We examined the effects of root and litter exclusion on the rate of soil CO2 efflux and microbial biomass using trenching and tent separation techniques in a secondary forest (SF) and a pine (Pinus caribaea Morelet) plantation in the Luquillo Experimental Forest in Puerto Rico. Soil surface CO2 efflux was measured using the alkali trap method at 12 randomly-distributed locations in each treatment (control, root exclusion, litter exclusion, and both root and litter exclusion) in the plantation and the SF, respectively. We measured soil CO2 efflux every two months and collected soil samples at each sampling location in different seasons to determine microbial biomass from August 1996 to July 1997. We found that soil CO2 efflux was significantly reduced in the litter and root exclusion plots (7-year litter and/or root exclusion) in both the secondary forest and the pine plantation compared with the control. The reduction of soil CO2 efflux was 35.6% greater in the root exclusion plots than in the litter exclusion plots in the plantation, whereas a reversed pattern was found in the secondary forest. Microbial biomass was also reduced during the litter and root exclusion period. In the root exclusion plots, total fungal biomass averaged 31.4% and 65.2% lower than the control plots in the plantation and the secondary forest, respectively, while the total bacterial biomass was 24% and 8.3% lower than the control plots in the plantation and the secondary forest, respectively. In the litter exclusion treatment, total fungal biomass averaged 69.2% and 69.7% lower than the control plots in the plantation and the secondary forest, respectively, while the total bacterial biomass was 48% and 50.1% lower than the control plots in the plantation and the secondary forest, respectively. Soil CO2 efflux was positively correlated with both fungal and bacterial biomass in both the plantation the secondary forest. The correlation between soil CO2 efflux and active fungal biomass was significantly higher in the plantation than in the secondary forest. However, the correlation between the soil CO2 efflux and both the active and total bacterial biomass was significantly higher in the secondary forest than in the plantation in the day season. In addition, we found soil CO2 efflux was highly related to the strong interactions among root, fungal and bacterial biomass by multiple regression analysis (R2 > 0.61, P < 0.05). Our results suggest that carbon input from aboveground litterfall and roots (root litter and exudates) is critical to the soil microbial community and ecosystem carbon cycling in the wet tropical forests.

Restoration of California native perennial grassland is often initiated with cultivation to reduce the density and cover of non‐native annual grasses before seeding with native perennials. Tillage is known to adversely impact agriculturally cultivated land; thus changes in soil biological functions, as indicated by carbon (C) turnover and C retention, may also be negatively affected by these restoration techniques. We investigated a restored perennial grassland in the fourth year after planting Nassella pulchra, Elymus glaucus, and Hordeum brachyantherum ssp. californicum for total soil C and nitrogen (N), microbial biomass C, microbial respiration, CO2 concentrations in the soil atmosphere, surface efflux of CO2, and root distribution (0‐ to 15‐, 15‐ to 30‐, 30‐ to 60‐, and 60‐ to 80‐cm depths). A comparison was made between untreated annual grassland and plots without plant cover still maintained by tillage and herbicide. In the uppermost layer (0‐ to 15‐cm depth), total C, microbial biomass C, and respiration were lower in the tilled, bare soil than in the grassland soils, as was CO2 efflux from the soil surface. Root length near perennial bunchgrasses was lower at the surface and greater at lower depths than in the annual grass–dominated areas; a similar but less pronounced trend was observed for root biomass. Few differences in soil biological or chemical properties occurred below 15‐cm depth, except that at lower depths, the CO2 concentration in the soil atmosphere was lower in the plots without vegetation, possibly from reduced production of CO2 due to the lack of root respiration. Similar microbiological properties in soil layers below 15‐cm depth suggest that deeper microbiota rely on more recalcitrant C sources and are less affected by plant removal than in the surface layer, even after 6 years. Without primary production, restoration procedures with extended periods of tillage and herbicide applications led to net losses of C during the plant‐free periods. However, at 4 years after planting native grasses, soil microbial biomass and activity were nearly the same as the former conditions represented by annual grassland, suggesting high resilience to the temporary disturbance caused by tillage.

The anthropogenic rise in atmospheric CO2 is expected to impact carbon (C) fluxes not only at ecosystem level but also at the global scale by altering C cycle processes in soils. At the Swiss Canopy Crane (SCC), we examined how 7 years of free air CO2 enrichment (FACE) affected soil CO2 dynamics in a ca. 100‐year‐old mixed deciduous forest. The use of 13C‐depleted CO2 for canopy enrichment allowed us to trace the flow of recently fixed C. In the 7th year of growth at ~550 ppm CO2, soil respiratory CO2 consisted of 39% labelled C. During the growing season, soil air CO2 concentration was significantly enhanced under CO2‐exposed trees. However, elevated CO2 failed to stimulate cumulative soil respiration (Rs) over the growing season. We found periodic reductions as well as increases in instantaneous rates of Rs in response to elevated CO2, depending on soil temperature and soil volumetric water content (VWC; significant three‐way interaction). During wet periods, soil water savings under CO2‐enriched trees led to excessive VWC (>45%) that suppressed Rs. Elevated CO2 stimulated Rs only when VWC was ≤40% and concurrent soil temperature was high (>15 °C). Seasonal Q10 estimates of Rs were significantly lower under elevated (Q10=3.30) compared with ambient CO2 (Q10=3.97). However, this effect disappeared when three consecutive sampling dates of extremely high VWC were disregarded. This suggests that elevated CO2 affected Q10 mainly indirectly through changes in VWC. Fine root respiration did not differ significantly between treatments but soil microbial biomass (Cmic) increased by 14% under elevated CO2 (marginally significant). Our findings do not indicate enhanced soil C emissions in such stands under future atmospheric CO2. It remains to be shown whether C losses via leaching of dissolved organic or inorganic C (DOC, DIC) help to balance the C budget in this forest.

Forest soil is a major component of terrestrial ecosystems for carbon sequestration and plays an important role in the global carbon cycle. Soil carbon flux and soil carbon pools were investigated in a poplar plantation chronosequence over a rotation in northwest China. Based on continuous field observation in 2007, the results showed that mean soil CO2 efflux rate was 5.54, 4.81, and 3.93 μmol CO2 m−2 s−1 for stands of 2-, 8-, and 15-year-old, respectively, during the growing season. Significant differences in soil respiration of three age classes were mainly because soil temperature, carbon allocation, and fine root growth changed greatly with stand age. Multiple regression analysis suggested that soil temperature and fine root biomass in the upper layer could explain 78–85% of the variation in soil respiration. Mineral soil C stock at 0–40 cm depth was 55.77, 55.09, and 58.14 t ha−1 in the 2-, 8-, and 15-year-old stands, respectively. The average rate of soil C sequestration was 0.13 t ha−1 year−1 following afforestation on former crop lands. Although the plantations had similar management practices and soil types since their establishment, many biotic and abiotic factors such as root biomass and turnover rate, soil condition of the plantations had undergone marked changes at different development stages, which could result in the remarkable differences in soil carbon flux and storage over a rotation. Our results highlight the importance of the development stage within a rotation of poplar plantation in assessment of soil carbon budget.

Increased partitioning of carbon (C) to fine roots under elevated [CO2], especially deep in the soil profile, could alter soil C and nitrogen (N) cycling in forests. After more than 11 years of free‐air CO2 enrichment in a Liquidambar styraciflua L. (sweetgum) plantation in Oak Ridge, TN, USA, greater inputs of fine roots resulted in the incorporation of new C (i.e., C with a depleted δ13C) into root‐derived particulate organic matter (POM) pools to 90‐cm depth. Even though production in the sweetgum stand was limited by soil N availability, soil C and N contents were greater throughout the soil profile under elevated [CO2] at the conclusion of the experiment. Greater C inputs from fine‐root detritus under elevated [CO2] did not result in increased net N immobilization or C mineralization rates in long‐term laboratory incubations, possibly because microbial biomass was lower in the CO2‐enriched plots. Furthermore, the δ13CO2 of the C mineralized from the incubated soil closely tracked the δ13C of the labile POM pool in the elevated [CO2] treatment, especially in shallower soil, and did not indicate significant priming of the decomposition of pre‐experiment soil organic matter (SOM). Although potential C mineralization rates were positively and linearly related to total SOM C content in the top 30 cm of soil, this relationship did not hold in deeper soil. Taken together with an increased mean residence time of C in deeper soil pools, these findings indicate that C inputs from relatively deep roots under elevated [CO2] may increase the potential for long‐term soil C storage. However, C in deeper soil is likely to take many years to accrue to a significant fraction of total soil C given relatively smaller root inputs at depth. Expanded representation of biogeochemical cycling throughout the soil profile may improve model projections of future forest responses to rising atmospheric [CO2].

Indonesia lost more tropical forest than all of Brazil in 2012, mainly driven by the rubber, oil palm, and timber industries. Nonetheless, the effects of converting forest to oil palm and rubber plantations on soil organic carbon (SOC) stocks remain unclear. We analyzed SOC losses after lowland rainforest conversion to oil palm, intensive rubber, and extensive rubber plantations in Jambi Province on Sumatra Island. The focus was on two processes: (1) erosion and (2) decomposition of soil organic matter. Carbon contents in the Ah horizon under oil palm and rubber plantations were strongly reduced up to 70% and 62%, respectively. The decrease was lower under extensive rubber plantations (41%). On average, converting forest to plantations led to a loss of 10 Mg C ha?1 after about 15 years of conversion. The C content in the subsoil was similar under the forest and the plantations. We therefore assumed that a shift to higher δ13C values in plantation subsoil corresponds to the losses from the upper soil layer by erosion. Erosion was estimated by comparing the δ13C profiles in the soils under forest and under plantations. The estimated erosion was the strongest in oil palm (35 ± 8 cm) and rubber (33 ± 10 cm) plantations. The 13C enrichment of SOC used as a proxy of its turnover indicates a decrease of SOC decomposition rate in the Ah horizon under oil palm plantations after forest conversion. Nonetheless, based on the lack of C input from litter, we expect further losses of SOC in oil palm plantations, which are a less sustainable land use compared to rubber plantations. We conclude that δ13C depth profiles may be a powerful tool to disentangle soil erosion and SOC mineralization after the conversion of natural ecosystems conversion to intensive plantations when soils show gradual increase of δ13C values with depth.

We investigated the effects of three elevated atmospheric CO2 levels on a Populus deltoides plantation at Biosphere 2 Laboratory in Oracle Arizona. Stable isotopes of carbon have been used as tracers to separate the carbon present before the CO2 treatments started (old C), from that fixed after CO2 treatments began (new C). Tree growth at elevated [CO2] increased inputs to soil organic matter (SOM) by increasing the production of fine roots and accelerating the rate of root C turnover. However, soil carbon content decreased as [CO2] in the atmosphere increased and inputs of new C were not found in SOM. Consequently, the rates of soil respiration increased by 141% and 176% in the 800 and 1200 μL L?1 plantations, respectively, when compared with ambient [CO2] after 4 years of exposure. However, the increase in decomposition of old SOM (i.e. already present when CO2 treatments began) accounted for 72% and 69% of the increase in soil respiration seen under elevated [CO2]. This resulted in a net loss of soil C at a rate that was between 10 and 20 times faster at elevated [CO2] than at ambient conditions. The inability to retain new and old C in the soil may stem from the lack of stabilization of SOM, allowing for its rapid decomposition by soil heterotrophs.

Incorporating crop residues and biochar has received increasing attention as tools to mitigate atmospheric carbon dioxide (CO2) emissions and promote soil carbon (C) sequestration. However, direct comparisons between biochar, torrefied biomass, and straw on both labile and recalcitrant soil organic matter (SOM) remain poorly understood. In this study, we explored the impact of biochars produced at different temperatures and torrefied biomass on the simple C substrates (glucose, amino acids), plant residues (Lolium perenne L.), and native SOM breakdown in soil using a 14C labeling approach. Torrefied biomass and biochars produced from wheat straw at four contrasting pyrolysis temperatures (250, 350, 450, and 550 °C) were incorporated into a sandy loam soil and their impact on C turnover compared to an unamended soil or one amended with unprocessed straw. Biochar, torrefied biomass, and straw application induced a shift in the soil microbial community size, activity, and structure with the greatest effects in the straw‐amended soil. In addition, they also resulted in changes in microbial carbon use efficiency (CUE) leading to more substrate C being partitioned into catabolic processes. While overall the biochar, torrefied biomass, and straw addition increased soil respiration, it reduced the turnover rate of the simple C substrates, plant residues, and native SOM and had no appreciable effect on the turnover rate of the microbial biomass. The negative SOM priming was positively correlated with biochar production temperature. We therefore ascribe the increase in soil CO2 efflux to biochar‐derived C rather than that originating from SOM. In conclusion, the SOM priming magnitude is strongly influenced by both the soil organic C quality and the biochar properties. In comparison with straw, biochar has the greatest potential to promote soil C storage. However, straw and torrefied biomass may have other cobenefits which may make them more suitable as a CO2 abatement strategy.

Global climate warming may induce a positive feedback through increasing soil carbon (C) release to the atmosphere. Although warming can affect both C input to and output from soil, direct and convincing evidence illustrating that warming induces a net change in soil C is still lacking. We synthesized the results from field warming experiments at 165 sites across the globe and found that climate warming had no significant effect on soil C stock. On average, warming significantly increased root biomass and soil respiration, but warming effects on root biomass and soil respiration strongly depended on soil nitrogen (N) availability. Under high N availability (soil C:N ratio < 15), warming had no significant effect on root biomass, but promoted the coupling between effect sizes of root biomass and soil C stock. Under relative N limitation (soil C:N ratio > 15), warming significantly enhanced root biomass. However, the enhancement of root biomass did not induce a corresponding C accumulation in soil, possibly because warming promoted microbial CO2 release that offset the increased root C input. Also, reactive N input alleviated warming-induced C loss from soil, but elevated atmospheric CO2 or precipitation increase/reduction did not. Together, our findings indicate that the relative availability of soil C to N (i.e., soil C:N ratio) critically mediates warming effects on soil C dynamics, suggesting that its incorporation into C-climate models may improve the prediction of soil C cycling under future global warming scenarios.

Cellulosic biofuel from forest thinning operations is a potential renewable energy source in regions with overstocked forests such as those in western United States. However, it is possible that biomass removal can deplete nutrients from soil, which can alter soil respiration (Rs) and exoenzyme properties, and potentially impact tree growth. This study evaluates the impact of biomass removal on Rs and exoenzyme properties and the capacity of soil amendments to counteract any potential effects. At two study locations, we created four post‐thinning biomass retention levels: full biomass removal (0×), full biomass retention (1×), double biomass retention (2×), and a no‐thin treatment. Four soil amendment treatments were applied to each biomass retention level: N fertilizer (F), biochar (B), fertilizer plus biochar (FB), and an untreated control (C). We evaluated treatment effects on Rs and activity of four exoenzymes to represent C‐cycling, N‐release, and P‐release processes. Biomass retention levels had no effect on Rs (p = .42) or exoenzyme activities (p > .29). Variation in exoenzyme activity was explained by location, season, soil organic matter, soil moisture content, and temperature. Variation in Rs was explained by the same variables, in addition to C‐cycling exoenzyme activity and soil pH. Soil amendments had no effect on exoenzyme activities (p > .49), and no main effect on Rs (p = .48), though amendments influenced Rs differently at each location (p = .02). Short‐term findings suggest small‐diameter biomass removal for cellulosic biofuel production will not impact Rs and exoenzyme properties, and paired with our tree growth study, provide evidence that biofuel systems are a feasible renewable energy source in the western North America.

The mechanistic understanding of warming and nitrogen (N) fertilization, alone or in combination, on microbially mediated decomposition is limited. In this study, soil samples were collected from previously harvested switchgrass (Panicum virgatum L.) plots that had been treated with high N fertilizer (HN: 67 kg N ha?1) and those that had received no N fertilizer (NN) over a 3‐year period. The samples were incubated for 180 days at 15 °C and 20 °C, during which heterotrophic respiration, δ13C of CO2, microbial biomass (MB), specific soil respiration rate (Rs: respiration per unit of microbial biomass), and exoenzyme activities were quantified at 10 different collections time. Employing switchgrass tissues (referred to as litter) with naturally abundant 13C allowed us to partition CO2 respiration derived from soil and amended litter. Cumulative soil respiration increased significantly by 16.4% and 4.2% under warming and N fertilization, respectively. Respiration derived from soil was elevated significantly with warming, while oxidase, the agent for recalcitrant soil substrate decomposition, was not significantly affected by warming. Warming, however, significantly enhanced MB and Rs indicating a decrease in microbial growth efficiency (MGE). On the contrary, respiration derived from amended litter was elevated with N fertilization, which was consistent with the significantly elevated hydrolase. N fertilization, however, had little effect on MB and Rs, suggesting little change in microbial physiology. Temperature and N fertilization showed minimal interactive effects likely due to little differences in soil N availability between NN and HN samples, which is partly attributable to switchgrass biomass N accumulation (equivalent to ~53% of fertilizer N). Overall, the differential individual effects of warming and N fertilization may be driven by physiological adaptation and stimulated exoenzyme kinetics, respectively. The study shed insights on distinct microbial acquisition of different substrates under global temperature increase and N enrichment.

Tropical forest conversion to agricultural land leads to a strong decrease of soil organic carbon (SOC) stocks. While the decrease of the soil C sequestration function is easy to measure, the impacts of SOC losses on soil fertility remain unclear. Especially the assessment of the sensitivity of other fertility indicators as related to ecosystem services suffers from a lack of clear methodology. We developed a new approach to assess the sensitivity of soil fertility indicators and tested it on biological and chemical soil properties affected by rainforest conversion to plantations. The approach is based on (non-)linear regressions between SOC losses and fertility indicators normalized to their level in a natural ecosystem. Biotic indicators (basal respiration, microbial biomass, acid phosphatase), labile SOC pools (dissolved organic carbon and light fraction) and nutrients (total N and available P) were measured in Ah horizons from rainforests, jungle rubber, rubber (Hevea brasiliensis) and oil palm (Elaeis guineensis) plantations located on Sumatra. The negative impact of land-use changes on all measured indicators increased in the following sequence: forest < jungle rubber < rubber < oil palm. The basal respiration, microbial biomass and nutrients were resistant to SOC losses, whereas the light fraction was lost stronger than SOC. Microbial C use efficiency was independent on land use. The resistance of C availability for microorganisms to SOC losses suggests that a decrease of SOC quality was partly compensated by litter input and a relative enrichment by nutrients. However, the relationship between the basal respiration and SOC was non-linear; i.e. negative impact on microbial activity strongly increased with SOC losses. Therefore, a small decrease of C content under oil palm compared to rubber plantations yielded a strong drop in microbial activity. Consequently, management practices mitigating SOC losses in oil palm plantations would strongly increase soil fertility and ecosystem stability. We conclude that the new approach enables quantitatively assessing the sensitivity and resistance of diverse soil functions to land-use changes and can thus be used to assess resilience of agroecosystems with various use intensities.

A novel procedure was developed for direct quantitative isolation of microbial DNA from soil. This technique was used to evaluate microbial DNA pools in soils of contrasting types (chernozems and brown forest soils) under different anthropogenic loads. A strong correlation was found between microbial biomass and DNA contents in soils of different types (R 2= 0.799). The ratio of soil CO2 emission rate to the amount of extractable DNA in the soil was shown to reflect the physiological state of the soil microbial community; this ratio can be used as an ecophysiological parameter similarly to the metabolic quotient qCO2.

Soil organic matter (SOM) mineralization processes are central to the functioning of soils in relation to feedbacks with atmospheric CO2 concentration, to sustainable nutrient supply, to structural stability and in supporting biodiversity. Recognition that labile C‐inputs to soil (e.g. plant‐derived) can significantly affect mineralization of SOM (‘priming effects’) complicates prediction of environmental and land‐use change effects on SOM dynamics and soil C‐balance. The aim of this study is to construct response functions for SOM priming to labile C (glucose) addition rates, for four contrasting soils. Six rates of glucose (3 atm% 13C) addition (in the range 0–1 mg glucose g?1 soil day?1) were applied for 8 days. Soil CO2 efflux was partitioned into SOM‐ and glucose‐derived components by isotopic mass balance, allowing quantification of SOM priming over time for each soil type. Priming effects resulting from pool substitution effects in the microbial biomass (‘apparent priming’) were accounted for by determining treatment effects on microbial biomass size and isotopic composition. In general, SOM priming increased with glucose addition rate, approaching maximum rates specific for each soil (up to 200%). Where glucose additions saturated microbial utilization capacity (>0.5 mg glucose g?1 soil), priming was a soil‐specific function of glucose mineralization rate. At low to intermediate glucose addition rates, the magnitude (and direction) of priming effects was more variable. These results are consistent with the view that SOM priming is supported by the availability of labile C, that priming is not a ubiquitous function of all components of microbial communities and that soils differ in the extent to which labile C stimulates priming. That priming effects can be represented as response functions to labile C addition rates may be a means of their explicit representation in soil C‐models. However, these response functions are soil‐specific and may be affected by several interacting factors at lower addition rates.
