Quantitative analyses have been made of the dietary cholesterol requirement for the growth of the larvae of Musca domestica. The larvae will not grow on diets to which no cholesterol is added, a few pupae and adults are obtained when the concentration of cholesterol is 0·05 μmol/g of diet, but the concentration has to be raised to 0·36 μmol/g of diet before the maximum numbers of pupae and adults are obtained. Further addition of cholesterol above 0·36 μmol/g diet did not have any significant effect on the weight and growth of the larvae. However, the ratios of the cholesterol to phospholipid fractions recovered from the larvae increased rapidly when the concentration of cholesterol in the diet was raised from 0·05 to 0·56 μmol/g of diet. Above this concentration only a slight increase in the ratios was observed. Larvae reared on diets containing 0·05 μmol cholesterol/g of diet contain only 25 per cent of the cholesterol content of the larvae reared on the diets containing more than 0·28 μmol of cholesterol/g of diet, the cholesterol content being expressed relative to the weight of the larvae,The absence of cholesterol synthesis has been demonstrated in the larvae by feeding [4-14C] cholesterol. The specific activity of the cholesterol recovered from the larvae is the same as that of cholesterol added to the diet. Irrespective of the cholesterol concentration of the larval diet, approximately 97 per cent of the radioactivity recovered from the larvae behaved as free cholesterol, less than 1 per cent as cholesterol esters and the rest as unidentified ‘polar sterols’. Larvae of the saw-toothed grain beetle, Oryzaephilus surinamensis, were reared individually on meridic diets from hatching to adult emergence at 32±0·25°C and 75% r.h. These larvae developed little, if at all, and failed to carry out larval-pupal ecdysis on a diet devoid of leucine. Survival increased as the concentration of dietary l-leucine increased from 0·0 to 3·0 mg/g and rate of development, as l-leucine increased to 8·0 mg/g. With concentrations of dietary l-leucine greater than 8·0 mg/g, the rate of development of Oryzaephilus surinamensis decreased.  相似文献

Meridic and oligidic diets suitable for the continuous culture of the aphid Myzus persicae were developed through modifications of a holidic diet. These included the addition of various amounts of crude nutrients and the replacement of defined nutrients by crude nutrients. The highest level of aphid growth (mean weights of 600 to 800 μg) was maintained (for 45 successive generations) on a meridic diet made by supplementing a holidic diet with 2.0% yeast extract (NBC).Continuous culture, at a level of growth (400 to 600 μg) comparable to that on the unsupplemented holidic diet (formulated with 34 defined nutrients), was supported by an oligidic diet containing only 10 weighed ingredients: 15 g sucrose, 2.5 g enzymatic casein hydrolysate (NBC), 2 g yeast extract, 123 mg MgSO4·7H2O, 100 mg ascorbic acid, 10 mg niacin, 5 mg Ca pantothenate, 2.5 mg pyridoxine, 2.5 mg thiamine, and appx. 1.5 gm K2HPO4·3H2O (pH 6.8) per 100 ml of diet.Yeast extract at 2.0% provided adequate amounts of choline chloride, biotin, folic acid and inositol, but not of niacin, pantothenate, pyridoxine, and thiamine. Enzymatic casein hydrolyzate at 2.5% could replace the 20 defined amino acids of the holidic diet. Diets with both yeast extract and casein hydrolysate did not have to be supplemented with trace minerals (Cu, Fe, Mn and Zn). Yeast extract at 2.5% provided sufficient amounts of trace minerals, amino acids, and B-vitamins to sustain numerous successive generations, albeit at a low level of growth (200 to 300 μg). The simplicity and low cost of oligidic diets makes them attractive for aphid studies in which nutrition is not the primary consideration.  相似文献

Fifth instar Manduca sexta growth response to injected doses of canavanine was concentration-dependent over a range of 0·5 to 2·0 mg/g body weight. Twenty-four hr after injection of 14C-guanidinooxy-d,l-canavanine, M. sexta larvae incorporated approximately 3·6% of the labelled l-canavanine into protein of non-gut tissue. Adult M. sexta mortality was related to the level of injected canavanine over a range of 2 to 8 mg/g body weight. Injection of as little as 2 mg canavanine/g body weight caused hyperactivity in adult M. sexta. Arginine, able to negate the toxic effects of canavanine during larval growth, was only marginally capable of overcoming canavanine effects on larval-pupal ecdysis.  相似文献

Larvae from two populations of Heliothis zea were reared on artificial diets containing various sterols, which supported suboptimal growth, and their tissue sterols were characterized in order to determine how these dietary sterols are utilized by this insect. The sterols studied included Δ5,7-sterols (7-dehydrocholesterol or ergosterol), Δ8-sterols (lanosterol and/or 24-dihydrolanosterol), and a Δ5-sterol (4,4-dimethylcholesterol). Although larvae did not develop on 4,4-dimethylcholesterol, those fed primarily Δ8-4,4,14-trimethylsterols developed to the third instar. When the latter sterols were spared with cholesterol, the larvae reached the sixth instar and contained 4,4,14-trimethylsterols as well as cholesterol in their tissues. When larvae were fed 7-dehydrocholesterol, <1% of the larvae from one population developed to the sixth instar and these larvae contained 7-dehydrocholesterol as their principal sterol. The other larvae successfully completed their larval stage when they were transferred from the diet containing 7-dehydrocholesterol (or no sterol) to a diet containing cholesterol within at least 9 days. The sterol composition of larvae transferred from a diet containing cholesterol to a diet containing 7-dehydrocholesterol, after they had reached 60% of their final weight, was 54% cholesterol and 46% 7-dehydrocholesterol. The major sterol isolated from the tissues of the larvae fed ergosterol was also 7-dehydrocholesterol. Therefore, although the larva of H. zea can dealkylate and saturate the side chain of the Δ5,7,22-24β-methylsterol, it carries out little metabolism of the B ring of the nucleus. These studies demonstrate that, when Δ5,7- or Δ8-sterols are the principal sterols in the diet of H. zea, they are absorbed and incorporated into its tissues, although they slow the rate of growth and may prevent complete development of the larva.  相似文献

—The isoelectric point, surface charge and Km for choline of choline acetyltransferase from different species were determined. Choline acetyltransferase from mouse and monkey brain was resolved into three molecular forms with isoelectric points at 7·1, 7·5, 8·4 and 7·0, 7·35, 8·35 respectively, whereas choline acetyltransferase from the electric organ of Torpedo and from rabbit brain showed a molecular form with isoelectric point 6·6 and 6·9, respectively. With the exception of rabbit brain enzyme, there was a good correlation between the isoelectric points and surface charges of the different choline acetyltransferases. The Km's for choline were 0·66, 0·88, 0·92 and 3·5 mM for monkey, mouse, rabbit and Torpedo choline acetyltransferase respectively. The separated molecular forms of mouse and monkey enzymes did not show any significant difference in their affinity for choline.  相似文献

The transport of sterols incorporated into the lecithin bilayer of small unilamellar liposomes through a model membrane was studied. A two-chamber diffusion cell containing liposomes with incorporated [4-14C]cholesterol or β-[4-14C]sitosterol in the donor chamber and liposomes with unlabeled cholesterol in the receiver chamber was used. The permeability coefficients of the sterols through silastic rubber membranes which served as a model membrane were measured. The permeability for cholesterol incorporated into liposomes in a phosphatidyl choline/cholesterol molar ratio of 1 : 1, produced by sonication for 1 h, and subsequent centrifugation at 100000 × g for 1 h, was 1.6 · 10?8 cm sec?1. Dilution of the liposome suspension did not change the permeability coefficient significantly. The permeability coefficient of sitosterol incorporated into liposomes was about 4-times smaller than that of cholesterol. These results suggest that the sterols were delivered to the silastic membrane by the intact liposomes and that free solute was not involved in the transport to the membrane to a significant degree. The large differences in the permeability coefficients between cholesterol and sitosterol indicate that an aqueous interfacial barrier was crossed by the sterol during the delivery to the membrane.  相似文献

The fatty acid composition of Pieris brassicae was measured from larvae reared on four different diets. Pieris can alter the composition of fatty acids in the diet through selective incorporation and synthesis. Oleate is preferentially accumulated on artificial diets (15·9 per cent in diet, 43·8 per cent in neutral lipid (NL) of fifth instar larvae), but not equally on natural diets (18·1 per cent in Brassica napus, 25·6 per cent in the NL of fifth instar larvae). Incorporation of linolenate appears to depend on the concentration of both linolenate and linoleate in the diet. With dietary levels of 35·7% linolenate and 32·2% linoleate, fifth instar larvae contain 12·2 and 16·0 per cent, respectively, of these acids. With 45·8% linolenate and 12·5% linoleate in the diet, fifth instar larvae contain 44·1 and 11·6 per cent of these acids, respectively, in the NL. Palmitoleate is actively synthetized on the artificial diets; with trace amounts of dietary palmitoleate, fifth instar larvae have 9·3 per cent of this acid in the NL. Pieris regulates the uptake of linoleate from the diet at the intestinal wall as was shown by linoleic acid-1-14C, and is unable to convert dietary linoleate to any of the 18-carbon analogues. The female apparently accumulates linolenate into egg phospholipids on the artificial diet, but in general the fatty acid composition of the eggs resembles that of the fat body.  相似文献

The occurrence of a factor in soybean which deters feeding of the silkworm was demonstrated. The deterrent was not extractable with ether or ethanol, but was extracted with 90% ethanol or hot water. Preliminary experiments showed that the deterrent was adsorbed on charcoal, but neither on activated alumina nor in a steam distillate. Autoclaving did not inactivate the deleterious effect. A crude extract of the deterrent showed a marked negative feeding response by including 0·4 mg/g of the dry diet.Inclusion of the crude extract in the diet retarded the growth of larvae, whereas larvae grew well when receiving the 90% ethanol-washed meal. Feeding response for various soybean meals and proteins were compared, and better larval growth has generally obtained when the feeding response was more positive. It is, however, inconclusive whether or not the deleterious principle possesses some physiologically toxic effect.The feeding behaviour of newly hatched larvae was observed in the presence of favourable and/or unfavourable diets, and a brief discussion is made concerning the feeding behaviour of this insect.  相似文献

The dispersion of methoxychlor and Rhodamine B dye in the Chalk River, Canada, was measured by chromatography and fluorometry. The accumulation of methoxychlor was studied in laboratory and field experiments.The river was treated for 15 min with an oil solution of methoxychlor (0·79 μg/litre). The numbers of drifting simuliid larvae reached maximum values 60 min after the start of treatment at stations 275 m and 550 m downstream from the point of application. The peak numbers decreased with distance downstream, and most of the larvae began to drift after the methoxychlor had passed by.No methoxychlor could be detected in water, larvae or moss collected from the Chalk River before the experiment, but drifting larvae caught after the treatment contained residues ranging from 0·24 to 2·57 mg/kg.The larvae which began drifting later generally contained more methoxychlor than those drifting soon after treatment. S. venustum began drifting sooner than S. vittatum, and the mean sizes of drifting larvae of both species tended to increase as time passed. Residues of methoxychlor ranging from 7·4 to 34·6 μg/kg were detected in moss and grasses in the river for up to 8 weeks after the treatment.Laboratory experiments indicated that simuliid larvae concentrate particulate formulations of methoxychlor more efficiently than ethanol ones. Residue values in the larvae reached 2310 μg/kg.Trichoptera larvae concentrated methoxychlor to levels up to 1563 μg/kg, and ethanol formulations to higher levels than particulate ones.  相似文献

To evaluate the relationship between the FMO3 genotype and dietary trimethylamine (TMA) precursors on TMA-N content in eggs, laying hens were fed with choline (500 and 4000 mg/kg diet) and, in a later trial, with rapeseed cake (0, 100 and 300 g/kg diet). Hens were genotyped for non-synonymous A/T polymorphism at position nt1034 of the cDNA of the FMO3 gene, which is associated with elevated TMA-levels in egg yolks. Genotypes were equally distributed among the experimental diets. TMA was extracted from yolks and measured as TMA-N by a colorimetric method. As expected, hens of the TT-genotype had high amounts of TMA-N in egg yolks when fed either 4000 mg choline/kg diet or the rapeseed cake diet, while hens of the AA-genotype showed low levels of TMA-N. Heterozygous AT-hens had a slightly increased level of TMA-N when fed with 4000 mg choline/kg diet but not with the rapeseed cake feed. Rapeseed cake can be fed to AA and AT hens without leading to fishy tainted eggs.  相似文献

Krebs II ascites cells have a low affinity uptake system for choline (Km = 36 μM, Vm = 76 nmol/min per 2·108 cells). Choline entered the cells and was rapidly phosphorylated (95% of total intracellular soluble label). Trans acceleration of labeled choline from cells preloaded with radiolabeled choline and postincubated in the presence of unlabeled choline indicates that choline transport in Krebs II ascites cells is carrier mediated. Ethanolamine competed for the choline carrier. The uptake was reduced by hemicholinium-3, iodoacetamide and ouabain. The mechanism of choline transport in Krebs Ii ascites cells is in agreement with a linear transport model.  相似文献

研究常见饲料原料中胆碱的生物学效价,为渔用配合饲料配制提供基础数据。试验选取团头鲂平均体重(3.50.1)g 990尾,以酪蛋白、明胶等原料配制4个氯化胆碱添加水平(0、1030、1230、1430 mg/kg)的纯合饲料,并在1030 mg/kg的基础上分别添加7种常见饲料原料(鱼粉、豆粕、菜粕、棉粕、次粉、麸皮、米糠),使得各组配合饲料中胆碱含量为1230 mg/kg,共11组,每组3重复,饲养8周。根据肝脏胆碱沉积量和试验鱼胆碱摄食量,评定团头鲂幼鱼对不同浓度的胆碱利用率和这7种原料中胆碱的生物学效价。结果表明:在4个纯和饲料中,肝脏胆碱沉积量和增重率随着饲料胆碱水平从0到1230 mg/kg饲料的增加而显著上升(P0.05),并在饲料胆碱水平从1230到1430 mg/kg饲料的增加而差异不显著(P0.05);在同一胆碱水平(1230 mg/kg)的条件下,原料组的增长率均高于对照组;团头鲂幼鱼对鱼粉、豆粕、菜粕、棉粕、次粉、麸皮和米糠的胆碱生物学效价分别为87.42%、112.54%、76.84%、98.00%、95.91%、43.88%、91.5%。分析可知,团头鲂生产饲料中尚需要额外添加氯化胆碱方能满足其对胆碱的需要,实际添加量与饲料所使用的原料有关。    相似文献

Qualitative and quantitative analyses have been made of the phospholipid composition of haemolymph from larvae of Musca domestica and of the two lipoprotein fractions separated from it by agarose-gel electrophoresis. The effect of rearing the larvae on defined diets containing adequate choline, no added choline, and choline plus 2-aminobutan-1-ol on this composition has been studied.The haemolymph lipoproteins have a phospholipid pattern similar to that of the unfractionated haemolymph. The chief component is phosphatidylethanolamine. Diglycerides and free sterols are the major neutral lipids present in the haemolymph and the separated lipoproteins.The different diets cause changes in the various phospholipids present in the two lipoproteins which are similar to those that occur in other tissues of the larvae. Choline deficiency increased the proportion of the haemolymph phospholipid that is associated with the lipoprotein having the slower electrophoretic mobility. The results are compared with those obtained from other insects and vertebrates and the rôle of the lipoprotein in choline deficiency in the housefly is discussed.  相似文献

The fate of radiolabelled campesterol, sitosterol and 24-methylenecholesterol fed in chemically-defined diets to honey bee (Apis mellifera L.) workers was determined. At various intervals, sterols of prepupae, newly emerged adults and queens were analyzed qualitatively, quantitatively and radiochemically and it was determined that there was not sufficient radioactivity associated with cholesterol and/or desmosterol in any of the samples to verify that any of the three C28 and C29 sterols was dealkylated and converted to cholesterol. Similarly, there was no evidence for the conversion of campesterol or sitosterol to 24-methylenecholesterol. It was concluded that the major portion of the sterols incorporated into the tissues of the brood larvae originated from the worker bees used to establish the colony. There is good evidence supporting the premise that the workers can make available sterols from their endogenous pools to the nutrient in the hive and that they can replenish these sterols with those from the artificial diet. The queen is also able to replenish sterols utilized in egg production from those obtained by the workers from the artificial diet, and at the end of nine weeks queens contained more than four times as much sterol, on a ‘μg sterol per g fresh weight’ basis, than was found in fertile queens at the beginning of the test period.  相似文献

On diets devoid of l-isoleucine or of l-valine, these larvae developed little, if at all, and died before reaching the larval-pupal ecdysial stage. Graded concentrations of l-isoleucine up to 6·0 mg/g or of l-valine up to 5·0 mg/g increased the rate of development and survival of this insect. The lipids of the adults and of several immature stages of the southwestern corn borer, Diatraea grandiosella, were studied after they were fed natural corn stalks or artificial diets. Linoleic acid (18:2) was the major fatty acid of the neutral lipids in both the natural and the artificial diets, but aleic acid (18:1) was the principal neutral lipid in all insect stages. Also, linoleic acid and oleic acid were the principal acids in the insect phospholipids of all stages. The content of linoleic acid in the natural diet was also high, but that in the artificial diet appeared to be much too low for insect requirements. Phosphatidyl choline (PC) and phosphatidyl ethanolamine (PE) were the major phospholipids in all growth stages. Thus, in larvae diapausing in the field, the unsaturated fatty acid content of PC was 59·3 per cent, primarily 16:1 and 18:1, and PE was 87·4 per cent, primarily 18:1, 18:2, and 18:3, and the fatty acids in the number 1- and 2-positions of PC were 53·6 and 97·2 per cent unsaturated, respectively. The haemolymph of diapausing southwestern corn borer larvae contained primarily glycerides but also had some PC and PE. Fat body from diapausing larvae contained primarily 16:0, 16:1, and 18:1 in a ratio of 1 : 1 : 2. Thus lipids of the southwestern corn borer do not reflect dietary lipids as closely as do other insects studied.  相似文献

—A method to achieve labelling of the acetylcholine stores of the brain under ideal physiological conditions is described. To this end, mice fed on a choline free diet were supplied with deuterium labelled choline in the drinking water. Labelled and unlabelled choline in plasma and in the brain as well as labelled and unlabelled acetyicholine in the brain were measured by a gas chromatographic-mass spectrometric method. It was found that after 1–25 days on the deuterium choline diet, substantial amounts of the plasma choline and brain acetylcholine were displaced by deuterium choline and deuterium acetylcholine, respectively. Already on the first day, the mole ratio of deuterium choline/total choline in plasma was 0·22, and it approached a maximum of 0·57 on the 14th day. The mole ratios of deuterium acetylcholine/total acetylcholine in the brain were slightly but significantly lower than those of deuterium choline/total choline in plasma 1–14 days, but asymptotically approached the mole ratios of deuterium Ch/total Ch in plasma by 25 days. Intact brains submitted to incubation at room temperature for 10 min increased their total choline content by about 500 per cent. Concurrently, in brains from animals kept on a deuterium choline diet for 1–2 days, the level of deuterium choline rose only by 50 per cent after incubation. Deuterium choline levels increased, however, by 200–300 per cent in the brains from animals kept on the deuterium diet for longer time periods. On the basis of these data it is suggested that: (a) choline in plasma is partly supplied from the food and partly from endogenous sources; (b) plasma choline rapidly equilibrates (less than one day) with a pool of Ch in the brain which is responsible for biosynthesis of acetylcholine; (c) the size of this choline pool is in the order of 34–40 nmol/g.  相似文献

Choline is required to make essential membrane phospholipids. It is a precursor for the biosynthesis of the neurotransmitter acetylcholine and also is an important source of labile methyl groups. Mammals fed a choline-deficient diet develop liver dysfunction; however, choline is not considered an essential nutrient in humans. Healthy male volunteers were hospitalized and fed a semisynthetic diet devoid of choline supplemented with 500 mg/day choline for 1 wk. Subjects were randomly divided into two groups, one that continued to receive choline (control), and the other that received no choline (deficient) for three additional wk. During the 5th wk of the study all subjects received choline. The semisynthetic diet contained adequate, but no excess, methionine. In the choline-deficient group, plasma choline and phosphatidylcholine concentrations decreased an average of 30% during the 3-wk period when a choline-deficient diet was ingested; plasma and erthrocyte phosphatidylcholine decreased 15%; no such changes occurred in the control group. In the choline-deficient group, serum alanine aminotransferase activity increased steadily from a mean of 0.42 mukat/liter to a mean of 0.62 mukat/liter during the 3-wk period when a choline-deficient diet was ingested; no such change occurred in the control group. Other tests of liver and renal function were unchanged in both groups during the study. Serum cholesterol decreased an average of 15% in the deficient group and did not change in the control group. Healthy humans consuming a choline-deficient diet for 3 wk had depleted stores of choline in tissues and developed signs of incipient liver dysfunction. Our observations support the conclusion and choline is an essential nutrient for humans when excess methionine and folate are not available in the diet.  相似文献
