Cytogeographical variability among 564 plants from 26 populations of Turnera sidoides subsp. pinnatifida in mountain ranges of central Argentina was analysed with meiotic chromosome counts and flow cytometry and is described at regional and local scales. Populations were primarily tetraploids (2n = 4x = 28), although diploid (2n = 2x = 14), hexaploid (2n = 2x = 42), and mixed populations of diploids and triploids (2n = 3x = 21) were also found. Diploids, triploids, and hexaploids were fewer in number and restricted to narrow areas, while tetraploids were the most common and geographically widespread cytotype. Diploids grew at higher altitudes and in colder and wet locations; tetraploids had the broadest ecological spectrum, while hexaploids occurred at the lowest altitudes and in drier conditions. The cytotypes were also spatially segregated at a microgeographical scale. Diploids grew in the piedmont, tetraploids were in the adjacent valley, and in the contact zone of both cytotypes, patches of diploids and triploids were found. At a regional scale, the distribution of the cytotypes may be governed by a combination of ecological and historical variables, while segregation in the contact zone may be independent of the selective environment because the cytotypes are unable to coexist as a result of reproductive exclusion. To investigate pigeons as a potential source of pathogenic yeast species, 47 samples of pigeon droppings and 322 samples from pigeon cloacae were evaluated. The samples were also collected from trees located near the pigeon habitats, in the city of Fortaleza, Ceará, Northeast Brazil. In addition, we evaluated the in vitro antifungal susceptibility of these environmental Cryptococcus strains to amphotericin B, azoles and caspofungin. C. neoformans var. neoformans (n = 10), C. laurentii (n = 3), Candida spp. (n = 14), Rhodotorula mucilaginosa (n = 6) and Trichosporon sp. (n = 3) were isolated from pigeon droppings. In contrast, only Candida spp. (n = 4), Trichosporon sp. (n = 3) and R. mucilaginosa (n = 2) were recovered from cloacae specimens. Only Candida glabrata (n = 1) was recovered from plant samples. Azole resistance was detected in only one environmental strain of Cryptococcus, which was resistant to itraconazole (MIC = 1 μg/ml). As expected, all Cryptococcus strains were resistant to caspofungin. In summary, the present study confirms that urban pigeons are a potential source of Cryptococcus spp. and other pathogenic yeasts. Additionally, antifungal resistance was observed in one environmental strain of Cryptococcus neoformans var. neoformans in Northeast Brazil.  相似文献

Interspecific hybridization is a primary cause of extensive morphological and chromosomal variation and plays an important role in plant species diversification. However, the role of interploidal hybridization in the formation of hybrid swarms is less clear. Epidendrum encompasses wide variation in chromosome number and lacks strong premating barriers, making the genus a good model for clarifying the role of chromosomes in postzygotic barriers in interploidal hybrids. In this sense, hybrids from the interploidal sympatric zone between E. fulgens (2n = 2x = 24) and E. puniceoluteum (2n = 4x = 56) were analyzed using cytogenetic techniques to elucidate the formation and establishment of interploidal hybrids. Hybrids were not a uniform group: two chromosome numbers were observed, with the variation being a consequence of severe hybrid meiotic abnormalities and backcrossing with E. puniceoluteum. The hybrids were triploids (2n = 3x = 38 and 40) and despite the occurrence of enormous meiotic problems associated with triploidy, the hybrids were able to backcross, producing successful hybrid individuals with broad ecological distributions. In spite of the nonpolyploidization of the hybrid, its formation is a long‐term evolutionary process rather than a product of a recent disturbance, and considering other sympatric zones in Epidendrum, these events could be recurrent.  相似文献

Palytoxin (PTX), one of the most potent and chemically complex marine toxins, is predominantly found in zoanthid corals and sporadically in dinoflagellates. Its biosynthesis and metabolic pathways are largely unknown. However, the widespread occurrence of the toxin in phylogenetically distinct marine organisms is consistent with its production by microorganisms and subsequent accumulation in the food chain. To investigate a possible microbial origin, bacteria from two zoanthid corals (Palythoa caribaeorum, Zoanthus pulchellus) and one sponge (Neofibularia nolitangere) were isolated. More than 250 bacteria were screened for hemolysis using a newly developed PTX-screening assay of which 7% showed PTX-like hemolytic activity. 16S rRNA gene sequencing revealed that these bacterial isolates belonged to strains of Bacillus cereus group (n = 11) as well as the genera Brevibacterium (n = 4) and Acinetobacter (n = 2). The results indicate the presence of Na+/K+-ATPase toxins and possibly PTX in hemolytic bacteria from P. caribaeorum.  相似文献

Three Australian native animal species yielded 60 samples composed of three indigenous ticks. Hosts included twelve koalas, two echidnas and one wombat from Victoria, and ticks were of the species Ixodes tasmani (n = 42), Bothriocroton concolor (n = 8) and B. auruginans (n = 10), respectively. PCR screening and sequencing detected a species of Coxiella, sharing closest sequence identity to C. burnetii (>98%), in all B. auruginans, as well as a species of Rickettsia, matching closest to R. massiliae, in 70% of the same samples. A genotype sharing closest similarity to Rickettsia bellii (>99%) was identified in three female B. concolor collected from one of the echidnas. Three samples of I. tasmani, taken from three koalas, yielded different genotypes of Rickettsiella. These results represent the first detection of the three genera in each tick species and identify a high level of previously undetected bacterial diversity in Australian ticks.  相似文献

Intertribal somatic hybrids between Brassica napus (2n = 38, AACC) and a dye and medicinal plant Isatis indigotica (2n = 14, II) were obtained by fusions of mesophyll protoplasts. From a total of 237 calli, only one symmetric hybrid (S2) and five asymmetric hybrids (As1, As4, As6, As7 and As12) were established in the field. These hybrids showed some morphological variations and had very low pollen fertility. Hybrids S2 and As1 possessed 2n = 52 (AACCII), the sum of the parental chromosomes, and As12 had 2n = 66 (possibly AACCIIII). Hybrids As4, As6 and As7 were mixoploids (2n = 48–62). Genomic in situ hybridization analysis revealed that pollen mother cells at diakinesis of As1 contained 26 bivalents comprising 19 from B. napus and 7 from I. indigotica and mainly showed the segregation 26:26 at anaphase I (AI) with 7 I. indigotica chromosomes in each polar group. Four BC1 plants from As1 after pollinated by B. napus resembled mainly B. napus in morphology but also exhibited some characteristics from I. indigotica. These plants produced some seeds on selfing or pollination by B. napus. They had 2n = 45 (AACCI) and underwent pairing among the I. indigotica chromosomes and/or between the chromosomes of two parents at diakinesis. All hybrids mainly had the AFLP banding patterns from the addition of two parents plus some alterations. B. napus contributed chloroplast genomes in majority of the hybrids but some also had from I. indigotica. Production of B. napusI. indigotica additions would be of considerable importance for genome analysis and breeding.  相似文献

We developed four microsatellite DNA loci to test for multiple paternity of black rockfish, Sebastes inermis, from the Seto Inland Sea of Japan. All loci showed a high degree of polymorphism (number of alleles per locus = 10–14, expected heterozygosity = 0.80) and discriminating power (probability of identity index = 3.71 × 10−6, exclusion probability = 0.999) in unrelated wild specimens (n = 32). Genotypic assignment of five dams (109–220 mm in total length) and 50 embryos from each dam (n = 50) indicated that four dams were mated with a single sire. Only for one dam and three of her embryos we could not exclude multiple paternity.  相似文献

Length–weight relationships (LWR) were estimated for five most abundant fish species occurring in the Utinga State Park within the metropolitan area of Belém, State of Pará, Northern Brazil. Data were obtained from specimens of Curimata knerii (n = 75), Gasteropelecus levis (n = 54), Hemigrammus rodwayi (n = 82), Hyphessobrycon bentosi (n = 69), and Pristobrycon calmoni (n = 76) collected in July 2013 with seines (1.5 mm mesh), sieves (1 mm mesh), and gill nets (40–120 mm mesh) in the main waterbodies. All LWRs are novel for science, increasing knowledge on biological information of the Neotropical freshwater fish.  相似文献

Mayflies (Insecta: Ephemeroptera) are common inhabitants of streams throughout the Appalachian Mountains. Headwater mayfly assemblages were evaluated with respect to regional landuse disturbances (coal mining and residential) in eastern Kentucky, USA. Estimates of mayfly taxa richness and relative abundance were compared at 92 sites represented by least-disturbed reference (REF; n = 44), residential only (RESID; n = 14), mixed residential and mining (MINED/RESID; n = 14), and mining only (MINED; n = 20) landuse categories. A total of 48 species from 27 genera and 9 families were identified; Ephemerella, Epeorus, Ameletus, Cinygmula, and Paraleptophlebia comprised the core 5 genera most frequently encountered at REF sites. These same genera (among others) were often reduced or extirpated from other landuse categories. Mean mayfly richness and relative abundance were significantly higher at REF sites compared to all other categories; MINED sites had significantly lower metric values compared to RESID and MINED/RESID sites. Relative mayfly abundance was most strongly correlated to specific conductance (r = 0.72) compared to total habitat score (r = 0.59), but relationships varied depending on landuse category. Non-metric multidimensional scaling (for mayfly taxa) and principal components analysis (for environmental variables) separated REF sites strongly from most other sites. The results indicate that expected mayfly communities are disappearing from streams where mining disturbance and residential development has occurred and because of the long-term impacts incurred by both landuses, recovery is uncertain.  相似文献

Reproductive interference can shape regional distribution patterns in closely related species, if prezygotic isolation barriers are weak. The study of such interaction could be more challenging in nuptial gift‐giving species due to the direct nutritional effects on both sexes of both species during copulation. We mapped the distribution of two sister bush‐cricket species, Pholidoptera aptera and Pholidoptera transsylvanica, at the northern margin of their overlapping ranges in Europe, and with a behavioral experiment, we tested the possibility of heterospecific mating. We found a very rare coexistence of species locally (0.5%, n = 391 sites) with mostly mutually exclusive distribution patterns, resulting in a mosaic pattern of sympatry, whereas they occupied the same climate niche in forest‐dominated mountain landscape. Over 14 days of a mating experiment with seven mixed groups of conspecifics and heterospecifics (n = 56 individuals in total), the number of received spermatophores per female was 3–6 in P. aptera and 1–7 in P. transsylvanica. In total, we found 8.1% of heterospecific copulations (n = 99 transferred spermatophores with genetic identification of the donor species), while we also confirmed successful transfer of heterospecific sperms into a female's reproductive system. Because bush‐cricket females also obtain required nutrition from a heterospecific spermatophylax what should increase their fitness and fecundity, we suggest that their flexibility to mate with heterospecifics is beneficial and drives reproductive interference. This may substantially limit the reproductive success of the less frequent species (P. transsylvanica), coupled with eventual detrimental effects from hybridization, and result in the competitive exclusion of that species from their areas of coexistence.  相似文献

为探讨国产毛茛科(Ranunculaceae)驴蹄草属(Caltha L.)植物的细胞学特征,对驴蹄草(C.palustris L.)3个居群和花葶驴蹄草(C.scaposa Hook.f.&Thoms.)5个居群进行了细胞学研究。驴蹄草贵州纳雍居群的染色体数目为2n=32(四倍体),两个云南中甸居群的染色体数目均为2n=64(八倍体)。花葶驴蹄草四川红原、康定、石渠居群的染色体数目均为2n=32(四倍体),该数目为首次报道;西藏林芝和云南德钦居群的染色体数目均为2n=64(八倍体)。驴蹄草的染色体比花葶驴蹄草大。这两种植物的32或64条染色体分别以4条或8条为单位大致能够排列为8组同源染色体,但同一组内的染色体经常具有明显的异形性(heteromorphy),不同居群的核型组成多少具有差异。同时,还分析了驴蹄草和花葶驴蹄草的不同倍性细胞型在我国的地理分布式样。  相似文献

Length–weight relationships (LWRs) were evaluated for Badis badis (n = 25), Sperata seenghala (n = 26), Labeo gonius (n = 34), Rasbora rasbora (n = 30), Bagarius bagarius (n = 24), Gagata cenia (n = 27), Glyptothorax stoliczkae (n = 24) and Channa orientalis (n = 28) from the Ravi River tributary in North India. Altogether 218 samples of eight species were obtained between May and November 2014 using cast nets and gill nets. LWRs for these species were unknown to FishBase, and new maximum lengths were recorded for two of these species.  相似文献

Mitotic chromosome numbers of 32 populations belonging to 23 species of the genus Lessingianthus H.Rob. (Vernonieae, Asteraceae) were determined. The chromosome number of all examined plants was found to be based on x = 16. The numbers observed varied from 2n = 32 to 2n = 176. The results include the first report of the chromosome number for 11 species: L. lanatus (2n = 32), L. varroniifolius (2n = 32), L. cataractarum (2n = 64), L. intermedius (2n = 64), L. argenteus (2n = 96), L. centauropsideus (2n = 96), L. profusus (2n = 96), Lessingianthus sp. nov. 1 (2n = 96), Lessingianthus sp. nov. 2 (2n = 128), L. robustus (2n = 160), and L. macrocephalus (2n = 176). New chromosome numbers were found in the four other species: L. rubricaulis, L. laniferus, and L. sellowii were tetraploid with 2n = 64, while L. oxyodontus was hexaploid with 2n = 96. B chromosomes were observed in L. coriaceus and L. varroniifolius. Lessingianthus macrocephalus (2n = 11x = 176) is reported as the first case of an odd polyploid and the higher chromosome number of Lessingianthus. The significance of the results is discussed in relation to chromosomal data available for the genus.  相似文献

The English grain aphid, Sitobion avenae (Fabricius), is one of the most important insect pests causing substantial yield losses in wheat production in China and other grain-growing areas in the world. The efficient utilization of wheat genes for resistance to English grain aphid (EGA) provides an efficient, economic and environmentally sound approach to reduce the yield losses. In the present study, the wheat line C273 (Triticum durum AABB, 2n = 4x = 28), is resistant to EGA in greenhouse and field tests. To identify the resistance gene, designated RA-1 temporarily, C273 was crossed with susceptible genotype Poland 305 (T. polonicum, AABB, 2n = 4x = 28). The F1, F2 and F2:3 lines were tested with EGA in the field and greenhouse. The results indicated that RA-1 is a single dominant gene, closely linked to the microsatellite markers (SSR) Xwmc179, Xwmc553 and Xwmc201 on chromosome 6AL at genetic distances of 3.47, 4.73 and 7.57 cM, respectively. The three SSR markers will be valuable in marker-assisted selection for resistance to EGA as well as for cloning this gene in the future.  相似文献

We evaluated the uterus and ovaries of owl monkeys (Aotus azarai infulatus) via gynecological ultrasound examination. We evaluated the subjects in 2 different time periods. The first period (P1) was characterized by the absence of mating, with daily examinations, during 4 mo (n = 10). At the end of P1, we paired the subjects for 30 d, but without ultrasonographic evaluation. The second period (P2) was characterized by the presence of mating, with examinations once a week, during 7 consecutive months (n = 9). We evaluated the uterus and ovaries in sagittal and transverse scans, using a 5–12 MHz linear array probe. The uterine volume (UV) was directly proportional to the number of previous parturitions. The right ovary volume (RtOV) is greater than the left (LtOV) in P1 and P2. There is a positive correlation (p < 0.05) between the females’ mass, RtOV (r = 0.28) and LtOV (r = 0.16).  相似文献

The n‐alkane composition in the leaf cuticular waxes of natural populations of Bosnian pine (Pinus heldreichii), Austrian pine (P. nigra), and Macedonian pine (P. peuce) was compared for the first time. The range of n‐alkanes was wider in P. nigra (C16 – C33) than in P. heldreichii and P. peuce (C18 – C33). Species also diverged in abundance and range of dominant n‐alkanes (P. heldreichii: C23, C27, and C25; P. nigra: C25, C27, C29, and C23; P. peuce: C29, C25, C27, and C23). Multivariate statistical analyses (PCA, DA, and CA) generally pointed out separation of populations of P. nigra from populations of P. heldreichii and P. peuce (which were, to a greater or lesser extent, separated too). However, position of these species on the basis of n‐alkane composition was in accordance neither with infrageneric classification nor with recent molecular and terpene investigations.  相似文献

Salmonella enterica in cattle production systems may be associated with important human and animal disease issues. However, tremendous diversity exists among Salmonella recovered, and more information is needed about strains of greatest potential health concern, particularly those that are multidrug resistant (MDR). By characterizing Salmonella isolates from commercial feedlot pens, this study aimed to evaluate the strain diversity and prevalence of MDR Salmonella from different types of composite pen samples. Antimicrobial susceptibility profiles, serotype, and presence or absence of the integron-encoded intI1 gene were determined for 530 Salmonella isolates recovered using composite rope (n = 335), feces (n = 59), and water (n = 136) samples from 21 pens in 3 feedlots. The study investigated only pens with available isolates from multiple sample types. Most isolates (83.0%) of the 19 Salmonella serotypes identified were susceptible or intermediately susceptible to all the antimicrobials evaluated. Resistance to sulfisoxazole (14.9%), streptomycin (3.8%), and tetracycline (3.6%) were the most common. None of the isolates tested positive for a class 1 integron, and only 2.5% were resistant to multiple antimicrobials. All the MDR isolates, namely, serotypes Uganda (n = 9), Typhimurium (n = 2), and Give (n = 2), were resistant to at least five antimicrobials. Most MDR isolates (n = 11) were from two pens during 1 week within one feedlot. Overall, many Salmonella isolates collected within a pen were similar in terms of serotype and antimicrobial susceptibility regardless of sample type. However, MDR Salmonella and rare serotypes were not recovered frequently enough to suggest a general strategy for appropriate composite sampling of feedlot cattle populations for Salmonella detection and monitoring.  相似文献

Chromosome rearrangements may result in both decrease and increase of chromosome numbers. Here we have used comparative chromosome painting (CCP) to reconstruct the pathways of descending and ascending dysploidy in the genus Boechera (tribe Boechereae, Brassicaceae). We describe the origin and structure of three Boechera genomes and establish the origin of the previously described aberrant Het and Del chromosomes found in Boechera apomicts with euploid (2n = 14) and aneuploid (2n = 15) chromosome number. CCP analysis allowed us to reconstruct the origin of seven chromosomes in sexual Bstricta and apomictic B. divaricarpa from the ancestral karyotype (n = 8) of Brassicaceae lineage I. Whereas three chromosomes (BS4, BS6, and BS7) retained their ancestral structure, five chromosomes were reshuffled by reciprocal translocations to form chromosomes BS1‐BS3 and BS5. The reduction of the chromosome number (from x = 8 to x = 7) was accomplished through the inactivation of a paleocentromere on chromosome BS5. In apomictic 2n = 14 plants, CCP identifies the largely heterochromatic chromosome (Het) being one of the BS1 homologues with the expansion of pericentromeric heterochromatin. In apomictic B. polyantha (2n = 15), the Het has undergone a centric fission resulting in two smaller chromosomes – the submetacentric Het′ and telocentric Del. Here we show that new chromosomes can be formed by a centric fission and can be fixed in populations due to the apomictic mode of reproduction.  相似文献

Ability to survive the low pH of the human stomach is considered be an important virulent determinant. It was suggested that the unique acid tolerance of Shigella boydii 18 CDPH, the strain implicated in a 1998 outbreak, may have played an important role in surviving the acidic food (bean salad). The strain was capable of inducing arginine-dependent acid-resistance (ADAR) pathway. This pathway was assumed to be absent in Shigella sp. Here, we have examined occurrence and efficacy of ADAR pathway in 21 S. boydii strains obtained from the American Type Culture Collection (ATCC) along with strains of S. flexneri (n = 7), S. sonnei (n = 4), and S. dysenteriae (n = 2). The eight S. boydii strains were able to induce ADAR to survive the acid challenge at pH 2.0; additional 8 strains could tolerate acid challenge at pH 2.5 but not at pH 2.0. The remaining five S. boydii strains were not able to induce ADAR pathway and could not survive acid challenge even at pH 2.5. ADAR pathway also appears to be present in all four Shigella sp. Shigella ADAR pathway was induced when cells were grown under partial oxygen pressure while its expression in E. coli required mere fermentative growth on glucose.  相似文献
