Recent DNA sequence analyses indicated that Nigrosabulum globosum is a cleistothecial representative of the Bionectriaceae in the Hypocreales, but morphological characters supporting this relationship are unknown. Using light and electron microscopy we followed the development of the ascomata of this species, from the formation of gametangia through to the development of mature ascospores, and observed a series of characters that confirmed its hypocrealean affinities. These included the formation of a gel-filled centrum during early stages of ascoma development, the subsequent appearance of hyaline peridial tissue enclosed within a layer we interpret as representing a melanized uniloculate stroma, apically derived paraphyses, and an ascogenous system that gives rise to asci that were both cylindrical to clavate and globose. Ascospores, previously reported to be smooth, were ornamented with a honeycomb-like reticulum and were able to germinate within the ascoma. The carbonaceous outer (stromatic) walls of the mature, grit-like cleistothecia indicate possible resistance to UV radiation and desiccation. Furthermore, the complement of germinated ascospores would enable mature ascomata to function as propagules that could quickly initiate new growth when transferred to fresh substrate. Our reexamination of N. globosum also provides data that support the hypothesized close relationship with other bionectriaceous, cleistothecial coprophiles, i.e., species of Hapsidospora, and Bulbithecium in particular.  相似文献

A single, permineralized ascoma resembling a pseudothecium assignable to the Pleosporales is described from the Eocene Appian Way fossil locality on Vancouver Island, British Columbia. The ascoma is globose, ostiolate, and erumpent on a fragment of the bark from an unidentified seed plant. Basally arranged asci contain large, multicelled, obovate ascospores within a single cavity or locule enclosed by a two-layered pseudoparenchymatous tissue that ostensibly represents ascostroma. Given this interpretation of the specimen's morphological features, Margaretbarromyces dictyosporus gen. sp. nov. represents the first report of a corticolous pleosporalean ascoma in the fossil record.  相似文献

Sclerotinia species are sexually reproducing ascomycetes. In the past S. minor and S. sclerotiorum, have been assumed to be homothallic because of the self-fertility of colonies derived from single ascospores. S. trifoliorum has previously been shown to be bipolar heterothallic due to the presence of four self-fertile and four self-sterile ascospores within a single ascus [Uhm, J.Y., Fujii, H., 1983a. Ascospore dimorphism in Sclerotinia trifoliorum and cultural characters of strains from different-sized spores. Phytopathology 73: 565–569]. However, isolates of S. minor and S. sclerotiorum were proven to be homothallic ascomycetes, by self-fertility of all eight ascospores within an ascus. Apothecia were raised from all eight ascospores of a single tetrad from four isolates of S. minor and from an isolate of S. sclerotiorum, indicating that inbreeding may be the predominant breeding mechanism of S. minor. Ascospores from asci of S. minor and S. sclerotiorum were predominantly monomorphic, but rare examples of ascospore dimorphism similar to S. trifoliorum were found.  相似文献

Data from microscopic morphology, single-spore cultures, and DNA analyses of teleomorphs and anamorphs support the recognition of five species of Prosthecium with Stegonsporium anamorphs on Acer: P. acerinum sp. nov., the teleomorph of S. acerinum; P. acerophilum comb. nov., formerly known as Dictyoporthe acerophila; P. galeatum comb. nov., originally described as Massaria galeata; P. opalus sp. nov.; and P. pyriforme sp. nov., the teleomorph of S. pyriforme s. str. The morphology of both type specimens and freshly collected material was investigated. The teleomorphs have brown ellipsoidal ascospores with five distosepta and often a longitudinal distoseptum. The anamorphs of all species described here belong to Stegonsporium; their connection to the Prosthecium teleomorphs was demonstrated by morphology and DNA sequences of single spore cultures derived from both ascospores and conidia. The anamorphs and teleomorphs of all five Prosthecium species are described and illustrated by LM images, and a key to these species is provided. As perceived from this work, S. pyriforme is restricted to Europe and does not occur in North America, whereas S. acerinum is restricted to North America, not found in Europe. The host associations given in the literature are revised and evidence is provided that only A. opalus, A. pseudoplatanus, and A. saccharum are confirmed hosts of Prosthecium with Stegonsporium anamorphs. Molecular phylogenetic analyses of tef1, ITS rDNA, and partial nuLSU rDNA sequences confirm that the species with Stegonsporium anamorphs are closely related to P. ellipsosporum, the generic type species. Stilbospora macrosperma is confirmed as the anamorph of P. ellipsosporum by DNA data of single spore isolates obtained from both ascospores and conidia.  相似文献

采用扫描电镜对秦岭北坡楼观台地区的厚皮、薄皮两种类型栓皮栎软木进行细胞微观构造观察分析,并与欧洲栓皮槠进行比较,以阐明厚皮、薄皮栓皮栎软木的相关特性,为中国栓皮栎软木的合理利用提供依据。结果表明:(1)两种类型栓皮栎软木细胞的排列结构较一致,均由内部中空的封闭型薄壁细胞紧密排列组成;在弦切面上呈蜂窝状排列,径切面和横切面上呈砖墙状排列;在径切面上,软木细胞侧高整齐地排列成行,且与树干轴向垂直;在横切面上,软木细胞侧高整齐地处于以树干轴为中心散发出来的射线上。(2)栓皮栎软木细胞大小、细胞壁和侧壁褶皱等受生长季节的影响;从软木细胞形态特征上看,厚皮类型软木细胞壁薄、细胞体积大,其软木质量优于薄皮类型。(3)与欧洲栓皮槠比较,发现厚皮类型栓皮栎早软木细胞棱柱高较小(20.6μm vs.(对比)30~40μm),软木细胞壁略厚(1.7μm vs.1~1.5μm),细胞实体积(细胞壁体积占细胞总体积比例)略大(18.75%vs.10%),厚皮类型栓皮栎软木比欧洲栓皮槠的软木质量差一些。(4)受树皮生长应力的影响,两种类型栓皮栎软木细胞侧高壁上多发生褶皱,早软木细胞褶皱严重,晚软木细胞没有褶皱,但在早晚软木交界或含有杂质处褶皱特别严重,表明厚皮类型软木细胞的侧壁褶皱程度高于薄皮类型。(5)对细胞形态特征及软木特性等的分析表明,薄皮类型栓皮栎软木质量比厚皮类型差,未来对软木资源的开发利用应更注重厚皮类型。  相似文献

The new genus Corylomyces, isolated from the surface of a hazelnut (Corylus avellana) in the French Pyrenees, is described, illustrated and compared with morphologically similar taxa. It is characterised by tomentose, ostiolate ascomata possessing long necks composed of erect to sinuose hairs, and one- or two-celled, opaque, lunate to reniform ascospores. Analyses of the SSU and LSU fragments rDNA gene sequences support its placement in the Lasiosphaeriaceae (Sordariales).  相似文献

Two new species ofConiochaeta, isolated from Japanese soils, are described and illustrated:C. cephalothecoides, which is characterized by dark brown ascomata clothed with short setae, cylindrical asci and ovoid to almond-shaped or pyriform ascospores with a longitudinal germ slit; andC. dumosa, which is characterized by dark brown ascomata clothed with short setae and dense hyphal hairs, cylindrical asci and ellipsoid-fusoid ascospores with a longitudinal germ slit. These species are distinguished from most species of the genus by the unique cephalothecoid peridium of their ascomata. Neosartorya indohii and N. tsurutae, two new Neosartorya species isolated from tropical rainforest soil in the Amazonian area, Brazil, are described and illustrated. Neosartorya indohii is characterized by its spreading growth on Czapeks and malt extract agars, light yellow cleistothecia, broadly lenticular ascospores with two conspicuously serrate-incised equatorial crests and tuberculate convex surfaces, and globose to subglobose conidia with a smooth wall. Neosartorya tsurutae is characterized by its spreading growth on Czapeks and malt extract agars, white cleistothecia, broadly lenticular ascospores with four equatorial crests and rugulose-ruminate convex surfaces, and ovoid to broadly ellipsoidal conidia with a smooth wall.  相似文献

Two morphologically similar groups of ascomycetes with globose to subglobose perithecia, elongate necks, unitunicate asci floating freely at maturity, and hyaline ascospores currently placed in Calosphaeria s. lat. and Ceratostomella s. lat., respectively, are studied. The Calosphaeria-like fungi have groups of perithecia growing between cortex and wood, arranged in circular groups with converging necks and piercing the cortex in a common point; the asci with a shallow apical ring and U- to horseshoe-shaped hyaline ascospores are compared with Calosphaeria pulchella, the type species of the genus. Conidiogenesis of the investigated Calosphaeria-like fungi is holoblastic-denticulate; ramichloridium-like and sporothrix-like conidiophores and conidia were formed in vitro. Ascospore and ascus morphology, structure of the ascal apex, ascogenous system, mode of conidiogenesis and the large subunit rRNA sequences of this group differ considerably from C. pulchella and both groups are unrelated. Thus a new genus, Tectonidula, is described with two accepted species, T. hippocrepida and T. fagi; they are separated by ascospore and ascus morphology and holoblastic-denticulate conidiogenesis from the core species of Calosphaeria. The placement of Tectonidula among perithecial ascomycetes is discussed. The relationship of Tectonidula with Barbatosphaeria and two ramichloridium-like hyphomycetous genera Rhodoveronaea and Myrmecridium is investigated. Three species formerly attributed to Ceratostomella are studied. The revision of the herbarium type specimen and fresh material of Ceratostomella ligneola revealed that it is conspecific with Ceratostomella ampullasca and Ceratostomella similis. The LSU phylogeny clearly separated C. ligneola from Ceratostomella s. str. and morphologically similar Lentomitella. On the basis of molecular sequence data and detailed comparison of morphology of asci, ascospores and ascogenous system the genus Natantiella is described for C. ligneola with C. ampullasca and C. similis as its synonyms. Natantiella produced sterile mycelium in vitro.  相似文献

The new genus and species Teracosphaeria petroica is described for a perithecial ascomycete and its anamorph occurring on decayed wood collected in New Zealand. The fungus produces immersed, non-stromatic ceratosphaeria-like perithecia in nature, with hyaline, septate ascospores produced in unitunicate, non-amyloid asci. The anamorph produced in vitro is phialophora-like with lightly pigmented phialides terminating in flaring, deep collarettes that are often noticeably brown with conspicuous periclinal thickening. Phylogenetic analysis of LSU rDNA sequence data indicates that this fungus is distinct from morphologically similar fungi classified in the Chaetosphaeriales, the Trichosphaeriales or the Magnaporthaceae. It forms a monophyletic group with recently described, chaetosphaeria-like ascomycetes, such as the pyrenomycete genus Mirannulata, and shows affinity with the anamorphic species Dictyochaeta cylindrospora. The usefulness of describing anamorph genera for morphologically reduced anamorphs, when anamorph characteristics are actually part of the holomorph diagnosis, is discussed. An apparently contradictory example of the so-called Cordana and Pseudobotrytis anamorphs of Porosphaerella spp. is also discussed.  相似文献

The closely related genera Carestiella, Schizoxylon and Stictis (Stictidaceae, Ostropales, Ascomycota) are revised in northern Scandinavia, and a revised phylogenetic hypothesis, including all seven species of this group in the area, is presented. A key to the species of Stictis s. lat. (including Carestiella, Schizoxylon and Stictis) in Northern Scandinavia is presented. S. albescens and S. confusum are described as new to science, and the new name S. brunnescens is introduced for S. mollis ssp. populorum, when treated as a distinct species. C. socia and S. albescens are the only representatives of their genera in northern Scandinavia, but a total of five species of Stictis are present in the area (S. brunnescens, S. confusum, S. mollis, S. populorum and S. radiata). Substantial within-species variation in ascoma morphology and wall pigmentation is revealed and discussed, and found to correlate with fungal lifestyles; optional lichenization apparently affects several details in fungal anatomy and morphology.  相似文献

Erysiphe cumminsiana andE. galeopsidis, which have immature asci in the current season, have been recorded from Japan, but the ascospores of both fungi have not been described. In the present experiments, some observations before and after overwintering were made on the cleistothecia ofE. cumminsiana on three species ofCacalia and two species ofLigularia, andE. galeopsidis onGeranium thunbergii. After overwintering, the former fungus developed six to eight, rarely four spores in an ascus and the latter fungus always four spores in an ascus. 采用半薄切片技术和组织化学染色法对宁夏枸杞大孢子发生和雌配子体发育过程中的细胞结构变化及营养物质积累特征进行了观察。结果表明,(1)宁夏枸杞为中轴胎座,多室子房,倒生胚珠,单珠被,薄珠心类型。(2)位于珠心表皮下的孢原细胞可直接发育为大孢子母细胞,减数分裂后形成直线型大孢子四分体,合点端第一个大孢子发育为功能大孢子,胚囊发育类型为蓼型,具有珠被绒毡层。(3)初形成的胚囊外周组织中没有营养物质积累,成熟胚囊时期出现了大量的淀粉粒且呈珠孔端明显多于合点端的极性分布特征。(4)助细胞的珠孔端具有明显的丝状器结构,呈PAS正反应表现出多糖性质,成熟胚囊具有承珠盘结构。  相似文献

Taphrina entomospora is one of the few species of the genus described on native plants of the Southern Hemisphere and also one of the few leaf pathogens known on Nothofagus species. The anatomical changes it produces on N. pumilio leaves, and its morphology, cytology, and sporogenesis were studied. The fungus is a perennial species that overwinters as mycelium in the foliar buds and infects the developing leaves, so the whole blade develops the disease symptoms. Interveinal areas of the leaves become chlorotic, thickened and rounded. Palisade parenchyma fails to develop, with spongy parenchyma developing as packed, rounded, isodiametric cells with little intercellular space. The mycelium is subcuticular, dikaryotic, and produces ascogenous hyphae, asci, and ascospores as described for other species in the genus. Before ascus discharge, ascospores bud in a regular, unique way. The life-cycle of T. entomospora is compared with other representative taxa in the genus and the distribution of this pathogen is discussed.  相似文献

Discula betulina is a foliar pathogen on birch (Betula) and Gnomonia intermedia is found on overwintered birch leaves. Perithecia of G. intermedia developed in vitro on colonies of D. betulina isolated from birch tissues in late summer, and single ascospores of G. intermedia consistently developed into colonies similar to D. betulina, producing typical D. betulina conidia. Isolates of D. betulina could be grouped into two mating types, which produced fertile perithecia of G. intermedia when mated with each other. Mycelia from single-ascospore and single-conidial isolates were inoculated onto shoots of downy birch, causing lesions and die-back from which D. betulina was consistently isolated. ITS region ribosomal DNA sequence analysis confirmed the results of the morphological and mating studies, and found that the closest known relatives of G. intermedia/D. betulina are Gnomoniella nana and Sirococcus clavigignenti-juglandacearum. The conclusion from these studies is that D. betulina is the anamorph of G. intermedia.  相似文献

Chaetosphaeria tortuosa is described as the newly discovered teleomorph of Menispora tortuosa, based on specimens from Canada and the Czech Republic, and single spore isolations from both morphs. The fungus produces superficial, more or less globose, papillate, dark brown to black smooth perithecia (200–)220–250 × (220–)230–260 μm. The asci are unitunicate, 8-spored, cylindrical-fusiform, (110–)120–133(–145) × 12–14 with a distinct apical, nonamyloid annulus 1–1.5 μm high, 3.5–4 μm wide. The ascospores are fusiform, 19–24 × 5–6 μm, hyaline, 3-septate, smooth, and 2-seriate in the ascus. The morphology of the teleomorph and anamorph are similar to that of C. ovoidea (anamorph: M. glauca), differing in dimensions of asci and ascospores, and in the disposition and morphology of the phialides of the anamorphs. The generic concept and phylogeny of Menispora is briefly discussed, and a key to the 11 species currently accepted in the genus is provided.  相似文献
