of drought, the resulting shrinkage beingaccompanied by reduced root hydraulic conductance, less contactwith drying soil, and less water loss from the plant to thesoil. Phosphorus and nitrogen uptake capacities were assessed during36–58 d drying cycles to determine whether the abilityof sagebrush (Artemisia tridentata Nutt.) to absorb these nutrientschanged as the roots were subjected to increasing levels ofwater stress. Water was withheld from mature plants in large(6 I) containers and the uptake capacity of excised roots insolution was determined as soil water potentials decreased from–0.03 MPa to –5.0 MPa. Phosphorus uptake rates of excised roots at given substrateconcentrations increased as preharvest soil water potentialsdecreased to –5.0 MPa. Vmax and Km also increased as soilwater potentials declined. Declining soil water potentials depressednitrogen uptake at set substrate concentrations, but uptakecapacity, calculated as the sum Vmax for both NH+4+NO3,did not change significantly with drying. The sum Vmax correlatedwith root nitrogen concentration. Root uptake capacity for nitrogen and phosphorus was extremelystable under severe water stress in this aridland shrub. Maintenanceof uptake capacity, coupled with a previously demonstrated abilityto conduct hydraulic lift, may enable A. tridentata better tomaintain nitrogen and phosphorus uptake as soil water availabilitydeclines. These mechanisms may be important in the ability ofA. tridentata to maintain growth, complete reproduction, andgain an advantage against competitors late in the season whenthe soil layers with higher nutrient availability are dry.
