Biochemical evidence was obtained for an increase in acid phosphatase activity in the larval fat body of Calliphora erythrocephala during larval and pharate pupal instars. This observation is in conflict with published data indicating a decreasing enzyme activity in late third stage larvae. Centrifugation and filtration studies showed that the pH of the homogenisation medium has a strong influence on the solubilisation of acid phosphatase and its distribution in homogenate components. Differences in biochemical techniques including the pH value may explain the discrepancy between the published results and the present findings.The observed increase in acid phosphatase activity is related to the activity of the lysosomal system in the period immediately preceding pupal-adult apolysis.  相似文献

Before both larval moults in Drosophila melanogaster, the chitin in the cuticle is digested to a significant degree by the moulting fluid. A spurt of chitinase activity appears just before each ecdysis, drops sharply after the first ecdysis, and begins to rise again just about the time that chitin degradation becomes evident. The level of enzyme activity/mg of soluble protein reached just before the second ecdysis is about twice that reached before the first, and this declines gradually after the ecdysis until puparium formation. Chitinase activity is measured with a viscometric assay on a chitosan substrate.The enzyme activity is stable, with no loosely bound cofactor. Data also exist supporting the presence of more than one enzyme fraction in Drosophila with chitinase activity.  相似文献

The alfalfa weevil Hypera postica is a serious economic pest in most alfalfa grown in many countries worldwide. Digestive α-amylase and pectinase activities of larvae were investigated using general substrates. Midgut extracts from larvae showed an optimum activity for α-amylase against starch at acidic pH (pH 5.0). α-Amylase from larval midgut was more stable at mildly acidic pH (pH 5–6) than highly acidic and alkaline pH. The enzyme showed its maximum activity at 35°C. α-Amylase activity was significantly decreased in the presence of Ca2+, Mg2+ and sodium dodecylsulfate. On the contrary, K+ and Na+ did not significantly affect the enzyme activity. Zymogram analysis revealed the presence of one band of α-amylase activity in in-gel assays. Pectinase activity was assayed using agarose plate and colorimetric assays. Optimal pH for pectinase activity in the larval midgut was determined to be pH 5.0. Pectinase enzyme is more stable at pH 4.0–7.0 than highly acidic and alkaline pH. However, the enzyme was more stable at slightly acidic pH (pH 6.0) when incubation time increased. Maximum activity for the enzyme incubated at different temperatures was observed to be 40°C. Optimum pH activity for α-amylase and pectinase is not completely consistent with the pH prevailing in the larval midgut. This is the first report of the presence of pectinase activity in H. postica.  相似文献

α-Amylase activity appears during gut differentiation of plutei of the sand dollar Dendraster excentricus. The activity increases coincidentally with that of a β-1, 3-glucanase. The α-amylase appears to have maltase activity. Glucose in the surrounding seawater depresses the increase in β-glucanase activity but does not affect the normal α-amylase increase. This suggests that the glucose repression of β-glucanase activity may be quite specific and not result from some general effect of glucose on metabolism. Sodium dodecyl sulfate-acrylamide gel electrophoresis resolves the larval α-amylase into components of 52,000 and 76,000 molecular weight. The α-amylase from the intestines of adult sand dollars resolves as a single component of 57,000 molecular weight. The pH curves for adult and larval α-amylases are also different and it is probable that adult and larval α-amylases are different proteins.  相似文献

The activity of the lysosomal marker enzyme acid phosphatase in the larval fat body of Calliphora erythrocephala increases during development, but not at the same rate throughout the tissue. During the feeding stage, the posterior region has a higher acid phosphatase activity than the anterior region. When the larvae cease feeding on the 5th day of development, the acid phosphatase activity of the inactive anterior lobe increases rapidly in a mosaic-cell pattern. When 4-day-old feeding stage larvae are starved, this increase occurs one day earlier than normally. After the emptying of the gut, the acid phosphatase activity of all the anterior cells both in normal and in starved larvae exceeds that of those in the posterior region.Transplantation experiments indicate that the induction of acid phosphatase activity in the fat body during normal development, especially in the anterior region, is caused by a change in the internal environment when the larvae cease feeding. Both RNA and protein synthesis are involved in this induction process. Inductive factors are present in 5-day-old larvae as well as during formation of the puparium. The competence of the feeding-stage fat cells to develop high acid phosphatase activity is acquired before the actual induction takes place.  相似文献

During the programmed cell death of intersegmental muscles in Manduca sexta, the activities of several enzymes begin to decrease at different times. The rate of loss of these activities also varies among organelles and is dependent on other factors. Myofilament proteins begin to disappear as early as 4 days before the emergence of the adult. An enzyme digesting ATP at acid pH increases 3-fold in total activity, and more than 10-fold in relative activity over the same period.  相似文献

Ethanolamine kinase was partially purified from the larvae of Culex pipiens fatigans and its properties were studied. The enzyme was separated from choline kinase by acetic acid precipitation at pH 5.0 of a 13,000g supernatant of the larval homogenate. Alkaline phosphatase activity was removed from the enzyme preparation by the acid treatment followed by ammonium sulfate fractionation. The enzyme was localized in the cytosolic fraction and had a requirement for Mg2+ as a cofactor. The Km values for ethanolamine and ATP were 4 × 10?4 and 1.54 × 10?4m, respectively. The affinity of the enzyme for nucleotide triphosphates was in the order, ATP > ITP > GTP while UTP and CTP were poorly utilized. p-Chloromercuribenzoate and N-ethylmaleimide inhibited the enzyme activity and reduced glutathione protected the enzyme from their inhibition. Choline and serine had no effect on the enzyme activity. The enzyme had a molecular weight of 44, 000 daltons as determined by gel filtration chromatography. Eggs contained the highest specific activity of the enzyme while adult insects had the highest total enzyme activity.  相似文献

Purified chicken intestinal alkaline phosphatase is active at pH 8 to 9, but becomes rapidly inactivated with change of pH to 6 or less. Also, a solution of the inactivated enzyme at pH 4.5 rapidly regains its activity at pH 8. In the range of pH 6 to 8 a solution of purified alkaline phosphatase consists of a mixture of active and inactive enzyme in equilibrium with each other. The rate of inactivation at lower pH and of reactivation at higher pH increases with increase in temperature. Also, the activity at equilibrium in the range of pH 6 to 8 increases with temperature so that a solution equilibrated at higher temperature loses part of its activity on cooling, and vice versa, a rise in temperature shifts the equilibrium toward higher activity. The kinetics of inactivation of the enzyme at lower pH and the reactivation at higher pH is that of a unimolecular reaction. The thermodynamic values for the heat and entropy of the reversible inactivation and reactivation of the enzyme are considerably lower than those observed for the reversible denaturation of proteins. The inactivated enzyme at pH 4 to 6 is rapidly reactivated on addition of Zn ions even at pH 4 to 6. However, zinc ions are unable to replace magnesium ions as cocatalysts for the enzymatic hydrolysis of organic phosphates by alkaline phosphatase.  相似文献

The larval midgut of the Colorado beetle, Leptinotarsa decemlineata contains cathepsin B, D and H activity detected by use of haemoglobin, synthetic substrates specific for each enzyme, pH at which the substrate was maximally hydrolysed and effects of potential activators and inhibitors on proteolytic activity. Cysteine proteases cathepsin B, and H were activated by thiol compounds and inhibited by iodoacetamide, TLCK and epoxysuccinyl-leucyl-amido(guanidino)butane (E-64) a cysteine specific proteinase inhibitor. Cathepsin B was distinguished from H by hydrolysis of benzoyloxycarbonyl-Ala-Arg-Arg-methoxynaphthylamide, a cathepsin B specific substrate and inhibition of substrate hydrolysis by leupeptin. Cathepsin H activity, detected using the specific substrate arginine-naphthylamide, was insensitive to leupeptin. Cathepsin D had maximal activity at pH 4.5 and was inhibited by pepstatin, an aspartic proteinase inhibitor.  相似文献

Papilio machaon (Linnaeus) (Lepidoptera: Papilionidae) feeds on different plant species of the family Apiaceae. It also rarely feeds on the Rutaceae family such as rue (Ruta chalepensis) and fennel. In the current study, α-amylase activity of the larval midgut was investigated and characterised. Also, the effect of the wheat seed extracts on the larval midgut was explored. Results showed that the amylase activity was present in the midgut. Gel assays showed that more than one amylase band (three) was present in the larval midgut showing their importance in the insect feeding. Characterisation of the amylase showed that this enzyme was active in a wide range of the pHs. However, optimal pH for the enzyme activity was alkaline pH (pH 8.0). The effect of wheat seed extracts on the amylase activity showed that the insect amylase is highly sensitive to wheat protein extract. These results showed that wheat seed extract have a good potential to be explored more in order to purify its products and these products could be used in IPM programme in order to combat insect pest.  相似文献

Highly purified pyruvic decarboxylase (EC 4.1.1.1) from wheat germ catalyses the decarboxylation of hydroxypyruvate. A kinetic analysis of the activity of the enzyme with pyruvate and hydroxypyruvate as substrates suggests that a single enzyme is involved. The kinetics of decarboxylation are autocatalytic. The time lag before maximum activity is reached is affected by the concentration of hydroxypyruvate and the pH. The question whether or not hydroxypyruvate is a natural substrate for the enzyme remains unresolved, but it may be significant that at physiological pH (ca 7.5) the enzyme shows optimum activity with hydroxypyruvate, but negligible activity with pyruvate.  相似文献

Midgut and fatbody mitochondria from fifth larval instar Manduca sexta display a membrane-associated transhydrogenase that catalyzes a reversible hydride ion transfer between NADP(H) and NAD(H). The NADPH-forming activity occurs as a nonenergy- or energy-linked activity with energy for the latter derived from either electron transport-dependent NADH or succinate utilization, or ATP hydrolysis by Mg++-dependent ATPase. During the ten-day developmental period preceding the larval-pupal molt (fifth larval instar), significant peaks in the mitochondrial transhydrogenase activities of midgut and fatbody tissues were noted and these peaks were coincident with the onset of wandering behavior and with the fifty-fold increase in ecdysone 20-monooxygenase (E20-M) activity previously reported for M. sexta midgut. Since E20-M preferentially uses NADPH in catalyzing ecdysone conversion to the physiologically active molting hormone, 20-hydroxyecdysone, the physiological and developmental significance of the mitochondrial, NADPH-forming energy-linked transhydrogenations were made apparent. Moreover, that the increases in all transhydrogenase activities resulted from de novo enzyme synthesis were indicated by the cycloheximide-dependent reductions in these activities.  相似文献

When a bleached strain of Euglena is maintained in a medium containing very low con centrations of phosphate, the acid phosphatase activity increases. The increase in acid phosphatase activity is prevented by Actinomycin D and by p-fluorophenylalanine (PFA), indicating that the increased activity is due to de novo synthesis of acid phosphatase. When phosphate is replenished, the acid phosphatase activity decreases to the level characteristic of uninduced cells before there is any appreciable cell division. When cell division resumes in the presence of PFA, the level of acid phosphatase activity remains approximately constant. This indicates that there are two different phosphatases: a constitutive enzyme, whose synthesis is insensitive to the presence of PFA, and an induced enzyme, whose synthesis is sensitive to PFA. These enzymes are not equally sensitive to changes in pH and in fluoride concentration, thus permitting them to be assayed individually in whole toluene-treated cells. Induced cells also acquire the ability to remove phosphate from the medium very rapidly.  相似文献

Clostridium beijerinckii (“Clostridium butylicum”) NRRL B592 and NRRL B593 were grown in batch cultures without pH control. The use of more sensitive and accurate procedures for the determination of solvents in cultures led to the recognition of the onset of solvent production about 2 h earlier than the previously assigned point and at a higher culture pH for both strains. Reliable assays for solvent-forming enzyme activities in cell extracts have also been developed. The results showed that activities of solvent-forming enzymes in strain NRRL B592 started to increase about 1 h before the measured onset of solvent production and that the increase in activities of solvent-forming enzymes was not simultaneous. The degree of increase of these enzyme activities for both strains ranged from 2- to 165-fold, with acetoacetate decarboxylase and butanol-isopropanol dehydrogenase showing the largest activity increases. However, the pattern of increase of enzyme activities differed significantly in the two strains of C. beijerinckii. When an increase in solvent-forming enzyme activities was first detected in strain NRRL B592, the culture pH was at 5.7 and the concentrations of total acetic and butyric acids were 5.2 and 3.6 mM, respectively. For strain NRRL B593, the corresponding pH was 5.5. Thus, the culture conditions immediately preceding the expression of solvent-forming enzyme activities differed significantly from those that have been correlated with the production of solvents at later stages of growth.  相似文献

When plasma from third instar larvae of the fleshfly, Sarcophaga barbarta, was diluted tenfold with distilled water, lipoproteins precipitated out. After centrifuging, the water supernatant was rendered 30, 50, and 65% to ammonium sulphate, and it was found that the 50% fraction contained 95% of the tyrosinase activity in all the fractions, the enzyme being present in its inactive form or proenzyme. The proenzyme was activated by mixing it with activator isolated from the larval cuticle. After addition of activator there followed a lag period before the rapid phase of activation, the duration of the lag being dependent upon the concentration of both proenzyme and activator. The final activity attained was dependent upon the concentration of proenzyme but was independent of the activator concentration.The level of proenzyme in the plasma rose steadily throughout the third larval instar reaching a maximum in 7 day larvae, formation of the puparium commencing about 24 hr later, the rounded-off white stage (r.o.). At the r.o. and golden-brown stage (1 hr later) the level was still maximal, but 12 hr later at the dark-brown puparial stage no proenzyme was isolatable from the plasma, all the enzyme at this stage behaving as active enzyme.The vast majority (95%) of the proenzyme isolated from plasma in the larval stages and at the r.o. white stage was present in the 50% ammonium sulphate fraction, whereas 1 hr later at the golden-brown stage only 33% of the proenzyme was found in the 50% fraction, 62% now being found in the 65% fraction. At the dark-brown puparial stage 12 hr later, not only was there a further redistribution, but all the enzyme behaved as active enzyme. It is suggested that these changes in the distribution and behaviour of the proenzyme indicate that, in vivo, activation of the enzyme in the blood has taken place over the period r.o. white to the golden-brown to dark-brown puparial stage.  相似文献

The growth rate of insects may vary in response to shifty environments. They may achieve compensatory growth after a period of food restriction followed by ad libitum food, which may further affect the reproductive performance and lifespan of the resulting phenotypes. However, little is known about the physiological mechanisms associated with such growth acceleration in insects. The present study examined the metabolic rate, the antioxidant enzyme activity and the gene expression of adult Cryptolaemus montrouzieri (Coleoptera: Coccinellidae) after experiencing compensatory growth during its larval stages. Starved C. montrouzieri individuals achieved a similar developmental time and adult body mass as those supplied with ad libitum food during their entire larval stage, indicating that compensatory growth occurred as a result of the switch in larval food regime. Further, the compensatory growth was found to exert effects on the physiological functions of C. montrouzieri, in terms of its metabolic rates and enzyme activities. The adults undergoing compensatory growth were characterized by a higher metabolic rate, a lower activity of the antioxidant enzymes glutathione reductase, catalase, and superoxide dismutase and a lower gene expression of P450 and trehalase. Taken together, the results indicate that although compensatory growth following food restriction in early larval life prevents developmental delay and body mass loss, the resulting adults may encounter physiological challenges affecting their fitness.  相似文献

We report on the presence of arachidonic acid in larval and adult tissues of the primary screwworm, Cochliomyia hominivorax and of the secondary screwworm, C. macellaria. Arachidonic acid is present in the phospholipids of whole animal extracts of both species. This fatty acid appears to be accumulated during the larval stages, because proportions of arachidonic acid were higher in adults than in larvae. These insects probably obtain the arachidonic acid from dietary phospholipids. We also report on a phospholipase A2 activity in midgut preparations from third instars of the primary screwworm. Phospholipase A2 is responsible for hydrolyzing fatty acids from the sn-2 position of dietary phospholipids to release essential fatty acids. The screwworm enzyme is similar to mammalian digestive phospholipase A2s because it depends on calcium for high catalytic activity, it is sensitive to the site-specific inhibitor oleyloxyethylphosphorylcholine, and it interacts with heparin. We further characterized the screwworm midgut phospholipase A2 by altering the reaction conditions, including reaction time, radioactive substrate concentration, protein concentration, pH and temperature. We speculate that the biological significance of this enzyme relates to acquiring essential fatty acids, including arachidonic acid, from dietary phospholipids.  相似文献

A simple and rapid extraction procedure was developed to determine simultaneously the molting hormone (MH) and juvenile hormone (JH) activity in a single insect tissue sample. From the onset of the last larval stage to adult eclosion of the greater wax moth, Galleria mellonella, three JH peaks were noted: at the time of the sixth larval ecdysis, 1 day before the seventh larval ecdysis, and at the time of adult eclosion. Three MH peaks were recorded for the male: at 1 day before the sixth larval ecdysis, 1 day before the seventh larval ecdysis, and 2 days after pupation. In the female, a fourth peak was shown at the time of adult eclosion. This fourth peak exhibits the highest molting hormone activity of all samples, 1600 Musca units/g of fresh tissue or an equivalent of 5.6 μg/g of ecdysterone. Eighty per cent of this MH accumulated in the ovary. The significance of MH and JH titers as related to the endocrine regulation of development is discussed in the light of this finding.  相似文献

Juvenile hormone esterase activity has been found during the intramoult period of each larval stadium in Trichoplusia ni. The activity is indistinguishable from that occurring during the final larval stadium, on the basis of its four isoelectric forms and kinetic data. The 4th- to 5th-instar intramoult peak in activity also occurs in other Lepidoptera (Heliothis virescens, Spodoptera exigua, Manduca sexta and Hyphantria cunea). Further, the final species also possessed a peak of activity during the intramoult period to the penultimate larval instar. The findings have important implications for the current concept that the function of juvenile hormone esterase is, by reason of its anti-juvenile hormone action, an enzyme of the last larval instar which enables metamorphosis to begin.  相似文献

The starch hydrolyzing enzyme amyloglucosidase (AMG) from Rhizopus was immobilized onto the protonated salt (TS) and basic (TB) forms of chemically synthesized poly(o-toluidine) (POT) using adsorption and covalent binding. The polymers were activated with glutaraldehyde prior to covalent bonding. The immobilization efficiency was affected by the pH of the immobilization medium, contact time and amount of enzyme. After immobilization, the pH and temperature were changed to conditions under which the enzyme is most active. Immobilized AMG was more stable with respect to changes in pH and increases in temperature compared to free AMG. The immobilized enzyme retained high catalytic activity after multiple uses and showed enhanced stability with storage compared to free enzyme.  相似文献
