Different ATPases may control the various functional changes that spermatozoa undergo just prior to fertilization, with the enzyme's specific location within the cell reflecting its function. The activities of Mg2+-ATPase, Ca2+Mg2+-ATPase, Na+K+-ATPase and Ca2+-ATPase were determined for head plasma membranes (HPM) and sperm body membrane (SBM) from both fresh (n = 4) and cryopreserved bovine spermatozoa (n = 4) and fresh homogenized whole spermatozoa (HWS) (n = 6). No activity of Ca2+Mg2+-ATPase was found in any preparation from spermatozoa. Ca2+-ATPase was detected in fresh SBM and HWS but not in HPM. Activity of Mg2+-ATPase and Na+K+-ATPase was higher in HPM than HWS or SBM (P < 0.01). Cryopeserving the whole sperm reduced the activities of all three enzymes, but Na+K+-ATPase was more sensitive to cryopreservation than Mg2+-ATPase (P ≤ 0.05). Enzyme location suggests that Ca2+-ATPase may be associated with events in the flagellum, while Mg2+-ATPase and Na+K+-ATPase may affect functions in the sperm head. Cryopreservation-induced damage to ATPases might be involved in reducing the fertilizing ability of cryopreserved spermatozoa.  相似文献

In reconstituted human red blood cells a difference was found in (Ca2+ + Mg2+)-ATPase activity and in Ca2+ efflux at 37°C, depending on the side of the membrane at which the monovalent cations K+ and Na+ were placed. Under the conditions used, (Ca2+ + Mg2+)-ATPase activity and Ca2+ efflux was highest when K+ (35 ± 0.5 mM (± S.E.), mean of four experiments) was at the inside and Na+ (130 mM) at the outside of the ghost membrane.  相似文献

Tissue-specific age-dependent changes were observed in Na+K+-, Ca2+-, and Mg2+-ATPase activities in tropical tasar silkworm, Antheraea mylitta Drury. Maximum enzyme activity was recorded in all the tissues on day 12 (before spinning) in control group of animals. In testis, Na+K+-, Ca2+-, and Mg2+-ATPase activities gradually increased from day 2 to day 12 during fifth larval age and level was maintained up to adult eclosion while, in ovary, a marked decline was noted up to day of adult emergence. Further, a significant and sharp rise was found in ATPase activity in silk gland tissue up to day 12 and afterwards a drastic fall was noted on day 15 (end of spinning) during fifth larval age.Administration of T4 to fifth stage larvae (1 hr old) at doses 0.5–2.0 μg/g significantly elevated the Na+K+-, Ca2+-, and Mg2+-ATPase activities in larval and pupal gonads in a dose-dependent fashion. But, in moths, the enhancement was very much confined to Na+K+- and Ca2+-ATPase in testes and only Ca2+-ATPase in ovaries. Again, in silk glands thyroxine (0.5–2.0 μg/g) caused a significant rise in the all ion-dependent ATPase activities only during the fifth larval stage. Interestingly, higher doses of T4 (4.0 μg/g) caused a significant reduction in Na+K+-, Ca2+- and Mg2+-ATPase in all the tissues almost all the days studied so far. However, lower doses of T4 (0.1 and 0.25 μg/g) remained ineffective in altering the different ion-specific ATPase activities. This study suggests, that mammalian thyroxine has a metabolic influence showing biphasic nature of action in tasar silkworm ATPase system.  相似文献

Antibodies raised in rabbits against the purified erythrocyte membrane Ca2+ pumping ATPase were affinity-purified using an ATPase-Sepharose column. Addition of a few molecules of the purified antibody per molecule of ATPase was sufficient to inhibit the ATPase activity. Extensively washed ghosts or preincubated pure ATPase sometimes develop an appreciable Mg2+-ATPase activity. In such cases, the antibodies inhibited the Mg2+-ATPase as well as the Ca2+-ATPase. This is consistent with the hypothesis that a portion of the Mg2+-ATPase activity of ghosts is derived from the Ca2+-ATPase. When nitrophenylphosphatase activity was observed, both Mg2+ - and Ca2+-stimulated activities were observed. Only the Ca2+ activity was inhibited by the antibodies, confirming that this activity is due to the Ca2+ pump, and suggesting that the Mg2+-nitrophenylphosphatase is due to a separate enzyme. Amounts of antibody comparable to those which inhibited the Ca2+-ATPases had no effect on the Na+-K+-ATPase; 4-fold higher amounts of antibody significantly stimulated the Na+-K+-ATPase, but this effect of the antibody was not specific: Immunoglobulins from the nonimmune serum also significantly stimulated the Na+-K+-ATPase.In resealed erythrocyte membranes, antibodies incorporated into the ghosts inactivated the Ca2+-ATPase, while antibodies added to the outside had no significant effect.  相似文献

Hepatic Na+-K+-ATPase and Mg2+-ATPase activities of male green lizards declined during the maturation phase (juvenile to 1-year-old) and stabilized thereafter. On the other hand, the Ca2+-ATPase activity of the liver declined during the later half of the life span (1-year-old to 2–4-year-old). Starvation stress induced a decline in hepatic Na+-K+-ATPase and Mg2+-ATPase activities of juvenile lizards and caused an increase in 1-year-old and 2–4-year-old counterparts. The Ca2+-ATPase activity declined only in starved 1-year-old lizards. Following cold stress, the hepatic Na+-K+-ATPase activity of juvenile lizards showed a higher degree of decline than 2–4-year-old counterparts. The Mg2+-ATPase activity declined in cold-stressed juvenile lizards, but the parameter was elevated in similarly treated 1-year-old lizards. On the other hand, the increase in Ca2+-ATPase activity in response to cold stress was confined only to 2–4-year-old lizards.  相似文献

Na+,K+-activated ATPase activity in tick salivary glands increases during the rapid stage of tick feeding paralleling similar increases in dopamine and cAMP-stimulated fluid secretion. High concentrations of cyclic AMP increase Na+,K+-ATPase activity in a plasma membrane-enriched fraction from the salivary glands of rapidly feeding ticks. Cyclic AMP-dependent protein kinase inhibitor protein blocks activation of Na+,K+-ATPase activity at low but not high concentrations of cAMP indicating that both activator and inhibitor modulator phosphoproteins of Na+,K+-ATPase activity exist in the plasma membrane-enriched fraction.ATPase activity in the plasma membrane-enriched fraction is not measurable in the absence of Mg2+, Ca2+ and Na+. Ca-stimulated nucleotidase activity is highest with ATP serving as the preferred substrate in a series including CTP, UTP, GTP and ADP. Calcium, Mg2+ stimulated ATPase activity is activated further by calmodulin and partially inhibited by low concentration of vanadate, trifluoperazine and oligomycin. Results suggest that the plasma membrane-enriched fraction of tick salivary glands contains both Ca2+-ATPase activity and oligomycin-sensitive Ca2+, Mg2+-ATPase activities, the latter likely from a small amount of mitochondria in the partially purified organelle fraction.  相似文献

This study examined the status of sarcolemmal Na+/K+-ATPase activity in rat heart under conditions of Ca2+-paradox to explore the existence of a relationship between changes in Na+/K+-pump function and myocardial Na+ as well as K+ content. One min of reperfusion with Ca2+ after 5 min of Ca2+-free perfusion reduced Na+/K+-ATPase activity in the isolated heart by 53% while Mg2+-ATPase, another sarcolemmal bound enzyme, retained 74% of its control activity. These changes in sarcolemmal ATPase activities were dependent on the duration and Ca2+ concentration of the initial perfusion and subsequent reperfusion periods; however, the Na+/K+-ATPase activity was consistently more depressed than Mg2+-ATPase activity under all conditions. The depression in both enzyme activities was associated with a reduction in Vmax without any changes in Km values. Low Na+ perfusion and hypothermia, which protect the isolated heart from the Ca2+-paradox, also prevented reperfusion-induced enzyme alterations. A significant relationship emerged upon comparison of the changes in myocardial Na+ and K+ content to Na+/K+-ATPase activity under identical conditions. At least 60% of the control enzyme activity was necessary to maintain normal cation gradients. Depression of the Na+/K+-ATPase activity by 60-65% resulted in a marked increase and decrease in intracellular Na+ and K+ content, respectively. These results suggest that changes in myocardial Na+ and K+ content during Ca2+-paradox are related to activity of the Na+/K+-pump; the impaired Na+/K+-ATPase activity may lead to augmentation of Ca2+-overload via an enhancement of the Na+/Ca2+-exchange system.  相似文献

Cadmium (≤ 50 μM) decreases the heat resistance (39°C) of the activity of frontal cilia in the Anodonta cygnea gills incubated in dechlorinated tap water, while in the presence of added 2 mM Ca2+ the minimal acting concentration of cadmium rises up to 100 μM.2. The inhibitory effect of Cd2+ (1.5 mM) on the ATPase activity measured in the gill microsomal fraction is temperature dependent and increases as follows: ouabain insensitive Na2+- or K+-ATPase (no inhibition), Ca2+-ATPase (50% inhibition), Mg2+-ATPase (100% inhibition).3. Cadmium itself (≤ 50 μM) added to microsomal suspension stimulates the H+-sensitive ATP hydrolysis resembling on its pH-dependence the Mg2+- but not Ca2+-ATPase activity.4. Cd2+ can mimic the effect of Mg2+ as a cofactor required for activation of the ouabain-insensitive Na+- or K+-ATPase. Monovalent cations fail to activate the ATPase when Mg2+ is substituted by Ca2+.5. One of the mechanisms underlying the toxicity of Cd2+ to Anodonta gills could be based upon an interaction of Cd2+ with Mg2+-ATPase followed by suppression of the ciliary activity.  相似文献

Experiments on the effects of varying concentrations of Ca2+ on the Mg2+ + Na+-dependent ATPase activity of a highly purified preparation of dog kidney (Na+ + K+)-ATPase showed that Ca2+ was a partial inhibitor of this activity. When Ca2+ was added to the reaction mixture instead of Mg2+, there was a ouabain-sensitive Ca2+ + Na+-dependent ATPase activity the maximal velocity of which was 30 to 50% of that of Mg2+ + Na+-dependent activity. The apparent affinities of the enzyme for Ca2+ and CaATP seemed to be higher than those for Mg2+ and MgATP. Addition of K+, along with Ca2+ and Na+, increased the maximal velocity and the concentration of ATP required to obtain half-maximal velocity. The maximal velocity of the ouabain-sensitive Ca2+ + Na+ + K+-dependent ATPase was about two orders of magnitude smaller than that of Mg2+ + Na+ + K+-dependent activity. In agreement with previous observations, it was shown that in the presence of Ca2+, Na+, and ATP, an acid-stable phosphoenzyme was formed that was sensitive to either ADP or K+. The enzyme also exhibited a Ca2+ + Na+-dependent ADP-ATP exchange activity. Neither the inhibitory effects of Ca2+ on Mg2+-dependent activities, nor the Ca2+-dependent activities were influenced by the addition of calmodulin. Because of the presence of small quantities of endogenous Mg2+ in all reaction mixtures, it could not be determined whether the apparent Ca2+-dependent activities involved enzyme-substrate complexes containing Ca2+ as the divalent cation or both Ca2+ and Mg2+.  相似文献

The effects of thyroxine on the activity of different ATPases (Na+-K+, Ca2+, and Mg2+) in fat body cells of the silkworm, Bombyx mori, were investigated during different developmental stages. In both sexes the maximum enzyme activity was observed in the fat body cells of day 7 last instar larva (the day before spinning). Na+-K+, Ca2+-, and Mg2+-ATPase activity in the fat body markedly declined after pupation and continued to decrease in day 1 adults. Injection of thyroxine (T4) at doses of 1.0 and 2.0 μg/g during fifth instar significantly elevated all ATPase activities in the larval, pupal, and adult stages in both sexes. At a dose of 0.5 μg/g, T4 had no effect on day 2 fifth instar larva, although it increased the ATPase activity at the other stages investigated. A higher dose (3.0 μg/g) caused a significant reduction in enzyme activity in all stages with the exception of day 2 fifth instar larva. Thus, the repression of enzyme activity with the higher dose and the elevation of enzyme activity with the lower dose establish the biphasic nature of T4 action on the ATPase system in fat body cells of the silkworm. Alterations in the activity of ATPases, peroxidases and lipoxygenases were studied during early stages of post-harvest deterioration of cassava (Manihot esculenta Crantz cv. Oyolu) root tubers. The peak activities of Ca2+-ATPase, (Ca2++Mg2+)-ATPase, Na++K+-ATPase, Mg2+-ATPase and peroxidases were observed after the first 24 h and thereafter decreased. The activity of lipoxygenase was biphasic, probably depicting two distinct isoforms expressed during deterioration. The results indicate that ATPases and peroxidases have a role in the post-harvest deterioration of cassava tuber, but the participation of lipoxygenases seems unlikely.  相似文献

The effect of alcohol on enzymes involved in energy metabolism of nervous tissue were analyzed, in vivo after acute and chronic ethanol administration to rats and in vitro by addition of 50 mM and 100 mM ethanol to the medium of cultured nerve cells: chick neurons, chick glial cells, a neuronal cell line (MT17) and a glial tumoral cell line (C6). The parameters we measured were (Na+,K+), Mg2+ and ecto Ca2+,Mg2+ ATPase activities involved in transport phenomena and enolase activities (non neuronal NNE and neuron specific enolase NSE) as markers of nerve cell maturation. In vivo, after chronic ethanol administration (Na+,K+) ATPase activity was increased while Mg2+ dependent activity was not affected. Enolase activity was decreased. Acute ethanol administration decreased (Na+,K+) ATPase activity, while Mg2+ dependent activity was not affected. In cultured nerve cells ethanol effect was dose, time and cell type dependent; alterations of the cell membrane by trypsinization of the tissue before seeding modifies the effect of ethanol on the enzymes we analyzed. Our results suggest that alcohol effect on nerve cells depends mainly on the lipoprotein structure of the cell membranes which may have different properties from one cell type to another.  相似文献

Trifluoperazine dihydrochloride-induced inhibition of calmodulin-activated Ca2+-ATPase and calmodulin-insensitive (Na+ + K+)- and Mg2+-ATPase activities of rat and human red cell lysates and their isolated membranes was studied. Trifluoperazine inhibited both calmodulin-sensitive and calmodulin-insensitive ATPase activities in these systems. The concentration of trifluoperazine required to produce 50% inhibition of calmodulin-sensitive Ca2+-ATPase was found to be slightly lower than that required to produce the same level of inhibition of other ATPase activities. Drug concentrations which inhibited calmodulin-sensitive ATPase completely, produced significant reduction in calmodulin-insensitive ATPases as well. The data presented in this report suggest that trifluoperazine is slightly selective towards calmodulin-sensitive Ca2+-ATPase but that it is also capable of inhibiting calmodulin-insensitive (Na+ + K+)-ATPase and Mg2+-ATPase activities of red cells at relatively low concentrations. Thus the action of the drug is not due entirely to its interaction with calmodulin-mediated processes, and trifluoperazine cannot be assumed to be a selective inhibitor of calmodulin interactions under all circumstances.  相似文献

Ca2+ inhibited the Mg2+-dependent and K+-stimulated p-nitrophenylphosphatase activity of a highly purified preparation of dog kidney (Na+ + K+)-ATPase. In the absence of K+, however, a Mg2+-dependent and Ca2+-stimulated phosphatase was observed, the maximal velocity of which, at pH 7.2, was about 20% of that of the K+-stimulated phosphatase. The Ca2+-stimulated phosphatase, like the K+-stimulated activity, was inhibited by either ouabain or Na+ or ATP. Ouabain sensitivity was decreased with increase in Ca2+, but the K0.5 values of the inhibitory effects of Na+ and ATP were independent of Ca2+ concentration. Optimal pH was 7.0 for Ca2+-stimulated activity, and 7.8–8.2 for the K+-stimulated activity. The ratio of the two activities was the same in several enzyme preparations in different states of purity. The data indicate that (a) Ca2+-stimulated phosphatase is catalyzed by (Na+ + K+)-ATPase; (b) there is a site of Ca2+ action different from the site at which Ca2+ inhibits in competition with Mg2+; and (c) Ca2+ stimulation can not be explained easily by the action of Ca2+ at either the Na+ site or the K+ site.  相似文献

N-Ethylmaleimide was employed as a surface label for sarcolemmal proteins after demonstrating that it does not penetrate to the intracellular space at concentrations below 1·10?4 M. The sarcolemmal markers, ouabain-sensitive (Na+ + K+)-ATPase and Na+/Ca2+-exchange activities, were inhibited in N-ethylmaleimide perfused hearts. Intracellular activities such as creatine phosphokinase, glutamate-oxaloacetate transaminase and the internal phosphatase site of the Na+ pump (K+-p-nitrophosphatase) were not affected. Almost 20% of the (Ca2+ + Mg2+)-ATPase and Ca2+ pump were inhibited indicating the localization of a portion of this activity in the sarcolemma. Sarcolemma purified by a recent method (Morcos, N.C. and Drummond, G.I. (1980) Biochim. Biophys. Acta 598, 27–39) from N-ethylmaleimide-perfused hearts showed loss of approx. 85% of its (Ca2+ + Mg2+-ATPase and Ca2+ pump compared to control hearts. (Ca2+ + Mg2+)-ATPase and Ca2+ pump activities showed two classes of sensitivity to vanadate ion inhibition. The high vanadate affinity class (K12 for inhibition approx. 1.5 μM) may be localized in the sarcolemma and represented approx. 20% of the total inhibitable activity in agreement with estimates from N-ethylmaleimide studies. Sucrose density fractionation indicated that only a small portion of Mg2+-ATPase and Ca2+-ATPase may be associated with the sarcolemma. The major portion of these activities seems to be associated with high density particles.  相似文献

Because intracellular [Na+] is kept low by Na+/K+-ATPase, Na+ dependence is generally considered a property of extracellular enzymes. However, we found that p94/calpain 3, a skeletal-muscle-specific member of the Ca2+-activated intracellular “modulator proteases” that is responsible for a limb-girdle muscular dystrophy (“calpainopathy”), underwent Na+-dependent, but not Cs+-dependent, autolysis in the absence of Ca2+. Furthermore, Na+ and Ca2+ complementarily activated autolysis of p94 at physiological concentrations. By blocking Na+/K+-ATPase, we confirmed intracellular autolysis of p94 in cultured cells. This was further confirmed using inactive p94:C129S knock-in (p94CS-KI) mice as negative controls. Mutagenesis studies showed that much of the p94 molecule contributed to its Na+/Ca2+-dependent autolysis, which is consistent with the scattered location of calpainopathy-associated mutations, and that a conserved Ca2+-binding sequence in the protease acted as a Na+ sensor. Proteomic analyses using Cs+/Mg2+ and p94CS-KI mice as negative controls revealed that Na+ and Ca2+ direct p94 to proteolyze different substrates. We propose three roles for Na+ dependence of p94; 1) to increase sensitivity of p94 to changes in physiological [Ca2+], 2) to regulate substrate specificity of p94, and 3) to regulate contribution of p94 as a structural component in muscle cells. Finally, this is the first example of an intracellular Na+-dependent enzyme.  相似文献

The seawater cations, Na+, K+, Mg2+, and Ca2+, each stimulated MnO2-reductase activity of whole cells and cell extracts of Bacillus 29. Concentrations of Na+ and K+ which stimulated whole cells and cell extracts maximally were equivalent to those in two- to fivefold diluted seawater. Cell-extract activity was strongly stimulated by Ca2+ and Mg2+ up to a concentration of 0.01 M Mg2+ and 0.002 M Ca2+, with little additional stimulation above these concentrations. Whole-cell activity was stimulated biphasically with increasing concentrations of Ca2+ and Mg2+. Comparison of the effects of individual cations or mixtures of them at concentrations equivalent to their concentration in fivefold diluted seawater showed that more activity was obtained with 0.01 M Mg2+ or 0.002 M Ca2+ than with 0.1 M Na+, and more with 0.1 M Na+ than with 0.0022 M K+. Fivefold diluted seawater permitted as much or more activity as solutions of individual or synthetic mixtures of the cations. Pre-exposure experiments showed that the ionic history of whole cells was important to their ultimate activity. The MnO2-reductase activity of induced whole cells exhibited a temperature optimum near 40 C. Cell extracts had different temperature optima (Topt), depending on whether induced glucose-linked activity (Topt = 25 C), uninduced glucose-linked, ferricyanide-dependent activity (Topt = 30 C), or uninduced ferrocyanide-linked activity (Topt = 40 C) were being measured.
