Rifampin-resistant (Rifr) mutants were isolated spontaneously from Bacillus subtilis strain 168. A fraction of the mutants did not grow on a minimal medium. A high concentration of one of the L-amino acids (glutamic acid, glutamine, arginine, proline, aspartic acid, or asparagine) was required to restore their growth on the medium. Further analysis of one of the mutants (strain RF 161) suggested that the mutant is unable to use ammonia as a nitrogen source and requires amino acids instead. Activity of glutamate synthase was not detected in the crude extract of the mutant. The Rifr mutation was closely located to cysA and the drug resistance was cotransformed with the property of amino acid requirement at 100% frequency. All revertants to prototrophy tested showed the rifampin-sensitive (Rifs) property. The activity of the DNA-dependent RNA polymerase of the mutant was resistant to rifampin. It is concluded that some alteration of RNA polymerase may cause absence of the activity of an enzyme involved in the nitrogen metabolism. 相似文献

Summary Nicotine, administered by sub-cutaneous injection over a period of six weeks, was found to be ineffective in breaking male mouse chromosomes as judged by dominant lethal tests. Variation of different features of populations of streptomycin-sensitive and streptomycin-resistant forms of M. purpurea var. violacea, an organism producing gentamicin was studied. The population of the initial streptomycin-sensitive culture was characterized by high homogeneity with respect to the cultural, morphological and some physiological properties. The variation of the features, such as the colony size, pigment formation, auxotrophic mutations, antibiotic production significantly increased in populations grown on media with streptomycin. Mutants differing from the initial strain by a complex of cultural, morphological and physiological features and in particular the antibiotic production were isolated from populations of the streptomycin-resistant variants. 相似文献

The parameters of reproduction were studied in American minks that were homo- and heterozygous for the Black crystal mutation (Cr). The given mutation that changes the hair pigmentation was obtained de novo duration selection of American minks for tame behavior. Apart from changed color, the following reproduction disorders were detected in the Black crystal mutants, as compared to standard animals: (1) delayed entry into the reproductive season in females; (2) greater percentage in both homo- and heterozygous animals of females bearing no offspring; (3) greater early postnatal lethality indices. The most marked suppressing effect on the development of offspring was detected in homozygous females (Cr/Cr), whose progeny perished within a few days after birth. An analysis of reproduction in mutants and standard animals is presented. An attempt was made to explain the conceivable mechanisms that form the basis of disorders detected. 相似文献

Lonidamine, a dichlorinated derivative of indazole-3-carboxylic acid, has recently been indicated as an antiproliferative agent being able to reduce mitotic activity of tumor cells. We have evaluated lonidamine effect on proliferating, non tumor cells choosing as a model the male germ cells obtained from cultured seminiferous epithelium explants. The obtained germ cells are able to duplicate in vitro and we have found that lonidamine, at low doses, induces a significative inhibition of the incorporation of labelled thymidine into the duplicating germ cells. The effect seems to be specific for the germ cells since lonidamine does not affect duplicative ability of the somatic cells of the seminiferous tubules and of muscle fibroblasts. 相似文献

Flavodoxin (Fld) is a bacterial electron-transfer protein that possesses flavin mononucleotide as a prosthetic group. In the genomes of the Pseudomonas species, the mioC gene is the sole gene, annotated Fld, but its function remains unclear. In this study, phenotype microarray analysis was performed using the wild-type and mioC mutant of pathogenic Pseudomonas aeruginosa PAO1. Our results showed that the mioC mutant is very resistant to oxidative stress. Different antibiotics and metals worked differently on the sensitivity of the mutant. Other pleiotropic effects of mutation in the mioC gene, such as biofilm formation, aggregation ability, motility and colony morphology, were observed under iron stress conditions. Most of the phenotypic and physiological changes could be recovered in the wild type by complementation. Mutation of the mioC gene also influenced the production of pigments. The mioC mutant and mioC over-expressed complementation cells, over-produced pyocyanin and pyoverdine, respectively. Various secreted chemicals were also changed in the mutant, which was confirmed by (1) H NMR analysis. Interestingly, physiological alterations of the mutant strain were restored by the cell-free supernatant of the wild type. The present study demonstrates that the mioC gene plays an important role in the physiology of P.?aeruginosa and might be considered as a suitable drug target candidate in pathogenic P.?aeruginosa. 相似文献

A non-overlapping generation model is proposed which links Wright's adaptive topography concept to the rather inexact notion of Darwinian fitness as survival and reproduction. In general, evolution is seen to weight proportionate increases in survival twice as greatly as proportionate increases in fertility. Certain special cases are also delineated in which measurements of survival and fertility receive equal weight in the fitness equations. In addition, some implications of the present study for Wright's shifting balance theory and models based on Fisher's reproductive value are discussed. 相似文献

A Yersinia pestis fur mutation was constructed by insertionally disrupting the fur open reading frame. Analysis of a Fur-regulated beta-galactosidase reporter gene revealed a loss of iron regulation as a result of the fur mutation. trans complementation with the cloned Y. pestis fur gene restored iron regulation. The expression of most iron-regulated proteins was also deregulated by this mutation; however, a number of iron-repressible and two iron-inducible polypeptides retained normal regulation. Mutations in fur or hmsH, a gene encoding an 86-kDa surface protein required for hemin storage, increased the sensitivity of Y. pestis cells to the bacteriocin pesticin. Interestingly, the Y. pestis fur mutant lost temperature control of hemin storage; however, expression of the HmsH polypeptide was not deregulated. When grown with excess iron, a Y. pestis fur mutant possessing the 102-kb pigmentation locus exhibited severe growth inhibition and a dramatic increase in the number of spontaneous nonpigmented chromosomal deletion mutants present at late log phase. These results suggest that the Fur protein of Y. pestis is an important global regulator and that a separate Fur-independent iron regulatory system may exist. 相似文献

Models of the evolution of gynodioecy assume that inbreeding affects male and female fertility equally and ignore quantitative variation in sex expression. The objectives of this study were to assess inbreeding effects, genetic background, and plant maturity on male and female fertility and the mechanism of male sterility inheritance for Nemophila menziesii (Hydrophyllaceae). Frequency of male-sterile flowers, number of anthers and ovules, and percentage of viable pollen were measured on plants from different pedigrees and five inbreeding levels (F = 0, 0.0625, 0.25, 0.5, and 0.75). Quantitative variation in male sterility was evident. As inbreeding increased, anther and ovule number decreased; the effect on anther number was greater than on ovule number. Pedigrees varied in number of male-sterile flowers and inbreeding effects. Frequency of male-sterile flowers was greatest among first flowers. No trade-off between male and female fertility was detected. A model attributing male sterility to a cytoplasmic locus and restoration to male fertility to a nuclear locus accounted for the distribution of complete sterility and hermaphroditism over the pedigrees. This study suggests that models of the evolution and maintenance of gynodioecy should allow for quantitative variation in male and female fertility components due to inbreeding, pedigree, and plant maturity. 相似文献

Pleiotropic effects of a null mutation in the c-fos proto-oncogene. 总被引:39,自引:0,他引:39
The c-fos proto-oncogene has been implicated as a central regulatory component of the nuclear response to mitogens and other extracellular stimuli. Embryonic stem cells targeted at the c-fos locus have been used to generate chimeric mice that have transmitted the mutated allele through the germline. Homozygous mutants show reduced placental and fetal weights and significant loss of viability at birth. Approximately 40% of the homozygous mutants survive and grow at normal rates until severe osteopetrosis, characterized by foreshortening of the long bones, ossification of the marrow space, and absence of tooth eruption, begins to develop at approximately 11 days. Among other abnormalities, these mice show delayed or absent gametogenesis, lymphopenia, and altered behavior. Despite these defects, many live as long as their wild-type or heterozygous littermates (currently 7 months). These data indicate that c-fos is not required for the growth of most cell types but is involved in the development and function of several distinct tissues. 相似文献

At different parts of the world, Red Seaweeds are one component of human diets especially at Southeast Asia. Red Seaweeds structurally contain bioactive molecules so; we studied the effect of Chondrus crispus on increasing the male albino rat fertility. Twelve male albino rats are used in this study as two group pre-treated group and post- treated one each with 6 animals. The pretreated group was dissected before the post-treated group injection. Each post treated rat injected intramuscular with 1 mg of Chondrus crispus with dose 0.1 ml/ twice per week for 48 day (Mukhtar et al., 2013).The results showed that increasing on the total testosterone levels insignificantly, sperm motility significantly, and decreasing in both FSH and DPPH levels insignificantly and significantly for the MDA levels in the post-treated group. The morphological appearance and histological examination for the sperm, testis and liver were normal as the pretreated group. The molecular studies showed absence of any DNA fragmentation for the testis of both group.The Red Seaweed has an enhanced effect in the testicular function of the animal which might increase their fertility and sexual activities. 相似文献

Ochi (Agric. Biol. Chem. 51:829-835, 1987) has isolated a relaxed mutant of Streptomyces antibioticus, designated relC49, relC49 accumulates significantly lower levels of ppGpp than the parent stain, IMRU3720. At its maximum, the ppGpp level in relC49 was only one-fourth that observed in strain IMRU3720. Interestingly, a burst of ppGpp synthesis between 18 and 22 h of growth in IMRU3720 coincided with the onset of actinomycin production in that strain. As shown previously, the activity in protein synthesis of ribosomes from strain IMRU3720 decreases with the age of the culture. The decrease in activity was less pronounced in cultures of relC49. relC49 mycelium contains reduced levels of phenoxazinone synthase, a key enzyme involved in actinomycin biosynthesis. The rel mutation prevents the normal increase in the activity of one of the other enzymes required for production of the antibiotic, 3-hydroxyanthanilate-4-methyltransferase, and a third enzyme, actinomycin synthetase I, appears to be completely absent from relC49 mycelium. Levels of phenoxazinone synthease mRNA were examined by RNA dot blotting with the cloned phenoxazinone synthase gene as a probe. mRNA levels for phenoxazinone synthase were dramatically reduced in relC49 compared with strain IMRU3720. These results are discussed in terms of the possible regulation of the onset of actinomycin production by ppGpp. 相似文献
