Viola uliginosa is an endangered species throughout its range in Central Europe. Based on an unusual combination of morphological characters, including a reported lack of cleistogamous (CL) flowers, it was previously placed in the monotypic subsection Repentes, section Viola. Available individual phylogenetic analyses identify V. uliginosa as a lineage deeply embedded in section Viola, but hitherto its exact position was unknown. Here we confirm the occurrence of cleistogamy in V. uliginosa. Unlike other species of section Viola, Vuliginosa produced both chasmogamous and CL flowers at the same time, not at different times of the year (seasonal cleistogamy). The two flower types differed conspicuously in morphology and microstructure, especially in size and shape of the pistil and stamens as confirmed by scanning electron microscopy. Secondly, we resolved the phylogenetic position of Vuliginosa as a lineage deeply embedded in subsection Rostratae by means of a multigene multispecies coalescent analysis using *BEAST. In this analysis, Vmirabilis and Vuliginosa were successive sisters to the rest of subsection Rostratae and, because allopolyploids exist between Vmirabilis and the last one, subsection Rostratae needs to comprise all these taxa to be monophyletic. Sequences of the nuclear region ITS and the variable molecular markers ISSR were used to estimate the phylogeny of the section Melanium of the genus Viola. We confirm that the so-called pansies form a derived and monophyletic group. Two floral characters, the upturned side petals and the large size of pollen grains appear to be synapomorphies in Melanium. The Melanium species are very closely related, as shown by the reduced genetic variation compared to the other sections of Viola. Our analyses suggest x=5 or x=7 as the possible base chromosome number of the section Melanium. Polyploidy and hybridization would have played an important role in the evolutionary history of this clade resulting in a wide range of chromosome number. The low genetic differentiation and the complex cytological evolution suggest that diversification in Melanium is the result of a reticulate evolution and rapid radiation in Europe and Northern Africa.  相似文献

Viola tricolor is a pseudometallophyte covering heavy-metal-polluted and non-polluted areas. The species is a member of the evolutionarily young sect. Melanium of Viola. In this study, we sought to determine whether the karyotype of V. tricolor is stable with respect to chromosome structure or is altered depending on environmental conditions (heavy-metal-polluted vs. non-polluted areas). We established the karyotypes of plant material originating from a Zakopane meadow (non-metallicolous population) and from the Bukowno mine waste heap (metallicolous population), showing evident interpopulation differentiation in chromosome type (2M+20 m+2sm+2st vs. 18 m+8sm), in the number, size, and distribution of rDNA loci (25S and 5S), and also in chromosome mutations, mainly fission of chromosomes into acentric fragments and translocation of the fragments. Variable numbers of both 25S and 5S rDNA loci were distributed at different positions of the chromosomes and not on specific pairs of chromosomes. The results clearly indicate that the karyotype of V. tricolor results from the unstable genetic structure of the species. This character, typical for relatively young evolutionary groups, proves its membership to the Melanium section considered to be young within the genus Viola.  相似文献

Abstract

This integrated study provides new insights into pollen and seed morphology and pollen heteromorphism of four closely related annual taxa of Viola sect. Melanium. The plant material, both fresh and dried, was collected in Italy and studied using light and scanning electron microscope. Palynological data for V. hymettia together with a detailed comparative analysis of seed morphology and micromorphology of the four species are reported for the first time. Results of this work highlight some pollen and seed features as useful diagnostic characters. The pollen size proves to be of diagnostic value to easily separate V. kitaibeliana, having the smallest pollen grains, from the others, especially from V. arvensis with the largest ones. Exine ornamentation is microreticulate, showing no relevant differences among species. We can partially confirm the diagnostic value of the prevailing pollen morph as it can be useful only for V. arvensis (five-aperturate) versus V. tricolor and V. hymettia (four-aperturate). The macro- and micromorphology of seeds provide additional useful distinguishing characters. Particularly, seed size was found to be a good delimitating character, especially to distinguish V. kitaibeliana (with the smallest seeds) from V. arvensis, and easy to be measured with no need of particular equipment.  相似文献

  • Viola reichenbachiana (2= 4= 20) and Vriviniana (2= 8= 40) are closely related species widely distributed in Europe, often sharing the same habitat throughout their overlapping ranges. It has been suggested in numerous studies that their high intraspecific morphological variability and plasticity might have been further increased by interspecific hybridisation in contact zones, given the sympatry of the species and the incomplete sterility of their hybrid. The aims of this study were to: (i) confirm that V. reichenbachiana and Vriviniana have one 4x genome in common, and (ii) determine the impact of hybridisation and introgression on genetic variation of these two species in selected European populations.
  • For our study, we used 31 Viola populations from four European countries, which were analysed using AFLP and sequencing of a variable plastid intergenic spacer, trnH‐psbA.
  • Our analysis revealed that V. reichenbachiana exhibited larger haplotype diversity, having three species‐specific haplotypes versus one in Vriviniana. The relationships among haplotypes suggest transfer of common haplotypes into Vriviniana from both V. reichenbachiana and hypothetically the other, now extinct, parental species. AFLP analysis showed low overall genetic diversity of both species, with Vriviniana showing higher among‐population diversity. None of the morphologically designated hybrid populations had additive AFLP polymorphisms that would have indicated recent hybridisation. Also, kinship coefficients between both species did not indicate gene flow. Vriviniana showed significant population subdivision and significant isolation by distance, in contrast to V. reichenbachiana.
  • The results indicate lack of gene flow between species, high influence of selfing on genetic variability, as well as probably only localised introgression toward Vriviniana.

Abstract

The authors report the use of nuclear [internal transcribed spacer (ITS) sequences 1 and 2] and chloroplast DNA (trnS-trnG intergenic spacer sequences) in Viola subsect. Viola for separate tracking of maternal lineages and detecting dubious herbarium specimens. A phylogenetic investigation carried out on ITS data after removal of all material with possible hybrid origin showed that V. hirta is a monophyletic unit, whereas V. odorata includes at least V. collina and V. jaubertiana, as well as three sequences of V. alba subsp. dehnhardtii from literature. In some, among the sampled individuals, the morphological attribution to one species is contradicted by nuclear DNA, which indicated a wide distance from other non-specific individuals from different locations and closer proximity to a different species. Chloroplast DNA data for the same individuals, on the contrary concurred with morphological evidence. These findings confirm document univocal correlation between our chosen chloroplast sequences and the studied taxa at species or subspecies level; these sequences have the appropriate variability range to be employed for detection of the maternal lineage of unknown Viola samples.  相似文献

Despite a number of recent molecular phylogenetic studies on Phlomoides, in terms of trichome morphology the genus is still among the most poorly studied taxa in the family Lamiaceae. In order to test the utility of trichome characters for delimitation of sections, subsection and species of Phlomoides, we examined trichomes of 64 species representing all recognized sections and subsections using stereomicroscopy and scaning electron microscopy. Two basic types of trichomes could be identified: non‐glandular and glandular. Both trichome types can be simple or branched. The glandular trichomes were sessile, short stalked or long stalked. Different kinds of branched trichomes were observed in most species of P. sect. Phlomoides, i.e. symmetrically stellate, stellate with a central long branch, bi‐ or trifurcate. The species of P. sect. Filipendula were mostly covered by simple trichomes. Moreover, variation in trichome characters appears to have particular value, not only for the classification at sectional or subsectional rank, but also for delimitation of species within each section. For example, all studied species of P. subsect. Fulgentes are characterized by various kinds of stellate trichomes, while the trichome variability in P. subsect. Tetragonae was sufficiently high for species discrimination. An ancestral character state reconstruction was performed in order to investigate the evolution of trichome types and it revealed the following evolutionary trends in trichome characters of Phlomoides: 1) branched trichomes are primitive in Phlomoides as compared to simple ones, 2) long simple non‐glandular trichomes are derived as compared to short simple ones and 3) the presence of stalked glandular trichomes is advanced as compared to subsessile or sessile ones.  相似文献

Comprising ca. 200 species, Saxifraga sect. Ciliatae is the most species-rich section of Saxifraga s.str., whose center of diversity is in the Tibeto-Himalayan region. The infra-sectional classification of sect. Ciliatae is still in debate due to the high level of species richness, as well as remarkable variations of habitat, morphology, physiology and life cycles. Subdivisions of this section proposed in various taxonomic systems have not been adequately tested in previous phylogenetic studies, partly due to low taxonomic sampling density, but also to the use of few DNA markers. In order to achieve a more robust infra-sectional classification of sect. Ciliatae, complete chloroplast genomes of 94 taxa from this section were analyzed, of which 93 were newly sequenced, assembled and annotated. The length of the 94 plastomes of sect. Ciliatae taxa range from 143,479 to 159,938 bp, encoding 75 to 79 unique protein-coding genes (PCGs). Analyses of the 94 plastomes revealed high conservation in structural organization, gene arrangement, and gene content. Gene loss and changes of IR boundaries were detected but in extremely low frequency. The molecular phylogenetic tree from concatenated PCGs and complete chloroplast genome sequences exhibited high resolution and support values and confirms that sect. Ciliatae is monophyletic. Three well-supported clades were revealed within the section that agree relatively well with the subsectional taxonomy of Gornall (1987), but some minor modifications should be made. Firstly, the monotypic subsection Cinerascentes should be abandoned and its constituent species, Scinerascens, assigned to subsect. Gemmiparae. Secondly, subsections Rosulares and Serpyllifoliae should be merged and become subsect. Rosulares. Section Ciliatae thus comprises: subsect. Hirculoideae Engl. & Irmsch.; subsect. Rosulares Gornall; subsect. Gemmiparae Engl. & Irmsch.; subsect. Flagellares (C. B. Clarke) Engl. & Irmsch. and subsect. Hemisphaericae (Engl. & Irmsch.) Gornall.  相似文献

Forty-nine morphological characters were scored or measured on 44 populations (376 individuals) of Viola subsect. Viola from the West Carpathians and adjacent areas (Slovakia, Czech Republic, Austria and Hungary). The presence of six species, namely V. alba (represented by subsp. alba), V. ambigua, V. collina, V. hirta, V. odorata and V. suavis s.l. was revealed based on pollen fertility, cytological and morphometric analyses. The morphological characters traditionally used to delimit taxa within the subsection and those revealed by our study as most reliable are widely discussed. A key for identifying the taxa and most common hybrids of subsection Viola occurring in the West Carpathians is presented. Chromosome counting and flow cytometry were used to determine the ploidy levels of the populations studied. All individuals of V. alba subsp. alba, V. collina, V. hirta and V. odorata were tetraploid, while those of V. ambigua and V. suavis s.l. were octoploid.  相似文献

20 Bupleurum L. taxa were investigated for micromorphological typology of the fruit surfaces with SEM. Considerably different types of sculpturing at species and subspecies level and similarities between related taxa were observed. A dichotomic key was prepared using the diagnostic surface patterns. Reticulate (sect. Bupleurum), rugose (sect. Aristata Godron subsect. Aristata) and rugulose-colliculate-verrucate (subsect. Juncea Briq.) types of surface structures were distinguished. Micromorphological characteristics may be valuable additional taxonomical criteria for the delimitation of infrageneric groups of species.  相似文献

Vochysia palmirana is described and illustrated as a new species. It is a member of sect.Vochysiella subsect.Decorticantes. Morphological characters, AFLP markers and flow cytometry were used to investigate the morphological and genetic variability and differentiation of Viola reichenbachiana and V. riviniana in non‐metallicolous (NM) and metallicolous (M) populations. The aims were to clarify the taxonomic status of plants occurring in ore‐bearing areas, to determine any relationship in V. reichenbachiana and V. riviniana from sites not polluted with heavy metals, and to examine the genetic variability and differentiation of M and NM populations of both species. Multivariate analyses based on morphological characters showed significant differences between V. reichenbachiana and V. riviniana from non‐polluted sites, high levels of intra‐ and inter‐population variability, and the occurrence of inter‐specific hybrids. Plants from M populations showed hybrid characters but also fell within the range of V. riviniana or V. reichenbachiana. There were no significant differences in relative genome size between plants from polluted areas and V. riviniana from NM populations. Bayesian analysis of population genetic structure based on AFLP markers distinguished two main groups: V. reichenbachiana and V. riviniana together with the M populations. That analysis also revealed the occurrence of populations of inter‐specific hybrids from non‐polluted areas. Further Bayesian analysis of V. riviniana including NM and M populations separated all the studied M populations from NM populations. We conclude that plants forming the M populations are well adapted to a metal‐polluted environment, and could be considered as stabilised introgressive forms resulting from unidirectional (asymmetric) introgression toward V. riviniana.  相似文献

Adonis sect.Consiligo is split into three subsections based on morphology, growth pattern, pollen and fruit morphology. For all species distribution maps are provided and discussed with regard to their ecology and climatic requirements. These data reveal additional characters that support the classification of the taxa. Most species of the section grow in continental Eurasiatic steppes and mountain steppe habitats. Only the species of subsect.Amurenses grow in Far Eastern temperate forests and prefer a more oceanic climate. The taxonomic position of these strongly divergent species is supported by the distributional and ecological data. The evolution ofAdonis sect.Consiligo occurred under strong climatic control. During evolution the taxa could only slightly widen their ecological amplitude and, thus, ecology remained rather stable except that of the subsect.Amurenses. Ecological and distributional data can be used to validate taxonomy and phylogeny and reveal additional informations.  相似文献

通过ITS序列对21种中国外来苋属植物进行系统进化关系研究。通过ITS序列种间、种内遗传距离分析,发现苋属种间变异为0~0.055 1,种内变异为0~0.009 2。使用TAXON DNA软件分析ITS序列种间、种内变异的分布图看出规律,结果表明苋属ITS序列的种间变异适中,种间变异明显大于种内变异。采用最大似然法(ML)构建的系统树将中国苋属分为5或6个进化支(根据自展支持率取值不同)。异株苋亚属长芒苋和苋亚属刺苋聚类在一起,西部苋和糙果苋单独成为一个进化支。苋亚属中苋组苋亚组反枝苋和绿穗苋亚组鲍氏苋有着更近的亲缘关系,苋组苋亚组尾穗苋和绿穗苋亚组绿穗苋、繁穗苋等亲缘关系更近。白苋亚属分为2或3个类群,根据自展支持率取值不同,合被苋可以和白苋、北美苋并为一支,也可以单独成为一支。综上所述,该文认为苋属经典分类体系中3亚属或2~3组的分类地位不成立,建议中国苋属采取5组2亚组或6组2亚组的分类体系。5组2亚组分别由长芒苋组、糙果苋组、苋组(苋亚组和绿穗苋亚组)、白苋组和凹头苋组组成。其中,合被苋也可从白苋组分出,单独构成1组,形成6组2亚组的分类体系。表明该序列对苋属大部分种类分类效果较好,对西部苋和糙果苋复合群,绿穗苋复合群以及白苋亚属的分类价值不高。  相似文献

Studies of the anatomical structure of the stem and leaf, with special emphasis on the organization of the vascular system, has been carried out on 13 Aconitum species from Aconitum subgenera Aconitum, Anthora and Lycoctonum. All investigated species show a more or less mesomorphic anatomical structure, typical for other Ranunculaceae. Hence, these species have similar trilacunar three‐trace organization of the nodal vascular system. In the stem the vascular system is open (with weakly developed cambium) or closed collateral, and incomplete (consisting of the bundles were represented), large complete, middle‐size complete and small incomplete or with weakly developed xylem. The number of vascular bundles in petioles appear to have no taxonomical value. Nevertheless, it was found that the spatial organization of these vascular bundles in the petiole are of taxonomic importance. As a result, the investigated species can be divided into four main groups congruent with the current sectional and subgeneric division of the genus. The only exception was A. × cammarum (A. sect. Acomarum) in which the is identical to that of A. sect. Cammarum. The most primitive vascularization is found in A. anthora, while the most advanced one is found in A. variegatum. The highly differentiated and distinct nodal anatomy of A. anthora suggests a high, plausibly subgeneric, taxonomical rank of this species.  相似文献

Fresh leaves of 150 individuals from 15 populations representing five putative species of Stachys sect. Fragilicaulis, subsect. Fragiles distributed in the Zagros mountains in western Iran were analyzed for isoenzyme variation. Four enzyme systems (peroxidase, esterase, superoxide dismutase and ascorbate peroxidase) have been studied and a total of 32 bands representing seven putative loci have been analyzed by UPGMA using the Euclidean distances. UPGMA analysis resulted in the recognition of three distinct clusters among the considered populations. The average value of Shannon diversity index (H′) estimated for each population ranged from 0.05 to 2.17. The mean number of bands per isoenzyme ranges from 1.60 to 2.50. The value of Euclidean distance ranges from 0.85 to 3.79. The analysis of isoenzyme variation patterns suggests recognizing following species in Stachys sect. Fragilicaulis subsect. Fragiles: Stachys benthamiana (including Stachys megalodonta), Stachys kurdica (including Stachys ballotiformis) and Stachys asterocalyx. These data also suggest paying more attention to geographical distribution of taxa in Stachys for their taxonomic delimitation. This study provides another strong support for application of electrophoretic analysis of isoenzymes in taxonomy at species level.  相似文献

We present a comprehensive phylogeny derived from nuclear ribosomal DNA (nrDNA) for 214 samples representing 98 species and five varieties, including 44 species and five varieties native to China. Our collection of 25 species and five varieties (44 samples) covering all five sections of the genus (Comber) distributed in China also were included in the internal transcribed spacer (ITS) database. This study incorporates previous research with an emphasis on Chinese species, including the controversial subsection, Sinomartagon 5c Comber. In the phylogenetic tree obtained by maximum parsimony (PAUP) and maximum likelihood (RAxML) analyses, the samples were divided into four major groups. Our results suggest that the subsection (subsect.) 5c Comber should be classified into the true subsect. 5c and the section (sect.) Lophophorum. And the latter was divided into three subsections (subsect. Lophophorum I, subsect. Lophophorum II, and subsect. Lophophorum III). Based on molecular phylogenetic analysis and fluorescence in situ hybridization, we report that L. henryi and L. rosthornii are closely related, and we propose their classification into subsect. Leucolirion 6a. Our results support Comber’s subdivision of sect. Leucolirion, which was primarily based on bulb color. Chinese species were divided into five sections: sect. Martagon, sect. Archelirion, sect. Leucolirion, sect. Sinomartagon, and sect. Lophophorum. These findings contribute to our understanding of the phylogeny, origin, and classification of Lilium.  相似文献

  • The development of different pollen morphs by one specimen – pollen heteromorphism – occurs in ca. one‐third species of the genus Viola. Melanium section species (pansies) stand out in producing the widest range of apertures among Viola species. Aperture number decreases with elevation increase, and faster germination of five‐aperturate pollen as compared with three‐aperturate has previously been postulated.
  • We re‐examined pollen heteromorphism in the context of its viability, and made correlations with elevation (>1500 m a.s.l. versus <1500 m a.s.l.), soil type (metalliferous versus non‐metalliferous; MET versus NMET) and chromosome number based on selected study criteria of ca. 20% karyologically and morphologically strongly differentiated but genetically closely related pansies.
  • A total of 79% of analysed species were heteromorphic, forming three‐ to six‐colp or ate pollen per individual flower. Mean aperture number and pollen viability were not affected by soil type (MET versus NMET). Mean aperture number was also not influenced by elevation or species chromosome number. Positive correlations were established between aperture number and pollen viability, negative between pollen viability and elevation (increasing altitude of 100 m decreased pollen viability by 0.4%) and lack of correlation between chromosome number and pollen viability.
  • The varied frequencies of different pollen morphs among species are not under the general pressure of ecological conditions, as previously postulated for the species of Melanium section. Rather, this trait in pansies, similar to other floral characters (e.g. long, curved nectar spur, ‘landing platform’, posterior petals with nectar guides), is adaptive but dependent on the breeding system (inbreeding versus outbreeding) of the individual species.

The mating systems operating in seven species of Marasmius collected recently in northern Thailand were determined. Marasmius cremeus belonging to sect. Sicci subsect. Siccini ser. Leonini, and Marasmius straminiceps belonging to sect. Marasmius subsect. Sicciformes are described as new species. Five members of sect. Marasmius were tetrapolar (bifactorial), viz. M. apatelius, M. guyanensis, M. nigrobrunneus, M. ruforotula, and M. straminiceps. Two members of sect. Sicci were bipolar (unifactorial), viz., M. corneri (syn. M. incarnatus) and M. cremeus. Our data support the hypothesis that the mating system is consistent within infrageneric taxa. The seven species that are herein described, illustrated, and compared with phenetically similar species represent the first reports for this genus in Thailand.  相似文献
