Plastid sequences of the atpB-rbcL spacer and rbcL gene itself were used to evaluate their respective potential in reconstructing the phylogeny of 15 taxa from the tribe Rubieae (Rubiaceae). From our previous analyses using the atpB-rbcL spacer, the 15 selected taxa represent most of the variability of the tribe. Since this group is considered to be relatively recent (Upper Tertiary), it should allow the study of early dynamics of nucleotide substitutions in recent divergences. The results show that the spacer and rbcL inferred phylogenies are not totally congruent; the spacer trees are more similar to interpretations of morphological data. A comparative analysis of the pattern of nucleotide substitution of these two sequences in the Rubieae shows that (1) the overall rate of substitution is similar in the spacer and in rbcL, and the rate of synonymous substitution in rbcL is much higher; (2) the level of homoplasy is higher in rbcL than in the spacer matrix which shows a higher phylogenetic structure; and (3) the pattern of transition and transversion substitutions is different in the two sequences, and is not linear in rbcL. As a result of these observations, we suggest that (1) the spacer is evolving relatively slowly because of unsuspected, and phylogenetically important; selective constraints on its sequence; and (2) in the rbcL sequence, many sites, free of constraint, are changing at high rate, and some of these sites seem to have undergone multiple substitutions even in this recent tribe. A new genus of broad‐nosed weevils belonging to the tribe Naupactini (Coleoptera: Curculionidae) is described, based on six species from the Páramo–Puna subregion (Andean region) of South America. Three species are new and three have been transferred from the genus Asymmathetes (Asymmathetes nigrans, Asymmathetes rugicollis, and Asymmathetes vulcanorum). The new genus is recognized by the black, denuded, and shiny tegument, the well‐developed pre‐epistome, the elytral base curved backwards on middle, the reduction of the hindwings, and the widely separated procoxae, closer to the anterior than to the posterior margin of the prosternum. A cladistic analysis of the six species of the new genus plus five out‐groups (Amitrus, Amphideritus, Asymmathetes, Melanocyphus, and Trichocyphus), using 49 morphological characters, resulted in a single cladogram. © 2012 The Linnean Society of London, Zoological Journal of the Linnean Society, 2012, 166 , 54–71.  相似文献

Haplodontium zangii X.R.Wang &; J.C.Zhao, a new moss species from Xizang, China, is described and illustrated. Previously, specimens of H. zangii have been identified as Mielichhoferia himalayana Mitt. However, H. zangii is distinctly different from M. himalayana in having excurrent costae with short awns (vs long denticulate awns), short-pyriform capsules, 0.8–1?mm (vs long-pyriform capsules, 2.5–3?mm), and densely papillose exostome teeth (vs smooth or vertically striped exostome teeth). Haplodontium zangii is similar to H. macrocarpum (Hook.) J.R.Spence, which was traditionally placed in Mielichhoferia Nees &; Hornsch. as M. macrocarpa (Hook.) Bruch &; Schimp. The main differences between H. zangii and H. macrocarpum are in the morphology of the leaves, capsules, guide cells, and stomata. Mielichhoferia himalayana and another Chinese species of Mielichhoferia, M. sinensis Dix., are also transferred to Haplodontium Hampe, a new genus in the bryoflora of China, as H. himalayanum (Mitt.) X.R.Wang &; J.C.Zhao and H. sinensis (Dix.) X.R.Wang &; J.C.Zhao. A morphological comparison and a key to the three species of Haplodontium in China as well as to H. macrocarpum, a species that is likely to be found in China, are provided.  相似文献

Phylogenetic relationships within the palaeotropical tribe Lepisoroideae (Polypodiaceae) were investigated by studying sequence variation of four plastid DNA regions: rbcL, rps4 plus rps4‐trnS IGS, trnL intron plus trnL‐F IGS, rbcL‐atpB IGS plus part of atpB. In total, over 4000 nucleotides were sequenced for 39 species. Seven well‐supported clades were found in the analyses of the combined data set. We provide a new classification of Lepisoroideae by integrating phylogenetic results and known variation of morphological characters. The two small genera Neocheiropteris and Tricholepidium are supported as monophyletic, the genus Paragramma is resurrected and the genera Lepisorus, Neolepisorus, Lemmaphyllum and Lepidomicrosorium are re‐circumscribed. We proposed 14 new combinations, among which Caobangia is treated as a synonym of Lemmaphyllum. The genus Curcuma L. is commonly used as spices, medicines, dyes and ornamentals. Owing to its economic significance and lack of clear‐cut morphological differences between species, this genus is an ideal case for developing DNA barcodes. In this study, four chloroplast DNA regions (matK, rbcL, trnH‐psbA and trnL‐F) and one nuclear region (ITS2) were generated for 44 Curcuma species and five species from closely related genera, represented by 96 samples. PCR amplification success rate, intra‐ and inter‐specific genetic distance variation and the correct identification percentage were taken into account to assess candidate barcode regions. PCR and sequence success rate were high in matK (89.7%), rbcL (100%), trnH‐psbA (100%), trnL‐F (95.7%) and ITS2 (82.6%) regions. The results further showed that four candidate chloroplast barcoding regions (matK, rbcL, trnH‐psbA and trnL‐F) yield no barcode gaps, indicating that the genus Curcuma represents a challenging group for DNA barcoding. The ITS2 region presented large interspecific variation and provided the highest correct identification rates (46.7%) based on BLASTClust method among the five regions. However, the ITS2 only provided 7.9% based on NJ tree method. An increase in discriminatory power needs the development of more variable markers.  相似文献

Phylogenetic analyses of molecular data (COI, 16SrDNA, 18SrDNA, and 28SrDNA) show that the hesionid genus Gyptis Marion & Bobretzky in Marion, 1874, and the tribe Gyptini Pleijel, 1998, are nonmonophyletic as currently delineated. We introduce the new tribe Amphidurini and the new genus Neogyptis to accommodate these new findings. Amphidurini is sister to Gyptini and Ophiodromini and includes Amphiduros Hartman, 1959, Amphiduropsis, Pleijel, 2001, Neogyptis gen. nov. , and, possibly, Parahesione Pettibone, 1956. Morphologically, Amphidurini can be separated from Gyptini by the distally, rather than subdistally, inserted ventral cirri, and Neogyptis by the combination of this feature, the presence of a median antenna, and a distal ring with papillae on the proboscis. We redescribe and transfer the four species Gyptis crypta Pleijel, 1993, Gyptis mediterranea Pleijel, 1993, Gyptis plurisetis Hilbig, 1992, and Gyptis rosea (Malm, 1874) from Gyptis to Neogyptis gen. nov. , and describe five new species from shallow waters in Belize, Hong Kong, off Vladivostok, and deep‐sea hydrothermal vents in the Lau Basin off Fiji.  相似文献

The aphid genus Blackmania gen. nov. is described. In the genus, B. eastopi sp. nov. associated with Polygonum equisetiforme (Polygonaceae) from Israel and Cyprus is described and illustrated. Morphologically, the new genus is similar to the genus Acaudella in respect to the lack of a cauda. Acaudella puchovi, associated with Atraphaxis caucasica (=A. buxifolia) and A. spinosa (Polygonaceae) from Uzbekistan and Israel, is re‐described and the apterous viviparous female is figured for the first time. The lectotype and paralectotypes of A. puchovi are also designated. An identification key to known species of the tribe Macrosiphini without cauda is provided. The morphological separation of B. eastopi gen. nov., sp. nov. from A. puchovi is visualized using principal component analysis.  相似文献

Species of the tribe Adramini (Tephritidae: Trypetinae) are usually slender, and some specific species have eyes borne at the ends of their long stalks. This tribe is mainly distributed in the tropics and subtropics of the Afrotropical, Oriental and Australasian Regions; relatively few species occur in the Palearctic and Nearctic Regions. The phylogeny of the tribe Adramini is presented here based on analysis of morphological and molecular information (DNA sequences of nuclear 28S rDNA, mitochondrial COI and COII, and 16S rDNA genes) for its representative species in most genera. Three monophyletic groups (Adrama‐com‐group, Pelmatops‐com‐group and Dimeringophrys‐com‐group) were discovered in the combined morphological and molecular tree. The results showed moderate support for the monophyly of Adramini and strong support for most of its genera. However, Euphranta appears to be polyphyletic. Sapadrama, Celidodacus and Euphranta are basal taxon, and Coelopacidia, Soita and Trypanophion are closely related to the stalk‐eyed fruit flies (Pelmatops + Pseudopelmatops). A hypothesis regarding the morphology–function relationships for two main groups of Adramini (Adrama‐group and Pelmatops‐group) with different evolving probabilities is inferred. Sapadrama is proposed be removed from Adramini; a new genus, Ichneumonomacula Chen gen. n. and a new species Ichneumonmacula wangyongi Chen sp. n., are recognized and described, and a key to recognize the genera of Adramini around the world is provided.  相似文献

Generic boundaries among the genera Cheilosporum, Haliptilon, and Jania—currently referred to the tribe Janieae (Corallinaceae, Corallinales, Rhodophyta)—were reassessed. Phylogenetic relationships among 42 corallinoidean taxa were determined based on 26 anatomical characters and nuclear SSU rDNA sequence data for 11 species (with two duplicate plants) referred to the tribe Corallineae and 15 species referred to the tribe Janieae (two species of Cheilosporum, seven of Haliptilon, and six of Jania, with five duplicate plants). Results from our approach were consistent with the hypothesis that the tribe Janieae is monophyletic. Our data indicate, however, that Jania and Haliptilon as currently delimited are not monophyletic, and that Cheilosporum should not be recognized as an independent genus within the Janieae. Our data resolved two well‐supported biogeographic clades for the included Janieae, an Indian‐Pacific clade and a temperate North Atlantic clade. Among anatomical characters, reproductive structures reflected the evolution of the Janieae. Based on our results, three genera, Cheilosporum, Haliptilon, and Jania, should be merged into a single genus, with Jania having nomenclatural priority. We therefore propose new combinations where necessary of some species previously included in Cheilosporum and Haliptilon.  相似文献

Phylogenetic relationships of the subfamily Combretoideae (Combretaceae) were studied based on DNA sequences of nuclear ribosomal internal transcribed spacer (ITS) regions, the plastid rbcL gene and the intergenic spacer between the psaA and ycf3 genes (PY-IGS), including 16 species of eight genera within two traditional tribes of Combretoideae, and two species of the subfamily Strephonematoideae of Combretaceae as outgroups. Phylogenetic trees based on the three data sets (ITS, rbcL, and PY-IGS) were generated by using maximum parsimony (MP) and maximum likelihood (ML) analyses. Partition-homogeneity tests indicated that the three data sets and the combined data set are homogeneous. In the combined phylogenetic trees, all ingroup taxa are divided into two main clades, which correspond to the two tribes Laguncularieae and Combreteae. In the Laguncularieae clade, two mangrove genera, Lumnitzera and Laguncularia, are shown to be sister taxa. In the tribe Combreteae, two major clades can be classified: one includes three genera Quisqualis, Combretum and Calycopteris, within which the monophyly of the tribe Combreteae sensu Engler and Diels including Quisqualis and Combretum is strongly supported, and this monophyly is then sister to the monotypic genus Calycopteris; another major clade includes three genera Anogeissus, Terminalia and Conocarpus. There is no support for the monophyly of Terminalia as it forms a polytomy with Anogeissus. This clade is sister to Conocarpus. Electronic Publication  相似文献

Abstract One application of DNA barcoding is species identification based on sequences of a short and standardized DNA region. In plants, various DNA regions, alone or in combination, have been proposed and investigated, but consensus on a universal plant barcode remains elusive. In this study, we tested the utility of four candidate barcoding regions (rbcL, matK, trnHpsbA, and internal transcribed spacer (ITS)) as DNA barcodes for discriminating species in a large and hemiparasitic genus Pedicularis (Orobanchaceae). Amplification and sequencing was successful using single primer pairs for rbcL, trnH‐psbA, and ITS, whereas two primer pairs were required for matK. Patterns of sequence divergence commonly showed a “barcoding gap”, that is, a bimodal frequency distribution of pairwise distances representing genetic diversity within and between species, respectively. Considering primer universality, ease of amplification and sequencing, and performance in discriminating species, we found the most effective single‐region barcode for Pedicularis to be ITS, and the most effective two‐region barcode to be rbcL + ITS. Both discriminated at least 78% of the 88 species and correctly identified at least 89% of the sequences in our sample, and were effective in placing unidentified samples in known species groups. Our results suggest that DNA barcoding has the potential to aid taxonomic research in Pedicularis, a species‐rich cosmopolitan clade much in need of revision, as well as ecological studies in its center of diversity, the Hengduan Mountains region of China.  相似文献

An extended molecular phylogenetic analysis of Uvaria (Annonaceae) is presented, using maximum parsimony, maximum likelihood and Bayesian methods, based on sequences of four plastid DNA regions (matK, psbA‐trnH spacer, rbcL and trnL‐F). The additional taxa include the monotypic West African genus Balonga, the monotypic South‐East Asian genus Dasoclema and seven Australian representatives of the genus Melodorum. The results indicate that all of these taxa are nested within a well‐supported clade otherwise consisting of Uvaria species, indicating that their taxonomic treatment needs to be reassessed. The distinguishing morphological characteristics of the taxa are re‐evaluated and interpreted as specialized adaptations of the basic Uvaria structure. The genus Uvaria is accordingly extended following the transfer of these species, necessitating six new nomenclatural combinations and two replacement names. © 2010 The Linnean Society of London, Botanical Journal of the Linnean Society, 2010, 163 , 33–43.  相似文献

ABSTRACT

Pselaphotumulus Owens and Carlton, gen. nov., is described as the sixth genus in the tribe Pselaphini known from New Zealand. Three new species are described: Pselaphotumulus aorerei, sp. nov., Pselaphotumulus dubius, sp. nov. and Pselaphotumulus unus, sp. nov.. Three species, Pselaphus cavelli (Broun 1893), Pselaphus oviceps (Broun 1917) and Pselaphus urquharti (Broun 1917) are transferred to Pselaphotumulus, nov. combs. Lectotypes from type series in the New Zealand Broun Collection (Natural History Museum, London) are designated for these three species. Habitus photographs, distributional maps and line drawings of diagnostic characters are provided for each species. A key to species is provided. Searches of museum collections have not yielded representatives outside of New Zealand, suggesting that this is the first endemic genus in the tribe Pselaphini described from the New Zealand’s main islands, specifically, the South Island. Pselaphotumulus species exhibit restricted distribution patterns that approximately coincide with the Pacific/Indo Australian Plate boundary.  相似文献

Tricalcaria Han gen. nov. and its type species Tricalcaria stueningi Han sp. nov. are described from China. Morphological characters, including those of the male and female genitalia, are figured. The main diagnostic characters of Tricalcaria are: hind tibia with three spurs in both male and female; ventral margin of costal protrusion bearing a series of long, hook‐like, sclerotized setae in the male genitalia; female papilla analis modified and bearing over part of its surface broad blade‐like sclerotized setae. The wing pattern, venation, and genitalia of this genus are compared with those of the most closely related genera, Tristeirometa Holloway, 1997, Hypocometa Warren, 1896, and Phthonoloba Warren, 1893. Tribal placement is discussed, with the conclusion that the new genus should be placed in the tribe Trichopterygini.  相似文献
