Microsatellite loci were isolated from a Mahonia aquifolium cultivar. We describe the variability of 10 loci in invasive European and native North American M. aquifolium and their transspecies amplification in native Mahonia repens and Mahonia pinnata from North America and one species of the related genus Berberis (Berberis vulgaris), native to Europe. The markers should be useful to reveal the genetic origin of invasive Mahonia populations and differences in the genetic make up between invasive and native populations.  相似文献

The ability of an insect to disperse to new habitat patches is difficult to quantify, but key to the establishment and persistence of populations. In this study, we examined dispersal of the phytophagous chrysomelid beetle, Galerucella calmariensis, which is currently being introduced into North America for the biological control of purple loosestrife (Lythrum salicaria), an aggressive wetland weed. We used a mark, release, and recapture approach to determine how rates of colonization of host patches by this beetle are influenced by the distance of the patch from the source of dispersers, and by the presence of conspecifics at the patch. We released color-coded beetles at six distances from a long, linear patch of purple loosestrife that was divided into segments with and without conspecifics. We observed initial flight directions as beetles left the release points and collected all beetles that settled at the target patch. We found a bias in initial flight toward the target for distances up to 50 m. Over the 7 days of the experiment, beetles arrived at the target from all release points, including the farthest release point, 847 m away. G. calmariensis was strongly attracted to conspecifics when settling after dispersal; 86% of the 582 recovered beetles came from the segments inhabited by conspecifics. The probability of an individual arriving at the patch declined steeply with release distance. This relationship fits a model in which beetles move in a random direction and stop if they intercept the target patch, and where beetles are lost at a constant rate with distance travelled. The dispersal and patch-colonizing behavior of G. calmariensis is likely to have important consequences for the biological control program against purple loosestrife. Two Fix mutants of pea (Pisum sativum L.) which are unable to fix molecular nitrogen, E135f (sym-13) and Sprint-2Fix (sym-31), were crossed to create the doubly homozygous recessive line, named RBT (sym-13, sym-31). The ultrastructural organization of nodules of the RBT line was compared with that of each of the two parental mutant lines and with the original wild-type genotypes of the cultivars Sparkle and Sprint-2. It was shown that the RBT line is similar to the mutant line Sprint-2Fix in having abnormal symbiosome composition and bacteroids with relatively undifferentiated morphology. Because the phenotypic manifestation of the sym-31 mutant allele suppresses the phenotypic manifestation of the sym-13 mutant allele, it is concluded that the function of the gene Sym-31 (which is mutated in the Sprint-2Fix line) is necessary at an earlier stage of symbiosome development than the gene Sym-13 (which is mutant in the E135f line). Canopy CO2 concentrations in a tropical rainforest in French Guiana were measured continuously for 5 days during the 1994 dry season and the 1995 wet season. Carbon dioxide concentrations ([CO2]) throughout the canopy (0.02–38 m) showed a distinct daily pattern, were well-stratified and decreased with increasing height into the canopy. During both seasons, daytime [CO2] in the upper and middle canopy decreased on average 7–10 μmol mol−1 below tropospheric baseline values measured at Barbados. Within the main part of the canopy (≥ 0.7 m), [CO2] did not differ between the wet and dry seasons. In contrast, [CO2] below 0.7 m were generally higher during the dry season, resulting in larger [CO2] gradients. Supporting this observation, soil CO2 efflux was on average higher during the dry season than during the wet season, either due to diffusive limitations and/or to oxygen deficiency of root and microbial respiration. Soil respiration rates decreased by 40% after strong rain events, resulting in a rapid decrease in canopy [CO2] immediately above the forest floor of about 50␣μmol mol−1. Temporal and spatial variations in [CO2]canopy were reflected in changes of δ13Ccanopy and δ18Ocanopy values. Tight relationships were observed between δ13C and δ18O of canopy CO2 during both seasons (r 2 > 0.86). The most depleted δ13Ccanopy and δ18Ocanopy values were measured immediately above the forest floor (δ13C = −16.4‰; δ18O = 39.1‰ SMOW). Gradients in the isotope ratios of CO2 between the top of the canopy and the forest floor ranged between 2.0‰ and 6.3‰ for δ13C, and between 1.0‰ and 3.5‰ for δ18O. The δ13Cleaf and calculated c i/c a of foliage at three different positions were similar for the dry and wet seasons indicating that the canopy maintained a constant ratio of photosynthesis to stomatal conductance. About 20% of the differences in δ13Cleaf within the canopy was accounted for by source air effects, the remaining 80% must be due to changes in c i/c a. Plotting 1/[CO2] vs. the corresponding δ13C ratios resulted in very tight, linear relationships (r 2 = 0.99), with no significant differences between the two seasons, suggesting negligible seasonal variability in turbulent mixing relative to ecosystem gas exchange. The intercepts of these relationships that should be indicative of the δ13C of respired sources were close to the measured δ13C of soil respired CO2 and to the δ13C of litter and soil organic matter. Estimates of carbon isotope discrimination of the entire ecosystem, Δe, were calculated as 20.3‰ during the dry season and as 20.5‰ during the wet season. The aim of this study was to investigate quality coding of blend ratios of binary mixtures by olfactory receptor cells in the spiny lobster. Three odorants (adenosine-5′-monophosphate, l-glutamate, and taurine) at 0.1–100 μmol · l−1 and seven blend ratios of each of their binary mixtures at a total concentration of 100 μmol · l−1 were used. The olfactory cells recorded (n = 48) evoked across-neuron patterns for single odorants that were well separated from each other. Across-neuron patterns varied with stimulus concentration but less than with stimulus type. Blend ratios of the three mixtures evoked across-neuron patterns that were orderly placed within a continuum between those elicited by the components. Mixture interactions, defined as a lack of independent effects by a mixture's components, occurred in 25, 24 and 37% of responses to blend ratios of glutamate/taurine, adenosine-5′-monophosphate/taurine, and glutamate/adenosine-5′-monophosphate, respectively. These mixture interactions did not have a large enough effect on the across-neuron patterns for the mixtures such they would be novel relative to those of the single components. These results suggest that despite mixture interactions the quality of individual compounds is not lost when mixed. Sieve-tube exudate protein (STEP) from Ricinus communis L. seedlings consists of a characteristic set of more than 100 different polypeptides, against which a complex antiserum was raised. This antiserum cross-reacted with dominant protein species (molecular weights 10–30 kDa) present in the sieve-tube exudate and, to a lesser extent, with proteins in tissue extracts of Ricinus and a wide range of other plant species. For further elucidation of the nature of individual STEPs in the sieve tubes the anti-STEP serum was used to screen a cDNA expression library constructed from Ricinus cotyledon mRNA. Two clones that differed in the 3′ untranslated region encoded a protein of 11 kDa which showed striking homology to bacterial and eucaryotic glutaredoxin sequences. Glutaredoxin activity was confirmed for the recombinant protein after overexpression in Escherichia coli and characterised in detail in sieve-tube exudate. Michaelis Menten constants (K m) for reduced glutathione and cysteine were 2 mM and 50 μM, respectively. Besides l-cysteine, dehydroascorbate and protein disulphides were also reduced by the activity present in the sieve-tube exudate. Glutathione, which is the obligate donor of reduced thiols for glutaredoxin, was present in sieve-tube sap in millimolar concentrations (up to 3 mM) with a ratio of total to oxidised glutathione of 3:1. It is suggested that glutaredoxin and glutathione in sieve tubes prevent oxidative damage and may be involved in redox regulation of sieve-tube proteins. Whole body calcium influx, branchial calcium efflux, and renal Ca2+ excretion were measured in rainbow trout (Oncorhynchus mykiss) exposed to hypercapnia. These experiments were performed to assess the potential impact on Ca2+ balance of the changes in gill morphology known to accompany respiratory acidosis in this species. After 48 h of hypercapnia, gill filamental chloride cell fractional area was significantly reduced. Despite this reduction and the presumed involvement of the chloride cell in calcium influx, whole body calcium influx was increased after 12 h of hypercapnia and remained elevated for 48 h. Branchial calcium efflux was unaltered during hypercapnia exposure, whereas renal Ca2+ excretion was elevated over preflux values only at 6 h of hypercapnia. Measurement of the kinetics of whole body calcium influx after 48 h of hypercapnia revealed a significant increase in the maximal uptake rate of Ca2+, yet the affinity constant of Ca2+ uptake was unaffected. Measurements of high-affinity Ca2+ -ATPase activities and ATP-dependent Ca2+ transport of gill basolateral membrane vesicles revealed that the ATP-dependent Ca2+ extrusion mechanism of the gills was not affected by hypercapnia. The results of the present study clearly show that the reduced chloride cell surface area that accompanies hypercapnia in trout does not impair calcium homeostasis. Although adjustments to the basolateral membrane high affinity Ca2+ transporter do not appear to play a role, the mechanism(s) underlying the maintenance of calcium homeostasis under hypercapnic conditions are unresolved. Effects of egg size and parental quality on lapwing Vanellus vanellus chick survival were studied in southwestern Sweden over 6 years. Chicks from large eggs were heavier at hatching and survived significantly better than those from small eggs. To control for the confounding effect of parental quality on egg size and chick survival, we performed a cross-fostering experiment during 2 years, exchanging clutches between nests with large and small eggs. In control clutches, chicks from large eggs survived better than those from small eggs, but we found no significant difference in chick survival between exchanged clutches. Thus, egg size did not affect chick survival independently of parental quality. Fledging success increased with parental age and/or experience, and with female body mass. Whether seed consumers affect plant establishment is an important unresolved question in plant population biology. Seed consumption is ubiquitous; at issue is whether seedling recruitment is limited by safe-sites or seeds. If most seeds inhabit sites unsuitable for germination, post-dispersal seed consumption primarily removes seeds that would otherwise never contribute to the population and granivory has minimal impacts on plant abundance. Alternatively, if most seeds ultimately germinate before they lose viability, there is greater potential for seed consumption to affect plant recruitment. Of the many studies on seed consumption, few ask how seed loss affects seedling recruitment for species with long-lived seed banks. We examined post-dispersal seed predation and seedling emergence in bush lupine (Lupinus arboreus), a woody leguminous shrub of coastal grasslands and dunes in California. We followed the fate of seeds in paired experimental seed plots that were either protected or exposed to rodent granivores in grassland and dune habitats. Significantly more seeds were removed by rodents in dunes than grasslands. In dunes, where rodent granivory was greatest (65% and 86% of seeds removed from plots by rodents in two successive years), there is a sparse seed bank (6.6 seeds m−2), and granivory significantly reduced seedling emergence (in the same two years, 18% and 19.4% fewer seedlings emerged from exposed versus protected plots), suggesting seed rather than safe-site limited seedling recruitment. In contrast, rodents removed an average of 6% and 56% of seeds from grassland plots during the same two years, and the grassland seed bank is 43-fold that of the dunes (288 seeds m−2). Even high seed consumption in the second year of the study only marginally influenced recruitment because seeds that escaped predation remained dormant. Burial of seeds in both habitats significantly reduced the percentage of seeds removed by rodents. In the forest understorey, shrubs usually reproduce vegetatively rather than sexually, but the relative contribution of these two reproductive modes may vary with temporal changes in environmental conditions (e.g., light, substrates of forest floor) that are closely related to forest dynamics. To evaluate the occasional changes in reproductive modes of the clonal understorey shrub Hydrangea paniculata, the reproductive characteristics (i.e., flowering, seedling establishment, vegetative propagation, reproductive size and age) were investigated in four different developmental stages in a Japanese beech forest (early gap, EG; late gap, LG; building, BU; mature, MA). Flowering individuals occurred only in EG and LG, and a much greater number of seedlings was observed in both EG and LG than in BU and MA, mainly because canopy gaps provided sufficient light for flowering and suitable substrates (fallen trees and exposed mineral-soil) for seedling establishment. The flowering individuals originated from plants that had persisted in the shaded understorey until gap formation. In contrast to sexual reproduction, clonal fragmentation increased with forest development in the order LG, EG, BU, MA. This is because, in later stages of forest dynamics (BU and MA), large individuals, some of which had previously flowered in gap stages, were layered on the forest floor and subsequently produced a substantial number of clonal fragments by separation of the buried branches from the main stems which had decayed. This resulted in a large number of clonal fragments originating from a single mother plant. These results suggest that the reproductive modes of H. paniculata is strongly influenced by the changes in environmental conditions with respect to the dynamics of canopy trees. Reproduction from seed in gap stages, despite their short period, provide new genets in established populations, whereas vegetative propagation via fragmentation in closed-canopy stages enhance the stability of the population until the next disturbance.  相似文献

The influence of environmental variation on the demography of clonal organisms has been poorly studied. I utilise a matrix model of the population dynamics of the intertidal zoanthid Palythoa caesia to examine how density dependence and temporal variation in demographic rates interact in regulating population size. The model produces realistic simulations of population size, with erratic fluctuations between soft lower and upper boundaries of approximately 55 and 90% cover. Cover never exceeds the maximum possible of 100%, and the population never goes to extinction. A sensitivity analysis indicates that the model’s behaviour is driven by density dependence in the fission of large colonies to produce intermediate sized colonies. Importantly, there is no density-dependent mortality in the model, and density dependence in recruitment, while present, is unimportant. Thus it appears that the main demographic processes which are considered to regulate population size in aclonal organisms may not be important for clonal species. This investigation examined the influence of soil moisture and associated parameters on the cold hardiness of the Colorado potato beetle (Leptinotarsa decemlineata Say), a temperate-zone species that overwinters in terrestrial burrows. The body mass and water content of adult beetles kept in sand at 4 °C varied over a 16-week period of diapause according to the substratum's moisture content. Changes in body water content, in turn, influenced the crystallization temperature (range −3.3 to −18.4 °C; n = 417), indicating that environmental moisture indirectly determined supercooling capacity, a measure of physiological cold hardiness. Beetles held in dry sand readily tolerated a 24-h exposure to temperatures ranging from 0° to −5 °C, but those chilled in sand containing as little as 1.7% water (dry mass) had elevated mortality. Thus, burrowing in dry soils not only promotes supercooling via its effect on water balance, but may also inhibit inoculative freezing. Mortality of beetles exposed to −5 °C for 24 h was lower in substrates composed of sand, clay and/or peat (36–52%) than in pure silica sand (78%) having an identical water content (17.0% dry mass). In addition to moisture, the texture, structure, water potential, and other physico-chemical attributes of soil may strongly influence the cold hardiness and overwintering survival of burrowing insects. In the Hawaiian Islands, native Heteropogon contortus (pili grass) is being replaced by alien grasses, one of which is Pennisetum setaceum (fountain grass). Both grasses depend on seeds for population growth. To help understand factors promoting the spread of the alien and decline of the native, we investigated the effects of physical disturbance, nutrient addition, and seed supplementation on seedling recruitment in experimental field plots. In the first year, our field site experienced an unusual drought, and seedling recruitment was greater for H. contortus than for P. setaceum under all treatments. Disturbance increased recruitment of H. contortus seedlings during some sampling periods. Recruitment was not significantly increased by seed additions for either species despite our finding of only 49 and 4 seeds m–2 in the seed bank for H. contortus and P. setaceum, respectively. In the first year, most P. setaceum seedlings died between monthly surveys. We resurveyed our field plots in a second, wetter year and found the pattern was reversed: recruitment of P. setaceum seedlings was greater than H. contortus seedlings in most treatments. Greenhouse comparisons of seedling survival under three drought regimes (water every 5,7 and 10 days) revealed that H. contortus seedlings tolerate drought better than P. setaceum seedlings. Seedling recruitment for these species in the leeward Hawaiian Islands appears to be primarily dependent on water availability, with the alien having the advantage in wetter years. Once seedlings of the long-lived alien become established, the alien seems capable of maintaining its dominance over H. contortus, even during periods of drought.  相似文献

Photoperiodic time measurement regulating larval diapause in the pitcher-plant mosquito, Wyeomyia smithii, varies in a close relationship with latitude. The critical photoperiod mediating the maintenance and termination of diapause is positively correlated with latitude (r 2 = 0.977) among six populations from southern (30–31° N), intermediate (40° N), and northern (46–49° N) latitudes in North America. The developmental response to unnaturally short and to unnaturally long photoperiods declines with increasing latitude, so that longer critical photoperiods are associated with a downward rather than a lateral shift in the photoperiodic response curve. Exotic light and dark cycles of varying period (T) with a short (10 h) photophase and a scotophase ranging from 14 (T = 24) to 62 (T = 72) h, reveal two geographic patterns: a decline in perturbability of the photoperiodic clock with increasing latitude, and no change with latitude in the 21-h period of rising and falling development with increasing T. These results show (1) that there is a rhythmic component to photoperiodic time measurement in W. smithii, (2) that the period of this rhythm is about 21 h in all populations, and (3) that more northern populations show decreasing responsiveness to photoperiod and increasing stability against perturbation by exotic period lengths (T > 24). Previous studies on W.␣smithii indicate that this single temperate species of a tropical and subtropical genus has evolved from south to north. We therefore conclude that the evolution of increasing critical photoperiod in W. smithii during its adaptive radiation into North America has more likely involved the amplitude and not the period of the underlying circadian pacemaker. The post-dispersal fate of seeds and fruit (diaspores) of three vertebrate-dispersed trees, Crataegus monogyna, Prunus mahaleb and Taxus baccata, was studied in the Andalusian highlands, south-eastern Spain. Exclosures were used to quantify separately the impact of vertebrates and invertebrates on seed removal in relation to diaspore density and microhabitat. The three plant species showed marked differences in the percentage of diaspores removed, ranging from only 5% for C. monogyna to 87% for T. baccata. Although chaffinches (Fringilla coelebs) fed on diaspores, rodents (Apodemus sylvaticus) were the main vertebrate removers of seed and fruit. Two species of ant (Cataglyphis velox and Aphaenogaster iberica) were the only invertebrates observed to remove diaspores. However, the impact of ants was strongly seasonal and they only removed P. mahaleb fruit to any significant extent. While removal of seed by rodents was equivalent to predation, ants were responsible for secondary dispersal. However, their role was limited to infrequent, small-scale redistribution of fruit in the vicinity of parent trees. Rodents and ants differed in their use of different microhabitats. Rodents foraged mostly beneath trees and low shrubs and avoided open areas while the reverse was true of ants. Thus, patterns of post-dispersal seed removal will be contigent on the relative abundance and distribution of ants and rodents. Studies which neglect to quantify separately the impacts of these two guilds of seed removers may fail to elucidate the mechanisms underlying patterns of post-dispersal seed removal. The coincidence of both increased seed deposition by the main avian dispersers (Turdus spp.) and increased seed predation with increasing vegetation height suggested that selection pressures other than post-dispersal seed predation shape the spatial pattern of seed dispersal. Rather than providing a means of escaping post-dispersal seed predators, dispersal appears to direct seeds to microhabitats most suitable for seedling survival. Nevertheless, the reliance of most vertebrate-dispersed trees on regeneration by seed and the absence of persistent soil seed banks imply that post-dispersal seed predators may exert a strong influence on the demography of the plants whose seeds they consume. Even where microsites are limited, the coincidence of the most suitable microhabitats for seedling establishment with those where seed predation is highest provide a means by which selective seed predators can influence community composition. Temperature dependent changes in the mode of energy metabolism and in acid-base status were studied in the range from −1.7 to 26 °C in two populations of Arenicola marina collected in summer as well as in winter from intertidal flats of the North Sea (boreal) and the White Sea (subpolar). Extreme temperatures led to an accumulation of anaerobic end products, indicating the existence of both a low and a high critical temperature, beyond which anaerobic metabolism becomes involved in energy production. In summer animals from the North Sea the high critical temperature was found at temperatures above 20 °C, and the low critical temperature below 5 °C. Latitudinal or seasonal cold adaptation lead to a more or less parallel shift of both high and low critical temperature values to lower values. Between critical temperatures intracellular pH declined with rising temperature. Slopes varied between −0.012 and −0.022 pH- units/°C. In summer animals from the North Sea, the slope was slightly less than in White Sea animals, but differences appeared independent of the season. However, slopes were no longer linear beyond critical temperatures. A drop in intracellular pH at low temperatures coincided with the accumulation of volatile fatty acids in the body wall tissue of North Sea animals. A failure of active pHi adjustment is held responsible for the reduced ΔpHi/ΔT at temperatures above the high critical temperature. Extracellular pH was kept constant over the whole temperature range investigated. The ability of North Sea animals to adapt to temperatures beyond the critical temperature is poor compared to White Sea specimens. The larger range of temperature fluctuations at the White Sea is seen as a reason for the higher adaptational capacity of the subpolar animals. A hypothesis is proposed that among other mechanisms critical temperature values are set by an adjustment of mitochondrial density and thus, aerobic capacity. Accepted: 20 August 1996  相似文献

A method based upon targetting of intro-gressed markers in a Phomopsis-resistant line (R) of cultivated sunflower, issuing from a H. argophyllus cross was used to mark the Phomopsis resistance regions. Our study was based upon 203 families derived from a cross between an inbred line susceptible to Phomopsis (S1) and the introgressed resistant line (R). Families were checked for Phomopsis resistance level in a design with replicated plots and natural infection was re-inforced by pieces of contaminated stems. Thirty four primers were employed for RAPD analysis. Out of 102 polymorphic fragments between (S1) and H. argophyllus, seven were still present in (R) suggesting that they marked introgressions of H. argophyllus into (R). The plants were scored for the presence or absence of 19 fragments obtained from five primers, and the relationships between the presence/absence of fragments in plants and Phomopsis resistance/susceptiblity in the progenies was determined by using an analysis of variance. We found that at least two introgressed regions, as well as favourable factors from sunflower, contributed to the level of Phomopsis resistance in cultivated sunflower. Receptors monitoring muscle force innervate the opener muscle apodeme in the walking legs of the blue crab, Callinectes sapidus. Biocytin backfills reveal 9–15 bipolar neurons with somata as large as 60 μm positioned at the distal end of the apodeme. Sensory endings insert into the apodeme and are in series with the opener muscle. The axons of these neurons form the opener apodeme sensory nerve that merges with the most distal branch of the opener motor nerve. Recordings reveal that the receptors are not spontaneously active nor do they respond to passive muscle stretch. Isometric muscle contraction evoked by stimulating the opener excitor motor neuron is the adequate stimulus for receptor firing. Most significant is the finding that during contraction, over a wide range of forces, the firing rate of individual receptors closely parallels the rate of change of isometric force. The peak instantaneous frequency typically occurs at the force derivative maximum, but not at maximum force development. Variation in fruit production and pre-dispersal seed predation by Argyresthia conjugella was studied in␣four populations of Sorbus aucuparia in northern Sweden.␣The number of infructescences, fruits per infructescence, consumed seeds and developed unattacked seeds per fruit were scored in marked trees from 1984 to 1990. The results showed that the number of fruits produced in each population determined the number of seed predators occurring in the host population, as the yearly number of seed predators was significantly and positively correlated with yearly number of fruits, in all but one population. The seed predators showed a delay in response to variation in number of fruits produced. This lag in response resulted in a large proportion of fruits being attacked and seeds consumed in a bad fruiting year that followed a good fruiting year, and vice versa. The proportion of fruits attacked and seeds consumed was largest in the population showing the greatest between-year variation in fruit production and lowest in the population showing the lowest between-year variation in fruit production. Furthermore, the individuals within the former population were synchronised, while they were not in the latter population. These results contradict one of the possible explanations of mast-seeding, where large synchronised between-year variation is supposed to reduce pre-dispersal seed predation. Instead, differences in attraction of the seed predator to differences in fruit crop size could explain the observed difference in seed predation between the two populations with opposite fruiting patterns. Within each population, irrespective of year, the proportion of fruits attacked and seeds consumed was independent of a tree's fruiting display. Therefore, trees with high fruit production, despite harbouring the largest number of seed predators, produced the largest number of developed seeds in absolute numbers, compared to trees that produced few fruits.
