Abstract. Pressure-volume measurements were made on Artemisia tridentata Nutt. ssp. tridentata samples rehydrated for 0, 1.5, 3, 6 or 24 h. Increasing rehydration time caused a significant increase in osmotic potential at turgor loss, cell elasiticity, and the relative water content at turgor loss, and a significant decrease in pressure potential at saturation. Osmotic potential at saturation was changed significantly by rehydration, but no consistent trend was observed. The symplastic water fraction did not differ significantly among treatments. The increase in the osmotic potential at turgor loss did not correspond with decreasing cell elasticity or synthesis of solutes. Instead, the leaf solute content remained constant suggesting a redistribution of solutes between the symplast and apoplast. Using non-rehydrated samples for pressure-volume analysis introduced errors in estimates of the symplastic water fraction, osmotic potential at full turgor, and the relative water content at turgor loss. These errors are due to uncertainties in the determination of saturated weights. 相似文献

Two methods of rehydrating red pine ( Pinus resinosa Ait.) shoots for pressure-volume (PV) analysis were compared to clarify the effects of rehydration on estimated tissue water relations of shoots and fascicles. The commonly employed cut-shoot method was compared to rehydration by means of water uptake through the roots of intact plants. Cut-shoot rehydration and increased duration of rehydration significantly decreased estimates of tissue elasticity and relative water content at zero turgor for both shoots and fascicles. Rehydration of cut shoots for 2 days significantly increased the slope of the linear region of shoot pressure-volume (PV) curves and decreased estimates of the apoplastic water fraction. Changes in these and estimates of other water relations attributes were correlated with increased initial water content during rehydration. Estimated apoplastic water fraction was higher for needle fascicles than shoots despite the large amount of woody stem tissue contained in shoots. Fascicle water status strongly influenced shoot water relations, in part due to apparent apoplastic loading with water of fascicles during rehydration. 相似文献

The occurrence of mature individuals of Pseudotsuga menziesii in stands of Arctostaphylos species mark the initial stages of mixed evergreen forest invasion into chaparral in central coastal California. We planted
two cohorts of P. menziesii seeds at three sites under stands of two Arctostaphylos species and Adenostoma fasciculatum in order to determine whether first-year seedling emergence and survival, particularly during the regular summer drought,
underlie the spatial distribution of mature trees observed in chaparral. Regardless of the chaparral species they were planted
under, P. menziesii seeds that were not protected from vertebrate predation displayed very little emergence and no survival. In contrast, emergence
of P. menziesii that were protected from vertebrate predators was much higher but still did not significantly differ among the three chaparral
species. However, survival of protected seedlings under Arctostaphylos glandulosa was much greater than under A. fasciculatum, with intermediate survival under Arctostaphylos montana. While mortality of protected seedlings due to insect herbivory, fungal infection, and disturbance displayed no consistent
patterns, summer drought mortality appeared to drive the patterns of survival of P. menziesii under the different chaparral species. These emergence, mortality, and survival data suggest that spatial patterns of P. menziesii recruitment in chaparral are driven by first-year summer drought seedling mortality, but only in years when seeds and seedlings
are released from vertebrate predation pressure. Because the first-year drought mortality and survival patterns of P. menziesii seedlings differed strongly depending on the chaparral species, we examined the additional hypothesis that these patterns
are associated with differences in the availability of soil moisture under different chaparral species. Both higher survival
and lower drought mortality of P. menziesii seedlings were associated with higher soil water potential under Arctostaphylos stands during the summer drought, especially in the subsurface soil. The data suggest that Arctostaphylos stands, particularly stands of A. glandulosa, ameliorate xeric summer conditions to a degree that facilitates first-year establishment of P. menziesii and strongly influences spatial distribution of mature trees.

The primary objective of this study was to assess metabolomics for its capacity to discern biological variation among 10 full-sib families of a Douglas-fir tree breeding population, replicated on two sites. The differential accumulation of small metabolites in developing xylem was examined through metabolite profiles (139 metabolites common to 181 individual trees) generated by gas chromatography mass spectrometry and a series of statistical analyses that incorporated family, site, and tree growth and quantitative phenotypic wood traits (wood density, microfibril angle, wood chemistry and fiber morphology). Multivariate discriminant, canonical discriminant and factor analyses and broad-sense heritabilities revealed that metabolic and phenotypic traits alike were strongly related to site, while similar associations relating to genetic (family) structure were weak in comparison. Canonical correlation analysis subsequently identified correlations between specific phenotypic traits (i.e. tree growth, fibre morphology and wood chemistry) and metabolic traits (i.e. carbohydrate and lignin biosynthetic metabolites), demonstrating a coherent relationship between genetics, metabolism, environmental and phenotypic expression in wood-forming tissue. The association between cambial metabolites and tree phenotype, as revealed by metabolite profiling, demonstrates the value of metabolomics for systems biology approaches to understanding tree growth and secondary cell wall biosynthesis in plants. 相似文献

The present article investigates the chemical composition of volatiles of essential oil (EO) and headspace (HS) fraction, as well as biological activities of EO obtained from needles with twigs of Pseudotsuga menziesii var. menziesii cultivated in Serbia. The major class of compounds was monoterpene hydrocarbons with α-terpinolene, sabinene and β-pinene (EO), and sabinene, α-terpinolene and β-pinene (HS) as the dominant volatiles. Tested EO exhibited mostly low antimicrobial potential against investigated strains (ATCC and respiratory isolates), where MICs ranged 1.25–20.00 mg/mL. Nevertheless, based on presented results, where antimicrobial testing was done for the first time on human respiratory system isolates, there is a potential of this EO to be used as an adjuvant in the treatment of human respiratory infections, especially those caused by Pseudomonas aeruginosa or Candida albicans strains. Regarding toxicological evaluation, EO showed moderate toxicity in Artemia salina toxicity bioassay (LC50=347.41, after 24 h) as well as week toxicity against Drosophila melanogaster with the ability only to moderately delay larval and pupal development. 相似文献

Abstract. The purpose of this paper is to examine a technique for estimating the weight at full saturation (Ws ) from pressure-volume (P-V) analysis of non-rehydrated plant tissue at various water deficits. Tissue samples are typically rehydrated prior to P-V analysis to determine Ws , necessary to calculate many tissue water parameters. However, several studies have indicated that artificial rehydration may significantly alter P-V relationships, such as the plateau effect, resulting in erroneous measurements of tissue elasticity and osmotic potentials. The results of this study suggest that linear regression of P-V data at and above the turgor loss point may be used to extrapolate Ws from non-rehydrated samples at various moisture deficits, thus eliminating the plateau effect and other potential rehydration problems. Determination coefficients and standard errors of the Y-intercept indicated a strong linear relationship between tissue fresh weight and water potential (Ψ), and a high degree of predictability of Ws in all but one of the species-treatment combinations evaluated in this study, despite predawn Ψ as low as - 1.0 MPa. 相似文献

Vesicular-arbuscular mycorrhizae (VAM) were common in seedlings of Pseudotsuga menziesii and Tsuga heterophylla grown in a greenhouse soil bioassay in soils collected from the Oregon Coast Range. Although root samples were heavily colonized
by ectomycorrhizal fungi (EM), VAM colonization was observed in the cortical cells of both secondary and feeder roots. Vesicles,
arbuscules, and hyphae typical of VAM occurred in 48% of 61 P. menziesii and 25% of 57 T. heterophylla seedlings. The ecological significance of VAM presence in the Pinaceae, as well as interactions among VAM, EM, and the plant
host, deserve future investigation.

Here we describe a simple method for pulse-labeling tree seedlings with 13CO2(gas), and then apply the method in two related experiments: t (i) comparison of carbon allocation patterns between t Betula papyrifera Marsh. and t Pseudotsuga menziesii (Mirb.) Franco, and t (ii) measurement of one-way belowground carbon transfer from t B. papyrifera to t P. menziesii. Intraspecific carbon allocation patterns and interspecific carbon transfer both influence resource allocation, and consequently development, in mixed communities of t B. papyrifera and t P. menziesii.In preparation for the two experiments, we first identified the appropriate 13CO2(gas) pulse-chase regime for labeling seedlings: a range of pulse (100-mL and 200-mL 99 atom%13 CO2(gas)) and chase (0, 3 and 6 d) treatments were applied to one year-old t B. papyrifera and t P. menziesii seedlings. The amount of 13CO2 fixed immediately after 1.5 h exposure was greatest for both t B. papyrifera (40.8 mg excess 13C) and t P. menziesii (22.9 mg excess 13C) with the 200-mL pulse, but higher 13C loss and high sample variability resulted in little difference in excess13 C content between pulse treatments after 3 d for either species. The average excess 13C root/shoot ratio of t B. papyrifera and t P. menziesii changed from 0.00 immediately following the pulse to 0.61 and 0.87 three and six days later, which reflected translocation of 75% of fixed isotope out of foliage within 3 d following the pulse and continued enrichment in fine roots over 6 d. Based on these results, the 100-mL CO2(gas) and 6-d chase were considered appropriate for the carbon allocation and belowground transfer experiments.In the carbon allocation experiment, we found after 6 d that t B. papyrifera allocated 49% (average 9.5 mg) and t P. menziesii 41% (average 5.8 mg) of fixed isotope to roots, of which over 55% occurred in fine roots in both species. Species differences in isotope allocation patterns paralleled differences in tissue biomass distribution. The greater pulse labeling efficiency of t B. papyrifera compared to t P. menziesii was associated with its two-fold and 13- fold greater leaf and whole seedling net photosynthetic rates, respectively, 53% greater biomass, and 35% greater root/shoot ratio.For the carbon transfer experiment, t B. papyrifera and t P. menziesii were grown together in laboratory rootboxes, with their roots intimately mingled. A pulse of 100 mL13 CO2(gas) was applied to paper birch and one-way transfer to neighboring t P. menziesii was measured after 6 d. Of the excess 13C fixed by t B. papyrifera, 4.7% was transferred to neighboring t P. menziesii, which distributed the isotope evenly between roots and shoots. Of the isotope received by t P. menziesii, we estimated that 93% was taken up through belowground pathways, and the remaining 7% taken up by foliage as13 CO2(gas) respired by t B. papyrifera shoots. These two experiments indicate that t B. papyrifera fixes more total carbon and allocates a greater proportion to its root system than does t P. menziesii, giving it a competitive edge in resource gathering; however, below-ground carbon sharing is of sufficient magnitude that it may help ensure co-existence of the two species in mixed communities. 相似文献

We measured carbon (respiration, photosynthesis, and production) and nitrogen allocation to male and female cones of Rocky Mountain Douglas-fir (Pseudotsuga menziesii var. glauca) to quantify gender-specific: (1) resource allocation to reproduction, and (2) contribution to carbon costs of reproduction via photosynthesis. We also measured foliar photosynthesis and nitrogen concentration ([N]) near and far from female cones to examine the relationship between reproduction and foliar physiology. Over one growing season, male cones required only 8% of all carbon allocated to reproduction, with females consuming the remaining 92%. Female cones, however, had maximum instantaneous refixation rates of 54%, which, integrated over the season, offset 6% of their total carbon requirements, while male cones were completely dependent on vegetative tissues for carbon. Male cones received 22% of all nitrogen allocated to reproduction and female cones received the remaining 78%. Foliage near female cones had elevated photosynthesis during the early stages of cone development and consistently lower [N] than foliage far from cones. Although female cones may photosynthesize, the annual sum of carbon fixed by reproductive structures is minor in comparison to the total carbon allocated to production and respiration. 相似文献

Bud quiescence release, considered as the ultimate dormancy breaking phase, was achieved in Pseudotsuga menziesii (Mirb.) Franco by a 9-week cold (5°C) treatment, under short daylength (9 h) followed by a transfer to mild temperature (22°C) under long daylength (16 h). Indole-3-acetic acid (IAA), abscisic acid (ABA), zeatin-type (Z) and isopentenyladenine-type (iPA) cytokinin (CK) levels were measured by means of an ELISA technique performed on HPLC-fractionated extracts of terminal and axillary buds. During the cold period, all hormones except IP-type CK levels decreased, whereas the opposite observation was made after transfer to mild temperature and long daylength, when buds started to grow. Some other immunoreactive compounds were also detected and quantified. The ABA-glucosyl ester (ABA-GE) level pattern was similar to that of ABA, but no accumulation occurred at mild temperatures. A putative IAA conjugate, more polar than IAA, was also detected. Its level increased transiently like IAA in terminal buds and, to a lesser extent, in axillary buds during the 10th week of the experiment. In terminal buds, isopentenyladenosine ([9R]-iP) was released by alkaline hydrolysis of a polar immunoreactive compound detected with anti-[9R]iP antibodies. This compound accumulated during the cold period and quickly dropped at 22°C. Relationships between environmental conditions and endogenous hormones are discussed. 相似文献

We studied the effect of ectomycorrhizal fungi on bacterial communities colonizing roots of Douglas fir (Pseudotsuga menziesii). Mycorrhizal tips were cleaned of soil and separated based on gross morphological characteristics. Sequencing of the internal transcribed spacers of the nuclear rRNA gene cluster indicated that the majority of the tips were colonized by fungi in the Russulaceae, with the genera Russula and Lactarius comprising 70% of the tips. Because coamplification of organellar 16S rRNA genes can interfere with bacterial community analysis of root tips, we developed and tested a new primer pair that permits amplification of bacterial 16S rRNA genes but discriminates more effectively against organellar sequences than commonly used bacterial primer sets. We then used terminal restriction fragment length polymorphism (T-RFLP) and sequence analysis of the 16S rRNA gene to examine differences in bacterial communities associated with the mycorrhizal tips. Cluster analysis of T-RFLP profiles indicated that there were different bacterial communities among the root tips; however, the communities did not seem to be affected by the taxonomic identity of the ectomycorrhizal fungi. Terminal restriction fragment profiling and sequencing of cloned partial 16S rRNA genes indicated that most bacteria on the ectomycorrhizal tips were related to the Alphaproteobacteria and the Bacteroidetes group. 相似文献

Our previous attempt on in vitro fertilization (IVF) in conifers resulted in pollen tube penetration of female gametophytes,
but because of the rapid decline in egg viability, no further interaction occurred. In this report, we describe for the first
time that IVF has been achieved in conifers. Using Douglas fir (Pseudotsuga menziesii), we describe a two-step process which involved induction of pollen tubes in culture followed by introduction of isolated
female gametophytes at the tips of growing pollen tubes. Pollen tubes penetrated the introduced isolated female gametophytes
at various places, but a number of tubes entered the egg cell through the neck cells similar to the in vivo condition. Under
our current culture conditions, longevity of pollen tubes and eggs has been improved resulting in the release of sperms, fusion
of gametes, and initial formation of the proembryo. Continued plasmolysis of the egg limited the number of successful gametic
interactions. IVF has been accomplished in flowering plants in several ways, but the gametophyte-gametophyte IVF system described
in this paper is unique. IVF offers a novel breeding technology that takes advantage of the sexual reproductive route. When
coupled with hybridization and genetic transformation, IVF could result in the development of stable novel genotypes of economically
superior trees.

Seedlings of Douglas-fir, Pseudotsuga menziesii (Mirb.) Franco, and ponderosa pine, Pinus ponderosa Dougl. ex Laws., were grown in a controlled environment and fertilized with nutrient solutions containing 150 ppm (+N), or
0 ppm nitrogen (−N). These treatments greatly altered seedling growth, and the concentrations of N and carbohydrates in their
tissues. Metabolically active tissues, such as roots, incubated with a limited supply of O2 became hypoxic faster and synthesized more ethanol than less active tissues, such as needles. All tissues that were incubated
for 4 h in N2 synthesized ethanol. Needles incubated in N2 and light had much lower quantities of ethanol than needles in N2 and dark, suggesting that O2 from photosynthetsis limited internal anoxia. Most tissues from +N seedlings synthesized greater quantities of ethanol in
N2 anoxia than tissues from −N seedlings, probably because they were able to produce more enzymes with a greater availability
of N. However, this increase in ethanol synthesis between N treatments was most pronounced in the phloem. Ethanol and soluble
sugar concentrations were negatively related in needles and positively related in roots of N+ seedlings, but not −N seedlings.
Starch concentrations had no effect on the amount of ethanol produced by any tissue. Regardless of N treatments, all tissues
from ponderosa pine produced more N2-induced ethanol than Douglas-fir, in part because its tissues contained different concentrations of soluble sugars and N
as a consequence of phenological differences between the species. However, ponderosa pine tissues may also maintain greater
quantities of anaerobic enzymes, or their isozymes than Douglas-fir.

Douglas fir (Pseudotsuga menziesii) variety glauca (DFG) but not the variety viridis (DFV) showed symptoms of manganese (Mn) toxicity in some field sites. We hypothesized that these two varieties differed in Mn metabolism. To test this hypothesis, biomass partitioning, Mn concentrations, subcellular localization and 54Mn-transport were investigated. Total Mn uptake was three-times higher in DFG than in DFV. DFV retained > 90% of 54Mn in roots, whereas > 60% was transported to the shoot in DFG. The epidermis was probably the most efficient Mn barrier since DFV contained lower Mn concentrations in cortical cells and vacuoles of roots than DFG. In both varieties, xylem loading was restricted and phloem transport was low. However, sieve cells still contained high Mn concentrations. DFV displayed higher biomass production and higher shoot : root ratios than DFG. Our results clearly show that both varieties of Douglas fir differ significantly in Mn-uptake and allocation patterns rendering DFG more vulnerable to Mn toxicity. 相似文献

Abstract Leaf water relations characteristics of creosote bush, Larrea tridentata, were studied in view of previous reports that its leaves commonly experience zero or negative turgor under dry conditions. Leaf turgor loss point () was determined by a pressure-volume method for samples subjected to a hydration procedure and for untreated samples. Hydration caused to increase by as much as 3 M Pa. Hydration of samples also caused changes in other leaf water relations characteristics such as symplastic solute content, tissue elasticity and symplasmic water fraction, but total leaf solute content was unchanged. Comparison of our field plant water potential data with values of obtained by the two methods resulted in predictions of turgor loss during part or all of a diurnal cycle based on hydrated samples, and turgor maintenance (at least 0.3 MPa) based on untreated samples. Pooled data for obtained from both partially hydrated and untreated samples showed that L. tridentata maintains fairly constant levels of turgor over a wide range of leaf water potential. Dilution of cell contents by apoplastic water introduced significant errors in psychrometric determinations of osmotic potential in both frozen and thawed leaf tissue and expressed cell sap. Use of these values of osmotic potential resulted in predictions of zero turgor at all plant water potentials measured in the field. 相似文献

The gravitational component of water potential contributes a standing 0.01 MPa m?1 to the xylem tension gradient in plants. In tall trees, this contribution can significantly reduce the water potential near the tree tops. The turgor of cells in buds and leaves is expected to decrease in direct proportion with leaf water potential along a height gradient unless osmotic adjustment occurs. The pressure–volume technique was used to characterize height‐dependent variation in leaf tissue water relations and shoot growth characteristics in young and old Douglas‐fir trees to determine the extent to which growth limitation with increasing height may be linked to the influence of the gravitational water potential gradient on leaf turgor. Values of leaf water potential (Ψl), bulk osmotic potential at full and zero turgor, and other key tissue water relations characteristics were estimated on foliage obtained at 13.5 m near the tops of young (approximately 25‐year‐old) trees and at 34.7, 44.2 and 55.6 m in the crowns of old‐growth (approximately 450‐year‐old) trees during portions of three consecutive growing seasons. The sampling periods coincided with bud swelling, expansion and maturation of new foliage. Vertical gradients of Ψl and pressure–volume analyses indicated that turgor decreased with increasing height, particularly during the late spring when vegetative buds began to swell. Vertical trends in branch elongation, leaf dimensions and leaf mass per area were consistent with increasing turgor limitation on shoot growth with increasing height. During the late spring (May), no osmotic adjustment to compensate for the gravitational gradient of Ψl was observed. By July, osmotic adjustment had occurred, but it was not sufficient to fully compensate for the vertical gradient of Ψl. In tall trees, the gravitational component of Ψl is superimposed on phenologically driven changes in leaf water relations characteristics, imposing potential constraints on turgor that may be indistinguishable from those associated with soil water deficits. 相似文献
