The accumulation of 59Fe and 58Co from water by plaice (Pleuronectes platessa L.) has been studied in tissues, and their levels of accumulation related to stable element concentrations. The retention of both radionuclides from labelled Nereis and labelled pellets has also been examined, and the loss of activity from intraperitoneal injections followed over a three-month-period. Biological half-times of the exchange with water have been calculated for many organs, although the rate of accumulation for some tissues could only be expressed as a linear function; however, for both radionuclides the direct accumulation from water resulted in concentration factors of less than 1 % of the values inferred from the stable element data. The relevance of such observations to heavy metal pollution at the higher trophic levels is discussed.  相似文献

Some further studies on the accumulation and retention of 64Zn and 54Mn by plaice, Pleuronectes platessa L., have been made. Direct accumulation from water, by eggs and larvae, is insufficient to account for the concentrations of these metals in young fish. It appears, therefore, that, from the earliest stages, food constitutes the major pathway for accumulation. Stable element analyses of liver, muscle, and bone samples from fish of differing weights and ages show that their concentrations, in adult fish, remain fairly constant. On this basis it has previously been estimated that biological half-times of these metals via food should be shorter than those observed after accumulation from labelled water. Experiments following the loss of the isotopes from fish which had been fed labelled polychaete worms support this view.  相似文献

The accumulation of inorganic mercury from sea water by plaice eggs, larvae, and adult fish has been studied using 203HgCl2 as a tracer. The isotope was rapidly accumulated and the levels of accumulation have been related to stable element concentrations. High concentration factors were attained by many organs, but the distribution of the 203Hg was markedly different from that of the stable element. Whereas the largest fraction of the body burden of mercury is contained by muscle, only a slow rate of accumulation into this tissue was observed. In addition, the mercury in fish muscle is of the methyl form: no evidence for the methylation of the tracer was obtained.  相似文献

The dynamics of Na+, K+, and proline accumulation in various organs of non nodulated Vigna sinensis and Phaseolus aureus was followed during their acclimation to two levels of salinities for a period of 35 days and was correlated to the vegetative growth of the two species. The rate of Na+ and K+ absorption is at a maximum during the first 15 to 20 days of culture. K+ absorption is not completely inhibited even at 100 mM NaCl although the endogenous Na+ largely surpasses that of K+ in certain organs. Low salinity rather accelerates K+ absorption in both species. The relative growth rates (RGR) correlate with the rate of Na+ and K+ accumulation. At low salinity (10 mM NaCl), the RGR of V. sinensis is greater than that of P. aureus. However, at high salinity (100 mM NaCl) the RGR is the same for both species. The growth of the younger parts of the two species is not arrested by salt treatment. Very high accumulation of Na+ is avoided in organs with less vacuolated tissues. At no time does the endogenous K : Na ratio in these organs fall below 1.0. Certain organs, especially the roots, hypocotyls, and the lower parts of the stems are capable of storing large quantities of Na+. In V. sinensis, the accumulated Na+ and K+ are evenly distributed among the various organs while in P. aureus they are rather concentrated in the roots. External salinity creates water deficiency in the younger plant parts and as a consequence, proline accumulates especially in the youngest aerial organs - more in P. aureus than in V. sinensis. The accumulation of this amino acid in both the species is dependent on time and correlates directly, not only with the water deficit, but also with the K+ contents. In contrast, it does not seem to depend directly on the endogenous Na+ content. The relative salt tolerance of the two species and the possible role of K+, Na+ and proline in the osmotic adjustments of the two species under saline conditions are discussed.  相似文献

Species specificity of osmotic and ion regulation in five species of acipenserids connected in varying degrees with sea conditions is investigated. The objects of study were freshwater middle-Volga sterlet Acipenser ruthenus; Siberian sturgeon A. baerii, inhabiting the Lena River and migrating to the sea near the river mouth; and the following migratory acipenserids of the Volga-Caspian Basin (regular diadromous migrations): the Russian sturgeon A. gueldenstaedtii, the great sturgeon Huso huso, and the starred sturgeon A. stellatus. The study was carried out on immature specimens (of age 2+). The level of osmolarity and concentrations of cations (Na+, K+, Ca2+, and Mg2+) were determined in blood serum, urine, and liquid from the cavity of spiral valve in fish from the fresh water (control) and after their adaptation (period of seven to ten days) to sea water with salinity 12.5–12.7 and 18‰. The increase was found in the functional activity of the mechanism of osmotic and ion homeostasis in sturgeons according to the increase of the environmental salinity of a particular species. The freshwater sterlet adapted to the sea environment as osmoconformer. The other studied species were capable of maintaining a relative stability of osmolarity and ion concentration in blood serum at different levels while adapting to the sea environment; they changed from hyperosmotic (in fresh water) to hypoosmotic type of regulation. The kidney plays a more significant role in the removal of excessive Na+ in sturgeons than in bony fish. An increase in the sodium-reabsorption and magnesium-secretion functions of the kidney as well as the sodium-absorption function of the intestines was observed in sturgeons, relative to the increase of environmental salinity.  相似文献

The decay of luminescence in the 6–600-μs range following a microsecond flash has been studied in Chlorella. The decay is highly polyphasic; three kinetic components are outlined, in confirmation of the results of K. L. Zankel (1971, Biochim. Biophys. Acta 245, 373–385).Extrapolation of the decay to zero dark time suggests that a unique metastable species C?+, resulting from photochemical charge separation in the System II reaction center, is the substrate of the recombination reaction which gives rise to luminescence.The fast (5–10 μs) and medium (50–70 μs) phases of the decay denote different stabilization steps, preceding relaxation of the centers by electron and proton transduction to the photosynthetic chain.NH2OH specifically inhibits the fast phase and enhances the medium phase. This effect is explained by assuming that the fast phase results from electron transfer from the water splitting system Z to the oxidized primary donor Y.3-(3,4-Dichlorophenyl)-1,1-dimethylurea (DCMU), in the presence of NH2OH elicits another fast phase. It is believed that DCMU affords a parasitic stabilization of C?+ by forming a complex with Q?.  相似文献

The uptake and translocation of 59Fe applied to leaves of Zea mays L. is studied with special reference to the effect of dimethylsulfoxide (DMSO). 59Fe is deposited on corn leaves as droplets of solution of ferrous sulfate or ferric nitrate (1 mM. The uptake of 59Fe is affected by the associated anion; the penetration is more important with the nitrate than with the sulfate. The translocation of 59Fe from the treated part during 24 hours is very low in all the experiments. The exsorption of 59Fe taken up, from the site of application in different solutions (FeSO4, 7H2O; EDTA Na) concerns only a low percentage of 59Fe present in the treated part. DMSO (0.5 and 1 %) increases the uptake but not the translocation of 59Fe applied as sulfate; it seems to have no effect when iron is applied as nitrate. The increase during 24 hours reaches between 32 % and 53 % in seven experiments with sulfate. The effect also appears in an experiment conducted during four weeks, with several applications of ferrous sulfate during this time. This effect of DMSO is discussed: it is mainly explained by the great hygroscopicity of this solvent, therefore the effect would in part depend on the solubility of salts in concentrated DMSO and of the climatic conditions: relative humidity and temperature of the air.  相似文献

The accumulation of organic mercury from sea water by plaice eggs, larvae and adult fish has been studied using CH3203HgCl as a tracer. The isotope was rapidly accumulated, the largest fraction being taken up by muscle tissue. High concentration factors were attained by many internal organs, particularly blood, spleen, and kidney. Longer biological half-times than previous estimates with 203HgCl2 were obtained: the possible consequences of inorganic mercury accumulated from sea water being excreted at the rate for methylmercury have been calculated.  相似文献

Blood from ducks parasitized with Plasmodium lophurae and normal duck blood were incubated with sodium 14C-1-acetate. After release of the parasites from infected red blood cells (RBC) and concurrent treatment of normal blood, lipids were extracted from cellular material and plasma and lipid classes separated by thin-layer chromatography. Specific activity (dpm/mg lipid) of lipid classes was measured quantitatively by liquid scintillation radioassay and gravimetric analysis. The data indicated that the parasite within the RBC incorporated 14C-labeled lipid precursors.Experiments employing sodium 14C-1-acetate in two concentrations, 50 μCi 14C in 0.91 μmole sodium acetate/50 ml blood and 500 μCi 14C in 9.1 μmole sodium acetate/50 ml blood (1.82 × 10?5M and 1.82 × 10?4M), showed higher 14C incorporation into parasitized blood than normal blood preparations at the higher substrate concentration at 5 hr of incubation. At 1.82 × 10?5 M 14C-1-acetate, the highest specific activity in P. lophurae was associated with lipid alcohols. Monoglycerides and diglycerides were significantly labeled. At the higher acetate concentration (1.82 × 10?4M), monoglyceride and diglyceride lipid classes had the highest specific activity in preparations of partially purified P. lophurae.Lipids of plasma from parasitized blood incubated for 5 hr with both concentrations of labeled acetate exhibited the highest specific activity in the free fatty acid class and sterols.At 24 hr of incubation, the lipids of partially purified P. lophurae had increased specific activity in free fatty acids, diglycerides, monoglycerides, phospholipids, and triglycerides.In plasma from parasitized blood incubated 24 hr with 14C-1-acetate, the highest specific activity and greatest percent of 14C incorporation was found in free fatty acids.  相似文献

Spergularia marina (L.) Griseb. is. a rapidly growing, annual, coastal halophyte. Because of its small size, it is suitable for isotope studies of ion transport well beyond the seedling stage. The purpose of this report is to establish the similarities and differences between 22Na+ and 42K+ uptake in S. marina and in more commonly used mesophytic crop species. Vegetative plants were used 18 days after transfer to solution culture. Plants were grown either on Na+-free medium or on 0.2 × sea water. 22Na+ uptake was linear with time for several hours. The rate was relatively insensitive to external concentration between 1 and 180 mol Na+ m?3, particularly in Na+-free plants. Transport to the shoot accounted for 40 to 70% of the total uptake, dependent on salinity but largely independent of time. 42K+ uptake decreased with increasing salinity in Na+-free plants and increased in 0.2 × sea water plants. Both uptake and transport to the shoot were non-linear with time, upward concavity suggesting recovery from a manipulative and/or osmotic injury. Steady state root contents were compared with predicted contents based on cortical cell electrical potentials using the Nernst equation. Reasonable agreement was found in all cases except Na+ content of 0.2 × sea water plants, in which active efflux was indicated. Uptake studies conducted in the presence of chemical modifiers (dicyclohexylcarbodiimide, dinitrophenol and fusicoccin) showed responses of 42K+ uptake as expected from studies on agronomic species, and implied the presence of a similar active uptake here despite the appearance of equilibrium. Active Na+ uptake was suggested at low Na+ levels. We conclude that S. marina is a promising experimental system combining the rapid nutrient acquisition strategy of agionomically important annuals with a high degree of salt tolerance.  相似文献

Di-deuterated and di-tritiated 16,16-dimethyl-trans-Δ2-PGE1 has been synthesized and used for development of a GC-MS method for quantitation of corresponding unlabelled drug in patient plasma. Although these carrier/internal standard molecules only contain 2 deuterium atoms the lower limit of detection at each injection is as low as about 40 pg.The maximum plasma levels of this drug following administration of vaginal suppositories used in clinical studies (1 mg 16,16-dimethyl-trans-Δ2-PGE1 methyl ester in 0.8 g Witepsol S-52) were 100–350 pg/ml i.e. in the same order of magnitude as earlier seenf ro 16,16-dimethyl-PGE2.  相似文献

以羊草幼苗为研究对象,通过调整全营养培养基(CK,0.05 mmol/L Fe2+、0.015 mmol/L Zn2+)中铁或者锌含量设置0、10倍、20倍Fe2+(Zn2+)浓度处理Fe0(Zn0)、Fe10(Zn10)、Fe20(Zn20),以及在高铁培养基中单独添加0.15 mmol/L Zn2+或同时添加10 mmol/L Ca2+、5 mmol/L Mg2+、20 mmol/L K+处理,测定培养6 d后幼苗生长指标和矿质元素含量、以及高铁(Fe20)处理下幼苗根中抗氧化指标和相关基因表达量,探究不同浓度Fe2+、Zn2+对羊草幼苗生长、矿质元素吸收积累及抗氧化指标、基因表达的影响。结果表明:(1)缺锌(Zn0)显著抑制羊草幼苗鲜重的增加和Zn元素的积累,但促进Fe、Mg元素的积累;高浓度锌(Zn10、Zn20)显著促进幼苗叶片生长和Zn元素的积累;缺铁(Fe0)显著抑制幼苗的根长、鲜重和Fe元素的积累,促进Mg、Zn元素的积累;高浓度铁(Fe10、Fe20)显著抑制羊草幼苗根叶生长、根毛发育和Ca、Zn、Mg、K元素的积累。(2)增加Zn2+和Ca2+、Mg2+、K+浓度无法恢复高铁胁迫对幼苗生长的抑制作用。(3)高浓度铁(Fe20)处理羊草幼苗48 h后,根部过氧化物酶、超氧化物歧化酶、过氧化氢酶、抗坏血酸过氧化物酶、谷胱甘肽还原酶活性和丙二醛、抗坏血酸、还原型谷胱甘肽含量显著升高;烟酰胺合成酶基因、过氧化物酶基因表达量显著下调,植物类萌发素蛋白基因表达量显著上调。研究发现,羊草幼苗生长发育和矿质元素积累对环境中Zn2+浓度变化不敏感,却受到环境中高浓度Fe2+的显著抑制,并造成严重的氧化胁迫伤害,这种伤害无法在添加Zn2+或同时添加Ca2+、Mg2+、K+的条件下恢复。  相似文献

The ascidians, the so-called sea squirts, accumulate high levels of vanadium, a transition metal. Since Henze first observed this physiologically unusual phenomenon about one hundred years ago, it has attracted interdisciplinary attention from chemists, physiologists, and biochemists. The maximum concentration of vanadium in ascidians can reach 350 mM, and most of the vanadium ions are stored in the + 3 oxidation state in the vacuoles of vanadium-accumulating blood cells known as vanadocytes. Many proteins involved in the accumulation and reduction of vanadium in the vanadocytes, blood plasma, and digestive tract have been identified. However, the process by which vanadium is taken in prior to its accumulation in vanadocytes has not been elucidated. In the present study, a novel vanadium-binding protein, designated VBP-129, was identified from blood plasma of the vanadium-rich ascidian Ascidia sydneiensis samea. Although VBP-129 mRNA was transcribed in all A. sydneiensis samea tissues examined, the VBP-129 protein was exclusively localized in blood plasma and muscle cells of this ascidian. It bound not only to VO2+ but also to Fe3+, Co2+, Cu2+, and Zn2+; on the other hand, a truncated form of VBP-129, designated VBP-88, bound only to Co2+, Cu2+ and Zn2+. In a pull-down assay, an interaction between VanabinP and VBP-129 occurred both in the presence and the absence of VO2+. These results suggest that VBP-129 and VanabinP function cooperatively as metallochaperones in blood plasma.  相似文献

The accumulation levels of anthropogenic 90Sr and 134Cs and 137Cs radionuclides in the marsh frog have been studied in the areas of the Beloyarskii water-storage reservoir (an industrial storm-water discharge channel of the nuclear power station) and the Verkhnii Tagil water-storage reservoir (the Tagil River down-stream of the dam). No significant distinction in the radionuclide accumulation (90Sr and 137Cs) depending on the amphibian sex and age is detected. Comparable levels of the accumulation of radionuclides in the marsh frog, when compared to the other representatives of the water ecosystem, are estimated. An assumption of the presence of some unidentified source of radioactive contamination of marsh frogs has been made; cesium-137 may be transferred from it to the Tagil River by the frogs.  相似文献

Changes of hematocrit, hemoglobin, and blood plasma glucose and Na+ and K+ content in response to hypoxia were studied in three Black Sea fish species. It was shown that in response to hypoxia in low-mobile rock perch Scorpaena porcus L., hematocrit and the blood plasma glucose level increased, while the content of K+ in erythrocytes decreased and the content of Na+ increased. In moderately mobile sea carp Diplodus annularis L. autogenic hypoxia caused a rise of hematocrit and blood plasma glucose. In actively swimming jack mackerel Trachurus mediterraneus ponticus Aleev, only considerable increase of Na+ content was revealed in hypoxia. The obtained results indicate that fish with different mobility under hypoxic conditions use different adaptation mechanisms. The value and direction of changes of the chosen parameters can be used to determine resistance of fish to oxygen deficit.  相似文献

1-Palmitoyllysophosphatidylcholine has been mixed in equimolar amounts with specifically deuterated palmitic acid and the structural properties of the lipid/water phase have been studied by 2H- and 31P-nuclear magnetic resonance. The order profile of the free palmitic acid is very similar to that of the parent compound 1,2-dipalmitoyl-sn-glycero-3-phosphocholine at temperatures above the gel-to-liquid crystal phase transition. The bending of the sn-2 chain which is typical for diacyl lipids is not observed for the free palmitic acid. The mixture of lysolipid and palmitic acid exhibits well-defined quadrupole splittings even at temperatures below the gel-to-liquid crystal phase transition. Hence it is possible for the first time to establish an order profile in the gel-state of the lipid bilayer phase. Between carbon atoms 5 to 12 the palmitic acid chain is found to assume the extended all-trans conformation with a very small contribution from gauche defects. Towards the methyl terminal a distinct increase in the gauche probability can be noted. The motion of the phosphocholine headgroup was also studied by 2H- and 31P-NMR using selectively deuterated 1-palmitoyllysophosphatidylcholine. The headgroup has a considerably larger motional freedom in the mixture of lysolipid and palmitic acid than in 1,2-dipalmitoyl-sn-glycero-3-phosphocholine. In addition, the average headgroup conformations are also different in the two systems.  相似文献

A radioimmunoassay (RIA) for the determination of antipyrine directly in plasma and saliva has been developed using 4-125I-iodoantipyrine as the radioligand. The method showed excellent agreement (r=0.98) with a recently reported RIA for antipyrine using 3H-antipyrine as the radioligand. The interassay coefficient of variation for the 125I-RIA did not exceed 7.6% and the mean recovery of antipyrine added to plasma or saliva was 101±1.73 (S.E.) over a range of 1.5 to 30 μg/ml. By virtue of its simplicity and low cost, the 125I-RIA for antipyrine offers an attractive method for the routine determination of antipyrine levels and subsequent calculation of its half-life in man.  相似文献

The interaction between the plant hormone, 3-indoleacetic acid (IAA), and some phospholipids in CDCL3 has been studied by 1H, 13C and 31P nuclear magnetic resonance (NMR) spectroscopy. Upon interaction with IAA, significant changes occurred in resonance positions of the phospholipid head group nuclei. Alteration of the fatty acid composition influenced the effects of IAA on these nuclei. These effects were observed in the ethanolamine and phosphate groups of the phosphatidylethanolamines, and in the choline, phosphate and glycerol groups of the phosphatidylcholines. Changes in resonance positions of the phospholipid head group nuclei were used for the determination of dissociation constants (Kd). In all cases, Kd values were approx. 10?2 molal for 1 : 1 interaction. The NMR results suggest an interaction orientation in which the aromatic ring system of IAA interacts with the quaternary nitrogen function of the head group, and the phosphate group becomes hydrogen-bonded to the NH or carboxyl proton of 1AA.  相似文献

With the successful development of methods for the isolation and purification of ethanol-insoluble cinnamic acid esters in mint it became possible to initiate kinetic, isotopic studies on purified, ‘insoluble’ derivatives of caffeic acid, ferulic acid and p-coumaric acid. Pulse-feeding experiments were conducted with 14CO2, phenylalanine-U-14C and cinnamic acid-3-14C. The ferulic acid derivative exhibited a significant turnover as compared to the other insoluble derivatives which were relatively stable. Time-course tracer studies were performed to compare the turnover of soluble caffeic acid derivatives with ‘insoluble’ forms of caffeic acid. Caffeic acid associated with a macromolecular fraction consistently showed a higher specific activity than either soluble caffeic acid or the caffeic acid associated with a second insoluble derivative.  相似文献

Iron, which occurs in sea water as particulate ferric hydroxide, is accumulated to high concentrations by the common mussel, Mytilus edulis (L.). The kinetics of the accumulation and excretion of iron in Mytilus has been studied using 59Fe-labelled ferric hydroxide and the tissue distribution and identification at the sub-cellular level determined by analytical electron microscopy. Iron-59 accumulates in linear proportion to the sea-water concentration and is found in all tissues; the concentration factors for viscera, kidneys, gills, muscle = mantle is 25: 6: 4: 1, respectively. The particulate iron is taken up by pinocytosis by special epithelial cells in the gills, gut, kidney and possibly labial palps and held in membrane-bound vesicles, unaccompanied by mucus in the case of gills and kidney, but with mucus present in the digestive diverticulum and mid-gut cells. There is no free iron within the cytoplasm. Approximately 30 % of the iron presented to the gut is not absorbed being voided with the faeces. The absorbed iron is exocytosed and then passed on to amoebocytes in the haemolymph for transport to other tissues, a major portion being excreted by transfer to the byssal threads.  相似文献
